Embryonic, larval, and juvenile development of a small cyprinid species, Tanichthys albonubes, is described from laboratory-reared specimens. The eggs, measuring 1.0–1.2 mm in diameter, were demersal, almost spherical in shape, transparent and unpigmented, with a pale yolk without oil globules. Hatching occurred 45–53 h after fertilization at 25.5°–26.9°C. The newly hatched larvae, measuring 2.2–2.6 mm in body length (BL), had melanophores on the head and body. In particular, a dark vertical streak occurring posterior to the otic capsule and melanophores above the eyes were distinctive. The yolk was completely absorbed at 3.4 mm BL. Notochord flexion was initiated at 5.0 mm BL and finished at 6.0 mm BL. Aggregate numbers of all fin rays were completed at 11 mm BL. Squamation was initiated at 8.4 mm BL and completed at 13 mm BL. Although the eggs of T. albonubes resembled those of other small danionin species, including Aphyocypris chinensis, Chela dadiburjori, Danio rerio, Devario malabaricus, Gobiocypris rarus, Hemigrammocypris rasborella, and Horadandia atukorali, they differed from those of A. chinensis, C. dadiburjori, G. rarus, and Horadandia atukorali in having a wider perivitelline space. The larvae and juveniles of T. albonubes were similar to those of the aforementioned seven species plus Danio albolineatus, Danio kerri, and Devario sp. (cf. D. aequipinnatus) in general morphology. However, the early life stage morphology of T. albonubes differed from them in having a dark vertical streak posterior to the otic capsule and melanophores above the eyes in the yolk sac larval stage, and a dark lateral streak with an unpigmented area just above the former on the body, a dark blotch on the caudal fin, and reddish dorsal, anal, and caudal fins during the postflexion larval and juvenile stages.  相似文献

 This study redescribes Bregmaceros mcclellandi Thompson, 1840, based on one specimen (74.4 mm SL) from the Bay of Bengal and 66 specimens (30.0–84.7 mm SL) from Mumbai (Bombay), India, because the type specimens have apparently been lost. The present specimens are characterized by having black dorsal, pectoral, and caudal fins and show the following morphology: caudal fin slightly forked; body chromatophores present mainly at the dorsal part; no scales on cheek; vertebrae 52–55 (13–15 + 38–41); dorsal rays 52–59; anal rays 54–60; pectoral rays 18–20; caudal rays 27–31 (principal rays 14); transverse scales 14–15. In the 66 Mumbai specimens, it was confirmed that the distinctive black fin pigmentation developed sequentially with growth, with complete pigmentation first on the anterior lobe of the dorsal fin, then simultaneously on the posterior lobe of the dorsal fin, the caudal fin, and the pectoral fin, and last, on the anal fin. This species is known only from the Arabian Sea, Bay of Bengal, and Gulf of Thailand. A review of 16 nominal Bregmaceros species indicates that, besides B. mcclellandi, the distinctive dark fin pigmentation is found in B. atripinnis (Tickell), B. atlanticus Goode and Bean, B. japonicus Tanaka, and B. lanceolatus Shen. B. atripinnis is considered a junior synonym of B. mcclellandi, and the others are clearly distinct from B. mcclellandi. Comments are made on some of the characters to more fully characterize the species and for reference in future revisionary and phylogenetic studies. The bz-m1 mutation in maize was one of the first to arise by direct transposition of the chromosome-breaking Ds element from its original or `standard' location in chromosome 9S to a known locus in the same chromosome arm. Thus, elucidation of its structure should shed light on the nature of the original Ds element described by McClintock in 1948. The Ds insertion in bz-m1 has been reported to be only 1.2 kb long – much shorter than other chromosome-breaking Ds elements that have been described. We have characterized here the Ds element in our bz-m1 stocks and have confirmed by genetic and molecular tests that, in the presence of Ac, it acts as a chromosome breaker. The Ds insertion at bz-m1 is 1260 bp long. Besides its normal 5′ and 3′ ends, it contains an internal 3′ end at the same junction as the chromosome-breaking double Ds element that has been found in several sh mutations. Thus, it appears to have arisen from the 4.1-kb double Ds by internal deletion of 2.9 kb. Because the element has lost one internal 5′ end, but retains the chromosome-breaking properties of double Ds, we have named it sesqui-Ds (sDs). The origin, structure and properties of sDs vis-à-vis double Ds support the hypothesis that double Ds corresponds to the chromosome-breaking Ds element at the `standard' location in 9S. The genus Culter, found only in East Asia (China, Vietnam, Korea and Russia), is a primary group of freshwater fishes in the subfamily Cultrinae (Cyprinidae). Of a total of 517 specimens studied, 235 were Culter alburnus from different localities and were used for morphometric analysis in this study. 17 landmarks were used in geometric morphometrics (GM) analysis of shapes. 13 traditional morphometrics (TM) and 21 truss-based morphometrics (TBM) characteristics were measured. The results of discriminant analysis and cluster analysis showed that four species-groups could be divided into separate groups. They were (1) C. alburnus + C. recurviceps, (2) C. dabryi dabryi + C. oxycephalus + C. dabryi shinkainensis, (3) C. oxycephaloides, (4) C. mongolicus mongolicus + C. mongolicus elongates + C. mongolicus qionghaiensis. The six populations of C. alburnus (Basilewsky, Nouv Mém Soc Imp Nat Moscou, 10:215–263, 1855), from Lake Poyanghu, River Songhuajiang, River Liaohe, River Jialingjiang, River Lijiang and Lake Taihu, should be classified as different geographical populations, but not subspecies. C. mongolicus elongates (He and Liu, Zool Res, 1(4):483–485, 1980) from Lake Chenghaihu and C. mongolicus qionghaiensis (Ding, Acta Zootaxon Sin, 15(2):246–250, 1990) from Lake Qionghaihu should be classified as subspecies. C. dabryi shinkainensis differed greatly from C. dabryi dabryi in body forms, and we suggest that C. dabryi shinkainensis should be changed to species, instead of subspecies, namely Culter shinkainensis (Yih and Chu, Acta Hydrobiol Sin, 2:170–199, 1959).  相似文献

REC114 is one of 10 genes known to be required for the initiation of meiotic recombination in Saccharomyces cerevisiae. It is transcribed only in meiosis, and our previous sequence analysis suggested the presence of an intron in the 3′ end of the gene. Hypotheses in the literature have suggested, because of its unusual location, either that the putative intron in REC114 is likely to be necessary for expression, or that there may actually be no intron present. This work demonstrates that REC114 does have an intron and is one of only three genes in yeast with introns located in the 3′ end. Furthermore, the 3′ splice site utilized in REC114 is a very rare AAG sequence; only three other genes in yeast use this nonconsensus sequence. The splicing of REC114 does not require MER1, a gene known to be involved in meiosis-specific RNA processing. In fact, an intronless copy of REC114 can complement a null rec114 mutation. Thus, it does not appear that the intron is essential for expression of REC114. Although the intron is not absolutely required for meiotic function, it is conserved in evolution; two other species of yeast contain an intron at the same location in their REC114 genes. The eggs, measuring 0.9–1.0 mm in diameter, were demersal, almost spherical in shape, transparent and unpigmented, with a pale yellow yolk without oil globules. Hatching occurred 49–56 h after fertilization at 26.2°–27.3°C. The newly hatched larvae, measuring 2.9–3.1 mm in body length (BL) with 17 + 19–20 = 36–37 myomeres, had melanophores on the head and body. A cement organ on the forehead for adhering to objects during the yolk sac and early preflexion larval stages was distinctive. The yolk was completely absorbed at 3.6–4.0 mm BL. Notochord flexion was initiated at 5.1–5.6 mm BL and finished at 7.1 mm BL. Aggregate numbers of all fin rays were completed at 14 mm BL. Squamation was initiated midlaterally on the anterior trunk at 14 mm BL and completed at 27 mm BL. Although the eggs of I. auropurpureus resembled those of the closely related species Chela dadiburjori, Danio rerio, and Devario malabaricus, they differed from those of Danio rerio and Devario malabaricus in having a narrower perivitelline space. The larvae and juveniles of I. auropurpureus were also similar to those of C. dadiburjori, Danio rerio, and Devario malabaricus in general morphology, but they differed from the latter three species in having a series of dark blotches laterally on the body in the juvenile stage. Moreover, I. auropurpureus differed from C. dadiburjori in having more myomeres and a near-single row of melanophores on the body along the dorsal midline from the yolk-sac to early postflexion larval stages, from Danio rerio in having a cement organ on the forehead during the yolk-sac and early preflexion larvae, and a single melanophore on the lower eye margin in the early yolk-sac larvae, and from Devario malabaricus in having a single melanophore on the lower eye margin in the early yolk-sac larvae. The presence of a cement organ on the forehead indicates a close relationship among the genera Inlecypris, Chela, and Devario.  相似文献

Two novel non-allelic mutants that were unable to fix nitrogen (Fix) were obtained after EMS (ethyl methyl sulfonate) mutagenesis of pea (Pisum sativum L.). Both mutants, SGEFix–1 and SGEFix–2, form two types of nodules: SGEFix–1 forms numerous white and some pink nodules, while mutant SGEFix–2 forms white nodules with a dark pit at the distal end and also some pinkish nodules. Both mutations are monogenic and recessive. In both lines the manifestation of the mutant phenotype is associated with the root genotype. White nodules of SGEFix–1 are characterised by hypertrophied infection threads and infection droplets, mass endocytosis of bacteria, abnormal morphological differentiation of bacteroids, and premature degradation of nodule symbiotic structures. The structure of the pink nodules of SGEFix–1 does not differ from that of the parental line, SGE. White nodules of SGEFix–2 are characterised by “locked” infection threads surrounded with abnormally thick plant cell walls. In these nodules there is no endocytosis of bacteria into host-cell cytoplasm. The pinkish nodules of SGEFix–2 are characterised by virtually undifferentiated bacteroids and premature degradation of nodule tissues. Thus, the novel pea symbiotic genes, sym40 and sym33, identified after complementation analysis in SGEFix–1 and SGEFix–2 lines, respectively, control early nodule developmental stages connected with infection thread formation and function. A new balitord fish, Hemimyzon sheni n. sp. was collected from the Tar-Ju River basin, Taitung County, southeast Taiwan. The new endemic species can be distinguished from other congeneric species by a combination of the following features: (1) dorsal fin rays 3 + 8, pectoral fin rays 13+11 (total 24); (2) lateral-line scales 78–80 and predorsal scales 31–32; (3) somewhat pointed head shape; (3) small pelvic fin which does not extend to rear vertical of dorsal fin; (4) position of anus close to anal fin base; and (5) predorsal region with many very small light brown marks and unmarked pectoral fin. A diagnostic key to three nominal, endemic species of Hemimyzon from Taiwan is also provided in this paper.  相似文献

A new species of Dipcadi (Hyacinthaceae) that is allied to D. concanense (Dalzell) Baker but differs in its small flowers (13 – 18 mm long vs 35 – 47 mm long) and funnel shaped perianth tube (5 – 6  ×  5 – 6 mm vs 18 – 27  ×  4 – 5.5 mm) is described as D. goaense. The new species is apparently endemic, because it is known only from the type locality in Goa state of India. The type locality is at the foothills of Western Ghats and the habitat is a soil covered, lateritic, open area.  相似文献

Monokaryons of Coprinus cinereus constitutively form small spores (oidia) in the aerial mycelium. Some strains also produce large, inflated single cells (chlamydospores) at the agar/air interface, and hyphal aggregates (hyphal knots) that can develop into sclerotia. Monokaryons show various reactions upon transformation with heterologous A mating type genes. Production of oidia in such A-activated transformants is repressed in the dark and induced by blue light. Five of six monokaryons tested following transformation with A genes showed induced production of hyphal knots and sclerotia in the dark, and at least three strains showed enhanced chlamydospore production in the dark. Continuous incubation under blue light inhibited formation of hyphal knots, sclerotia and chlamydospores in both competent monokaryons and in A-activated transformants. On artificial medium and on a 12 h light/12 h dark regime, A-activated transformants of one distinct monokaryon (218) formed fruit-body primordia that were arrested in development before karyogamy. Our studies show that A mating type genes control all major differentiation processes in Coprinus, but whether developmental processes can proceed depends on the genetic background of the strain. We show that circular plasmids containing perfect palindromic regions of 2 × 1.1 kb can be propagated in sbcC strains of Escherichia coli, a result that is at variance with the well known observation that λ DNA cannot tolerate palindromic regions larger than 2 × 265 bp. However, a significant fraction of these palindrome-containing plasmids can be recovered from E. coli strains either as linear molecules with hairpins at their ends or as head-to-head dimers, both in a RuvC-and RusA-independent manner. Our results suggests that large palindromes may form cruciforms in E. coli. However, palindrome-associated DNA rearrangements occur by a process that does not require any known cruciform resolvase activity. The genus Oryza to which cultivated rice belongs has 24 species (2n = 24 or 48), representing seven genomes (AA, BB, CC, EE, FF, BBCC and CCDD). The genomic constitution of five of these species is unknown. These five species have been grouped into two species complexes, the tetraploid ridleyi complex (O. ridleyi, O.␣longiglumis) and the diploid meyeriana complex (O.␣granulata, O. meyeriana, O. indandamanica). To evaluate the genomic structure of these species in terms of divergence at the molecular level vis-à-vis other known genomes of Oryza, we used the total genomic DNA hybridization approach. Total genomic DNA (after restriction digestion) of 79 accessions of 23 Oryza species, 6 related genera, 5 outgroup taxa (2 monocots, 3 dicots) and 6 F1s and BC1s derived from crosses of O.␣sativa with wild species were hybridized individually with 32P-labeled total genomic DNA from 12 Oryza species: O. ridleyi, O. longiglumis, O. granulata, O.␣meyeriana, O. brachyantha, O. punctata, O. officinalis, O. eichingeri, O. alta, O. latifolia, O. australiensis, and O.␣sativa. The labeled genomic DNAs representing the ridleyi and meyeriana complexes cross-hybridized best to all the accessions of their respective species, less to those representing other genomes of Oryza and related genera, and least to outgroup taxa. In general, the hybridization differential measured in terms of signal intensities was >50-fold under conditions that permit detection of 70–75% homologous sequences, both in the presence and in the absence of O. sativa DNA as competitor. In contrast, when total DNAs representing other Oryza genomes were used as probes, species of the O.␣ridleyi and O.␣meyeriana complexes did not show any significant cross-hybridization (<5%). These results demonstrate that the genome(s) of both of these complexes are highly diverged and distinct from all other known genomes of Oryza. We, therefore, propose new genomic designations for these two species complexes: GG for the diploid O. meyeriana complex and HHJJ for the allotetraploid O. ridleyi complex. The results also suggest that the uniqueness of these genomes is not restricted to species-specific highly repetitive DNA sequences, but also applies to dispersed sequences present in single or low to moderate copy numbers. Furthermore these appear to share relatively more genome-specific repeat sequences between themselves than with other genomes of rice. The study also demonstrates the potential of total genomic DNA hybridization as a simple but powerful tool, complementary to existing approaches, for ascertaining the genomic makeup of an organism. Upeneus australiae sp. nov. is described on the basis of 14 specimens collected from Australian waters. Although it belongs to the seven-spined group of Upeneus, now comprising five species, U. australiae is distinguished from its congeners by the combination of the following characters: 6–7 + 16–18 (modally 7 + 17) gill rakers, ectopterygoids without teeth, five and six black oblique bars on upper and lower caudal lobes, respectively, and peritoneum transparent with small black spots. The eggs, measuring 2.1–2.4 mm in diameter, were demersal, almost spherical in shape, transparent and unpigmented, with a pale yellow yolk without oil globules. Hatching occurred 39–45 h after fertilization at 26.8°–27.4°C. The newly hatched larvae, measuring 4.8–5.1 mm in body length (BL) with 22 + 17 = 39 myomeres, were characterized by melanophores already deposited on the eyes. The eggs of B. canarensis resembled those of the related danionin species Candidia barbatus, Opsariichthys uncirostris uncirostris, Zacco platypus, Z. sieboldii, and Z. temminckii. Although the larvae of B. canarensis were also similar to those of the foregoing species in general morphology, they differed in having a straight notochord tip and pigmentation on the eyes at hatching and the almost entire absence of melanophores on the ventral body surface from the yolk sac to postflexion larval stages. In all organisms glutathione-conjugate transporters (GS-X pumps) mediate the detoxification of a number of xenobiotics by removing them from the cytosol. In addition, GS-X pumps appear to play a role in the processing of endogenous compounds. We have isolated a novel genomic clone from Arabidopsis thaliana that encodes a putative GS-X pump, AtMRP4, which is part of a recently defined gene family. The derived amino acid sequence shares high levels of similarity (55–63%) with human, yeast, and other Arabidopsis homologues. The expression of the different members of the AtMRP gene family in Arabidopsis cell suspensions after treatment with chemicals that modify glutathione metabolism (compounds that induce different types of stress and that act as herbicide antidotes – safeners – in monocotyledonous species) revealed that the members of this gene family are differentially regulated. Lignosus hainanensis sp. nov., is described and illustrated on the basis of collections originating from tropical forest in Hainan Province, southern China. Both the morphology and phylogeny support this new species. It is characterized by its centrally stipitate basidiocarps arising from a distinct sclerotium, yellowish brown to cinnamon-brown pileal surface, cream to cream-buff pore surface, trimitic hyphal system with clamped generative hyphae, and oblong ellipsoid to cylindrical basidiospores. Lignosus hainanensis may be confused with L. rhinocerus, also occurring in southern China, but L. rhinocerus can be distinguished by its smaller pores (6–8 per mm) and smaller and distinctly broadly ellipsoid basidiospores (4–5 × 2.8–3 μm).  相似文献

Neolamprologus cancellatus, a new cichlid species, is described on the basis of eight specimens from the Zambian coast of Lake Tanganyika. The new species is characterized by a subtruncate caudal fin and a body that is slender (depth 22.3–25.2% in standard length) and easily distinguishable from its congeners by having 7–8 anal fin spines, 34–37 scales in longitudinal line, 33 total vertebrae, and a gridlike body pattern on a pale brownish body color. This species is only known to inhabit the shallow (2–7 m depth), rocky bottom of Wonzye Point in the southern part of the lake.  相似文献

Juveniles of three eleotrid Butis species (B. butis, B. humeralis, and B. koilomatodon) are described; their occurrence patterns were examined in Sikao Creek, a mangrove estuary located in southern Thailand. Juveniles of each species were distinguished by the following characters: B. butis with no bands on body and pale pelvic fins; B. humeralis with no bands on body and densely pigmented pelvic fins; and B. koilomatodon with 5–6 regular bands on body and a fleshy process (preorbital knob) on the snout. Although B. butis shared the aforementioned characters with B. amboinensis found in the same estuary, the former was distinguished from the latter by having a greater number of pectoral fin rays (18–21 vs. 17) and a deeper caudal peduncle. Distribution patterns of the three Butis species in Sikao Creek were distinguishable from each other. Smaller B. butis [mean ± SD = 22.7 ± 16.9 mm in standard length (SL), n = 32] occurred in the upper reach of the estuary, while larger specimens (52.4 ± 26.2 mm SL, n = 18 and 51.5 ± 29.7 mm SL, n = 10, respectively) were found in the middle and lower reaches and none in the marine area. In B. humeralis and B. koilomatodon, only juveniles were caught except for one adult specimen each. Juveniles (8.9–16.5 mm SL, n = 79) of B. humeralis occurred in the upper and middle reaches and the marine area. B. koilomatodon juveniles (9.9–13.7 mm SL, n = 30) were distributed in all areas from the lower to upper reaches.  相似文献

A new coprophilous species, Coprinellus radicellus, is presented and described on the basis of morphological characters and a species phylogeny inferred from ITS1–5.8S–ITS2 and beta-tubulin gene sequences. The species is characterized by lageniform pileo- and caulocystidia, 8- to 11-μm long ellipsoid basidiospores with a central germ-pore, globose cheilocystidia, an often rooting stipe, the lack of pleurocystidia and velar elements. These characters distinguish C. radicellus from all other described species in subsection Setulosi. It comes closest to C. brevisetulosus (Arnolds) Redhead, Vilgalys & Moncalvo and C. pellucidus (P. Karst.) Redhead, Vilgalys & Moncalvo both morphologically and phylogenetically, but is distinct from both. Another five representatives of morphologically recognizable groups of subsection Setulosi were included in the phylogenetic analyses and found were distinct from C. radicellus, both morphologically and phylogenetically. To date, this new species is known only from Scandinavia, which is surprising in view of the uniform geographical distributions of most setulose Coprinellus species. A key to coprophilous taxa of Coprinellus with pileo- and caulocystidia is presented. Three new combinations are proposed.  相似文献

The genes encoding the ApaLI (5′-G^TGCAC-3′), NspI (5′-RCATG^Y-3′), NspHI (5′-RCATG^Y-3′), SacI (5′-GAGCT^C-3′), SapI (5′-GCTCTTCN1^-3′, 5′-^N4GAAGAGC-3′) and ScaI (5′-AGT^ACT-3′) restriction-modification systems have been cloned in E.␣coli. Amino acid sequence comparison of M.ApaLI, M.NspI, M.NspHI, and M.SacI with known methylases indicated that they contain the ten conserved motifs characteristic of C5 cytosine methylases. NspI and NspHI restriction-modification systems are highly homologous in amino acid sequence. The C-termini of the NspI and NlaIII (5′-CATG-3′) restriction endonucleases share significant similarity. 5mC modification of the internal C in a SacI site renders it resistant to SacI digestion. External 5mC modification of a SacI site has no effect on SacI digestion. N4mC modification of the second base in the sequence 5′-GCTCTTC-3′ blocks SapI digestion. N4mC modification of the other cytosines in the SapI site does not affect SapI digestion. N4mC modification of ScaI site blocks ScaI digetion. A DNA invertase homolog was found adjacent to the ApaLI restriction-modification system. A DNA transposase subunit homolog was found upstream of the SapI restriction endonuclease gene.
