Plants regenerated from tobacco (Nicotiana tabacum L. cv. Wisconsin 38) cells that were adapted to 428 mM NaCl were found to have hexaploid or near-hexaploid chromosome numbers compared to the normal tetraploid, 2N(2C)=4X=48 chromosome numbers of plants regenerated from unadapted cells. Even though cells with chromosome numbers other than hexaploid were found in the cell population only hexaploid plants were regenerated. The hexaploid condition may impart some karyotypic stability that allows more efficient morphogenic activity. The hexaploid condition could not be correlated with several phenotypic alterations associated with plants regenerated from adapted cells, including male sterility and increased salt tolerance.  相似文献

Summary Growth reduction or cessation is an initial response of Atriplex nummularia L. cells to NaCl. However, A. nummularia L. cells that are adapted to 342 and 428 mM NaCl are capable of sustained growth in the presence of salt. Cells that are adapted to NaCl exhibit a reduced rate of division compared to unadapted cells. Unlike salt adapted cells of the glycophyte Nicotiana tabacum L., A. nummularia L. cells do not exhibit reduced rate of cell expansion after adaptation. However, the cell expansion rate of unadapted A. nummularia L. cells is considerably slower than that of unadapted glycophyte cells and this normally low rate of cell expansion may contribute to the enhanced capacity of the halophyte to tolerate salt. Turgor of NaCl adapted cells was equivalent to unadapted cells indicating that the cells of the halophyte do not respond to salt by osmotic over adjustment as reported for the glycophyte tobacco (Binzel et al. 1985, Plant Physiol. 79:118–125).  相似文献

Most cells of the male PTK2 cell line contain one nucleolus, and they are diploid. However, a proportion of cells have more than one nucleolus. Cells with two and three nucleoli were isolated and cloned into viable populations. Greater than 90% of the cells in these clonal populations maintained the abnormal nucleolar number of the originally isolated cell. Karyotype analysis of cells with two and three nucleoli demonstrated that the cells were respectively tetraploid and hexaploid. It was concluded that in PTK2 cells, nucleolarity is a good index of ploidy even if the ploidy level is abnormal. Furthermore, long term monitoring of the tetraploid cells demonstrated virtually no tendency towards reversion to the diploid condition as suggested by other studies in Potorous.  相似文献

Ploidy variation of pronamide-treated maize calli during long term culture   总被引:1,自引:0,他引:1  
Summary Anther-derived calli of corn were treated with 10 M pronamide for 2, 3 and 4 days. The ploidy level of the calli was then evaluated using flow cytometry, at different times after the treatment. Untreated haploid calli did not change in ploidy level for 97 days but by 466 days, there were up to 50% diploid or higher ploidy cells thus showing that spontaneous doubling may occur during corn calli subculture with this genotype. Pronamide treatment did increase the percentage of diploid and tetraploid cells and by 466 days, all of the lines showed an additional change toward higher ploidy levels. This change may be due to spontaneous chromosome doubling or to differential cell cycle times of cells with different ploidy levels. The ploidy level of plants regenerated from the cultures was determined by counting the guard cell chloroplast numbers and the correlation with the ploidy level of the cultures was r2=0.84. These studies show that pronamide treatments can increase haploid maize callus chromosome numbers and that spontaneous chromosome doubling can occur with time in maize callus.  相似文献

Tetraploid and hexaploid silkworms, Bombyx mori L. (Lepidoptera Bombycidae) were induced by applying a cold shock to diploid and triploid eggs at the first cleavage stage. Male meiosis in the primary spermatocytes of these silkworms having a different ploidy was observed. In the polyploid cells, chromosome bridges, 2 for the tetraploid and 3 for the hexaploid, occurred between the newly formed daughter nuclei at telophase. Observation in the living spermatocytes showed that tetraploid cells needed a longer time than the diploid ones to complete the first meiotic division. The delay in the cell division may be responsible for the high sterility of the tetraploid males.  相似文献

Twelve taxa of theOxalis tuberosa alliance were analysed and found to share the same basic chromosome number x = 8. The karyotypes are composed by small metacentric and submetacentric chromosomes. Different ploidy levels were found among the taxa: there were 9 diploids, 1 tetraploid, 1 hexaploid and 1 octoploid. The last ploidy level corresponds toO. tuberosa, the only tuber bearing taxon found so far in the alliance. Cytotaxonomic evidence and evolutionary considerations suggest to classify theO. tuberosa alliance in sect.Herrerea.  相似文献

Background

Evolution of sturgeons and paddlefishes (order Acipenseriformes) is inherently connected with polyploidization events which resulted in differentiation of ploidy levels and chromosome numbers of present acipenseriform species. Moreover, allopolyploidization as well as autopolyploidization seems to be an ongoing process in these fishes and individuals with abnormal ploidy levels were occasionally observed within sturgeon populations. Here, we reported occurrence of Siberian sturgeon (Acipenser baerii) male with abnormal ploidy level for this species, accessed its ploidy level and chromosome number and investigate its potential sterility or fertility in comparison with normal individuals of sterlet (A. ruthenus), Russian sturgeon (A. gueldenstaedtii) and Siberian sturgeon (A. baerii).

Results

Acipenser ruthenus possessed 120 chromosomes, exhibiting recent diploidy (2n), A. gueldenstaedtii and A. baerii had ~245 chromosomes representing recent tetraploidy (4n), and A. baerii male with abnormal ploidy level had?~?368 chromosomes, indicating recent hexaploidy (6n). Genealogy assessed from the mtDNA control region did not reveal genome markers of other sturgeon species and this individual was supposed to originate from spontaneous 1.5 fold increment in number of chromosome sets with respect to the number most frequently found in nature for this species. Following hormone stimulation, the spontaneous hexaploid male produced normal sperm with ability for fertilization. Fertilization of A. baerii and A. gueldenstaedtii ova from normal 4n level females with sperm of the hexaploid male produced viable, non-malformed pentaploid (5n) progeny with a ploidy level intermediate to those of the parents.

Conclusion

This study firstly described occurrence of hexaploid individual of A. baerii and confirmed its autopolyploid origin. In addition to that, the first detailed evidence about fertility of spontaneous hexaploid sturgeon was provided. If 1.5 fold increment in number of chromosome sets occurring in diploids, resulted triploids possess odd number of chromosome sets causing their sterility or subfertility due to interference of gametogenesis. In contrast, 1.5 fold increment in number of chromosome sets in naturally tetraploid A. baerii resulted in even number of chromosome sets and therefore in fertility of the hexaploid specimen under study.  相似文献

Summary Fifty-eight somatic hybrid plants, produced both by chemical (11) and electrical fusion (47) of protoplasts of dihaploid Solanum tuberosum and S. brevidens, have been analysed by molecular, cytological and morphological methods. The potentially useful euploid plants constituted 34% of the total, of which 20% were tetraploid and 14% hexaploid; the remainder were aneuploid at the tetraploid, hexaploid and octoploid levels. Analysis of chloroplast DNA showed that 55% of hybrids contained chloroplasts from S. brevidens and 45% from S. tuberosum. Hexaploids, the products of three protoplasts fusing together, were analyzed with specific DNA probes, and this revealed that nuclear genome dosages could be either 21 S. tuberosumS. brevidens, or vice-versa. Chloroplast types of hexaploids were not influenced by nuclear genome dosage, and all six possible combinations of genome dosage and chloroplast types were found amongst tetraploids and hexaploids. To examine the morphology of the hybrid population and its possible relation to the chromosome number and chloroplast DNA type, 18 morphological characteristics were measured on greenhouse-grown plants and analyzed by principal component and canonical variate analyses. Both analyses showed that nuclear ploidy has the most prominent influence on the overall morphology of the hybrids. Differential parental genome expression in the morphology of the hybrids is discussed. These results provide useful data on the range of genetic combinations that can be expected to occur amongst somatic hybrid plants.  相似文献

The present study was an attempt to develop an in vitro colchicine chromosome doubling protocol to restore the fertility of an F1 interspecific hybrid in Lilium. Basal scale segments of Lilium?×?formolongi?×?Oriental hybrid (FO) bulblets were pre-cultured for three durations (6, 15 and 25 days) and soaked in three colchicine concentrations (0.00, 1.25 and 2.50 mM) for 18, 24 and 36 h. To separate mixoploids, three cycles of adventitious bud induction were performed. The ploidy levels of the surviving plantlets were detected by flow cytometry at 30–31 weeks after induction and were confirmed by chromosome counts. The results indicated that the pre-culture duration, colchicine concentration and exposure time all had significant impacts on the tetraploid induction rate. The preferred procedure was to pre-culture the segments for 6 days and then treat them with 1.25 mM colchicine for 24 h. The morphological traits of rosette leaves were significantly different between the tetraploid and diploid plants. The adult tetraploid plants had considerably longer and wider leaves, larger flowers, and delayed flowering time (8 days later) than did diploid plants. Pollen viability tests and backcross trials of FO hybrids demonstrated fertility restoration at the tetraploid level. This protocol provides a feasible method for inducing fertile tetraploid FO hybrids for further breeding.  相似文献

Summary Somatic hybrid plants of various ploidy levels obtained after chemical fusion between two dihaploid clones of potato Solanum tuberosum L. have been analysed by cytological, morphological and molecular methods. The hybrid nature of tetraploid and hexaploid plants and the genome dosage in hexaploid hybrids were confirmed by Giemsa C-banding. Tetraploid and hexaploid hybrids showed numerical as well as structural chromosome mutations. The latter occurred mainly in the nuclear organizing chromosome. The tetraploid hybrids were more vigorous than the dihaploid parents as demonstrated by an increase in height, enlargement of leaves, increase in the number of internodes, restored potential for flowering and increased tuber yield. The grouping of tetraploid somatic hybrids into various classes on the basis of leaf morphology revealed that plants with a full chromosome complement were more uniform than aneuploids. Many hexaploid somatic hybrids were also more vigorous than the dihaploid parents and could be grouped into two different classes on the basis of floral colour and tuber characteristics, the differences being due to their different dosage of parental genomes. Most of the tetraploid somatic hybrids showed pollen development halted at the tetrad stage as one of the parental clones contained a S. Stoloniferum cytoplasm. However, one tetraploid plant produced pollen grains with high viability. The chloroplast genome in the hybrid plants was determined by RFLP analysis. All of the hybrids had a cpDNA pattern identical to one parent, which contained either S. Tuberosum or S. Stoloniferum cpDNA. A slight preference for S. Tuberosum plastids were observed in hybrid plants. No correlation between pollen development and plastid type could be detected.  相似文献

Belonging to the genus Cenchrus with 16–22 species, Cenchrus ciliaris L. (syn. Pennisetum ciliare (L.) Link, buffelgrass) is a perennial, common in warmer regions of both hemispheres, growing as a C4 grass in a wide range of habitats. In the present study we determined chromosome number and nuclear DNA content (2C DNA) for 28 natural populations collected from northern to southern Tunisia. Three ploidy levels were found: one tetraploid population (2n?=?4x?=?36), three pentaploid (2n?=?5x?=?45), and 24 hexaploid populations (2n?=?6x?=?54). The hexaploid chromosome number has already been reported for Tunisian populations of C. ciliaris but tetraploid and pentaploid (2n?=?45) are new for this area. The tetraploid population was found in the semi-arid north; pentaploids were mostly on the northern side of the arid region, while the hexaploids were located mainly in the arid southern Tunisian and Saharan region. 2C DNA values, assessed using flow cytometry, correlated with chromosome counts. Nuclear DNA content ranged from 2C?=?3.03 to 4.61 pg, revealing three ploidy levels corresponding to 4x, 5x, 6x, and mean 2C DNA amounts were of 3.03, 3.7 and 4.48 pg, respectively. Each cytotype produced viable pollen. Flow cytometric seed screening neither proved nor disproved apomixis. The most frequent hexaploid populations seem best adapted to arid conditions in southern Tunisia. The monoploid value, 1Cx, was constant. The existence of pentaploid cytotype suggests hybridization ability between tetraploids and hexaploids. It appears that polyploidization is the major evolutionary mechanism in the speciation of C. ciliaris.  相似文献

The genus Cynodon comprises ten species. The objective of this study was to evaluate the genetic diversity of Korean bermudagrasses at the morphological, cytological and molecular levels. Morphological parameters, the nuclear DNA content and ploidy levels were observed in 43 bermudagrass ecotypes. AFLP markers were evaluated to define the genetic diversity, and chromosome counts were made to confirm the inferred cytotypes. Nuclear DNA contents were in the ranges 1.42-1.56, 1.94-2.19, 2.54, and 2.77-2.85 pg/2C for the triploid, tetraploid, pentaploid, and hexaploid accessions, respectively. The inferred cytotypes were triploid (2n = 3x = 27), tetraploid (2n = 4x = 36), pentaploid (2n = 5x = 45), and hexaploid (2n = 6x = 54), but the majority of the collections were tetraploid (81%). Mitotic chromosome counts verified the corresponding ploidy levels. The fast growing fine-textured ecotypes had lower ploidy levels, while the pentaploids and hexaploids were coarse types. The genetic similarity ranged from 0.42 to 0.94 with an average of 0.64. UPGMA cluster analysis and principle coordinate analysis separated the ecotypes into 6 distinct groups. The genetic similarity suggests natural hybridization between the different cytotypes, which could be useful resources for future breeding and genetic studies.  相似文献

Approximately 70% of the angiosperm species are polyploid, an important phenomenon in the evolution of those plants. But ploidy estimates have often been hindered because of the small size and large number of chromosomes in many tropical groups. Since polyploidy affects cell size, morphometric analyses of pollen grains and stomata have been used to infer ploidy level. Polyploidy is present in many species of the Cerrado, the Neotropical savanna region in Central Brazil, and has been linked to apomixis in some taxa. Eriotheca gracilipes and Eriotheca pubescens are common tree species in this region, and present cytotypes that form reproductive mosaics. Hexaploid individuals (2n = 6x = 276) are polyembryonic and apomictic, while tetraploid and diploid individuals (2n = 2x = 92, 2n = 4x = 184) are sexual and monoembryonic. We tested whether morphometric analysis can be used to estimate ploidy levels in E. gracilipes and E. pubescens individuals. Pollen material from diploid and hexaploid individuals of E. gracilipes, and tetraploid and hexaploid individuals of E. pubescens, were fixed in 50% FAA, and expanded leaves were dried in silica gel. Pollen grains and stomata of at least five individuals from each population were measured. The results demonstrate that all measures were significantly different among cytotypes. Individuals with higher levels of ploidy (hexaploid) all presented measurements that were higher than those with lower levels (diploid and tetraploid). There was no overlap between ploidy levels in each species at 95% confidence interval. Thus, the size of the pollen grains and stomata are effective parameters for analysis of ploidy levels in E. gracilipes and E. pubescens.  相似文献

Prairie cordgrass plants (Spartina pectinata Bosc ex Link) were examined from 61 locations representing the geographic distribution of prairie cordgrass in the U.S. Using flow cytometry, the genome size of 183 individual plants of prairie cordgrass was determined, and the chromosome counts were obtained. Three distinct ploidy levels were observed: tetraploid ( $ \overline x = {1}.{56} {\text{pg}},{2}n = {4} \times = {4}0 $ ), hexaploid ( $ \overline x { = 2}.{33} {\text{pg}},{2}n = {6} \times = {6}0 $ ), and octoploid ( $ \overline {\text{x}} { = 3}.0{6} {\text{pg}},{2}n = {8} \times { = 8}0 $ ). In the sampled areas, the tetraploid populations extended from the East North Central to the New England regions of the U.S., while the octoploid cytotypes were mostly distributed in the West North Central regions. Populations of the tetraploids and octoploids were found in close proximity in the West North Central (Iowa and Kansas) and the West South Central (Oklahoma) regions. The hexaploid cytotype was found in one mixed population (4x?+?6x) occurring in Illinois. No statistically significant intraploidy genome size variation was found in the tetraploid populations, while significant intraploidy genome size variation was found in the octoploid populations. This study precisely defined the geographic distribution of cytotypes in prairie cordgrass throughout the different regions of the U.S. These results provide critical genome size and ploidy distribution information needed to design efficient breeding schemes for high yielding cultivars of prairie cordgrass with local adaption.  相似文献

During recent years, hazel cell suspension culture has been significantly considered as a new important source of paclitaxel. Artificial polyploidy alters different characters in plants which results in amplifying the secondary metabolites in medicinal plants. In this paper, the effects of tetraploidy induction on paclitaxel content and gene expression in hazel cell suspension culture were investigated. Various concentrations of colchicine and duration of exposure in solid and liquid media were examined and the ploidy level of cells was determined using flow cytometric analysis. The tetraploid cells were obtained from 0.2% colchicine in the exposure time of 5 and 6 days in solid medium and 0.3% colchicine in the exposure time of 3 and 4 days in liquid medium. Tetraploid cells were employed to prepare cell suspension. 3 µM of phenylalanine and 0.05 mM of vanadyl sulfate were added to both tetraploid and diploid (control) suspension to elicit paclitaxel induction. High performance liquid chromatography analysis demonstrated that the tetraploid cell suspensions produced paclitaxel of about [9.88 µg g?1 (DW)] 1.7-fold compared with diploid cells [5.74 µg g?1 (DW)]. The application of phenylalanine and vanadyl sulfate increased the concentration of paclitaxel in both diploid and tetraploid cells. Moreover, qRT-PCR analysis showed that the expression of GGPPS gene was significantly increased and PAL gene expression was altered after tetraploidization.  相似文献

Aim Newly formed polyploids experience problems of establishment and spread similar to those faced by newly introduced alien species. To understand the significance of polyploidy in biological invasions, we mapped the distribution of ploidy levels in Solidago gigantea Aiton in its native range in North America, and in Europe and East Asia where it is invasive. Location North America, Europe and East Asia. Methods Flow cytometry was used to measure ploidy levels in a total of 834 plants from 149 locations. Together with data from contributors and a literature review, ploidy‐level data were assembled for 336 locations. Cytogeographical maps from North America and Europe were prepared, incorporating new and previously published ploidy‐level data. Results In the native range, diploid, tetraploid and hexaploid plants were found, and also one triploid and one pentaploid plant (2n = 3x and 2n = 5x, respectively, each being new reports for this species). There was a high degree of geographical separation among the ploidy levels, and populations with mixed ploidy were rare. However, four zones were identified where plants of different ploidy could come into contact. In Europe and East Asia, only tetraploid plants were found. Main conclusions The geographical pattern in North America suggests that the ploidy levels are ecologically differentiated, although further investigations are needed to identify the nature of these differences. Alien populations appear to be exclusively tetraploid, but it is not clear whether this is because tetraploids were selectively introduced or because diploids were unsuccessful. In any case, comparisons between native and introduced populations need to account for ploidy level.  相似文献

Summary Transmission of extra genome chromosomes by three Vaccinium ashei (2n=6x=72)/V. corymbosum (2n=4x=48) pentaploid hybrids backcrossed to the hexaploid species V. ashei was examined. Chromosome numbers were determined for 36 and 31 progeny representing 5x × 6x and 6x × 5x type crosses, respectively. Chromosome numbers ranged from hypopentaploid (2n=4x+11=59) to hexaploid with means of 2n=66.2 for 5x × 6x progeny and 2n=68.0 for 6x × 5x progeny, representing overall extra genome chromosome gains of 3.3% and 33.3%, respectively. Extra chromosome number distributions for both the 5x × 6x and x × 5x progeny deviated significantly from the theoretical distribution assuming random chromosome transmission and were also found to be heterogeneous. The 2n=5x+9=69 class predominated in 6x × 5x progeny, while a predominate class was lacking in the 5x × 6x progeny. Higher than expected frequencies of plants with chromosome numbers near the pentaploid and hexaploid levels were found in the 5x × 6x progeny, whereas the frequency was only greater at the hexaploid number in 6x × 5x progeny. Present and previous results (Vorsa et al. 1986) indicate that extra genome chromosome transmission in oddploids can be influenced by selection at both gametophytic (pollen) and post-zygotic stages. However, post-zygotic selection may involve two different mechanisms acting concurrently: 1) chromosome imbalance due to aneuploidy and/or 2) endosperm imbalance referring to maternal: paternal genome ratios deviating from 21. Such a mechanism could result in differential transmission rates of extra genome chromosomes in oddploids when crosses are made to differing ploidy levels, and to reciprocal differences as well.  相似文献

Osmotin is a major protein which accumulates in tobacco cells (Nicotiana tabacum L. var Wisconsin 38) adapted to low water potentials. Quantitation of osmotin levels by immunoblots indicated that cells adapted to 428 millimolar NaCl contained 4 to 30 times the level of osmotin found in unadapted cells, depending on the stage of growth. Unadapted cells accumulated low levels of osmotin with apparent isoelectric points, (pl) of 7.8 and >8.2. Upon transfer of NaCl-adapted cells to medium without NaCl and subsequent growth for many cell generations, the amount of osmotin declined gradually to a level intermediate between that found in adapted and unadapted cells. NaCl-adapted cells grown in the absence of NaCl accumulated both pl forms; however, the form accumulated by cells adapted to NaCl (pl > 8.2) was most abundant. Adapted cells grown in the absence of NaCl exhibited absolute growth rates and NaCl tolerance levels which were intermediate to those of NaCl-adapted and unadapted cells. The association between osmotin accumulation and stable NaCl tolerance indicates that cells with a stable genetic change affecting the accumulation of osmotin are selected during prolonged exposure to high levels of NaCl. This stable alteration in gene expression probably affects salt tolerance.  相似文献

Although triploid Populus varieties have been used widely in timber and pulpwood production, the performance of economic traits in Populus with higher ploidy levels remains unknown due to a lack of germplasms with higher ploidy. In this study, we first successfully induced hexaploids in Populus by treating triploid leaf explants with colchicine in vitro. In total, 32 hexaploids were produced. The frequency of hexaploids was significantly affected by the interaction between colchicine concentration and exposure time. The highest hexaploid induction efficiency was 16.89% (±?2.26), which was achieved by treating explants with 0.04% colchicine for 7 days. Compared to triploids, hexaploids had thinner epidermal hair, larger stomata and protoplasts, and fewer chloroplasts, indicating that significant phenotypic changes accompanied an increase in ploidy level. These hexaploids are valuable for investigating the performance of economic traits in Populus with higher ploidy levels and have the potential to be used as parents to produce new tetraploid and pentaploid germplasms in Populus breeding programs.  相似文献

The existence of neopolyploidy in prairie cordgrass (Spartina pectinata Link) has been documented. The neohexaploid was discovered coexisting with tetraploids in central Illinois, and has been reported to exhibit competitiveness in the natural environment. It is hypothesized that the natural tetraploid cytotype produced the hexaploid cytotype via production of unreduced gametes. Meiosis I chromosome pairing was observed in tetraploid (2n?=?4x?=?40), hexaploid (2n?=?6x?=?60), and octoploid (2n?=?8x?=?80) accessions and the percentage of meiotic abnormality was determined. Significant differences in meiotic abnormality exist between tetraploid, hexaploid, and octoploid cytotypes. An elevated incidence of abnormal, predominantly trivalent pairing in the neohexaploid suggests that it may possess homologous chromosomes in sets of three, in contrast to the tetraploid and octoploid cytotypes, which likely possess homologous chromosomes in sets of two. Abnormal chromosome pairing in the hexaploid may result in unequal allocation of chromosomes to daughter cells during later stages of meiosis. Chromosome pairing patterns in tetraploid, hexaploid, and octoploid cytotypes indicate genome compositions of AABB, AAABBB, and AABBA′A′B′B′, respectively.  相似文献
