Ceratotheca triloba (Bernh.) Hook.f. commonly known as an African foxglove is an indigenous plant which occurs in most parts of South Africa. The species is commonly consumed as a leafy vegetable and utilized for its medicinal properties. Although the high nutritional value of the species and medicinal properties are well documented, information related to critical aspect of cultivation is currently limited. Therefore, this study aimed to evaluate the effect of vermicompost leachate (VCL) on growth, nutritional, phytochemical, and antioxidant levels in C. triloba at different growth stages under nutrient-deficient conditions. After in vitro germination, seedlings were grown in the greenhouse for 2 and 4 months under nitrogen (–N); phosphorus (–P); and potassium (–K) deficiency conditions, and were treated with VCL. Vermicompost leachate did not improve the growth of C. triloba plants under the nutrient-deficient conditions. Although –N-deficient plants with or without VCL caused a decline in growth parameters, they significantly enhanced phytochemicals in 2-month-old plants. In most cases, the application of VCL to –P- and –K-deficient plants improved the photosynthetic pigments, protein, and phenolic, as well as flavonoid accumulation. Harvesting time was also found to play a crucial role in the accumulation of evaluated parameters in nutrient-deprived plants. From these findings, it can be deduced that VCL has a potential to minimize the effect of nutrient deficiency especially under –P and –K deficiency in C. triloba plants.  相似文献

To determine the effects of vermicompost leachate (VCL) on resistance to salt stress in plants, young tomato seedlings (Solanum lycopersicum, cv. Ailsa Craig) were exposed to salinity (150 mM NaCl addition to nutrient solution) for 7 days after or during 6 mL L??1 VCL application. Salt stress significantly decreased leaf fresh and dry weights, reduced leaf water content, significantly increased root and leaf Na+ concentrations, and decreased K+ concentrations. Salt stress decreased stomatal conductance (gs), net photosynthesis (A), instantaneous transpiration (E), maximal efficiency of PSII photochemistry in the dark-adapted state (Fv/Fm), photochemical quenching (qP), and actual PSII photochemical efficiency (ΦPSII). VCL applied during salt stress increased leaf fresh weight and gs, but did not reduce leaf osmotic potential, despite increased proline content in salt-treated plants. VCL reduced Na+ concentrations in leaves (by 21.4%), but increased them in roots (by 16.9%). VCL pre-treatment followed by salt stress was more efficient than VCL concomitant to salt stress, since VCL pre-treatment provided the greatest osmotic adjustment recorded, with maintenance of net photosynthesis and K+/Na+ ratios following salt stress. VCL pre-treatment also led to the highest proline content in leaves (50 µmol g??1 FW) and the highest sugar content in roots (9.2 µmol g??1 FW). Fluorescence-related parameters confirmed that VCL pre-treatment of salt-stressed plants showed higher PSII stability and efficiency compared to plants under concomitant VCL and salt stress. Therefore, VCL represents an efficient protective agent for improvement of salt-stress resistance in tomato.  相似文献

Strategies to globally protect biological diversity are often hampered by an insufficient ecological knowledge about target species. This also applies to Gagea spathacea (Liliaceae), a ‘vulnerable’ woodland spring geophyte with a distribution largely restricted to the lowlands of Central Europe. We studied whether the species’ linkage to highly fertile forest soils is related to its high nitrogen (N) demands during its short developmental cycle. We hypothesized that the species exhibits a highly efficient N (re)cycling strategy, characterized by efficient resorption of N from the leaves and reallocation to bulbs at the end of the growing season. To test this assumption, we conducted a 15N tracer experiment and quantified 15N flows between soil, leaves, bulbs, and roots. Our findings support our hypothesis that G. spathacea is exceptionally efficient in recycling N, shown by the resorption of 68% of leaf N and its reallocation to bulbs at the end of the growing season. After 6 weeks of growth the plant showed a distinct shift in its N metabolism: The C:N ratio of leaves strongly increased and those of bulbs decreased, leaf 15N enrichment and recovery started to decrease, while total plant 15N recovery remained constant, indicating no further N uptake from the soil. Leaf N reallocation to bulbs was accompanied by a twofold increase of the bulbs’ biomass. Because of the stenoecious behaviour of G. spathacea, a careful protection and sustainable management of G. spathacea forest habitats is necessary, particularly in its Central European core area.  相似文献

The vulnerability of gastropods to their predators varies with life history traits such as morphology, body size, behavior, and growth rates as well as predator size. A recent study suggested that the invasive apple snail, Pomacea maculata, was considerably more vulnerable to crayfish predators than the native Florida apple snail, P. paludosa. The difference was hypothesized to be caused by the relatively small hatchling size of P. maculata. To test this hypothesis, we conducted a series of feeding assays designed to quantify maximum feeding rates and selective foraging of crayfish on apple snails. The rate at which crayfish killed individual P. maculata (i.e., kill rates) decreased with snail size, and kill rates on both species increased with crayfish size. Kill rates on juvenile P. maculata were higher than kill rates on size-matched hatchling P. paludosa, and crayfish fed selectively on P. maculata when offered mixed groups of size-matched snails. Further analyses revealed that hatchling P. paludosa possess shells 1.8× heavier than size-matched P. maculata suggesting differences in vulnerability to crayfish were consistent with interspecific differences in shell defenses. Differences in hatchling size and defensive traits in combination make crayfish kill rates on hatchling P. maculata approximately 15.4× faster than on hatchling P. paludosa, but the relative contribution of hatchling size to differences in apple snail vulnerability was >3× greater than the contribution of defensive traits.  相似文献

The purpose of this study was to determine the concentration of some metals (Al, Cd, Co, Cr, Cu, Fe, Mn, Ni, Pb, Zn, Ca and Mg) in soil of serpentine and limestone sites, their bioaccumulation and impact on some biochemical parameters in T. luanica, T. kosovarica and T. albanica plants. T. kosovarica and T. albanica grows in serpentine soil, while T. luanica grow in limestone soil. The research showed that concentrations of Cd, Co, Cr, Fe, Mn and Ni were significantly higher at serpentine soil sites in comparison with limestone sites, while concentrations of Pb, Cd, Co and Cr in bulbs, leaves and seeds were under the limit of detection. The concentration of Ni in plant samples of T. kosovarica was significantly higher in comparison with its concentration in T. albanica, but it was under the limit of detection in T. luanica. Moreover, concentrations of Al and Fe in leaves of T. kosovarica and T. albanica were higher in comparison with T. luanica. The concentration of Mg was significantly higher in T. kosovarica and T. albanica than in T. luanica. The δ-aminolevulinic acid dehydratase activity, malondialdehyde and glutathione contents in leaves of T. luanica were higher in comparison with T. kosovarica and T. albanica. In addition, the amounts of total chlorophyll and δ-aminolevulinic acid (ALA) in leaves of T. albanica were higher in comparison with T. kosovarica and T. luanica. Our findings show that target organs of metal accumulation in three Tulip species appears to be leaves?>?seeds?>?bulbs, while the biochemical parameters show that limestone sites represent a less stressful habitat for growing these plant species in comparison with serpentine sites.  相似文献

Changes in the levels of secondary compounds can trigger plant defenses. To identify phenolic compounds induced by Bemisia tabaci Middle East-Asia Minor 1 (MEAM1) in tobacco (Nicotiana tobacco L.), the content changes of 11 phenolic compounds in plants infested by B. tabaci MEAM1 or Trialeurodes vaporariorum were compared. The chlorogenic acid, catechin, caffeic acid, p-coumaric acid, rutin, and ferulic acid contents in B. tabaci MEAM1-infested tobacco plants increased significantly, having temporal and spatial effects, compared with uninfested control and T. vaporariorum infested plants. The contents were 4.10, 2.84, 2.25, 3.81, 1.46, and 1.91 times higher, respectively, than those in the control. However, a T. vaporariorum nymphal infestation just caused smaller chlorogenic acid, catechin, caffeic acid, and rutin contents increase, which were 2.33, 2.13, 1.59, and 3.19 times higher, respectively, than those in the control. In B. tabaci MEAM1 third-instar nymph-infested plants, chlorogenic acid, catechin, caffeic acid, and rutin increased more significantly in systemic than in local leaves. Salicylate-deficient plants inhibited the induction of the content of 10 phenolic compounds, but not caffeic acid, after a B. tabaci MEAM1 nymphal infestation. Thus, the elevated levels of phenolic compounds induced by B. tabaci MEAM1 were correlated with the salicylic acid signaling pathway and induced the responses of defense-related phenolic compounds.  相似文献

Bemisia tabaci Middle East-Asia Minor 1 (MEAM1) infestation adversely affected photosynthesis of host plants. In the current study, chlorophyll a fluorescence was measured to determine the effects of MEAM1 nymph infestation of tobacco local and systemic leaves on energy metabolism and electron transport of photosystemII(PSII). The results showed that the density of PSII reaction centres per excited cross section (CS) (RC/CS) of infested and systemic leaves was reduced at 14 and 20 days. In systemic leaves, the number of PSII closed reaction centres (1-qP) increased significantly at 14 and 20 days. Absorption flux per QA? reducing PSII reaction centre (RC) (ABS/RC), trapped energy flux per RC (TRo/RC), and electron transport per RC (ETo/RC) of infested and systemic leaves increased with MEAM1 nymph infestation. The most obvious increase in absorption flux per CS (ABS/CSo) and trapped energy flux per CS (TRo/CSo) of infested and systemic leaves occurred at 14 days. MEAM1 nymph infestation significantly reduced the energy required for PSII QA to be completely reduced (Sm) in tobacco leaves. These results suggested that MEAM1 nymph infestation caused changes in light-harvesting antenna system and deactivation of the reaction centre, resulting in the reduction of photons absorbed by reaction centres per unit area. MEAM1 nymph infestation, particularly the 3rd instar nymphs, decreased light utilization ability and increased excess excitation energy in tobacco leaves. With MEAM1 nymph infestation, the relative electron transport capacity of the entire electron transport chain decreased, and more light energy was used to reduce QA.  相似文献

Background and aims

Invasive weeds may exert negative impact on other plant species and soil processes. The observed negative impact of an invasive weed species may be driven by allelopathy or nutrient availability.

Methodology

Sorghum halepense is one of the worst invasive weeds in crop fields. We quantified the species richness in the S. halepense-invaded communities and communities not yet invaded by the weed. Sorghum soil and no-sorghum soil were analysed for total phenolics, microbial activity, available nitrogen (N) and organic carbon. Manipulative experiments were carried out to understand the interference potential of S. halepense. Soil was amended with root or shoot leachate of S. halepense, and its impact on plant growth and soil properties was studied.

Results

Out of four S. halepense-sites, lower plant species richness was observed in one site compared to uninvaded sites. S. halepense-invaded soil had higher levels of total phenolics and lower levels of available N. Higher inhibition in the root growth of Brassica juncea or Bidens pilosa was observed in root leachate-amended soil than shoot leachate-amended soil. Shoot leachate-amended soil had higher levels of total phenolics and available N than root leachate-amended soils. Significant reduction in the available N was observed in soil amended with root leachate. Significant decline in the total phenolics over a period of time was observed in soil amended with root leachate or shoot leachate of S. halepense. Higher CO2 release was observed 24 h after amending soil with root leachate or shoot leachate of S. halepense.

Conclusions

Sorghum halepense interference potential in its soil is likely due to lower levels of available N. Greater reduction in root dry weight of assay species in root leachate amended soil compared to shoot leachate amended soil was likely due to lower levels of available N in root leachate-amended soil. Relative interference potential of both root and shoot leachates or extracts should be evaluated in allelopathy bioassays and further experiments should be designed to distinguish the role of allelochemicals and nutrient availability in plant growth inhibition.

The effects of plant growth regulators (PGRs) and organic elicitors (OEs) on in vitro propagation of Eucomis autumnalis was established. Three-year-old ex vitro grown plants from organogenesis of E. autumnalis and somatic embryogenesis (previously reported protocol) of Drimia robusta were investigated for antibacterial activity. In vitro propagation from leaf explants of E. autumnalis was established using different PGRs and OE treatments for mass propagation, biomass production and bioactivity analysis to supplement the use of wild plant material. Prolific shoots (16.0?±?0.94 shoots per explant) were obtained with MS (Murashige and Skoog in Physiol Plant 15:473–497, 1962) medium containing 100 mg l?1 haemoglobin (HB), 10 µM benzyladenine (BA) and 2 µM naphthaleneacetic acid (NAA). The shoots were rooted effectively with a combination of 2.5 µM indole-3-acetic acid and 5.0 µM indole-3-butyric acid. The plantlets were successfully acclimatized in a vermiculite-soil mixture (1:1 v/v) in the greenhouse. Three-year-old ex vitro-grown E. autumnalis and D. robusta plants derived via organogenesis and somatic embryogenesis respectively exhibited antibacterial activity and varied with PGR and OE treatments, plant parts and bacteria. The leaves of E. autumnalis ex vitro-derived from a combination of HB, BA and NAA followed by the individual treatments of BA and HB gave the best antibacterial activities (<?1 mg ml?1: minimum inhibitory concentration from 0.098 to 0.78 mg ml?1) against all tested pathogenic bacteria (Bacillus subtilis, Enterococcus faecalis, Micrococcus luteus, Staphylococcus aureus, Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa). The bulbs of D. robusta ex vitro-derived from solid culture with 10 µM picloram, 1 µM thidiazuron and 20 µM glutamine exhibited good antibacterial activity against E. faecalis, M. luteus and S. aureus when compared with other treatments and mother plants. The ex vitro-grown E. autumnalis and D. robusta biomass produced with PGRs along with OE treatments confirmed a good potent bioresource and can be used as antibacterial agents. The in vitro plant regeneration of E. autumnalis and D. robusta protocols and ex vitro plants could be used for conservation strategies, bioactivity and traditional medicinal use.  相似文献

Biologically active substances and antioxidant activity of extracts from leaves and inflorescences of nine representatives of the genus Spiraea L. growing on the territory of the Far East of Russia were investigated. Widespread species of the genus Spiraea (S. salicifolia, S. media var. media, S. betulifolia and S. ussuriensis subsp. ussuriensis) have the highest levels of biologically active substances. The inflorescences of spiraeas there contain more flavonols (up to 3.9%), oxycinnamic acids (up to 1.2%), catechins (up to 5.7%) and saponins (up to 5.1%) compared to their leaves, and there are more tannins (up to 11.6%) in the leaves. Among the Far Eastern representatives of the genus Spiraea, S. betulifolia and S. beauverdiana (section Calospira), S. humilis and S. salicifolia (section Spiraria), S. pubescens and S. media var. media (section Chamaedryon) are promising antioxidants. Plants of the genus Spiraea probably contain water-soluble antioxidant compounds of phenolic type, because the antioxidant activity of aqueous extracts in the leaves and inflorescences of spiraeas is higher (0.16–2.79 mg/g) than that of water-alcoholic compounds (0.06–2.54 mg/g). The antioxidant activity in the leaves of spiraeas is generally higher than that in the inflorescence. A reliable positive correlation is observed between the antioxidant activity of aqueous extracts from the organs of spiraeas and a content of oxycinnamic acids.  相似文献

Present study was aimed to select a suitable Trichoderma isolate as candidate antagonist based on its efficacy in producing cell wall degrading enzymes (CWDEs), its mycoparasitism activity and expression of related genes against the red rot pathogen caused by Colletotrichum falcatum in sugarcane. For which, six different isolates of Trichoderma selected from our earlier studies (T. harzianum, T. asperullum) were evaluated based on their capability in releasing cell wall degrading enzymes individually and during antagonism with C. falcatum in dual plate. Amongst T. harzianum (T20) exhibited the greatest mycoparasitic potential against the C. falcatum, by producing higher concentration of  CWDEs viz., chitinase and β-1, 3-glucanase, slightly lower amounts of cellulase and protease with significant reduction in polygalacturonase produced by pathogen. Further microscopic observation on interaction of C. falcatum with the selected isolate of T. harzianum (T20) exhibited the mycoparasitic activity of antagonist over pathogen in dual culture and inhibition of C. falcatum pathogenesis in detached sugarcane leaves. In addition, expression pattern of eight genes coding various enzymes involved in mycoparasitism by T. harzianum over C. falcatum were analyzed using qRT-PCR in vitro and on sugarcane leaves. In in vitro interactions, five genes of  cell wall degrading enzymes viz., chitinase (chit33), endochitinase (endo42), β-1, 3-glucanase (glu), exochitinase 1 (exc1), exochitinase 2 (exc2), were upregulated during and after contact as compared to before contact, while three genes related with proteases such as alkaline proteinase (prb1), trypsin-like protease (Pra1), subtilin-like serine protease (ssp), genes were upregulated during the contact with C. falcatum and slightly down regulated after contact. In detached leaves, seven genes were potentially upregulated except subtilin-like serine protease, which was down regulated during interaction of C. falcatum and T. harzianum as compared to T. harzianum inoculation alone. All these biochemical and molecular results confirm the efficacy of T. harzianum (T20) against C. falcatum and justify the right selection of candidate antagonist for our further studies on identification of antifungal genes/proteins against C. falcatum in sugarcane.  相似文献

Alstonia scholaris (Dr C. Alston, 1685–1760) (Family Apocynaceae) (Chattim tree), commonly known as devil tree, is an evergreen tropical tree. The tree is native to India and also found in Sri Lanka, Southern China, throughout Malaysia to northern Australia. This plant is seriously damaged by formation of tumor like galls across the Kolkata city,West Bengal which affects its ornamental and medicinal value. Gall is formed by ovipositing adults of Pseudophacopteron alstonium Yang et Li 1983 (Hemiptera: Psyllidae: Phacopteronidae) and results in destruction of host plant. The nymphal stage undergoes moulting through first instar to third instar to reach the adult within galls. It is observed that highly infested leaves can bear 60–80 galls. The gallmaker Pseudophacopteron sp. stresses the host organ, and the host counters it with physiological activities supplemented by newly differentiated tissues. In infested leaves, chlorophyll and carbohydrate contents decreased sequentially with the age of the gall. There were no significant changes in protein and total amino acid content in gall tissue. But total lipid content was highest in mature galled leaves. Increased phenolic content after psylloid herbivory, which exerted oxidative stress on the host plants, was observed in gall infested leaves as compared to fresh ungalled leaves of Alstonia scholaris. Moisture content was highest in ungalled healthy leaves than the young galled, mature galled and perforated galled leaves.  相似文献

Species of Alternaria are serious plant pathogens, causing major losses on a wide range of crops. Leaf blight symptoms were observed on tomato leaves, and samples were collected from various regions. Isolation was done from symptomatic tomato leaves, and 15 representatives were selected from a collection of 65 isolates of Alternaria species. The virulence of Alternaria isolates was investigated on detached leaves (DL) and whole plants of tomato cv. Super strain B. A phylogenetic analysis was performed based on three partial gene regions, the glyceraldehyde-3-phosphate dehydrogenase (GAPDH), the RNA polymerase second largest subunit (RPB2) and the Alternaria major allergen gene (Alt a 1). The potentiality of Alternaria isolates to produce toxins was also investigated on the basis of thin-layer chromatography (TLC). Our investigations revealed that Alternaria isolates showed different levels of virulence either on tomato plants or DL. Based on the phylogeny of three genes, Alternaria isolates encompassed two species of small-spored morphospecies: A. alternata (14 isolates) and A. arborescens (single isolate). The produced toxins varied among Alternaria isolates with tenuazonic acid (TeA) being the most abundant mycotoxin produced by most isolates. This study highlighted on other Alternaria species in Egypt that might represent a serious concern for tomato producers as causal agents of leaf blight over other species, i.e. A. solani.  相似文献

The increase of soil-borne pathogens induced by phenolic acids that accumulate in continuous cropping soil reduces the yield and quality of crops. The aims of this study were to investigate (i) the biological control of Fusarium solani, in soil enriched with phenolic acids, by the inoculation of the endophytic fungus Phomopsis liquidambari, and (ii) the biocontrol mechanisms involved. Inoculation of P. liquidambari significantly inhibited the reproduction of F. solani. The prompt degradation of soil phenolic acids by P. liquidambari was determined, but no direct antagonism relationship was observed between P. liquidambari and F. solani, implying the alleviated stimulation of phenolic acids was a major factor in controlling F. solani. Moreover, the presence of glucose did not significantly impact the biocontrol function of P. liquidambari, and P. liquidambari inoculation significantly alleviated disease severity of peanut. Therefore, P. liquidambari could be an effective means to control F. solani in phenolic acids-rich continuous cropping soils.  相似文献
