Two entomophthoraceous species, found at different localities in Israel, caused epizootics onHypera variabilis Herbst. larvae during spring of 1976 and 1977.Zoophthora phytonomi (Arthur) Batko, emend.Ben-Ze’ev & Kenneth (syn.Entomophthora phytonomi Arthur) produced in the field only conidia, whereasConidiobolus osmodes Drechsler formed only dark, rough-walled resting spores that had been erroneously thought in the past to belong toZ. phytonomi. In culture,Z. phytonomi, isolated from conidia, produced subhyaline, smooth resting spores, whereasC. osmodes from resting spores produced rotund conidia, which proved pathogenic toH. variabilis larvae and adult houseflies. 1. In fresh chloroplasts, three b-type cytochromes exist. These are b-559HP (λmax, 559 nm; Em at pH 7, +370 mV; pH-independent Em), b-559LP (λmax, 559 nm; Em at pH 7, +20 mV; pH-independent Em) and b-563 (λmax, 563 nm; Em at pH 7, ?110 mV; pH-independent Em). b-559HP may be converted to a lower potential form (λmax, 559 nm; Em at pH 7, +110 mV; pH-independent Em).2. In catalytically active b-f particle preparations, three cytochromes exist. These are cytochrome f (λmax, 554 nm; Em at pH 7, +375 mV, pK on oxidised cytochrome at pH 9), b-563 (λmax, 563 nm; Em at pH 7, ?90 mV, small pH-dependence of Em) and a b-559 species (λmax, 559 nm, Em at pH 7, +85 mV; pH-independent Em).3. Cytochrome bd is a terminal component of the respiratory chain of Escherichia coli catalyzing reduction of molecular oxygen to water. It contains three hemes, b558, b595, and d. The detailed spectroelectrochemical redox titration and numerical modeling of the data reveal significant redox interaction between the low-spin heme b558 and high-spin heme b595, whereas the interaction between heme d and either hemes b appears to be rather weak. However, the presence of heme d itself decreases much larger interaction between the two hemes b. Fitting the titration data with a model where redox interaction between the hemes is explicitly included makes it possible to extract individual absorption spectra of all hemes. The α- and β-band reduced-minus-oxidized difference spectra agree with the data published earlier ([22] J.G. Koland, M.J. Miller, R.B. Gennis, Potentiometric analysis of the purified cytochrome d terminal oxidase complex from Escherichia coli, Biochemistry 23 (1984) 1051-1056., and [23] R.M. Lorence, J.G. Koland, R.B. Gennis, Coulometric and spectroscopic analysis of the purified cytochrome d complex of Escherichia coli: evidence for the identification of “cytochrome a1” as cytochrome b595, Biochemistry 25 (1986) 2314-2321.). The Soret band spectra show λmax = 429.5 nm, λmin ≈ 413 nm (heme b558), λmax = 439 nm, λmin ≈ 400 ± 1 nm (heme b595), and λmax = 430 nm, λmin = 405 nm (heme d). The spectral contribution of heme d to the complex Soret band is much smaller than those of either hemes b; the Soret/α (ΔA430A629) ratio for heme d is 1.6.  相似文献

Insects are important for humanity; play role in crop pollination, and biocontrol of harmful pests. The red palm weevil, Rhynchophorus ferrugineus, is a major pest of date palms and has become a serious threat. Scientists needs ample numbers of insects for bioassays to explore control options. The alga Spirulina platensis, is enriched by protein, natural vitamins, minerals, and amino acids, stimulate the development of organisms that feed on it. I assessed the value of Spirulina as a nutritional supplement for red palm weevil larvae by adding its various percentages to the artificial diet. Once a week, the larvae were removed from the containers, washed with distilled water, dried, weighed using an electronic scale, returned to a new container, and supplied with Spirulina mixed fresh diet. Larvae fed with lower concentrations showed vigorous growth and significant weight gain. Particularly, larvae fed 0.5%, 1%, and 2% Spirulina powder supplementation to their diet were healthier and gained more weight than larvae reared with >5% concentration. Overall 40% mortality was recorded in larvae fed with 10% concentration. Higher concentrations were lethal, and all larvae died within two weeks when fed 20% Spirulina. The present research findings indicate that Spirulina used in concentrations from 0.5% to below 5% had a beneficial effect on red palm weevil larval growth but a detrimental effect and even mortality was recorded when used ≥5%.  相似文献

A Brazilian Bacillus thuringiensis subspecies israelensis, toxic to Diptera, including mosquitoes, was found also to show toxicity to the coleopteran boll weevil Anthonomus grandis Boheman at an equivalent level to that of the standard coleopteran-active B. thuringiensis subspecies tenebrionis T08017. Recombinant B. thuringiensis strains expressing the individual Cyt1Aa, Cry4Aa, Cry4Ba and Cry11Aa toxins from this strain were assessed to evaluate their potential contribution to the activity against A. grandis, either alone or in combination. Whilst individual toxins produced mortality, none was sufficiently potent to allow calculation of LC50 values. Combinations of toxins were unable to attain the same potency as the parental B. thuringiensis subsp. israelensis, suggesting a major role for other factors produced by this strain.  相似文献

Factorial treatments of entomopathogenic nematodes (EPN) and composted, manure mulches were evaluated for two years in a central Florida citrus orchard to study the post-application biology of EPN used to manage the root weevil, Diaprepes abbreviatus. Mulch treatments were applied once each year to study the effects of altering the community of EPN competitors (free-living bactivorous nematodes) and antagonists (nematophagous fungi (NF), predaceous nematodes and some microarthro-pods). EPN were augmented once with Steinernema riobrave in 2004 and twice in 2005. Adding EPN to soil affected the prevalence of organisms at several trophic levels, but the effects were often ephemeral and sometimes inconsistent. EPN augmentation always increased the mortality of sentinel weevil larvae, the prevalence of free-living nematodes in sentinel cadavers and the prevalence of trapping NF. Subsequent to the insecticidal effects of EPN augmentation in 2004, but not 2005, EPN became temporarily less prevalent, and fewer sentinel weevil larvae died in EPN-augmented compared to non-augmented plots. Manure mulch had variable effects on endoparasitic NF, but consistently decreased the prevalence of trapping NF and increased the prevalence of EPN and the sentinel mortality. Both temporal and spatial abundance of NF were inversely related to the prevalence of Steinernema diaprepesi, whereas Heterorhabditis zealandica prevalence was positively correlated with NF over time. The number of weevil larvae killed by EPN was likely greatest in 2005, due in part to non-target effects of augmentation on the endemic EPN community in 2004 that occurred during a period of peak weevil recruitment into the soil.  相似文献

The red palm weevil (RPW), Rhynchophorus ferrugineus, is an important pest of palms, and difficult to control by conventional methods. Therefore, microbial control is an alternative strategy for controlling RPW. Herein, a total of 15 entomopathogenic fungi (EPFs) were subjected to primary pathogenicity screening against last stage of RPW larvae. The preliminary data showed that four Beauveria bassiana isolates (JEF-484, 158, 462 and 507) and one Isaria fumosorosea isolate (JEF-014) resulted in 100 % mortality within 5–10 days post inoculation (d.p.i.), respectively. According to the time required for RPW mortality, JEF-484, 158, 462 and 014 were further subjected to bioassays using 107 conidia/ml suspensions by spraying method. Based on the results, JEF-484 showed the highest mortality and shortest LT50 on the last stage of RPW larvae, followed by JEF-158. The two isolates also showed good conidial production and high thermal stability compared to the other isolates. Therefore, JEF-484 and JEF-158 were selected for bioassays against RPW egg and the last larval stage with different concentrations of 105, 106 and 107 conidia/ml conidial suspensions by spraying method. For the bioassay at the egg stage, JEF-158 showed a significantly higher ovicidal effect than JEF-484. In the larval bioassay, both EPF isolates showed a dosage-dependent effect on the RPW larvae. JEF-484 caused higher mortality in RPW larvae than JEF-158. In summary, the combination of the 2 promising EPF isolates might provide an opportunity for the practical microbial control of RPW at different life stages in palm tree fields.  相似文献

Abstract: The western corn rootworm (Diabrotica virgifera virgifera LeConte, Col.; Chrysomelidae) is an alien invasive species in Europe. It is a univoltine species with eggs that overwinter in the soil and larvae that hatch in spring. Three larval instars feed on maize roots, which can cause plant lodging and yield loss of economic importance. Adults emerge between mid‐June and early August and can reduce yields through intensive silk feeding. In order to provide a thorough understanding of the population dynamics of this invasive pest species in the invaded European region, complete age specific life‐tables were constructed in two maize fields in southern Hungary assessing the significance of natural mortality factors acting on D. v. virgifera populations. This information provides a rational basis for devising sustainable integrated pest management programmes, in particular, by enabling the identification of vulnerable pest age intervals for the timely application of various management tools. The life‐table for D. v. virgifera in Europe resulted in a total mortality of about 99% from the egg stage in the autumn to the emergence of adult females in the following year (KTotal = 2.48), which is comparable with North America. The highest reduction of D. v. virgifera numbers resulted from the mortality in first instar larvae (94% marginal death rate) and from the unrealized fecundity (80%). However, only the variation in mortality between years can change the generational mortality and thus influence population growth. High variation in the marginal death rate between fields and years was found in the second and third instar larval stages, and in the overwintering egg stage. These mortality factors therefore have the potential to cause changes in the total generational mortality. Furthermore, the life‐table suggested that a high fecundity could compensate for a high generational mortality and would lead to population increase.  相似文献

(i) Compounds activating the microsomal electron transfer oxidative reactions, e.g., the mixed function oxidase (aminopyrine, aniline), the Δ9-desaturase (stearyl-CoA), and lipid peroxidation reaction (iron pyrophosphate), cause a decrease in the steady-state reduced level of cytochrome b5. (ii) In the absence of substrates, the kox for cytochrome b5 was the same whether reduced by NADH or NADPH (about 0.045 S?1, indicating that no distinction exists between the cytochrome b5 involved in NADH-driven and NADPH-driven microsomal reactions which utilize this hemoprotein. (iii) The agents activating the oxidative pathways affect the first-order rate constant for cytochrome b5 oxidation (kox), but the apparent first-order rate constant obtained for reduction (kred) of cytochrome b5 by NADPH is still more than 10 times the kox, and the kred obtained with NADH is still more than 100 times the kox. (iv) Of the compounds used, only stearyl-CoA caused a decrease in the NADH-supported steady-state reduced level of cytochrome b5. This effect is probably due to a detergent-like action of stearyl-CoA on the membrane proteins, interfering with some interactions (e.g., NADPH-cytochrome c reductase with cytochrome P-450; NADH-cytochrome b5 reductase with cytochrome b5). (v) Based upon the kinetic and steady-state measurements it is concluded that substrate-induced changes in the steady-state reduced level of cytochrome b5 are evidence for a decrease in the population of this hemoprotein available to the reductase due to competition with other more favored acceptors, (vi) Measurements using the duration of the reduced state and rates of electron flow through cytochrome b5 reveal that normally about 60% of the NADH-derived reducing equivalents go through cytochrome b5 while only about one electron in nine passes through this cytochrome when NADPH is the source of reducing equivalents. Substrates of the various pathways alter the proportion of electrons passing through cytochrome b5 depending upon their activating or inhibiting action on cytochrome b5-dependent or -independent reactions.  相似文献

The red imported fire ant,Solenopsis invicta Buren, was identified as a key predator of the boll weevil,Anthonomus grandis Boheman. Although key factor analysis does not guarantee that the key factor is the causative agent, the evidence of boll weevil mortality was so clear that the fire ant could be identified as causing the mortality. Ant predation on immature boll weevils during the summer of 1981 averaged 84%. When this predation was combined with other natural mortality factors, the weevil population density throughout the season never exceeded levels that required control measures.  相似文献

Larvae, pupae, and adults of the carrot weevil (Listronotus oregonensis) were infected and killed by the three entomophagous nematodes (Steinernema feltiae, S. bibionis, and Heterorhabditis heliothidis) under controlled conditions. Third-stage larvae were more susceptible than pupae or adults. S. feltiae and S. bibionis were the most aggressive nematode species, causing larval mortality after 24-48 hours in both continuous and 2-hour contact with nematode suspension. The nematodes multiplied sufficiently in all insects at all stages of development; however, production of infective-stage larvae per host cadaver was variable.  相似文献

The cabbage seedpod weevil, Ceutorhynchus obstrictus (Marsham) (Coleoptera: Curculionidae), is a widely distributed invasive pest of cruciferous crops in North America. Control measures rely mostly on the application of insecticides but alternative control strategies such as classical biological control are under evaluation. To investigate the impact of parasitoids and other mortality factors on C. obstrictus populations, life table studies were conducted between 2005 and 2007 in 13 winter oilseed rape fields in Switzerland, part of the native range. Under field conditions females only realized approximately 50% of their potential lifetime fecundity, varying between 96 and 631 eggs per individual. Total generational mortality was higher than 99.6% in each year. Overwintering mortality of adults was the major population limiting factor, contributing approximately 50% to the overall generational mortality of C. obstrictus, whereas factors acting on the immature stages were responsible for the remaining 50%. Among the mortality factors of the immature stages, egg, larval and pupal mortality contributed 9–12%, 25–28% and 4–6% to the generational mortality, respectively. Larval ecto-parasitism on its own accounted for 7–15% of the generational mortality. Comparison of life table data presented here with that from North America will be invaluable for elucidating the mortality factors that regulate C. obstrictus populations in Europe, the region of origin.  相似文献

Fitness associated with pesticide resistance was assessed in twospotted spider mites, Tetranychus urticae Koch, using life table analysis. Two strains resistant to fenpyroximate (FR) and pyridaben (PR), and one susceptible strain (S) were included in this study. Two reciprocal crosses (FR♂xS♀ and PR♂xS♀) were established by conventional crossing procedure. Intrinsic rate of natural increase (rm) was used as a comparative parameter of reproductive fitness among 3 parent strains (FR, PR, and S) and two reciprocal crosses. Sensitivity analysis was performed to evaluate significances of biological factors contributing to rm. The rm values were not significantly different from each other, with an exception of FR♂xS♀ cross and ranged from 0.250 to 0.274 per day. Sensitivity analysis revealed that birth rate (b) and mortality (μ) effects could account for the effect on rm in the two resistant strains. Contributions of b and μ to rm was markedly different in two resistant strains; high contribution in b and low in μ. Age at the first reproduction (t) and age at the last reproduction (tω) made roughly equal contributions to rm between the strains and the crosses. This study clearly showed that how rm was affected by various biological factors and provided a basic analytical tool for pesticide resistant management.  相似文献

Bruchid larvae cause major losses in grain legume crops throughout the world. Some bruchid species, such as the cowpea weevil, are pests that damage stored seeds. Plants synthesize a variety of molecules, including proteinaceous proteinase inhibitors, to defend themselves against attack by insects. In this work, a trypsin inhibitor (DMTI-II) isolated from Dimorphandra mollis seeds was tested for anti-insect activity against Callosobruchus maculatus larvae. The inhibitor produced ca. 67% mortality to this bruchid when incorporated into an artificial diet at a level of 1%. The doses necessary to cause 50% mortality (LD50) and to reduce weight by 50% (ED50) for DMTI-II were ca. 0.50% and 0.60%, respectively. The action of DMTI-II on C. maculatus larvae may involve the inhibition of trypsin-like activity of larval midgut extracts, the absence of digestion by midgut preparations or with a mixture of pepsin and papain, and its association with a chitin column and chitinous structure in the midgut of this insect.  相似文献

We analyzed the population dynamics of a burrowing owl (Athene cunicularia) colony at Mineta San Jose International Airport in San Jose, California, USA from 1990–2007. This colony was managed by using artificial burrows to reduce the occurrence of nesting owls along runways and within major airport improvement projects during the study period. We estimated annual reproduction in natural and artificial burrows and age-specific survival rates with mark–recapture techniques, and we estimated the relative contribution of these vital rates to population dynamics using a life table response experiment. The breeding colony showed 2 distinct periods of change: high population growth from 7 nesting pairs in 1991 to 40 pairs in 2002 and population decline to 17 pairs in 2007. Reproduction was highly variable: annual nesting success (pairs that raised ≥1 young) averaged 79% and ranged from 36% to 100%, whereas fecundity averaged 3.36 juveniles/pair and ranged from 1.43 juveniles/pair to 4.54 juveniles/pair. We estimated annual adult survival at 0.710 during the period of colony increase from 1996 to 2001 and 0.465 during decline from 2002 to 2007, but there was no change in annual survival of juveniles between the 2 time periods. Long-term population growth rate (λ) estimated from average vital rates was λa = 1.072 with λi = 1.288 during colony increase and λd = 0.921 (Δλ = 0.368) during decline. A life table response experiment showed that change in adult survival rate during increasing and declining phases explained more than twice the variation in growth rate than other vital rates. Our findings suggest that management and conservation of declining burrowing owl populations should address factors that influence adult survival. © 2011 The Wildlife Society.  相似文献

Acute toxicity and sublethal effects of six commonly used insecticides, i.e., malathion, carbaryl, imidacloprid, cypermethrin, dimethoate, and monocrotophos, were evaluated through biological and life table parameters of Zygogramma bicolorata Pallister in laboratory. Concentration of these insecticides was within the minimum ranges of recommended field rate. Among the insecticides tested for acute toxicity, monocrotophos and imidacloprid caused the highest mortality of third instars and prolonged the development time of treated larvae. Fecundity and egg viability were also reduced in monocrotophos-treated group. Sublethal toxicity (carryover effect) of insecticides was evaluated through life table analysis of F1 progenies developed from surviving third instars treated for acute toxicity experiment. Survivorship was prolonged to 117 days in carbaryl treated group. Monocrotophos prolonged the overall immature development time compared to other insecticidal treatments and untreated control. Moreover, the lowest female survival, and the lowest value of life indices parameters, i.e., m x , R 0, r m, and λ, was evident in monocrotophos-exposed groups compared to that in other tested insecticides. However, mean generation time (T c) and doubling time (DT) were significantly prolonged in the insecticide-treated groups compared to those in the untreated group. A significantly greater number of females were produced in control groups than those in treated with malathion and sex ratio (proportion of male) was computed as 0.34 and 0.37, respectively. Based on the present study, it can be concluded that none of the tested insecticides can be classified as safe to Z. bicolorata. However, comparisons among the tested insecticides showed that malathion was less toxic compared to other insecticides tested.  相似文献

The developmental stages of the beech weevil, Rhynchaenus fagi L. are proposed as suitable material on which to base a practical study on some aspects of population dynamics. The life history of the weevil is explained, illustrating the convenience of the immature stages for the study of development and mortality in a population. The factors normally causing mortality during each of the developmental stages are described. Results obtained from the collection of a random sample of infested leaves are included as an example of the simplicity with which data can be obtained to illustrate the concept of the life table. The limitations of such a study as well as the possibilities for changing emphasis are briefly discussed.  相似文献

The virulence of different entomopathogenic nematode strains of the families Steinernematidae and Heterorhabditidae, isolates from Catalonia (NE Iberian Peninsula), and their symbiotic bacteria was assessed with regard to the larvae and adults of the hazelnut weevil, Curculio nucum L. (Coleoptera: Curculionidae). The nematode strains screened included one Steinernema affine, five Steinernema feltiae, one Steinernema carpocapsae, one Steinernema sp. (a new species not yet described) and one Heterorhabditis bacteriophora. The pathogenicity of all the strains of nematodes was tested on larvae and only four of them on adults of the hazelnut weevil. Larval mortality ranged from 10% with S. affine to 79% with Steinernema sp. Adult mortality was higher in S. carpocapsae, achieving 100% adult weevil mortality. The pathogenicity of the symbiotic bacteria Xenorhabdus bovienii, X. kozodoii, X. nematophila and Photorhabdus luminescens was studied in larvae and adults of C. nucum. In the larvae, X. kozodoii showed a LT50 of 22.7 h, and in the adults, it was 20.5 h. All nematodes species except S. affine tested against larvae showed great potential to control the insect, whereas S. carpocapsae was the most effective for controlling adults.  相似文献

The abundance and diversity of potential carabid predators of Sitona lineatus (L.) in field beans were monitored between 1980 and 1983. The absolute abundance of the large predatory species was determined in mark-recapture experiments and a radio-labelling technique was used to estimate the level of predation in the field. Using previously published population data for S. lineatus, the importance of predation in the population dynamics of the weevil was estimated. The mortality of weevil larvae due to predation varied from 0.6%– 10.5% while that of adult weevils varied from 2.6%– 23.8%. It appeared that carabids played a significant role in the population dynamics of S. lineatus and, in years when abundant, could reduce the population of larvae and overwintering adults by more than 30%.  相似文献

Abandonment of traditional land-use practices can have strong effects on the abundance of species occurring in agricultural landscapes. However, the precise mechanisms by which individual performance and population dynamics are affected are still poorly understood. To assess how abandonment affects population dynamics of Succisa pratensis we used data from a 4-year field study in both abandoned and traditionally grazed areas in moist and mesic habitats to parameterize integral projection models. Abandoned populations had a lower long-term stochastic population growth rate (λ S = 0.90) than traditionally managed populations (λ S = 1.08), while λ S did not differ between habitat types. The effect of abandonment differed significantly between years and had opposed effects on different vital rates. Individuals in abandoned populations experienced higher mortality rates and lower seedling establishment, but had higher growth rates and produced more flower heads per plant. Population viability analyses, based on a population survey of the whole study area in combination with our demographic models, showed that 32 % of the populations face a high risk of extinction (>80 %) within 20 years. These results suggest that immediate changes in management are needed to avoid extinctions and further declines in population sizes. Stochastic elasticity analyses and stochastic life table response experiments indicated that management strategies would be most effective if they increase survival of small plants as well as seedling establishment, while maintaining a high seed production. This may be achieved by varying the grazing intensity between years or excluding grazers when plants are flowering.  相似文献
