Highly tolerogenic bovine gamma globulin (BGG), a thymus-dependent antigen, was prepared by biologic filtraration in vitro. It readily induced tolerance in vivo in BALB/c mice and also rendered their nonadherent lymph node cells tolerant after in vitro incubation. Biologic filtration in vitro was carried out by incubating 2.5 × 107 lymph node cells with 10 mg of nontolerogenic BGG in 10 ml of Eagle's medium containing 2% normal mouse serum at 37 °C for 6 hr. The BGG-containing medium was clarified by centrifugation and was used without further dilution.For tolerance induction in vitro, lymph node cells were separated into adherent and nonadherent populations on Falcon plastic. These cells were incubated for 0–18 hr at 37 °C with biologically filtered BGG (bBGG). After incubation, the cells were washed three times and (2–2.5) × 107 nonadherent or 4 × 106 adherent cells were injected iv with their untreated counterpart into lethally irradiated mice which had received 106 bone marrow cells. The recipients were then challenged with 300 μg of aggregated BGG, and tolerance was assayed by the elimination of labeled BGG, rosette formation, and passive hemagglutination. Spleen cells were similarly treated for comparison. Our findings show that tolerance was not induced in vitro in adherent lymph node cells. However, in the nonadherent populations, those from the lymph node but not the spleen were rendered tolerant. The acquisition of tolerance in vitro was gradual. It was dependent upon the length of exposure to bBGG and required at least 6 hr.  相似文献

In vitro culture techniques have been used to compare the direct (IgM) plaqueforming cell (PFC) response to heterologous erythrocytes (RBC) by normal mouse spleen cells and spleen cells from mice injected intravenously with 5 × 104 RBC ten days previously [low dose primed (LDP)]. Although LDP mice fail to undergo a significant primary PFC response, their spleen cells are capable of a secondary or enhanced PFC response in vitro. The secondary PFC response is shown to be a function of: (A) an increase in the frequency of immunocompetent cells or units (IU) due to in vivo priming, and (B) an increased number of PFC generated per IU subsequent to in vitro stimulation. The latter increase is shown to be mediated through a shorter PFC doubling-time during logarithmic expansion of the PFC population. Analysis of nonadherent spleen cell dose response experiments indicate that two nonadherent cell types interact in the secondary response. Subsequent cocultivation experiments suggested that both of these cell types must be “primed” to allow induction of a secondary response. Although adherent cells are required for the secondary response, normal splenic adherent cells serve as equivalent substitutes for LDP adherent cells.  相似文献

The role of adherent cells in the regulation of lymphocyte DNA, RNA, and protein synthesis was investigated. Splenic adherent cells suppressed DNA synthesis of spleen cells. The reverse was true of lymph node adherent cells, i.e., DNA synthesis was less in the absence of adherent cells. Histocompatibility was not required for interaction between adherent and nonadherent cells. The regulatory adherent cell in either case was not θ positive. Removal of splenic adherent cells decreased RNA and protein synthesis of nonadherent spleen cells. RNA synthesis by lymph node cells increased when adherent cells were removed, but protein synthesis was lower. Autoradiographic analysis revealed that removal of adherent spleen cells allowed a greater number of nonadherent cells to respond to the presence of mitogen. Adherent cells were observed to regulate DNA synthesis of B lymphocytes also. Lymph node adherent cells amplified DNA synthesis in both nonadherent spleen and lymph node cells, while splenic adherent cells suppressed splenic nonadherent cells but stimulated nonadherent lymph node cells. We feel the data are compatible with the idea that at least two functionally distinct adherent cell populations exist. A mechanism is suggested to explain the data.  相似文献

The plaque-forming cell (PFC) response of long-term radiation induced allogeneic bone marrow chimeric (ABMC) mice has been shown to be markedly deficient. The nature of the cellular deficiency of the primary PFC response was investigated using in vitro culture techniques. Adherent spleen cells from ABMC or DBA/2 mice support equally well the development of PFC from nonadherent DBA/2 spleen cells. Nonadherent cells prepared from ABMC mice when cocultivated with DBA/2 adherent cells showed a minimal response. However, the addition of activated DBA/2 T cells to cultures containing adherent cells from DBA/2 mice and nonadherent cells from ABMC mice completely reconstituted the in vitro response to sheep erythrocytes. Therefore a cellular deficiency of the humoral immune system of ABMC mice was shown to be associated with the thymus-derived lymphocyte pool.  相似文献

LPS-induced immune unresponsiveness has been shown to be related to an impaired production of differentiation signal factor(s). The mechanism underlying this phenomenon was analyzed. The in vitro anti-SRBC response of spleen cells from normal mice was not suppressed by addition of LPS-treated spleen cells, ruling out a possible implication of active suppressor cells. Immune responsiveness concomitant with TRF production was restored in LPS-sensitized cells upon addition of 2-ME. The role of adherent and nonadherent cells was also investigated; both cell populations from LPS-treated mice were able to collaborate with their normal counterparts showing that the defective TRF production results from synergistic effects of LPS-induced alterations concerning both adherent and T-cell populations.  相似文献

The transfer of lymphocytes from NIH mice infected with Leishmania tropica major to nonimmune syngeneic mice, which had received 600 rad irradiation, markedly altered the course of subsequent Leishmania mexicana and L.t. major infections in these animals, when compared with infections in irradiated mice reconstituted with lymphocytes from uninfected animals. Some resistance to L. mexicana could be transferred with unfractionated lymphocytes, mixed nylon wool adherent, and nonadherent lymphocytes and to a lesser extent by nylon wool nonadherent cells alone but not by the adherent population. When the recipients of “immune” lymphocytes were infected with L.t. major the resulting lesions were either small and did not ulcerate, or grew rapidly, ulcerated, and healed. The course of L.t. major in the recipients depended on whether the donor of “immune” lymphocytes had a healed or a healing lesion. Immunity to L.t. major was largely associated with the nylon wool nonadherent population.  相似文献

Responsiveness of rabbit spleen cells to anti-allotype antibody was measured in terms of increased thymidine incorporation. Incorporation was enhanced after removal of cells which had ingested or had adhered to magnetic particles. B lymphocytes, prepared from spleen cells by the removal of adherent cells and of RTLA bearing T cells, were more responsive to anti-allotype antibody than were the original spleen suspensions. This increase could not be explained by enrichment in B cells. It was concluded that an adherent cell suppressed B cell transformation. The addition of 2-mercaptoethanol to the cell cultures stimulated with mitogen augmented the incorporation of thymidine. Adherent cells interfered with 2-mercaptoethanol potentiation in the response to anti-allotype antibody but not in the response to Con A. Fractionation of spleen cells, over glass bead columns, yielded nonadherent and adherent cell populations. The responsiveness of nonadherent cells to anti-allotype induced thymidine incorporation was two to six times that of unfractionated cells. The responsiveness of nonadherent cells to stimulation by anti-allotype antibody was reduced after addition of adherent cells. Findings were discussed in terms of the inhibitory role played by adherent cells on anti-allotype antibody induced responsiveness of rabbit B cells and of the possible participation of a third cell type which functions as a promotor of mitogenic T cell stimulation.  相似文献

The cultured cells prepared from the spleens and peritoneal exudate cells of the C3H/HeJ strain of mice produce very little or no interferon (IFN) by stimulation of bacterial lipopolysaccharide (LPS). However, the cells taken from LPS-non-responder C3H/HeJ mice which had been infected with Mycobacterium bovis bacillus Calmette-Guérin (BCG) prior to the experiment were capable of producing IFN in culture in the presence of LPS. The peritoneal exudate cells of BCG-primed C3H/HeJ mice were separated into adherent cell and nonadherent cell populations by their adhesiveness to plastic culture dishes. IFN production required the presence of both these cell populations in the same culture, and the IFN activities produced were mainly IFN-gamma. The cultures with nonadherent cells and fixed adherent cells still produced IFN, but the cell cultures reconstituted with the BCG-primed cell population and unprimed cell population produce little if any IFN-gamma. Moreover, when both of the populations were cultured in Marbrook culture vessels separated by a membrane filter, the cultures produced very little or no IFN-gamma. These results indicate that there is a mechanism of IFN-gamma induction by LPS which requires the direct contact between adherent cells and nonadherent cells without the participation of any soluble factor(s) from the adherent cells. The producer cells were mainly in the nonadherent cell population. Previous treatment of nonadherent cells with anti-Thy-1.2 antibody, anti-Lyt-1.1 antibody, anti-L3T4 antibody, or anti-asialo-GM1 antibody and complement diminished the ability of the cells for LPS-induced IFN production with the help of adherent cells. Therefore, it is concluded that both T cells (presumably L3T4+T cells) and asialo-GM1+ natural killer cells in the BCG-primed C3H/HeJ cell cultures produced IFN-gamma in the presence of LPS, and the production was supported by the function of macrophages.  相似文献

Freshly isolated or overnight cultured peripheral blood mononuclear cells from immune or nonimmune animals had natural cytolytic activity against bovine herpesvirus 1 (BHV-1)-infected tumor target cells. No lysis was demonstrated against tumor target cells alone. This natural cytolytic activity was present in mononuclear cells from the spleen, lymph node, and peripheral blood but little or no cytolytic activity was detected in bone marrow or thymus cells. When monoclonal antibodies and complement to deplete bovine mononuclear cell subpopulations from the nonadherent cells were used, results indicated the effector cell was not a T cell, B cell, or activated monocyte. From nonadherent populations separated on density gradients, it was determined that the effector cells were large, low density mononuclear cells. These results indicate the nonadherent effector cells mediating lysis of BHV-1-infected xenogeneic adherent target cells were large null lymphocytes and/or immature monocytes.  相似文献

The contribution of lymphotoxin to guinea pig leukocyte natural cytotoxicity was evaluated with [3H]TdR release and colony-inhibition assays of 104C1 benzo(a)pyrene in vitro-transformed and tumorigenic, tumor-specific transplantation antigen-negative, syngeneic strain 2/ N fibroblasts. Cytolethal 3H-release activities of mitogen (PHA)1-stimulated nonimmune and ovalbumin (OA) immune as well as OA-stimulated OA immune unfractionated, adherent (macrophage-enriched) and nonadherent peritoneal leukocytes are qualitatively similar. 3H release is maximal by 48 hr, increases with antigen or mitogen concentration, is greatest with unfractionated leukocytes, and is least with adherent macrophages. Lymphotoxin produced by peritoneal leukocytes, alone or in combination with the leukocytes does not or only minimally induces 3H release even after 6 days of incubation with guinea pig target cells although guinea pig lymphotoxin possesses cytolytic activity as indicated by 3H release from αL929 mouse tumor cells. In contrast to the absent or very weak cytolytic activity of guinea pig lymphotoxin for the guinea pig target cells nonimmune macrophages, nonadherent leukocytes, and lymphotoxin all exhibit readily detectable colony-inhibitory (CI) activity for the syngeneic tumor cells. Macrophage and lymphotoxin CI, moreover, are additive, whereas nonadherent leukocyte and lymphotoxin CI are synergistic. The latter may be due to additional lymphotoxin induced by target cell antigens or other mechanisms of target cell stimulation of effector lymphoid cells and result from very high local levels of lymphotoxin released by the effector cells. Lymphotoxin CI, furthermore, can be cytostatic or cytolethal as indicated by resumption of 104C1 but not αL929 colony growth following removal of lymphotoxin, indicating that natural cell-mediated cytotoxicity consists of lymphotoxin-dependent and -independent cytostatic and cytolethal effector mechanisms.  相似文献

We have fractionated human peripheral blood lymphocytes (PBL) into subclasses with an affinity technique that takes advantage of the specific interaction between the plant lectin, Lens culinaris agglutinin (Lentil-PHA), and its cell surface receptors. PBL were incubated in plastic tubes or petri dishes coated with a gelatin layer to which lentil-PHA had been coupled using a water soluble carbodiimide compound. Adherent cells were recovered by melting the gelatin, and bound lectin was removed by exposing the cells to an appropriate sugar hapten. Approximately 25 to 50% of PBL specifically adhered to gelatin-coated tubes derivatized with lentil-PHA. Binding studies with 125I-labeled lentil-PHA showed that, compared to unfractionated PBL which bound 2.0 × 106 lentil-PHA molecules per cell, adherent cells bound more (3.7 × 106/cell), and nonadherent cells bound less (1.1 × 106/cell) lentil-PHA. By contrast, there were no differences among these groups for binding of other plant lectins. When stimulated in vitro by optimal mitogenic concentrations of lentil-PHA, lymphocytes adherent to lentil-PHA responded better than their nonadherent counterparts whereas the two groups responded the same to stimulation by the leukoagglutinin from Ph. vulgaris. The data indicate that plant lectins may be used to isolate PBL subclasses with different structural and functional properties.  相似文献

The involvement of adherent splenic cells in the production of deficient lymphocyte responses during the acute phase of experimental Chagas' disease was investigated. When cultured together, purified adherent splenocytes from mice acutely infected with Trypanosoma cruzi caused a significant reduction in the responses of normal mouse spleen cells to T and B cell-specific mitogens. Similar observations were made when infected mouse adherent splenocytes were co-cultured with normal mouse nonadherent cells. Exchange of adherent cells in infected mouse spleen cells suspensions for adherent cells from uninfected mice resulted in increased responses to stimulation with the T and B cell mitogens tested. Treatment of infected mouse cell suspensions with indomethacin improved the responsiveness of these cells to the mitogens. These results support the concept that the immunosuppression that is characteristic of experimental acute Chagas' disease is at least in part mediated by an adherent cell population and is dependent on a prostaglandin-mediated mechanism.  相似文献

In the present study we investigated some of the cellular mechanisms for the generation of macrophage-activating factor(s) (MAF) in immune responses to tumor antigens. C3H/HeN mice were immunized to syngeneic MH134 hepatoma or MCH-1-A1 fibrosarcoma by intradermal inoculation of viable tumor cells, followed by the surgical resection of the tumor. Spleen and lymph node cells from these tumor-immune mice were stimulated in vitro with the corresponding tumor cells, and supernatant from such a culture was tested for an ability to activate macrophages to exert their cytostatic and cytolytic activities as detected on tumor cells unrelated to immunizing tumors. Peritoneal adherent cells as a macrophage source, which were preincubated with supernatant from co-culture of tumor-unimmunized normal spleen and lymph node cells plus tumor cells, failed to exhibit any significant antitumor effect on unrelated X5563 tumor cells, whereas the addition of supernatant from cultures containing immune lymphocytes to adherent cells resulted in appreciably potent cytostatic and cytolytic effects on X5563 tumor cells, indicating the generation of MAF in culture supernatant. The activation of tumor-immune spleen and lymph node cells for MAF generation was tumor-specific, because anti-MH134- and anti-MCH-1-A1-immune lymphocytes produced MAF by the stimulation with the respective but not with the other alternative tumor cells. Such MAF production was abolished by treatment of tumor-immune spleen and lymph node cells with anti-Thy-1.2 or anti-Lyt-1.1 but not with anti-Lyt-2.1 antibody plus complement before culturing. These results indicate that the tumor-specific Lyt-1+2- T cell subset has a crucial role in generating MAF by which an adherent cell population as a source of macrophages acquires the potential for inducing a cytolytic as well as a cytostatic effect on tumor cells.  相似文献

The stimulator cells for the activation of cytotoxic T lymphocyte (CTL) precursor cells were studied in vivo and in microculture systems with limiting concentrations of helper factor (interleukin-2). We found in both cases that mixtures of irradiated spleen cells from athymic and euthymic donors of different haplotypes activated CTL responses preferentially against alloantigens which were carried by the euthymic donors. This applied also if athymic and euthymic donors shared parts of the H-2 gene complex. Stimulator cells from athymic and euthymic donors activated, on the other hand, equally strong responses in microcultures which were supplemented with additional helper factor or in conventional macrocultures. This and direct cell mixing experiments showed that the ineffective stimulation by the nude spleen cells in vivo and under conditions of limiting helper factor was not mediated by active suppression. Our experiments are therefore best explained by the assumption that cytotoxic responses under conditions of limiting helper factor require a close proximity between CTL precursor cells and helper T cells. This proximity may be most efficiently provided by the receptors of the CTL precursor cells when helper T cells serve as stimulator cells. Lymph node cells were consistently inferior to spleen cells as stimulator cells, and the nylon wool nonadherent fraction of spleen cells was on the average also inferior to unfractionated spleen cells, indicating that the stimulator T cells belong to a spleen seeking and partly nylon wool adherent T-cell subpopulation or require an additional cell type for optimal stimulation.  相似文献

Strategies to produce resistance to infection with Leishmania braziliensis in BALB/c mice are described. Mice infected with virulent parasites were used as spleen cell donors (adherent/nonadherent cells) with which to immunize naive recipients which were themselves later challenged with the organism. Immunization with both adherent and nonadherent spleen cells (but not serum) in the presence of adjuvant led to protection. In the former case it seems that an immunogenic form of parasite antigen presented in the context of MAC-1+ adherent cells was responsible. In contrast immunization with nonadherent spleen cells depended upon the presence of Thy-1.2+ Lyt-1+ cells in the spleen cell preparation from infected animals. Immunization with adherent cells, but not with nonadherent cells, led to the development of a population of Thy-1.2+ spleen cells capable of adoptively transferring resistance to naive mice.  相似文献

Picryl chloride induces contact hypersensitivity in mice, accompanied by spleen cell sensitization that is demonstrable in vitro by specific antigen-induced formation of leukocyte adherence inhibition factor (LAIF). This cellular activity was detected only up to 7 days after sensitization; thereafter the spleen cells appeared to be unreactive with the antigen. The cells were still normally reactive with the mitogen concanavalin A. Antigen reactivity of such “late” cells was restored by passage through a glass-bead column (provided resulting nonadherent cells were reconstituted with normal macrophages), and the restored reactivity was again suppressed by the eluted glass-bead-adherent cells. Suppression was antigen specific. Separation of T and B lymphocytes by affinity chromatography, after glass-bead treatment of sensitized spleen cells, showed that two subpopulations of B cells—those responsible for producing LAIF as well as those suppressing LAIF production by T cells—were glassbead adherent. This was extended by showing directly with anti-Thy-1.2 serum that B cells producing LAIF and suppressor T cells were glass adherent. Thus two suppressive cell populations, and the B cell producing LAIF, were glass adherent while the T-cell LAIF producer was not. Tests for adoptive transfer of cutaneous hypersensitivity in vivo demonstrated the relevance of many of the above observations to conditions in the whole animal. “Late” spleen cells from sensitized mice could not transfer hypersensitivity but this property was restored by glass-bead passage. The eluted adherent cells suppressed transfer. Both adoptive transfer and its suppression were antigen specific.  相似文献

The capacity of human monocytoid cell lines and peripheral blood monocytes to modulate their expression of plasminogen receptors has been assessed. After PMA stimulation, THP-1 or U937 monocytoid cells were separated into adherent and nonadherent populations. Plasminogen bound to adherent cells with similar capacity and affinity as to nonstimulated cells. In contrast, the nonadherent cells bound plasminogen with 5-17-fold higher capacity (without a change in affinity). This increase was selective as urokinase bound with similar affinity and capacity to the adherent and nonadherent populations. Upregulation of plasminogen receptors on the nonadherent monocytoid cells was rapid, detectable within 30 min, and reversible, adhesion of the nonadherent cells resulted in a sixfold decrease in plasminogen binding within 90 min. The increase in plasminogen binding to the nonadherent cells was associated with a marked increase in their capacity to generate plasmin activity from cell-bound plasminogen. PMA stimulation of human peripheral blood monocytes increased their expression of plasminogen receptors by two- to fourfold. This increase was observed in both adherent and nonadherent monocytes. Freshly isolated monocytes maximally bound 5.0 x 10(5) plasminogen molecules per cell, whereas monocytes cultured for 18 h or more maximally bound 1.7 x 10(7) molecules per cell, a 30-fold difference in receptor number. These results indicate that both monocytes and monocytoid cell lines can rapidly and markedly regulate their expression of plasminogen binding sites. As enhanced plasminogen binding is correlated with an increased capacity to generate plasmin, an enzyme with broad substrate recognition, modulation of plasminogen receptors may have profound functional consequences.  相似文献

Summary Lymphocytes from patients with transitional cell carcinoma (TCC) of the urinary bladder are more cytotoxic to bladder tumor cells than to a variety of control cells. This disease-related cytotoxicity has previously been shown to involve several mechanisms and different types of effector cells. To analyze further the nature of the effector cells operative in this system, peripheral blood lymphocytes from eight TCC patients were stimulated in vitro with TCC extract and cultured in the presence of interleukin 2 and allogeneic feeder cells. When tested for cytotoxicity in vitro on a target cell panel including both adherent and nonadherent cell lines, the lymphocytes killed a broad spectrum of targets in a major histocompatibility complex (MHC)-unrestricted fashion. When cloned by limiting dilution, clones were obtained which displayed a more restricted pattern of target cell killing. Some of the clones were highly but not exclusively selective for TCC-derived target cells. Phenotypically, these cells resembled mature T cells of CTL-type (CD8+/CD4). They also expressed the CD3/5 T cell antigen receptor complex but target cell killing was not MHC-restricted. The results of various inhibition experiments suggested that the CD3/TCR complex was involved in the cytotoxicity exhibited by these effector cells. However, its precise role in target cell recognition and the identification of the tumor cell structures recognised by the effector cells require further studies.  相似文献

Summary Nonadherent cells of the bone marrow of C3H/HeN mice were incubated for 3 days with the culture supernatant of an L-929 cell line containing macrophage-colony-stimulating factor. Approximately, 70% of the cells became phagocytic, adherent to plastic dishes and positive for nonspecific esterase staining. The adherent cells exhibited a weak tumoricidal activity against MM48 syngeneic mammary carcinoma cells, and the cytotoxicity was strongly augmented by the addition of bacterial lipopolysaccharide to the cytotoxicity assay. The cytotoxicity induced by lipopolysaccharide was also shown to be mediated by Thy1.2 and asialo-GM1+ cells, and was abrogated by the addition of carrageenan. Macrophage-colony-stimulating-factor-producing (D66) and nonproducing (A23) variants were separated from the MM48 tumor line in in vitro culture following limiting dilution. There was no difference between these two variants in either the in vitro growth rate or the susceptibility to macrophage-mediated cytotoxicity. C3H/HeN mice inoculated i.p. with D66 survived longer than did those inoculated i.p. with A23. C3H/HeN mice bearing D66 or A23 as an ascitic form were given i.p. injections of Nocardia rubra cell wall skeleton (N-CWS). N-CWS significantly prolonged the survival period of mice bearing D66, whereas it exhibited no apparent antitumor effect on mice bearing A23. The increase in the cell number of D66 in the peritoneal cavity was significantly retarded, compared with that of A23. In contrast, the number of peritoneal macrophages increased more in D66-bearing mice than in A23-bearing mice. The increase in the peritoneal macrophage number was further augmented by an i.p. injection of N-CWS. Peritoneal macrophages of D66-bearing mice exhibited apparent tumoricidal activity against MM48 tumor cells in the presence of lipopolysaccharide, and the cytotoxicity was significantly augmented by i.p. injection of N-CWS. On the other hand, the responsiveness of peritoneal macrophages to lipopolysaccharide was found to be poor in A23-bearing mice and the tumoricidal activity was only weakly augmented by N-CWS. These results strongly suggest that M-CSF plays an important role not only in the maturation of macrophage progenitors but also in the induction and the accumulation of activated macrophages. Nonadherent spleen cell populations exposed in vitro to ribonucleic acid-rich preparations from mouse macrophages that had been incubated with human γ-globulin (RNA : HGG) were able to produce specific antibody, as measured by rosette-forming cells, in lethally irradiated (800 R), reconstituted, syngeneic mice. Exposure of the RNA to anti-HGG serum abrogated its ability to initiate antibody synthesis, as did monospecific anti-human γ chain and anti-human κ chain serum. Normal rabbit serum, anti-bovine albumin serum, anti-human μ chain or anti-human λ chain serum, when substituted for anti-HGG serum had no effect. Thus, the presence of both γ heavy chains and κ light chains of the antigen in the RNA moiety was indicated. Although both an adherent and a nonadherent cell were required by HGG to stimulate rosetteforming cells in irradiated mice, the need for the adherent cell was eliminated when RNA : HGG was substituted for HGG.
