The effects of GA3, 24-epibrassinolide (EBL), and their combination on morphogenesis of Arabidopsis thaliana (L.) Heynh seven-day-old seedlings were studied. Four plant lines were analyzed: wild type Ler and ga4-1 mutant, belonging to the Landsberg erecta ecotype and wild type Col and det2 mutant, both of the Columbia ecotype. In ga4-1 and det2, GA4/1-and brassinosteroid-deficient mutants, the highest hypocotyl growth response to the lack of hormones was noted. The cotyledon shape and size were dependent on EBL, and the root length was both GA3-and EBL-regulated, indicating organ specificities in the responses to these hormones. Simultaneous treatment of dark-grown plants with GA3 and EBL exerted an additive stimulatory effect on the root growth of det2, reduced the inhibitory effect of EBL on hypocotyl elongation of ga4-1, and enhanced the effect of EBL on hypocotyl and cotyledon elongation of det2.  相似文献

The role of gibberellins in plant morphology under selective light was studied. A comparison of the effects of green and blue light on growth, development, and hormonal balance was performed for two Arabidopsis thaliana (L.) Heynh. (ecotype Landsberg erecta lines: wild type Ler and its ga4-1 mutant with suppressed GA4/1 synthesis. The absence of active GA4/1 from ga4-1 mutant determined its retarded growth, slowed passing through developmental phases, suppressed apical dominance, and reduced seed productivity. The retarded growth and development of the mutant was related to changed hormonal balance in them as compared to wild-type line: IAA content and the IAA/ABA ratio were declined, zeatin riboside and ABA accumulated. Green light retarded stem elongation and branching, reduced leaf specific surface density and plant seed productivity, and retarded plant transition to reproduction to a greater degree at GA4/1 deficit (ga4-1) than at its normal content (Ler).  相似文献

We studied the role of cryptochrome 1 (CRY1) and phytochromes in the photomorphogenetic responses of plants to the middle-wavelength region of photosynthetically active radiation. A comparison was performed of green light (GL) action on growth, GA activity and IAA and ABA contents during seedling deetiolation of two Arabidopsis thaliana (L.) Heynh lines of Landsnerg erecta ecotype (wild type Ler and mutant hy4) and of Phaseolus vulgaris L. It was shown that a growth responses of Ler hypocotyls to GL of 515 nm and Ler cotyledons to GL of 542 nm were weaker than those of the hy4 mutant defected in the CRY1 synthesis. Far-red light (730 nm) neutralized the effect of GL (533 nm) on the phytohormone balance in the primary kidney bean leaves. The data obtained permit a supposition that plants possess several photoregulatory systems functioning under GL of higher (515 nm) and lower emission energy (542–553 nm). A possible operation of GL receptors, other than cryptochrome 1 and phytochromes, is discussed.  相似文献

Auxin and two phytochrome-interacting factors, PHYTOCHROME-INTERACTING FACTOR4 (PIF4) and PIF5, play crucial roles in the enhancement of hypocotyl elongation in transgenic Arabidopsis thaliana plants that overproduce LOV KELCH PROTEIN2 (LKP2).

Abstract

LOV KELCH PROTEIN2 (LKP2) is a positive regulator of hypocotyl elongation under white light in Arabidopsis thaliana. In this study, using microarray analysis, we compared the gene expression profiles of hypocotyls of wild-type Arabidopsis (Columbia accession), a transgenic line that produces green fluorescent protein (GFP), and two lines that produce GFP-tagged LKP2 (GFP-LKP2). We found that, in GFP-LKP2 hypocotyls, 775 genes were up-regulated, including 36 auxin-responsive genes, such as 27 SMALL AUXIN UP RNA (SAUR) and 6 AUXIN/INDOLE-3-ACETIC ACID (AUX/IAA) genes, and 21 genes involved in responses to red or far-red light, including PHYTOCHROME-INTERACTING FACTOR4 (PIF4) and PIF5; and 725 genes were down-regulated, including 15 flavonoid biosynthesis genes. Hypocotyls of GFP-LKP2 seedlings, but not cotyledons or roots, contained a higher level of indole-3-acetic acid (IAA) than those of control seedlings. Auxin inhibitors reduced the enhancement of hypocotyl elongation in GFP-LKP2 seedlings by inhibiting the increase in cortical cell number and elongation of the epidermal and cortical cells. The enhancement of hypocotyl elongation was completely suppressed in progeny of the crosses between GFP-LKP2 lines and dominant gain-of-function auxin-resistant mutants (axr2-1 and axr3-1) or loss-of-function mutants pif4, pif5, and pif4 pif5. Our results suggest that the enhancement of hypocotyl elongation in GFP-LKP2 seedlings is due to the elevated level of IAA and to the up-regulated expression of PIF4 and PIF5 in hypocotyls.

A homologue of the Arabidopsis gene FUSCA3 (FUS3), isolated from the protocorm-like body (PLB) of Rosa canina and designated RcFUS3, encodes 331 amino acid residues. It was shown that RcFUS3 is specifically expressed in the PLB of R. canina and that its subcellular localization is in the nucleus. The Arabidopsis fus3-3 mutant phenotype could be rescued by over-expression of RcFUS3, suggesting that RcFUS3 has a function similar to that of Arabidopsis FUS3. Over-expression of RcFUS3 in wild type Arabidopsis resulted in a decrease in endogenous GA and CTK levels, an increase in ABA and IAA contents, starch grain accumulation in the cotyledon and hypocotyl, failure of cotyledon extension and abnormal elongation of the hypocotyl, abnormal stomatal and pavement cells, an increase in branch numbers, prolongation of the growth cycle, and morphological changes in floral organs. Interestingly, over-expression of RcFUS3 in homozygous form resulted in premature degradation of the tapetum, indehiscent anthers and hypogenetic stamens, causing complete male sterility in Arabidopsis; this is the first observation that over-expression of a gene (RcFUS3) homologous to FUS3 can lead to male sterility and starch grain accumulation in Arabidopsis.  相似文献

Abstract

Casein kinase 2 (CK2) is a conserved tetrameric kinase composed of two α and two β subunits. The inhibition of CK2 activity usually results in severe developmental deficiency. Four genes (CKA1CKA4) encode CK2 α subunit in Arabidopsis. Single mutations of CKA1, CKA2, and CKA3 do not affect the normal growth of Arabidopsis, while the cka1 cka2 cka3 triple mutants are defective in cotyledon and hypocotyl growth, lateral root development, and flowering. The inhibition of CKA4 expression in cka1 cka2 cka3 background further reduces the number of lateral roots and delays the flowering time. Here, we report the characterization of a novel knockout mutant of CKA4, which exhibits various developmental defects including reduced primary root and hypocotyl elongation, increased lateral root density, delayed cotyledon expansion, retarded rosette leaf initiation and growth, and late flowering. The examination of the cellular basis for abnormal root development of this mutant revealed reduced root meristem cells with enhanced RETINOBLASTOMA-RELATED (RBR) expression that promotes cell differentiation in root meristem. Moreover, this cka4-2 mutant accumulates higher anthocyanin in the aerial part and shows an increased expression of anthocyanin biosynthetic genes, suggesting a novel role of CK2 in modulating anthocyanin biosynthesis. In addition, the complementation test using primary root elongation assay as a sample confirms that the changed phenotypes of this cka4-2 mutant are due to the lack of CKA4. Taken together, this study reveals an essential role of CK2 α4 subunit in multiple developmental processes in Arabidopsis.

Hormonal regulation and symbiotic relationships provide benefits for plants to overcome stress conditions. The aim of this study was to elucidate the effects of exogenous methyl jasmonate (MeJA) application on root hydraulic conductivity (L) of Phaseolus vulgaris plants which established arbuscular mycorrhizal (AM) symbiosis under two water regimes (well-watered and drought conditions). The variation in endogenous contents of several hormones (MeJA, JA, abscisic acid (ABA), indol-3-acetic acid (IAA), salicylic acid (SA)) and the changes in aquaporin gene expression, protein abundance and phosphorylation state were analyzed. AM symbiosis decreased L under well-watered conditions, which was partially reverted by the MeJA treatment, apparently by a drop in root IAA contents. Also, AM symbiosis and MeJA prevented inhibition of L under drought conditions, most probably by a reduction in root SA contents. Additionally, the gene expression of two fungal aquaporins was upregulated under drought conditions, independently of the MeJA treatment. Plant aquaporin gene expression could not explain the behaviour of L. Conversely, evidence was found for the control of L by phosphorylation of aquaporins. Hence, MeJA addition modified the response of L to both AM symbiosis and drought, presumably by regulating the root contents of IAA and SA and the phosphorylation state of aquaporins.  相似文献

The aim of the present study was to establish a regeneration system via de novo organogenesis from different types of non-meristematic explants of Passiflora cristalina. Leaf, hypocotyl, root segments, cotyledons, and endosperm of P. cristalina seeds were inoculated in Murashige and Skoog (MS)-basal medium, supplemented with different concentrations of 6-Benzyladenine (BA), Thidiazuron (TDZ), or Kinetin (KIN). BA was found to be the most efficient cytokinin in induction of de novo organogenesis from most the explants used in the study. The highest frequencies of adventitious bud formation in the hypocotyl and cotyledon explants were observed in medium supplemented with 1.0 mg L?1 BA. For leaf and endosperm segments, the best concentration was 2.0 mg L?1 BA; while for root segments, the highest mean values were observed with 1.0 mg L?1 KIN. The different morphogenetic responses obtained from each explant source were characterized using light microscopy. P. cristalina revealed a remarkable organogenic potential, with superior production of adventitious shoots compared with the other Passiflora species evaluated elsewhere. These results will be helpful to establish a reproducible and reliable micropropagation protocol, as well as to implement conservationist and biotechnological-based genetic breeding strategies for this wild Passiflora species.  相似文献

Brassinosteroids (BRs) are a group of plant hormones and the bioactive BR, brassinolide (BL), is causally implicated in promoting cell elongation and cell proliferation. In Arabidopsis, the biosynthesis of BL is essential for hypocotyl etiolation in the dark, and application of bioactive BRs can promote both hypocotyl and root elongation, although high concentrations of applied BRs result in inhibition of root elongation. A non-steroidal structure consisting of four stereoisomers was designed to contain subunits bearing key functional groups mimicking those of BL. The bioactivity of each of these individual stereoisomers was tested using the Arabidopsis thaliana det2-1 mutant line, which is deficient in BL, and thus does not etiolate in the dark. Application of BL at each of 0.1, 1.0, and 10.0 µM promotes hypocotyl elongation in dark-grown det2-1 plants while simultaneously inhibiting elongation of their primary root. In contrast, the mimetic structures, when applied to dark-grown det2-1 plants, promote hypocotyl elongation without negatively affecting primary root elongation. In fact, two of the mimetic structures, applied at a 10 µM concentration, significantly promoted both hypocotyl and root elongation. Correlation of this contrasting behavior with the configurations of the hydroxylated stereocenters of the mimetics is described. This is the first example of a non-steroidal BL mimetic where the biological activities of individual stereoisomers were tested and compared.  相似文献

Imaging analysis was carried out during the gravitropic response of etiolated Arabidopsis hypocotyls, using an IAA19 promoter fusion of destabilized luciferase as a probe. From the bright-field images we obtained the local deflection angle to the vertical, A, local curvature, C, and the partial derivative of C with respect to time, \(\partial C/\partial t\). These were determined every 19.9 µm along the curvilinear length of the hypocotyl, at ~10 min intervals over a period of ~6 h after turning hypocotyls through 90° to the horizontal. Similarly from the luminescence images we measured the luminescence intensity of the convex and concave flanks of the hypocotyl as well as along the median of the hypocotyl, to determine differential expression of auxin-inducible IAA19. Comparison of these parameters as a function of time and curvilinear length shows that the gravitropic response is composed of three successive elements: the first and second curving responses and a decurving response (autostraightening). The maximum of the first curving response occurs when A is 76° along the entire length of the hypocotyl, suggesting that A is the sole determinant in this response; in contrast, the decurving response is a function of both A and C, as predicted by the newly-proposed graviproprioception model (Bastien et al., Proc Natl Acad Sci USA 110:755–760, 2013). Further, differential expression of IAA19, with higher expression in the convex flank, is observed at A?=?44°, and follows the Sachs’ sine law. This also suggests that IAA19 is not involved in the first curving response. In summary, the gravitropic response of Arabidopsis hypocotyls consists of multiple elements that are each determined by separate principles.  相似文献

Changes in agronomic characters and the profile of various endogenous phytohormones during tuber development were studied in Dioscorea opposite (Chinese yam) cv. Guihuai 16. Tuber development exhibited a sigmoidal growth pattern according to the changes in tuber agronomic characters. The growth cycle of yam tuber could be divided into three stages: initiation stage, enlargement stage, and maturation stage. Moreover, the enlargement stage could be separated into three phases—slow growth phase, rapid growth phase, and late growth phase. Endogenous changes in phytohormones were associated with developmental changes in the tubers. The pulses of bioactive gibberellins (such as GA3 and GA4) were measured in tubers. The highest contents of GA3 and GA4 were reached 90 days after field planting, corresponding to the beginning of the rapid growth phase of tuber enlargement. Changes in trans-zeatin (tZ), jasmonic acid (JA), indole-3-acetic acid (IAA), and abscisic acid (ABA) levels were also observed, and seemed to be related to tuber enlargement at different phases. Continuous increases in JA and tZ contents accompanied tuber enlargement. Transient pulses of both IAA and ABA contents were also observed at the start of tuber rapid growth. Additionally, a second peak level of IAA was detected at the tuber maturation stage. These results suggest GAs play a key role at the beginning of the tuber rapid growth stage, and there is a close relationship between whole tuber enlargement and the contents of JA and tZ. Moreover, it is suggested that IAA and ABA also may be linked to the beginning of tuber rapid growth, and IAA also seems to be correlated to late tuber maturation.  相似文献

This study investigated the role of hydrogen sulfide (H2S) in the regulation of the ascorbate (AsA) and glutathione (GSH) metabolism by jasmonic acid (JA) in the leaves of Arabidopsis thaliana by using H2S scavenger hypotaurine (HT) and H2S synthetic mutant (SALK_041918, designated Atl-cdes). The results showed that JA significantly increased the H2S content, the activities of L-cysteine desulfhydrase (L-CDes), D-cysteine desulfhydrase (D-CDes), ascorbate peroxidase (APX), glutathione reductase (GR), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), L-galactono-1,4-lactone dehydrogenase (GalLDH) and γ-glutamylcysteine synthetase (γ-ECS), the ratio of AsA to dehydroascorbate (DHA), and decreased the content of malondialdehyde (MDA) and H2O2 in the wild type of A. thaliana, compared to control. The above effects of JA except the increased activities of L-CDes and D-CDes were suppressed by addition of HT. However, JA and HT+JA had no significant effects on the ratio of reduced GSH to oxidized GSH (GSSG) in the wild type of A. thaliana. Application of HT to the control decreased H2S content, AsA/DHA ratio, and activities of APX, GR, DHAR, MDHAR, γ-ECS, and GalLDH, but had no effects on MDA content, activities of L-CDes and D-CDes, and GSH/GSSG ratio. In the H2S synthetic mutant, JA had no obvious effects on above mentioned parameters except the D-CDes activity compared with the control. Our results suggest that JA-induced H2S, which is a signal that leads to the up-regulation of the AsA and GSH metabolism.  相似文献

In order to determine the most suitable in vitro tissue culture and plant regeneration conditions for the small flowered willow herb (Epilobium parviflorum Schreb), various explants were cultured on semi-solid MS media containing factorial combinations of plant growth regulators. Callus induction from hypocotyl, cotyledon, petiole and leaf explants was achieved on media containing 2,4-dichlorophenoxy acetic acid (2,4-D) and kinetin (KIN). All other growth regulator combinations [□-naphtaleneacetic acid (NAA) ± benzylaminopurine (BAP), NAA ± thidiazuron (TDZ), indol acetic acid (IAA) ± Zeatin (ZEA)] tested failed to respond. The best results with cotyledon- and petiole- derived callus were obtained from MS medium supplemented with 1.0 mg l?1 2,4-D + 0.1 mg l?1 KIN and 2.0 mg l?1 2,4-D + 0.2 mg l?1 KIN. It was observed that B5 basal medium was more effective than MS basal medium for producing seedling and the most effective seed sterilizing solution was 25 % (v/v) sodium hypochlorite (NaOCl). No plant regeneration was observed in either callus induction or during the subculturing stage. This is the first report on in vitro tissue culture study within the genus Epilobium.  相似文献

Low temperatures limit plant growth, development, and reproductive success. A series of complex adaptive responses in plants evolved to withstand this environmental challenge. Here, eight accessions of Elymus nutans, which originated in Tibet at altitudes between 3720 and 5012 m above sea level, were used to identify heritable adaptations to chilling stress. Dynamic responses of phytohormone, sugar, and gene expression levels related to chilling tolerance were analyzed. During the initial stage of chilling stress (0–24 h), some high-altitude E. nutans accessions exhibited rapid increases in abscisic acid (ABA), jasmonic acid (JA), and zeatin content. This coordinated with decreases in the levels of auxin (IAA), salicylic acid (SA), gibberellins (GA), and the ethylene precursor 1-aminocyclopropane-1-carboxylic acid (ACC). EnCBF9 and EnCBF14 expression in the high-altitude accessions, Baqing, Xainza, Damxung, and Ali, increased within 1 h of chilling exposure, while chilling induction of EnCOR14a was detected after 3 h of chilling stress. Accessions from high altitudes displayed an increased sucrose and raffinose accumulation and a reduced degradation of chlorophyll under chilling stress. After 24–120 h of chilling exposure, plant adaptation to the chilling treatment was associated with a lower accumulation of ABA and moderate rise of zeatin, IAA, GA, ACC, SA, and JA. EnCBF9, EnCBF14, and EnCOR14a genes were down-regulated during the late stage of chilling stress. Taken together, the dynamic responses of phytohormones and sugars, and the higher expression of the EnCBFs and EnCOR genes play critical roles in the acclimation to chilling in high-altitude accessions of E. nutans, thereby allowing them to achieve higher chilling tolerance.  相似文献

The role of jasmonic acid (JA) in plant photomorhogenesis under green light (GL) was studied. The effect of GL of different intensity (8.1 and 18.1 W/m2) with or without 1 μM JA treatment on growth of plants and photosynthetic pigment level was compared for two types of Arabidopsis thaliana (L.) Heynh. Landsberg erecta ecotype plants: Ler, the wild type, and hy4, a mutant. A much more pronounced growth of hypocotyls and cotyledons of Ler plants in GL was observed compared to that of hy4 with suppressed cryptochrome 1 (cry1), a GL photoreceptor. Treatment with JA in GL caused retardation of hypocotyl and cotyledon growth of Ler plants; however, it stimulated their growth in hy4 plants. JA reduced the chlorophyll a and total carotenoids levels in cotyledons of Arabidopsis plants in GL. Blocked GL signal transduction due to the absence of cry1 in hy4, as well as the higher intensity of GL reduced the negative effect of exogenous JA on growth of cotyledons and photosynthetic pigments. The rice EMS-derived mutant leaf adaxialized 1 (lad1) was isolated based on its upward rolling leaf phenotype. Besides the adaxially rolled leaf, many other agronomic traits were also compromised in lad1. The rolling trait was characterized by a noticeable alteration of bulliform cells in the adaxial side of the leaves. Map-based cloning showed a single nucleotide substitution in the promoter region of the KAN1 gene in lad1 mutant. Further, over-expressing and CRISPR/cas9-edited knockdown transgenic plants confirmed that KAN1 was responsible for the mutant phenotype of lad1. Yeast two-hybrid and bimolecular fluorescence complementation assay demonstrated that KAN1 can interact with the auxin response factors ARF3, ARF7 and ARF15. Physiologically, the contents of auxin (IAA), abscisic acid (ABA), jasmonic acid (JA) and gibberellin (GA) were all significantly increased in the lad1 mutant. Moreover, the GA3 content dramatically decrease in wild-type, but increased in lad1 under IAA induction. Additionally, the expression levels of several IAA and GA biosynthesis and responsive-related genes and genes involved in leaf polarity determination were altered in lad1. Therefore, we hypothesized that KAN1/ARFs protein complexes act as auxin-dependent regulatory units that play a conserved role in leaf development.  相似文献

The effects of the salt stress (200 mM NaCl) and exogenous jasmonic acid (JA) on levels of osmolytes and flavonoids in leaves of four-week-old Arabidopsis thaliana L. plants of the wild-type (WT) Columbia-0 (Col-0) and the mutant jin1 (jasmonate insensitive 1) with impaired jasmonate signaling were studied. The increase in proline content caused by the salt stress was higher in the Col-0 plants than in the mutant jin1. This difference was especially marked if the plants had been pretreated with exogenous 0.1 μM JA. The sugar content increased in response to the salt stress in the JA-treated WT plants but decreased in the jin1 mutant. Treatment with JA of the WT plants but not mutant defective in jasmonate signaling also enhanced the levels of anthocyanins and flavonoids absorbed in UV-B range in leaves. The presence of JA increased salinity resistance of the Col-0 plants, since the accumulation of lipid peroxidation products and growth inhibition caused by NaCl were less pronounced. Under salt stress, JA almost did not render a positive effect on the jin1 plants. It is concluded that the protein JIN1/MYC2 is involved in control of protective systems under salt stress.  相似文献

Brassinosteroids (BRs) are plant hormones that affect diverse aspects of plant development. Various BR-biosynthetic or BR-signaling mutants contribute to BR functions and signaling events in many plant species. The BR receptor brassinosteroid-Insensitive 1 (BRI1) plays critical roles in BR signaling. We previously identified a weak bri1 mutant allele, bri1-120, that has a mutation site in the extracellular domain of BRI1. Here, genetic suppressor screening revealed that a PHYB gene mutation led to suppression of ethyl methanesulfonate (EMS)-mutagenized bri1-120. The morphology of bri1-120phyB-1 indicated that compact and rounded phenotypes of bri1-120 were suppressed. However, BR sensitivity of the bri1-120phyB-1 was only recovered in hypocotyl elongation, and overexpression of PHYB in bri1-120 did not enhance bri1-120 phenotypes. To further investigate the relationship between BR and light signalings, we examined the seed germination pattern and hypocotyl growth of bri1-120phyB-1 as compared to that of each single mutant under various light conditions. Seed germination in bri1-120phyB-1 was higher than in both the single mutants. Hypocotyl length in bri1-120phyB-1 was intermediate between that of bri1-120 and phyB-1, whereas sensitivity to red light in bri1-120phyB-1 remained the same as in phyB-1. These results suggest that BR and light signalings affect diverse cellular responses both together and independently, depending on the specific cellular processes.  相似文献
