【背景】前期结果表明,DDT降解菌株Chryseobacterium sp. PYR2可高效去除土壤中的DDT等污染物,具有潜在的应用价值,但该菌对植物的影响尚不清楚。【目的】探讨菌株Chryseobacterium sp. PYR2对植物的促生作用及其机理,为后续开发DDT降解及植物促生双效功能菌剂提供理论依据。【方法】配制该菌株的不同梯度稀释菌悬液,用纸卷发芽法和盆栽法研究菌悬液对小麦种子萌发和植株生长的影响;Salkowski法测定PYR2合成吲哚-3-乙酸(Indole-3-acetic acid,IAA)量;单因素实验研究不同培养条件对菌株生长及IAA合成的影响;液相色谱-串联质谱-多反应监测(LC-MS/MS-MRM)方法分析IAA在PYR2菌体内的生物合成途径。【结果】PYR2菌悬液可明显提高小麦种子萌发率并促进小麦植株的生长,小麦的侧根数、株高、鲜重、干重等指标均明显提高。该作用是由于菌株PYR2可以合成植物生长激素IAA。最适IAA合成条件:温度30°C,pH 7.0-8.0,盐浓度0.5%,L-色氨酸50mg/L。代谢液中检测到色醇、色胺和吲哚-3-乙酰胺3种中间代谢产物,推测PYR2体内存在3条IAA合成途径,分别为吲哚-3-丙酮酸(IPy A)、TAM和IAM途径。【结论】菌株PYR2对小麦具有明显的促生效果,是由于其具有多条高效合成IAA的代谢途径,表明其在农药污染土壤的生物修复及作物种植中具有潜在的应用前景。  相似文献

【目的】研究微嗜酸寡养单胞菌(Stenotrophomonas acidaminiphila)CW117中磷酸吡哆胺氧化酶基因pnpox(phosphopyridoxamine oxidase,pnpox)在维生素B6(VB6)合成中的贡献及对黄曲霉毒素B1(aflatoxin B1,AFB1)的降解活性。【方法】采用基因插入突变方式,对菌株CW117中磷酸吡哆胺氧化酶基因pnpox进行突变,得到突变菌株。通过高效液相色谱法(high performance liquid chromatography,HPLC)检测突变株对AFB1的降解活性,以及突变株中吡哆醇和吡哆醛的合成情况,确定基因pnpox在寡养单胞菌体内VB6合成中的贡献和黄曲霉毒素降解代谢作用。【结果】成功构建了磷酸吡哆胺氧化酶基因突变子pnpox::pK19mobΩ2HMB,突变子吡哆醛的合成量较野生型菌株显著减少,吡哆醇合成量与野生型菌株无显著性差异;同时,突变子与野生型株CW117对AFB1的降解活性未发现显著性差异。【结论】菌株CW117中磷酸吡哆胺氧化酶在吡哆醛合成的过程中起着重要作用,该基因突变会导致VB6的严重缺乏,影响寡养单胞菌正常生长,但该基因对CW117降解黄曲霉毒素无显著性贡献。  相似文献

【目的】研究hfq基因在Mesorhizobium huakuii 7653R抵抗外界不利环境和共生固氮中的功能特性。【方法】利用pK19mob同源重组方法构建7653R hfq基因的插入失活突变株7653RΔhfq,并构建互补菌株7653RΔhfq-C,对hfq在压力胁迫和共生固氮中的功能特性进行研究。【结果】与野生型7653R相比,突变株7653RΔhfq的生长速率降低,热激处理后致死率升高;hfq突变影响了7653R中部分sRNA的表达;在4.5%乙醇和50 mmol H_2O_2生长胁迫下,突变株适应性明显较野生型差。另外,接种突变株的紫云英结瘤能力和固氮酶活性都明显降低。【结论】hfq基因作为重要的转录后调控因子,在7653R抵御外界胁迫环境和与宿主紫云英的共生固氮中发挥了重要作用。  相似文献

【背景】植物-微生物联合修复技术在减轻环境胁迫作用、增强植物抗性、改善矿区景观等方面发挥重要作用,其中根际促生菌可与植物相互作用,促进植物生长,增强植物对非生物胁迫的耐受性。【目的】鉴定铜尾矿中分离得到的2株根际细菌,研究其对植物生长的促生特性,测定2株菌的接种对于煤矸石胁迫下香根草的影响。【方法】对铜尾矿中分离得到的2株菌进行16S rRNA基因序列鉴定和扫描电子显微镜观察,对菌株的促生特性进行测定,并将其分别接种于覆土5 cm的煤矸石和掺土10%的煤矸石中生长的香根草根际,测定2个月后香根草的理化指标和生理学指标。【结果】菌株P5-11和P5-19经鉴定均为草螺菌(Herbaspirillum),2株菌均具有固氮、溶磷、产吲哚乙酸(Indole-3-Acetic Acid,IAA)和产铁载体的特性,其中P5-19的产吲哚乙酸能力约为P5-11的2倍,具有较好的促生能力;2株菌均能提高香根草的株高、生物量、氮积累量和抗氧化酶活性,并降低丙二醛积累量。【结论】2株草螺菌均具有良好的促生特性,能够促进煤矸石胁迫下香根草的生长发育,这不仅为促生菌肥的研制提供了优良菌种,也为香根草在矿区生态恢复中的应用提供了参考价值。  相似文献

粪产碱菌的Tn5转座诱变及吲哚乙酸生物合成特性的研究   总被引:5,自引:0,他引:5  
粪产碱菌(Alcaligenes faecalis)A1501的吲哚乙酸(IAA)合成需要外源色氨酸参与。在不含色氨酸的限制性培养基中,A1501能良好生长,但不能合成IAA,表明在A1501中存在一条依赖于色氨酸的IAA合成途径。A1501的IAA合成具有菌体密度依赖特性。采用Tn5转座诱变技术构建A1501的突变库,从3500多株Tn5转染子中分离到一株色氨酸营养缺陷型突变株AT63。该Tn5突变株在不含色氨酸的限制性培养基上不能生长,但仍能进行IAA的生物合成,每毫升菌体密度等于10的突变株菌体的IAA合成量为224μg。对突变株AT63的研究表明在A1501中至少存在两条IAA合成途径:一条以色氨酸为合成前体,另一条以吲哚-3-磷酸甘油为前体。Southern杂交结果表明突变株中Tn5插入位点可能位于编码色氨酸合成酶基因上。  相似文献

【目的】为了进一步鉴定铜绿假单胞菌转录调控因子σ~(38)对2个拷贝吩嗪合成基因簇(phz A1-G1和phz A2-G2)的具体调控方式并推定介导绿脓菌素合成代谢的可能调控机制。【方法】根据铜绿假单胞菌基因组信息,利用同源重组原理构建rpo S基因缺失突变株Δrpo S以及克隆全长rpo S基因作互补分析;再以单一吩嗪基因簇缺失突变株Δphz1和Δphz2为出发菌株,分别构建rpo S缺失突变株Δrpo Sphz1和rpo S插入突变株Δrpo Sphz2,测定并比较野生株及相关突变株的绿脓菌素合成量,初步推定σ~(38)因子对2个不同吩嗪基因簇表达的调控方式。【结果】在GA培养基中,突变株Δrpo S的绿脓菌素合成量比野生株显著增加;互补分析证实,σ~(38)可使突变株Δrpo S的绿脓菌素降低并接近野生株PAO1水平;与对照株Δphz1相比,突变株Δrpo Sphz1的绿脓菌素合成量因σ~(38)因子缺失而显著减少;而与对照株Δphz2相比,突变株Δrpo Sphz2的绿脓菌素合成量因σ~(38)因子缺失显著增加。【结论】转录调控因子σ~(38)对铜绿假单胞菌绿脓菌素的合成代谢的确具一定的负调控作用;结合已报道的研究结果,初步推定:σ~(38)因子通过负调控吩嗪基因簇phz1,正调控吩嗪基因簇phz2的表达实现对绿脓菌素合成代谢的调控。  相似文献

一株来自大棚温室甜椒根际的绿针假单胞菌Pseudomonas chlororaphis G-05,可分泌抗生物质吩嗪-1-羧酸,并具有抑制辣椒疫霉的生物防治功效。【目的】为了系统研究该菌株的生物防治功能及抗生物质合成与分泌机制。【方法】首先通过生化法和16S rDNA同源比对法对该菌株进行系统分类的初步鉴定,再根据基因的同源性从G-05基因组DNA中克隆长1.4 kb的gacS基因的部分保守区段,采用抗庆大霉素基因(gentamycin resistance cassette, aacC1)插入失活的策略构建了该基因突变株G-05S。【结果】在King’ s B(KMB)或PPM培养基中,突变株G-05S合成吩嗪-1-羧酸的能力受到明显抑制。然而,突变株G-05S分泌的吲哚乙酸与野生株相比无显著差异。互补实验表明,gacS基因的表达可以使突变株G-05S的吩嗪-1-羧酸的合成恢复到野生株水平。【结论】由此推测,GacS(Global activator sensor )对不同次生代谢物的调控具有特异性。  相似文献

【目的】由青枯雷尔氏菌(Ralstonia solanacearum)引起的植物青枯病是一种毁灭性土传病害。胞外多糖(extracellular polysaccharides,EPS)是青枯雷尔氏菌关键的致病因子之一。通过构建胞外多糖缺失突变株,研究胞外多糖在青枯病致病中的作用。【方法】从青枯雷尔氏菌FJAT-91的基因组中克隆出胞外多糖合成结构基因epsD同源臂,克隆至自杀性质粒p K18mobsacB,再将庆大霉素抗性基因(Gm)插入同源臂中间,获得重组质粒p K18-epsD。将重组质粒转化至青枯雷尔氏菌FJAT-91感受态细胞中,通过同源重组敲除epsD基因,获得EPS合成缺失的突变株FJAT-91Δeps 。研究突变株与野生菌株在菌落形态、胞外多糖合成、运动能力、定殖能力的差异性。【结果】突变菌株FJAT-91ΔepsD与出发菌株FJAT-91相比:胞外多糖产量显著减少,生长较慢;泳动能力(swimming motility)和群集运动能力(swarming motility)显著降低;在番茄苗根部和茎部的定殖能力显著降低;弱化指数(AI)为0.905,鉴定为无致病力菌株。【结论】胞外多糖在青枯雷尔氏菌的致病中起着关键的作用,本课题研究成果为开发植物疫苗提供了优良的材料与研究基础。  相似文献

【目的】获得江苏沿海滩涂盐生药用植物中华补血草内生及根际具有1-氨基环丙烷-1-羧酸(ACC)脱氨酶活性的细菌,研究其遗传多样性和潜在促生活性。【方法】从中华补血草和根际土壤分离筛选具有ACC脱氨酶活性的菌株,对其ACC脱氨酶活性定量检测,通过16S r RNA基因序列分析确定菌株系统发育地位。同时研究其固氮、溶磷、产植物生长素吲哚乙酸(IAA)及耐盐能力。【结果】分离筛选获得18株具有ACC脱氨酶活性的内生与根际细菌,定量检测发现其中有13株菌的ACC脱氨酶含量在20 nmolα-KA/(mg Pr·h)以上,有11株菌可以固氮,7株菌能够解磷,9株菌产生IAA。菌株的Na Cl盐耐受范围多数在0–13%之间。16S r RNA基因测序表明,活性菌株分属于7个属,多样性丰富,节杆菌属(Arthrobacter)为优势类群。其中菌株KLBMP 5180为节杆菌属的潜在新种。【结论】江苏沿海滩涂盐生药用植物中华补血草共生环境中具有丰富多样的具ACC脱氨酶活性的菌株,并存在潜在新物种资源,具有进一步研究价值。  相似文献

For mass isolation of plant metaphase chromosomes, cultured cells of wheat (Triticum monococcum) and parsley (Petroselinum hortense) were synchronized by hydroxyurea and colchicine treatment. This synchronization procedure resulted in high mitotic synchrony, especially in suspension cultures of parsley in which 80% of the cells were found to be at the metaphase stage. Mitotic protoplasts isolated from these synchronized cell cultures served as a source for isolation of chromosomes. The described isolation and purification method yielded relatively pure chromosome suspension. The uptake of the isolated plant chromosomes into recipient wheat, parsley, and maize protoplasts was induced by polyethylene-glycol treatment. Summary Turfgrass, like other major crop species, is recalcitrant to manipulation in vitro. To perform efficient genetic transformation of turfgrass, it is necessary to optimize tissue culture conditions. In most reports, plant tissue culture techniques have been applied to propagate a single cultivar or several cultivars in one species of turfgrass. In this experiment, four turfgrass genera were used, namely common bermudagrass, Cynodon dactylon [L.] Pers. (California origin); red fescue, Festuca rubra L. var. rubra ‘Shadow’; perennial ryegrass, Lolium perenne L. ‘Barbal’; and Kentucky bluegrass, Poa pratensis L. ‘Merion.’ Mature seeds were surface-sterilized and cultured on basal Murashige and Skoog (MS) media supplemented with 30–250 μM 2,4-dichlorophenoxyacetic acid (2,4-D) for callus induction. Regeneration media consisted of MS supplemented with 5–10 μM 6-benzyladenine (BA). Among the genera, Poa had the higest callus induction percentage (CIP) regardless of 2,4-D concentration, followed by Cynodon, Lolium, and Festuca, respectively. Cynodon and Lolium had the highest callus regeneration percentage (CRP) and overall regeneration rate (ORR). Festuca had a poor CIP, CRP, and ORR compared to other studied genera. Cynodon produced the highest shoot number per explant. Based on the results of the present study, MS medium supplemented with 60 μM 2,4-D (for callus induction) and 7.5 μM BA (for regeneration) can be used in multi-generic transformation studies with the genera used.  相似文献

The enterobacterial pathogen Erwinia chrysanthemi causes soft rot diseases on a wide range of plants, including the model plant Arabidopsis thaliana. This bacterium proliferates in the host by secreting a set of pectin degrading enzymes responsible for symptom development. In addition, survival of this bacterium in planta requires two high-affinity iron acquisition systems mediated by siderophores and protective systems against oxidative damages, suggesting the implication by both partners of accurate mechanisms controlling their iron homeostasis under conditions of infection. In this review, we address this question and we show that ferritins both from the pathogen and the host are subtly implicated in the control of this interplay.  相似文献

Studies of induced systemic resistance using strains of plant growth-promoting rhizobacteria (PGPR) have concentrated on the use of individual PGPR as inducers against multiple diseases of a single crop. To date, few reports have examined the potential of PGPR strain mixtures to induce systemic resistance against diseases of several different plant hosts. The objective of this study was to select mixtures of compatible PGPR strains with the capacity to elicit induced systemic resistance in four hosts. The specific diseases and hosts tested in this study included: bacterial wilt of tomato (Lycopersicon esculentum) caused by Ralstonia solanacearum, anthracnose of long cayenne pepper (Capsicum annuum var. acuminatum) caused by Colletotrichum gloeosporioides, damping off of green kuang futsoi (Brassica chinensis var. parachinensis) caused by Rhizoctonia solani, and cucumber mosaic virus (CMV) on cucumber (Cucumis sativus). To examine compatibility, seven selected PGPR strains were individually tested for in vitro antibiosis against all other PGPR strains and against three of the tested pathogens (R. solanacearum, C. gloeosporioides, and R. solani). No in vitro antibiosis was observed among PGPR strains or against pathogens. Twenty-one combinations of PGPR and seven individual PGPR were tested in the greenhouse for induced resistance activity. Results indicated that four mixtures of PGPR and one individual strain treatment significantly reduced the severity of all four diseases compared to the nonbacterized control: 11 mixtures reduced CMV of cucumber, 16 mixtures reduced bacterial wilt of tomato, 18 mixtures reduced anthracnose of long cayenne pepper, and 7 mixtures reduced damping off of green kuang futsoi. Most mixtures of PGPR provided a greater disease suppression than individual PGPR strains. These results suggest that mixtures of PGPR can elicit induced systemic resistance to fungal, bacterial, and viral diseases in the four hosts tested.  相似文献

Rapid bendings of the pulvinus inMimosa pudica, of the trap lobes inDionaea muscipula andAldrovanda vesiculosa, and of the tentacle in Drosera are triggered by action potentials in their motor cells. The action potential ofMimosa may be a C1-spike, and that ofDionaea andAldrovanda may be a Ca2+-spike. Propagation of action potentials in the petiole or motor organ is thought to be electrotonie, cell-to-cell, transmission. The Ca 2+ release from unidentified organelles in the pulvinus or the Ca2+ influx of the cells in the trap with the action current and activation of contractile fibrillar network having ATPase activity in the cytoplasm must be involved in the rapid bending. Contractions of fibrils may open pores in the membrane of the motor cells upon activation. Outward bulk flow of the vacuolar sap through these pores, due to the pressure inside the cell, must result in turgor loss of the motor cells and then the bending of the organ.  相似文献

Cuticles from the adaxial surface of Citrus aurantium L. leaves and from the pericarp of Lycopersicon esculentum L. and Capsicum annuum L. were isolated enzymatically and their oxygen permeability was determined. Isolated cuticles were mounted between a gaseous and an aqueous compartment with the physiological outer side of the membrane facing the gaseous compartment. Permeability for oxygen was characterized by permeability (P) and diffusion (D) coefficients. P and D were independent of the driving force (gradient of oxygen concentration) across the cuticle, thus, Henry's law was obeyed. P values for the diffusion of oxygen varied between 3·10-7 (Citrus), 1.4·10-6 (Capsicum), and 1.1·10-6 (Lycopersicon) m·s-1. Extraction of soluble lipids from the cuticles increased the permeability. By treating the cutin matrix and the soluble lipids as resistances in series, it could be demonstrated that the soluble lipids were the main resistance for oxygen permeability in Citrus cuticles. However, in Lycopersicon and Capsicum, both the cutin matrix and the soluble lipids determined the total resistance. P values were not affected by either the proton concentration (pH 3–9) or the cations (Na+, Ca2+) present at the morphological inner side of the cuticles. It is concluded that the water content of cuticles does not affect the permeability properties for oxygen. Partition coefficients indicated a high solubility of oxygen in the cuticle of Citrus. Cucumber (Cucumis sativus L.) and pear (Pyrus domestica Medik.) fruit proplastids, and pea (Pisum sativum L., cv. Meteor) leaf chloroplasts, extracted by osmotic rupture of protoplasts isolated after degradation of the cell walls by cellulase and pectinase, agglutinated in the presence of Con A. Agglutination of cucumber proplastids was inhibited by anti-Con A and by methyl D-gluco/manno pyranosides but not by methyl D-galactopyranoside. Fluorescein isothiocyanate-conjugated Con A (FITC-Con A) rendered agglutinated clumps fluorescent. If cellulase was omitted from the macerating medium, Con A-mediated agglutination did not occur even if proplatids were subsequently incubated with cellulase. Proplastids and chloroplasts extracted by conventional mechanical disruption methods were not agglutinated by Con A and did not acquire fluorescence with FITC-Con A. The amount of pectin held in cell walls by ionic bonds only was determined by extraction with cyclohexanediamine tetraacetic acid (CDTA) at room temperature, to remove calcium ions without degrading the galacturonan chains. Enzymic degradation was avoided by extracting the cell walls with phenol-acetic acid-water during preparation. From cell walls of celery petioles, cress hypocotyls and tomato and cucumber pericarp CDTA extracted 64–100 mg g-1 pectin, leaving 80–167 mg g-1 uronic acid in the residue. An additional extraction at high ionic strength was used to make the galacturonan chains more flexible and thus detach any pectins held by steric interactions, but the amount released in this way was small. Most of the residual uronic acid polymers could be extracted by cold alkali and remained soluble on neutralisation, showing that it was not water-insolubility that prevented their extraction with CDTA. Optimum growth conditions and inoculation regimes were determined for severalFrankia strains isolated from both Alnus and Casuarina host plants. Growth conditions were estabilished that allowed a reduction in generation time to less than 15 hours for certain Alnus derivedFrankia. Differences in plant growth response were observed with differing inoculum levels and soil mixtures. Elite strains of Alnus derivedFrankia were isolated that elicited similar growth reponses in allAlnus species tested; however, differences were observed betweenFrankia strains and plant growth response of variousCasuarina species tested.  相似文献
