Methadone exhibits an antagonistic effect toward steroidogenesis which lies prior to progesterone in the biosynthetic pathway in isolated rat adrenal cells. Levels of adenosine cyclic 3′–5′ monophosphate are depressed in a dose dependent fashion in ACTH stimulated cells as is steroidogenesis in cells stimulated with N6O2-dibutyryl adenosine cyclic 3′–5′ monophosphate. Stimulation produced by the ACTH analog, O-nitrophenyl sulfenyl ACTH, is also inhibited by methadone. The participation of adenosine cyclic 3′–5′ monophosphate as an obligatory messenger in ACTH stimulated steroidogenesis is discussed with respect to the pharmacological properties of methadone in this system.  相似文献

In order to investigate the mechanism of glucose repression of the N-acetylglucosamine metabolic enzymes in Candidaalbicans, an obligatory aerobic yeast, the activities of the following inducible enzymes were assayed: the N-acetylglucosamine uptake, N-acetylglucosamine kinase and glucosamine-6-phosphate deaminase. In the presence of glucose or other sugars e.g. succinate and glycerol, synthesis of these enzymes took place at a normal rate, suggesting that the hexose produces no catabolite repression in this organism. On the contrary, strong inhibition by glucose was observed on the activities of N-acetylglucosamine uptake and deaminase in N-acetylglucosamine-grown cells of Saccharomycescerevisiae, a facultative aerobe. From the results, it is concluded that “glucose effect” or catabolite repression is absent in Candidaalbicans, a pathogenic strain of yeast.  相似文献

Alkali-inactivated pig liver pyruvate kinase, type L, and a cyanogen bromide fragment from the same enzyme were shown to be phosphorylated by (32P)ATP and cyclic 3′,5′-AMP-stimulated protein kinase. In both cases the rate of phosphorylation was higher than with the native enzyme. Pyruvate kinases types A and M were not phosphorylated under the same conditions. From the 32P-labelled cyanogen bromide fragment (32P)phosphorylserine was isolated. The electrophoretic pattern of (32P)phosphopeptides obtained on partial acid hydrolysis of the fragment indicated that the phosphorylated site of the fragment was identical with that of the native pyruvate kinase.  相似文献

Solid-state microelectrodes for measuring intracellular Cl? activity (aiCl) were made by sealing the tips of tapered glass capillaries (tip diameter 0.3 μm), coating them under vacuum with a 0.2–0.3 μm thick layer of spectroscopic grade silver, and sealing them (except for the terminal 2–5 μm of the tip) inside tapered glass shields. 106 microelectrodes had an average slope of 55.0 ± 0.6 mV (S.E.) per decade change in αCl. Tip resistance was (77.1 ± 3.1 × 109ω (n=30). Electrode response was rapid (10–20 s), was unaffected by HCO3?, H2PO42? or protein, and remained essentially unchanged over a 24-h period. αiCl in frog sartorius muscle fibers and epithelial cells of bullfrog small intestine was measured in vitro. In both tissues, αiCl significantly exceeded the value corresponding to equilibrium distribution of Cl? across the cell membrane.  相似文献

The addition of cyclic AMP to the shaking medium of cells disaggregated from pseudoplasmodia of Dictyostelium discoideum suppressed the accumulation of cell-bound phosphodiesterase which normally occurs (1) after disaggregation. The suppression was not secondarily brought about by its possible inhibitory effect of cyclic AMP on protein synthesis or by its stimulating effect on the release of the enzyme into the medium. The effect was reversible and specific to cyclic AMP. On the other hand, the inhibitory effect of cyclic AMP on the disaggregation-induced inactivation of UDP-galactose transferase was not apparent in the initial period, but thereafter it slowed down the decrease in the enzyme activity. These results indicate that exogenous cyclic AMP mimics at least in part the regulatory effects of cell-to-cell contact on certain enzymes.  相似文献

Partially purified nucleotide fraction of moss containing [14C]-labelled putative adenosine 3′, 5′ -cyclic monophosphate (cAMP) and marker authentic [3H] -cAMP was characterized by chemical deamination and also by the enzymatic hydrolysis with beef heart cyclic nucleotide phosphodiesterase. A significant conversion of marker authentic [3H] -cAMP into [3H] -inosine 3′, 5′ -cyclic monophosphate (cIMP) and [3H] -5′ adenosine monophosphate was observed by respective treatments. In contrast, the [14C] -labelled putative cAMP from control and theophylline-treated moss tissue was insensitive to chemical deamination and enzymatic hydrolysis. Apparently, the [14C] -labelled product which comigrates with authentic [3H] -cAMP does not represent true cAMP. Both the methods employed for characterization of the labelled putative cAMP were sensitive enough to detect picomole quantities of authentic [3H] -cAMP. Lack of detectability of prelabelled [14C] -cAMP in our preparations implies that the tissue may contain authentic cyclic AMP below the picomole levels. Thus, the attributed physiological role to adenosine 3′, 5′ -cyclic monophosphate in moss tissue appears somewhat skeptical.  相似文献

The accumulation of 45Ca2+ by intact mouse mastocytoma cells was examined before and after treatment of the cells with N6,O2′-dibutyryladenosine 3′,5′, cyclic monophosphate and theophylline to inhibit growth. In the presence of phosphate either glycolysis, respiration or ATP supported 45Ca2+ uptake by the cells and in each case the accumulated 45Ca2+ appeared to be retained by mitochondria. Inhibition of growth by drug treatment for 20h increased subsequent 45Ca2+ accumulation when cells were incubated with 45CaCl2, succinate and phosphate. Since prior drug treatment did not increase 45Ca2+ accumulation with glucose, ATP or malate the drugs appeared to increase 45Ca2+ accumulation by affecting succinate metabolism.  相似文献

To investigate the mechanism of serine protease stimulation of rat ovarian adenylate cyclase, a variety of synthetic protease inhibitors were used. These inhibitors blocked trypsin, chymotrypsin and hCG stimulation of adenylate cyclase in nearly the same manner. The inhibition of hormone stimulated adnylate cyclase could not be explained by a loss of [125I]hCG binding. Cholera toxin and epinephrine stimulation of adenylate cyclase were similarly inhibited, whereas basal and fluoride-stimulated activities were only affected by higher doses of the inhibitors. The results suggest that adenylate cyclase in the ovary may be regulated by membrane protease activity.  相似文献

Crude preparations of cyclic adenosine 3′, 5′-monophosphate phosphodiesterase were activated 1.5 to 2 fold by incubation with ATP, Mg2+ and cyclic AMP in a reaction which was both, time and temperature dependent. Cyclic AMP phosphodiesterase remained in an activated state upon filtration of the enzymatic preparation through Sephadex G-25 and ion-exchange chromatography. Activation of the enzyme in the presence of [γ 32P]ATP resulted in a significant amount of [32P] protein-bound radioactivity. Reversible deactivation of cyclic AMP phosphodiesterase was enhanced by Mg2+ and was accompanied by the release of [32P] protein bound radioactivity. The evidence is consistent with a mechanism for controlling cyclic AMP phosphodiesterase through phosphorylation-dephosphorylation sequence.  相似文献

A procedure is described which unequivocally demonstrates the presence of adenosine 3′:5′-cyclic monophosphate in Phaseolus vulgaris. Its concentration was determined spectrophotometrically at 2·6–9·2 nmol g?1 of tissue (dry wt) for 6-day-old seedlings and about one-tenth of this in 13-day-old plants.  相似文献

The sarcolemmal membranes isolated from rat skeletal muscle are capable of incorporating 32P from [γ?32P]ATP. The membrane protein phosphorylation requires Mg2+. Cyclic AMP, cyclic GMP and their dibutyrul derivatives showed no marked effect on sarcolemmal phosphorylation.The Mg2+-dependent 32P labeling was significantly enhanced by Na+. The rate of Na+ -stimulated 32P incorporation was quite rapid reaching steady state levels within 5 s at 0 °C. K+ reduced the Na+ -stimulated 32P-incorporation but enhanced the 32Pi release. This inhibitory effect of K+ on Na+ -stimulated 32P incorporation was prevented by the cardiac glycoside, ouabain.The Na+ -dependent 32P labeling showed substrate dependency and the Na+ site was saturable. The apparent Km for ATP was 2 · 10?5 M. The optimum pH for 32P labeling was between 7 and 8.Na+ -dependent membrane phosphorylation showed a direct relationship with the (Na+ + K+ATPase activity. The high turnover rate of 32P intermediate (12 000 min ?1) suggested its functional significance in the overall transport ATPase reaction sequence.The predominate portion (> 90%) of the phosphorylated membrane complex was sensitive to acidified hydroxylamine and to alkaline pH suggesting an acylphosphate nature of the phosphoprotein.Sodium dodecyl sulfate-polyacrylamide gel electrophoresis revealed that 32P incorporation occurred predominately into a 108 000 dalton subunit which is a major protein component of sarcolemmal membranes. A very low level of 32P incorporation was also observed into a 25 000 dalton subunit and Ca2+ slightly enhanced the phosphorylation of this component.The size (Mr 108 000) and some properties of the sarcolemmal phosphoprotein are closely similar to other (Na+ + K+ATPase preparations reported so far.  相似文献

The validity of using the binding-protein method for determining cyclic AMP in purified and partially purified extracts of Phaseolus tissues has been examined and confirmed. Measurement of cyclic AMP concentration by binding-protein gave similar results to those obtained by direct spectrophotometry of purified extracts. A cyclic AMP binding-protein and adenylate cyclase were demonstrated in Phaseolus extracts. Isolated intact chloroplasts were shown to possess adenylate cyclase activity but persistent cyclic AMP phosphodiesterase activity obviated quantitative assessment.  相似文献
