CO2 exchange were measured on pea seedlings (Pisum sativum L. var. Bördi) cultivated from seeds imbibed either in water (C-plants) or in gibberellic acid (GA3) at the concentration of 25 g/1 (GA-plants), and then grown under 17 W/m2 blue light (B-plants) or 11 W/m2 red light (R-plants).When measured under the same light conditions as during growth the net photosynthesis (APS) rate in B-plants was about twice higher than that in R-plants. Dark respiration (DR) rate was 70% higher in B- than in R-plants. Red light retarded the development of photosynthetic activity, but GA3 suppressed this effect. The hormone enhanced net photosynthesis and dark respiration to the same extent.When measured under saturating white light net photosynthesis rate of C-plants was also two times higher in B-plants than in R-plants. Growth conditions had only a slight effect on the APS of GA-plants under white light. APS rates of GA-plants grown under red light were higher under white light than those of C-plants, but lower than those of plants grown under blue light.We assume that blue light induced formation of plants that were adapted to higher light intensity: red light had an opposite effect, whereas gibberellic acid induced formation of plants that were adapted to medium light intensity.  相似文献

Photosynthetic activity, the content of various photosynthetic pigments, and the chloroplast ultrastructure were examined in the leaves of cucumber (Cucumis sativus L.) and pea (Pisum sativum L.) plants of different ages grown under red light (600–700 nm, 100 W/m2). In pea leaves tolerant to red-light irradiation, chloroplast ultrastructure did not essentially change. In the first true leaves of cucumber plants susceptible to red-light irradiation, we observed a considerable increase in the number and size of plastoglobules, the appearance of chloroplasts lacking grana or containing only infrequent grana, and stromal thylakoids. In the upper leaves of 22-day-old cucumber plants, the chloroplast structure was essentially similar to that of the control chloroplasts in white light, and we therefore suppose that these plants have acclimated to red light.  相似文献

The activity of pea chloroplast elongation factor G (EF-G), a nuclear-coded protein required for the elongation cycle of chloroplast protein synthesis, is regulated in response to light. In pea seedlings germinated and grown under continuous white or red light, EF-G specific activity reaches a maximum between days 10 to 15, and then decreases. EF-G activity is almost undetectable in extracts from dark-grown seedlings. When 13-day dark-grown pea seedlings are transferred to light, EF-G specific activity reaches a higher value after 2 to 3 days than observed in seedlings grown under continuous light. The small and large subunits of ribulose bisphosphate carboxylase continue to accumulate after EF-G specific activity has reached maximum levels. Cytoplasmically synthesized components of the chloroplast protein synthetic apparatus, such as EF-G, may help coordinate cytoplasmic and nuclear events with chloroplast gene expression during light-induced chloroplast differentiation.  相似文献

Detached leaves of white clover (Trifolium repens L.) were keptfor 1 h under various conditions of temperature, oxygen concentrationand light intensity. Rates of photosynthesis were measured whereappropriate and then ribulosebisphosphate carboxylase oxygenase(RuBPCO) was extracted rapidly and its initial activity measuredimmediately. The extracted activity increased with increased intensity ofillumination of the leaves. Where leaves were pretreated atlow light intensity, the lower the temperature of the leavesthe higher the extracted activity of RuBPCO. At high light intensitytemperature did not affect the activity of subsequently extractedRuBPCO but the light intensity which was necessary for maximumactivity increased with temperature. Activity of RuBPCO fromleaves pretreated in the dark was least when CO2 was low andtemperature high. Leaves, pretreated at low temperatures andhigh light intensity in 20% O2, yielded higher activity in extractsthan leaves pretreated under similar conditions but in 2% O2.A relatively weak temperature response of photosynthesis atlow irradiances was associated with a decrease in extractableRuBPCO activity with increasing temperature. A strong temperaturedependence of the oxygen inhibition of photosynthesis was associatedwith lower extractable RuBPCO activity in leaves pretreatedat low oxygen concentration at low temperatures. With leavesfrom plants grown at low temperatures prior to treatment ofleaves, oxygen inhibition of photosynthesis was less temperaturedependent and activity of RuBPCO in extracts was not decreasedby low O2 at low temperatures. Differences in the activationof RuBPCO appear to influence photosynthesis and account foran absence of oxygen inhibition of photosynthesis at low temperaturesin plants grown in warm conditions. The effects of gibberellic acid on lignification in seedlings of a dwarf and a tall cultivar of pea (Pisum sativum) grown under red or white light or in the darkness, were studied. Gibberellic acid (10−6-10−4 m) promoted stem elongation in both light and dark and increased the percentage of lignin in the stems of the light-grown dwarf pea. The gibberellin had no effect on the lignin content of the tall pea although high concentrations (10−4 m) promoted growth of the tall plants. Time course studies indicated that the enhanced lignification in the gibberellin-treated dwarf plants occurred only after a lag period of several days. It was concluded that gibberellic acid-enhanced ligmification had no direct relation to gibberellic acid-promoted growth. The activity of phenylalanine ammonia-lyase (E.C. 4.3.1.5) was higher in gibberellin-treated dwarf plants grown under white or red light than in untreated dwarf plants. Gibberellic acid had no detectable effect on the activity of this enzyme when the plants were grown in darkness, just as it had no effect on lignification under dark conditions. The data suggest that in gibberellin-deficient peas the activity of phenylalanine ammonia-lyase is one of the limiting factors in lignification.  相似文献

Wheat (Triticum aestivum L. cv. HD 2285) was grown in control (C) and heated (H) open top chambers (OTCs) for entire period of growth and development till maturity. The mean maximum temperature of the entire period was 3 °C higher in H-compared to C-OTCs. Net photosynthetic rate (P N) measured at different temperature (20–40 °C) of C-and H-grown plants showed greater sensitivity to high temperature in H-plants. P N measured at respective growth temperature was lower in H-compared to C-plants. The CO2 and irradiance response curves of photosynthesis also showed lesser response in H-compared to C-plants. The initial slope of P N versus internal CO2 concentration (P N/C i) curve was lower in H-than C-plants indicating ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) limitation. In irradiance response curve, the plateau was lower in H-compared to C-plants which is interpreted as RuBPCO limitation. RuBPCO content in the leaves of C-and H-plants, however, was not significantly different. Ribulose-1,5-bisphosphate carboxylase (RuBPC) initial activity was lower in H-plants, whereas activity of fully activated enzyme was not affected, indicating a decrease in activation state of the enzyme. This was further substantiated by the observed decrease in RuBPCO activase activity in H-compared to C-plants. RuBPCO activase was thus sensitive even to moderate heat stress. The decrease in P N under moderate heat stress was mainly due to a decrease in activation state of RuBPCO catalysed by RuBPCO activase.  相似文献

Limitation of photosynthesis and light activation of ribulose,1,5-bisphosphate carboxylase (RuBPCO) were examined in the 5thleaf of seedlings of red clover (Trifolium pratense L. cv. Renova)for 5 d following an increase in photosynthetic photon fluxdensity (PPFD) from 200 to 550µmol quanta m–2 s–1.Net photosynthesis and its stimulation at 2.0 kPa O2 initialactivity of rapidly extracted RuBPCO, standard activity of RuBPCOafter incubation of the extracts in the presence of CO2, Mg2+,and inorganic phosphate and contents of soluble protein, starch,soluble sugars, and various photosynthetic metabolites weredetermined. Photosynthesis decreased and starch content increased.No decrease in photosynthesis was found if, when PPFD was increased,all leaves except the investigated 5th leaf were removed, suggestingthat the decrease in photosynthesis was due to accumulated carbohydrates.The stimulation of photosynthesis at 2.0 kPa O2 did not decreaseand the ratio of the total foliar steady-state contents of triosephosphate to 3-phosphoglycerate increased suggesting that thedecrease in photosynthesis was not due to limiting inorganicphosphate in chloroplasts. Intercellular CO2 partial pressureand RuBP content were not decreased. Nevertheless, the ratioof photosynthesis to initial RuBPCO activity decreased, suggestingthat the catalysis per active RuBPCO site was decreased. Theincrease in PPFD in the growth cabinet and the PPFD at whichleaves were preconditioned for 1 h, affected not only initialactivity but also the standard activity of RuBPCO. The resultssuggest that a varying proportion of RuBPCO was bound to membranesand was contained in the insoluble fraction of the extracts.A comparison of photosynthesis with extracted RuBPCO activitysuggested that membrane bound RuBPCO did not contribute to photosyntheticCO2 fixation and that the binding and release to and from membranesmodulated actual RuBPCO activity in vivo. Dark-grown pea seedlings (Pisum sativum L.) were irradiated for a short period each day with low intensity red light (662 nm), red light immediately followed by far red light (730 nm), or far red light alone. Other plants were transferred to a white light regime (14 hours light/10 hours dark). There was no change in the amount of RNA in the tissue on a fresh weight basis after the various treatments. However, compared with dark-grown seedlings, those plants irradiated with red light showed an increase in the net RNA content per stem apex. In addition there was a two- to three-fold increase in ribosomal RNA of the etioplasts relative to the total ribosomal RNA. These increases were comparable to those found in plants grown in the white light regime. The changes were much smaller if the dark-grown plants were irradiated either with red light followed by far red light, or with far red light alone. Thus continuous light is not essential for the production of ribosomal RNA in plastids, and the levels of ribosomal RNA found in chloroplasts can also be attained in etioplasts of pea leaves in the dark provided the leaf phytochrome is maintained in its active form.  相似文献

通过对烟草植株覆盖白、红、黄、蓝、紫色滤膜获得不同光质,研究了烟草叶片在7~70d的生长发育期内,不同光质处理对烟叶组织结构特征、核酮糖1,5-二磷酸羧化酶/加氧酶(Rubisco)羧化酶活性、Rubisco基因(rbc)表达及其活化酶(Rca)基因(rca)表达的影响。结果表明,与黄膜处理下生长的烟叶相比,红、蓝、紫膜处理下生长的烟叶有较高的叶片厚度、栅栏组织厚度、海绵组织厚度、栅栏细胞密度和较小的组织空隙率。此外,红、蓝、紫膜处理的叶片有较高的Rubis-co羧化酶活性和净光合速率及较强的rbc和rca基因表达。实验结果表明不同光质对烟草叶片的组织结构特征有显著影响,光质可能通过影响Rubisco羧化酶活性进而影响叶片光合效率,而光质、叶片组织结构和光合效率之间存在某种程度的相互联系。  相似文献

It has been suggested that field experiments which increase UV-B irradiation by a fixed amount irrespective of ambient light conditions (‘square-wave’), may overestimate the response of photosynthesis to UV-B irradiation. In this study, pea (Pisum sativum L.) plants were grown in the field and subjected to a modulated 30% increase in ambient UK summer UV-B radiation (weighted with an erythemal action spectrum) and a mild drought treatment. UV-A and ambient UV control treatments were also studied. There were no significant effects of the UV-B treatment on the in situ CO2 assimilation rate throughout the day or on the light-saturated steady-state photosynthesis. This was confirmed by an absence of UV-B effects on the major components contributing to CO2 assimilation; photosystem II electron transport, ribulose 1,5-bisphosphate regeneration, ribulose 1,5-bisphosphate carboxylase/oxygenase carboxylation, and stomatal conductance. In addition to the absence of an effect on photosynthetic activities, UV-B had no significant impact on plant biomass, leaf area or partitioning. UV-B exposure increased leaf flavonoid content. The UV-A treatment had no observable effect on photosynthesis or productivity. Mild drought resulted in reduced biomass, a change in partitioning away from shoots to roots whilst maintaining leaf area, but had no observable effect on photosynthetic competence. No UV-B and drought treatment interactions were observed on photosynthesis or plant biomass. In conclusion, a 30% increase in UV-B had no effects on photosynthetic performance or productivity in well-watered or droughted pea plants in the field.  相似文献

Activation state of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) is an important parameter determining the rate of net photosynthesis (P N) in situ for which no information is available with reference to altitude. We analyzed activation state along with P N in three plant species and their cultivars grown at low (LA, 1 300 m) and high (HA, 4 200 m) altitudes. No significant change in P N and the initial activity of RuBPCO was obtained with reference to altitude. However, activation state of RuBPCO was reduced significantly in the HA plants as compared to the LA ones. Hence low partial pressure of CO2 prevailing at HA might be responsible for the lower activation state of RuBPCO.  相似文献

The carboxylating activity and content of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO, EC 4.1.1.39), and other soluble proteins in young seedlings and mature leaves of Lutescens-758, a drought-sensitive cultivar of soft spring wheat Triticum aestivum L., were studied under the conditions of drought and subsequent rehydration. Seedlings and mature plants preliminarily treated with the cytokinin-like compound kartolin-4 were compared to untreated plants. Drought-induced decrease in RuBPCO activity should be attributed not only to proteolytic decomposition of the enzyme protein itself but also to a partial inhibition of its catalytic activity. The decrease in RuBPCO activity was larger than that in RuBPCO content. Water stress induced a marked decrease in the soluble protein content. Kartolin-4 increased the resistance to drought.  相似文献

The influence of blue, red and white light and gibberellic acid (GA3) on gibberellin-like activity in tissue extracts of leaves, stems and roots was investigated during growth of pea seedlings (Pisum salivum L. cv. Bördi). Higher GA-like activity was found in leaves and stems of pea plants that were growing in blue light than in those under red or white light. The activity of NADP and O2 photoreduction by water is essentially higher in chloroplasts isolated from pea seedlings (Pisum sativum L.) grown under blue light as compared with that from plants grown under red light. In contrast, the photoreduction of NADP and O2 with photosystem I only is practically the same or even lower in chloroplasts isolated from plants grown under blue light. The addition of plastocyanin does not affect the rate or the extent of NADP photoreduction by water in the chloroplasts isolated from plants grown under blue light, whereas it sharply activates NADP reduction in the chloroplasts isolated from plants grown under red light. The extent of the light-induced oxidation of cytochrome f is appreciably higher in chloroplasts isolated from plants grown under blue light. Cytochrome b559 plays the predominant role in the oxidoreductive reactions of these chloroplasts. Furthermore, the fluorescence measurements indicate more effective transfer of excitation energy from chlorophyll to the photosystem II reaction center in chloroplasts isolated from plants grown under blue light.  相似文献

Red light-emitting diodes (LEDs) are a potential light source for growing plants in spaceflight systems because of their safety, small mass and volume, wavelength specificity, and longevity. Despite these attractive features, red LEDs must satisfy requirements for plant photosynthesis and photomorphogenesis for successful growth and seed yield. To determine the influence of gallium aluminium arsenide (GaAlAs) red LEDs on wheat photomorphogenesis, photosynthesis, and seed yield, wheat (Triticum aestivum L., cv. 'USU-Super Dwarf') plants were grown under red LEDs and compared to plants grown under daylight fluorescent (white) lamps and red LEDs supplemented with either 1% or 10% blue light from blue fluorescent (BF) lamps. Compared to white light-grown plants, wheat grown under red LEDs alone demonstrated less main culm development during vegetative growth through preanthesis, while showing a longer flag leaf at 40 DAP and greater main culm length at final harvest (70 DAP). As supplemental BF light was increased with red LEDs, shoot dry matter and net leaf photosynthesis rate increased. At final harvest, wheat grown under red LEDs alone displayed fewer subtillers and a lower seed yield compared to plants grown under white light. Wheat grown under red LEDs+10% BF light had comparable shoot dry matter accumulation and seed yield relative to wheat grown under white light. These results indicate that wheat can complete its life cycle under red LEDs alone, but larger plants and greater amounts of seed are produced in the presence of red LEDs supplemented with a quantity of blue light.  相似文献

Three types of transgenic plants of Solanum tuberosum cvs. Kamyk and Oreb, and Nicotiana tabacum cvs. Maryland Mammoth and Trapezond were selected according to intensity of introduced ipt gene expression and resulting amount of synthesised cytokinins (CKs). In comparison with controls, original transgenic regenerants grown in vitro showed a massive increase of CK contents, in tobacco by 379 % and in potato by 159 % (MAS). Potato grown in soil from tubers of transgenic plants demonstrated a moderate increase (44 %) of CK contents (MOD). Transgenic tobacco grown from seeds in vitro did not show any significant change in CK contents (NOT). Initial (RuBPCi and RuBPOi) and total (RuBPCt) activities of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO), and the activity of phosphoenolpyruvate carboxylase (PEPC) were not significantly affected by the transformation in the NOT plants. In the MOD plants, the RuBPCO activities were stimulated by up to 34 % whereas the PEPC activity was decreased by 17 %. On the other hand, all the measured enzyme activities were 32 – 91 % lower in the MAS. Leaf area, fresh and dry masses, and chlorophyll and soluble protein contents also went down with increasing CK amounts in the transformants. Dependence of RuBPCi/RuBPOi and RuBPCt/PEPC ratios on the relative CK amounts in transgenic plants revealed that the individual enzyme activities were not affected uniformly. Endogenous CK contents in the MAS thus apparently exceeded an optimum needed for positive effects on many physiological traits and became a stress factor for such plants.  相似文献

Cytokinins are plant growth regulators that induce shoot formation, inhibit senescence and root growth. Experiments with hydroponically grown tobacco plants, however, indicated that exogenously applied cytokinin led to the accumulation of proline and osmotin. These responses were also associated with environmental stress reactions, such as salt stress, in many plant species. To test whether increased endogenous cytokinin accumulation led to NaCl stress symptoms, the gene ipt from Agrobacterium tumefaciens, encoding isopentenyl transferase, was transformed into Nicotiana tabacum cv. SR-1 under the control of the light-inducible rbcS-3A promoter from pea. In high light (300 mol PPFD m-2 s-1), ipt mRNA was detected and zeatin/zeatin glucoside levels were 10-fold higher than in control plants or when transformants were grown in low light (30 mol PPFD m-2 s-1). High light treatment was accompanied by increased levels of proline and osmotin when compared to low light grown transformed and untransformed control plants. The time courses of the synthesis of diamine oxidase in pea plants grown for 14 days either in the light or in the dark are similar with the highest increase in activity occurring in the cotyledons and in the shoots during the first 6 to 8 days. Plants grown in the dark showed a 2- to 3-fold higher enzyme activity than plants grown in the light. Pea diamine oxidase could bein vivo efficiently inhibited by substrate analogues 1,4-diamino-2-butanone and 1,5-diamino-3-pentanone. The first compound inhibited proportionally to its concentration the growth of etiolated pea plants, but its instability makes an unequivocal interpretation of the results difficult. On the other hand, 1,5-diamino-3-pentanone a stable and more efficient diamine oxidase inhibitor depressed the growth of pea seedlings only at concentrations as high as 5 mM and 10 mM, at which the growth of cress seedlings not containing diamine oxidase was also strongly depressed. The results obtained indicate that tryptamine oxidation catalyzed by diamine oxidase is not involved in the main metabolic pathway leading from tryptophan to indoleacetate in pea plants.  相似文献

Plant growth, chlorophyll (Chl) content, photosynthetic gas exchange, ribulose-1,5-bisphosphate carboxylase (RuBPCO) enzyme activity, and Chl fluorescence in radish (Raphanus sativus var. longipinnatus) plants were examined after turnip mosaic virus (TuMV) infection. Plant fresh mass, dry mass, Chl content, net photosynthetic rate (P N), transpiration rate (E), stomatal conductance (g s), and RuBPCO activity were significantly lower in infected plants after 5 weeks of virus infection as compared to healthy plants. The 5-week virus infection did not induce significant differences in intercellular CO2 concentration (C i, photochemical efficiency of photosystem 2, PS2 (Fv/Fm), excitation capture efficiency of open PS2 reaction centres (Fv'/Fm'), effective quantum efficiency of photosystem 2 (ΔF/Fm'), and photochemical quenching (qP), but non-photochemical quenching (qN) and alternative electron sink (AES) were significantly enhanced. Thus the decreased plant biomass of TuMV-infected plants might be associated with the decreased photosynthetic activity mainly due to reduced RuBPCO activity.  相似文献
