The effects of long-term NaCl and KCl treatment on plant growth and antioxidative responses were investigated in Chenopodium album, a salt-resistant species widely distributed in semi-arid and light-saline areas of Xinjiang, China. Growth parameters [plant height, branch number, leaf morphology and chlorophyll (Chl) content], the level of oxidative stress [superoxide anion radical (O2 ), hydrogen peroxide (H2O2) and malondialdehyde (MDA) concentrations], activity of antioxidant enzymes [superoxide dismutase (SOD), catalase (CAT), peroxidase (POX)], the contents of non-enzymatic antioxidants [carotenoids (Car) and ascorbic acid (AsA)] and expression of selected genes were investigated. Plants were grown in the presence of 0, 50, and 300 mM NaCl or KCl for 2 months. Growth was stimulated by 50 mM NaCl or KCl, maintained stable at 300 mM NaCl, but was inhibited by 300 mM KCl. Three hundred mM NaCl did not affect O2 , H2O2, MDA, Car and AsA, but increased the activities of SOD, CAT and POX compared to the controls. RT-PCR analysis suggested that expression of some genes encoding antioxidant enzymes could be induced during long-term salt stress, which was consistent with the enzyme activities. Treatment with 300 mM KCl was associated with elevated oxidative stress, and significantly decreased Car and AsA contents. These results suggest that an efficient antioxidant machinery is important for overcoming oxidative stress induced by treatment with high NaCl concentrations in C. album. Other strategies of ion regulation may also contribute to the differential tolerance to Na and K at higher concentrations.  相似文献

To elucidate the physiological mechanism of chilling stress mitigated by cinnamic acid (CA) pretreatment, a cucumber variety (Cucumis sativus cv. Jinchun no. 4) was pretreated with 50 μM CA for 2 d and was then cultivated at two temperatures (15/8 and 25/18 °C) for 1 d. We investigated whether exogenous CA could protect cucumber plantlets from chilling stress (15/8 °C) and examined whether the protective effect was associated with the regulation of antioxidant enzymes and lipid peroxidation. At 2 d, exogenous CA did not influence plant growth, but induced the activities of some antioxidant enzymes, including superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), guaiacol peroxidase (GPX, EC 1.11.1.7), glutathione peroxidase (GSH-Px, EC 1.6.4.2) and ascorbate peroxidase (APX, EC 1.11.1.11) in cucumber leaves, and it also elevated the contents of reduced glutathione (GSH) and ascorbate (AsA). When CA was rinsed and the CA-pretreated seedlings were exposed to different temperatures, the antioxidant activities in leaves at 3 d had undergone additional change. Chilling increased the activities of CAT, GSH-PX, APX, GSH and AsA in leaves, but the combination of CA pretreatment and chilling enhanced the antioxidant activities even more. Moreover, chilling inhibited plant growth and increased the contents of malonaldehyde (MDA), superoxide radical (O2) and hydrogen peroxide (H2O2) in cucumber leaves, and the stress resulted in 87.5% of the second leaves being withered. When CA pretreatment was combined with the chilling stress, we observed alleviated growth inhibition and decreased contents of MDA, H2O2 and O2 in comparison to non-pretreated stressed plants, and found that the withered leaves occurred at a rate of 25.0%. We propose that CA pretreatment increases antioxidant enzyme activities in chilling-stressed leaves and decreases lipid peroxidation to some extent, enhancing the tolerance of cucumber leaves to chilling stress.  相似文献

Pigeonpea [Cajanus cajan (L.) Millsp.] is a waterlogging-sensitive legume crop. We studied the effect of waterlogging stress on hydrogen peroxide (H2O2) content, lipid peroxidation and antioxidant enzyme activities in two pigeonpea genotypes viz., ICPL-84023 (waterlogging resistant) and MAL-18 (waterlogging susceptible). In a pot experiment, waterlogging stress was imposed for 6 days at early vegetative stage (20 days after sowing). Waterlogging treatment significantly increased hydrogen peroxide accumulation and lipid peroxidation, which indicated the extent of oxidative injury posed by stress conditions. Enzyme activities of peroxidase (POX), catalase (CAT), ascorbate peroxidase (APX), superoxide dismutase (SOD) and polyphenol oxidase (PPO) increased in pigeonpea roots as a consequence of waterlogged conditions, and all the enzyme activities were significantly higher in waterlogged ICPL-84023 than in MAL-18. POX activity was the maximum immediately after imposing stress, therefore, it was suggested to be involved in early scavenging of H2O2, while rest of the enzymes (CAT, APX, SOD and PPO) were more important in late responses to waterlogging. Present study revealed that H2O2 content is directly related to lipid peroxidation leading to oxidative damage during waterlogging in pigeonpea. Higher antioxidant potential in ICPL-84023 as evidenced by enhanced POX, CAT, APX, SOD and PPO activities increased capacity for reactive oxygen species (ROS) scavenging and indicated relationship between waterlogging resistance and antioxidant defense system in pigeonpea.  相似文献

The effects of foliar spraying with spermidine (Spd) on antioxidant system in tomato (Lycopersicon esculentum Mill.) seedlings were investigated under high temperature stress. The high temperature stress significantly inhibited plant growth and reduced chlorophyll (Chl) content. Application of exogenous 1 mM Spd alleviated the inhibition of growth induced by the high temperature stress. Malondialdehyde (MDA), hydrogen peroxide (H2O2) content and superoxide anion (O2) generation rate were significantly increased by the high temperature stress, but Spd significantly reduced the accumulation of reactive oxygen species (ROS) and MDA content under the stress. The high temperature stress significantly decreased glutathione (GSH) content and activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), ascorbate peroxidase (APX), glutathione reductase (GR), monodehydroascorbate reductase (MDHAR) and dehydroascorbate reductase (DHAR), but increased contents of dehydroascorbic acid (DHA), ascorbic acid (AsA), and oxidized glutathione (GSSG) in tomato leaves. However, Spd significantly increased the activities of antioxidant enzymes, levels of antioxidants and endogenous polyamines in tomato leaves under the high temperature stress. In addition, to varying degrees, Spd regulated expression of MnSOD, POD, APX2, APX6, GR, MDHAR, DHAR1, and DHAR2 genes in tomato leaves exposed to the high temperature stress. These results suggest that Spd could change endogenous polyamine levels and alleviate the damage by oxidative stress enhancing the non-enzymatic and enzymatic antioxidant system and the related gene expression.  相似文献

The aim of the study was to relate the effects of deficiency and excess of Mn with the generation of reactive oxygen species (ROS) and altered cellular redox environment in mulberry (Morus alba L.) cv. Kanva-2 plants. Mn deficiency symptom appeared as mild interveinal chlorosis in middle leaves. Mn-excess did not produce any specific symptom. Leaf water potential (Ψ) was increased in Mn-deficient and Mn-excess mulberry plants. Mn-deficient leaves contained less Mn, less chloroplastic pigments and high tissue Fe, Zn and Cu concentrations. Starch content was increased with increasing Mn supply. While reducing sugar content increased in Mn-deficient and Mn-excess plants as well, non-reducing sugars remained unaffected in Mn-deficient plants and decreased in Mn-excess plants. Moreover, study of antioxidative responses, oxidative stress (H2O2 and lipid peroxidation) and cellular redox environment [dehydroascorbate (DHA)/ascorbic acid (AsA) ratio] in Mn-stressed mulberry plants was also undertaken. Both hydrogen peroxide and lipid peroxidation were enhanced in the leaves of Mn-deficient plants. Increased H2O2 concentration in Mn-excess leaves did not induce oxidative damage as indicated by no change in lipid peroxidation. The ratio of the redox couple (DHA/AsA) was increased both in Mn-deficient or Mn-excess plants. The activities of superoxide dismutase (EC 1.15.1.1) and catalase (EC 1.11.1.6) increased in Mn-deficient plants. The activity of ascorbate peroxidase (EC 1.11.1.11) increased with increasing Mn supply. The results suggest that deficiency or excess of Mn induces oxidative stress through enhanced ROS generation and disturbed redox couple in mulberry plants.  相似文献

Hydrogen peroxide (H2O2), an active oxygen species, is widely generated in many biological systems and mediates various physiological and biochemical processes in plants. In this study, we demonstrated that exogenous H2O2 was able to improve the tolerance of wheat seedlings to salt stress. Treatments with exogenous H2O2 for 2 days significantly enhanced salt stress tolerance in wheat seedlings by decreasing the concentration of malondialdehyde (MDA), the production rate of superoxide radical (O2 ), and increasing the activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX), and the concentration of glutathione (GSH) and carotenoids (CAR). To further clarify the role of H2O2 in preventing salt stress damage, CAT and ascorbate (AsA), the specific H2O2 scavengers, were used. The promoting effect of exogenous H2O2 on salt stress could be reversed by the addition of CAT and AsA. It was suggested that exogenous H2O2 induced changes in MDA, O2 , antioxidant enzymes and antioxidant compounds were responsible for the increase in salt stress tolerance observed in the experiments. Therefore, H2O2 may participate in antioxidant enzymes and antioxidant compounds induced tolerance of wheat seedlings to salt stress. The results also showed that exogenous H2O2 had a positive physiological effect on the growth and development of salt-stressed seedlings.  相似文献

The purpose of this study was to elucidate whether exogenous spermidine (Spd) protection of tomato (Solanum lycopersicum L.) seedlings under salinity-alkalinity stress is associated with antioxidant enzymes in the chloroplast. The effects of exogenous Spd on antioxidant enzyme activity and antioxidant content in the chloroplast were evaluated in seedlings of salt-sensitive ecotype (Zhongza 9) grown in a 75 mM salinity-alkalinity solution, with or without 0.25 mM Spd foliar spraying. Results showed that salinity-alkalinity stress increased MDA content, superoxide anion O2?- generation rate, superoxide dismutase (SOD), ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR) activities and ratio of AsA/DHA and reduced contents of ascorbate (AsA), dehydroascorbate (DHA), AsA+DHA, glutathione (GSH), oxidized glutathione (GSSG), GSH+GSSG, dehydroascorbate reductase (DHAR) activity and ratio of GSH/GSSG in chloroplasts. The exogenous Spd application combined with salinity-alkalinity stress decreased the O2?- generation rate and MDA content compared to salinity-alkalinity stress alone. The exogenous Spd also increased AsA-GSH cycle components and increased all antioxidant enzyme activities in most cases. Therefore, exogenous Spd alleviates salinity-alkalinity stress damage using antioxidant enzymes and non-enzymatic systems in chloroplasts.  相似文献

The influence of varied Mg supply (10-1000 micromolar) and light intensity (100-580 microeinsteins per square meter per second) on the concentrations of ascorbate (AsA) and nonprotein SH-compounds and the activities of superoxide dismutase (SOD; EC 1.15.11) and the H2O2 scavenging enzymes, AsA peroxidase (EC 1.11.1.7), dehydroascorbate reductase (EC 1.8.5.1), and glutathione reductase (EC 1.6.4.2) were studied in bean (Phaseolus vulgaris L.) leaves over a 13-day period. The concentrations of AsA and SH-compounds and the activities of SOD and H2O2 scavenging enzymes increased with light intensity, in particular in Mg-deficient leaves. Over the 12-day period of growth for a given light intensity, the concentrations of AsA and SH-compounds and the activities of these enzymes remained more or less constant in Mg-sufficient leaves. In contrast, in Mg-deficient leaves, a progressive increase was recorded, particularly in concentrations of AsA and activities of AsA peroxidase and glutathione reductase, whereas the activities of guaiacol peroxidase and catalase were only slightly enhanced. Partial shading of Mg-deficient leaf blades for 4 days prevented chlorosis, and the activities of the O2.− and H2O2 scavenging enzymes remained at a low level. The results demonstrate the role of both light intensity and Mg nutritional status on the regulation of O2.− and H2O2 scavenging enzymes in chloroplasts.  相似文献

Salinity, a severe environmental factor, has limited the growth and productivity of crops. Many compounds have been applied to minimize the harmful effects of salt stress on plant growth. An experiment was conducted to investigate the interactive effects of exogenous ascorbic acid (AsA) and gibberellic acid (GA3) on common bean (Phaseolus vulgaris L. cv. Naz) seedlings under salt stress. The changes of growth parameters, photosynthetic and non-photosynthetic pigments and potassium content showed that the addition of 1 mM AsA and/or 0.05 mM GA3 considerably decreased the oxidative damage in common bean plants treated with 200 mM NaCl. The NaCl-stressed seedlings exposed to AsA or GA3, specifically in their combination, exhibited an improvement in sodium accumulation in both roots and shoots, as compared to NaCl-treated plants. NaCl treatment increased hydrogen peroxide (H2O2) content and lipid peroxidation indicated by accumulation of malondialdehyde (MDA), whereas the interaction of AsA with GA3 decreased the amounts of MDA and H2O2. In the meantime, interactive effect of these substances enhanced protein content and the activity of the antioxidant enzyme, guaiacol peroxidase, in common bean plants under salt stress. It was concluded that synergistic interaction between AsA and GA3 could alleviate the adverse effects of salinity on P. vulgaris seedlings.  相似文献

The mechanism of growth amelioration in salt-stressed pennyroyal (Mentha pulegium L.) was investigated by exogenous application of penconazole (PEN). Seven weeks after sowing, seedlings were treated with increasing NaCl concentrations (0, 25, 50, and 75 mM) with or without PEN (15 mg l?1) and were harvested randomly at different times. Results showed that some growth parameters and the relative water content (RWC) decreased under salt stress, while lipid peroxidation, H2O2 content, activities of superoxide dismutase (SOD; EC 1.15.1.1), peroxidase (POX; EC 1.11.1.7), polyphenol oxidase (PPO; EC 1.10.3.1), catalase (CAT; EC 1.11.1.6), and ascorbate peroxidase (APX; EC 1.11.1.1) remarkably increased. Exogenous application of PEN increased some growth parameters, RWC, antioxidant enzyme activities, and H2O2 content, but the effects of PEN were more significant under salt stress conditions. PEN treatment also decreased lipid peroxidation. These results suggest that PEN-induced tolerance to salt stress in M. pulegium plants may be related to regulation of antioxidative responses and H2O2 level.  相似文献

Ascorbic acid (AsA) is the most abundant antioxidant in plants and plays a role in responding to oxidative stress. It has been shown that AsA plays a role in protecting against abiotic stresses. Rice seedlings stressed with 5 μM CdCl2 showed typical Cd toxicity (chlorosis and increase in malondialdehyde content). Rice seedlings pretreated with heat shock at 45°C (HS) or H2O2 under non-HS conditions resulted in the increase in ascorbic acid (AsA) content and the AsA/dehydroascorbate ratio in rice leaves. Exogenous application of AsA or L-galactonone-1, 4-lactone (GalL), a biosynthetic precursor of AsA, under non-HS conditions, which resulted in an increase in AsA content in leaves, enhanced subsequent Cd tolerance of rice seedlings. Pretreatment with imidazole, an inhibitor of NADPH oxidase, under HS conditions significantly decreased H2O2 and AsA contents in leaves and reduced subsequent Cd tolerance of rice seedlings. We also observed that pretreatment with lycorine, which is known to inhibit the conversion of GalL to AsA, significantly inhibited HS-induced AsA accumulation in leaves and reduced HS-induced protection against subsequent Cd stress of rice stress. It appears that HS- or H2O2-induced protection against subsequent Cd stress of rice seedlings is mediated through AsA. The time-course analyses of HS in rice seedlings demonstrated that the accumulation of H2O2 preceded the increase in AsA. Based on the data obtained in this study, it could be concluded that the early accumulation of H2O2 during HS signals the increase in AsA content, which in turn protects rice seedlings from oxidative damage caused by Cd.  相似文献

As a major antioxidant in plants, ascorbic acid (AsA) plays a very important role in the response to aluminum (Al) stress. However, the effect of AsA on the mitigation of Al toxicity and the mechanism of nitrate nitrogen (NO3 ?–N) uptake by plants under Al stress are unclear. In this study, a hydroponic experiment was conducted using peak 1 A rice (sterile line, Indica) with weaker resistance to Al and peak 1 superior 5 rice (F1 hybrid, Indica) with stronger resistance to Al to study the effects of exogenous AsA on the physiological and biochemical responses to NO3 ?–N uptake by rice roots exposed to 50 μmol L?1 Al. Al stress induced increases in the concentrations of H2O2 and malondialdehyde (MDA) and in the activities of antioxidant enzymes [such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX)]. Plasma membrane (PM) H+-ATPase and H+-pump activities, endogenous AsA content and NO3 ?–N uptake in rice roots decreased under Al stress. After treatment with 2 mmol L?1 exogenous AsA combined with Al, concentrations of H2O2 and MDA in roots notably decreased, and endogenous AsA content and activities of SOD, POD, CAT, and APX in rice roots increased significantly; furthermore, the interaction of PM H+-ATPase and the 14-3-3 protein was also enhanced significantly compared with that in control plants without AsA treatment, which clearly increased NO3 ?–N uptake. Based on all of these data, the application of AsA significantly reduced the accumulation of H2O2 and MDA and increased the activities of PM H+-ATPase and the H+-pump by increasing the endogenous AsA content, the antioxidant enzyme activities, and the interaction of PM H+-ATPase and the 14-3-3 protein in the roots of the two rice cultivars under Al stress, thereby improving the uptake of NO3 ?–N in rice.  相似文献

以‘拉丁诺’白三叶(Trifolium repens cv.‘Ladino’)为试验材料,研究外源H2S处理对PEG6 000(聚乙二醇)模拟干旱胁迫下白三叶叶片相对含水量(RWC)、膜脂过氧化、活性氧成分、抗氧化酶、抗坏血酸-谷胱甘肽循环代谢和非酶抗氧化物质的影响,以揭示H_2S调控白三叶抗旱性的生理机制。结果显示:(1)0.2 mmol/L的外源NaHS(H_2S供体)能显著提高干旱胁迫下白三叶的叶片相对含水量,维持显著较低的电解质渗透率(EL)和丙二醛(MDA)含量。(2)与直接干旱胁迫相比,干旱胁迫下外源添加NaHS处理的白三叶叶片内超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性显著增强,抗坏血酸-谷胱甘肽循环代谢中关键酶抗坏血酸过氧化物酶(APX)、脱氢抗坏血酸还原酶(DHAR)、单脱水抗坏血酸还原酶(MDHAR)和谷胱甘肽还原酶(GR)活性及其抗氧化中间产物抗坏血酸(AsA)、谷胱甘肽(GSH)含量也显著提高。(3)叶片类黄酮、总酚和原花青素的含量在一定的胁迫时间范围内亦显著增加,并伴随着活性氧成分O_2~(-·)产生速率和H_2O_2水平降低。研究认为,外源H2S能通过促进干旱胁迫下白三叶体内的多重抗氧化防御能力来提高其幼苗的抗旱性。  相似文献

Sphaerophysa kotschyana is a Turkish endemic and endangered plant that grows near Salt Lake, in Konya, Turkey. However, little is known about the ability of this plant to generate/remove reactive oxygen species (ROS) or its adaptive biochemical responses to saline environments. After exposure of S. kotschyana to 0, 150, and 300 mM NaCl for 7 and 14 days, we investigated (1) the activities and isozyme compositions of the antioxidant enzymes superoxide dismutase (SOD), catalase (CAT), peroxidase (POX), ascorbate peroxidase (APX), and glutathione reductase (GR); (2) the oxidative stress parameters NADPH oxidase (NOX) activity, lipid peroxidation (MDA), total ascorbate (tAsA) content, and total glutathione content (tGlut); and (3) ROS levels for superoxide anion radical (O 2 ·? ), hydrogen peroxide (H2O2), hydroxyl radicals (OH·), and histochemical staining of O 2 ·? and H2O2. H2O2 content increased after 14 days of salt stress, which was consistent with the results from histochemical staining and NOX activity measurements. In contrast, oxidative stress induced by 150 mM NaCl was more efficiently prevented, as indicated by low malondialdehyde (MDA) levels and especially at 7 days, by increased levels of SOD, POX, APX, and GR. However, at 300 mM NaCl, decreased levels of protective enzymes such as SOD, CAT, POX, and GR, particularly with long-term stress (14 days), resulted in limited ROS scavenging activity and increased MDA levels. Moreover, at 300 mM NaCl, the high H2O2 content caused oxidative damage rather than inducing protective responses against H2O2. These results suggest that S. kotschyana is potentially tolerant to salt-induced damage only at low salt concentrations.  相似文献

To provide a theoretical basis for revealing the mechanism of winter leaf reddening in evergreen species, the relationships between winter leaf reddening, reactive oxygen species (ROS) and the antioxidant system of Buxus microphylla ‘Wintergreen’ were studied. The pigment changes, ROS production, lipid peroxidation and antioxidants activities of sun leaves during the reddening and regreening processes were investigated, using green shade leaves as controls. The carotenoids in the sun leaves increased linearly with reddening but decreased with the regreening. There was no significant difference in either the superoxide anions (O 2 ) or malondialdehyde (MDA) changes between the sun and shade leaves, and their O 2 contents were positively correlated with MDA. In contrast to the shade leaves, the sun leaves showed a trend in which the hydrogen peroxide (H2O2) changes were closely related to the reddening process and positively correlated with carotenoids content but not with the MDA content. A similar trend was observed for catalase (CAT) and ascorbate peroxidase (APX) activity between the sun and shade leaves, but superoxide dismutase (SOD) and peroxidase (POD) activity and the ascorbate (AsA) content differed between these two ecotypes. Furthermore, the sun leaves had higher CAT activity and AsA content than the corresponding shade leaves. These results suggested that H2O2 might play an important role in the winter reddening of sun leaves by promoting the accumulation of carotenoids. In addition, SOD, POD and AsA probably play a photoprotective role in winter-red sun leaves, while the changes in O 2 , CAT and APX were independent of winter leaf reddening and were more likely responses to stress caused by low temperatures.  相似文献

采用盆栽实验,对干旱胁迫下黄土高原4种乡土禾草冰草、长芒草、无芒隐子草和白羊草叶片过氧化氢(H2O2)、丙二醛(MDA)含量、抗氧化酶超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)、抗坏血酸过氧化物酶(APX)、谷胱甘肽还原酶(GR)、脱氢抗坏血酸还原酶(DHAR)、单脱氢抗坏血酸还原酶(MDHAR)、谷胱甘肽过氧化物酶(GPX)活性和非酶抗氧化物质还原型抗坏血酸(AsA)、还原型谷胱甘肽(GSH)、类胡萝卜素(Car)含量进行了测定。 结果表明:随着干旱胁迫程度的加剧,4种乡土禾草叶片H2O2、MDA含量均呈增加趋势,这说明它们均遭受了干旱所造成的氧化胁迫,且干旱程度越大其遭受的氧化胁迫也越大。由于4种乡土禾草均为禾本科植物并生存于相同的生态环境中,它们在抗氧化特性上具有一定共性。在60%FC和45%FC干旱胁迫下,4种乡土禾草均可以通过增加抗氧化酶SOD、CAT、APX、GR、DHAR、MDHAR、GPX活性和非酶抗氧化物质AsA含量来抵御干旱所造成的氧化胁迫。由于种属差异,4种乡土禾草的抗氧化特性也存在差异。在60%FC和45%FC干旱胁迫下,冰草、无芒隐子草和白羊草还通过增加非酶抗氧化物质Car含量增强抗氧化能力,长芒草和白羊草则还可通过增加POD活性抵御干旱。在60%FC干旱胁迫下,冰草还可通过增加非酶抗氧化物质GSH含量提高其抗氧化性。采用隶属函数法对4种乡土草种抗氧化能力的综合评价表明,冰草的抗氧化能力最强,其次为无芒隐子草和白羊草,长芒草的抗氧化能力最差。  相似文献

The aim of this study is to investigate the impacts of exogenous salicylic acid (SA) pretreatments on hydrogen peroxide (H2O2) accumulation, protein oxidation, and H2O2-scavenging enzymes in leaves of Cd-treated flax seedlings. Cd-enhanced H2O2 levels were related to increased activities of guaiacol peroxidase (POX, EC 1.11.1.7) and ascorbate peroxidase (APX, EC 1.11.1.11), and were independent of changes in catalase (CAT, EC 1.11.1.6) and superoxide dismutase (SOD, EC 1.15.1.1) activities. In control flax seedlings, exogenous SA pretreatments inhibited the activity of CAT, resulted in an enhanced production of H2O2 suggesting that SA requires H2O2 to initiate an oxidative stress. However, although leaves of Cd-free flax seedlings pretreated with SA accumulated in vivo H2O2 by 1.2-fold compared with leaves of Cd-only exposed ones; the damage to growth and proteins after the exposure to Cd was significantly less, indicating that SA can regulate the Cd-induced oxidative stress. Moreover, the Cd-treated seedlings primed with SA exhibited a higher level of total antioxidant capacities and increased activities of H2O2-detoxifying enzymes.  相似文献

We studied the effect of water stress imposed at anthesis and pre-anthesis stages on oxidative stress and antioxidant activity in four wheat cultivars, two hexaploid Triticum aestivum cultivars, drought resistant cv. C 306 and drought susceptible cv. Hira, and two tetraploid cultivars, T. durum cv. A 9-30-1 and T. dicoccum cv. HW 24. Water stress decreased relative water content (RWC), membrane stability index (MSI), and increased H2O2 and malondialdehyde (MDA) contents as well as activity of superoxide dismutase (SOD), catalase (Cat) and peroxidase (POX) in all the genotypes at all the stages. Both the tetraploid cultivars showed higher RWC, MSI and SOD activity, and lower H2O2 and MDA contents under water stress than hexaploid ones. Cat and POX activities were highest in C 306.  相似文献

The role of exogenous methyl jasmonate (MeJA) in alleviating drought stress was investigated on Huangguogan. Except for intercellular CO2 concentration, MeJA had little effect on net photosynthetic rate, stomatal conductance, and transpiration rate under drought stress. Compared with drought stress, MeJA significantly alleviated the decrease of chlorophyll content. However, chlorophyll a/b ratio was significantly increased. MeJA significantly increased proline and soluble sugar contents, significantly decreased the O2 and H2O2 levels, and increased SOD and POD activities. In addition, the MDA content of drought stress was the highest of all treatments. MeJA significantly reduced MDA content in drought-stressed Huangguogan leaves. Although the Ascorbic acid (AsA) contents of 500 and 1000 mg L?1 MeJA treatments were lower than that of 250 mg L?1 MeJA, but all concentration of MeJA treatments delayed the decline of AsA content. Therefore, MeJA could induce drought stress tolerance by increasing the osmotic adjustment substances and antioxidant activities.  相似文献

Effect of salinity on Hyacinth bean, Lablab purpureus (HA-4 cultivar) was evaluated in 10-day old seedlings with 100–500 mM NaCl over 72 h of exposure. The stress reduced dry and fresh weight, leaf surface area, root and shoot length, total chlorophyll, and RWC. Oxidative stress markers, H2O2, glutathione, TBARS, proline, ascorbic acid, total phenols, and total soluble sugar contents were significantly elevated. Salinity enhanced antioxidant enzymes, POX, and GR activities and reduced that of CAT in concentration and time dependent manner in leaves. Antioxidant enzymes in roots showed inverse relationship with concentration and time of exposure. Metabolic enzyme β-amylase activity increased in both leaves and roots. Acid phosphatase decreased in leaves and elevated in roots. Intensity of constitutive isozymes correlated with in vitro levels under stress, but the protein band patterns differed from controls. Lablab showed reasonable tolerance up to 300 mM NaCl, but leaves and roots differed in their response.  相似文献
