We investigated the effects that habitat variation has on the structure and dynamics of a hybrid zone between two closely related crickets in Connecticut. A collecting protocol was developed in which crickets were sampled from characteristic habitats on either side of the hybrid zone and from two distinct habitat types within the zone. Presumptive pure Gryllus pennsylvanicus were sampled from fields in northwestern Connecticut and represent “inland” populations. “Pure” Gryllus firmus were sampled from beaches along the coast and represent the “coastal” populations. Crickets from within the hybrid zone were sampled from two different soil types: the “loam” populations from loamy soils and the “sand” populations from sandy soils. Moreover, an attempt was made to identify closely adjacent sand and loam localities to determine the scale of habitat variation and its possible effects on hybrid-zone structure. In general, there was little variation in morphological traits or in allozyme and mtDNA genotype frequencies among localities from within each of the four habitat types. Between each of the closely situated sand and loam localities within the hybrid zone, however, there were very significant differences in each of these sets of markers. In addition, crickets from hybrid-zone populations were tested for reproductive isolation. The asymmetric outcome of hybrid crosses that exists across the zone (Harrison, 1983) also exists on a finer ecological scale within the zone. Thus, this hybrid zone is a mosaic of strikingly differentiated populations. The dynamics of hybrid zones with mosaic structures are discussed in contrast to the traditional clinal models. The data are also discussed in light of the semipermeable nature of species boundaries. The extent to which a species boundary is permeable varies not only from one genetic marker to the next, but also with the ecological and geographic context of species interaction.  相似文献

Studies of hybridization and introgression and, in particular, the identification of admixed individuals in natural populations benefit from the use of diagnostic genetic markers that reliably differentiate pure species from each other and their hybrid forms. Such diagnostic markers are often infrequent in the genomes of closely related species, and genomewide data facilitate their discovery. We used whole‐genome data from Illumina HiSeqS2000 sequencing of two recently diverged (600,000 years) and hybridizing, avian, sister species, the Saltmarsh (Ammodramus caudacutus) and Nelson's (A. nelsoni) Sparrow, to develop a suite of diagnostic markers for high‐resolution identification of pure and admixed individuals. We compared the microsatellite repeat regions identified in the genomes of the two species and selected a subset of 37 loci that differed between the species in repeat number. We screened these loci on 12 pure individuals of each species and report on the 34 that successfully amplified. From these, we developed a panel of the 12 most diagnostic loci, which we evaluated on 96 individuals, including individuals from both allopatric populations and sympatric individuals from the hybrid zone. Using simulations, we evaluated the power of the marker panel for accurate assignments of individuals to their appropriate pure species and hybrid genotypic classes (F1, F2, and backcrosses). The markers proved highly informative for species discrimination and had high accuracy for classifying admixed individuals into their genotypic classes. These markers will aid future investigations of introgressive hybridization in this system and aid conservation efforts aimed at monitoring and preserving pure species. Our approach is transferable to other study systems consisting of closely related and incipient species.  相似文献

The hybrid zone between eastern and western grass snakes, which has been independently hypothesized to be due to secondary contact, is investigated using univariate and multivariate analysis of a large number of independent, quantitative characters. Investigation of the patterns of geographic variation show that 5 out of 6 character systems show a congruent pattern of “stepped clinal” change across the zone. Insofar as this case is concerned, secondary contact ‘hybrid’ zones are associated with concordant changes in many character systems and “speciation” involves the origin of a large number of small differences in most character systems rather than the major reorganisation of a single system.  相似文献

Herein we report the first genetic comparison among species in the genus Eulimnadia. Multilocus genotypic patterns (using six allozyme loci) were compared for a total of 2277 clam shrimp from nine populations from Arizona and New Mexico. Seven of these populations were morphologically typed as Eulimnadia texana Packard and two were typed as Eulimnadia diversa Mattox. All populations were hermaphrodite-biased, and highly inbred (inbreeding coefficients ranging from 0.33 to 0.98). Genetic distances showed the two species to be within the range described for other arthropods. One of the two E. diversa populations appeared to be a hybrid between E. texana and E. diversa, showing electrophoretic patterns similar to both species, although morphologically, they were typed as E. diversa. A phenogram (generated using coancestry distances and a neighbor joining algorithm) placed this hybrid population half-way between these two species, and a breakdown of individuals within this hybrid population (based on allozyme scores) indicated individuals very similar to the second E. diversa population, and two groups of apparent hybrid individuals. Therefore, the distinction between these two species is questionable due to their apparent hybridization in this area of Arizona. Genetic population structuring was noted among the seven E. texana pools. Estimated migration rates were less than one migrant per generation. Even in the geographically close pools in New Mexico, which were separated by only hundreds of meters, significant sub-structuring was noted, and estimates of migration rate were less than two migrants per generation.  相似文献

The oribatid mite Carabodes reticulatus Berlese, 1913 is redescribed from typical and topotypical material. A new species, C. tyrrhenicus, widespread in Sardinia and other Tyrrhenian islands, is also described on the basis of morphological evidence. Scanning electron microscopy was used to study intra- and inter-specific variation, in order to determine diagnostic characters. The morphology-based taxonomy was tested through allozyme analysis of the two species and of two closely related species, namely C. coriaceus C. L. Koch, 1836 and C. arduinii Valle, 1955. Allozyme data are largely congruent with morphological evidence, indicating that the four entities represent well-differentiated evolutionary lineages. Molecular results show large inter-specific genetic differentiation, suggesting that these taxa arose from ancient cladogenetic events.  相似文献

Recent taxonomic revisions in the genus Diaphanosoma have made the uncertainty in species discrimination with morphological characters obvious. Therefore species characterization on a genetic basis seems to be required.Here we examined the genetic structure of two Diaphanosoma mongolianum populations and three Diaphanosoma brachyurum populations in Central Europe by allozyme electrophoresis. A genetic differentiation between both species was evident. 5 out of 9 tested loci carried diagnostic alleles. Both species differed in their habitat choice: D. mongolianum was adapted to higher trophic levels than D. brachyurum. Co-occurrence was observed in a eutrophic lake. Populations sampled from their preferred habitats showed high genotype diversities.Most genotype frequency distributions were in agreement with Hardy-Weinberg expectations and temporal (between years) changes in genotypic compositions were not common. This indicated annually occurring bouts of sexual reproduction, similar to intermittent Daphnia populations. Genetic differentiation among lakes suggested restricted gene flow.  相似文献

Two large experiments designed to detect allozyme-associated heterosis for growth rate in Drosophila melanogaster were performed. Heterosis associated with allozyme genotypes may be explained either by functional overdominance at the allozyme loci, or closely linked loci; or by genotypic correlations between allozyme loci and loci at which deleterious recessive alleles segregate. Such genotypic correlations would be favored by consanguineous mating, small effective population size, population mixing and strong natural or artificial selection. D. melanogaster is outbred, has large effective population size and there is little evidence for genotypic disequilibria. Therefore it would be unlikely to show allozyme heterosis due to genotypic correlations. In the first experiment I estimated the genotypic values of 97 replicated genotypes. In the second experiment, 500 individuals were raised in a fluctuating, stressful environment. In neither experiment was there any consistent evidence for allozyme heterosis in size or development rate, fluctuating asymmetry for size or in tendency to deviate from the population mean. In the first experiment, heterosis explained less than 5.6% of the genetic variance in growth characters. In the second, heterosis explained less than 0.1% of the phenotypic variance in growth characters. Outside of the molluscs, species which show allozyme heterosis have population structures or histories which tend to promote genotypic correlations. There is little evidence that functional overdominance is responsible for observations of allozyme-associated heterosis.  相似文献

The genus Rhoptropus consists of six species of diurnal, chiefly rupicolous, geckos that are endemic to the arid and semi-arid zones of Namibia and southern Angola. Sixteen skeletal and scalation characters and six allozyme characters analysed cladistically yield a single most parsimonious pattern of relationships: (( R. afer, R. bradfieldi ) ( R. taeniostictus (R. boultoni (R. barnardi, R. biporosus )))). Support for the placement of R. taeniostictus is tenuous because of missing allozyme data, but the remainder of the topology, especially the basal dichotomy, is well supported. Morphological and allozyme data are congruent and a phenetic analysis of genetic distances among the taxa is consistent with the phylogeny. The diversification of the genus probably took place in the Mid- to Late Tertiary and both basal and distal cladogenic events may be related to the evolution of the Namib Desert and movement of the Namib sands. Rates of evolution among the lineages of Rhoptropus appear to be unequal and R. afer , a species largely restricted to horizontal sheet rock habitats, has accumulated a large number of autapomorphic traits.  相似文献

We describe a narrow hybrid zone between the mottled sculpin (Cottus b. bairdi) and the Blue Ridge sculpin (C. caeruleomentum). Seven characters (dorsal fin rays, pectoral fin rays, caudal base band condition, male spawning coloration, and one frequency and two fixed allozyme differences) distinguish the two taxa in the hybrid zone. C. caeruleomentum and C. b. bairdi diverged in these characters in allopatry as indicated by their distribution on opposite sides of the Atlantic-Ohio divide. However, a stream capture placed these two taxa in secondary contact in Wills Creek, Potomac drainage (Atlantic slope). Allozyme data indicate the presence of post-F(1) hybrids in the zone of secondary contact. Changes in allozymes, morphology, and spawning coloration along a transect in Wills Creek reveal the hybrid zone is less than 20 river kilometers in length. Estimates of root mean square dispersal and gene flow tentatively suggest that selection is operating in the Wills Creek hybrid zone. C. b. bairdi and C. caeruleomentum are maintaining their identity in seven distinguishing characters on opposite ends of the hybrid zone revealing these two taxa are independent evolutionary lineages.  相似文献

Marked external and osteological differences exist between the related cyprinids Couesius plumbeus and Rhinichthys cataractae. C. plumbeus differs from R. cataractae in several external features, such as eye size and mouth shape, any one of which permits identification of individuals. C. plumbeus has postcleithral and nasal bones while R. cataractae does not. Many differences in bone shape also exist. Natural hybrids between the two species from Upper and Lower Kananaskis reservoirs, southwestern Alberta, are intermediate in the average of their external and osteological characters. Some characters in the hybrids, however, are so variable that they fall within the range of both parental species. An individual hybrid may resemble one parental species in one diagnostic character and the other parental species in another. The largest individuals known of the three forms from the Kananaskis system are hybrids.  相似文献

Allozyme electrophoresis, a partial nucleotide sequence of a mitochondrial gene for cytochrome oxidase I (COI) and discriminant analysis of shell morphometric characters were used to study the relationships among the Sicilian marine gastropods of the Patella genus. Allozyme and mtDNA markers unequivocally distinguished the species and were very useful markers in correctly classifying the different species when morphological characters overlapped each other. Several allozyme loci and many nucleotide positions were diagnostic of species. In contrast, the discriminant analysis of simple morphometric shell characters failed to adequately discriminate the species, suggesting that environmental factors influence colouration and morphological patterns in the Patella species. Our results underline the importance of a genetic approach, as compared to a morphological approach, in discriminating the Mediterranean Patella species.  相似文献

The shape of hybrid zones provides insight into genetic isolation between the evolutionary lineages involved: the greater the limitation to gene flow, the farther along the unimodal to bimodal continuum. We study hybrid zone modality in a group of closely related species at a variety of levels of genetic divergence. We explore the degree and the geography of hybridization in 12 transects of Triturus newts, for nine of the ten secondary contact zones in Europe, using nuclear and mitochondrial encoded genetic data and morphological characteristics. Species status is verified by examining the correlation between the three marker systems in a geographical context. At 40 allozyme loci, two transects classify as unimodal, three as bimodal and two as intermediate, while the signal for hybridization is weak or absent in five transects. One zone studied in duplicate was classified as intermediate in one region and as bimodal in another region. mtDNA introgression is frequent and extends beyond nuclear introgression in two transects. Morphology provides additional evidence for hybridity, including one transect for which the signal of nuclear gene flow is weak. Compared to simulations allowing panmixia, the observed allozyme transitions at contact zones show a deficit of backcrossing to various degrees. Over all transects, there is a weak negative relationship between the level of hybridization and allozyme genetic distance for species pairs, consistent with Bateson–Dobzhansky–Muller effects. This observation, based upon highly comparable data for a single genus, supports conclusions derived from analyses over a wide variety of other taxa. © 2014 The Authors. The distribution of multilocus genotypes found within a natural hybrid zone is determined by the sample of genotypes present when the hybrid zone first formed, by subsequent patterns of genetic exchange between the hybridizing taxa, and by drift and selection within each of the hybrid zone populations. We have used anonymous nuclear DNA restriction fragment polymorphisms (RFLPs) to characterize the array of multilocus genotypes present within a well-studied hybrid zone between two eastern North American field crickets, Gryllus pennsylvanicus and Gryllus firmus. These crickets hybridize along a zone of contact that extends from New England to Virginia. Previous studies have shown that both premating and postmating barriers exist between the two cricket species, but the absence of diagnostic morphological and allozyme markers has made it difficult to assess the consequences of these barriers for genetic exchange. Analyses based on four diagnostic anonymous nuclear markers indicate that hybrid zone populations in Connecticut contain few F1 hybrids, and that nonrandom associations persist among nuclear gene markers, between nuclear and cytoplasmic markers, and between molecular markers and morphology. Field cricket populations within the hybrid zone are not “hybrid swarms” but consist primarily of crickets that are very much like one or the other of the parental species. Despite ample opportunity for genetic exchange and evidence for introgression at some loci, the two species remain quite distinct. Such a pattern appears to be characteristic of many natural hybrid zones.  相似文献

Hybrid zones provide interesting systems to study genetic and ecological interaction between different species. The correct identification of hybrids is necessary to understand the evolutionary process involved in hybridization. An oak species complex occurring in Mexico formed by two parental species, Quercus crassifolia H. & B. and Q. crassipes H. & B., and their putative hybrid species, Q. dysophylla, was analyzed with molecular markers (random amplified polymorphic DNA [RAPDs]) and morphological tools in seven hybrid zones (10 trees per taxa in each hybrid zone) and two pure sites for each parental species (20 trees per site). We tested whether geographic proximity of hybrid plants to the allopatric site of a parental species increases its morphological and genetic similarity with its parent. Seventeen morphological traits were measured in 8700 leaves from 290 trees. Total DNA of 250 individuals was analyzed with six diagnostic RAPD primers. Quercus crassifolia differed significantly from Q. crassipes in all the examined characters. Molecular markers and morphological characters were highly coincident and support the hypothesis of hybridization in this complex, although both species remain distinct in mixed stands. Clusters and a hybrid index (for molecular and morphological data) showed that individuals from the same parental species were more similar among themselves than to putative hybrids, indicating occasional hybridization with segregation in hybrid types or backcrossing to parents. Evidence does not indicate a unidirectional pattern of gene flow.  相似文献

Hybridization phenomena in anurans have traditionally been studied through morphological comparisons, under the assumption that various hybrids (e.g., F1‘s, backcrosses) are predictably intermediate to parental species. We critically evaluate this assumption by examining morphology in genetically categorized hybrids between the treefrogs Hyla cinerea and H. gratiosa. A total of 202 frogs from a hybridizing population in Alabama were assayed for allozyme and mitochondrial DNA genotype and for a large suite of osteological characters. Discriminant analyses demonstrated distinct morphological separation between the genetically “pure” parental species. Morphometric analyses of genetically identified hybrids showed: 1) an extreme range of phenotypic expression within F1 and backcross classes, and 2) no apparent directional parental bias on the F1 phenotype. Had morphology alone been used as a guide, over 40 percent of the individuals with known hybrid ancestry would have been misclassified as “pure” parental species, and about 25 percent of the backcross individuals would not have been distinguished from F1‘s. These results exemplify the utility of joint comparisons of morphology and genotypic constitution in studies of natural hybridization, and they emphasize the limitations inherent in describing hybrid classes solely by morphological criteria.  相似文献

The stability of complex patterns of geographic variation was investigated by assessing the congruence between multivariate ordinations derived from randomly chosen real characters. Two series of populations were analysed representing two situations with complex patterns of geographic variation. The first, a ‘Eurasian’ series of populations, showed a strongly structured hierarchical pattern, the second, an ‘eastern’ series of populations, showed a more subtle complex pattern of smooth clines and steps. The characters were selected from a total of 81 (Eurasian) or 61 (eastern) within-population independent characters from six different systems. The congruence between ordinations of the geographical populations was measured by the rotational fit statistic, R2. Three procedures were used to compare ordinations based on from two to up to 80 characters randomly chosen to give: A, completely independent character sets; B, subsets compared to the total set; and C, potentially overlapping sets. All three procedures showed that congruence between the ordinations was asymptotic in relation to character number. This relationship was described by one of two mathematical models (procedure B did not result in a hyperbolic model as found with simple patterns of geographic variation). Generally speaking, once a sufficient number of characters are used, the complex patterns of geographic variation are stable, reliable and predictive and not substantially influenced by character choice. The strongly structured hierarchical pattern required 15 or so characters to achieve reliability whilst the more subtle patterns required 20 or so characters. However, the addition of further characters does improve reliability in both cases. The greater percentage of variance portrayed by three-dimensional ordinations compared to two-dimensional ordinations is achieved at the cost of lower congruence when a sufficient number of characters are used. If case studies of geographic variation were to adopt these procedures (preferably using completely independent character sets; procedure A) the reliability of their results would be indicated).  相似文献

Species in cryptic complexes tend to be very difficult, if not impossible, to identify using morphological characters. One such complex is the spruce budworm (Choristoneura fumiferana Clemens, 1865) species group, an economically important group of Nearctic forest pests. Morphological, ecological, behavioural and genetic characters have been studied to try to understand the taxonomy of this group better, but diagnostic character states differ in frequency rather than being complete replacements between each species. We used mitochondrial DNA (mtDNA), together with a new morphology‐based character system that focuses on forewing colour components, to determine if one or a combination of character sources can be used for species diagnoses within the spruce budworm complex. We characterized 47 forewing morphometric measurements and sequenced a 470 bp region of cytochrome c oxidase I mtDNA for 111 ingroup individuals comprising five taxa within the complex. Larval host association and coloration or adult pheromone attraction were used as the prior method for grouping individuals. Our results showed that linear discriminant analysis of morphometric wing characters gave unique clusters for all species on the first and second canonical axes, except for a partial overlap between C. fumiferana and C. biennis, which are not sympatric in nature. In contrast, mtDNA distinguished C. fumiferana, C. pinus pinus Freeman, 1953 and a group of western species, but the three western species (C. occidentalis Freeman, 1967 , C. biennis Freeman, 1967 and C. lambertiana Busck, 1915) shared mtDNA haplotypes. On the basis of the linear discriminant analysis of the combined character set, this study supports the application of both morphology and mtDNA within a framework of integrative taxonomy as the most accurate method for species identification. Furthermore, it demonstrates the utility of quantitative colour analysis, which may be particularly helpful for groups in which colour characters are difficult to divide into discrete units due to intergrading hues.  相似文献
