In higher plants the vacuolar K(+)-selective (VK) channel was identified solely in guard cells. This patch-clamp study describes a 40 pS homologue of the VK channel in Beta vulgaris taproot vacuoles. Reciprocal gene exchange between cultivated sugar beet and wild beets in seed production areas is probably the reason for the occurence of weed beets in sugar beet production fields. Therefore, when releasing transgenic sugar beet plants into the environment, gene transfer to wild beets ( Beta vulgaris ssp. maritima ) has to be considered. In this study the transfer of BNYVV- (beet necrotic yellow vein virus) resistance and herbicide-tolerance genes from two transgenic sugar beet lines that were released in field experiments in 1993 and 1994 in Germany to different wild beet accessions was investigated. In order to evaluate the consequences of outcrossing, manual pollinations of emasculated wild beet plants with homozygous transgenic sugar beet plants were performed. In the resulting hybrids the transgenes were stably inherited according to Mendelian law. Gene expression in leaves and roots of the hybrids was in the same range as in the original transgenic sugar beet plants. Moreover, it was found that in one of the wild beet accessions, transfer and expression of the BNYVV resistance gene did considerably increase the level of virus resistance.  相似文献

Commercial strains of sugar beet, and breeders' lines of Beta vulgaris and B. vulgaris subsp. maritima , were exposed to infection with SBY and SBYN strains of beet yellows virus, using varying numbers of Myzus persicae as vectors, so as to cause a wide range of symptoms. The severity of the symptoms was assessed by scoring for superficial veinal necrosis ('etch'). All varieties of sugar beet tested were susceptible to infection, but the severity of the symptoms varied, particularly between the cultivated and wild beet types.
The scores for severity of symptoms made in the glasshouse were positively correlated with similar scores made in a field experiment using two cultivated and two wild beet types. The symptom scores were also positively correlated with losses in root and sugar yields caused by the virus.  相似文献

Bacteroides amylophilus has growth requirements for Na(+), PO(4) (3-), K(+), and small quantities of Mg(2+). No requirement could be shown for Ca(2+) in media previously found growth-yield-limiting for Bacteroides succinogenes. Deletion of Co(2+), Mn(2+), Cl(-), or SO(4) (2-) did not affect growth. Quantitative studies indicate that Na(+), K(+), and PO(4) (3-) have differing effects on the growth of B. amylophilus. A concentration of sodium and potassium ions affects both growth rate and growth yield, whereas a phosphate concentration markedly affects growth yield, but affects growth rate only slightly, if at all. The sodium requirement of B. amylophilus is absolute. It cannot be replaced by K(+), Li(+), Rb(+), or Cs(+). The latter three monovalent cations are toxic to B. amylophilus if supplied to the organism at Na(+)-replacing concentrations. K(+) is inactive at similar concentrations. The K(+) requirement of B. amylophilus may be satisfied by Rb(+). The concentration of Na(+) required by B. amylophilus for abundant growth suggests that B. amylophilus should be considered a slightly halophilic organism. The results suggest that Na(+) may be a more frequent requirement among terrestial bacteria obtained from relatively low-salt environments than has been previously believed.  相似文献

Sugar beet plants (Beta vulgaris L. cv. F58-554H1) were germinated and cultured under standardized environmental conditions. The effects of K deficiency on photosynthetic and respiratory CO2 exchange rates of attached leaves were studied under conditions of low Na supply by withholding both Na and K from the culture medium at cut-off (28 days after planting). Compost has been proposed as a means of simultaneously diverting organic materials from landfills while producing a valuable product that improves tilth, organic matter content and nutrient supply of agricultural soils. Composts manufactured from different source materials may have markedly different properties however, even if they meet all regulatory requirements. We compared the capacity of composts made from three different combinations of organic wastes (horse manure and bedding, mink farm wastes, municipal solid waste (MSW) and sewage sludge) along with clarifier solids from a chemo-thermomechanical pulp mill, to enhance the growth of tomato (Lycopersicon esculentum L.) seedlings grown in nutrient-poor organic potting soil. Germination and seedling emergence of tomatoes, cress (Lapidium sativum L.) or radish (Raphanus sativus L.) were tested to assess phytotoxicity of the four amendments. Mink farm compost and horse manure compost stimulated root and shoot growth of tomato seedlings but MSW compost and pulp mill solids were strongly inhibitory. MSW compost and unamended potting soil also inhibited seedling emergence and pulp mill solids produced stunting and deformities in radish and cress seedlings. Both toxic constituents and nutrient imbalances may be responsible for the growth-inhibiting effects of these amendments. Application of pulp mill solids to agricultural soil without composting may lead to deleterious effects on vegetable crops.  相似文献

Soil amendment with two types of composts: animal manure (AC) and vegetable waste (VC) induced composts have potential to alleviate Cd toxicity to maize in contaminated soil. Therefore, Cd mobility in waste water irrigated soil can be addressed through eco-friendly and cost effective organic soil amendments AC and VC that eventually reduces its translocation from polluted soil to maize plant tissues. The comparative effectiveness of AC and VC at 3% rate were evaluated on Cd solubility, its accumulation in maize tissues, translocation from root to shoot, chlorophyll contents, plant biomass, yield and soil properties (pH, NPK, OM). Results revealed that the addition of organic soil amendments significantly minimized Cd mobility and leachability in soil by 58.6% and 47%, respectively in VC-amended soil over control. While, the reduction was observed by 61.7% and 57%, respectively when AC was added at 3% over control. Comparing the control soil, Cd uptake effectively reduced via plants shoots and roots by 50%, 46% respectively when VC was added in polluted soil. However, Cd uptake was decreased in maize shoot and roots by 58% and 52.4% in AC amended soil at 3% rate, respectively. Additionally, NPK contents were significantly improved in polluted soil as well as in plant tissues in both composts amended soil Comparative to control, the addition of composts significantly improved the maize dry biomass and chlorophyll contents at 3% rate. Thus, present study confirmed that the addition of animal manure derived compost (AC) at 3% rate performed well and might be consider the suitable approach relative to vegetable compost for maize growth in polluted soil.  相似文献

An assay was developed to characterize the kinetic parameters of the Na(+)-K+ pump of rat erythrocytes under conditions as physiological as possible. Changes in the red cell Na+ and Rb+ content were determined in Na+ media (containing 2.5 mM inorganic phosphate (PO4) as a function of cell Na+ (2-8 mmol/l) and extracellular Rb+ (0.2-5 mM). Evaluation of the data revealed that under these conditions the Na(+)-K+ pump mediates, in addition to forward running 3 Nai+: 2 Rbo+ exchange, 1 Ki+:Rbo+ exchange and pump reversal (3 Nao+:2 Ki+ exchange). The two latter modes of Na(+)-K+ pump operation are accelerated by PO4 and lowering of cell Na+. At physiological cation and PO4 concentrations, 1Ki+:Rbo+ exchange contributes by 30-60% to total ouabain-sensitive Rb+ uptake. Thereby, the stoichiometry of ouabain-sensitive Na+ net-extrusion to Rb+ uptake is reduced to values between 1.0 and 0.5. Only at cell Na+ contents above 20 mmol/l the Na+:Rb+ stoichiometry approaches the value of 3:2 = 1.5. At certain constellations of Nai+ and Rbo+ the Na(+)-K+ pump cannot perform any net-transport of Na+ and K+ (Rb+). These equilibrium points are not far from those expected from thermodynamic considerations. The results demonstrate that in normal rat erythrocytes the reversible reaction cycle of the Na(+)-K+ pump runs in several modes of operation. The "abnormal" modes complicate the interpretation of unidirectional fluxes mediated by the Na(+)-K+ pump.  相似文献

A 7-year study located in Prince Edward Island, Canada, examined the influence of compost and manure on crop yield and nematode populations. The compost used in this study consisted of cull waste potatoes, sawdust, and beef manure in a 3:3:1 ratio, respectively. No plant-parasitic nematodes were detected in samples collected from windrow compost piles at 5- and 30-cm depths prior to application on field plots. Low population densities of bacterial-feeding nematodes were recovered from compost windrows at the 5-cm depth. Field plots of potato (Solanum tuberosum cv. Kennebec) received compost applied at 16 metric tonnes per hectare, or beef manure applied at 12 metric tonnes per hectare. An adjacent trial with barley (Hordeum vulgare cv. Mic Mac) received only the compost treatment. In both trials the experimental design was a complete randomized block with four replicates. Data averaged over seven growing seasons indicated that population levels of root-lesion nematodes (primarily Pratylenchus penetrans) were higher in root-zone soil in potato plots treated with either compost or manure compared to the untreated control plots. The soil amendments did not affect root-knot nematode (Meloidogyne hapla) population densities in the potato plots, but clover-cyst nematodes (Heterodera trifolii) were more numerous in the root-zone soils of barley treated with compost compared to the untreated plots. Numbers of bacterial-feeding nematodes (primarily Diplogaster lheritieri) were greater in soil in potato plots treated with manure and in soil around barley roots than in untreated plots. Total yields of potato tubers averaged over seven growing seasons increased by 27% in the plots treated with either compost or manure. Grain yields of barley also were increased by 12% when compost was applied. These results indicated that organic amendments increased crop yields, but the impacts on different nematode species varied and usually increased soil population levels.  相似文献

Plant species differ in nutrient uptake efficiency. With a pot experiment, we evaluated potassium (K) uptake efficiency of maize (Zea mays L.), wheat (Triticum aestivum L.), and sugar beet (Beta vulgaris L.) grown on a low-K soil. Sugar beet and wheat maintained higher shoot K concentrations, indicating higher K uptake efficiency. Wheat acquired more K because of a greater root length to shoot dry weight ratio. Sugar beet accumulated more shoot K as a result of a 3- to 4-fold higher K influx as compared to wheat and maize, respectively. Nutrient uptake model NST 3.0 closely predicted K influx when 250 mg K kg?1 were added to the soil, but under-predicted K influx under low K supply. Sensitivity analysis showed that increasing soil solution K concentration (CLi) by a factor of 1.6–3.5 or buffer power (b) 10- to 50-fold resulted in 100% prediction of K influx. When both maximum influx (Imax) and b were increased by a factor of 2.5 in maize and wheat and 25 in sugar beet, the model could predict measured K influx 100%. In general, the parameter changes affected mostly calculated K influx of root hairs, demonstrating their possible important role in plant K efficiency.  相似文献

Gene flow and introgression from cultivated to wild plant populations have important evolutionary and ecological consequences and require detailed investigations for risk assessments of transgene escape into natural ecosystems. Sugar beets (Beta vulgaris ssp. vulgaris) are of particular concern because: (i) they are cross-compatible with their wild relatives (the sea beet, B. vulgaris ssp. maritima); (ii) crop-to-wild gene flow is likely to occur via weedy lineages resulting from hybridization events and locally infesting fields. Using a chloroplastic marker and a set of nuclear microsatellite loci, the occurrence of crop-to-wild gene flow was investigated in the French sugar beet production area within a 'contact-zone' in between coastal wild populations and sugar beet fields. The results did not reveal large pollen dispersal from weed to wild beets. However, several pieces of evidence clearly show an escape of weedy lineages from fields via seed flow. Since most studies involving the assessment of transgene escape from crops to wild outcrossing relatives generally focused only on pollen dispersal, this last result was unexpected: it points out the key role of a long-lived seed bank and highlights support for transgene escape via man-mediated long-distance dispersal events.  相似文献

Membrane-bound pyrophosphatase of the hyperthermophilic bacterium Thermotoga maritima(Tm-PPase), a homologue of H(+)-translocating pyrophosphatase, was expressed in Escherichia coli and isolated as inner membrane vesicles. In contrast to all previously studied H(+)-PPases, both native and recombinant Tm-PPases exhibited an absolute requirement for Na(+) but displayed the highest activity in the presence of millimolar levels of both Na(+) and K(+). Detergent-solubilized recombinant Tm-PPase was thermostable and retained the monovalent cation requirements of the membrane-embedded enzyme. Steady-state kinetic analysis of pyrophosphate hydrolysis by the wild-type enzyme suggested that two Na(+) binding sites and one K(+) binding site are involved in enzyme activation. The affinity of the site that binds Na(+) first is increased with increasing K(+) concentration. In contrast, only one Na(+) binding site (K(+)-dependent) and one K(+) binding site were involved in activation of the Asp(703) --> Asn variant. Thus, Asp(703) may form part of the K(+)-independent Na(+) binding site. Unlike all other membrane and soluble PPases, Tm-PPase did not catalyze oxygen exchange between phosphate and water. However, solubilized Tm-PPase exhibited low but measurable PP(i)-synthesizing activity, which also required Na(+) but was inhibited by K(+). These results demonstrate that T. maritima PPase belongs to a previously unknown subfamily of Na(+)-dependent H(+)-PPase homologues and may be an analogue of Na(+),K(+)-ATPase.  相似文献

The organic produce industry is gaining popularity with consumers because of the perception for healthier foods and the environmental benefits of this agricultural practice. Common amendments in organic agriculture include compost and compost tea, the latter being a relatively new product in North America. The main objective of this study was to assess the effectiveness of ruminant and municipal solid waste compost and compost teas made from these composts on selected soil, leaf and fruit parameters of raspberries. Generally, foliar compost tea application was as effective as compost addition in raspberry production. Concentrations of K in leaf and fruits were significantly lower (p?=?0.05) in compost tea treated raspberries. Furthermore, the compost tea increased leaf Na compared to compost amendments which suggested that raspberries preferentially take up Na via foliar applications compared to root Na applications. The yield, total antioxidant capacity of fruit, and vitamin C content of fruit were not affected by treatment but differed greatly among years. Precipitation also varied greatly among years as did soil nutrient concentrations and may have influenced yield, total antioxidant capacity, and vitamin C content.  相似文献

A new genetic character of the beet Beta vulgaris L., named stem color, was described and studied genetically. This character was shown to be dominant trait and monogenically inherited. The first-year beet plants with the genotype Stc/_have red leafstalks, weakly colored central rib, and colored storage root; however, the root itself is not colored. The second-year plants have a red-colored low third of the floral shoot. The plants with the genotype stc/stc are uncolored. The Stc gene was localized to the first linkage group at a distance of 17.5 +/- 2.1 crossing over units from the gene B (Bolting), which controls the annual-perennial habit of beet.  相似文献

The rate of CO2 gas exchange, transpiration, stomatal conductivity, and efficiency of the use of water were assessed in leaves of the sugar beet (Beta vulgaris var. Saccharifera (Alef) Krass.) sprayed with 40% methanol, against the background of increasing water deficiency in soil. A decrease in the negative impact of water deficiency in experimental plants was shown to be due to the larger stress-resistance of the photosynthetic apparatus, higher rate of photosynthesis, and more effective use of water.  相似文献

The relation between the level of genetic variation and the rate of recombination per physical unit was investigated in sea beet (Beta vulgaris subsp. maritima). The rate of recombination per physical unit was estimated indirectly through marker density in an RFLP linkage map of sugar beet. From this map, we also selected RFLP markers covering two of the nine chromosomes in Beta. The markers were used to estimate the level of genetic variation in three populations of sea beet, two from Italy and one from England. Two estimates of genetic variation were employed, one based on the number of alleles in the sample and the other on heterozygosity. A statistically significant positive correlation was found between recombination rate and genetic variation. Several theoretical explanations for this are discussed, background selection being one. A correlation similar to this has been observed previously in Drosophila, one that was higher than what we obtained for Beta. This is consistent with various biological differences between the two species.  相似文献

Previous work with membrane vesicles has demonstrated an absolute dependence on K+ for proton translocation by the inorganic pyrophosphatase (H(+)-PPase: EC 3.6.1.1) from the vacuolar membrane (tonoplast) of higher plants. Using intact vacuoles from sugar beet (Beta vulgaris) storage tissue, we have monitored PP1-dependent currents by patch clamp in 'whole vacuole' mode. Serial K+ substitutions were made at both tonoplast faces. The results show that K+ activation occurs only at the cytosolic face.  相似文献

Introgression arising from crop-to-wild gene flow provides novel sources of genetic variation in plant species complexes. Hybridization within the Beta vulgaris species complex is of immediate concern; crop lineages ( B .  vulgaris ssp. vulgaris ) hybridize easily with their wild relatives ( B .  vulgaris ssp. maritima ) thereby threatening wild beet gene diversity with genetic swamping. Hybridization 'hotspots' occur in European seed production areas because inland ruderal wild beets occur and reproduce in sympatry with cultivated beets. We studied gene flow occurring between seed-producing cultivars and ruderal wild B .  vulgaris in southwestern France to determine whether feral beets, arising from unharvested cultivated seed, represent an opportunity for crop-to-wild gene flow. We surveyed 42 inland ruderal beet populations located near seed production fields for nucleo-cytoplasmic variation and used a cytoplasmic marker diagnostic of cultivated lines. Occurrence of cultivated-type cytoplasm within ruderal populations clearly reflected events of crop seed escape. However, we found no genetic signatures of nuclear cultivated gene introgression, which suggests past introgression of cultivated cytoplasm into a wild nuclear background through seed escape rather than recent direct pollen flow. Overall, patterns of genetic structure suggested that inland ruderal wild beet populations act as a metapopulation, with founding events involving a few sib groups, followed by low rates of seed or pollen gene flow after populations are established. Altogether, our results indicate that a long-lived seed bank plays a key role in maintaining cultivated-type cytoplasm in the wild and highlight the need for careful management of seed production areas where wild and cultivated relatives co-occur.  相似文献

Sugar beet seedlings (Beta vulgaris L. cv. Monohill) were cultivated for 3 weeks at different root and shoot temperatures and the plasma membranes (PM) from roots were purified by aqueous two-phase partitioning and analyzed for lipid composition and ATPase activities. Lipid analyses, undertaken immediately after PM purification from the roots, showed that a low root zone temperature (10 degrees C) decreased the ratio between the major lipids phosphatidylcholine (PC) and phosphatidylethanolamine (PE). A low temperature in the root environment increased the mol% of PE and decreased the mol% of phosphatidic acid (PA), independent on the shoot growth temperature. A low temperature also decreased the mol% of linoleic acid (18:2) and increased mol% of linolenic acid (18:3) in the analyzed lipid classes, especially in PC and PE. The ratio between acyl chain lipids and protein generally increased in PM from roots grown at 10 degrees C, compared with higher temperature. The changes in lipid composition correlated with changes in ATPase activities, detected as hydrolyses of MgATP. The kinetic parameters, K(m) and V of the PM H(+)ATPase in roots increased at a low cultivation temperature, independent on shoot temperature. Moreover, Arrhenius analyses showed that the transition temperature was independent of both root or shoot growth temperature at 10-24 degrees C, whereas the activation energy of the ATPase was dependent on the growth temperature of the root, and independent on shoot temperature. Thus, acclimation processes can take place in roots, irrespective of the shoot temperature.  相似文献

Na(+) uptake by plant roots has largely been explored using species that accumulate little Na(+) into their shoots. By way of contrast, the halophyte Suaeda maritima accumulates, without injury, concentrations of the order of 400 mM NaCl in its leaves. Here we report that cAMP and Ca(2+) (blockers of nonselective cation channels) and Li(+) (a competitive inhibitor of Na(+) uptake) did not have any significant effect on the uptake of Na(+) by the halophyte S. maritima when plants were in 25 or 150 mM NaCl (150 mM NaCl is near optimal for growth). However, the inhibitors of K(+) channels, TEA(+) (10 mM), Cs(+) (3 mM), and Ba(2+) (5 mM), significantly reduced the net uptake of Na(+) from 150 mM NaCl over 48 h, by 54%, 24%, and 29%, respectively. TEA(+) (10 mM), Cs(+) (3 mM), and Ba(2+) (1 mm) also significantly reduced (22)Na(+) influx (measured over 2 min in 150 mM external NaCl) by 47%, 30%, and 31%, respectively. In contrast to the situation in 150 mm NaCl, neither TEA(+) (1-10 mM) nor Cs(+) (0.5-10 mM) significantly reduced net Na(+) uptake or (22)Na(+) influx in 25 mM NaCl. Ba(2+) (at 5 mm) did significantly decrease net Na(+) uptake (by 47%) and (22)Na(+) influx (by 36% with 1 mM Ba(2+)) in 25 mM NaCl. K(+) (10 or 50 mM) had no effect on (22)Na(+) influx at concentrations below 75 mM NaCl, but the influx of (22)Na(+) was inhibited by 50 mM K(+) when the external concentration of NaCl was above 75 mM. The data suggest that neither nonselective cation channels nor a low-affinity cation transporter are major pathways for Na(+) entry into root cells. We propose that two distinct low-affinity Na(+) uptake pathways exist in S. maritima: Pathway 1 is insensitive to TEA(+) or Cs(+), but sensitive to Ba(2+) and mediates Na(+) uptake under low salinities (25 mM NaCl); pathway 2 is sensitive to TEA(+), Cs(+), and Ba(2+) and mediates Na(+) uptake under higher external salt concentrations (150 mM NaCl). Pathway 1 might be mediated by a high-affinity K transporter-type transporter and pathway 2 by an AKT1-type channel.  相似文献
