Winter rye (Secale cereale L. cv Musketeer) grown at 5 °C/250 µmol photons m–2 s–1 exhibited a relative reduction state of PS II comparable to that of rye grown at 20 °C but high light (800 µmol photons m–2 s–1) (1-qP = 0.32) whereas winter rye grown at 20 °C/250 µmol photons m–2 s–1 exhibited values of 1-qP ( 0.15) comparable to plants grown at 5 °C but low light (50 µmol photons m–2 s–1). The apparent size of the electron donor pool to PS I, estimated either in vivo or in vitro in the presence of methylviologen by A820 was positively correlated with the relative reduction state of PS II under the steady-state growth conditions. Immunoblotting of rye thylakoid polypeptides indicated that the relative contents of Lhcb1, Lhcb2, D1, Cyt f, PC, PsaA/PsaB heterodimer and the -subunit of ATPase complex exhibited minimal changes on a Chl basis. In contrast, a 2-fold increase in plastoquinone A content was associated with increasing growth irradiance at growth temperatures of either 5 or 20 °C. We suggest that the increases in the apparent size of the electron donor pool to PS I associated with rye grown at either 5 °C/250 µmol photons m–2 s–1or 20 °C/800 µmol photons m–2 s–1 may be explained by an increased thylakoid plastoquinone A content, coupled with possible enhanced PS I cyclic electron transport and/or increased capacity for electron donation from the stroma to the intersystem electron transport chain. The results are discussed with respect to photosynthetic adjustment to changes in PS II excitation pressure in winter rye.  相似文献

The growth yield of the PUFA-producing marine microalgaIsochrysis galbana ALII-4 grown in a light limited chemostat, was measured under a wide variety of conditions of incident irradiance (I O ) and dilution rates (D). The experiments were conducted under laboratory conditions at 20 °C under continuous light. D ranged from 0.0024 to 0.0410 h–1 at three intensities of Io (820, 1620 and 3270 µmol photon m–2 s–1) close to those found in outdoor cultures. A maximum efficiency max = 0.616 g mol photon–1 was obtained at I O = 820 µmol photon m–2 s–1 and D = 0.030 h–1 and the maximum capacity of the biomass to metabolize the light harvested was found to be 13.1 µmol photon g–1 s–1. Above this value, a significant drop in the system efficiency was observed. A new approach based in the averaged irradiance is used to assess the photon flux absorbed by the biomass.  相似文献

Laurencia brongniartii is usually found at depths below 4 m, but can be found in shallow subtidal areas in crevices and on the walls of a coral reef in Amami Oshima Island, Kagoshima Prefecture, Japan, where irradiances were significantly lower than those at similar depths in open water. In preparation for the possible cultivation of this species for its antibiotic compounds, the effects of temperature and irradiance on photosynthesis and growth were measured. Photosynthesis and growth rates of L. brongniartii explants were highest at 26 and 28 °C, which closely corresponded to temperatures found during August to late December when it was most abundant. The estimated maximum photosynthesis rate (P max) was 4.41 mol photon m–2 s–1 at 26 °C and 4.07 mol photon m–2 s–1 at 28 °C. Saturating irradiance occurred at 95 mol photon m–2 s–1 at 26 °C and 65 mol photon m–2 s–1 at 28 °C. In contrast, growth experiments at 41.7 mol photon m–2 s–1 caused bleaching of explants and the maximum growth rate observed during the study was 3.02 ± 0.75% day–1 at 28 °C and 25 mol photon m–2 s–1. The difference in the saturating irradiance for photosynthesis and the irradiance that caused bleaching in growth experiments suggests that long-term exposure to high irradiance was detrimental and should be addressed before the initiation of large scale cultivation.  相似文献

Tobacco plantlets were cultured in vitro under high (200 µmol m–2 s–1) or low (60 µmol m–2 s–1) irradiance with or without saccharose in the medium. Light microscopy and image analysis were used to evaluate the effect of these culture conditions on leaf anatomy. Addition of saccharose resulted in thicker leaves (all leaf layers) and larger mesophyll cells under both growth irradiances. Various irradiance affected leaf anatomy differently when plantlets had been cultivated in presence or absence of saccharose in the medium. While under high irradiance in presence of saccharose leaf thickness and number of chloroplasts per cell section were increased, plantlets grown under high irradiance in absence of saccharose had thinner leaves and less chloroplasts per cell section. Three Bromeliaceae species of the medium Orinoco basin, Venezuela, were compared in their light-use characteristics. The bromeliads studied were two species of pineapple, i.e. the wild species Ananas ananassoides originating from the floor of covered moist forest, and the primitive cultivar Panare of Ananas comosus mostly cultivated in semi-shaded palm swamps, and Pitcairnia pruinosa, a species abundant in highly sun exposed sites on rock outcrops. Ananas species are Crassulacean acid metabolism (CAM) plants, P. pruinosa is C3 plant. Plants were grown at low daily irradiance (LL = 1.3 mol m–2 d–1 corresponding to an incident irradiance of 30 mol m–2 s–1) and at high irradiance (HL = 14.7 mol m–2 d–1 or 340 mol m–2 s–1), and CO2 and H2O-vapour gas exchange and photochemical (qP) and non-photochemical quenching (qNP) of chlorophyll a fluorescence of photosystem 2 (PS2) were measured after transfer to LL, medium irradiance (ML = 4.1 mol m–2 d–1 or 95 mol m–2 s–1) and HL. All plants showed flexible light-use, and qP was kept high under all conditions. LL-grown plants of Ananas showed particularly high rates of CAM-photosynthesis when transferred to HL and were not photoinhibited.  相似文献

The responses to irradiance of photosynthetic CO2 assimilation and photosystem 2 (PS2) electron transport were simultaneously studied by gas exchange and chlorophyll (Chl) fluorescence measurement in two-year-old apple tree leaves (Malus pumila Mill. cv. Tengmu No.1/Malus hupehensis Rehd). Net photosynthetic rate (P N) was saturated at photosynthetic photon flux density (PPFD) 600-1 100 (mol m-2 s-1, while the PS2 non-cyclic electron transport (P-rate) showed a maximum at PPFD 800 mol m-2 s-1. With PPFD increasing, either leaf potential photosynthetic CO2 assimilation activity (Fd/Fs) and PS2 maximal photochemical activity (Fv/Fm) decreased or the ratio of the inactive PS2 reaction centres (RC) [(Fi – Fo)/(Fm – Fo)] and the slow relaxing non-photochemical Chl fluorescence quenching (qs) increased from PPFD 1 200 mol m-2 s-1, but cyclic electron transport around photosystem 1 (RFp), irradiance induced PS2 RC closure [(Fs – Fo)/Fm – Fo)], and the fast and medium relaxing non-photochemical Chl fluorescence quenching (qf and qm) increased remarkably from PPFD 900 (mol m-2 s-1. Hence leaf photosynthesis of young apple leaves saturated at PPFD 800 mol m-2 s-1 and photoinhibition occurred above PPFD 900 mol m-2 s-1. During the photoinhibition at different irradiances, young apple tree leaves could dissipate excess photons mainly by energy quenching and state transition mechanisms at PPFD 900-1 100 mol m-2 s-1, but photosynthetic apparatus damage was unavoidable from PPFD 1 200 mol m-2 s-1. We propose that Chl fluorescence parameter P-rate is superior to the gas exchange parameter P N and the Chl fluorescence parameter Fv/Fm as a definition of saturation irradiance and photoinhibition of plant leaves.  相似文献

Spirulina platensis (= Arthrospira fusiformis) was isolated from Lake Chitu, a saline, alkaline lake in Ethiopia, where it forms an almost unialgal population. Optimum growth conditions were studied in a turbidostat. Cultures grown in modified Zarrouk's medium and exposed to a range of light intensities (20–500 µmol photons m–2s–1) showed a maximum specific growth rate (µmax) of 1.78 d–1. Quantum yield for growth (µ) was 3.8% at the optimum light for growth of 330 µmol photons m–2s–1, and ranged from 2.8 to 9.4%. With increase in irradiance, the chlorophyll a concentration decreased, and the carotenoids/chlorophyll a ratio increased by a factor of 2.4. The phosphorus to carbon ratio (P/C) showed some variation, while the nitrogen to carbon ratio (N/C) remained relatively constant, thus causing fluctuations in the N:P ratio (7–11) of cells. An optimum N:P ratio of about 7 was attained in cells growing at the optimum light for growth. Results from the continuous culture experiments agreed well with maximum values of photosynthetic efficiency given in the literature for natural populations of S. platensis in the soda lakes of East Africa, Lake Arenguade (Ethiopia), and Lake Simbi (Kenya).  相似文献

Biomass and eicosapentaenoic acid (EPA) productivities were investigated in a flat panel airlift loop reactor ideally mixed by static mixers. Growth with ammonium, urea and nitrate as nitrogen source were performed at different aeration rates. Cultures grew on ammonium but the decay of pH strongly inhibited biomass increase. On urea biomass productivity reached 2.35 g L–1d–1at an aeration rate of 0.66 vvm (24 h light per day, 1000 mol photon m–2s–1). Aeration rates between 0.33 vvm and 0.66 vvm and maximal productivities on urea were linearly dependent. Productivity on nitrate never exceeded 1.37 g L–1d–1. In the range of maximum productivity photosynthesis efficiency of 10.6% was reached at low irradiance (250 mol photon m–2s–1). Photosynthesis efficiency decreased to 4.8% at 1000 mol photon m–2s–1. At these high irradiances the flat panel airlift reactor showed a 35% higher volume productivity than the bubble column. At continuous culture conditions the influence of CO2concentration in the supply air was tested. Highest productivities were reached at 1.25% (v/v) CO2where the continuous culture yielded 1.04 g L–1d–1(16 h light per day, 1000 mol photon m–2s–1). The average EPA content amounted to 5.0% of cell dry weight, that resulted in EPA productivities of 52 mg L–1d–1(continuous culture, 16 h light per day) or 118 mg L–1d–1(batch culture, 24 h light per day).  相似文献

In vitro cultures of Nephrolepis exaltata and Cordyline fruticosa were stored at 5°, 9° or 13°C, at a low irradiance (3–5 mol m–2 s–1) or in darkness. Prior to storage the cultures were subjected to 18°, 21°, 24° or 27°C and 15, 30 or 45 mol m–2 s–1 in a factorial combination.The optimal storage conditions for Nephrolepis were 9°C in complete darkness. These cultures were still transferable to a peat/perlite mixture at the end of the experimental period of 36 months.The optimal storage conditions for Cordyline were 13°C and a low light level (±3–5 mol m-2 s-1). When the pre-storage conditions were normal growth room conditions (24°C and 30 mol m-2 s-1), in vitro cultures could be stored for 18 months. With the most favourable pre-storage treatment (18°C and 15 mol m-2 s-1) some cultures still had green shoots after 36 months of storage, but did not survive transfer to peat/perlite.Pre-conditioning before storage was most favourable for Nephrolepis, and not that important, but still favourable, for Cordyline. There was an interaction between pre-storage temperature and pre-storage irradiance. AxenicTrentepohlia odorata was cultured at three different NH4Cl levels (3.5 × 10–2, 3.5 × 10–3, 3.5 × 10–4 M) and three different light intensities (48, 76, 122 µmol m–2 s–1). Chloride had no effect on growth over this range of concentration. High light intensity and high NH4Cl concentration enhanced the specific growth rate. The carotenoid content increased under a combination of high light intensity and low N concentration. WhenD. bardawil was exposed to the same combination of growth conditions, there was an increase in its carotenoid content. The light saturation and the light inhibition constants (K s andK i, respectively) for growth, and the saturation constant (K m) for NH4Cl were determined. TheK s andK i values were higher inT. odorata (66.7 and> 122 mol m–2 s–1, respectively) than inD. bardawil (5.1 and 14.7 µmol m–2 s–1, respectively). Grain sorghum [Sorghum bicolor (L.) Moench. cvs. TX430 and KS82] was grown in a Haynie very fine sandy loam (coarse-silty, mixed, superactive, calcareous, mesic Mollic Udifluvents) under constant 47 % shade or full irradiance in a greenhouse under two watering regimes to see the combined and individual effects of low irradiance (LI) and low water (LW) on the sorghum genotypes. Under the high-irradiance (HI) and high-water (HW) treatment (control) and the LI-HW treatment, TX430 grew taller than KS82. Both LI and LW reduced several times the fresh and dry masses. Under the control conditions, TX430 reached its maximum net photosynthetic rate (P Nmax) of 28.93 mol m–2 s–1 at a photosynthetic photon flux density (PPFD) of 1 707 mol m–2 s–1, and KS82 reached its P Nmax of 28.32 mol m–2 s–1 at a PPFD of 2 973 mol m–2 s–1. The fact that TX430 had P Nmax under a lower PPFD than KS82 may relate to its taller growth under LI conditions. Hence genotypes of sorghum might be selected for low irradiance using curves relating P N to PPFD.  相似文献

The effects of irradiance and growth phase on the concentration ofascorbic acid (AA) were examined in Isochrysis sp.(T.ISO),one of the most popular microalgal species used in aquaculture. Across fourdifferent irradiances (15, 40, 100 and 200 mol m–2s–1 with continuous illumination; i.e., lightconditionstypically found in hatcheries), the average % AA in cultures during logarithmicphase ranged from 0.38% (15 mol m–2s–1) to 0.49% (100 mol m–2s–1) of cell dry weight. Average % AA values forcultures at 40 and 100 mol m–2s–1 reduced by more than half to 0.16% and 0.20%,respectively with the onset of stationary phase.  相似文献

Summary Selaginella lepidophylla, the resurrection plant, curls dramatically during desiccation and the hypothesis that curling may help limit bright light-induced damage during desiccation and rehydration was tested under laboratory conditions. Restraint of curling during desiccation at 25° C and a constant irradiance of 2000 mol m–2 s]t-1 significantly decreased PSII and whole-chain electron transport and the Fv/Fm fluorescence yield ratio following rehydration relative to unrestrained plants. Normal curling during desiccation at 37.5°C and 200 mol m–2 s–1 irradiance did not fully protect against photoinhibition or chlorophyll photooxidation indicating that some light-induced damage occurred early in the desiccation process before substantial curling. Photosystem I electron transport was less inhibited by high-temperature, high-irradiance desiccation than either PSII or whole-chain electron transport and PSI was not significantly affected by restraint of curling during desiccation at 25°C and high irradiance. Previous curling also helped prevent photoinhibition of PSII electron transport and loss of whole-plant photosynthetic capacity as the plants uncurled during rehydration at high light. These results demonstrate that high-temperature desiccation exacerbated photoinhibition, PSI was less photoinhibited than PSII or whole-chain electron transport, and stem curling ameliorated bright light-induced damage helping to make rapid recovery of photosynthetic competence possible when the plants are next wetted.  相似文献

This study reports the effects of light availability during the acclimatization phase on photosynthetic characteristics of micropropagated plantlets of grapevine (Vitis vinifera L.) and of a chestnut hybrid (Castanea sativa × C. crenata). The plantlets were acclimatized for 4 weeks (grapevine) or 6 weeks (chestnut), under two irradiance treatments, 150 and 300 mol m–2 s–1 after in vitro phases at 50 mol m–2 s–1. For both treatments and both species, leaves formed during acclimatization (so-called `new leaves') showed higher photosynthetic capacity than the leaves formed in vitro either under heterotrophic or during acclimatization (so-called `persistent leaves'), although lower than leaves of young potted plants (so-called `greenhouse leaves'). In grapevine, unlike chestnut, net photosynthesis and biomass production increased significantly with increased light availability. Several parameters associated with chlorophyll a fluorescence indicated photoinhibition symptoms in chestnut leaves growing at 300 mol m–2 s–1. The results taken as a whole suggest that 300 mol m–2 s–1 is the upper threshold for acclimatization of chestnut although grapevine showed a better response than chestnut to an increase in light.  相似文献

Understanding how photosynthetic capacity acclimatises when plants are grown in an atmosphere of rising CO2 concentrations will be vital to the development of mechanistic models of the response of plant productivity to global environmental change. A limitation to the study of acclimatisation is the small amount of material that may be destructively harvested from long-term studies of the effects of elevation of CO2 concentration. Technological developments in the measurement of gas exchange, fluorescence and absorption spectroscopy, coupled with theoretical developments in the interpretation of measured values now allow detailed analyses of limitations to photosynthesisin vivo. The use of leaf chambers with Ulbricht integrating spheres allows separation of change in the maximum efficiency of energy transduction in the assimilation of CO2 from changes in tissue absorptance. Analysis of the response of CO2 assimilation to intercellular CO2 concentration allows quantitative determination of the limitation imposed by stomata, carboxylation efficiency, and the rate of regeneration of ribulose 1:5 bisphosphate. Chlorophyll fluorescence provides a rapid method for detecting photoinhibition in heterogeneously illuminated leaves within canopies in the field. The object of this work was to determine, using a full-factorial experiment, the influence of temperature, irradiance and salinity on growth and hepatotoxin production by Nodularia spumigena, isolated from Lake Alexandrina in the south-east of South Australia. Higher levels of biomass (determined as particulate organic carbon, POC), toxin production and intracellular toxin concentration per mg POC were produced under light limited conditions (30 mol m–2 s–1) and at salinities equal to or greater than those experienced in Lake Alexandrina. Both highest biomass and total toxin production rates were recorded at temperatures equal to or greater than those of the lake (20 and 30°C). The temperature at which maximum biomass and toxin production was recorded decreased from 30°C for cultures grown at 30 mol m–2 s–1 to 20°C when grown at 80 mol m–2 s–1. In contrast, intracellular toxin per mg POC was highest at the lowest growth temperature, 10°C, at both 30 and 80 mol m–2 s–1. It appears that the optimum temperature for biosynthetic pathways used in the production of toxin is lower than the optimum temperature for those pathways associated with growth. Intracellular toxin levels were higher in cells cultured at 10°C/30 mol m–2 s–1 whereas the majority of the toxin was extracellular in cells grown at 30°C/30 mol m–2 s–1. This implies that the highest concentration of toxin in lake water would occur under high temperature and high irradiance conditions. Individual environmental parameters of salinity, irradiance and temperature were all shown to influence growth and toxin production. Notwithstanding, the overall influence of these three parameters on toxin production was mediated through their effect upon growth rate.  相似文献

The increase in growth, determined by dry weight gain, of rice (Oryza sativa L.) and maize (Zea mays L.) caused by foliar applications of 9--L(+) adenosine, a putative second messenger elicited by triacontanol, was studied under different environmental conditions. Maize seedlings cultured in the greenhouse under approximately 100 mol m–2s–1 of light prior to treatment with L(+) adenosine did not respond unless they received supplemental light (250–300 mol m–2s–1) after treatment. Exposure of rice seedlings growing for 16 h at 150 mol m–2s–1 to short periods of 450 mol m–2s–1 (< than 20 min) had no effect on the positive response of rice to L(+) adenosine; however, exposure for 60 min or more increased the growth of rice and obviated the effect of L(+) adenosine. Rice seedlings treated with L(+) adenosine at different times during the day responded only when treated 9 to 12h after initiation of the photoperiod. Normal growing temperatures under different light intensities had little or no direct effect on the response of plants to L(+) adenosine.  相似文献

Transformed Nicotiana plumbaginifolia plants with constitutive expression of nitrate reductase (NR) activity were grown at different levels of nitrogen nutrition. The gradients in foliar NO 3 content and maximum extractable NR activity observed with leaf order on the shoot, from base to apex, were much decreased as a result of N-deficiency in both the transformed plants and wild type controls grown under identical conditions. Constitutive expression of NR did not influence the foliar protein and chlorophyll contents under any circumstances. A reciprocal relationship between the observed maximal extractable NR activity of the leaves and their NO 3 content was observed in plants grown in nitrogen replete conditions at low irradiance (170 mol photons·m–2 ·s–1). This relationship disappeared at higher irradiance (450 mol photons·m–2·S–1) because the maximal extractable NR activity in the leaves of the wild type plants in these conditions increased to a level that was similar to, or greater than that found in constitutive NR-expressors. Much more NO 3 accumulated in the leaves of plants grown at 450 mol photons·m–2·s–1 than in those grown at 170 mol photons·m–2·s–1 in N-replete conditions. The foliar NO 3 level and maximal NR activity decreased with the imposition of N-deficiency in all plant types such that after prolonged exposure to nitrogen depletion very little NO 3 was found in the leaves and NR activity had decreased to almost zero. The activity of NR decreased under conditions of nitrogen deficiency. This regulation is multifactoral since there is no regulation of NR gene expression by NO 3 in the constitutive NR-expressors. We conclude that the NR protein is specifically targetted for destruction under nitrogen deficiency. Biochemical and biophysical parameters, including D1-protein turnover, chlorophyll fluorescence, oxygen evolution activity and zeaxanthin formation were measured in the marine seagrassZostera capricorni (Aschers) in response to limiting (100 mol·m–2·–1), saturating (350 mol·m–2·s–1) or photoinhibitory (1100 mol·m–2·s–1) irradiances. Synthesis of D1 was maximal at 350 mol·m–2·s–1 which was also the irradiance at which the rate of photosynthetic O2 evolution was maximal. Degradation of D1 was saturated at 350 mol·m–2·s–1. The rate of D1 synthesis at 1100 mol·m–2·s–1 was very similar to that at 350 mol·m–2·s–1 for the first 90 min but then declined. At limiting or saturating irradiance little change was observed in the ratio of variable to maximal fluorescence (Fv/Fm) measured after dark adaptation of the leaves, while significant photoinhibition occurred at 1100 mol·m–2·s–1. The proportion of zeaxanthin in the total xanthophyll pool increased with increasing irradiance, indicative of the presence of a photoprotective xanthophyll cycle in this seagrass. Adjustment in leaf area : mass ratio called leaf area ratio (LAR) is one of the strategies to optimize photon harvesting. LAR was recorded for 10 genotypes of Hevea brasiliensis under high irradiance and low temperature and the genotypes were categorized into two groups, i.e. high LAR and low LAR types. Simultaneously, the growth during summer as well as winter periods, photosynthetic characteristics, and in-vitro oxidative damage were studied. Low LAR (19.86±0.52 m2 kg–1) types, recorded an average of 18.0 % chlorophyll (Chl) degradation under high irradiance and 7.1 % Chl degradation under low temperature. These genotypes maintained significantly higher net photosynthetic rate (P N) of 10.4 mol(CO2) m–2 s–1 during winter season. On the contrary, the high LAR (24.33±0.27 m2 kg–1) types recorded significantly lower P N of 4 mol(CO2) m–2 s–1 and greater Chl degradation of 37.7 and 13.9 % under high irradiance and low temperature stress, respectively. Thus LAR may be one of the physiological traits, which are possibly involved in plant acclimation process under both stresses studied.  相似文献
