Low temperature tolerance was investigated in the imbibed seedof 15 seed lots compnsmg seven cultivars of Lactuca sativa L.During rapid cooling (20 °C h–1) some seeds of allseed lots survived to –16 °C but none to –20°C. The majority of seed lots retained over 50 per centviability above –14 °C due to isolation of the embryofrom external ice by the endosperm, and subsequent embryo super-cooling.Certain seed lots, including all three seed lots of cv. TomThumb, showed high mortality at temperatures above –10°C. Correlation of mortality with the formation of externalice suggested that the endosperm is not an effective nucleationbarrier in these seed lots. Survival to –20 °C was increased at slower coolingrates (6 to 1 °C h–1) due to freeze desiccation ofthe embryo, but seed lots varied considerably in their toleranceof specific cooling rates. A model to explain this variationwas developed incorporatmg (1) seed lot super-cooling limittemperature, (2) the rate at which freeze dehydration of thesupercooled embryo took place, (3) the moisture content at whichnucleation (at –20 °C) was no longer certain and (4)the.initial equilibrium moisture content of the fully imbibedseed. Factors (1), (2) and (3) were found to be relatively constant,but low (or artificially reduced) seed moisture content wasclosely correlated with high survival at natural cooling rates.Seed size fractions of similar moisture content from a singlecultivar showed that more small seeds survive cooling at 3 °Ch–1 to –20 °C than larger seed. Seed with pierced endosperms or ineffective nucleation barrierswere capable of surviving to at least –10 °C if cooledslowly (1 °C h–1) but were killed by rapid (20 °Ch–1) cooling. In previous work, we demonstrated that there was an optimummoisture level for seed storage at a given temperature (Vertucciand Roos, 1990), and suggested, using thermodynamic considerations,that the optimum moisture content increased as the storage temperaturedecreased (Vertucci and Roos, 1993b). In this paper, we presentdata from a two year study of aging rates in pea (Pisum sativum)seeds supporting the hypothesis that the optimum moisture contentfor storage varies with temperature. Seed viability and vigourwere monitored during storage under dark or lighted conditionsat relative humidities between 1 and 90%, and temperatures between-5 and 65°C. The optimum moisture content varied from 0·015g H2O g-1 d.wt at 65°C to 0·101 g H2O g-1 d.wt at15°C under dark conditions and from 0·057 at 35°Cto 0·092 g H2O g-1 d.wt at -5°C under lighted conditions.Our results suggest that optimum moisture contents cannot beconsidered independently of temperature. The effect of alternating temperatures on the times taken byseeds of lentil (Lens culinaris Medikus) to germinate was investigatedusing a two-way temperature-gradient plate. Between 5 and 25°C,warmer temperatures increased the rate of germination. Variationamong the individual seeds in the times required for germinationat different constant temperatures within this range were describedwell by a log-normal distribution of thermal times, accumulatedabove a base temperature of 1·5°C. Even with amplitudesas great as 20°C, no effect of alternation per se on thethermal time required for germination was detected—whetherthe cool temperature was applied for 8 or 16 h d-1. Similarly,in alternating temperature regimes where the minimum temperatureof the diurnal cycle was between 0°C and the base temperature,the thermal times required for germination (where no thermaltime accrued during the periods when temperature was below Tb)were in close agreement with those values provided by the modeldetermined at warmer constant temperatures. However, where theminimum temperature applied was < 0°C the germinationof all but the earliest germinators was delayed beyond modelpredictions, and more so where the sub-zero minimum temperaturewas applied for 16 rather than 8 h d-1. The results, therefore,contradict the view that alternation in temperature per se reducesthe thermal time required for seed germination. Seed conditioning and germination in witchweed (Striga asiatica(L.) Kuntze) were temperature-dependent. With higher conditioningtemperatures, shorter conditioning time was required for germinationwith terminal dl-strigol (strigol) treatment at 30 °C. Maximumgermination (80–100%) was obtained by conditioning inwater at 20, 25, 30 and 35 °C for 14, 7, 5 and 3 d, respectively,and terminally treating with 10–6 M strigol at 30 °C.Seeds conditioned in 10–8 M strigol instead of water germinatedmuch less with the same terminal strigol treatment. Generally,conditioning was slower when seeds were conditioned in strigolrather than water. The reduction in germination rate by pretreatmentin strigol or pretreatment at low temperatures could be overcomeby increasing the terminal strigol concentration in the germinationtest. Conditioned seeds did not germinate at 10 and 15 °Cwith a terminal 10–6 M strigol treatment but yielded closeto maximum germination at 25, 30 and 35 °C with the sameterminal strigol treatment. To obtain maximum germination, boththe minimum conditioning temperature and the minimum germinationtemperature for conditioned seeds were 20 °C. The calanoid copepod, Eudiaplomus graciloides, was reared fromegg to adult on uni-algal diets (0.1. 0.5 and 2.5 mg dry wt1–1) using the green alga, Chlamydomonas reinhardtii,as food, or on a mixed diet consisting of Lake Esrom water filteredthrough a plankton net with pore size 45 µm and supplementedwith C. reinhardtii (2.5 mg dry wt 1–1). On the mixeddiet at 21.0°C growth in body dry wt (W, µg dry wt)was exponential, and the growth constants were 0.21 day–1in the early to mid juvenile stage (N1 - C4) and 0.11 day–1in the late juvenile to early adult stage (C4-A). At 14.5°Cthe corresponding growth rate constants were 0.10 and 0.08 day–1.Similar growth rates were found at uni-algal concentrationsof 0.5 and 2.5 mg dry wt I–1, and it was argued that thethreshold concentration for growth in Eudiaptomus was closeto 0.1 mg dry wt I–1. The clearance (C, ml h–1)of copepodites was measured on the uni-algal diets. The constantsof the regression (C = aWb) were: a = 0.125, b = 0.858 (2000C. reinhardtii ml–1), a = 0.068, b = 0.849 (10 000), a= 0.028, b = 0.875 (50 000). Ingestion rates were calculatedfrom the clearances and the average algal concentrations. Atthe three food levels the average daily rations were 30, 67and 125% of body dry wt. The respiration rate (R, nl O2 h–1)was measured in individuals reared on the mixed diet. The constantsof the regression (R = aWb) were: a = 4.82, b = 1.07 (nauplii,14.5°C), a = 4.17, b = 0.904 (copepodites and adults, 14.5°C),a = 6.87, b = 0.757 (copepodites and adults, 21.0°C). Nosignificant difference in the respiration rate of copepoditesreared on uni-algal diets and the mixed diet could be demonstrated.Energy budgets were calculated. The assimilation efficiencyand the gross growth efficiency of copepodites decreased markedlywith increasing food concentration, the net growth efficiencyvaried from an average of 0.44 at the lowest algal concentrationto 0.60 on the mixed diet. The response of the germination of seeds of Barbarea vema (Mill.)Aschers, Brassica chinensis L., Brassica juncea (L.) Czern.& Coss., Brassica oleracea L. var. gongylodes L., Camelinasaliva (L.) Crantz, Eruca saliva Mill., Lepidium sativum L.,Nasturtium officinale R. Br., and Rorippa palustris (L.) Besserto white fluorescent light of different photon flux densitiesapplied for different daily durations in a diurnal alternatingtemperature regime of 20 °C/30 °C (16 h/8 h) was quantifiedby linear relations between probit percentage germination andthe logarithm of photon dose, the product of photon flux densityand duration. The low energy reaction, in which increasing dosepromotes germination, was detected in all the seed populationsbut in Barbarea vema and Brassica Juncea the lowest photon doseapplied (10–5–2 and 10–5 7 mol m–2 d–1,respectively) was sufficient to saturate the response. Comparisons,where possible, between photoperiods demonstrated reciprocity,i.e. germination was proportional to photon dose irrespectiveof photoperiod, for the low energy reaction in Brassica oleracea(1 min d–1 to 1 h d–1), Camelina saliva (1 min d–1to 8 h d–1), Eruca saliva (1 min d–1 to 24 h d–1),Lepidium sativum (I min d–1 to 8 h d–1) and Rorippapalustris (1 min d–1 to 8 h d–1), but not in Brassicachinensis and Nasturtium officinale. The high irradiance reaction,in which increasing dose inhibits germination, was detectedin Barbarea vema, Brassica chinensis, Brassica juncea, Brassicaoleracea, and Camelina saliva. The minimum dose at which inhibitionwas detected was lO–0–3 mol m–2 d–1.These results are discussed in the context of devising optimallight regimes for laboratory tests intended to maximize germination The response of germination to photon dose was also quantifiedwith 3 x 10–4 M GA2, co-applied (Brassica chinensis, Camelinasaliva, and Lepidium sativum) and with 2 x 10–2 M potassiumnitrate co-applied (Brassica chinensis). In the latter casepotassium nitrate had no effect in the dark and inhibited germinationin the light, but GA2, promoted germination substantially inall three species. Variation amongst seeds in the minimum photondose required to stimulate germination was not affected by co-applicationof GA2, in Brassica chinensis and Camelina saliva, whereas seedsof Lepidium salivum showed a narrower distribution of sensitivitiesto the low energy reaction in the presence of GA2 Barbarea vema (Mill.) Photoinhibition of Seed Germination in Mediterranean Maritime Plants   总被引:4,自引:1,他引:3  
Photoinhibition of seed germination was shown for Allium staticiforme,Brassica tournefortii, Cakile maritima and Otanthus maritimus,all plant species inhabiting sandy coasts of the MediterraneanSea. Germination of A. staticiforme was found to be typicallyMediterranean in regard to its temperature range (>0–20°C), while B. tournefortii germinated optimally at intermediatetemperatures (15–25 °C). Light sensitivity was morepronounced in the latter species and 50 % inhibition of seedgermination was obtained with photon flux densities of approximately0.1 and 0.015 mol m–2 d–1 in A. staticiforme andB. tournefortii, respectively. However, the slopes of the regressionlines of germination plotted against the logarithm of whitelight flux density are similar in both species. From monthlyexperiments performed under fluctuating conditions of temperatureand light, simulating the elimate of Athens throughout the year,optimal germination response peaked in ‘winter’conditions for A. staticiforme, while a bimodal pattern wasobserved for B. tournefortii (maxima at the warm ends of therainy season). In contrast to the other plants, a fifth speciestested, Crithmum maritimum, showed an absolute light requirement,probably related to the rocky habitats of the species; the optimumgermination period coincided with the rainy season. Pot experimentswith seeds of the five species buried at various depths confirmedthat maximum emergence is favoured by shallow depths (0.5–1cm). It is concluded that seed germination in maritime plantsof the Mediterranean rim is mediated by a photoinhibition mechanismwhich can be considered an adaptation strategy against surfaceseedling establishment at the harsh, sandy or shingle, sea coasthabitats. 86Rb uptake was examined in two species of unicellular greenalgae, Chlamydomonas nivalis isolated from snow, and a cellwall-less mutant of the temperate freshwater Chlamydomonas reinhardii.In C. reinhardii cells grown at 20°C and cooled rapidlyto 0°C, 86Rb uptake was abolished. Cells cooled rapidlyto –5°C in the absence of ice accumulated 86Rb veryrapidly but the time course of this uptake suggested non-selectiveaccumulation through a damaged plasmalemma. Cells grown at 8°Cwere viable, able to divide and motile; they showed no signsof cold-shock and 86Rb uptake, albeit slow, was measurable at–5°C in the absence of extracellular ice. Cells ofC. nivalis grown at 20°C were damaged at sub-zero temperaturesalthough they did show an enhanced 86Rb uptake at 0°C. Cellsgrown at 5°C were able to accumulate 86Rb from media undercooledto -5°C in the absence of extracellular ice, and again showedenhanced uptake at 0°C. Maximum daily consumption was estimated for Mysis relicta fedad libitum rations of Daphnia pulex at 4,10,15 and 18°C.Gut-residence time was also evaluated for M.relicta fed clado-ceranprey at 4, 10 and 157deg;C. Mean daily consumption (g dry weightof Daphnia g–1 dry weight of Mysis day–1) rangedfrom 6% at 4%C to 12% at 10°. At 18°C, Mysis feedingrate declined to 9% day1. Mean, weight-adjusted consumptionrates exhibited a ‘dome-shaped’ response in relationto water temperature. Consumption rate was highest at 10°Cand lowest at 4°C. Estimated Q10 was more sensitive from4 to 10°C (Q10= 3) than from 10 to 15°C (Q10=1.2). Gut-residencetime for Mysis was inversely related to water temperature, implyingthat evacuation rate increases linearly with water temperature.Feeding and gut-evacuation rates become disassociated at watertemperatures >10°C. As water temperature increased above1°C, relative evacuation rate increased, whereas feedingrate declined. Kennedy, C. D. and Gonsalves, F. A. N. 1988. H+ efflux and trans-rootpotential measured while increasing the temperature of solutionsbathing excised roots of Zea mays.—J. exp. Bot. 39: 37–49. Novel temperature-ramp procedures have been used to measureH+ efflux and trans-root potential of excised roots of Zea mays(var. Fronica). Two types of experiment were performed: (1),increasing temperature from 17°C, and (2), pre-cooling theroots to 1°C before starting the temperature ramp. The ratesof increase of temperature for H+ efflux and trans-root potentialexperiments were 0·5 and 2·1°C min–1respectively The H+ scans revealed strong sharp maxima at 30°C and 32°C,for non-pre-cooled and pre-cooled roots respectively, the latterbeing significantly smaller. The trans-root potential scansfor the pre-cooled roots showed a corresponding maximum at 30°C,which was inhibited by KCN (1-0 mmol dm–3) with or withoutSHAM (10 mmol dm–3), or Hg2+ (1, 10, 100 µmol dm–3)in the bathing solutions. Some of the evidence suggests thatthese maxima are associated with electrogenic H+ pumping, mediatedby a plasma membrane-bound ATPase. However, no correspondingmaximum was observed in the trans-root potential scans for non-pre-cooledroots, the potential remaining at about — 75 m V from20°C to 35°C. As there is a 7-fold increase in H+ effluxbetween 20°C and 30°C, the relationship between netH+ efflux and electrogenic proton pumping in these roots isby no means clear. Some possibilities are considered here. Pre-cooled and non-pre-cooled roots show clear maxima in thetrans-root potential scans at about 46°C, at which temperaturethere is a slight net H+ influx. Immature Glycine max (L.) Merrill seeds, initially between 50and 450 mg f. wt, were grown and matured successfully in vitro.Excised seeds were floated in a liquid medium containing 5 percent sucrose, minerals and glutamine in flasks incubated at25 °C under 300 to 350 µE m–2 s–1 fluorescentlight. During 16 to 21 d in culture, seeds grew to a matured. wt of 100 to 600 mg per seed at an average rate of 5 to 25mg d. wt per seed d–1 depending on initial size. Growthrates were maximal during the first 8 to 10 d in vitro but declinedwith loss of green colour in the cotyledons. Seed coats rupturedwith rapid cotyledon expansion during the first 2 d in culture.Embryos were tolerant to desiccation and 80 to 90 per cent germinatedif removed from culture before complete loss of green colour.The growth of excised seeds in vitro exceeded the growth ofseeds in detached pods, but when windows were cut in pods topermit direct exposure of seeds to the medium, seed growth wascomparable. Measurements were made using GC/MS SIM1 of the effects of temperatureon cis,trans-ABA levels in developing ovules and embryos oftwo pea genotypes contrasted in seed size. These effects werethen related to differences in the growth of the pods, seeds,embryos, and testae. In both genotypes high temperatures hastenedthe onset and rate of logarithmic and then linear growth, greatlyshortening the duration of pod and seed development but withoutgreatly altering seed size. Cis,trans-ABA was most concentratedxin the ovules immediately after fertilization. It also accumulatedin the embryo, more rapidly in the larger-seeded line, duringseed maturation. The stage when accumulation in the embryo beganwas the same irrespective of temperature. Accumulation ceasedwhen the pods started to desiccate. The effects of differentconstant temperatures on the maximum levels of embryo cis,trans-ABAwere relatively small and confounded in one genotype by variationin ovule abortion and in the other by differences in the stagewhen cis,trans-ABA accumulation ceased. However, when plantswere transferred from 13 °C to 29 °C at two differentstages during seed maturation, further seed growth was greatlyinhibited coincident with a substantial increase in embryo cis-trans-ABA.The results suggested a role for cis,trans-ABA in the controlof cotyledon enlargement during the linear phase of seed growth.  相似文献

The levels of K$ and total electrolyte leakage into leachingsolution from imbibed seeds of Pisum sativum L. and Phaseolusvulgaris L. have been followed to determine the relationshipof K$ loss to the level of total electrolyte loss, and its relationto the physiological condition of the seed. The extent and rate of loss of both K$ and total electrolytesvaried with both seed condition and the conditions imposed bythe experiment. K$ accounted for between 25 and 50% of the totalelectrolyte leakage. The relationship was at all times significantlycorrelated, but changed with time and with experimental conditions. The effect of immersion, which increased electrolyte leakagedue to soaking injury, masked differences in seeds of varyingfield performance. Imbibition at either 5 °C or 40 °C before leaching wasshown to increase both K$ and total electrolyte loss in comparisonto seeds imbibed at 25 °C. These results suggest that the ‘conductivity test’,which is used as a measure of seed deterioration, may need tobe re-examined in terms of experimental procedure and interpretation.  相似文献

Respiratory activity and seasonal changes in carbohydrate contentof the storage organs of Rubus chamaemorus L. have been investigated.Leaf dark respiration rate increases in a non-linear mannerfrom 0·7 mg CO2 evolved dm–2 h–1 at 0 °Cto 4·6 rng CO2 evolved dm–2 hh–1 at 30 °C.Root and rhizome respiration rates increase from 1 µ1O2 uptake g–1 fresh weight h–1 at 0.7 ° C to10 µ10, uptake g–1 f. wt h–1 at 20 °C.Rhizome carbohydrate reserves decline from a September peakof 33 per cent alcohol insoluble d. wt to 16 per cent in May. Diurnal temperature fluctuations induced change in soya bean-pod[Glycine max (L.) Merr.] carbon exchange rate (CER, where positiveCER represents CO2 evolution). CER appeared to depend linearlyon temperature. Linear regressions of CER on temperature interceptedthe temperature axis at 5°C (i.e. zero CER at 5°C).Slopes of these regressions (i.e. temperature sensitivity) changedover the season. The CER-temperature sensitivity coefficient,K, (calculated from observed values of CER. pod temperatureand temperature intercept) rose from less than 0·02 mgCO2 h–1 pod–1 °C–1 during early pod-flll,peaked at over 0·04 mg CO2 h–1 pod–1 °C–1at mid pod-fill, and then declined during late pod-fill andmaturation. Centropages abdominalis is a neritic, omnivorous, temporallyabundant copepod present throughout the subarctic Pacific andits marginal seas. The two main objectives of this study wereto determine how temperature influences the development of C.abdominalis and whether growth rates of in situ populationsmay be limited by available food. At 6.9°C, median developmenttime from eggs laid to 50% adults was 42 days and the averageweight-specific somatic growth rate was 0.17 day–1. At4.6°C, median development time to adult was 59 days (projected)and growth rate averaged 0.08 day–1, suggesting that 4.6°Cmay be approaching the lower temperature for development andgrowth in this species. The functional relationship betweendevelopment time and temperature was established over the temperaturerange in which this species occurs. The in situ adult growthrates between 10 and 13°C averaged 0.14 day–1 andwere generally lower than the laboratory-reared juvenile growthrates, which may indicate that adult C. abdominalis are foodlimited in the field during summer and autumn.  相似文献

Post-embryonic development time, egg production rate and adultbody size of calanoid copepod Eurytemora affinis from Lake Ohnuma,Japan, were determined under six temperature-food conditions(103,5 x 103, 104 and 5 x 104 cells ml–1 at 15°C,5 x 104 cells ml–1 at 10and20°C) in the laboratory.The measured parameters varied with both temperature and foodconcentration. Development time from hatching to adult femalewas 9.2, 11.4 and 22.8 days at 20, 15 and 10°C respectively,at the highest food concentration. The males developed to adultat one to two days earlier than the females. An effect of foodshortage on development time occurred at the lowest food concentration.This development time was 24.8 days even at 15°C, beingtwice as long as that at the highest food concentration. Prosomelength of these food-limited females was approximately 75% ofwell-fed ones, which reduced by only 10% with increasing temperaturefrom 10 to 20°C. Clutch size (C, eggs clutch–1) ofwell-fed individuals depended on prosome length of the adultfemale (L, mm), and was expressed as an equation: C = 65.2 L383. Clutch size of individuals reared at less than 104 cellsml–1, however, mostly laid below the estimated curve,especially at the lowest food concentration being only 10% ofthat at the highest food concentration. These results suggestthat food availability is a more important factor affectingpopulation growth of E.affinis in Lake Ohnuma than variationof temperature.  相似文献

Plants of six contrasting genotypes of barley were raised fromvernalized (imbibed at 1 °C for 30 d) or non-vernalizedseeds and grown in 12 different controlled environments comprisingfactorial combinations of three photoperiods (10, 13 and 16h d–1), two day temperatures (18 and 28 °C) and twonight temperatures (5 and 13 °C). Except at longer daysfor Athenais or Arabi Abiad, the 28 °C day temperature wasgenerally supra-optimal and delayed awn emergence. In Arabi Abiad,however, significant responses to temperature were not detected.The low temperature pre-treatment of the seeds reduced the subsequenttime to awn emergence in Athenais and the autumn-sown genotypesAger, Arabi Abiad and Gerbel B, especially in longer days, buteither had no effect or tended to delay awn emergence in thespring-sown types Emir and Mona. Seasonal changes in the photosynthesis and primary productionof Microcystis aeruginosa Kütz. were investigated in LakeKasumigaura during 1981–1982. Microcystis always showeda light-saturated photosynthesis-light curve. Both Pmax andthe initial slope of the photosynthesis-light curve of Microcystisin early summer were very high, so it was concluded that Microcystisutilized both low and high light intensities efficiently. ThePmax of Microcystis was found to be a function of the watertemperature except in August and September. The linear regressionon the temperature-Pmax relationship discontinued at 11°C,where the Pmax value dropped; Microcystis did not photosynthesizebelow 4°C. The initial slope of the curve was also descendingbelow 11°C. It is suggested that Microcystis changes itsphysiological properties below 11°C. The highest value ofgross production calculated for M. aeruginosa was 5.4 gC m–2d–1 in July; the annual gross production was estimatedto be 300 gC m–2year–1 (i.e., 40% of the total primaryproduction in this lake).  相似文献
