Zygophyllum dumosum Boiss. is a perennial Saharo-Arabian phytogeographical element and a dominant shrub on the rocky limestone southeast-facing slopes of the Negev desert. The plant is highly active during the winter, and semideciduous during the dry summer, i.e., it sheds its leaflets, while leaving the thick, fleshy petiole green and rather active during the dry season. Being resistant to extreme perennial drought, Z. dumosum appears to provide an intriguing model plant for studying epigenetic mechanisms associated with drought tolerance in natural habitats. The transition from the wet to the dry season was accompanied by a significant decrease in nuclear size and with posttranslational modifications of histone H3 N-terminal tail. Dimethylation of H3 at lysine 4 (H3K4)—a modification associated with active gene expression—was found to be high during the wet season but gradually diminished on progression to the dry season. Unexpectedly, H3K9 di- and trimethylation as well as H3K27 di- and trimethylation could not be detected in Z. dumosum; H3K9 monomethylation appears to be prominent in Z. dumosum during the wet but not during the dry season. Contrary to Z. dumosum, H3K9 dimethylation was detected in other desert plants, including Artemisia sieberi, Anabasis articulata and Haloxylon scoparium. Taken together, our results demonstrate dynamic genome organization and unique pattern of histone H3 methylation displayed by Z. dumosum, which could have an adaptive value in variable environments of the Negev desert.  相似文献

We assessed dung and track counts for indexing brocket deer abundance in seasonal habitats on a ranch where hunting is not practiced in the Bolivian lowlands. Surveys were replicated along four 10-km trails (totaling 180 km in the wet season and 90 km in the dry season) through four semideciduous forest habitats. Dung pellets and tracks were identified as belonging to Mazama gouazoubira or M. americana by size and shape. Pellet groups lasted more than 1 month during the dry season, but decayed within 1–2 weeks in the wet season. Mean density estimates based on dung counts varied widely between seasons for gray brockets (from 6.77±4.0 to 30.57±23.5 ind/km2; mean±SD) but not for reds (from 3.52±4.6 to 6.98±7.2 ind/km2). These values were probably too high due to underestimation of daily deposition rate, and were reduced during the wet season because of dung decay. We found consistently more dung in the dry season and more tracks in the wet season. Sightings of red brockets were too few for line-transect analysis (n=6), but those of gray brockets (n=42) produced an overall estimate of 5.6 ind/km2 (95% CI=3.5–9.0 ind/km2). Different estimates indicated that gray brockets were more abundant than reds in all situations, except perhaps in the riverine forest. Environmental factors affected these indices of abundance differently, and while we recommend the use of dung counts in dry-season scenarios, we think that index reliability should be assessed locally before conducting population comparisons.  相似文献

Spatiotemporal variations in microbial gene abundances were investigated to identify potential zones of methanotroph and methanogen biomass in a peat bog in Sarobetsu-genya wetland. The abundances of the bacterial and archaeal 16S rRNA genes, pmoA, and mcrA were 107–109, 107–108, 104–106, and 104–107 copies g−1 dry peat, respectively. Correlation analysis based on microbial gene abundances and environmental factors showed that the spatiotemporal distributions of the abundances of the four microbial genes in peat layers were similar. The mcrA abundance showed a significant negative correlation with the dissolved organic carbon content and a significant positive correlation with the peat temperature. The pmoA abundance was not detectable during the spring thaw when the lowest peat temperature at a depth of 50 cm was recorded. At a depth of 200 cm, the peat temperature exceeded 6°C throughout the year, and the mcrA abundance exceeded 104 copies g−1 dry peat. These results indicate that the seasonal microbial activity related to methane should be evaluated in not only the shallow but also the deep peat layers in order to elucidate the methane dynamics in boreal wetlands.  相似文献

The effect of formulation, fungal concentration, type and seasonal changes on the mortality of the tick Amblyomma variegatum was investigated. A previous study demonstrated high pathogenicity of strains of the fungi Beauveria bassiana and Metarhizium anisopliae against the ticks Rhipicephalus appendiculatus and Amblyomma variegatum (Kaaya et al. J Invertebr Pathol 1996; 67: 15–20). The present study was undertaken to explore possible additive or synergistic effects of the two fungi on A. variegatum. The effects of oil and water formulations at different concentrations of each fungus and combination of the two on the mortality of A. variegatum in the laboratory and in the field during the wet and dry seasons were determined and compared. The oil formulation performed better in all assays, with highest tick mortality of 92% occurring during the wet season at conidia concentration of 1 × 1010 conidia/ml of the mixed fungi compared to 49% for the water formulation at similar conidia concentration. However, at the same conidial concentration during the dry season, mortalities in the field were relatively low with the mixture of the fungi recording 24% and 17% tick mortality for the oil and water formulation respectively. The effect of infecting the ticks with a cocktail of the two fungi was inconclusive under more controlled conditions in the laboratory but field results under both wet and dry seasons indicated significant differences between the separate and mixed fungi infections. The results demonstrate a potential of cocktail formulations in the control of ticks and possibly of other arthropod pests.  相似文献

The bacterial community compositions in Chenopodium album and Stellaria media seeds recovered from soil (soil weed seedbank), from bulk soil, and from seeds harvested from plants grown in the same soils were compared. It was hypothesized that bacterial communities in soil weed seedbanks are distinct from the ones present in bulk soils. For that purpose, bacterial polymerase chain reaction denaturing gradient gel electrophoresis (PCR–DGGE) fingerprints, made from DNA extracts of different soils and seed fractions, were analyzed by principal component analysis. Bacterial fingerprints from C. album and S. media seeds differed from each other and from soil. Further, it revealed that bacterial fingerprints from soil-recovered and plant-harvested seeds from the same species clustered together. Hence, it was concluded that microbial communities associated with seeds in soil mostly originated from the mother plant and not from soil. In addition, the results indicated that the presence of a weed seedbank in arable soils can increase soil microbial diversity. Thus, a change in species composition or size of the soil weed seedbank, for instance, as a result of a change in crop management, could affect soil microbial diversity. The consequence of increased diversity is yet unknown, but by virtue of identification of dominant bands in PCR–DGGE fingerprints as Lysobacter oryzae (among four other species), it became clear that bacteria potentially antagonizing phytopathogens dominate in C. album seeds in soil. The role of these potential antagonists on weed and crop plant growth was discussed.  相似文献

Summary A heavy metal resistant bacterium, Bacillus circulans strain EB1 showed a high cadmium biosorption capacity coupled with a high tolerance to this metal when grown in its presence. Bacillus circulans EB1 cells grown in the presence of 28.1 mg cadmium/l were capable of removing cadmium with a specific biosorption capacity of 5.8 mg Cd/g dry wt biomass in the first 8 h. When the cells were pre-conditioned with low concentrations of cadmium in pre-grown medium, the uptake was increased to 6.7 mg Cd/g dry wt biomass. The maximum uptake of␣cadmium was during mid-logarithmic phase of growth. The resting cells (both wet and dry) of EB1 were also able to biosorb cadmium. Specific biosorption capacities of wet and dry biomass were 9.8 and 26.5 mg Cd/g dry wt biomass, respectively. Maximum cadmium removals by both wet and dry cells were at pH 7.0. The results showed that the cadmium removal capacity of resting cells was markedly higher than that of growing cells. Since both growing and resting cells had a high biosorption capacity for cadmium, EB1 cells could serve as an excellent biosorbent for removal of cadmium from natural environments.  相似文献

In the present study, antiproliferative, apoptotic and especially neurotoxic effects of Caulerpa racemosa var. cylindracea dry and wet extracts on mouse neuroblastoma cell line, NA2B were investigated by neurotoxicity screening test (NST). C. racemosa var. cylindracea wet and dry extracts were obtained by methanol (MT) extraction. The effect of the extracts on viability and proliferation was measured by MTT. NA2B cells were induced to differentiate using 1 μM dcAMP and the amount of inhibition of growing neurites in different dilutions (50, 35, 25, 15, 10 and 5 μl/ml) by extracts was measured. The number of apoptotic cells was computed by TUNEL method using cells in culture. It was found that majority of the cells died with dry extract above the level of 15 μl/ml due to the MT effect. Below this level, on the other hand, presence of cell death and antiproliferative effect was noted due to the toxic effects of C. racemosa var. cylindracea which was independent of MT. In all doses of wet extracts, similar but less prominent dose-dependent effects were observed. Below the level of 15 μl/ml, mild toxic effect presented itself with neurite inhibition. In addition to the toxic, apoptotic and antiproliferative effects of C. racemosa var. cylindracea, its neurotoxic effects possessing property at low concentrations which manifesting itself by neurite inhibition was also showed. This species offers a potential for developing new drugs due to its antiproliferative, toxic and apoptotic effects. Nevertheless, its neurotoxic effect is a factor to be considered as multifunctional agents especially in neuronal metabolism.  相似文献

The endophytic mycobiota of leaves and stems of the mistletoe Phoradendron perrottettii and its host tree Tapirira guianensis, two physiologically connected plant species of the Brazilian savannah in southeastern Brazil, were investigated to evaluate host and organ recurrence among endophytes. Leaves and stems of P. perrottettii and leaves of T. guianensis were sampled in the dry and wet season. Stems of T. guianensis were also sampled in the wet season. Endophytes were isolated by an adapted trituration and particle filtration protocol. A total of 1,615 isolates representing 99 species and 20 sterile morphotypes were recovered; 64 morphospecies occurred as singletons. The number of isolates and species was higher in the wet season. Leaves of P. perrottettii were less densely colonized than other organs studied, but were the most species-rich. Conversely, stems of T. guianensis yielded more isolates but were less species-rich. Both plants were found to harbor similar but distinguishable endophytic assemblages. The Jaccard’s index of similarity between the fungal assemblages of both plants was 0.82, higher than found for other plants in similar habitats. The fungal species composition seemed to be influenced by the collection season and organ type, as demonstrated by multivariate correspondence analysis. Paraconiothyrium brasiliense, P. sporulosum and Verticillium leptobactrum were the dominant species in P. perrottettii. In leaves of T. guianensis, Pseudocercospora sp., Phomopsis sp. 1 and Lecanicillium psalliotae were the most frequent, while Stagonospora sp. 1 and Phomopsis sp. 1 were the dominant endophytes in its stems. The results indicated that some of the dominant endophytic taxa isolated in this study colonize different hosts and plant organs while others seem to exhibit a high degree of host or organ recurrence. This study represents the first evaluation of diversity of fungal endophytes in natural vegetation of the Brazilian savannah and contributes information about the distribution and possible specificity of endophytes in tropical dicotyledoneous plants.  相似文献

Growth of Ruta graveolens shoots was induced when Bacillus sp. cell lysates were added to the culture medium. Elicitation of coumarin by this lysate was also very effective; the concentrations of isopimpinelin, xanthotoxin and bergapten increased to 610, 2120 and 1460 μg g−1 dry wt, respectively. It also had a significant effect on the production of psoralen and rutamarin (680 and 380 μg g−1 dry wt) and induced the biosynthesis of chalepin, which was not detected in the control sample, up to 47 μg g−1 dry wt With lysates of the Pectobacterium atrosepticum, their effect on growth was not so significant and had no effect on the induction of coumarin accumulation. But elicitation with this lysate was much more effective for inducing the production of furoquinolone alkaloids; the concentrations of γ-fagarine, skimmianine, dictamnine and kokusaginine rose to 99, 680, 172 and 480 μg g−1 dry wt, respectively.  相似文献

The structure and demographic processes were compared in shrub communities to test the effects of vegetation succession on population growth, fecundity and abundance of the dwarf birch (Betula nana L.), which is a rare and endangered plant species in Poland and a glacial relict in Central Europe. The effects of Ledum palustre L. and Vaccinium uliginosum L. were studied in the Linje nature reserve in Chełmińskie Lake District (northern Poland), in three permanent plots on a peat bog. Vegetative growth and reproduction of B. nana were lower in plant communities dominated by L. palustre and V. uliginosum, than in a reference site. Fecundity was also lower, despite the fact that the percentage share of potentially fertile age groups was similar in all study sites. Mortality of ramets was independent of vegetation, both for juvenile and mature stages. The results confirm that B. nana is intolerant of shade, and it is more abundant in vegetation without competitors. Light limitation can lead to its decline, primarily by a decrease in vegetative growth. Sexual reproduction may be negatively affected by shade, but it plays only small role in population growth. Butterfly larvae can destroy inflorescences, and thus contribute to low effectiveness of sexual reproduction. Increasing density of shrubs and trees in peat bogs can reduce the abundance of dwarf birch, and can lead to the extinction of its local populations.  相似文献

A genetic transformation system has been developed for callus cells of Crataegus aronia using Agrobacterium tumefaciens. Callus culture was established from internodal stem segments incubated on Murashige and Skoog (MS) medium supplemented with 5 mg l−1 Indole-3-butyric acid (IBA) and 0.5 mg l−1 6-benzyladenine (BA). In order to optimize the callus culture system with respect to callus growth and coloration, different types and concentrations of plant growth regulators were tested. Results indicated that the best average fresh weight of red colored callus was obtained on MS medium supplemented with 2 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) and 1.5 mg l−1 kinetin (Kin) (callus maintenance medium). Callus cells were co-cultivated with Agrobacterium harboring the binary plasmid pCAMBIA1302 carrying the mgfp5 and hygromycin phosphotransferase (hptII) genes conferring green fluorescent protein (GFP) activity and hygromycin resistance, respectively. Putative transgenic calli were obtained 4 weeks after incubation of the co-cultivated explants onto maintenance medium supplemented with 50 mg l−1 hygromycin. Molecular analysis confirmed the integration of the transgenes in transformed callus. To our knowledge, this is the first time to report an Agrobacterium-mediated transformation system in Crataegus aronia.  相似文献

Tripartite interactions among Paenibacillus lentimorbus NRRL B-30488 (B-30488), Piriformospora indica DSM 11827 (DSM 11827) and their consortia (B-30488:DSM 11827:: 1:1) with native rhizobial population in the rhizosphere of Cicer arietinum L. (Chick pea) was tested for enhancing nodulations and plant growth promotion. Number of nodules and dry weight per plant significantly enhanced (P = 0.05), which is further evident by N, P, and K uptake by plants and were found to be maximum in B-30488 treated followed by B-30488: DSM 11827 and DSM 11827, as compared with uninoculated control, in 60 days grown chickpea plants. Microbial community structure in the rhizosphere of the four treatments was assessed, using Biolog Eco and MT plates. Principal component analysis (PCA) of carbon source utilization pattern on Biolog Eco plates did not show any clustering among the four samples indicating that in case of individually DSM 11827 and B-30488 treated chickpea rhizosphere there was significant change in microbial community structure, compared with lesser changes in un-inoculated and B-30488 and DSM 11827 consortium treated chickpea rhizosphere microflora. Additional carbon sources tested using Biolog MT plates, higher activity of lignin, chitin, and cellulose utilizing microbial communities in the rhizosphere being stimulated by root exudates treated with B-30488 alone or in consortia with DSM 11827, and, in turn, should encourage beneficial symbiotic or mutualistic microorganisms that can act as plant growth promoting and biocontrol agents.  相似文献

This study investigated the factors affecting in vitro flowering of Perilla frutescens. The shoots regenerated from cotyledonary and hypocotyl explants cultured on Murashige and Skoog (MS) medium supplemented with benzyladenine (BA) and indole-3-acetic acid, each at 0.5 mg l−1, were excised and transferred to MS medium containing 30 g l−1 of sucrose, 8.25 g l−1 of ammonium nitrate, and 1.0 mg l−1 of BA. After 40 d of culture, 86.2% of shoots flowered and most of which self-fertilized in vitro and produced mature fruits with viable seeds. These seeds were germinated and plants were grown to maturity and flowered in soil under greenhouse conditions. The in vitro flowering system reported in this study may facilitate rapid breeding of P. frutescens and offers a model system for studying the physiological mechanism of flowering.  相似文献

The ecophysiological variabilities in the ectohydrolytic enzyme profiles of the three species of Pseudoalteromonas, P. citrea, P. issachenkonii, and P. nigrifaciens, have been investigated. Forty-one bacteria isolated from several invertebrates, macroalgae, sea grass, and the surrounding water exhibited different patterns of hydrolytic enzyme activities measured as the hydrolysis of either native biopolymers or fluorogenic substrates. The activities of the following enzymes were assayed: proteinase, tyrosinase, lipase, amylase, chitinase, agarase, fucoidan hydrolase, laminaranase, alginase, pustulanase, cellulase, β-glucosidase, α- and β-galactosidases, β-N-acetylglucosaminidase, β-glucosaminidase, β-xylosidase, and α-mannosidase. The occurrence and cell-specific activities of all enzymes varied over a broad range (from 0 to 44 μmol EU per hour) and depended not only on taxonomic affiliation of the strain, but also on the source/place of its isolation. This suggests ‘specialization’ of different species for different types of polymeric substrates as, for example, all strains of P. citrea and P. issachenkonii hydrolyzed alginate and laminaran, while strains of P. nigrifaciens were lacking the ability to hydrolyze most of the algal polysaccharides. The incidence of certain enzymes such as fucoidan hydrolases, alginate lyases, agarases, and α-galactosidases might be strain specific and reflect its particular ecological habitat. In order to assess the long-term impacts of saline groundwater irrigation to Haloxylon ammodendron, one of the main shrubs in the Tarim desert highway ecological shelterbelt, we irrigated the H. ammodendron seedlings with progressive saline groundwater (3–30 g L−1, simulation environment in the Tarim desert highway ecological shelterbelt) and investigated the diurnal variations of chlorophyll a (Chl a) fluorescence parameters, such as maximal quantum yield of photosystem II (PSII) photochemistry (Fv/Fm), quantum yield of photochemical energy conversion in PSII (YII), the apparent rate of electron transport at the PSII level (ETR), photochemical quenching coefficient (qP), non-photochemical quenching (NPQ), quantum yield of nonregulated non-photochemical energy loss in PSII (YNO) and quantum yield of regulated non-photochemical energy loss in PSII (YII), at approximately 2-h intervals. Fv/Fm with 5 g L−1 (S2) was lower than that with 2 g L−1 (S1) but a little higher than 20 g L−1 (S5), respectively. Under the low light [photosyntheticallyactive radiation (PAR) ≤ 250 μmol m−2 s−1, at 08:00, 10:00 and 20:00 h of the local time], S1 kept the lowest YII and the highest YNPQ; while under the high light (PAR ≥ 1500 μmol m−2 s−1), the YII performed S1>S2>S5, and the reverse YNPQ; under mild light (250 μmol mt-2 s−1 ≤ PAR ≤ 1500 μmol m−2 s−1), S1 remained the highest YII, no matter the light and the salinity, the similar YNO almost occurred basically. The results showed that the sand-binding plant H. ammodendron could regulate its energy-utilizing strategies. The S2 might be the most suitable salinity of the irrigation water for H. ammodendron in the Tarim desert highway ecological shelterbelt in the northwest of China.  相似文献

Allophycocyanin (APC) is a minor component of phycobiliproteins in cyanobacteria and red algae. This paper describes a simple and inexpensive extracting method for isolating APC from Spirulina (Arthrospira) platensis with high efficiency. The crude phycobiliprotein extract was pretreated by ammonium sulfate fractionation. Then, by adding hydroxylapatite into crude phycobiliprotein extract dissolved in 20 mM phosphate buffer (pH 7.0), APC was selectively adsorbed by hydroxylapatite but C-phycocyanin (C-PC) was not. The hydroxylapatite was collected and APC was extracted from the crude phycobiliprotein extract. Then, the enriched APC was washed off from the hydroxylapatite using 100 mM phosphate buffer (pH 7.0). In this simple extracting method it was easy to remove C-PC and isolate APC in large amounts. The absorbance ratio A 650/A 280 of extracted APC reached 2.0. The recovery yield was 70%, representing 4.61 mg · g−1 wet weight. The extracted APC could be further purified by a simple anion-exchange chromatography with a pH gradient from 5.6 to 4.0. The absorbance ratio A 650/A 280 of the purified APC reached 5.0, and the overall recovery yield was 43%, representing 2.83 mg · g−1 wet weight. Its purity was confirmed by native polyacrylamide gel electrophoresis (PAGE) and sodium dodecyl sulfate-PAGE.  相似文献

The effects of Aporrectodea caliginosa earthworms on both carbon dioxide (CO2) accumulation in and emissions from soil, as well as the simultaneous impact of earthworms on soil microbiological properties were investigated in a microcosm experiment carried out over 5.5 months. Concentration of CO2 in soil air was greater at a depth of 15 cm when compared with a depth of 5 cm, but varied during the season both in control and earthworm-inhabited chambers. Peaks of CO2 concentrations at both depths occurred in both treatments during August, approximately 80 days after the experiment started. Generally, the presence of earthworms increased the CO2 concentration at 15-cm depth. Larger CO2 emissions were consistently recorded in conjunction with higher amounts of CO2 in soil air when chambers were inhabited by earthworms. The total CO2 emissions during the experimental period covering 161 days were estimated at 118 g CO2-C m−2 and 99 g CO2-C m−2 from chambers with and without earthworms respectively. Moreover, the presence of earthworms increased microbial biomass in the centre and at the bottom of chambers, and enhanced both dehydrogenase activity and nitrifying enzyme activity in the soils. We suggest that the effect of earthworms on both the enhanced soil accumulation of CO2 as well as emissions of CO2 was mostly indirect, due to the impacts of earthworms on soil microbial community.  相似文献

Bacillus thuringiensis (Bt) Berliner is a promising agent for microbial control of agriculturally and medically important insects. This study aimed at searching for Bt strains encoding Cry proteins that act more efficiently against fall armyworm. Thirty Bt strains were isolated from soil samples in Pernambuco State and evaluated through bioassays. Among these, strain I4A7 was the most efficient against the fall armyworm, Spodoptera frugiperda (J. E. Smith, 1797) (Lepidoptera: Noctuidae), and thus it was characterized by biochemical sodium dodecyl sulfate–polyacrylamide gel electrophoresis (SDS–PAGE) and molecular (polymerase chain reaction (PCR) and sequencing reaction) methods. The protein pattern of this strain on a SDS–PAGE was similar to that of B. thuringiensis israelensis (Bti). Moreover, I4A7 cry DNA sequence showed high identity (99–100%) to genes cry4Aa, 4Ba, 10Aa, 11Aa, cyt1Aa and cyt2B from Bti. The toxicity of the newly isolated Bti-like strain upon S. frugiperda should be considered as this strain might be used in combination with other Bt strains, such as B. thuringiensis var. kurstaki (Btk). The dwarf pomegranate (Punica granatum L. var. nana) is a dwarf ornamental plant that has the potential to be the model plant of perennial fruit trees because it bears fruits within 1 year of seedling. We established an Agrobacterium-mediated transformation system for the dwarf pomegranate. Adventitious shoots regenerated from leaf segments were inoculated with A. tumefaciens strain EHA105 harboring the binary vector pBin19-sgfp, which contains neomycin phosphotransferase (npt II) and green fluorescent protein (gfp) gene as a selectable and visual marker, respectively. After co-cultivation, the inoculated adventitious shoots were cut into small pieces to induce regeneration, and then selected on MS medium supplemented with 0.5 μM α-naphthaleneacetic acid (NAA), 5 μM N6-benzyladenine (BA), 0.3% gellan gum, 50 mg/l kanamycin, and 10 mg/l meropenem. Putative transformed shoots were regenerated after 6–8 months of selection. PCR and PCR-Southern blot analysis revealed the integration of the transgene into the plant genome. Transformants bloomed and bore fruits within 3 months of being potted, and the inheritance of the transgene was confirmed in T1 generations. The advantage of the transformation of dwarf pomegranate was shown to be the high transformation rate. The establishment of this transformation system is invaluable for investigating fruit-tree-specific phenomena.  相似文献
