Every week, for 20 weeks, the growth of naturally occurring grain storage fungi on wheat infested with the three commonest British grain storage mites,Acarus siro, Glycyphagus destructor andTyrophagus longior, was compared with that on uninfested wheat.The number of colonies of theAspergillus glaucus group per gram were always less on grain infested with mites than on uninfested grain.Penicillium spp. were also less numerous on grain which was infested withA. siro but did not appear to be affected by the other mites. In contrast, two fungi which are pathogenic to mites,Aspergillus restrictus andWallemia sebi, were more abundant in the presence of certain mites. The former was associated withG. destructor, the latter withG. destructor andA. siro.The three species of mites either feed on theA. glaucus group andPenicillium spp., or inhibit them by an unknown secretion. Pathogenic fungi are probably avoided. Mites are therefore an important variable in studies on fungal growth during grain drying and storage.  相似文献

Egg production and longevity of isolated pairs ofAcarus siro L., when fed on mycelial pellets of selected fungi from shake cultures, were compared with those on a control diet of wheatgerm and brewer's yeast. The mites were always less productive on the fungal diet. They did best on the yeastHansenula anomola and worst on the field fungusAlternaria alternata. There was very little difference between egg production on wheatgerm alone and that on the control diet of wheatgerm and yeast. On the control diet, males lived longer than females. Fungal diets usually shortened male lives, while in two-thirds of the cases female lifespans were unaffected.  相似文献

Twenty bisected grains of wheat infested with five pairs of the three commonest British grain-storage mites,Acarus siro L., Glycyphagus destructor (Shrank) andTyrophagus longior (Gervais), were examined every week for 20 weeks. Mite populations, the resulting damage to germ and endosperm, and visible fungal growth were observed at 20°C and relative humidities (r.h.) of 90% and 75%.At 90% r.h.,A. siro populations reached nearly 14000 per test-tube before slowly dropping to 5000. The mites ate the germ before the endosperm, leaving an impenetrable layer of crushed endosperm cells between these regions. TheG. destructor population reached only 800 before declining to 300; these mites ate over 75% of the germ and small amounts of endosperm.Tyrophagus longior populations rose to 2200 mites before crashing at week 12 to the initial population level; these mites ate over 75% of the germ and small amounts of endosperm.At 75% r.h., bothA. siro andT. longior populations were lower than at the higher r.h., peaking at 3000 and 1000 respectively and decreasing to 500 and 600 mites respectively.Glycyphagus destructor did markedly better than at 90% r.h., reaching 1500 before falling to 400. The damage at this humidity was slower to occur but was similar to that at 90% r.h. at the end of 20 weeks.At both humidities visible fungus was always less abundant on infested grain that uninfested grain.  相似文献

The identification of allergy‐causing mites is conventionally based on morphological characters. However, molecular taxonomy using ribosomal DNA (rDNA) may be particularly useful in the analysis of mite cultures and purified mite fractions in the production of allergenic extracts. Full‐length internal transcribed spacers (ITS1 and ITS2) were obtained from Dermatophagoides farinae, Dermatophagoides pteronyssinus, Dermatophagoides microceras and Euroglyphus maynei (Astigmata: Pyroglyphidae), Glycyphagus domesticus and Lepidoglyphus destructor (Astigmata: Glycyphagidae), Tyrophagus fanetzhangorum, Tyrophagus putrescentiae, Tyrophagus longior, Tyrophagus neiswanderi, Acarus farris and Acarus siro (Astigmata: Acaridae), and Blomia tropicalis (Astigmata: Echymopodidae), using mite‐specific primers. Polymerase chain reaction (PCR) products were digested with HpaII and RsaI restriction enzymes in order to produce species‐specific PCR restricted fragment length polymorphism (RFLP) profiles. A semi‐nested re‐amplification step was introduced before the RFLP in order to apply the method to environmental samples. Results demonstrate that rDNA sequences can be used for the unambiguous identification of mite species. The PCR–RFLP system allows the identification of species in purified mite fractions when the availability of intact adult mite bodies for morphological identification is limited. This reliable and straightforward PCR–RFLP system and the rDNA sequences obtained can be of use in the identification of allergy‐causing mite species.  相似文献

Traps have been used extensively to provide early warning of hidden pest infestations. To date, however, there is only one type of trap on the market in the U.K. for storage mites, namely the BT mite trap, or monitor. Laboratory studies have shown that under the test conditions (20 °C, 65% RH) the BT trap is effective at detecting mites for at least 10 days for all three species tested: Lepidoglyphus destructor, Tyrophagus longior and Acarus siro. Further tests showed that all three species reached a trap at a distance of approximately 80 cm in a 24 h period. In experiments using 100 mites of each species, and regardless of either temperature (15 or 20 °C) or relative humidity (65 or 80% RH), the most abundant species in the traps was T. longior, followed by A. siro then L. destructor. Trap catches were highest at 20 °C and 65% RH. Temperature had a greater effect on mite numbers than humidity. Tests using different densities of each mite species showed that the number of L. destructor found in/on the trap was significantly reduced when either of the other two species was dominant. Aeration is a promising alternative to the use of pesticides for the control of storage insects by cooling bulk grain, but its effectiveness against mite pests is neither fully understood nor optimised. For this reason, the productivity of three species of storage mites, Acarus siro, Lepidoglyphus destructor and Tyrophagus longior, was studied in a laboratory-based experiment at four combinations of temperature and humidity (10°C and 70% RH, 10°C and 80% RH, 20°C and 70% RH, 20°C and 80% RH) with and without an airflow (at 10 m3/h/tonne, equalling 2.5 l/s/tonne, in tubes containing 15 g of grain). This is the first time that a study has examined the three principal components of aeration separately from each other. The effect of these factors was different for each species. For A. siro, temperature was the most important factor, while airflow and humidity were of similar but lesser importance. For T. longior, temperature was more important than humidity, while the reverse was true for L. destructor. For these two species, airflow was the least important factor. The airflow decreased the productivity of L. destructor and T. longior but increased the productivity of A. siro. This increase in productivity confirms that, in practice, prevention of mite infestations, in particular A. siro, will require storage of grain at low temperature, relative humidity and moisture content. Fungi associated with two mite species,Acotyledon krameri andTyrophagus putrescentiœ, and their habitats were studied using 1% glucose-Czapek's and potato-dextrose agar media at 28±2°C. A total of 54 species and one variety belonging to 25 genera were recovered from different habitats of mite species; 36 species and one variety belonging to 18 genera were associated with mite bodies. Nearly most fungal species isolated from mite bodies were also encountered in their habitats. The most common species wereAspergillus flavus, A. fumigatus, A. niger, Mucor racemosus, Nectria hœmatococca. A. terreus, Scopulariopsis brevicaulis andTrichurus spiralis were isolated only from mite bodies whereasFusarium oxysporum, Penicillium chrysogenum, P. corylophilum, P. funiculosum andRhizopus stolonifer were recovered from the mite habitats.A. krameri individuals survived well onCunninghamella elegans, F. oxysporum andM. racemosus cultured on both type of media, whereasCylindrocarpon destructans was best forT. putrescentiœ survival.  相似文献

In storage facilities one can find grain either in stored grain mass or in grain residues in the store corners or machinery. Although it is claimed that grain residues are serious pest reservoirs since they harbor numbers of stored product arthropods and are connected via continuous emigration with grain mass, the documentation for this is not convincing. Therefore in 78 selected grain stores, we simultaneously sampled the grain mass and residues in order to compare concurrent mite communities in these two different habitats. We found 30 species in about 614 000 individuals in residues and 23 species in about 20 000 individuals in grain mass. Canonical correspondence analysis (CCA) of transformed abundance data showed differences in the communities of mites in grain mass and residues: (i) species associated to grain residues (e.g. Tyrophagus longior, Tydeus interruptus, Acarus farris and Cheyletus eruditus) and (ii) species associated to both grain mass and grain residues (e.g. Tarsonemus granarius, Acarus siro, Tyrophagus putrescentiae, Lepidoglyphus destructor and Cheyletus malaccensis). Although the residue samples had more mites and higher species diversity than the stored grain mass, no correlation in mite abundance and species numbers between samples from grain residues and grain mass was found, thereby indicating low connectivity of these two habitats.  相似文献

Both house dust and house dust mitesDermatophagoides pteronyssinus contained a wider range of fungi than laboratory mite cultures. In total, nine species of fungi were isolated fromD. pteronyssinus in house dust, and these included three xerophilic species (Eurotium amstelodami, Aspergillus penicillioides andWallemia sebi) commonly found in laboratory cultures ofD. pteronyssinus. It is concluded that mites do interact with a similar range of fungi in natural dust and in laboratory culture, but that the diversity of fungal species in the laboratory is reduced and the density of individual fungal species in culture exceeds that of house dust. In a second experiment, dust samples were incubated at room temperature with 75% relative humidity. The diversity of fungi invariably declined from up to 13 genera to the few species recorded in laboratory culture. This suggests that the dominance of xerophilic fungi in laboratory mite rearings is mediated primarily by low relative humidity, and the exclusion of air-borne spores.  相似文献

The adaptation of nine species of mites that infest stored products for starch utilization was tested by (1) enzymatic analysis using feces and whole mite extracts, (2) biotests, and (3) inhibition experiments. Acarus siro, Aleuroglyphus ovatus, and Tyroborus lini were associated with the starch‐type substrates and maltose, with higher enzymatic activities observed in whole mite extracts. Lepidoglyphus destructor was associated with the same substrates but had higher activities in feces. Dermatophagoides farinae, Chortoglyphus arcuatus, and Caloglyphus redickorzevi were associated with sucrose. Tyrophagus putrescentiae and Carpoglyphus lactis had low or intermediate enzymatic activity on the tested substrates. Biotests on starch additive diets showed accelerated growth of species associated with the starch‐type substrates. The inhibitor acarbose suppressed starch hydrolysis and growth of the mites. We suggest that the species with higher starch hydrolytic activity in feces were more tolerant to acarbose, and α‐amylase and α‐glucosidase of synanthropic mites are suitable targets for inhibitor‐based strategies of mite control. © 2009 Wiley Periodicals, Inc.  相似文献

Fungal feeding soil invertebrates feed on a wide spectrum of fungal species suggesting that mixed diets increase fitness. We investigated relationships between food preferences for seven saprophytic fungal species/forms and fitness parameters (mortality, growth, time to reproduction, reproduction, egg size) in two Collembola species, Folsomia candida and Protaphorura armata. The fungal species/forms studied included the wild type and a melanin-deficient form of Aspergillus fumigatus to investigate the role of melanin in collembolan nutrition. Also, three mixed diets consisting of a preferred fungal species (Cladosporium cladosporioides) and species of intermediate or low food quality were investigated. Both Collembola species preferred similar fungal species/forms as food. Food preference generally matched fitness parameters, i.e. growth and reproduction of Collembola was at a maximum when feeding on preferred fungi. This was not the case for A. fumigatus. The wild type and the melanin-deficient form ranked among the least preferred fungi. Growth and reproduction of Collembola were low when feeding on the wild type but high when feeding on the melanin-deficient form indicating that the Collembola misjudged the food quality of the latter in the preference tests. The results show for the first time that genes driving melanin syntheses (pksP) strongly affect the food quality of fungi for fungal feeding invertebrates. Feeding on mixed diets generally increased growth and reproduction of Collembola except when the diets included toxic species (Penicillium sp.). The results support the nutrient balance hypothesis and also show that the detection of toxic species in the diet is important. They indicate that the widespread generalist feeding mode of Collembola maximizes fitness if toxic fungal species are avoided. The fitness parameters growth, reproduction and time until onset of reproduction were correlated closely but egg volume, which also varied with fungal diet, correlated poorly with the other fitness parameters. Variation in egg size with fungal diet shows that the diet of Collembola may have transgenerational effects.  相似文献

Summary Lignin peroxidases produced byPhanerochaete chrysosporium have several important potential industrial applications based on their ability to degrade lignin and lignin-like compounds. A stirred tank reactor system for the production of lignin peroxidases is described here. Included in this study is an examination of the mechanics of pellet biocatalyst formation and the optimization of an acetate buffered medium. Higher levels of lignin peroxidase were obtained with acetate buffer compared to the other buffer systems tested. Concentrations of 0.05% (w/v) Tween 80 and 0.4 mM veratryl alcohol gave optimal lignin peroxidase activity in acetate buffered medium. In shake flask cultures, mycelial fragments in the inoculum aggregated into pellets during the first eight hours of incubation and thereafter increased in size through the eighth day. The agitation rate in shake flask cultures affected pellet size, the number of pellets formed, and lignin peroxidase activity. Transfer of fungal pellets from shake flask culture to a continuously oxygenated baffled stirred tank reactor (STR) resulted in production of high lignin peroxidase titres comparable to those of shake flask cultures when the agitation rate, oxygen dispersion and foaming were closely controlled.  相似文献

Aqueous extracts of fungal biomass ofAspergillus niger, A. flavus, Penicillium notatum, Fusarium oxysporum and the filtrates of their culture media were analysed for elicitation capability to enhance anthocyanin production in callus cultures ofDaucus carota. The mycelial extract ofA. flavus at the 2.5% level gave maximum elicitation, which resulted in a two-fold increase in anthocyanin with maximal productivity of 23.7% on a dry weight basis. Whereas the media filtrates ofA. flavus induced a 1.25-fold increase in anthocyanin production with a yield of 20.6% on a dry weight basis,P. notatum andF. oxysporum were not as effective asA. flavus orA. niger. The contact time and the concentration required for maximal elicitation of anthocyanin differed with the type of elicitor used. Qualitative analysis of anthocyanins revealed the presence of cyanidin glycosides in control and elicited cultures.  相似文献

Mites as Selective Fungal Carriers in Stored Grain Habitats   总被引:2,自引:0,他引:2  
Mites are well documented as vectors of micromycetes in stored products. Since their vectoring capacity is low due to their small size, they can be serious vectors only where there is selective transfer of a high load of specific fungal species. Therefore the aim of our work was to find out whether the transfer of fungi is selective. Four kinds of stored seeds (wheat, poppy, lettuce, mustard) infested by storage mites were subjected to mycological analysis. We compared the spectrum of micromycete species isolated from different species of mites (Acarus siro, Lepidoglyphus destructor, Tyrophagus putrescentiae, Caloglyphus rhizoglyphoides and Cheyletus malaccensis) and various kinds of stored seeds. Fungi were separately isolated from (a) the surface of mites, (b) the mites' digestive tract (= faeces), and (c) stored seeds and were then cultivated and determined. The fungal transport via mites is selective. This conclusion is supported by (i) lower numbers of isolated fungal species from mites than from seeds; (ii) lower Shannon–Weaver diversity index in the fungal communities isolated from mites than from seeds; (iii) significant effect of mites/seeds as environmental variables on fungal presence in a redundancy analysis (RDA); (iv) differences in composition of isolated fungi between mite species shown by RDA. The results of our work support the hypothesis that mite–fungal interactions are dependent on mite species. The fungi attractive to mites seem to be dispersed more than others. The selectivity of fungal transport via mites enhances their pest importance.  相似文献

Bacillus thuringiensis (Bt) toxins present a potential for control of pest mites. Information concerning the effect of Bt and its possible application to the biocontrol of synathropic mites is rare. The toxic effect of Bacillus thuringiensis var. tenebrionis producing Cry3A toxin was tested on the mites Acarus siro L., Tyrophagus putrescentiae (Schrank), Dermatophagoides farinae Hughes, and Lepidoglyphus destructor (Schrank) via feeding tests. Fifty mites were reared on Bt additive diets in concentrations that ranged from 0 to 100 mg g−1 under optimal conditions for their development. After 21 days, the mites were counted and the final populations were analyzed using a polynomial regression model. The Bt diet suppressed population growth of the four mite species. The fitted doses of Bt for 50% suppression of population growth were diets ranging from 25 to 38 mg g−1. There were no remarkable differences among species. Possible applications of Bt for the control of synanthropic mites are discussed.  相似文献

Background

Bacteria are associated with the gut, fat bodies and reproductive organs of stored product mites (Acari: Astigmata). The mites are pests due to the production of allergens. Addition of antibiotics to diets can help to characterize the association between mites and bacteria.

Methodology and Principal Findings

Ampicillin, neomycin and streptomycin were added to the diets of mites and the effects on mite population growth (Acarus siro, Lepidoglyphus destructor and Tyrophagus putrescentiae) and associated bacterial community structure were assessed. Mites were treated by antibiotic supplementation (1 mgg−1 of diet) for 21 days and numbers of mites and bacterial communities were analyzed and compared to the untreated control. Bacterial quantities, determined by real-time PCR, significantly decreased in antibiotic treated specimens from 5 to 30 times in A. siro and T. putrescentiae, while no decline was observed in L. destructor. Streptomycin treatment eliminated Bartonella-like bacteria in the both A. siro and T. putrescentiae and Cardinium in T. putrescentiae. Solitalea-like bacteria proportion increased in the communities of neomycin and streptomycin treated A. siro specimens. Kocuria proportion increased in the bacterial communities of ampicillin and streptomycin treated A. siro and neomycin and streptomycin treated L. destructor.

Conclusions/Significance

The work demonstrated the changes of mite associated bacterial community under antibiotic pressure in pests of medical importance. Pre-treatment of mites by 1 mgg−1 antibiotic diets improved mite fitness as indicated accelerated population growth of A. siro pretreated streptomycin and neomycin and L. destructor pretreated by neomycin. All tested antibiotics supplemented to diets caused the decrease of mite growth rate in comparison to the control diet.  相似文献

Laboratory studies revealed that the acarid mite,Tyrophagus casei Oudemans is an adequate diet for development and reproduction of the stigmaeid mite,Agistemus exsertus Gonzalez. A combination of pollen grains of castor beanRicinus communis L. andT. casei is a sufficient plant-animal diet for the development and reproduction of the phytoseiid mite,Amblyseius gossipi Elbadry. Among 39 fungal species and one variety belonging to 16 genera isolated from 116 healthy tomato fruits collected from markets in Assiut, Egypt, during 1994,Aspergillus niger was found to be the most prevalent, being isolated from 84.6% of the samples. Of 11 species recovered from 156 diseased tomatoes,Alternaria alternata, Rhizopus stolonifer andA. niger were the most common and isolated from 52.7%, 35.9% and 25.0% of the samples, respectively. Experiments for comparison of the effects of medium containing tomato juice with synthetic medium on the mycelial growth of nine fungal species indicated that, the tomato juice medium was more suitable for growth of all fungal species. The effect of five commercial fungicides and sodium hypochlorite when applied as post-harvest dips after inoculation was studied in laboratory againstA. alternata, A. niger andR. stolonifer. At 10 and 100 µg ml–1, neither of the fungicide caused a noticeable inhibition ofAlternaria rot. At 1000 µg ml–1 benlate, rovral and sumisclex completely preventedAlternaria andAspergillus rot, whereas cuprosan and ridomil were ineffective against rotting caused byA. niger. Rhizopus rot development was inhibited by over 50% with one treatment only (rovral at 1000 µg ml–1). Sodium hypochlorite has good curative properties against fruit rots especially those caused byA. alternata andA. niger.  相似文献

Adult female mites of the genusTrochometridium Cross possess a pair of internal sacs between the bases of legs III and IV, which are adapted for carrying spores, apparently ascospores, of an undetermined fungus. A three-way symbiotic relationship exists between the mite, the fungus, and various bees (and possibly other holometabolous insects) which nest in relatively dry alkaline soils. The mite transports spores of the fungus to suitable sites for germination — cells of ground-nesting bees containing a bee egg or young larva which dies as a result of development of the fungus and mite. The mite may also stimulate mycelial growth, possibly by killing the young bee or by secreting a substance when feeding. The fungus provides the preferred mycelial substrate on which the mite feeds and undergoes its life cycle. This mutualistic association between the mite and fungus is at the expense of the bee, which transports the other two entities to favorable sites for their development — its newly made and provisioned cells. The sporothecae and specialized association ofTrochometridium mites with a fungal pathogen or saprophyte of another host organism are compared with those ofSiteroptes mites, which are considerably better understood.  相似文献

In this paper the effects of environmental conditions on the mycelial growth of Ganoderma lucidum were investigated in shake flask cultures. The optimal temperature and pH were found to be around 30–35?°C and 4, respectively, in a glucose-ammonium chloride medium. The maximum mycelial concentration reached to around 350?mg/100?ml. The formation of mycelial pellets and their ultra structure was demonstrated to be greatly affected by cultivating conditions. Increasing surface aeration would be beneficial for mycelial growth. However, too high rotating speed in shake cultures had a detrimental effect on the formation of mycelial pellets and the optimum was found to be 100?rpm.  相似文献
