Anatomically preserved gymnosperm axes are relatively abundant in Permian localities of Antarctica, but their anatomy has rarely been studied in detail, which limits comparison with other Gondwanan morphotaxa. Here we describe a silicified trunk collected from the Upper Permian Buckley Formation at Coalsack Bluff, in the central Transantarctic Mountains. The trunk has a small heterogeneous pith approximately 4 mm in diameter containing conspicuous sclerotic nests, endarch primary xylem maturation, paired leaf traces, and secondary xylem of the Araucarioxylon type. Comparison with contemporaneous gymnosperm axes from Antarctica indicates that the Coalsack Bluff trunk represents a new Permian morphotaxon for the region. The anatomical characters of the pith and secondary xylem suggest an affinity with the genus Kaokoxylon Kräusel, previously reported from Permian and Triassic localities of Southern Africa, South America, India, and Australia.  相似文献

Isolations from oak symptomatic of Acute Oak Decline, alder and walnut log tissue, and buprestid beetles in 2009–2012 yielded 32 Gram-negative bacterial strains showing highest gyrB sequence similarity to Rahnella aquatilis and Ewingella americana. Multilocus sequence analysis (using partial gyrB, rpoB, infB and atpD gene sequences) delineated the strains into six MLSA groups. Two MLSA groups contained reference strains of Rahnella genomospecies 2 and 3, three groups clustered within the Rahnella clade with no known type or reference strains and the last group contained the type strain of E. americana. DNA–DNA relatedness assays using both the microplate and fluorometric methods, confirmed that each of the five Rahnella MLSA groups formed separate taxa. Rahnella genomospecies 2 and 3 were previously not formally described due to a lack of distinguishing phenotypic characteristics. In the present study, all five Rahnella MLSA groups were phenotypically differentiated from each other and from R. aquatilis. Therefore we propose to classify the strains from symptomatic oak, alder and walnut and buprestid beetles as: Rahnella victoriana sp. nov. (type strain FRB 225T = LMG 27717T = DSM 27397T), Rahnella variigena sp. nov. (previously Rahnella genomosp. 2, type strain CIP 105588T = LMG 27711T), Rahnella inusitata sp. nov. (previously Rahnella genomosp. 3, type strain DSM 30078T = LMG 2640T), Rahnella bruchi sp. nov. (type strain FRB 226T = LMG 27718T = DSM 27398T) and Rahnella woolbedingensis sp. nov. Tree-ring research in Ireland has typically been dominated by Quercus species, particularly Quercus petraea and Quercus robur. Recent years have seen a greater focus on multi-species reconstructions in Ireland but, due to difficulties with the hardness of the wood, missing/pinched rings and fused stems, Taxus baccata has not been included in these investigations. Despite these difficulties, a 31-tree, 204-year T. baccata chronology was successfully constructed from Killarney National Park, southwest Ireland. The chronology exhibits promising dendroclimatological potential, with climatic responsiveness equivalent to that of the other major Irish tree taxa, including Quercus. The chronology shows the strongest relationship with May–June precipitation from Muckross House synoptic station (1970–2007; r = 0.521, p < 0.01) and Valentia Observatory (1941–2007; r = 0.545, p < 0.01). November–April temperatures also exhibited a strong relationship with the chronology post-1970 (r = 0.605, p < 0.01 for Muckross House, r = 0.567, p < 0.01 for Valentia Observatory), but this relationship is not time stable and breaks down for the pre-1970 Valentia Observatory record. The long-lived nature of T. baccata, the exceptional preservation of wood and rings in this hard softwood species, as well as its prominence in Irish archaeology, all point to the potential to expand this chronology both spatially and temporally, and demonstrate T. baccata's potential in multi-site and multi-species tree-ring studies in the region.  相似文献

Bacteria of the genus Massilia often colonize extreme ecosystems, however, a detailed study of the massilias from the Antarctic environment has not yet been performed. Here, sixty-four Gram-stain-negative, aerobic, motile rods isolated from different environmental samples on James Ross Island (Antarctica) were subjected to a polyphasic taxonomic study. The psychrophilic isolates exhibited slowly growing, moderately slimy colonies revealing bold pink-red pigmentation on R2A agar. The set of strains exhibited the highest 16S rRNA gene sequence similarities (99.5–99.9%) to Massilia violaceinigra B2T and Massilia atriviolacea SODT and formed several phylogenetic groups based on the analysis of gyrB and lepA genes. Phenotypic characteristics allowed four of them to be distinguished from each other and from their closest relatives. Compared to the nearest phylogenetic neighbours the set of six genome-sequenced representatives exhibited considerable phylogenetic distance at the whole-genome level. Bioinformatic analysis of the genomic sequences revealed a high number of putative genes involved in oxidative stress response, heavy-metal resistance, bacteriocin production, the presence of putative genes involved in nitrogen metabolism and auxin biosynthesis. The identification of putative genes encoding aromatic dioxygenases suggests the biotechnology potential of the strains. Based on these results four novel species and one genomospecies of the genus Massilia are described and named Massilia rubra sp. nov. (P3094T = CCM 8692T = LMG 31213T), Massilia aquatica sp. nov. (P3165T = CCM 8693T = LMG 31211T), Massilia mucilaginosa sp. nov. (P5902T = CCM 8733T = LMG 31210T), and Massilia frigida sp. nov. (P5534T = CCM 8695T = LMG 31212T).  相似文献

We investigated the role of hydraulic conductivity, wood density, and xylem cavitation in the response of baldcypress (Taxodium distichum) seedlings to increased soil salinity and drought. One-year-old, greenhouse-grown seedlings were irrigated daily with a 100 mM (≈6‰) salt solution or once per week with fresh water (drought). Controls were irrigated daily with fresh water. Gas exchange rates of stressed plants were reduced by approximately 50% (salt) and 70% (drought), resulting in a 50–60% reduction in diameter growth for both treatments. Stem-specific hydraulic conductivity (KS native) of stressed plants was 33% (salt) and 66% (drought) lower than controls and we observed a strong positive correlation between KS native and gas exchange. In addition, we found a strong relationship between CO2 assimilation rate (A) and the soil-to-leaf hydraulic conductance (kL). The relationship was identical for all treatments, suggesting that our moderate salt stress (as well as drought) did not affect the photosynthetic biochemistry of leaves, but rather reduced A via stomatal closure. Lower KS native of stressed plants was associated with increased wood density and greater resistance to xylem cavitation. Xylem pressures causing 50% loss of hydraulic conductivity (P50) were ?2.88 ± 0.07 MPa (drought), ?2.50 ± 0.08 MPa (salt) and ?2.01 ± 0.04 MPa (controls). P50s were strongly correlated with wood density (r = ?0.71, P < 0.01) and KS native (r = 0.74, P < 0.01). These findings support the hypothesis that there is a significant trade-off between a plant's cavitation resistance and its hydraulic efficiency. The results of the present study indicate that stressed plants partitioned their biomass in a way that strengthened their xylem and reduced vulnerability to xylem cavitation. Hence, these seedlings could be better suited to be planted in environments with elevated soil salinity. For most parameters (especially P50), drought had an even more pronounced effect than salinity. This is important as nurseries could produce “stress-acclimated” seedlings simply by reducing irrigation amounts and would not have to contaminate the soils in their nursery beds with salt applications.  相似文献

Ramageoptera platycephala gen. et sp. nov., a new protopristocerine wasp, is described and figured from a female individual preserved in mid-Cretaceous amber from Tanai, northern Myanmar. Ramageoptera platycephala gen. et sp. nov. shows most of the known characters used to circumscribe the extinct subfamily Protopristocerinae, and highlights the putative status of stem-Pristocerinae that may occupy the Protopristocerinae. Ramageoptera platycephala gen. et sp. nov. is unique among the Protopristocerinae, at least, for possessing a flat and square shaped head; eyes glabrous and small; mandibles with four conspicuous sharp teeth; antenna with short scape; forewing with Rs + M vein absent; vein M straight; cell 2R1 distally opened; cell 1Cu closed, sub-equal to R cell; cell 2Cu open.  相似文献

One new foraminiferal genus, Rectoformata gen. nov., with two new species, has been described from a Permian limestone block in the Karakaya Complex in the South of the Balya district (Balikesir, northwestern Turkey). The type species of Rectoformata, Rectoformata tekini sp. nov., is characterized by a quadrangular transverse section having a calcareous microgranular wall with fine alveolar structures, whereas Rectoformata acari sp. nov. is recognized by a pentagonal transverse section having a calcareous microgranular wall with fine alveolar structures. The age of the described taxa is assigned as Midian ( = Capitanian) based on the association with small foraminifers and fusulinids. These new taxa are included in the family Chitralinidae, which is emended herein.  相似文献

Pirellula-like planctomycetes are ubiquitous aquatic bacteria, which are often detected in anoxic or micro-oxic habitats. By contrast, the taxonomically described representatives of these bacteria, with very few exceptions, are strict aerobes. Here, we report the isolation and characterization of the facultatively anaerobic planctomycete, strain PX69T, which was isolated from a boreal lake. Its 16S rRNA gene sequence is affiliated with the Pirellula-related Pir4 clade, which is dominated by environmental sequences retrieved from a variety of low-oxygen habitats. Strain PX69T was represented by ellipsoidal cells that multiplied by budding and grew on sugars, some polysaccharides and glycerol. Anaerobic growth occurred by means of fermentation. Strain PX69T grew at pH 5.5–7.5 and at temperatures between 10 and 30 °C. The major fatty acids were C18:1ω9c, C16:0 and C16:1ω7c; the major intact polar lipid was dimethylphosphatidylethanolamine. The complete genome of strain PX69T was 6.92 Mb in size; DNA G + C content was 61.7 mol%. Among characterized planctomycetes, the highest 16S rRNA gene similarity (90.4%) was observed with ‘Bythopirellula goksoyri’ Pr1d, a planctomycete from deep-sea sediments. We propose to classify PX69T as a novel genus and species, Lacipirellula parvula gen. nov., sp. nov.; the type strain is strain PX69T (=KCTC 72398T = CECT 9826T = VKM B-3335T). This genus is placed in a novel family, Lacipirellulaceae fam. nov., which belongs to the order Pirellulales ord. nov. Based on the results of comparative genome analysis, we also suggest establishment of the orders Gemmatales ord. nov. and Isosphaerales ord. nov. as well as an emendation of the order Planctomycetales.  相似文献

One-pot conversion with whole cells of bacteria was performed for biooxidation of meso monocyclic (3a–b) and bicyclic diols (3c–e) into corresponding chiral lactones of bicyclo[4.3.0]nonane structure (2a–b) as well as exo- and endo-bridged lactones with the structure of [2.2.1] (3c–d) and [2.2.2] (3e). Micrococcus sp. DSM 30771 was selected as biocatalyst with significant alcohol dehydrogenase activity. Among tested strains, microbial oxidation of meso diols 3a–e catalyzed by Micrococcus sp. afforded enantiomerically pure ((+)-(2S,3R)-2c (ee = 99%), (+)-(2S,3R)-2e (ee = 99%)) or enriched ((+)-(1S,5R)-2a (ee = 90%), (−)-(1S,5R)-2b (ee = 86%), (+)-(2S,3R)-2d (ee = 80%)) lactone moieties. Comparative study with respect to microbial cultivation as well as biooxidation was undertaken to verify agreement of secondary metabolite biosynthesis in different scales: from MTP (4 mL), across shake flask (100 mL) till bioreactor (4 L). The results from biotransformations showed quite similar dependence in oxidation of all substrates 3a–e in MTP and flasks as well, thereby confirmed the validity and reasonable approach of using MTP for preliminary studies.  相似文献

In order to detect the response of trees to environmental pollution in Shenyang during the last century, tree rings of two 100+ year old Chinese pine (Pinus tabulaeformis Carr.) were analyzed for nine trace element concentrations (K, Ca, Mg, Mn, Cu, Zn, Al, Pb and Cd) and compared to local environmental change. The concentrations of Cu, Zn and Al had significant (p < 0.01) increasing trends after 1940s and correlated significantly (p < 0.01), with increasing production of nonferrous metal from local industrial processing. Concentrations of nutrient element (K, Ca and Mg) showed significant (p < 0.01) increasing trend from heartwood to sapwood with declines in the last three decades. Significant correlation between climatic factors and element concentration in tree rings were also found, in which the temperature (November to May) and relative humidity in April and May showed significant positive correlation with Ca, Zn and Cu (p < 0.05) as well as Al (p < 0.01) concentrations in tree rings. April rainfall showed significant positive correlation with K and Zn (p < 0.05) as well as Al (p < 0.01) concentration in tree rings. The results suggest that tree rings from Chinese pine in Shenyang are sensitive to environmental change and have the potential to be used as a biomonitor for the environmental pollution. The results also demonstrate the feasibility of applying dendrochemical techniques to areas with limited samples.  相似文献

Secoisolariciresinol diglucosides (SDGs) (S,S)-SDG-1 (major isomer in flaxseed) and (R,R)-SDG-2 (minor isomer in flaxseed) were synthesized from vanillin via secoisolariciresinol (6) and glucosyl donor 7 through a concise route that involved chromatographic separation of diastereomeric diglucoside derivatives (S,S)-8 and (R,R)-9. Synthetic (S,S)-SDG-1 and (R,R)-SDG-2 exhibited potent antioxidant properties (EC50 = 292.17 ± 27.71 μM and 331.94 ± 21.21 μM, respectively), which compared well with that of natural (S,S)-SDG-1 (EC50 = 275.24 ± 13.15 μM). These values are significantly lower than those of ascorbic acid (EC50 = 1129.32 ± 88.79 μM) and α-tocopherol (EC50 = 944.62 ± 148.00 μM). Compounds (S,S)-SDG-1 and (R,R)-SDG-2 also demonstrated powerful scavenging activities against hydroxyl [natural (S,S)-SDG-1: 3.68 ± 0.27; synthetic (S,S)-SDG-1: 2.09 ± 0.16; synthetic (R,R)-SDG-2: 1.96 ± 0.27], peroxyl [natural (S,S)-SDG-1: 2.55 ± 0.11; synthetic (S,S)-SDG-1: 2.20 ± 0.10; synthetic (R,R)-SDG-2: 3.03 ± 0.04] and DPPH [natural (S,S)-SDG-1: EC50 = 83.94 ± 2.80 μM; synthetic (S,S)-SDG-1: EC50 = 157.54 ± 21.30 μM; synthetic (R,R)-SDG-2: EC50 = 123.63 ± 8.67 μM] radicals. These results confirm previous studies with naturally occurring (S,S)-SDG-1 and establish both (S,S)-SDG-1 and (R,R)-SDG-2 as potent antioxidants and free radical scavengers for potential in vivo use.  相似文献

The effect of salinity on leaf area and the relative accumulation of Na+ and K+ in leaves of the mangrove associate Hibiscus tiliaceus were investigated. Photosynthetic gas exchange characteristics were also examined under arid and non-arid leaf conditions at 0, 10, 20 and 30‰ substrate salinity. At salinities  40‰, plants showed complete defoliation followed by 100% mortality within 1 week. Salinities  30‰ were negatively correlated with the total leaf area per plant (r2 = 0.94). The reduction in the total plant leaf area is attributed to the reduction in the area of individual leaves (r2 = 0.94). Selective uptake of K+ over Na+ declined sharply with increasing salinity, where K+/Na+ ratio was reduced from 6.37 to 0.69 in plants treated with 0 and 30‰, respectively. Under non-arid leaf condition, increasing salinity from 0 to 30‰ has significantly reduced the values of the intrinsic components of photosynthesis Vc,max (from 50.4 to 18.4 μmol m−2 s-1), Jmax (from 118.0 to 33.8 μmol photons m−2 s−1), and VTPU (from 6.90 to 2.30 μmol m−2 s−1), while stomatal limitation to gas phase conductance (SL) increased from 14.6 to 38.4%. Water use efficiency (WUE) has subsequently doubled from 3.20 for the control plants to 8.93 for 30‰ treatment. Under arid leaf conditions, the stomatal factor (SL) was more limiting to photosynthesis than its biochemical components (73.4 to 26.6%, respectively, at 30‰). It is concluded that salinity causes a drastic decline in photosynthetic gas exchange in H. tiliaceus leaves through its intrinsic and stomatal components, and that the apparent phenotypic plasticity represented by the leaf area modulation is unlikely to be the mechanism by which H. tiliaceus avoids salt stress.  相似文献

A study was done on the taxonomy and morphology of plant parasitic nematodes (Tylenchida) found in a Guneid sugarcane factory field of Saccharum officinarum (sugarcane), in Gezira state, Sudan. The samples were collected from around the root zone of sugarcane stools at a depth of 10 to 15 cm. One new species belonging to Hoplolaimidae was identified and studied. Helicotylenchus zeidani sp. nov. is characterized by a smooth head and a conical tail with a projection. It is 0.72 mm (0.62 to 0.81 mm) long, a = 32.4 (25.6 to 36.6), c = 36.9 (29.2 to 53.1) with a medium sized body and shorter stylet.  相似文献

Megastigmane glycosides (15) together with seven (612) related known compounds were isolated from the whole plants of Gynostemma pentaphyllum. This study investigates Pb isotope ratios at low concentrations (parts per billion; ppb) in tree rings and soils in the Northern Athabasca Oil Sands Region (NAOSR), western Canada, to evaluate if: (1) climatic conditions influence on tree-ring Pb assimilation; and (2) such low Pb content allows inferring the regional Pb depositional history.Our results reflect the influence of winter snow cover and the importance of minimum temperature and precipitation in spring and summer on the bioavailability of Pb and its passive assimilation by trees in sub-arctic semi-humid climatic conditions. Winter conditions can influence the state of root systems that subsequently impacts the following growth period, while spring and summer conditions likely control microbial processes and water source, and may thus impact Pb assimilation by trees. Thus, the results of tree-ring Pb concentrations show interesting correlation with cumulated snow from November of the previous year to February (ρ = 0.53; P < 0.01; n = 36). Likewise, the 206Pb/207Pb ratios inversely correlate with minimum temperature from April to September (ρ = −0.67; P < 0.01; n = 40) and precipitation from May to August (ρ = −0.42; P < 0.01; n = 36). The isotopic results also suggest that the effects of climatic variations are superimposed by regional industrial Pb deposition: Western North American Aerosols (WNAA) and fugitive dust from the oil sands mining operations appear to be the most likely sources.Importantly, this study suggests that even at low Pb concentrations, tree-ring Pb isotopes are modulated by climatic conditions and potential input of regional and long-range transport of airborne Pb. These interpretations open the possibility of using Pb isotopes as an environmental tool for inferring the pollution history in remote regions, and improving our understanding of its natural cycle through the forest environment.  相似文献

A new toxin-producing marine diatom, Nitzschia bizertensis sp. nov., isolated from the Bizerte Lagoon (Tunisia, Southwest Mediterranean Sea) is, based on studies on eight different strains, characterized morphologically by light microscopy, transmission and scanning electron microscopy, and phylogenetically using the nuclear rDNA regions: SSU, ITS1, 5.8S, ITS2 and D1–D3 of the LSU. The species belongs to the sections Lanceolatae or Lineares as defined by Cleve and Grunow (1880). These sections are characterized by species having linear-lanceolate valves with an eccentric raphe where the fibulae does not extend into the valve, and are otherwise famous for the lack of characters useful for delineation of species. Nitzschia bizertensis differs from most other species in these sections by having a high density of interstriae. The morphological and phylogenetic studies and comparisons with previously described Nitzschia species showed Nitzschia bizertensis sp. nov. to be a new species. Batch culture experiments were conducted for estimations of maximum growth rate and production of domoic acid (DA). Maximum cellular DA content of the examined strains ranged from 2 × 10−4 to 3.6 × 10−2 pg cells−1. The total DA concentration (pg mL−1) was high already in exponential growth phase maybe due to reinoculation of “old” stationary phase cells, and increased into stationary growth phase where it reached a stationary level varying among the strains from ca. 4500 to 9500 pg mL−1. Nitzschia bizertensis represents a new domoic acid-producing diatom and is the second toxin producing Nitzschia species. The resolution of Nitzschia bizertensis and Nitzschia navis-varingica in different parts of the LSU phylogenetic tree, and the recovery of the Pseudo-nitzschia species phylogenetically distant from those two species suggests that the ability to produce DA either evolved multiple times independently or was lost multiple times.  相似文献

Two novel anaerobic alkaliphilic strains, designated as LacTT and LacVT, were isolated from the Prony Bay Hydrothermal Field (PBHF, New Caledonia). Cells were motile, Gram-positive, terminal endospore-forming rods, displaying a straight to curved morphology during the exponential phase. Strains LacTT and LacVT were mesophilic (optimum 30 °C), moderately alkaliphilic (optimum pH 8.2 and 8.7, respectively) and halotolerant (optimum 2% and 2.5% NaCl, respectively). Both strains were able to ferment yeast extract, peptone and casamino acids, but only strain LacTT could use sugars (glucose, maltose and sucrose). Both strains disproportionated crotonate into acetate and butyrate. Phylogenetic analysis revealed that strains LacTT and LacVT shared 96.4% 16S rRNA gene sequence identity and were most closely related to A. peptidifermentans Z-7036, A. namsaraevii X-07-2 and A. hydrothermalis FatMR1 (95.7%–96.3%). Their genome size was of 3.29 Mb for strain LacTT and 3.06 Mb for strain LacVT with a G + C content of 36.0 and 33.9 mol%, respectively. The ANI value between both strains was 73.2 %. Finally, strains LacTT (=DSM 100337 = JCM 30643) and LacVT (=DSM 100017 = JCM 30644) are proposed as two novel species of the genus Alkaliphilus, order Clostridiales, phylum Firmicutes, Alkaliphilus serpentinus sp. nov. and Alkaliphilus pronyensis sp. nov., respectively. The genomes of the three Alkaliphilus species isolated from PBHF were consistently detected in the PBHF chimney metagenomes, although at very low abundance, but not significantly in the metagenomes of other serpentinizing systems (marine or terrestrial) worldwide, suggesting they represent indigenous members of the PBHF microbial ecosystem.  相似文献

A new genus and species of anaxyelid wasp is described and figured from the mid-Cretaceous Burmese amber as Curvitexis kopylovi gen. et sp. nov. The placement of this new genus within the Anaxyelidae is corroborated by its wing venation and the configuration of its mesosoma. This new genus differs from all other Syntexinae from the mid-Cretaceous Burmese amber biota, inter alia, owing to its forewing with the vein 1Rs curved and longer than 1M, the crossvein 1r-rs absent, the vein 2Rs + M present; the hind wing with the abscissa 2M + Cu present, the vein m-cu absent, and the cell r closed. The recently described Paraxiphydria resinata Gao et al., 2022 is transferred to Anaxyelidae: Syntexinae, and the subfamily Paraxiphydriinae Gao et al., 2022 is synonymized under Syntexinae.  相似文献

The range of vertebrates that serve as intermediate hosts for parasites in the genus Sarcocystis remains incompletely defined. Here, we provide the first report of infections in treeshrews, describe the morphology of encysted parasites using light and transmission electron microscopy, and place this agent within a phylogenetic context by sequencing and comparing its 18 S ribosomal DNA to that of related parasites. Muscle infections were diagnosed in four of 45 wild treeshrews captured in the vicinity of Kunming, Yunnan Province, Mainland China. Thread-like cysts (10.773 ± 2.411 mm in length, 0.106 ± 0.009 mm in width) had walls (0.538–0.746 µm thick) that lacked perpendicular protrusions. The interior of the cyst was packed full with cyst merozoites, the shape of which was typical of Sarcocystis. The primary cyst wall consisted of a thin membrane supported by osmiophilic material, 31–60 nm in thickness. The ground substance was about 105–526 nm thickness. Cysts conformed to typical of ‘type 1’ sarcocysts. Freshly examined and frozen specimens did not differ in their cyst wall structure, however, the appearance of bradyzoites did differ: the conoid, rhoptries and micronemes were all visible in fresh bradyzoites; in stored bradyzoites, by contrast, the rhoptries appeared smaller, and although the conoid was visible, the micronemes were not. 18 S rRNA gene was distinct from any previously reported sequence in GenBank. Their genetic and morphological uniformity suggest that these parasites, derived from treeshrews, represent a single biological species, Sarcocystis tupaia, sp. nov.  相似文献
