Auxinic herbicides are widely used in agriculture to selectively control broadleaf weeds. Prolonged use of auxinic herbicides has resulted in the evolution of resistance to these herbicides in some biotypes of Brassica kaber (wild mustard), a common weed in agricultural crops. In this study, auxinic herbicide resistance from B. kaber was transferred to Brassica juncea and Brassica rapa, two commercially important Brassica crops, by traditional breeding coupled with in vitro embryo rescue. A high frequency of embryo regeneration and hybrid plant establishment was achieved. Transfer of auxinic herbicide resistance from B. kaber to the hybrids was assessed by whole-plant screening of hybrids with dicamba, a widely used auxinic herbicide. Furthermore, the hybrids were tested for fertility (both pollen and pistil) and their ability to produce backcross progeny. The auxinic herbicide-resistant trait was introgressed into B. juncea by backcross breeding. DNA ploidy of the hybrids as well as of the backcross progeny was estimated by flow cytometry. Creation of auxinic herbicide-resistant Brassica crops by non-transgenic approaches should facilitate effective weed control, encourage less tillage, provide herbicide rotation options, minimize occurrence of herbicide resistance, and increase acceptance of these crops.  相似文献

Streptomycin enhances the synthesis of anthocyanins and inhibits the synthesis of chlorophylls and the development of chloroplasts in dark-grown seedlings of cabbage (Brassica oleracea), mustard (Sinapis alba), tomato (Lycopersicon esculentum), and turnip (Brassica rapa) exposed to prolonged periods of irradiation in various spectral regions. These results suggest that the contribution of photosynthesis to light-dependent high irradiance reaction anthocyanin synthesis in seedlings of cabbage, mustard, tomato, and turnip is minimal, if any at all. So far, phytochrome is the only photoreceptor whose action in the control of light-dependent anthocyanin synthesis in seedlings of cabbage, mustard, tomato, and turnip has been satisfactorily demonstrated.  相似文献

Control of green peach aphid (Myzus persicae), a globally important pest, using plant‐derived oils is a promising alternative to conventional insecticides. Although various plant‐derived oils are potentially useful for insect control, dose–response studies and efficacy comparisons among oils have not been widely reported. Our objective was to compare M. persicae control by plant‐derived oils, focusing on oils derived from Brassicaceae species that exhibit rotational and environmental quality benefits. We thus applied sprays of emulsified ethyl esters from the seed oils of yellow mustard (Sinapis alba), oriental mustard (Brassica juncea) and rapeseed (Brassica napus) to M. persicae in a laboratory bioassay. A dose–response relationship was modelled for the S. alba spray yielding LD50/LD95 values of 18.2 ± 0.87/128.1 ± 5.10 μg ester per cm2 (P < 0.0001). Ethyl esters of oils from all three species and soybean (Glycine max) ethyl ester were compared to determine the efficacy of Brassicaceae oils relative to the dominant plant‐oil spray currently available. All ethyl esters were equally efficacious despite measured differences in fatty acid profiles among the oils. Oils derived from mustards B. juncea and S. alba are potentially useful feedstocks for the production of insecticidal sprays, and testing on additional insects is warranted.  相似文献

Wild relatives of Brassica are a rich reservoir of genes that are invaluable for the improvement of cultivated species. Sinapis alba is a close relative of crop Brassicas that possesses several desirable traits such as tolerance to Alternaria black spot disease, heat stress, insect pests and nematodes. This study is aimed at developing and characterizing hybrids between Brassica juncea and S. alba with the ultimate goal of transferring genes for tolerance to Alternaria brassicae and heat stress, the traits that are lacking in cultivated Brassica. We generated three hybrids between B. juncea and S. alba through protoplast fusion. The hybridity was confirmed through cytology and molecular markers. While two of the hybrids were symmetric, the third one was asymmetric and had greater resemblance to B. juncea. Hybrids showed some characteristic features of the parents and were fully male and female fertile and also set seeds upon back crossing with the parent species. In vitro leaf assay and field inoculation studies revealed that the hybrids are highly resistant to A. brassicae. Besides, hybrids set seeds at temperature of >?38 °C when parents failed to produce seeds indicating that hybrids possess heat tolerance. These stable hybrids provide a reliable genetic resource for transfer of genes from S. alba into cultivated Brassica species.  相似文献

Changes in the content of free sterols (FS), steryl esters (SE), steryl glucosides (SG) and acylated steryl glucosides (ASG) in germinating seeds of white mustard (Sinapis alba) were studied together with parrallel changes in specific activities of some enzymes involved in sterol conjugate transformation. It has been found that a distinct increase in the net SE content and a similar, but less pronounced, increase in SG content at the beginning of germination can be correlated with a distinctly earlier appearance of SE and SG synthesizing enzymes, i.e. triacylglycerol: sterol acyltransferase and UDPG: sterol glucosyltransferase in comparison with hydrolytic activities, i.e. SE hydrolase and SG hydrolase. Our results suggest that metabolism of SG and ASG takes place mainly in the cotyledons while SE metabolism takes place mainly in the roots.  相似文献

Homogenates or crude 300-16 000 g membrane fractions from Sinapis alba roots catalysed esterification of [4-14C]cholesterol with utilization of endogenous acyl sources. With acetone powder preparations cholesterol esterification was distinctly stimulated by a neutral lipid fraction isolated from S. alba roots. Among neutral lipids triacylglycerols were the most active in this process. Experiments with various acyl-labelled acylglycerols as acyl donors and non-labelled sterols as acceptors confirmed that triacylglycerols are directly utilized as the source of fatty acids for sterol esterification. Di- and mono-acylglycerols were much less effective.  相似文献

During the germination of Sinapis alba seed, alanine and some other amino acids were oxidized via the tricarboxylic acid cycle (TCA), although the pentose phosphate pathway (PPP) was also operative and may account for ca 50% of glucose oxidation. Soil-incorporated plant materials have been associated with reduction in soilborne pathogens and diseases. Mechanisms of the biocidal actions are complex and not well understood. A glasshouse experiment, a non replicated field demonstration, and a field experiment were conducted to determine volatile compounds after incorporation of various plant species and their effect on pest control. Cabbage (Brassica oleracea), canola (Brassica rapa), kale (Brassica oleracea var. acephala), lettuce (Lactuca sativa var. valmaine), two mustard varieties -Caliente (Brassica juncea) and Green wave (Brassica juncea), two radish varieties - Oil seed (Raphanus sativus var. oleiformis) and Cherriette (Raphanus sativus), common rye (Secale cereale), and sorghum Sudan grass (Sorghum bicolor var. sudanese) were used in the glasshouse experiment. Caliente 199 mustard (Brassica hirta) was planted in the field demonstration and white mustard (Sinapis alba) was used in the field experiment. Fresh plant materials were chopped manually in the glasshouse experiment and mechanically in the field studies at the flowering stage before incorporation in natural field soils. In the glasshouse experiment, the equivalent biomass dry weight ranged from a minimum of 573 g?m?2 for L. sativa var. valmaine to a maximum of 1851 g?m?2 for S. bicolor var. sudanese. The average biomass was 792 g?m?2 for B. hirta and 804 g?m?2 for S. alba in the two field studies, respectively. The glasshouse experiment used a loamy sand field soil inoculated with a natural fine sandy loam soil that was known to contain high populations of Verticillium dahliae. Soils at both field sites belonged to the sandy loam series, and efforts were made to maintain sufficient soil moisture for plant growth. Although the interest was to determine all volatile compounds in general, only methyl sulfide and dimethyl disulfide were identified and subsequently quantified. Depending on plant species and time of sampling (one to seven days after soil incorporation), 2.7 to 346.4?μg g ?1 plant dry weight for methyl sulfide and 0 to 283.2?μg g ?1 plant dry weight for dimethyl disulfide were found in the glasshouse experiment. In general, high concentrations of dimethyl disulfide and methyl sulfide appeared to have reduced V. dahliae colony counts in bioassay potato stem saps in the glasshouse experiment. However, the correlation was weak (R 2 ?=?0.31), but a relatively stronger correlation was obtained (R 2 ?=?0.58) when excluding B. oleracea and B. rapa from the regression. Dimethyl disulfide and methyl sulfide were nearly non-detectable in the field demonstration, consequently no disease assessment was made. In the field experiment, a production of 5.2?μg g ?1 plant dry weight for methyl sulfide and 1.2?μg g ?1 dry weight for dimethyl disulfide was found two days after soil incorporation of S. alba. Compared to the untreated control, total Fusarium oxysporum counts in field soil were significantly lower 39 days after S. alba incorporation. However, no significant impact was found on total Pythium counts. Soil population of citrus nematode (Tylenchulus semipenetrans) in the S. alba plots was significantly reduced to similar levels found in the untreated control 112 days after S. alba incorporation. Compared to the untreated control, soil density of non plant parasitic freeliving nematodes was higher 39 days after S. alba incorporation. The study demonstrated quantifiable production of methyl sulfide and dimethyl disulfide gases from a variety of plant species in glasshouse and natural field environments. Some beneficial effects against V. dahliae, F. oxysporum, and T. semipenetrans were observed. Additional studies are needed to further elucidate these complex chemical and biological interactions.  相似文献

Virus particles of isometric shape with a diameter of 26 nm were found in the sieve tubes and accompanying phloem cells in ultrathin sections prepared from the nerves of white mustard (Sinapis alba L.) leaves and roots infected with the beet mild yellowing virus (BMYV). BMYV particles were much more frequent in the roots ofSinapis alba plants. Isometric particles were not found in the leaves and roots of healthy mustard plants.  相似文献

Several HindIII monomer units of a tandemly repeated nuclear DNA sequence ofBrassica campestris andBrassica juncea (Cruciferae) have been cloned and sequenced. The monomer units, of 177 bp length, are AT-rich and share 88% homology between themselves and more than 65% homology with similar repeats of otherCruciferae likeBrassica oleracea, Sinapis alba andRaphanus sativus. Thus unlike the rapid divergence of tandemly repeated satellite DNA in other organisms, this DNA element is highly conserved thus indicating its importance.  相似文献

In Arabidopsis thaliana, vernalization promotes flowering by repressing the floral inhibitor FLOWERING LOCUS C (AtFLC). This repression is mediated through epigenetic modifications at the AtFLC locus, leading to gene silencing. Whether the well-known quantitative effect of vernalization is due to the degree of AtFLC repression and/or its stability after return to normal temperature conditions has not been clarified. The penetration, development, and reproduction of a California population of the sugarbeet cyst nematode, Heterodera schachtii, was observed on cultivars of cabbage (Brassica oleracea), phacelia (Phacelia tanacetifolia), buckwheat (Fagopyrum esculentum), oilseed radish (Raphanus sativus), and white mustard (Sinapis alba). With the exception of the nonhost, phacelia, all were readily penetrated by second-stage juveniles of H. schachtii. After 38 days at 25 C, no cysts were observed on phacelia cv. Angelia or on the oilseed radish cv. Nemex and Pegletta. Cyst production was low (<2.5 cysts/plant) on the buckwheat cv. Tardo and Prego and most of the oilseed radish cultivars. Cyst production was intermediate (5-14 cysts/plant) on most of the white mustard cultivars, and high on cabbage (20-110 cysts/plant). In microplot studies conducted over 133 days (approx. 450 degree-days, base 8 C), the reproductive index for H. schachtii was greater than 1.0 for cultivars of phacelia, oilseed radish, and white mustard as welt as in fallow treatments, indicating the need for further research on the use of these crops under field conditions.  相似文献

Host plant relations of the monophagous weevil Ceutorhynchus constrictus Marsh. (Coleoptera: Curculionidae: Ceutorhynchinae) feeding on garlic mustard, Alliaria petiolata (Bieb.) Cavara & Grande (Cruciferae) were studied in the laboratory. Most other crucifers were rejected in choice tests using garlic mustard as a reference plant, but Brassica nigra, Sinapis alba and Thlaspi arvense were as acceptable as the host plant. Flowering plants of Descurainia sophia were acceptable while young plants of this species were not. The most important feeding stimulants in extracts of garlic mustard were uncharged, water soluble compounds. The most abundant glucosinolate in garlic mustard, sinigrin, was a feeding stimulant, too. However, the feeding stimulatory activity of sinigrin was only expressed in the presence of still unidentified uncharged compounds from garlic mustard leaves. Host plant relations in monophagous crucifer-feeding insects is discussed in relation to the distinctness of glucosinolate patterns found in their host plants.
Zusammenfassung Ceutorhynchus constrictus Marsh. (Coleoptera: Chrysomelidae: Ceutorhynchinae) ist ein monophager Rüsselkäfer, der an Knoblauchhederich frisst. Das Wirtswahl-Verhalten dieses Käfers ist im Labor untersucht worden. Die meisten Crucifiren waren im Wahlversuche nicht akzeptiert, wenn Knoblauchhederich als Vergleichspflanze vorhanden war. Von Brassica nigra, Sinapis alba, und Thlaspi arvense wurden im Vergleich gleiche Mengen verzehrt wie von der Wirtspflanze. Blühende Descurainia sophia Pflanzen wurden, im Gegensatz zu Jungpflanzen der gleichen Art, angenommen. Die wichtichsten Phagostimulanten in Extrakten von Knoblauchhederich-Blättern waren ungeladene, wasserlösliche Substanzen. Das häufigste Glukosinolat im Knoblauchhederich, Sinigrin, war auch ein Phagostimulant. Doch war die phagostimulierende Wirkung von Sinigrin nur in Kombinationen mit noch nicht identifizierten, ungeladenen Substanzen aus Knoblauchhederich-Blätter nachweisbar. Wirtspfanzen-Beziehungen von monophagen Insekten werden diskutiert im Zusammenhang mit der Eigenart des Glukosinolat-Inhaltes ihrer Wirtspflanzen.

A total of 54 samples of Brassicaceae crops showing symptoms of mosaic, mottling, vein banding and/or leaf deformation were collected in Kyiv region (northern central part of Ukraine) in 2014–2015. A half of collected samples was found to be infected with Turnip mosaic virus (TuMV), and TuMV was detected in samples from Brassica oleracea var. capitata (cabbage), Raphanus sativus, Brassica juncea, Raphanus sp., Sinapis alba, Camelina sativa and Bunias orientalis (weed). The full‐length sequence of the genomic RNA of a Ukrainian isolate (UKR9), which was isolated from cabbage, was determined. Recombination analysis of UKR9 isolate showed that this isolate was an interlineage recombinant of world‐Brassica and Asian‐Brassica/Raphanus phylogenetic groups. This study shows for the first time the occurrence of TuMV in Ukraine.  相似文献

The lack of availability of sources of resistance against Alternaria brassicicola within the family Brassicaceae has made oilseed mustard plants a target for one of the most damaging and widespread fungal diseases, Alternaria black spot. Of the other non‐host‐resistant/tolerant plants, Sinapis alba, white mustard, is considered to be the most important apart from Arabidopsis. To understand the defence response of S. alba upon incompatible interaction with this pathogen, a functional genomic approach using cDNA amplified fragment length polymorphism was performed. The highly reproducible bands, found to be either more amplified or uniquely present in infected S. alba plants compared with non‐infected plants, were further subjected to comparative reverse Northern analysis in the incompatible white mustard (S. alba) and compatible India mustard (Brassica juncea L.) plants. The suppression of 46% of the genes in the compatible background indicates the possibility of effective and specific recognition of Alternaria in S. alba. Analysis of the 118 genes up‐regulated specifically in infected S. alba compared with B. juncea showed that 98 genes have similarity to proteins such as receptor‐like protein kinase genes, genes involved with calcium‐mediated signalling and salicylic acid‐dependent genes as well as other genes of known function in Arabidopsis. The apparent expression profile data were further confirmed for selected genes by quantitative real‐time polymerase chain reaction analysis. Classification of these genes on the basis of their induction pattern in Arabidopsis indicates that the expression profile of several of these genes was distinct in S. alba compared with B. juncea.  相似文献

A full-length cDNA clone (MB3) and three partial clones (MA1, MB1 and MB2) which encode myrosinase (thioglucoside glucohydrolase, EC 3.2.3.1) were isolated from a Sinapis alba (white mustard) cDNA library. Nucleotide sequence analysis of these clones revealed that they are encoded by a gene family. Southern blot analysis with gene-specific probes showed that the gene family consists of a least two subfamilies (MA and MB) each with several members both in S. alba and in Brassica napus (oilseed rape). In Arabidopsis thaliana (wall cress) only three myrosinase genes seem to be present. Northern blot analysis indicated that all the myrosinase mRNA species have the same size, approximately 1.95 kb.  相似文献

Alternaria blight is one of the most devastating diseases of rapeseed-mustard caused by a necrotrophic fungus Alternaria brassicae. Lack of satisfactory resistance resource in Brassica is still a main obstruction for developing resistance against Alternaria. In this study, we have selected Brassica juncea, Sinapis alba and Camelina sativa to understand and unravel the mechanism of disease resistance against Alternaria. Histopathological studies showed early onset of necrosis in B. juncea (1 dpi) and delayed in S. alba (2 dpi) and C. sativa (3 dpi) respectively. Early and enhanced production of hydrogen peroxide (H2O2) was observed in C. sativa and S. alba (6 hpi) when compared to B. juncea (12 hpi). An increase in catalase activity was observed in both C. sativa (36 % at 6 hpi) and S. alba (15 % at 12 hpi), whereas it significantly decreased in B. juncea at 6 hpi (23 %), 12 hpi (30 %) and 24 hpi (8 %). Gene expression analysis showed induction of PR-3 and PR-12 genes only in C. sativa and S. alba when compared to B. juncea suggesting their vital role for Alternaria resistance. In contrast, SA marker genes were significantly expressed in B. juncea only which provides evidence of hormonal cross talk in B. juncea during Alternaria infection thereby increasing its susceptibility.  相似文献

DNA topoisomerase was isolated for the first time from nucleoids of white mustard (Sinapis alba L.) chloroplasts. The enzyme had a molecular weight of 70 kDa; was ATP-independent, required the presence of mono- (K+) and bivalent (Mg2+) cations, and was capable of relaxing both negatively and positively supercoiled DNA. These results suggest that the enzyme isolated belongs to the type IB DNA topoisomerases.  相似文献
