We have applied the technique of saturation transfer electron paramagnetic resonance to study the rotational diffusion of spin labeled membrane bound cholinergic receptors from Torpedo marmorata. Two different spin labels were used: a spin labeled maleimide derivative which binds covalently to proteins and a long chain spin labeled acylcholine which binds reversibly with a high affinity to the receptor protein. The maleimide spin label has a motion whose rotational correlation time is τ2 > 10?3 sec. The long chain spin labeled acylcholine indicates slightly more motion (τ2 ? 10?4sec), but the nitroxide in this latter case is probably more loosely bound.  相似文献

Physical properties of pepsin-solubilized types I, II, III and V collagen have been measured in acid solution at 10°C. Our results indicate that types I, II and III collagen molecules undergo a monomer-aggregate equilibrium in solution whereas type V molecules appear to attract each other but do not undergo a similar monomer-aggregate equilibrium. Interstitial collagen monomers (I, II and III) have molecular weights between 280 × 103 and 289 × 103, translational diffusion coefficients between 0.820 × 10?7 and 0.845 × 10?7 cm2 s?1 and particle scattering factors at an angle of 175.5° and wavelength of 633 nm between 0.430 and 0.460. Type V collagen molecules after pepsin digestion were found to have a higher molecular weight (307 × 103), similar translational diffusion coefficient (0.860 × 10?7 cm2 s?1) and similar particle scattering factor at 175.5° (0.440) to the interstitial collagens. Theoretical bead models are discussed and suggest that changes in the translational diffusion coefficient were less sensitive to bending motions than were changes in the particle scattering factor at 175.5°C. Bend angles of 50° were shown to increase the particle scattering factor by 5% whereas a bend angle of greater than 125° was required to increase the translational diffusion coefficient by 5%. Models developed from idealized shapes seen by electron microscopy of rotary shadowed collagen molecules agreed best with experimental laser light scattering measurements when the bend angles were less than 90°.  相似文献

N-Phenylhydroxylamine is oxidized in aqueous phosphate buffer to nitrosobenzene, nitrobenzene, and azoxybenzene. Degradation is O2 dependent and shows general catalysis by H2PO4? (k1 = 2.3 M?2 sec?1) and PO4?3 (k2 = 2.3 × 105M?2 sec?1) or kinetically equivalent terms. Evidence is presented suggesting the intermediacy of a highly reactive species leading to these products.  相似文献

The reaction of hydroxyl radicals (?OH) and superoxide anions (O2?) with methional were investigated by pulse-radiolytic methods. The second-order rate constant for the attack of OH was determined at 8.2×109 M?1 sec?1. In the case of O2? a slow first-order decay rate of 5.2×103 sec?1 suggests a far less efficient reaction. The transient species were identified by comparison with published results of pulse radiolysis and EPR spectroscopy of model compounds. The mechanism for the oxidation of methional by OH was found to be more complex than a simple fragmentation reaction.  相似文献

In order to test the question if a pool of lipophilic ions may exist in black lipid membranes which cannot be detected by electrical relaxation measurements we have performed simultaneously measurements of the optical absorption of a lipophilic ion. The absorbance of membrane-bound dipicrylamine at 410 nm was measured with a sensitive spectrophotometer which can detect absorbance changes ? 4 · 10?5. A minimal concentration of about 6 · 1011 dipicrylamine ions per cm2 of the membrane could be detected with this instrument. The dipicrylamine concentration in the membrane obtained with the optical method Ntopt is compared with the concentrations Ntel obtained from simultaneous electrical relaxation measurements. Ntopt and Ntel agreed at low dipicrylamine concentrations (10?8–10?7 M in the aqueous phase) and showed saturation at higher concentrations (up to 5 · 10?6 M). In the saturation range Ntopt was maximally four times higher than Ntel. The significance of this difference is discussed together with general aspects of the saturation phenomenon.  相似文献

In an accompanying publication by Duckwitz-Peterlein, Eilenberger and Overath ((1977) Biochim. Biophys. Acta 469, 311–325) it is shown that the exchange of lipid molecules between negatively charged vesicles consisting of total phospholipid extracts from Escherichia coli occurs by the transfer of single lipid monomers or small micelles through the water. Here a kinetic interpretation is presented in terms of a rate constant, k?, for the escape of lipid molecules from the vesicle bilayer into the water. The evaluated rate constants are k?P = (0.86 ± 0.05) · 10?5s?1 and k?E = (1.09 ± 0.13) · 10?6s?1 for phospholipid molecules with trans-Δ9-hexadecenoate and trans-Δ9-octadecenoate, respectively, as the predominant acyl chain component. The rate constants are discussed in terms of the acyl chain and polar head group composition of the lipids.  相似文献

The lateral diffusion coefficients (D) of the molecular fluorescence probe 3,3′-dioctadecylindocarbocyanine iodide (DII) in the membrane of discoid erythrocyte ghosts has been measured with the photobleaching technique between 7°C and 40°C. A fluorescence microscope which allows bleaching experiments within small local fields (approx. 1 μm2) at high magnification (X1600) has been used for these measurements. The diffusion coefficient increases from D = 9 · 10?10cm2/s to D = 7.5 · 10?9cm2/s from 7 to 40°C. An increase in membrane fluidity between 12°C and 17°C indicates a conformational change of the lipid bilayer moiety in this temperature region. The diffusion coefficient measured in the regions between the spicules of echinocytes is appreciably smaller than in the untransformed discoid ghosts. In the myelin tubes originating from cells, the lateral diffusion is somewhat larger (about a factor of 2) than in the non-transformed ghosts. With the fluorescence probe technique the rate of growth of myelin tubes of 0.3 μm diameter has been estimated.  相似文献

The distribution of HCO3?-ATPase activity was studied in cell fractions prepared from homogenates of rat liver. The level of mitochondrial contamination in the microsomal fraction depended on the fractionation procedure and on the method of homogenization. With proper care, microsomes with undetectable mitochondrial contamination could be prepared. These microsomes had no detectable HCO3?-ATPase activity. Approximately 85 % of the total HCO3?-ATPase activity of the post 6000 x g · min supernatant was recovered in the mitochondrialfraction. The properties of this mitochondrial HCO3?-ATPase were not distinguishable from those of the various microsomal HCO3?-ATPase previously described by other investigators.  相似文献

Using a rapid pH electrode, measurements were made of the flash-induced proton transport in isolated spinach chloroplasts. To calibrate the system, we assumed that in the presence of ferricyanide and in steady-state flashing light, each flash liberates from water one proton per reaction chain. We concluded that with both ferricyanide and methylviologen as acceptors two protons per electron are translocated by the electron transport chain connecting Photosystem II and I. With methyl viologen but not with ferricyanide as an acceptor, two additional protons per electron are taken up due to Photosystem I activity. One of these latter protons is translocated to the inside of the thylakoid while the other is taken up in H2O2 formation. Assuming that the proton released during water splitting remains inside the thylakoid, we compute H+e? ratios of 3 and 4 for ferricyanide and methyl viologen, respectively.In continuous light of low intensity, we obtained the same H+e? ratios. However, with higher intensities where electron transport becomes rate limited by the internal pH, the H+e? ratio approached 2 as a limit for both acceptors.A working model is presented which includes two sites of proton translocation, one between the photoacts, the other connected to Photosystem I, each of which translocates two protons per electron. Each site presents a ≈ 30 ms diffusion barrier to proton passage which can be lowered by uncouplers to 6–10 ms.  相似文献

The molecular weight of the adenovirus type 2 hexon was calculated from sedimentation equilibrium, light scattering and sedimentation and diffusion experiments. The extinction coefficient, E1 cm1%, was determined to be 14.3 at 279 nm, from quantitative nitrogen and carbon analyses combined with the N,C content calculated from the amino acid composition. Other parameters determined were: the partial specific volume, \?gn = 0.738 cm3 g?1; the refractive index increment, (?n?c)T,P = 0.193 cm3 g?1 at 435.8 nm; the sedimentation coefficient, s20,w0 = 13.0 S; and the diffusion constant, D20, w0 = 3.32 × 10?7 cm2 s?1. All molecular weights were between 355,000 and 363,000. Crystal density measurements were made on native and glutaraldehyde cross-linked crystals and the molecular weights calculated from these data were compared with the precise molecular weight determined by physico-chemical methods.Only one polypeptide of molecular weight 120,000 was found in reduced, or reduced and alkylated, hexon. Four or six organomercurial molecules were bound per 120,000 molecular weight of native hexon upon titration with 2-chloromercuri-4-nitrophenol and 2-chloromercuri-4,6-dinitrophenol, respectively. With 5,5′-dithiobis (2-nitrobenzoic acid) only one SH-group per 120,000 could be titrated in native hexon, but after denaturation in 1% sodium dodeeyl sulphate five more SH-groups reacted per 120,000 molecular weight. Thus there are three identical polypeptides of molecular weight 120,000 per hexon of total molecular weight 360,000.  相似文献

Perfused rat hearts were treated with isoprenaline (10?6M) or ouabain (5.5 × 10?6M). The phosphate contents of troponin-I and myosin P light chains were established by radiolabelling with 32P; in the case of the light chains, direct chemical analysis of total and of specifically alkali-labile phosphate was also performed. Addition of isoprenaline caused phosphorylation of both troponin-I and myosin P light chains, reaching a maximum increment, after several minutes, of 1 mol/mol and 0.30 mol/mol, respectively. The Mg2+-ATPase activities, at saturating Ca2+ concentrations, of natural actomyosin isolated from treated hearts were significantly depressed, and an inverse correlation was established between the phosphate content of troponin-I and the Vmax[Ca2+] of this ATPase activity. The Ca2+ sensitivity of the Ca2+Mg2+-ATPase was also decreased. These changes were all reversed by an incubation permitting dephosphorylation of proteins by endogenous phosphatases.Treatment of hearts with ouabain caused no increment in troponin-I phosphorylation, but increased the P light chain phosphate content to a maximum of 0.30 mol/mol after some minutes. A positive correlation was evident between phosphate content of the light chains (in all experiments) and the maximum myosin Ca2+-ATPase activities. In addition, the Vmax[ATP] of the Ca2+Mg2+-ATPase of natural actomyosin was increased when light chain phosphorylation had occurred in the absence of troponin-I phosphorylation. P-light chain phosphorylation did not affect the Ca2+ sensitivity of Ca2+Mg2+-ATPase activity.We suggest that the effects of phosphorylation of troponin-I are to diminish thin filament sensitivity to Ca2+, and to decrease the efficiency of the transduction process along neighbouring actin monomers, such that the number of actin-myosin crossbridge interactions is decreased even in the presence of Ca2+ excess. Phosphorylation of P light chains of myosin has an activating effect on myosin Ca2+-ATPase activity, as well as on the rate of cross-bridge formation.  相似文献

The structure and dynamics of phosphatidylcholine bilayers containing chlorophyll were studied by X-ray diffraction and absorption polarization spectroscopy in the form of hydrated orientated multilayers below the thermal phase transition of the lipid chains and by nuclear magnetic resonance in the form of single-wall vesicles above the thermal transition. Our results show that (a) chlorophyll is incorporated into the phosphatidylcholine bilayers with its porphyrin ring located anisotropically in the polar headgroup layer of the membrane and with its phytol chain penetrating in a relatively extended form between the phosphatidylcholine fatty acid chains in the hydrocarbon core of the mixed bilayer membrane and (b) the intramolecular anisotropic rotational dynamics of the host phosphatidylcholine molecules are significantly perturbed upon chlorophyll incorporation into the bilayer at all levels of the phosphatidylcholine structure. These dynamics for the host phosphatidtlcholine fatty acid chains are qualitatively different from that of the incorporated chlorophyll phytol chains on a 10?9 ? 10?10s time scale in the ideally mixed two-component bilayer.  相似文献

The oxidation of unsaturated fatty acid micelles by the superoxide free radical (O?2), during γ irradiation in the presence of formate, is kinetically distinct from oxidation by hydroxyl free radicals (HO.). The evidence suggests that a direct reaction between (O?2) and lipid hydroperoxide initiates a chain oxidation process in the micelles. While tetranitromethane, which reacts rapidly with (O?2), protects the micelles from oxidation, active superoxide dismutase is no more effective than its apoprotein, due to lack of penetration of the micellar environment. We discuss these findings in the light of recent literature, and with reference to their possible significance for biological systems.  相似文献

In many instances the effect of superoxide (O2?) trapping agents in suppressing the net rate of O2 consumption of activated PMN's is not in accordance with theoretical expectations. We offer here an alternate explanation to those previously presented by Segal and Meshulam (FEBS Letters 100, 27–32) and Babior (Biochem. Biophys. Res. Comm. 91, 222–226). The paradoxical results previously presented can be explained by recognizing that shortly after activation of resting cells an O2 diffusion layer is established at or near the outer surface of these cells. The presence of this diffusion layer can markedly alter the anticipated stochiometric relationship between O2? trapped and apparent O2 consumed by these cells when they are exposed to O2? trapping agents.  相似文献

Diffusion of histamine, theophylline and tryptamine through planar lipid bilayer membranes was studied as a function of pH. Membranes were made of egg phosphatidylcholine plus cholesterol (1 : 1 mol ratio) in tetradecane. Tracer fluxes and electrical conductances were used to estimate the permeabilities to nonionic and ionic species. Only the nonionic forms crossed the membrane at a significant rate. The membrane permeabilities to the nonionic species were: histamine, 3.5 · 10?5cm · s?1; theophylline, 2.9 · 10?4cm · s?1; and tryptamine, 1.8 · 10?1cm · s?1. Chemical reactions in the unstirred layers are important in the transport of tryptamine and theophylline, but not histamine. For example, as pH decreased from 10.0 to 7.5 the ratio of nonionic (B) to ionic (BH+) tryptamine decreased by 300-fold, but the total tryptamine permeability decreased only 3-fold. The relative insensitivity of the total tryptamine permeability to the ratio, [B]/[BH+], is due to the rapid interconversion of B and BH+ in the instirred layers. Our model describing diffusion and reaction in the unstirred layers can explain some ‘anomalous’ relationships between pH and weak acid/base transport through lipid bilayer and biological membranes.  相似文献

The diffusion coefficients D(cm2 · s?1) of the sodium salts of a series of hydrophilic mono- and dicarboxylic acids, have been measured in the hydrophilic layers of phosphatidylcholine-water lamellar phases, as a function of phase hydration. At pH 9.0, the diffusion rates of the anionic (RCOO?) form of the acid exhibit a prominent increase within a narrow range of water content, specific to each anion. This high diffusion rate seems to occur when the Stokes diameter of an anion is equal to the thickness of the aqueous layer between the two planes formed by the quaternary ammonium groups of the choline phosphate dipoles of two facing layers of phosphatidylcholine molecules. This phenomenon demonstrates the importance of the spatial organization of successive binding sites in the rate constant of diffusional processes in hydrophilic channels.  相似文献

A steady-state competition system has been developed to investigate the reactions of the superoxide radical anion (O2?) with various peroxides, including the so-called Haber-Weiss reaction. Potassium superoxide dissolved in an oxygen-free solution of DMSO containing 18-dicyclohexyl-6-crown, is the source of O2?. High pressure liquid chromatography is used as an assay system for O2? reactivity, to detect and quantitate the yield of anthracene, formed as a major product in the reaction between O2? and 9,10-dihydroanthrancene. Decrease in anthracene yields, in the presence of peroxide, may be used to indicate a possible competing reaction between O2? and added peroxide. Complications involving peroxide-stimulated formation of anthraquinone derivatives are discussed. No evidence for a competing reaction between O2? and peroxide can be detected up to a 10-fold excess of peroxide over 9,10-dihydroanthracene.  相似文献

The lipid dynamics of the adrenocortical microsomal membranes was studied by monitoring the fluorescence anisotropy and excited state lifetime of a set of anthroyloxy fatty acid probes (2-, 7-, 9- and 12-(9-anthroyloxy)-stearic acid (AP) and 16-(9-anthroyloxy)palmitic acid (AS). It was found that a decreasing polarity gradient from the aqueous membrane interface to the membrane interior, was present. This gradient was not modified by the proteins, as evidenced by comparison of complete membranes and derived liposomes, suggesting that the anthroyloxy probes were not in close contact with the proteins. An important change of the value of the mean rotational relaxation time as a function of the position of the anthroyl ring along the acyl chain was evidenced. In the complete membranes, a relatively more fluid medium was evidenced in the C16 as compared to the C2 region, while the rotational motion appeared to be the most hindered at the C7–C9 level. In the derived liposomes, a similar trend was observed but the mobility was higher at all levels. The decrease of the mean rotational relaxation time was more important for 12-AS and 16-AP. Temperature dependence of the mean rotational relaxation time of 2-AS, 12-AS and 16-AP in the complete membranes revealed the existence of a lipid reorganization occurring around 27°C and concerning mainly the C16 region. The extent to which the acyl chain reacted to this perturbation at the C12 level depended on pH. The presence of proteins increased the apparent magnitude of this reorganization and also modified the critical temperature from approx. 23°C in the derived liposomes to approx. 27°C in the complete membranes. Thermal dependence of the maximum velocity of the 3-oxosteroid Δ54-isomerase, the second enzyme in the enzymatic sequence, responsible for the biosynthesis of the 3-oxo4-steroids in the adrenal cortex microsomes, was studied. The activation energy of the catalyzed reaction was found to be low and constant (2–5 kcal · mol?1) in the temperature range 16–40°C at pH 7.5, 8.5 and 9, corresponding to the minimum, intermediate and maximum rate, respectively. A drastic increase of the activation energy (20 kcal · mol?1) was observed at temperature below 16°C at pH 7.5. A correlated change of the pKESapp as function of temperature was detected; at 36°C pKESapp = 8.3 while at 13°C the value shifted to 8.7. The pH range of the group ionization was narrower at 13°C. In contrast with the behaviour of the 3β-hydroxy5-steroid dehydrogenase, the 3-oxosteroid Δ54-isomerase was apparently unaffected by the lipid reorganization at 27°C. It is suggested that this enzyme possesses a different and more fluid lipid environment than the bulk lipids.  相似文献

The stability of mouse uterine cytosol receptor-[3H]estradiol complex was evaluated in the presence of neutral salts of the Hoffmeister series. Marked increases in the rate of dissociation of the complex were observed with the more chaotropic anions (SCN?, ClO4?, NO3?, Br?), and the effects of these ions were greater at lower temperatures, where water assumes a more rigid structure. At higher temperatures F? and CH3COO?, which tend to stabilize water structure, led to retardation of the rate of dissociation of the hormone-receptor complex. There was essentially no change in steroid specificity in the presence of the markedly chaotropic salts. The perturbation of water structure adjacent to the steroid binding site is a factor to be considered in the isolation of steroid receptor complexes.  相似文献

From phosphomannose isomerase-less mutants of E. coli strains 08 and 09, rfe? derivatives were constructed by recombination with a Salmonella rfe? donor. In contrast to membranes from the parent E. coli strains, those from the rfe? recombinants did not synthesize the 08 or 09 mannan from GDP mannose in vitro. They could, however, be restored to biosynthetic activity with butanol extracts from the E. coli rfe+ bacteria. This indicated that the rfe mutation affects the synthesis of a hydrophobic acceptor.  相似文献
