Reproductive success of male insects commonly hinges both on their ability to secure copulations with many mates and also on their ability to inseminate and inhibit subsequent sexual receptivity of their mates to rival males. We here present the first investigation of sperm storage in Queensland fruit flies (Tephritidae: Bactrocera tryoni; a.k.a. 'Q-flies') and address the question of whether remating inhibition in females is directly influenced by or correlated with number of sperm stored from their first mates. We used irradiation to disrupt spermatogenesis and thereby experimentally reduce the number of sperm stored by some male's mates while leaving other aspects of male sexual performance (mating probability, latency until copulating, copula duration) unaffected. Females that mated with irradiated rather than normal males were less likely to store any sperm at all (50% vs. 89%) and, if some sperm were stored, the number was greatly reduced (median 11 vs. 120). Despite the considerable differences in sperm storage, females mated by normal males and irradiated males were similarly likely to remate at the next opportunity, indicating (1) number of sperm stored does not directly drive female remating inhibition and (2) factors actually responsible for remating inhibition are similarly expressed in normal and irradiated males. While overall levels of remating were similar for mates of normal and irradiated males, factors responsible for female remating inhibition were positively associated with presence and number of sperm stored by mates of normal but not irradiated males. We suggest seminal fluids as the most likely factor responsible for remating inhibition in female Q-flies, as these are likely to be transported in proportion to number of sperm in normal males, be uninfluenced by irradiation, and be transported without systematic relation to sperm number in irradiated males. 相似文献

The vapours of certain pure chemicals, typical of ripe fruits, elicited characteristic components of ovipositional behaviour from gravid Dacus tryoni (Froggat) in an olfactometer: the flies walked and flew upwind to the source of the vapour and then probed with their ovipositors. A range of alcohols, acids, ketones and esters having 2–6 carbon atoms were effective (1 and 10% of iso-butyric acid, n-butyric acid, methyl butyrate, ethyl butyrate, 2-butanone, ethyl lactate and ethyl acetate; and 10% concentrations of ethanol and 2-propanone). The most effective were 4–6 carbon acids, esters and ketones. Behavioural threshold for n-butyric acid vapour at 26°C was obtained from a 5×10–3% dilution in paraffin oil; maximum fly response occurred at about 200 times this concentration. Low concentrations of the 15-carbon sesquiterpene, -farnesene, were also very effective, despite its lower volatility. These results suggest that at least three different types of alfactory sensory neurones are involved in the identification of fruit attractants by gravid D. tryoni. 相似文献

Temperature can affect the survival rate through each immature stage of life history and hence the rate of survival from egg to adult emergence. This phenomenon was investigated experimentally by using both regimes of constant temperature and ones in which temperatures fluctuated on a daily basis with rates of heating and cooling approximating to those found in the field. In fluctuating regimes, the highest temperatures tolerated for a few hours on a daily basis were higher than those tolerable on a constant basis. The highest tolerable daily maximum was influenced to some extent by the daily minimum. Regimes near the tolerable threshold that were experienced during one stage of the life history adversely affected the survival rate during the next stage, even if the latter experienced optimal conditions. The results in dicate that, in nature, survival from egg to adult is unlikely if daily maxima prevail in the region of 38–40°C for more than 7 days.

Résumé La température peut affecter le taux de survie, pour chaque stade d'immaturité, dans l'histoire d'une vie et d'où le taux de survie de l'oeuf à son apparition adulte. Ce phénomène a été étudié à titre expérimental en utilisant les deux régimes à une température constante et l'un d'eux à des températures variées sur une base journalière avec un taux approximatif de chauffage et refroidissement comme ceux que l'on trouve dans ce domaine.Dans un régime de fluctuation, les températures les plus élevées pendant quelques heures sur une base journalière étaient plus fortes que celles tolérées sur une base constante.La maximum journalier de la température la plus élevée supportable était influencé, jusqu'à un certain point, par le minimum journalier.Les régimes qui ont fait l'expérience proche du seuil du supportable au cours d'un stade, dan l'histoire d'une vie, on affecté défavorablement le taux de survie du stade suivant, même si ce dernier a expérimenté les conditions optimal.Les résultats indiquent que, dans la nature, la survie d'oeuf à adulte est improbable si le maxima journalier de 38–40°C prédomine dans la région pendant plus de 7 jours.相似文献

Copula duration and sperm storage patterns can directly or indirectly affect fitness of male and female insects. Although both sexes have an interest in the outcome, research has tended to focus on males. To investigate female influences, we compared copula duration and sperm storage of Queensland fruit fly females that were intact, or had been incapacitated through decapitation or abdomen isolation. We found that copulations were far longer when females had been incapacitated, indicating that constraints imposed on copula duration by intact females had been relaxed. Repeatability of copula duration for males was very low regardless of female treatment, and this is also consistent with strong female influence. Number of sperm in the spermathecae was not influenced by female treatment, suggesting that female abdominal ganglia control the transport of sperm to these long-term storage organs. However, more sperm were found in the ventral receptacles of incapacitated females compared to intact females. Overall, results implicate cephalic ganglia in regulation of copula duration and short-term sperm storage in the ventral receptacle and abdominal ganglia in regulation of long-term sperm storage in the spermathecae. 相似文献

Evidence presented indicates that exposure of wild Queensland fruit files, Dacus tryoni (Froggatt), to a given host fruit type (e.g. pear) for 3 days causes females to attempt oviposition to a greater degree in that fruit type than in other fruit types (e.g. tomato, grape). The effects of exposure to a particular fruit type proved reversible, suggesting that D. tryoni females were capable of learning. Females exposed to pear for 3 days appeared to retain the effect of such exposure on acceptance of tomato for up to 4 days but appeared to retain the effect on acceptance of grape for less than 2 days. The possible significance of prior experience of females with a particular fruit type on future ability to discriminate among varying-quality specimens of that type is discussed.
Résumé Les éléments apportés montrent que l'exposition de D. tryoni Froggat à un type de fruit déterminé (par exemple, la poire) pendant 3 jours, conduit la femelle à tenter de pondre à une plus haute fréquence dans ce type de fruit que dans d'autres (par example; tomate ou raisin). Les effects de l'exposition à un type particulier de fruit étant réversibles, ceci suggère que les femelles de D. tryoni sont capables d'apprentissage. Des femelles exposées à des poires pendant 3 jours ont conservé l'effet d'une telle exposition pendant 4 jours en présence de tomates, mais moins de 2 jours en présence de raisin. Recovery time after experience of a given minimum temperature below torpor threshold is related to the value of that minimum, the length of time spent at that minimum, and the temperature prevailing during the recovery period above torpor threshold. A model can predict recovery time for flies experiencing a given temperature fluctuation if the length of time spent at the minimum is expressed as a proportion of LE50 at that minimum.The model has applications in defining the optimal protocol for chilling insects for use in the Sterile Insect Release Method. The model was confirmed by experiments showing that it is likely that flies will recover from non-lethal frosts before ant predators become active.

Un modèle mathématique permet d'estimer le temps de récupération après avoir subi une chute de température déterminée, en fonction du temps passé au minimum thermique exprimé comme une fraction du LE50 (temps nécessaire pour tuer 50% des mouches) à ce minimum.Ce modèle s'est trouvé étayé par des observations montrant qu'il est probable que les mouches se remettent des gelées sublétales avant la reprise d'activité des fourmis prédatrices. Various colors have been used as visual cues to trap insect pests. For example, yellow traps for monitoring and control of the oriental fruit fly (Bactrocera dorsalis) have been in use for a very long time. However, the chromatic cue of using color traps has never been meticulously investigated. In this study, the spectral sensitivities of the photoreceptors in the compound eyes of B. dorsalis were measured intracellularly, and the theory of receptor quantum catch was applied to study the chromatic cue of fly attracting. Responses to five wavelength categories with peak wavelengths of 370, 380, 490, and 510 nm, and one with dual peaks at 350 and 490 nm were recorded. Based on spectral sensitivities, six colored papers were chosen to test the color preference of the fly, and an additional UV preference test was done to confirm the effect of the UV stimuli. It was concluded that UV and green stimuli (spectra: 300-380 nm and 500-570 nm) would enhance the attractiveness of a colored paper to the oriental fruit fly, and blue stimuli (380-500 nm) would diminish the attractiveness. 相似文献

Caloric restriction (CR) has been widely accepted as a mechanism explaining increased lifespan (LS) in organisms subjected to dietary restriction (DR), but recent studies investigating the role of nutrients have challenged the role of CR in extending longevity. Fuelling this debate is the difficulty in experimentally disentangling CR and nutrient effects due to compensatory feeding (CF) behaviour. We quantified CF by measuring the volume of solution imbibed and determined how calories and nutrients influenced LS and fecundity in unmated females of the Queensland fruit fly, Bactocera tryoni (Diptera: Tephritidae). We restricted flies to one of 28 diets varying in carbohydrate:protein (C:P) ratios and concentrations. On imbalanced diets, flies overcame dietary dilutions, consuming similar caloric intakes for most dilutions. The response surface for LS revealed that increasing C:P ratio while keeping calories constant extended LS, with the maximum LS along C:P ratio of 21:1. In general, LS was reduced as caloric intake decreased. Lifetime egg production was maximized at a C:P ratio of 3:1. When given a choice of separate sucrose and yeast solutions, each at one of five concentrations (yielding 25 choice treatments), flies regulated their nutrient intake to match C:P ratio of 3:1. Our results (i) demonstrate that CF can overcome dietary dilutions; (ii) reveal difficulties with methods presenting fixed amounts of liquid diet; (iii) illustrate the need to measure intake to account for CF in DR studies and (iv) highlight nutrients rather than CR as a dominant influence on LS. 相似文献

The fruit fly Drosophila melanogaster has emerged as a popular model to investigate fundamental principles of neural circuit operation. The sophisticated genetics and small brain permit a cellular resolution understanding of innate and learned behavioural processes. Relatively recent genetic and technical advances provide the means to specifically and reproducibly manipulate the function of many fly neurons with temporal resolution. The same cellular precision can also be exploited to express genetically encoded reporters of neural activity and cell-signalling pathways. Combining these approaches in living behaving animals has great potential to generate a holistic view of behavioural control that transcends the usual molecular, cellular and systems boundaries. In this review, we discuss these approaches with particular emphasis on the pioneering studies and those involving learning and memory. 相似文献

Movements of mature male Bactrocera tryoni (Froggatt) (Diptera: Tephritidae) were observed individually in a wind tunnel under conditions of ‘cue-lure with wind’, ‘cue-lure with no wind’, ‘wind only’ and ‘no wind or cue-lure’. Further observations were made using a dense foliage array in the wind tunnel and a structured plume of cue-lure. Patterns of walking or flying were essentially the same in all of the first four treatments except that in the ‘cue-lure with wind’ treatment, over half of the flies moved in a consistent track upwind for at least 400 mm at some time during the first 5 min of observation. With clean wind, only 10% of the flies did this. The result was that mean net upwind displacement after 5 min in the ‘cue-lure with wind’ treatment significantly exceeded that in the other three treatments, the results of which did not differ significantly from each other. The upwind tracks were accomplished by either walking or flying (with or without stops) or by a combination of both. When the wind tunnel was filled with a dense foliage array, the results with cue-lure laden wind were similar to those obtained with the equivalent treatment without foliage, except that upwind tracks were predominantly in short stages. When flies were exposed to a structured plume of cue-lure odour (without foliage present), they did not apparently alter their behaviour on leaving or entering the plume, but some did make consistent upwind tracks while they were in the plume. 相似文献

Culture medium supplemented with wheat germ oil (WGO) causes physiological reactions, such as increased fecundity and mobility, in some insects. Although the impact of WGO on insect physiology is important, the mechanisms of these actions are poorly understood. In this paper, we test the hypothesis that the addition of WGO to medium developed for larval oriental fruit flies modulates gene expression in the corresponding adults. We separately reared larvae of Bactrocera dorsalis on diets lacking or supplemented with WGO, and analyzed for expressed proteins in the resulting adult males and females by 2D-electrophoresis. Analysis of the gels revealed significant changes in expression levels of >70 proteins, 64 of which were identified by mass spectrometric analysis on MALDI-TOF/TOF. Apparent changes in expression levels for 6 of these proteins were confirmed by quantitative real-time PCR, showing that the changes in mRNA expression were reflected in changes in protein expression. These findings support the hypothesis that one mechanism of WGO actions in insect nutrition is the modulation of gene expression. 相似文献

The potential of entomopathogenic nematodes, Heterorhabditis bacteriophora, Heterorhabditis zealandica and Steinernema khoisanae, to infect pupariating larvae, pupae and adults of Ceratitis capitata and Ceratitis rosa was investigated in laboratory bioassays. Pupariating larvae and adult flies were susceptible to nematode infection, with no infection recorded for the pupae. Pupariating larvae of C. capitata were generally more susceptible to infection than those of C. rosa. Significantly more larvae of C. capitata were infected by H. bacteriophora. For C. rosa, highest infectivity of larvae was obtained with H. zealandica. In contrast, adults of both species were highly infected by S. khoisanae. 相似文献

The effect of access to dietary protein (P) and the topical application of a juvenile hormone analogue (methoprene (M)) on mating behaviour of male melon fly Bactrocera cucurbitae was assessed in the laboratory and in field cages. Age, dietary protein and methoprene application increased the mating success and influenced the mating behaviour. Treatment with methoprene (M+) to protein-deprived (P−) males had only a modest effect on the acceleration of sexual maturity, but application of methoprene (M+) to protein-fed (P+) males greatly accelerated sexual maturity. Protein diet (P+) increased mating success of males in comparison to protein-deprived (P−) males. Protein and methoprene have a synergistic effect on mating behaviour, since M + P+ treated males exhibit reduced mating latency and achieved higher mating in younger ages than methoprene and/or protein-deprived males. Copulation duration was correlated with nutritional status and M + P+ males copulated longer at the age of advanced sexual maturity than M − P+ males. Our results suggest that in this species with a lek mating system, females discriminate between the males based on their sexual signals, which were influenced by protein in the adult diet, methoprene application and age. The results are discussed in the light of mating competitiveness of precocious treated young males and their relevance to Sterile Insect Technique application against this pest species. 相似文献

Four species of tephritid fruit flies, Ceratitis capitata, Bactrocera dorsalis, B. cucurbitae, and B. latifrons were evaluated for toxic, developmental, and physiological responses to the chemosterilant lufenuron. No significant mortality of laboratory strains of the first three species was observed after their exposure up to 50 μg/mL of lufenuron in agar adult diet, whereas B. latifrons adults fed with 50 μg/mL of lufenuron in the diet caused significant mortality compared to the control. Fertility of C. capitata adults fed on 50 μg/mL lufenuron-fortified diet between 7 and 12 days of age was approximately 46% of the no lufenuron control. Fertility of B. dorsalis and B. latifrons adults fed on 50 μg/mL lufenuron-incorporated diet was about 45% and 62% of the control, respectively. Lufenuron did not significantly affect fertility of B. cucurbitae adults. Lufenuron did not affect fecundity of C. capitata and B. dorsalis. Fecundity of B. cucurbitae and B. latifrons was not evaluated due to difficulty to count the eggs laid deep in the agar diet. Larvae fed on a liquid larval diet with ≤ 0.1 μg/mL of lufenuron were also evaluated. Pupal recovery, adult emergence, adult fliers, mating, egg hatch, and egg production of C. capitata were significantly decreased, while for B. dorsalis, pupal recovery, larval duration and adult emergence were affected. No effect of lufenuron on B. cucurbitae larvae was observed. B. latifrons was not performed because shortage of eggs at the time of this research. rosa are polyphagous agricultural pests originating from the African continent. The taxonomy of this group (the so-called Ceratitis FAR complex) is unclear. To clarify the taxonomic relationships, male and female-produced volatiles presumably involved in pre-mating communication were studied using comprehensive two-dimensional gas chromatography with time-of-flight mass spectrometry (GC×GC-TOFMS) followed by multivariate analysis, and gas chromatography combined with electroantennographic detection (GC-EAD). GC×GC-TOFMS analyses revealed sex specific differences in produced volatiles. Male volatiles are complex mixtures that differ both qualitatively and quantitatively but share some common compounds. GC-EAD analyses of male volatiles revealed that the antennal sensitivities of females significantly differ in the studied species. No female volatiles elicited antennal responses in males. The results show clear species-specific differences in volatile production and provide complementary information for the distinct delimitation of the putative species by chemotaxonomic markers. 相似文献

Recent studies in fruit flies have imposed dietary restriction (DR) by diluting yeast and have reported increased lifespan as the yeast-to-sugar ratio decreased. In this study, the effects of DR on the lifespan of Bactrocera dorsalis were investigated using constant-feeding diets with different yeast:sugar ratios and an intermittent-feeding diet in which flies ate every sixth day. Antioxidant enzyme activities and the malondialdehyde concentration were also measured in virgin females under constant-feeding DR protocols to investigate their relationships with lifespan. The results showed that B. dorsalis lifespan was significantly extended by DR, and carbohydrate-enriched diet may be important for lifespan-extension. Female flies lived significantly longer than males at all dietary levels under both feeding regimes, indicating no interaction between diet and sex in determining lifespan. Antioxidant enzyme activities increased with the amount of yeast increased in the diets (0–4.76%) between starvation and DR treatments, indicating that the antioxidants may have influences in determining lifespan in B. dorsalis under starvation and DR treatments. However, antioxidants cannot keep up with increased oxidative damage induced by the high yeast diet (25%). These results revealed that the extension of lifespan by DR is evolutionarily conserved in B. dorsalis and that yeast:sugar ratios significantly modulate lifespan in this species. 相似文献

Mortality data for non-acclimated and acclimated 3rd instar larvae and mid-term eggs of Bactrocera tryoni (Froggatt) (Diptera: Tephritidae) were obtained after immersing in hot water at 46 °C. Acclimation consisted of holding the larvae and eggs at 35 °C for 20 and 11 h respectively just prior to heat-treatment. The median lethal time (LT50) for acclimated larvae was found to be 6.9 min compared to 2.5 min for non-acclimated larvae. LT99.999 for acclimated larvae was 20.9 min compared to 8.7 min for non-acclimated larvae. LT50 for acclimated eggs was 5.0 min compared to 2.4 min for non-acclimated eggs. LT99.999 for acclimated eggs was 26.0 min compared to 6.6 min for non-acclimated eggs. For 3rd instar larvae, most acclimation effect on mortality had occurred by 8 h. A notable residual response was present 20 h after acclimation had occurred, reducing mortality at 46 °C for 4.5 min by roughly 25%. Mortality data at 46 °C were also obtained for non-acclimated and acclimated late instar larvae of Epiphyas postvittana (Walker) (Lepidoptera: Tortricidae). With this species, LT50 for acclimated larvae was 2.5 min compared to 1.1 min for non-acclimated larvae. LT99.999 for acclimated larvae was 9.5 min compared to 4.6 min for non-acclimated larvae. 相似文献

The complete mitochondrial genome of the oriental fruit fly Bactrocera dorsalis s.s. has been sequenced, and is here described and compared with the homologous sequences of Bactrocera oleae and Ceratitis capitata. The genome is a circular molecule of 15,915 bp, and encodes the set of 37 genes generally found in animal mitochondrial genomes. The structure and organization of the molecule is typical and similar to the two closely related species B. oleae and C. capitata, although it presents an interesting case of putative intra-molecular recombination. The relevance of the growing comparative dataset of tephritid complete mitochondrial genomes is discussed in relation to the possibility to develop robust assays for species discrimination in quarantine and agricultural monitoring practices, as well as basic phylogeography/population genetic studies. 相似文献

Allopatric speciation often yields ecologically equivalent sister species, so that their
secondary admixis enforces competition. The shores of Lake Tanganyika harbor about 120
distinct populations of the cichlid genus Tropheus, but only some are sympatric.
When alone, Tropheus occupies a relatively broad depth zone, but in sympatry,
fish segregate by depth. To assess the effects of competition, we studied the partial
co-occurrence of Tropheus moorii ‘Kaiser'' and
‘Kirschfleck'' with Tropheus polli. A previous study demonstrated via
standardized breeding experiments that some observed differences between Tropheus
‘Kaiser'' living alone and in sympatry with T. polli have a genetic
basis despite large-scale phenotypic plasticity. Using geometric morphometrics and neutral
genetic markers, we now investigated whether sympatric populations differ consistently in
body shape from populations living alone and if the differences are adaptive. We found
significant differences in mean shape between non-sympatric and sympatric populations,
whereas all sympatric populations of both color morphs clustered together in shape space.
Sympatric populations had a relatively smaller head, smaller eyes and a more anterior
insertion of the pectoral fin than non-sympatric populations. Genetically, however,
non-sympatric and sympatric ‘Kaiser'' populations clustered together to the
exclusion of ‘Kirschfleck''. Genetic distances, but not morphological
distances, were correlated with geographic distances. Within- and between-population
covariance matrices for T. moorii populations deviated from proportionality. It
is thus likely that natural selection acts on both phenotypic plasticity and heritable
traits and that both factors contribute to the observed shape differences. The consistency
of the pattern in five populations suggests ecological character displacement. 相似文献

Centrosome reproduction by duplication is essential for the bipolarity of cell division, but the molecular basis of this process is still unknown. Mutations in Saccharomyces cerevisiae CDC31 gene prevent the duplication of the spindle pole body (SPB). The product of this gene belongs to the calmodulin super-family and is concentrated at the half bridge of the SPB. We present a functional analysis of HsCEN3, a human centrin gene closely related to the CDC31 gene. Transient overexpression of wild-type or mutant forms of HsCen3p in human cells demonstrates that centriole localization depends on a functional fourth EF-hand, but does not produce mitotic phenotype. However, injection of recombinant HsCen3p or of RNA encoding HsCen3p in one blastomere of two-cell stage Xenopus laevis embryos resulted in undercleavage and inhibition of centrosome duplication. Furthermore, HsCEN3 does not complement mutations or deletion of CDC31 in S. cerevisiae, but specifically blocks SPB duplication, indicating that the human protein acts as a dominant negative mutant of CDC31. Several lines of evidence indicate that HsCen3p acts by titrating Cdc31p-binding protein(s). Our results demonstrate that, in spite of the large differences in centrosome structure among widely divergent species, the centrosome pathway of reproduction is conserved. 相似文献
