ABSTRACT. The sexual behaviour of male Glossina morsitans morsitans Westwood (Diptera: Glossinidae) towards decoys dosed with sex phero-mone (15,19,23-trimethylheptatriacontane) is described quantitatively from field observations. The flies responded with a standard behavioural pattern after landing. This involved: first, the perception of pheromone; second, orientation on the decoy and genitalia engagement; and third, a lengthy quiescence corresponding to natural copulation. The probability of a fly leaving a decoy decreased during the sequence and was least in the final 'copulatory' phase, when it remained constant under constant conditions. The initial, rapid rates at which flies left decoys were affected mostly by pheromone dose: the lower the dose, the higher the rate. The final, slow rate of leaving was unaffected by pheromone dose, being most affected by environmental stimuli, especially interference from other flies: at high fly densities, final rates of leaving were high. The results are discussed in the context of improving the potential for using pheromone and chemo-sterilant-dosed decoys for tsetse autosterilization.  相似文献

ABSTRACT. The sexual response of a Glossina morsitans morsitans Westw. male towards a decoy consisting of a dead male dosed with synthetic sex pheromone, was depressed by the presence of the cuticular lipids of the decoy male. However, addition of a C24 paraffin, not normally found on or in the cuticle of male or female G.m.morsitans , to a decoy male from which cuticular lipids were removed had a similar effect. Addition of either the C24 paraffin or an extract of male cuticular lipids to decoy females which possess natural sex pheromone in their cuticles, again depressed the sexual responses of males towards the decoys. The n-hexane solvent used in the application of compounds to decoys, itself depressed responses towards natural pheromone on a dead female at volumes in excess of 100μl, presumably because it mixed the surface waxes which masked the pheromone. Occlusion experiments implied that sex pheromone receptors are present on both the tarsi and tibiae of the male. No evidence was found for the existence of a specific mating deterrent on or in the cuticle of male G.m. morsitans.  相似文献

Abstract Reciprocal crosses were performed with Glossina swynnertoni and Glossina morsitans morsitans and with G. swynnertoni and Glossina morsitans centralis , using strains that carried marker genes in all three linkage groups. Glossina swynnertoni males can inseminate, but not fertilize, G.m.morsitans; all other crosses produced some fertile females. Hybridization did not cause sex ratio distortion among F{ flies. Most F and backcross females were fertile, but all F, males were sterile. Sterility among backcross males was also high (99% in BXj, 85% in Bx2, and about 50% in Bx3 to Bx5). Chromosome transmission by hybrid females usually conformed to Mendelian expectations, but genetic recombination was lower than observed in G.m.morsitans. The reduction in fertility among backcross females was not associated with heterozygosity in any linkage group. Sterility among hybrid and backcross males was associated with heterozygosity of sex chromosomes and probably autosomes. The results support the systematic placement of G.swynnertoni closer to G.mxentralis than to G.m.morsitans.  相似文献

The roles of visual and/or olfactory stimuli in eliciting mating responses from male Glossina morsitans morsitans Westwood were examined, using a system for automatically recording the number and duration of mating strikes made towards decoys, under controlled conditions. The results confirm that there is no olfactory component of the female sex recognition pheromone sensed by the male antennae, and the attraction of males to females appears to be visual. The absence of male-male mating strikes was the result of the absence of female sex-pheromone, rather than the presence of a repellent mating deterrent in the male cuticle. Experiments with coloured, artificial, sex-pheromone-dosed, cotton decoys showed that colour had only weak effects on attractiveness and number of encounters with decoys, and that no colour caused significant enhancement of mating responses over those shown to decoy females.  相似文献

ABSTRACT. Interspecific mating tests among seven tsetse species covering the 'morsitans' and 'palpalis' groups showed a degree of reaction that extends beyond the traditional taxonomic groupings. Tests with either live or dead females as targets produced essentially similar results which shows that differing responses of males are largely the consequence of differences in composition of the female surface cuticular paraffins and not of female behaviour. Gas chromatographic (GLC) analysis of cuticular paraffins tends to confirm this view. Responses of males of different species to dead conspecific males baited with the most active synthetic G.morsitans pheromone were variable. Comparison of behavioural responses and of GLC profiles of cuticular paraffins suggest that the species tested do not share a pheromone structure in common with G.morsitans. Mating tests with G.morsitans males using dead conspecific males baited with a variety of synthetic compounds provided evidence of the molecular configuration required to elicit a mating response. Synthetic 15,19-dimethyltritriacontane induced a suboptimal response in male G.morsitans but is a likely candidate for a sex pheromone in G.austeni. Although not conclusive, a comparison of behavioural evidence and GLC data has permitted speculation on the nature of sex pheromones in tsetse of the 'palpalis' group.  相似文献

Abstract.  Metabolic rate variation with temperature, body mass, gender and feeding status is documented for Glossina morsitans centralis . Metabolic rate [mean ± SE; VCO2= 19.78 ± 3.11 μL CO2 h−1 in males (mean mass = 22.72 ± 1.41 mg) and 27.34 ± 3.86 μL CO2 h−1 in females (mean mass = 29.28 ± 1.96 mg) at 24 °C in fasted individuals] is strongly influenced by temperature, body mass and feeding status, but not by gender once the effects of body mass have been accounted for. A significant interaction between gender and feeding status is seen, similar to patterns of metabolic rate variation documented in Glossina morsitans morsitans . Synthesis of metabolic rate-temperature relationships in G. m. centralis , G. m. morsitans and Glossina pallidipes indicate that biting frequency as well as mortality risks associated with foraging will probably increase with temperature as a consequence of increasing metabolic demands, although there is little evidence for variation among species at present. Furthermore, metabolic rate–body mass relationships appear to be similarly invariant among these species. These data provide important physiological information for bottom-up modelling of tsetse fly population dynamics.  相似文献

Abstract A denitrifying Cytophaga was isolated from soil enriched by anaerobic incubation with glucose, sulfide (S2−), nitrous oxide (N2O), and acetylene (C2H2). Such soil enrichments and pure cultures of the isolated Cytophaga reduced N2O rapidly even in the presence of a normally inhibitory concentration of C2H2 (4 kPa) providing S2− was present (8 μmol/g soil or 0.4 μmol/ml culture). Since C2H2 inhibition of the reduction of N2O is used as a tool in the assay of denitrification, the presence in large numbers of such a Cytophaga may influence the effectiveness of this assay especially in sulfidic environments.  相似文献

The cuticular wax composition of leaves has been analysed in three western European populations (Corsica, central Pyrenees, northern Alps) of Juniperus communis var. saxatilis Pall. (=  J. nana Willd., nom illeg.) and in one population of J. communis L. var. depressa Pursh. from North America (Sierra Nevada). Gas chromatography shows the presence of 13 alkanes in all samples ranging from C23 to C35 with important intraspecific polymorphism in alkane content. The dominant alkanes range from C33 to C35. Alkanes C21 and C22 were found only in Corsica and Sierra Nevada populations. Canonical discriminant analysis separated the J. communis L. var. depressa Pursh. of the population of Sierra Nevada from other populations of J. communis var. saxatilis Pall. on the basis of their higher C31 content and the constant presence of C21 and C22 alkanes. J. communis var. saxatilis Pall. populations from the Pyrenees are close to northern Alps populations characterized by high concentrations of C33, C34 and C35 alkanes. Abstract Two constitutive acetyl-CoA acetyltransferases (3-ketothiolases A and B) were purified from Alcaligenes eutrophus . Enzyme A was active with only acetoacetyl-CoA and 3-ketopentanoyl-CoA, whereas enzyme B was active with all the 3-ketoacyl-CoAs (C4−C10) tested. Enzyme A appeared to be a tetramer ( M r 70 000) with identical subunits ( M r 44 000) and enzyme B had a similar M r of 168 000 (containing M r 46 000 subunits). Enzymes A and B had isoelectric points of 5.0 and 6.4, respectively. The stoichiometry of the reactions catalysed by each enzyme was confirmed. K m values of 44 μM and 394 μM for acetoacetyl-CoA, and 16 μM and 93 μM for CoA, were determined with enzymes A and B, respectively. Enzymes A and B gave K m values of 1.1 mM and 230 μM, respectively, for acetyl-CoA. The condensation reaction was potently inhibited by CoA in both cases.  相似文献

ABSTRACT. The effects of age and hunger on the responses of male Glossina morsitans morsitans Westwood and G.pallidipes Austen to freeze-killed female decoys, were examined in the laboratory. In both species, activity, estimated as the total number of interactions between males and decoys, increased with both age and hunger. Interactions were divided into short-stay (<60 s) and long-stay, full copulatory responses. In both species, young, unfed males were significantly less likely to attempt to copulate with a decoy after encounter than were fed males. Among fed males the proportion of interactions that proceeded to full copulatory attempts did not change with increasing age, but decreased consistently with increasing hunger. At all ages and hunger levels, G.pallidipes were more active than G.m.morsitans. However, after encountering a decoy, G.pallidipes were less likely to attempt to copulate than G.m.morsitans. In both species the duration of copulatory attempts did not change with age, but declined with increasing hunger. Copulatory attempts by G.pallidipes were significantly shorter than those of G.m.morsitans. The results are discussed in relation to the behaviour of tsetse in response to control devices such as traps and targets.  相似文献

Abstract Measurements of residual haematin in males of Glossina morsitans morsitans Westwood reared in the laboratory at 25oC suggest that blood meal digestion is completed in 4 or 5 days after feeding. However, a high proportion of haematin is present as faecal matter 2 days after feeding and it is concluded that digestion is completed sooner than indicated by the regression of logio haematin on time. Therefore, low levels of residual haematin in field-caught tsetse provide no indication of the frequency with which they feed. For this reason the effects of feeding frequency upon various reproductive parameters in the laboratory have been examined. It is concluded that the best performance is achieved by G.m. morsitans females which ingest four blood meals per inter-larval period and that for a similar performance in G.pallidipes five blood meals are required. The extent to which such feeding frequencies are a reflection of feeding activity in the field are discussed in terms of the biochemical requirements to maintain a reproductive adult female tsetse in positive energy balance.  相似文献

Abstract Hyphomicrobium X was grown on a range of mixtures of methylamine and ethanol at various dilution rates in continuous cultures. It was shown that both substrates were utilized simultaneously at all dilution rates below μmax ethanol, the biomass being the sum of the biomass obtained during growth on the single substrates alone. No evidence was observed for a redistribution of the carbon-flow nor the ability to utilize the 'poorer' growth substrate-ethanol, at dilution rates greater than μmax ethanol. Work is presented which suggests that the assimilatory pathway enzymes for either the C1- or the C2-compound are regulated coordinately, but separately from the dissimilatory pathway enzymes associated with that compound.  相似文献

Plant cuticle is an extracellular lipid-based matrix of cutin and waxes, which covers aerial organs and protects them from many forms of environmental stress. We report here the characterization of CER8 / LACS1 , one of nine Arabidopsis long-chain acyl-CoA synthetases thought to activate acyl chains. Mutations in LACS1 reduced the amount of wax in all chemical classes on the stem and leaf, except in the very long-chain fatty acid (VLCFA) class wherein acids longer than 24 carbons (C24) were elevated more than 155%. The C16 cutin monomers on lacs1 were reduced by 37% and 22%, whereas the C18 monomers were increased by 28% and 20% on stem and leaf, respectively. Amounts of wax and cutin on a lacs1-1 lacs2-3 double mutant were much lower than on either parent, and lacs1-1 lacs2-3 had much higher cuticular permeability than either parent. These additive effects indicate that LACS1 and LACS2 have overlapping functions in both wax and cutin synthesis. We demonstrated that LACS1 has synthetase activity for VLCFAs C20–C30, with highest activity for C30 acids. LACS1 thus appears to function as a very long-chain acyl-CoA synthetase in wax metabolism. Since C16 but not C18 cutin monomers are reduced in lacs1 , and C16 acids are the next most preferred acid (behind C30) by LACS1 in our assays, LACS1 also appears to be important for the incorporation of C16 monomers into cutin polyester. As such, LACS1 defines a functionally novel acyl-CoA synthetase that preferentially modifies both VLCFAs for wax synthesis and long-chain (C16) fatty acids for cutin synthesis.  相似文献

The present work was carried out to evaluate how plant growth and cultural practices influence the amount and composition of cuticular waxes on leaves of rose cultivars. The total amount of cuticular wax per leaf area was higher for rose cultivar Apollo Parade than for Charming Parade. Both cultivars had waxes dominated by alkanes, with the major alkanes being the C31 and C33 homologues. Primary alcohols were the next most abundant constituent class, with C26 as the dominant homologue. Compared with Charming Parade, Apollo Parade had higher proportions of its total wax load as primary alcohols but lower acids and aldehydes. The proportion of alkanes in the total load on these cultivars was similar. Commercially produced roses are routinely treated with paclobutrazol (PBZ) to retard growth. PBZ treatments caused a 10% increase in total wax load and changes in the proportions of certain wax constituents within 11 days of application. Notable was an increase in the total proportion of acids in the total load 25 days after PBZ application, primarily because of increased C28 acids. An alternative method of retarding plant growth is production of roses under limited water availability. When Apollo Parade roses experienced periods of moderate drought stress during production, the wax load per leaf area increased 14 and 8% above control levels at 24 and 38 days after imposition of drought, respectively. Drought caused similar changes in the proportions of individual wax constituents as did PBZ application.  相似文献

Four ecotypes of Phragmites australis from different habitats in northwest China were examined to compare their photosynthetic characteristics. In a swamp ecotype, the Δ 13C value of leaf materials was −34.0‰, and bundle sheath cells contained a small amount of organelles and round-shaped chloroplasts, as being similar to typical C3 plants. In a dune ecotype, the Δ 13C value was −20.9‰ and bundle sheath cells contained oval-shaped chloroplasts with poorly-developed grana. In light and heavy salt meadow ecotypes, Δ 13C values were −30.6‰ and −35.6‰, respectively. The shape of bundle sheath chloroplasts in the light salt meadow ecotype was intermediate between those of the swamp and dune ecotypes. Abundance of bundle sheath organelles in the heavy salt meadow ecotype was intermediate. The swamp ecotype had photosynthetic enzyme activities typical of C3 type plants, whereas the dune ecotype had an increased activity of phosphoenolpyruvate carboxylase (PEPC), a key C4 enzyme, and a decreased ribulose 1,5-bisphosphate carboxylase (Rubisco) activity. The light salt meadow and heavy salt meadow ecotypes had substantial activities of PEPC, which indicates potential for C4 photosynthesis. These data suggest that this species evolved the C3-like ecotype in swamp environments and the C4-like C3-C4 intermediate in dune desert environments, and C3-like C3-C4 intermediates in salt environments.  相似文献

Carbon isotope ratio of leaf dry matter, δ 13C, was measured on species occurring within Baiyin desert community, consisting of valley, slope and ridge microhabitats, and within Shandan desert community, consisting of Gobi desert and seasonal flooded creek microhabitats, in Northwest China. δ 13C of C3 species increased with a decrease in soil water availability, suggesting that water-use efficiency (WUE) increased with decreasing soil moisture, whereas for all C4 species, δ 13C tended to decrease with decreasing soil water availability, suggesting that WUE also increased with decreasing soil moisture. Above results indicated that water-use pattern was conservative under drought for C4 and C3 plants. In this present study, C4 species' occurrences within different microhabitats were investigated and C4 plants were observed to be absent and/or scarce within relatively lower soil moisture microhabitats, whereas they occurred and/or even had a high abundance within relatively higher soil moisture microhabitats, suggesting limited moisture available was a key factor of limiting C4 distribution in arid region of Northwest China.  相似文献

Nodulated faba-beans ( Vicia faba L. var. minor) exhibiting high rates of N2 fixation (133 μmol C2H4 g−1 dry weight h−1), were subjected to water restriction. A loss of C2H2 reduction due to water stress was always associated with a decline of the leghemoglobin content for each of the 4 decreasing values of Ψmod. Electron micrographs showed ultrastructural alterations of the fixing tissue, which affected both partners and increased with the severity of water stress. In the nodule cytosol, the alkaline proteolysis approximately doubled when Ψmod decreased from −0.55 MPa to −1.55 MPa. Concomitantly, an increase of the nodule intracellular pH from 6.3 to 7.0 was observed. Proteolysis was due to serine proteases, exhibiting a pH-optimum of 8 and which actively degraded purified leghemoglobin in vitro (Km=100 μ M ). The degradation of leghemoglobin during water stress may contribute to the loss of C2H2 reduction and may affect the pattern of recovery upon rewatering.  相似文献

An open flow-through gas system was used to investigate the effect of plant age on nitrogenase activity in relation to root respiration (measured as CO2 release) and supra-ambient O2 levels in 24- to 51-day-old, nodulated Pisum sativum L. cv. Bodil. The effect of assaying plants repeatedly was also studied. The respiratory efficiency of nitrogenase [mol CO2 (mol C2H4)−1] and the relative decline in nitrogenase (EC 1.7.99.2) activity in response to introduction of C2H2 in the gas stream were unaffected by plant age. In contrast, the nitrogenase-linked respiration as a proportion of total root respiration increased with time. Accordingly, the specific respiration linked-to growth and maintenace of the noduled root system decreased with time. C2H2 reduction and root respiration were increased by supra-ambient O2 levels, but the tolerance to high O2 concentrations seemed to decrease with plant age. Repeated C2H2 assays on the same plants decreased their rate of growth and N accumulation: in addition, nitrogenase activity and root respiration were somewhat negatively affected. The results indicate that results from experiments with plants of different ages cannot always be directly compared, and that repeated C2H2 assays on the same plants should be applied with caution in physiological work.  相似文献
