Modern sugar cane varieties are derived from interspecific crosses involving as many as four species. Because a chromosome increase accompanies certain crosses and backcrosses, modern varieties have very high aneuploid chromosome numbers and complicated genetics. Despite this complexity, the chromosome behavior of some modern varieties approaches that of allopolyploids. In achieving homozygosity, therefore, such varieties should respond to inbreeding almost like diploids. The meiotic chromosome behavior of F1 hybrids and modern varieties indicates little or no genetic exchange between chromosomes ofSaccharum officinarum andS. spontaneum. Irradiation may break linkages between desirable and undesirableS. spontaneum genes not ordinarily broken by crossing-over between the chromosomes of the two species. The quick success of nobilizingS. spontaneum (recurrently back-crossing to “noble canes”) depends on a peculiar increase of the chromosomes ofS. officinarum. Experience with nobilizingS. spontaneum should not make breeders impatient when they turn to interspecific crosses unaccompanied by chromosome increases.  相似文献

We used a set of 16 SSR markers to check the identity of pure-species and hybrid clones in Vietnam’s Acacia auriculiformis, Acacia mangium, and acacia hybrid (A. mangium × A. auriculiformis) breeding programs. The statistics package HIest, applied to a large synthesized population, enabled accurate allocation of genotypes to the two pure species, F1 and F2 inter-specific hybrids and backcrosses, based on estimates of hybridity and heterozygosity. The hybridity status of putatively pure A. mangium and A. auriculiformis clones in adjacent clonal seed orchards was checked. Four out of 100 clones selected as A. mangium were found to be backcrosses (A. mangium × F1 inter-specific hybrid) while out of 96 clones selected as A. auriculiformis, two were F1 hybrids and two were backcrosses (A. auriculiformis × F1 hybrid). The markers were then applied to check the hybridity status of 160 putative acacia F1 hybrid genotypes that had been selected on morphological criteria from open-pollinated progenies collected from A. auriculiformis and A. mangium parents. Many selections based on morphology were found to be mistaken. Only thirteen of 63 clones originating from A. auriculiformis mothers were F1 hybrids, four were backcrosses, and the remaining 46 were pure A. auriculiformis. Fewer mistakes were evident for clones selected from A. mangium mothers, with 82 out of 89 clones confirmed as F1 hybrids, three as backcrosses, and four as pure A. mangium. The occurrence of F1 hybrids and backcrosses in pure-species seed orchards and their progeny shows that inter-species contamination is an issue requiring management in both pure-species and in hybrid breeding of these species in Vietnam. Examination of genetic distances among verified clones showed patterns of relatedness that were consistent with pedigree records. Implications for resource management as well as for breeding and clonal selection strategies are considered.  相似文献

Abstract

Ginseng is one of the most important herbs with valued pharmaceutical effects contributing mainly by the presence of bioactive ginsenosides in the roots. However, ginseng industry is impeded largely by its biological properties, because ginseng plants are slow-growing perennial herbs with lower yield. To increase the ginseng yield and amounts of ginsenosides, we developed an effective ginseng production system using the F1 progenies obtained from the interspecific reciprocal cross between two Panax species: P. ginseng and P. quinquefolius. Although hybrid plants show reduced male fertility, F1 hybrids with the maternal origin either from P. ginseng or P. quinquefolius displayed heterosis; they had larger roots and higher contents of ginsenosides as compared with non-hybrid parental lines. Remarkably, the F1 hybrids with the maternal origin of P. quinquefolius had much higher ginsenoside contents, especially ginsenoside Re and Rb1, than those with the maternal origin of P. ginseng. Additionally, non-targeted metabolomic profiling revealed a clear increase of a large number of primary and secondary metabolites including fatty acids, amino acids and ginsenosides in hybrid plants. To effectively identify the F1 hybrids for the large-scale cultivation, we successfully developed a molecular marker detection system for discriminating F1 reciprocal hybrids. In summary, this work provided a practical system for reciprocal hybrid ginseng production, which would facilitate the ginseng production in the future.

The role of hybridization through multi-specific synchronous spawning in the evolution of reef-building corals has been discussed since the 1990s, particularly for the genus Acropora. However, F1 hybrids have been reported as common in only one case in the Caribbean, with no evidence of mechanisms that would allow continuous reproduction of the hybrids. In this study, we report for the first time the fecundity of two F1 hybrid colonies produced experimentally from two Indo-Pacific species, A. intermedia and A. florida. These F1 hybrids spawned at the same time as the parental corals. Backcrossing and F1 hybrid crossing were successful in both directions. Furthermore, more than 90% self-fertilization was achieved in an F1 hybrid, although it was negligible in the parental corals. While it is possible that the F1 hybrid was a chimera, these results suggest that some products of interspecific hybridization may persist as the offspring of self-fertilizing F1 hybrids.  相似文献

Field studies and conceptual work on hybridization-mediated extinction risk in climate relicts are extremely rare. Nuphar pumila (Nymphaeaceae) is one of the most emblematic climate relicts in Europe with few isolated populations in the Alpine arc. The extent of introgression with related lowland and generalist species Nuphar lutea has never been studied using molecular methods. All biogeographical regions where N. pumila naturally occurs in the neighbourhood of the Alpine arc were sampled and studied using nuclear microsatellite markers. Furthermore, we used forward-in-time simulations and Approximate Bayesian Computation to check whether an introgression scenario fits with the observed admixture patterns and estimated the demographic parameters associated with this process. Our study confirms ongoing hybridization between N. pumila and N. lutea and validates it by the use of population models. More than 40 % of investigated N. pumila individuals were admixed and hybrids were found in over 60 % of studied populations. The introgression is bidirectional and is most likely a result of very recent gene flow. Our work provides strong evidence for rapid extinction risk and demographic swamping between specialized climatic relicts and closely related generalists. The remaining pure populations of N. pumila are rare in the Alpine arc and deserve high conservation priority.  相似文献

Abstract

Stem rust resistance genes Sr39 and Sr36 were transferred from Aegilops speltoides and Triticum timopheevii, respectively, to chromosome 2B of wheat. Genetic stocks RL6082 and RWG1 carrying Sr39 on a large and a shortened Ae. speltoides segments, respectively, and the Sr36-carrying Australian wheat cultivar Cook were used in this study. This investigation was planned to determine the genetic relationship between these genes. Stem rust tests on F3 populations derived from RL6082/Cook and RWG1/Cook crosses showed tight repulsion linkage between Sr39 and Sr36. The genomic in situ hybridization analysis of heterozygous F3 family from the RWG1/Cook population showed that the translocated segments do not overlap. Meiotic analysis on the F1 plant from RWG1/Cook showed two univalents at the metaphase and anaphase stages in a majority of the cells indicating absence of pairing. Since meiotic pairing has been reported to initiate at the telomere, pairing and recombination may be inhibited due to very little wheat chromatin in the distal end of the chromosome arm 2BS in RWG1. The Sr39-carrying large Ae. speltoides segment transmitted preferentially in the RL6082/Cook F3 population, whereas the Sr36-carrying T. timopheevii segment over-transmitted in the RWG1/Cook cross. Genotyping with the co-dominant Sr39- and Sr36-linked markers rwgs28 and stm773-2, respectively, matched the phenotypic classification of F3 families. The RWG1 allele amplified by rwgs28 was diagnostic for the shortened Ae. speltoides segment and alternate alleles were amplified in 29 Australian cultivars. Marker rwgs28 will be useful in marker-assisted pyramiding of Sr39 with other genes.

Interaction of photosystem I (PS I) complexes from cyanobacteria Synechocystis sp. PCC 6803 containing various quinones in the A1-site (phylloquinone PhQ in the wild-type strain (WT), and plastoquinone PQ or 2,3-dichloronaphthoquinone Cl 2 NQ in the menB deletion strain) and different numbers of Fe4S4 clusters (intact WT and FX-core complexes depleted of FA/FB centers) with external acceptors has been studied. The efficiency of interaction was estimated by measuring the light-induced absorption changes at 820 nm due to the reduction of the special pair of chlorophylls (P700 +) by an external acceptor(s). It was shown that externally added Cl 2 NQ is able to effectively accept electrons from the terminal iron-sulfur clusters of PS I. Moreover, the efficiency of Cl 2 NQ as external acceptor was higher than the efficiency of the commonly used artificial electron acceptor, methylviologen (MV) for both the intact WT PS I and for the FX-core complexes. The comparison of the efficiency of MV interaction with different types of PS I complexes revealed gradual decrease in the following order: intact WT?>?menB?>?FX-core. The effect of MV on the recombination kinetics in menB complexes of PS I with Cl 2 NQ in the A1-site differed significantly from all other PS I samples. The obtained effects are considered in terms of kinetic efficiency of electron acceptors in relation to thermodynamic and structural characteristics of PS I complexes.  相似文献

A sample of 12Mus (Leggada) triton Th. from the region of Bukavu (Democratic Republic of Congo) contains 5 ♂♂ and 7 ♀♀. 2N=32. All the autosomes are acrocentric. The sex-chromosomes of the ♂ are of the typeX—Y, theX beeing a big submetacentric (I.C.=0,4). Three ♀♀ possess two metacentricX, as expected. By four ♀♀, there is only one typicalX whose partner is acrocentric and as long as the long arm of a normalX. ThisX must have been arisen through the deletion of the short arm and is calledX ddc. The statistical analysis of the sample is compatible with this pattern:

Bruguiera hainesii (Rhizophoraceae) is one of the two Critically Endangered mangrove species listed in the IUCN Red List of Threatened Species. Although the species is vulnerable to extinction, its genetic diversity and the evolutionary relationships with other Bruguiera species are not well understood. Also, intermediate morphological characters imply that the species might be of hybrid origin. To clarify the genetic relationship between B. hainesii and other Bruguiera species, we conducted molecular analyses including all six Bruguiera species using DNA sequences of two nuclear genes (CesA and UNK) and three chloroplast regions (intergenic spacer regions of trnL-trnF, trnS-trnG and atpB-rbcL). For nuclear DNA markers, all nine B. hainesii samples from five populations were heterozygous at both loci, with one allele was shared with B. cylindrica, and the other with B. gymnorhiza. For chloroplast DNA markers, the two haplotypes found in B. hainesii were shared only by B. cylindrica. These results suggested that B. hainesii is a hybrid between B. cylindrica as the maternal parent and B. gymnorhiza as the paternal one. Furthermore, chloroplast DNA haplotypes found in B. hainesii suggest that hybridization has occurred independently in regions where the distribution ranges of the parental species meet. As the IUCN Red List of Threatened Species currently excludes hybrids (except for apomictic plant hybrids), the conservation status of B. hainesii should be reconsidered.  相似文献

In order to assist breeding and gene pool conservation in tropical Acacias, we aimed to develop a set of multipurpose SSR markers for use in both Acacia mangium and A. auriculiformis. A total of 51 SSR markers (developed in A. mangium and natural A. mangium x A. auriculiformis hybrid) were tested. A final set of 16 well-performing SSR markers were identified, six of which were species diagnostic. The markers were optimized for assay in four multiplex mixes and used to genotype range-wide samples of A. mangium, A. auriculiformis, and putative F1 hybrids. Simulation analysis was used to investigate the power of the markers for identifying the pure species and their F1, F2, and backcross hybrids. The six species diagnostic markers were particularly powerful for detecting F1 hybrids from pure species but could also discriminate the pure species from F2 and backcross progenies in most cases (97 %). STRUCTURE analysis using all 16 markers was likewise able to distinguish these cross types and pure species sets. Both sets of markers had difficulties in distinguishing F2 and backcross progenies. However, identifying F1 from pure species is the current primary concern in countries where these species are planted. The SSR marker set also has direct application in DNA profiling (probability of identity?=?4.1?×?10?13), breeding system analysis, and population genetics.  相似文献

Chromosome numbers for 98 plants ofF. pallens, 19 ofF. psammophila, F. belensis andF. vaginata, and 44 ofF. ovina (originating from Austria, the Czech Republic, Germany, Slovakia and Latvia) are given. In addition to theF. ovina andF. pallens groups, chromosome counts for the following taxa are also reported:F. alpestris (2n=14) reported for the first time in this work,F. amethystina subsp.amethystina (2n=28),F. brevipila (2n=42),F. cinerea (2n=28),F. rupicola subsp.rupicola (2n=42) andF. versicolor subsp.versicolor (2n=14).InF. pallens, two ploidy levels (2n=2x=14+0-1B, 2n=4x=28+0-1B) as well as two natural triploid plants (2n=21+0-1B), were found. In addition to the fourF. pallens types that have been distinguished in Austria, one new tetraploid type (F. pallens “scabrifolia”) from the Czech Republic and Germany is reported and its taxonomy is discussed. The distributions of the Oberösterreich-Niederösterreich and Pannonisches-HügellandF. pallens types outside of Austria are documented.Only the diploid chromosome number (2n=14) was found inF. psammophila andF. vaginata. Chromosome numbers forF. psammophila subsp.muellerstollii andF. belensis (both 2n=14) were determined here for the first time. Two ploidy levels, 2n=14+0-5B corresponding toF. ovina subsp.ovina and 2n=28 corresponding toF. ovina subsp.guestphalica andF. cf.duernsteinensis were confirmed inF. ovina. Differences in chromosome structure (simple and multiple secondary constrictions) betweenF. pallens as opposed toF. psammophila andF. vaginata are discussed. A complete survey of published chromosome counts for Central European species from theF. ovina andF. pallens groups is included.  相似文献

The reduction kinetics of the photo-oxidized primary electron donor P700 in photosystem I (PS I) complexes from cyanobacteria Synechocystis sp. PCC 6803 were analyzed within the kinetic model, which considers electron transfer (ET) reactions between P700, secondary quinone acceptor A1, iron-sulfur clusters and external electron donor and acceptors – methylviologen (MV), 2,3-dichloro-naphthoquinone (Cl2NQ) and oxygen. PS I complexes containing various quinones in the A1-binding site (phylloquinone PhQ, plastoquinone-9 PQ and Cl2NQ) as well as F X-core complexes, depleted of terminal iron–sulfur F A/F B clusters, were studied. The acceleration of charge recombination in F X-core complexes by PhQ/PQ substitution indicates that backward ET from the iron–sulfur clusters involves quinone in the A1-binding site. The kinetic parameters of ET reactions were obtained by global fitting of the P700 + reduction with the kinetic model. The free energy gap ΔG 0 between F X and F A/F B clusters was estimated as ?130 meV. The driving force of ET from A1 to F X was determined as ?50 and ?220 meV for PhQ in the A and B cofactor branches, respectively. For PQ in A1A-site, this reaction was found to be endergonic (ΔG 0?=?+75 meV). The interaction of PS I with external acceptors was quantitatively described in terms of Michaelis–Menten kinetics. The second-order rate constants of ET from F A/F B, F X and Cl2NQ in the A1-site of PS I to external acceptors were estimated. The side production of superoxide radical in the A1-site by oxygen reduction via the Mehler reaction might comprise ≥0.3% of the total electron flow in PS I.  相似文献

Background

Pinellia ternata is a Chinese traditional medicinal herb, used to cure diseases including insomnia, eclampsia and cervical carcinoma, for hundreds of years. Non-self-recognition in multicellular organisms can initiate the innate immunity to avoid the invasion of pathogens. A design for pathogen independent, heterosis based, fresh resistance can be generated in F1 hybrid was proposed.

Results

By library functional screening, we found that P. ternata genes, named as ptHR375 and ptHR941, were identified with the potential to trigger a hypersensitive response in Nicotiana benthamiana. Significant induction of ROS and Callose deposition in N. benthamiana leaves along with activation of pathogenesis-related genes viz.; PR-1a, PR-5, PDF1.2, NPR1, PAL, RBOHB and ERF1 and antioxidant enzymes was observed. After transformation into N. benthamiana, expression of pathogenesis related genes was significantly up-regulated to generate high level of resistance against Phytophthora capsici without affecting the normal seed germination and morphological characters of the transformed N. benthamiana. UPLC-QTOF-MS analysis of ptHR375 transformed N. benthamiana revealed the induction of Oxytetracycline, Cuelure, Allantoin, Diethylstilbestrol and 1,2-Benzisothiazol-3(2H)-one as bioactive compounds. Here we also proved that F1 hybrids, produced by crossing of the ptHR375 and ptHR941 transformed and non-transformed N. benthamiana, show significant high levels of PR-gene expressions and pathogen resistance.

Conclusions

Heterologous plant genes can activate disease resistance in another plant species and furthermore, by generating F1 hybrids, fresh pathogen independent plant immunity can be obtained. It is also concluded that ptHR375 and ptHR941 play their role in SA and JA/ET defense pathways to activate the resistance against invading pathogens.

Gram-positive, aerobic, non-motile, pale-yellow, and rodshaped bacterium, designated as Gsoil 188T, was isolated from the soil of a ginseng field in Pocheon, South Korea. A phylogenetic analysis based on 16S rRNA gene sequence comparison revealed that the strain formed a distinct lineage within the genus Brevibacterium and was most closely related to B. epidermidis NBRC 14811T (98.4%), B. sediminis FXJ8.269T (98.2%), B. avium NCFB 3055T (98.1%), and B. oceani BBH7T (98.1%), while it shared less than 98.1% identity with the other species of this genus. The DNA G + C content was 68.1 mol%. The predominant quinone was MK-8(H2). The major fatty acids were anteiso-C15:0 and anteiso-C17:0. The cell wall peptidoglycan of strain Gsoil 188T contained meso-diaminopimelic acid. The major polar lipids were phosphatidylglycerol, diphosphatidylglycerol, and an unidentified aminolipid. The physiological and biochemical characteristics, low DNA-DNA relatedness values, and taxonomic analysis allowed the differentiation of strain Gsoil 188T from the other recognized species of the genus Brevibacterium. Therefore, strain Gsoil 188T represents a novel species of the genus Brevibacterium, for which the name Brevibacterium anseongense sp. nov. is proposed, with the type strain Gsoil 188T (= KACC 19439T = LMG 30331T).  相似文献

We assembled a dataset tabulating the weights of Thai and Indonesian mangrove trees that we measured between 1982 and 2001. We selected four Thai study sites in Phang Nga, Ranong, Satun, and Trat Provinces and one site in eastern Indonesia on Halmahera Island in Maluku Province. The stands in Ranong Province and on Halmahera Island were in primary forests with data collected in the 1980s and the remaining stands were in secondary forests with data collected later. We collected 124 tree samples from ten species (Avicennia alba, Bruguiera cylindrica, B. gymnorrhiza, Ceriops tagal, Rhizophora apiculata, R. mucronata, Sonneratia alba, S. caseolaris, Xylocarpus granatum, and X. moluccensis) and measured the root weights of 32 individuals of nine species (A. alba, B. cylindrica, B. gymnorrhiza, C. tagal, R. apiculata, R. mucronata, S. alba, S. caseolaris, and X. granatum). All sampled trees were subjected to a standardized protocol to obtain aboveground weights. The trunks were divided into horizontal segments from which the leaves and branches were collected separately. Roots were collected by winching them out of the ground, by trench digging, or by complete excavation. Thus, we were able to compile the weights of the trunk, branches, leaves, and roots of each tree sampled. Aerial roots were included in root weight measurements, although they were collected above ground. We compiled separate lists of trunk diameters, trunk heights, heights of the lowest living branches, and the heights of aerial roots on the trunks of trees in different size categories. Our dataset includes a wide range of tree sizes (maximum trunk diameter 48.9 cm), geographical locations (1°10′N–12°24′N, 98°32′E–123°49′E) and organ weights (trunks, branches, leaves, and roots), and therefore should prove useful in future biomass studies of mangrove forests.  相似文献
