A point of source of light against a dark background is perceived by the human retina as a point image enhanced by off-axis points (rays if the source is polychromatic) of light scattered from objects along the optic axis. As a consequence of movement of the vitreous humor, scattering centers imbedded there impart of this scattering pattern a corresponding movement. A method has been devised to give the vitreous humor reproducible initial conditions and to record the observed relaxation of the scattering pattern to its new rest position. The vitreous humor is found to be overdamped, and the heretofore unreported shear elastic modulus has been determined. A striking gravitational effect is revealed by comparing observations along a horizontal optic axis with ones along a vertical optic axis. For the former, gravitational torque is found to dominate the elastic torque. The reason nature has developed a slow-responding gravitational sensor in the vitreous humor is not clear.  相似文献

The objective of this research is to design and optimise a mini/micro-channel based surface accumulator of Escherichia coli to be detected by acoustic wave biosensors. A computational research has been carried out using the state of the art software, CFD-ACE with water as bacteria bearing fluid. E. coli bacteria have been modelled as random discrete particles tracked by solving the Lagrangian equations. The design challenges are to achieve low shear force (pico-N), high concentration at accumulation and high enough Reynolds number to avoid bacteria swimming. A range of low Reynolds number (Re) from 28.2 to 58.3 has been considered along with the effects of particle–boundary interactions, gravity, Saffman lift and Magnus lift. About four orders of magnitude higher concentration at accumulation than the inlet concentration and lower shear force in the order of less than pico-N have been achieved in the optimised design with particles accumulating at a specific location under random particle–boundary interactions.  相似文献

The characterization of a dielectrophoretic/gravitational field-flow-fractionation (DEP/G-FFF) system using model polystyrene (PS) microbeads is presented. Separations of PS beads of different surface functionalization (COOH and none) and different sizes (6, 10, and 15 microm in diameter) are demonstrated. To investigate the factors influencing separation performance, particle elution times were determined as a function of particle suspension conductivity, fluid flow rate, and applied field frequency and voltage. Experimental data were analyzed using a previously reported theoretical model and good agreement between theory and experiment was found. It was shown that separation of PS beads was based on the differences in their effective dielectric properties. Particles possessing different dielectric properties were positioned at different heights in a fluid-flow profile in a thin chamber by the balance of DEP and gravitational forces, transported at different velocities under the influence of the fluid flow, and thereby separated. To explore hydrodynamic (HD) lift effects, velocities of PS beads were determined as a function of fluid flow rate in the separation chamber when no DEP field was applied. In this case, particle equilibrium height positions were governed solely by the balance of HD lift and gravitational forces. It was concluded that under the experimental conditions reported here, the DEP force was the dominant factor in controlling particle equilibrium height and that HD lift force played little role in DEP/G-FFF operation. Finally, the influence of various experimental parameters on separation performance was discussed for the optimization of DEP/G-FFF.  相似文献

Coral morphological variability reflects either genetic differences or environmentally induced phenotypic plasticity. We present two coral species that sense gravity and accordingly alter their morphology, as characterized by their slenderness (height to diameter) ratio (SR). We experimentally altered the direction (and intensity) of the gravitational resultant force acting along or perpendicular to the main body axis of coral polyps. We also manipulated light direction, in order to uncouple gravity and light effects on coral development. In the experiments, vertically growing polyps had significantly higher SR than their horizontal siblings even when grown in a centrifuge (experiencing different resultant gravitational forces in proximal and distal positions). Lowest SR was in horizontal side-illuminated polyps, and highest in vertical top-illuminated polyps. Adult colonies in situ showed the same pattern. Gravitational intensity also affected polyp growth form. However, polyp volume, dry skeleton weight and density in the various centrifuge positions, and in aquaria experiments, did not differ significantly. This reflects the coral's ability to sense altered gravity direction and intensity, and to react by changing the development pattern of their body morphology, but not the amount of skeleton deposited.  相似文献

The question of how rotation on a horizontal axis clinostat removes plants from the influence of the gravitational stimulus is answered. It is shown that appropriate horizontal axis clinostat rotation restricts the fall of intracellular particles to a quasi-circular path such that the position of the particle remains virtually stationary within cells. The displacement of the path of fall, due to centrifugal force, is then considered, and a method of determining the optimal rotation rate is developed from physical principles. This method selects the rotation rate which minimizes the volume of cytoplasm through which particles pass under the joint influence of centrifugal and gravitational forces. With the recognition that single axis clinostats are ineffective with large plants or for long experiments, a new type of clinostat is proposed on which intracellular conditions can be rendered virtually identical to those of plants in satellite free fall regardless of plant size or duration of experiment.  相似文献

Motivated by the lateral migration phenomena of fresh and glutaraldehyde-fixed red blood cells in a field flow fractionation (FFF) separation system, we studied the transverse hydrodynamic lift on a slightly flexible cylinder in a two-dimensional channel flow. The finite element method was used to analyze the flow field with the cylinder at different transverse locations in the channel. The shape of the cylinder was determined by the pressure on the surface of the cylinder from the flow field solution and by the internal elastic stress. The cylinder deformation and the flow field were solved simultaneously. The transverse lift exerted on the cylinder was then calculated. The axial and angular speed of the cylinder were iterated such that the drag and torque on the cylinder were nulled to represent a freely translating and rotating state. The results showed that the transverse lift on a deformable cylinder increased greatly and the equilibrium position moved closer to the center of the channel compared to a rigid cylinder. Also, with the same elastic modulus but a higher flow rate, a larger deformation and higher equilibrium location were found. The maximum deformation of the cylinder occurred when the cylinder was closest to the wall where a larger shear rate existed. The numerical results and experimental studies are discussed.  相似文献

在黄土塬区王东沟流域采集不同地形部位和不同土地利用方式下土壤样品,测定其颗粒组成、容重和饱和导水率,借助变异系数、非参数检验等方法研究了不同地形部位和不同利用方式对土壤物理性质的影响.结果表明:土壤物理性质在水平方向和沿垂直剖面都存在变异,但在同一地形部位或同一土地利用类型内,容重和颗粒组成基本相似.水平方向,饱和导水率属强变异,粘粒和砂粒含量属中等变异,粉粒含量和容重属弱变异;沿垂直剖面,土壤的粒级分布具有连续性,但0~25 cm的土壤容重和饱和导水率与下层土壤差异显著.沿坡面从上向下,土壤趋向粘重,但饱和导水率增大;其中上坡位和中坡位的土壤性质相似,在相关研究中可以进行合并.草地与其他利用方式下的土壤性质差异显著,主要受地形部位的影响;耕地和果园的土壤性质相似且不同于其他利用类型,表明人为干扰对土壤物理性质有重要影响.  相似文献

Blood cells suspended in shear flows exhibit much larger dispersive motions than those predicted by the Stokes-Einstein formula for Brownian diffusion. The lateral migration and the erratic motions of the 8 microns red blood cells (RBC) is thought to be analogous to a diffusive process. It is shown that the often cited convective-diffusion theory may not be an adequate model for describing the transverse migration of suspended cells in blood flow. A comprehensive review of both the classical theory and of contemporary work in particle transport is presented, with particular emphasis on low Reynolds number tube flows. The mechanisms of Taylor dispersion, the effects of Brownian perturbations on translational and rotational motions of the suspended particles in shear fields, and the influence of integratable and chaotic advections, are individually examined. The classical experiment by Segre and Silberberg (1962) lead us to believe that particle hydrodynamics may play an important role in transverse migrations. In this light, we have further examined the hydrodynamic aspects of the so-called "tubular pinch" effect, the lateral migration of rigid spheres. We have also discussed the transverse motions of liquid drops, and the reversibility of the organization of suspensions in transport. The convective accelerations in the entrance region of a tube can produce relative velocities between fluid medium and various type of particulates if there is a difference in density. The deformable RBC, an "active-type" particle, can provide feedback to the flow from both mass and momentum considerations; the more rigid platelet, a "passive-type" particle, will experience a much smaller relative velocity as compared to the RBC. We may expect that particles of different densities are transported to different equilibrium annular positions before entering the fully developed flow region. The erratic, lateral movement of suspended particulates in steady laminar tube flow can be described by the usual Lagrangian coordinates.  相似文献

The local exchange model developed by McNair et al. (1997) provides a stochastic diffusion approximation to the random-like motion of fine particles suspended in turbulent water. Based on this model, McNair (2000) derived equations governing the probability distribution and moments of the hitting time, which is the time until a particle hits the bottom for the first time from a given initial elevation. In the present paper, we derive the corresponding equations for the probability distribution and moments of the hitting distance, which is the longitudinal distance a particle has traveled when it hits the bottom for the first time. We study the dependence of the distribution and moments on a particle's initial elevation and on two dimensionless parameters: an inverse Reynolds number M (a measure of the importance of viscous mixing compared to turbulent mixing of water) and the Rouse number ?(a measure of the importance of deterministic gravitational sinking compared to stochastic turbulent mixing in governing the vertical motion of a particle). We also compute predicted hitting-distance distributions for two published data sets. The results show that for fine particles suspended in moderately to highly turbulent water, the hitting-distance distribution is strongly skewed to the right, with mode相似文献

We present a theoretical analysis of fluid flow and particle interactions in the cone-plate viscometer under conditions typically applied in biological studies. The analysis demonstrates that at higher shear rates, besides linear primary flow in the rotational direction, prominent non-linear secondary flow causes additional fluid circulation in the radial direction. Two parameters, the cone angle and Reynolds number, characterize flow in the viscometer over all ranges of shear rate. Our results indicate that secondary flow causes positional variations in: (i) the velocity gradient, (ii) the direction and magnitude of the wall shear stress at the plate surface, (iii) inter-particle collision frequency, (iv) magnitude and periodicity of normal and shear forces applied during particle-particle interactions, and (v) inter-particle attachment times. Thus, secondary flow may significantly influence cellular aggregation, platelet activation and endothelial cell mechanotransduction measurements. Besides cone-plate viscometers, this analysis methodology can also be extended to other experimental systems with complex non-linear flows.  相似文献

Bamboo sharks (Chiloscyllium plagiosum) are primarily benthic and use their relatively flexible pectoral and pelvic fins to rest on and move about the substrate. We examined the morphology of the pectoral fins and investigated their locomotory function to determine if pectoral fin function during both benthic station-holding and pelagic swimming differs from fin function described previously in leopard sharks, Triakis semifasciata. We used three-dimensional kinematics and digital particle image velocimetry (DPIV) to quantify pectoral fin function in five white-spotted bamboo sharks, C. plagiosum, during four behaviors: holding station on the substrate, steady horizontal swimming, and rising and sinking during swimming. During benthic station-holding in current flow, bamboo sharks decrease body angle and adjust pectoral fin angle to shed a clockwise fluid vortex. This vortex generates negative lift more than eight times that produced during open water vertical maneuvering and also results in an upstream flow that pushes against the posterior surface of the pectoral fin to oppose drag. In contrast, there is no evidence of significant lift force in the wake of the pectoral fin during steady horizontal swimming. The pectoral fin is held concave downward and at a negative dihedral angle during steady horizontal swimming, promoting maneuverability rather than stability, although this negative dihedral angle is much less than that observed previously in sturgeon and leopard sharks. During sinking, the pectoral fins are held concave upward and shed a clockwise vortex with a negative lift force, while in rising the pectoral fin is held concave downward and sheds a counterclockwise vortex with a positive lift force. Bamboo sharks appear to sacrifice maneuverability for stability when locomoting in the water column and use their relatively flexible fins to generate strong negative lift forces when holding position on the substrate and to enhance stability when swimming in the water column.  相似文献

Chiral objects in shear flow experience a chirality‐specific lift force. Shear flows past helices in a low Reynolds number regime were studied using slender‐body theory. The chirality‐specific lift forces in the vorticity direction experienced by helices are dominated by a set of helix geometry parameters: helix radius, pitch length, number of turns, and helix phase angle. Its analytical formula is given. The chirality‐specific forces are the physical reasons for the chiral separation of helices in shear flow. The presence of bacterial biofilms is detrimental in a wide range of healthcare situations especially wound healing. Physical debridement of biofilms is a method widely used to remove them. This study evaluates the use of microfluidic jet impingement to debride biofilms. In this case, a biofilm is treated as a saturated porous medium also having linear elastic properties. A numerical modeling approach is used to calculate the von Mises stress distribution within a porous medium under fluid-structure interaction (FSI) loading to determine the initial rupture of the biofilm structure. The segregated model first simulates the flow field to obtain the FSI interface loading along the fluid-solid interface and body force loading within the porous medium. A stress-strain model is consequently used to calculate the von Mises stress distribution to obtain the biofilm deformation. Under a vertical jet, 60% of the deformation of the porous medium can be accounted for by treating the medium as if it was an impermeable solid. However, the maximum deformation in the porous medium corresponds to the point of maximum shear stress which is a different position in the porous medium than that of the maximum normal stress in an impermeable solid. The study shows that a jet nozzle of 500 μm internal diameter (ID) with flow of Reynolds number (Re) of 200 can remove the majority of biofilm species.  相似文献

This study demonstrates an inexpensive and straightforward technique that allows the measurement of physical properties such as position, velocity, acceleration and forces involved in the locomotory behavior of nematodes suspended in a column of water in response to single wavelengths of light. We demonstrate how to evaluate the locomotion of a microscopic organism using Single Wavelength Shadow Imaging (SWSI) using two different examples.The first example is a systematic and statistically viable study of the average descent of C. elegans in a column of water. For this study, we used living and dead wildtype C. elegans. When we compared the velocity and direction of nematode active movement with the passive descent of dead worms within the gravitational field, this study showed no difference in descent-times. The average descent was 1.5 mm/sec ± 0.1 mm/sec for both the live and dead worms using 633 nm coherent light.The second example is a case study of select individual C. elegans changing direction during the descent in a vertical water column. Acceleration and force are analyzed in this example. This case study demonstrates the scope of other physical properties that can be evaluated using SWSI while evaluating the behavior using single wavelengths in an environment that is not accessible with traditional microscopes. Using this analysis we estimated an individual nematode is capable of thrusting with a force in excess of 28 nN.Our findings indicate that living nematodes exert 28 nN when turning, or moving against the gravitational field. The findings further suggest that nematodes passively descend in a column of water, but can actively resist the force of gravity primarily by turning direction.  相似文献

In conventional automated storage and retrieval systems (AS/RS), storage and retrieval (S/R) machine travels simultaneously in the horizontal and vertical directions. However, S/R machine cannot support overly heavy loads, such as sea containers, so a new AS/RS, called split-platform AS/RS (SP-AS/RS), was introduced and studied in recent years. The SP-AS/RS employs vertical and horizontal platforms, which move independently, and are capable of handling heavy loads. The vertical platform which represents an elevator (or lift) with the elevator’s lifting table carries the load up and down among different tiers and the horizontal platform which represents the shuttle carrier or the shuttle vehicle can access all cells of the tier in which it belongs to. Single command cycle (SC) and dual command cycle (DC) are two main operating modes in AS/RSs. However, travel time models in all previous articles related to the SP-AS/RS are only for the SC. In this study, we first present a continuous travel time model for the DC in the SP-AS/RS under input and output (I/O) dwell point policy and validate its accuracy by computer simulations. Our model and simulation results both show that the square-in-time rack incurs the smallest expected travel time. After comparing with the existing model for the SC, we find that the DC is better than the SC in terms of the expected travel time.  相似文献

Media perfusion bioreactor systems have been developed to improve mass transport throughout three-dimensional (3-D) tissue-engineered constructs cultured in vitro. In addition to enhancing the exchange of nutrients and wastes, these systems simultaneously deliver flow-mediated shear stresses to cells seeded within the constructs. Local shear stresses are a function of media flow rate and dynamic viscosity, bioreactor configuration, and porous scaffold microarchitecture. We have used the Lattice-Boltzmann method to simulate the flow conditions within perfused cell-seeded cylindrical scaffolds. Microcomputed tomography imaging was used to define the scaffold microarchitecture for the simulations, which produce a 3-D fluid velocity field throughout the scaffold porosity. Shear stresses were estimated at various media flow rates by multiplying the symmetric part of the gradient of the velocity field by the dynamic viscosity of the cell culture media. The shear stress algorithm was validated by modeling flow between infinite parallel plates and comparing the calculated shear stress distribution to the analytical solution. Relating the simulation results to perfusion experiments, an average surface shear stress of 5x10(-5)Pa was found to correspond to increased cell proliferation, while higher shear stresses were associated with upregulation of bone marker genes. This modeling approach can be used to compare results obtained for different perfusion bioreactor systems or different scaffold microarchitectures and may allow specific shear stresses to be determined that optimize the amount, type, or distribution of in vitro tissue growth.  相似文献

Legged locomotion of the Eurasian nuthatch Sitta europaea on horizontal and vertical substrates was examined using field observations and experiments. Although previous studies have reported that nuthatches use 'walking' on vertical substrates, we found that they usually used 'hopping' on both vertical and horizontal substrates. When climbing up a vertical substrate, the feet were staggered in position and small phase differences were observed between the left and right leg movements in the gait. In upward climbing, the body was inclined towards the substrate during the first stance phase similar to other tree-trunk climbers, but the tail was not used for helping body rotation unlike most tree-trunk climbers. The staggered position of the feet may allow the legs to play different roles in pulling towards and pushing away from the substrate. In downward climbing, the feet were staggered in position, but the phase difference was quite small. In field observations, the Eurasian nuthatch preferred to move vertically, rather than in an inclined direction.  相似文献

An electrochemical surface shear stress measurement was applied to a model of very thin unilateral arterial stenosis (height of 1/8 of the model pipe diameter with very smooth surface). Three dimensional wall shear stress distribution was measured under steady flow field from a relatively low Reynolds number, Re = 270, to a high Reynolds number, Re = 1200. There was a characteristic high and low wall shear distribution pattern around the stenosis. There were also remarkable high shear stress areas on the opposite wall and both side walls of the stenosis. It was clearly shown that three dimensional structure of the flow field, hence, the wall shear stress distribution, is affected by a minimal change on the arterial wall.  相似文献
