为了掌握柚木无性系光合生理与生长性状遗传变异规律,筛选出优良无性系。基于柚木无性系测定林,测定选自不同种源的19个无性系+1个对照的气体交换、叶绿素荧光参数和生长性状,分析柚木无性系光合生理特征及其与生长的关系。结果表明:气体交换、叶绿素荧光参数和树高在柚木不同无性系间、不同种源间都存在极显著差异,除了PSⅡ最大光化学效率(Fv/Fm),其他各性状都具有丰富的遗传变异,但净光合速率等光合生理参数与树高生长没有显著的相关性。结论:早期选出71-7、FS3为光合速率高和树高生长好的优良无性系,起源于印度的柚木资源可作为今后高光效育种选择的重点,需要对光合生理和生长测定进行综合评价才能准确地筛选优良无性系。  相似文献

以美洲黑杨与大青杨杂种无性系1年生扦插苗为材料,测定其低温胁迫中多个生理指标的变化。结果表明,6个杂种无性系SOD、POD和APX活性在低温胁迫中均表现出先上升后下降的趋势,而电解质渗漏率、活性氧含量、CAT和GR活性保持逐渐升高的趋势。无性系间差异显著,其中DU146和DU147电解质渗漏率和活性氧含量最低,而5种抗氧化酶活性均最高,表明其具有较强的耐寒性;而DU135和DU150电解质渗漏率和活性氧含量最高,而5种抗氧化酶活性均最低,表明其耐寒性差。  相似文献

土壤类型是影响植物分布和生产力的重要环境因素,但有关土壤环境异质性对植物抗逆性效应的研究非常缺乏。本研究以美洲黑杨 (Populus deltoids)为对象,以3种典型土壤类型(红壤、黄壤和黄棕壤)为栽培基质,在控制实验条件下,经过三个月的干旱胁迫(25%田间持水量)处理,测定了不同处理条件下美洲黑杨的气体交换速率、抗氧化能力、氮代谢特征、生物量积累与分配特征。研究结果表明,在红壤和黄壤条件下,与对照(75%田间持水量)相比,干旱胁迫显著降低了美洲黑杨各器官的生物量、光合速率、叶片氮同化酶的活性,显著增 加了叶片中过氧化氢、丙二醛和无机氮的含量。在黄棕壤条件下,干旱对美洲黑杨总生物量、光合速率、氮同化酶以及质膜完整性的负面 影响较小,这与其维持较高的超氧化物歧化酶,过氧化物酶和过氧化氢酶的活性相关,也与其生物量分配模式(如提高根冠比)密切相关。 由此可见,生长在黄棕壤条件下的美洲黑杨表现出较强的抗旱能力,这可能与其土壤母质中较高的土壤养分和良好的通气状况相关。因此,就土壤类型而言,与红壤和黄壤相比,黄棕壤提供的土壤环境条件有利于美洲黑杨的抗逆表现和栽培利用。  相似文献

以6年生的10个转TaLEA基因和1个非转基因小黑杨无性系为材料,对其生长量、干形、叶片性状、皮孔性状等15个指标进行测定分析。方差分析与遗传参数计算结果表明:11个小黑杨无性系间各性状达极显著差异水平(P<0.01),各指标表型变异系数处于1.92%~39.98%,遗传变异系数与表型变异系数接近,重复力处于0.621~0.987,表明基因的转入对无性系各性状的生长有一定的影响,但这种影响主要由遗传因素控制。表型相关分析结果表明转基因小黑杨生长量与干形性状、叶片性状、皮孔性状均呈不同程度的相关性,表明多指标共同作用,控制植株生长发育;利用综合评定法,当入选率为10%,初步选出XL-1和XL-9等2个优良无性系,2个无性系的树高、胸径分别比CK高16.55%和15.38%,比平均值高13.38%和19.46%,遗传增益分别为12.18%和17.42%。本研究为转基因杨树的良种选育提供重要的理论依据。  相似文献

黑杨无性系间长期水分利用效率差异的生理基础   总被引:8,自引:0,他引:8  
试验于2003年6月10日至8月20日在温室中进行,采用完全随机区组设计,4个水分处理(充分供水、轻度胁迫、中度胁迫、重度胁迫),6次重复.各水分处理严格控制浇水间隔:充分供水,每2d浇水1次;轻度胁迫,每3d浇水1次;中度胁迫,每4d浇水1次;重度胁迫,每5d浇水1次.每次浇水的量相同,500ml.试验材料为美洲黑杨杂交无性系,父本为哈佛杨(Populus deltoides Bartr.CI.‘Harvard'),母本为山海关杨(Populus deltoides Bartr.CV.‘shanhaiguanensis'),共10个无性系,编号为J1~J10.以当前优良品系108杨(Populus euramericana CV.‘114/69')和中林46杨(Populus deltoides Bartr.CV.‘Zhonglin 46')为对照,编号为CK1、CK2.测定了12个无性系间长期水分利用效率(WUEL)、光合参数(净光合速率Pn、蒸腾速率Tr、气孔导度Gs、瞬时水分利用效率WUEi、最大净光合速率Pmax、光饱和点LSP、羧化效率Ce)及气孔特征参数(气孔频度、气孔大小、上/下表皮气孔数目比)、根冠比的差异,并深入分析了光合参数与WUEL、气孔特征参数与WUEL、根冠比与WUEL及光合参数之间的相互关系.结果表明:无性系间WUEL存在差异,这种差异随水分胁迫的加剧而更加显著.无性系间WUEL的差异在4个水分处理下表现出一致性,即WUEL最优的都是J2,其次为J6、J7、J8、J9.高WUEL的J2等无性系其气孔频度、气孔大小、Gs、Tr、Pn和根冠比在所有无性系中都处于适中的位置.它们的上/下表皮气孔数目比大,Pmax、LSP、Ce明显优于对照.Pmax高、LSP高、Ce大,表明高WUEL的J2等无性系光合能力强.Gs、Tr、Pn适中,表明J2等无性系在保证具有高WUEi的同时能保持较强的光合能力.根冠比适中,表明J2等无性系的光合产物在根、冠之间分配合理,能有充足的根系分布保证水分的供应,同时又有较多的光合产物积累在地上部分.气孔特征参数的差异是导致无性系间光合能力和WUEi差异,并最终导致WUEL差异的一个主要因子.气孔频度和气孔大小适中的无性系,其Gs、Tr、Pn适中,WUEi较高;上/下表皮气孔数目比大的无性系则具有较强的光合能力.无性系间WUEL的差异是一系列具有差异的生理参数共同作用的结果,高WUEL的无性系通常光合能力强、WUEi高、根冠比适中.今后工作中选育高WUE的无性系时,除需关注单个生理参数的突出作用外,更应关注生理参数的协同作用.  相似文献

以白杨双杂交杂种新无性系为主要试验材料,在4种水分胁迫条件下对各无性系生理和生长指标进行分析,结果表明,土壤干旱胁迫对无性系净光合速率,蒸腾速度,气孔导度,叶温与气温差,细胞间CO2浓度等生理指标均产生显著影响。不同指标对水分胁迫的敏感性不同,无性系间在各项生理指标值及随水分胁迫发展而下降幅度均存在着显著差异。  相似文献

以不同地点30个白杨杂种无性系为材料,对其不同树龄的树高和胸径进行调查,对获得数据进行变异分析并构建生长曲线模型,方差分析结果表明地点间、无性系间、地点与无性系交互作用间均达到极显著差异水平(P0.01);树高和胸径表型变异系数变化范围分别为15.34%~23.80%和19.42%~31.88%;所有性状重复力较高,变化范围为0.797 7~0.985 9;模型构建结果表明,根据杨树无性系苗期树高和胸径构建的指数函数模型拟合度r2分别达0.82和0.87,拟合效果较好。利用模型对未测定数据进行估算,无论树高还是胸径,冠县无性系的最初生长量最高,而峰峰的树高和威县的胸径生长速率最快。基因型与环境具有明显的交互作用,不同无性系在相同地点表现差异较大,相同无性系在不同地点也呈现明显差异,表明无性系评价选择需要考虑环境因素,适地适树的进行选择。构建的模型可以为林木生长量预测提供理论依据。  相似文献

东北地区多个长白落叶松(Larix olgensis)种子园已经营建多年,但亲本生长情况往往被忽视。本研究以汪清林业局49个长白落叶松亲本无性系为材料,对其多个生长性状进行测定与遗传变异分析,方差分析结果表明49个无性系间的各测定性状差异均达极显著水平,各测定性状表型变异系数和遗传变异系数变化范围分别为7.71%~31.93%和2.05%~12.55%,重复力变化范围为0.44~0.76,高重复力、高变异系数有利于无性系的评价选择;树高、胸径等大部分生长性状的相关性达极显著水平,表明各生长性状在生长发育中相互影响;主成分分析结果表明第一主成分贡献率为61.44%,主要代表生物量性状;依托主成分分析结果对长白落叶松亲本无性系进行综合评价,初步筛选L76、L5、L7、L12和L4等5个优良无性系,这些无性系树高等性状遗传增益变化范围为3.55%~15.50%,入选无性系可以为长白落叶松良种选育提供材料。  相似文献

以平欧杂种榛3个品种(新榛1号、新榛2号、新榛3号)幼苗为材料,研究不同盐胁迫程度(对照、轻度、中度、重度)对幼苗生长性状、光合荧光特性以及根系构型的影响.结果表明: 3个品种幼苗的新稍长度、基径、叶面积,以及根、茎、叶、总生物量随着盐胁迫的增强而降低,根冠比则增加.中度、重度盐胁迫下3个品种幼苗的净光合速率较对照分别显著降低20.5%和43.2%.轻度、中度、重度盐胁迫下的蒸腾速率和气孔导度较对照分别降低2.0%、16.3%、32.0%和10.2%、35.7%、60.1%.随着盐胁迫的增强,胞间CO2浓度缓慢上升,水分利用效率呈先上升后下降的趋势,并在轻度盐胁迫下达到最大值.3个品种幼苗的初始荧光随着盐胁迫的增强而升高.最大荧光、最大光化学效率、潜在光化学活性、实际光化学效率、电子传递速率、光化学淬灭系数随着盐胁迫的增强而下降,非光化学淬灭系数则随着盐胁迫的增强先升高后降低.根系构型分析表明,盐胁迫导致3个品种幼苗的根系生物量、长度、表面积和体积下降.同一盐胁迫下,新榛2号各径级根系构型参数的下降幅度低于其他2个品种.平欧杂种榛幼苗生长性状、光合荧光特性以及根系构型参数受品种和盐胁迫程度的双重影响.盐胁迫下新榛2号表现出较强的生长与光合生理适应性,其耐盐性优于其他2个品种.  相似文献

为了解红松光合特性的遗传变异规律,本研究利用Lico-6400光合仪,对50个红松无性系的光合指标进行测定和分析。对PK27无性系进行光合指标日变化测定分析,发现PK27的净光合速率(Pn)、气孔导度(Gs)和蒸腾速率(Tr)日变化曲线均呈现典型双峰曲线,胞间CO2浓度(Ci)日变化曲线呈"V"字型曲线。对3个红松无性系的光响应和二氧化碳响应曲线测定表明,各曲线均呈"S"型,并且符合二次曲线模型。在饱和光强下,3个无性系的最大Pn变化范围为P27(15.00μmol·m~(-2)·s~(-1))~PK15(15.99μmol·m~(-2)·s~(-1))。饱和光强和饱和环境二氧化碳条件下(Ca),3个无性系最大Pn变化范围为25.08~27.35μmol·m~(-2)·s~(-1)。对50个红松无性系瞬时光合指标测定结果表明,50个无性系间Pn、Gs、Ci和Tr间均达极显著差异水平(p0.01),各光合指标表型变异系数变化范围为9.04%~38.25%,重复力变化范围为0.57~0.94,属于高重复力。相关性分析表明红松无性系光合指标间、光合指标与环境因子间均呈现极显著相关水平。该研究为红松遗传改良提供了理论依据。  相似文献

Hybrid poplars are an important renewable forest resource known for their high productivity. At the same time, they are highly vulnerable to water stress. Identifying traits that can serve as indicators for growth performance remains an important task, particularly under field conditions. Understanding which trait combinations translate to improved productivity is key in order to satisfy the demand for poplar wood in an uncertain future climate. In this study, we compared hydraulic and leaf traits among five hybrid poplar clones at 10 plantations in central Alberta. We also assessed the variation of these traits between 2‐ to 3‐year‐old branches from the lower to mid‐crown and current‐year long shoots from the mid to upper crown. Our results showed that (1) hybrid poplars differed in key hydraulic parameters between branch type, (2) variation of hydraulic traits among clones was relatively large for some clones and less for others, and (3) strong relationships between measured hydraulic traits, such as vessel diameter, cavitation resistance, xylem‐specific and leaf‐specific conductivity and leaf area, were observed. Our results suggest that leaf size could serve as an additional screening tool when selecting for drought‐tolerant genotypes in forest management and tree improvement programmes.  相似文献

We studied the way 12 traits responded to fish kairomones in a set of 16 Daphnia magna clones derived from four different habitats-two where daphnids co-occur with fish and two without fish. These clones differed widely in their response to predator kairomones, with none of the clones showing a significant response in all traits and all clones showing a response for at least one trait. Most of the clones showed a significant response in one to four traits, with no evidence for an association between different traits. Clones from fish habitats were slightly more responsive to the presence of fish kairomones than clones from fishless locations. We conclude that most clones show an induced response to the presence of their predators (fish) but that there is a large genetic variability with respect to the traits for which clones show a response. Our results indicate that the major distinction is not between inducible and noninducible genotypes but rather that the genotypes differ in the combination of traits for which they show inducible responses.  相似文献

Females of the hybrid Daphnia galeata × hyalina were isolated from a natural population in Lake Maarsseveen and from these individuals clones were cultured in the laboratory. Some clones were assumed to be migrating and some non-migrating. Life history experiments were performed with these clones in the presence and absence of fish kairomones. Results show that clones differed in life history strategy in concert with their presumed migration strategies which suggests a link between life history and behaviour. However, no sharp border exists between migrating and non-migrating clones since a gradual response pattern was found for the traits investigated. In the absence of fish kairomones, the relation between length at maturity and number of eggs was found to differ between the clones of the non-migrating and the migrating group. The latter had a lower number of eggs at similar length values. In the presence of fish kairomones, the length–clutch size relation was the same. However, non-migrating clones were smaller at the age at maturity and had fewer eggs. Although clones of both groups were thus found to differ significantly, overlap existed. Fibroblast cultures prepared from mice homozygous for a Robertsonian translocation (centric fusion) between autosomes 8 and 17 [Rb(8.17)] were used as donors in microcell-mediated chromosome transfer experiments. By using hamster recipient cells deficient in adenine phosphoribosyltransferase (APRT-) and selecting for expression of murine APRT (a chromosome 8 marker), microcell hybrids were isolated which retained only the mouse Rb(8.17) translocation in addition to the hamster chromosome complement. The translocation was stable in cells maintained under APRT+ selective pressure, and mouse marker traits encoded by genes on both chromosomes 8 and 17 segregated concordantly. A second family of hybrid clones was constructed by fusing microcells derived from wild-type mouse fibroblasts with APRT- hamster cells. Four of six clones analyzed retained only mouse chromosome 8. These studies demonstrated that microcell hybrids containing specific Robertsonian translocations as the only donor-derived genetic material can be obtained. Furthermore, a number of Robertsonian translocations between chromosomes which carry selectable markers (chromosomes 3, 8, and 11) and other autosomes have been described. By using fibroblast cultures prepared from mice containing these translocations as donors in microcell fusions, 18 of the 20 mouse chromosomes could be selectively fixed in different hybrid clones. Thus, a collection of 20 hybrid clones, each containing a single, specific mouse chromosome, can be constructed by using the strategy described in this report. The potential utility of such a monochromosomal hybrid panel is discussed.  相似文献

Summary Life-history variation was examined among members of theDaphnia longispina group, which consists ofD. galeata, D. hyalina, D. cucullata, and hybrids. Factorial experiments were conducted at two temperatures (14° and 20° C) and two food concentrations (0.2 and 1.0 mg Cl−1). Differences in life-history features (size at maturity, age at first reproduction, size of first clutch, offspring size in first clutch) under the different environmental conditions were assessed among eightDaphnia clones, which represented members of this species complex. Significant differences between parentals and hybrids for most life-history features were observed under various treatments; generally, hybrid clones showed intermediate life-history traits when compared with parentals. When comparisons were made among clones within a given species (i.e.D. galeata, D. galeata xcucullata, D. cucullata), clonal differences were also noted for certain life-history traits. The data are discussed with reference to the formation and maintenance of hybrid species complexes in nature. This paper is dedicated to the memory of Hans Georg Wolf, who died suddenly in May 1990 at the age of 39 years.  相似文献

Somatic fusion of mesophyll protoplasts was used to produce hybrids between the frost-tolerant species Solanum commersonii (2n=2x=24) and dihaploid S. tuberosum (2n=2x=24). This is a sexually incompatible combination due to the difference in EBN (Endosperm Balance Number, Johnston et al. 1980). Species with different EBNs as a rule are sexually incompatible. Fifty-seven hybrids were analysed for variation in chromosome number, morphological traits, fertility and frost tolerance. About 70% of the hybrids were tetraploid, and 30% hexaploid. Chloroplast counts in stomatal guard cells revealed a low frequency of cytochimeras. The frequency of aneuploids was relatively higher at the hexaploid level (hypohexaploids) than at the tetraploid level (hypotetraploids). The somatic hybrids were much more vigorous than the parents, and showed an intermediate phenotype for several morphological traits and moderate to profuse flowering. Hexaploid hybrid clones were less vigorous and had a lower degree of flowering than the tetraploid hybrid clones. All of the hybrids were female fertile but male sterile except for one, which was fully fertile and self-compatible. Many seeds were produced on the latter clone by selfing and on the male-sterile clones by crossing. The somatic hybrid plants showed an introgression of genes for frost tolerance and an adaptability to cold from S. commersonii. Therefore, the use of these somatic hybrids in breeding for and in genetic esearch on frost tolerance and cold-hardening is suggested.  相似文献

This study describes the characteristics of hybrids between two closely related rat myoblast lines, which differ both in the ability to express their program of differentiation and in the expression of neoplastic properties. Myogenic, nonneoplastic L6J1-S cells were hybridized with nonmyogenic, neoplastic L6J1-N1 cells. Six hybrid clones were isolated and expanded for analysis of myogenic competence, and four of these clones were also evaluated for parameters of transformation, including tumorigenicity, ability to clone in agar, and surface fibronectin. In addition to our analysis of isolated clones, we also assessed myogenic differentiation in colonies representing 226 early hybrid clones. Results of all these analyses demonstrate that the myogenic phenotype is retained and that the tumorigenic/transformed phenotype is suppressed in the hybrids. Furthermore, our results indicate that when the programs for myogenesis and neoplastic transformation are confronted within a single cell, they are expressed as mutually exclusive alternatives. In contrast to these results on myogenic X nonmyogenic L6 hybrids, it has been reported that isolated clones of A9 X L6 exhibited extinction of myogenic competence and retention of transformed properties. We have evaluated myotube formation in over 300 early hybrid clones between A9 and either diploid or subtetraploid L8 rat myoblasts. Our results demonstrate that all of these hybrid clones exhibit extinction regardless of the ploidy of the myoblast parent, and they further indicate that extinction is not a consequence of chromosome loss. These results support the conclusion that in A9 X L6 hybrids, the nonmyogenic, transformed phenotype is dominant.  相似文献

Asymmetric hybridization was conducted between wheat and Brorrats inermis keyss which is a distanfiy related intergeneric plant (belonging to different tribe) of wheat and possesses some favorable traits, such as resistant to cold, drought and disease. Protoplasts isolated from young embryo-derived calli of common wheat ( Triticura aestivum L., tv. 99P, (AABBDD), 2n = 42) were fused with UV-treated protoplasts isolated from young embryo-derived calli of Bromus inermis by PEG method. Three clones (No. 1 ~ No. 3) were regenerated from the fusion products and differentiated into albino seedlings. The clones and the seedlings were all verified as hybrids by chromosome counting, isozyme and RAPD analysis. Their isozyme and RAPD pattern contained the characteristic bands of both parents as well as new band(s). The chromosome numbers of albino were in the range of 42~54 with small chromosomes of Bromus inerm/s and chromosome fragments. The above results confirmed that hybrid albinos were obtained.  相似文献
