Termites adjust their response to entomopathogenic fungi according to the profile of fungal volatile organic compounds (VOCs). This study demonstrates the pathogenicity of Metarhizium anisopliae, Beauveria bassiana and Isaria fumosorosea (=Paecilomyces fumosoroseus) towards the Formosan subterranean termite, Coptotermes formosanus Shiraki (Isoptera: Rhinotermitidae). Using no-choice assays, M. anisopliae was found to be highly virulent (LT50 3.10 d) when compared to B. bassiana (LT50 6.62 d) and I. fumosorosea (LT50 12.39 d). Also using choice assays, the foraging behavior of C. formosanus was determined in the presence of pathogenic fungi. The highly pathogenic fungi (M. anisopliae) elicited a repellent response, causing most of the termites to forage in a safe zone farthest from the fungal source. This repellency resulted in relatively low mortality similar to the controls. The repellency of M. anisopliae conidia can be used to protect human belongings and timber from termites. While I. fumosorosea cultures were not repellent to C. formosanus workers, the termites were highly susceptible to infection. Electroantennographic responses of workers showed approximately 47% and 78% lower level of response to conidia of B. bassiana and I. fumosorosea, respectively, as compared to M. anisopliae. The VOC profile of repellent cultures of M. anisopliae mainly consisted of paraffins (60.97%), while the major proportion of the I. fumosorosea profile consisted of branched and cyclic alkanes (84.41%). From the above findings, we conclude that the incorporation of I. fumosorosea may increase the control potential of bait.  相似文献

The Formosan subterranean termites, Coptotermes formosanus, were incubated in soils collected from various locations on the island of Hawaii. More than 70% of the termites died within 2 weeks in seven of nine soil samples collected from Kamuela. In control soil termites remained alive after 4 weeks. Entomophthora coronata, Metarrhizium anisopliae, Aspergillus flavus, and an unidentified bacterium were isolated from 60 to 100% of dead termites incubated in the pernicious soils. E. coronata and M. anisopliae were pathogenic to C. formosanus. The innocuous soil from Hilo became pernicious to C. formosanus after it was inoculated with E. coronata or M. anisopliae.  相似文献

Although preference and utilization of clay have been studied in many higher termites, little attention has been paid to lower termites, especially subterranean termites. The Formosan subterranean termite, Coptotermes formosanus Shiraki, can modify its habitat by using clay to fill tree cavities. Here, the biological significance of clay on C. formosanus was investigated. Choice tests showed that significantly more termites aggre- gated in chambers where clay blocks were provided, regardless of colony group, observation period, or nutritional condition (fed or starved). No-choice tests showed that clay had no observable effect on survivorship, live or dry biomass, water content, and tunneling activity after 33-35 d. However, clay appeared to significantly decrease filter paper consumption (dry weight loss). Active particle (sand, paper, and clay) transport behavior was observed in both choice and no-choice tests. When present, clay was preferentially spread on the substrate, attached to the smooth surfaces of the containers, and used to line sand tunnels. Mechanisms and potential application of clay attraction are discussed.  相似文献

Enterobacter cloacae, one of the indigenous gut bacteria of the Formosan subterranean termite (Coptotermes formosanus), was genetically modified with a transposon Tn5 vector containing genes (tcdA1 and tcdB1) encoding orally insecticidal proteins from the entomopathogenic bacterium Photorhabdus luminescens subsp. laumondii TT01, a symbiont of the entomopathogenic nematode Heterorhabditis bacteriophora, for termite control. In the laboratory, termites were fed filter paper inoculated with the recombinant bacteria. The chromosomal expression of the introduced genes showed that there were insecticidal activities against termite workers and soldiers challenged with the transformed bacteria. After termites were fed recombinant bacteria, the termite mortality was 3.3% at day 5, and it increased from 8.7% at day 9 to 93.3% at day 29. All the dead termites contained the recombinant bacteria in their guts. Transfer of the recombinant bacteria occurred between donor workers (initially fed recombinant bacteria) and recipient workers (not fed). More than 20% of the recipient termites ingested recombinant bacteria within 2 h, and 73.3% of them had ingested recombinant bacteria after 12 h. The method described here provides a useful alternative for sustainable control of the Formosan subterranean termite (C. formosanus) and other social insects, such as the imported red fire ant (Solenopsis invicta).  相似文献

This study examined the effect of diet, experimental design, and length of time in the laboratory on intercolonial agonism among Formosan subterranean termite, Coptotermes formosanus Shiraki, colonies. In pairings of 12 C. formosanus Shiraki colonies collected in an urban forest, there was no significant reduction in survival of termites in 30 out of 59 colony pairs compared to colony controls, but there was <50% survival in 18 colony pairs and <10% survival in six colony pairs. There was no correlation between the level of aggressive behavior and the laboratory diet of the termites. Effect of bioassay design and length of time in the laboratory was evaluated in three colony pairs where tests were first conducted on the day of field collection, then colony pairs were retested every 7 days. Aggressive behavior decreased over time in both bioassays, but it tended to decrease more rapidly in the Petri dish tests. The rapid loss of agonism in groups of termites kept in the laboratory demonstrates that changes in environmental factors affect intercolonial agonism. The Formosan subterranean termite, Coptotermes formosanus Shiraki, is a costly invasive urban pest in warm and humid regions around the world. Feeding workers of the Formosan subterranean termite genetically engineered yeast strains that express synthetic protozoacidal lytic peptides has been shown to kill the cellulose digesting termite gut protozoa, which results in death of the termite colony. In this study, we tested if Melittin, a natural lytic peptide from bee venom, could be delivered into the termite gut via genetically engineered yeast and if the expressed Melittin killed termites via lysis of symbiotic protozoa in the gut of termite workers and/or destruction of the gut tissue itself. Melittin expressing yeast did kill protozoa in the termite gut within 56 days of exposure. The expressed Melittin weakened the gut but did not add a synergistic effect to the protozoacidal action by gut necrosis. While Melittin could be applied for termite control via killing the cellulose-digesting protozoa in the termite gut, it is unlikely to be useful as a standalone product to control insects that do not rely on symbiotic protozoa for survival.  相似文献

Separate groups of subterranean termites (Reticulitermes sp.) were exposed to whole cultures of Beauveria bassiana, Gliocladium virens, or Metarhizium anisopliae. Individuals were removed after varying time intervals and hindgut contents were plated onto potato dextrose agar. Viable spores first appeared in the hindguts within 8 hr of exposure. Fungi reisolated from the hindguts of diseased termites were pathogenic of healthy termites. Histological examination showed that invasion of the hemocoel by M. anisopliae occurred exclusively through direct invasion of the integument ca. 24 hr after death. B. bassiana invaded, primarily through the alimentary tract, ca. 12 hr prior to termite death.  相似文献

Cuticular hydrocarbon patterns of the Formosan subterranean termite, Coptotermes formosanus Shiraki, were similar among colonies from the same geographical location. Hydrocarbon patterns of Florida colonies were easily distinguished from those of Hawaii colonies by using canonical discriminant analysis. Groups of termites from the same colony did not fight one another when placed in an arena. Intercolonial aggression was not recorded among C. formosanuspopulations from Florida but three colonies from Hawaii fought with the other Hawaiian and three Florida colonies. Of the 12 colonies (six each from Florida and Hawaii) tested, 3 Florida colonies did not direct or receive aggression from any other colony. Cuticular hydrocarbon patterns were not correlated with agonistic behavior.  相似文献

Experiments were conducted to examine competitive interactions between the Formosan subterranean termite, Coptotermes formosanus Shiraki (FST), and the eastern subterranean termite, Reticulitermes flavipes (Kollar) (EST), using groups of termites with different worker:soldier proportions. Experiments were conducted using three connected test chambers: an FST chamber, an unoccupied center chamber, and an EST chamber. When groups of FST were comprised of 20% soldiers versus 2% EST soldiers, only 8% of center chambers were occupied exclusively by EST. When groups of FST were comprised of 10% soldiers versus 1% EST soldiers, 44% of center chambers were occupied exclusively by EST. When the only food source was located in the center chamber, 60% of center chambers were occupied by both species. FST did not completely displace EST in any of these experiments.  相似文献

Dogs were trained to detect Eastern subterranean termites, Reticulitermes flavipes (Kollar), using the United States Customs method of scent detection dog training modified with a food reward. Dogs were tested with various numbers of Eastern subterranean termites placed in vented PVC containers. Trained dogs were 95.93% accurate in finding > or = 40 Eastern subterranean termite workers (positive indications) and incorrectly indicated the presence of termites in 2.69% of the containers without termites. Multiple species of termites [dark southern subterranean, R. virginicus (Banks); Formosan subterranean, Coptotermes formosanus Shiraki; powderpost, Cryptotermes cavifrons Banks; and southeastern drywood termites, Incisitermes snyderi (Light)], were similarly evaluated. Dogs trained to locate Eastern subterranean termites were also 100% accurate in finding dark southern subterranean termites, 98.89% accurate in finding Formosan subterranean termites, 97.33% accurate in finding powderpost termites, and 88.89% accurate in finding southeastern drywood termites. Dogs were able to discriminate live termites from non-termite material. Trained dogs' false response rate was 25.33% to Eastern subterranean termite-damaged wood, 6.67% to American cockroaches, Periplaneta americana (L.), and 2.67% to Florida carpenter ants, Camponotus floridanus Buckley.  相似文献

The Formosan termite Coptotermes formosanus Shiraki is a well-known invasive pest that causes severe damage to wooden structures in many parts of the world. Although several studies examined its phylogeographic patterns using a few mitochondrial genes, the phylogenetic relationships among C. formosanus are poorly understood because of the small number of mutations known among its mitochondrial genes. To provide a useful genetic tool for further analyses, we analyzed the complete mitochondrial genome sequence of C. formosanus using specimens collected from three isolated islands in the Ryukyu Archipelago of Japan. The circular mitogenome of these termites consisted of genes encoding 22 transfer RNAs, two ribosomal RNAs, and 13 mitochondrial proteins, as is the case for most animal mitochondrial genomes. The G + C content was 34.1%, and the total length varied slightly between 16,234 and 16,236 base pairs. The complete mitochondrial genomes of the three populations were more than 99.9% identical to each other and showed differences at six nucleotide positions. The COII, 12S rRNA, and 16S rRNA genes that are commonly used for phylogenetic analyses revealed only one substitution or no substitutions. The mitogenome sequences determined here should contribute to the design of new molecular markers for the clarification of the historical distribution process of C. formosanus and for further phylogenetic analyses with this and related termite species.  相似文献

Use of proper application methods and formulations of termiticides are im- portant to reduce their negative impact to the environment. In this study, we conducted laboratory experiments to determine the effect of localized treatments with commercial dust and liquid formulations of fipronil against Formosan subterranean termites, Coptoter- mes formosanus Shiraki. The test arena consisted of a specially designed 16-chambered structure with a center chamber connected to 5 foraging chambers that themselves were connected to 10 additional foraging chambers. One peripheral chamber received a liquid or dust treatment and termites were released in the center chamber. Results showed that 〉 91% of the termites were dead within the 9-d test period despite the localized treatment of only 1 foraging chamber. Termites that were still alive after 9 d were transferred to an untreated dish and held for 10 more days. The majority of those termites were dead and the rest were moribund on day 19. Regardless of the specific dish treated, both formulations of fipronil were found to be highly efficacious. Termites did not exhibit repellency to either formulation. Our results suggest that localized (or spot) treatment with either commer- cially available dust or liquid formulations of fipronil can be a viable option for control of a termite infestation where complete soil drenching is not desirable.  相似文献

Indigenous gut bacteria of the Formosan subterranean termite (Coptotermes formosanus Shiraki, Isoptera: Rhinotermitidae) were used as shuttle systems to deliver, express and spread foreign genes in termite colonies. The gut bacterium Enterobacter cloacae was transformed with a recombinant plasmid (pEGFP) containing genes encoding ampicillin resistance and green fluorescent protein (GFP). In laboratory experiments, termite workers and soldiers from three colonies were fed with filter paper inoculated with transformed bacteria. Transformed bacteria were detected in termite guts by growing the entire gut flora under selective conditions and checking the cultures visually for fluorescence. We demonstrated that (1) transformed bacteria were ingested within a few hours and the GFP gene was expressed in the termite gut; (2) transformed bacteria established a persistent population in the termite gut for up to 11 weeks; (3) transformed bacteria were efficiently transferred throughout a laboratory colony, even when the donor (termites initially fed with transformed bacteria) to recipient (not fed) ratio was low; (4) transformed E. cloacae were transferred into soil; however, they did not accumulate over time and the GFP plasmid was not transferred to other soil bacteria. In the future, transgenic bacteria may be used to shuttle detrimental genes into termite colonies for improved pest control.  相似文献

While hybridization of an invasive species with a native species is a common occurrence, hybridization between two invasive species is rare. Formosan subterranean termites (Coptotermes formosanus) and Asian subterranean termites (C. gestroi) are both ecologically successful and are the two most economically important termite pests in the world. Both species have spread throughout many areas of the world due to human activity; however, their distributions overlap in only three narrow areas because of distinct ecological requirements. In south Florida, where C. formosanus and C. gestroi are both invasive, the dispersal flight seasons of both species overlapped for the first time on record in 2013 and 2014. Pairings of heterospecific individuals were readily observed in the field and C. gestroi males preferentially engaged in mating behavior with C. formosanus females rather than females from their own species. In the laboratory, heterospecific and conspecific pairings had an equal colony establishment rate, but heterospecific incipient colonies had twice the growth rate of conspecific incipient colonies, suggesting a potential case of hybrid vigor. As all pre-zygotic barriers were lifted between the two species in the field, the apparent absence of post-zygotic barriers in the laboratory raises the possibility for introgressive hybridization in south Florida. While laboratory observations remain to be confirmed in the field, and the alate hybrid fertility is currently unknown, our results raise a tangible concern about the hybridization of two major destructive pest species. Such hybridization would likely be associated with a new economic impact.  相似文献

Summary The Formosan subterranean termite (Coptotermes formosanus Shiraki) was first sighted in the cities of New Orleans and Lake Charles, Louisiana, in 1966. Its distribution in the historic French Quarter of New Orleans 23 years later (1989) and in 1991 and 1992 is reported here using counts of alates from light-traps. Light-trappings revealed a Quarter-wide distribution of the termite.C. formosanus shows an extended pause in flight behavior in the middle of the two-month flight season that is predictable and independent of weather. A colony-level mechanism, namely, developmental inhibition of nymphs by alates, may regulate this intermission in flight. The sex-ratio of light-trapped alates varies over time, but is generally female-biased, especially at the height of the flight season.  相似文献

Swarming by alates of the Formosan subterranean termite, Coptotermes formosanus Shiraki, occurs around dusk during April–June in Louisiana and adjoining states. Dispersal is the primary purpose for swarming. We studied the premating behavior, following swarming. Loss of wings, especially in females, was essential for successful formation of tandem pairs between the two sexes. No calling was observed and apparently a long-range volatile sex pheromone, reported in some other species of termites, may not be involved in mate attraction in C. formosanus. Males moving randomly use their antennae to detect a female, and, together with maxillary and labial palps, contact is maintained with her while in tandem. Previously mated males paired with virgin females showed tandem behavior. The incidence was very low when both males and females were previously mated and then paired. Males with complete ablation of antennae did not form tandem pairs. Ablation of 10 terminal antennal segments or the labial palps caused a significant increase in the time to initiate tandem behavior. Electrophysiological recordings from gustatory sensilla on both antennae and maxillary palps revealed increased neural activity in response to female abdominal tip extracts compared to activity elicited by a solvent control. Analysis of extracts of male and female abdominal tips using high-performance liquid chromatography showed a peak unique to the female extract. We discuss the presence of a nonvolatile chemical in female C. formosanus and its involvement in the specific premating behavior.  相似文献

This study was the first to use DNA sequencing data to infer the origin of a Formosan subterranean termite, Coptotermes formosanus Shiraki, infestation. Cytochrome oxidase II gene sequence was used to define eight specific maternal lineages from 14 geographic locations across the southeastern United States, Hawaii, and China. One maternal lineage, A, was unique to all the Atlanta collections and one New Orleans collection, which indicated that termites at these sites shared a common maternal ancestor. The impact of interstate commerce in terms of Formosan subterranean termite introductions is addressed as is the industry response needed to arrest the spread of these termites via commercial traffic in used railroad cross ties.  相似文献

The ability of termites to attack solid wood and plywood treated with quaternary ammonia compounds and common fire retardants was evaluated. The plywood and solid-wood specimens treated with either monoammonium phosphate (MAP), diammonium phosphate (DAP), ammonium sulfate (AS), didecyl dimethyl ammonium chloride (DDAC), or didecyl dimethyl ammonium tetrafluoroborate (DBF) were subjected to termite resistance tests using the subterranean termites Coptotermes formosanus Shiraki under laboratory conditions. The lowest mass losses and the highest termite mortalities were obtained for the solid-wood and plywood specimens treated with DDAC and DBF. Higher termite mortalities were seen in the plywood specimens treated with the fire retardants when compared to the solid-wood specimens. The MAP, DAP, and AS treatments lowered the mass losses in both solid-wood and plywood specimens in comparison with control specimens; however, DBF and DDAC protected specimens well against termite attack at both concentration levels tested.  相似文献
