Microbial processes within the rhizosphere of constructed wetlands are crucial to wastewater treatment, but the relation between microbial community diversity in rhizosphere, plant growth and water quality are unclear at present. The effects of plant growth, water C:N:P ratio and their interaction on microbial diversity in the rhizosphere were studied in synthetic wastewater in planted and unplanted wetlands during three different seasons. The physiological profile of microbial community-level in each wetland was assessed using substrate utilization patterns gathered via BIOLOG? ECO plates. Plant had a significant effect on AWCD parameter, since the planted wetlands usually had a higher the total microbial activity than the unplanted over the study period. The Shannon, Simpson and McIntosh indices in the planted wetlands were apparently higher than those in the unplanted wetlands under any C:N:P ratio influent condition especially in summer. It was also shown that the unplanted wetlands have a greater shift of the interstitial microbial community than the planted at different seasons, since plant rhizospheres produce a more ecologically stable system in order to resist against shifts in microbial community composition in response to C:N:P ratio change in wastewater. Principal component analysis and clustering analysis indicated that influent C:N:P ratio would induce similar microbial species in the planted wetlands and detach them from the unplanted wetlands.  相似文献

This study investigates the small-scale stratification of bacterial community composition and functional diversity in the rhizosphere of maize. Maize seedlings were grown in a microcosm with a horizontal mesh (53 M) creating a planar root mat and rhizosphere soil. An unplanted microcosm served as control. Thin slices of soil were cut at different distances from the mesh surface (0.2–5.0 mm) and analysed for bacterial community composition by PCR-DGGE (polymerase chain reaction-denaturing gradient gel electrophoresis) of 16S rDNA and tested for activities of different enzymes involved in C, N, P and S cycling. Bacterial community composition and microbial functional diversity were affected by the presence of the maize roots. The bacterial composition showed a clear gradient up to 2.2 mm from the root surface, while no such gradient was observed in the unplanted pot. Invertase and phosphatase activities were higher in the close vicinity of maize roots (0.2–0.8 mm), whereas xylanase activity was unaffected. This study shows that the changes in bacterial community composition and functional diversity induced by roots may extend several millimetres into the soil.  相似文献

Microorganisms that use nitrate as an alternative terminal electron acceptor play an important role in the global nitrogen cycle. The diversity of the nitrate-reducing community in soil and the influence of the maize roots on the structure of this community were studied. The narG gene encoding the membrane bound nitrate reductase was selected as a functional marker for the nitrate-reducing community. The use of narG is of special interest because the phylogeny of the narG gene closely reflects the 16S ribosomal DNA phylogeny. Therefore, targeting the narG gene provided for the first time a unique insight into the taxonomic composition of the nitrate-reducing community in planted and unplanted soils. The PCR-amplified narG fragments were cloned and analyzed by restriction fragment length polymorphism (RFLP). In all, 60 RFLP types represented by two or more clones were identified in addition to the 58 RFLP types represented by only one clone. At least one clone belonging to each RFLP type was then sequenced. Several of the obtained sequences were not related to the narG genes from cultivated bacteria, suggesting the existence of unidentified nitrate-reducing bacteria in the studied soil. However, environmental sequences were also related to NarG from many bacterial divisions, i.e., Actinobacteria and alpha, beta, and gamma proteobacteria. The presence of the plant roots resulted in a shift in the structure of the nitrate-reducing community between the unplanted and planted soils. Sequencing of RFLP types dominant in the rhizosphere or present only in the rhizosphere revealed that they are related to NarG from the Actinobacteria in an astonishingly high proportion.  相似文献

芒草作为第二代能源植物的代表,其生长过程中根际土壤细菌群落的结构与功能尚不清楚.本研究以种植5年的芒草(湘杂芒1号)为研究对象,选取裸地作为对照,采用16S rRNA基因Miseq测序技术研究其细菌群落组成,同时采用PICRUSt功能预测分析其功能.结果表明: 芒草根际细菌由变形菌门、酸杆菌门、放线菌门、绿弯菌门、拟杆菌门和芽单胞菌门等23个门、231个属的细菌组成,表现出群落组成的丰富性.细菌群落分析表明,种植湘杂芒1号改变了根际细菌群落结构,其细菌群落多样性低于裸地对照.PICRUSt功能预测分析表明,湘杂芒1号根际细菌主要涉及氨基酸运输和代谢、细胞壁/细胞膜/膜结构的生物合成、信号转导机制等24个基因功能家族,表现出功能上的丰富性,并有22个基因功能家族预测基因相对丰度高于裸地,表明种植湘杂芒1号提高了根际细菌功能.对氮、磷循环相关基因进行分析表明,种植湘杂芒1号改变了土壤氮、磷代谢能力.  相似文献

Sixty-six atrazine-degrading bacterial communities utilizing atrazine as sole N source and citrate as principal C source were isolated from unplanted and maize planted soils treated with atrazine. Ribosomal intergenic spacer analysis (RISA) fingerprints revealed that the genetic structure of atrazine-degrading bacterial communities was modified in the maize rhizosphere. To assess the underlying microbial diversity, 16S rDNA sequences amplified from each bacterial community were cloned. Libraries containing 660 16S rDNA clones were screened by restriction fragment length polymorphism (RFLP) analysis. In all, 63 clone families were identified. Rarefaction curves did not reach a clear saturation, indicating that the analysis of a greater number of clones would have revealed further diversity. Recovered 16S rDNA sequences were related to Actinobacteria, Bacteroidetes and Proteobacteria. The four dominant RFLP families were highly similar to Variovorax paradoxus, Burkholderia cepacia, Arthrobacter sp. and Bosea sp. The composition of most of the atrazine-degrading bacterial communities consisted of 2–7 different bacterial species. Various atrazine-degrading gene compositions were observed, two of these atzABCDEF, trzND and atzBCDEF, trzN being largely dominant. The first was more frequently detected in bacterial communities isolated from the maize rhizosphere whereas the second was more frequently detected in communities isolated from bulk soil. Monitoring of atrazine-degrading activity showed that 76% of the bacterial communities degraded up to 80% of the initially added atrazine within 15 days of culture. Altogether our results indicate that the maize rhizosphere has an impact on the genetic structure, the diversity and atrazine-degrading gene composition of the atrazine-degrading communities.  相似文献

Soils may comprise tens of thousands to millions of bacterial species. It is still unclear whether this high level of diversity is governed by functional redundancy or by a multitude of ecological niches. In order to address this question, we analyzed the reproducibility of bacterial community composition after different experimental manipulations. Soil lysimeters were planted with four different types of plant communities, and the water content was adjusted. Group-specific phylogenetic fingerprinting by PCR-denaturing gradient gel electrophoresis revealed clear differences in the composition of Alphaproteobacteria, Betaproteobacteria, Bacteroidetes, Chloroflexi, Planctomycetes, and Verrucomicrobia populations in soils without plants compared to that of populations in planted soils, whereas no influence of plant species composition on bacterial diversity could be discerned. These results indicate that the presence of higher plant species affects the species composition of bacterial groups in a reproducible manner and even outside of the rhizosphere. In contrast, the environmental factors tested did not affect the composition of Acidobacteria, Actinobacteria, Archaea, and Firmicutes populations. One-third (52 out of 160) of the sequence types were found to be specifically and reproducibly associated with the absence or presence of plants. Unexpectedly, this was also true for numerous minor constituents of the soil bacterial assemblage. Subsequently, one of the low-abundance phylotypes (beta10) was selected for studying the interdependence under particular experimental conditions and the underlying causes in more detail. This so-far-uncultured phylotype of the Betaproteobacteria species represented up to 0.18% of all bacterial cells in planted lysimeters compared to 0.017% in unplanted systems. A cultured representative of this phylotype exhibited high physiological flexibility and was capable of utilizing major constituents of root exudates. Our results suggest that the bacterial species composition in soil is determined to a significant extent by abiotic and biotic factors, rather than by mere chance, thereby reflecting a multitude of distinct ecological niches.  相似文献

A coupling of above-ground plant diversity and below-ground microbial diversity has been implied in studies dedicated to assessing the role of macrophyte diversity on the stability, resilience, and functioning of ecosystems. Indeed, above-ground plant communities have long been assumed to drive below-ground microbial diversity, but to date very little is known as to how plant species composition and diversity influence the community composition of micro-organisms in the soil. We examined this relationship in fields subjected to different above-ground biodiversity treatments and in field experiments designed to examine the influence of plant species on soil-borne microbial communities. Culture-independent strategies were applied to examine the role of wild or native plant species composition on bacterial diversity and community structure in bulk soil and in the rhizosphere. In comparing the influence of Cynoglossum officinale (hound's tongue) and Cirsium vulgare (spear thistle) on soil-borne bacterial communities, detectable differences in microbial community structure were confined to the rhizosphere. The colonisation of the rhizosphere of both plants was highly reproducible, and maintained throughout the growing season. In a separate experiment, effects of plant diversity on bacterial community profiles were also only observed for the rhizosphere. Rhizosphere soil from experimental plots with lower macrophyte diversity showed lower diversity, and bacterial diversity was generally lower in the rhizosphere than in bulk soil. These results demonstrate that the level of coupling between above-ground macrophyte communities and below-ground microbial communities is related to the tightness of the interactions involved. Although plant species composition and community structure appear to have little discernible effect on microbial communities inhabiting bulk soil, clear and reproducible changes in microbial community structure and diversity are observed in the rhizosphere. As the major crops in north China, spring crops are usually planted from April through May every spring and harvested in fall. Wheat is also a very common crop traditionally planted in fall or spring and harvested in summer year by year. This continuous cropping system exhibited the disadvantages of reducing the fertility of soil through decreasing microbial diversity. Thus, management of microbial diversity in the rhizosphere plays a vital role in sustainable crop production. In this study, ten common spring crops in north China were chosen sole-cropped and four were chosen intercropped with peanut in wheat fields after harvest. Denaturing gradient gel electrophoresis (DGGE) and DNA sequencing of one 16S rDNA fragment were used to analyze the bacterial diversity and species identification. DGGE profiles showed the bacterial community diversity in rhizosphere soil samples varied among various crops under different cropping systems, more diverse under intercropping system than under sole-cropping. Some intercropping-specific bands in DGGE profiles suggested that several bacterial species were stimulated by intercropping systems specifically. Furthermore, the identification of these dominant and functional bacteria by DNA sequencing indicated that intercropping systems are more beneficial to improve soil fertility. Compared to intercropping systems, we also observed changes in microbial community of rhizosphere soil under sole-crops. The rhizosphere bacterial community structure in spring crops showed a strong crop species-specific pattern. More importantly, Empedobacter brevis, a typical plant pathogen, was only found in the carrot rhizosphere, suggesting carrot should be sown prudently. In conclusion, our study demonstrated that crop species and cropping systems had significant effects on bacterial community diversity in the rhizosphere soils. We strongly suggest sorghum, glutinous millet and buckwheat could be taken into account as intercropping crops with peanut; while hulled oat, mung bean or foxtail millet could be considered for sowing in wheat fields after harvest in North China.  相似文献

The bacterial community composition in soil and rhizosphere taken from arable field sites, differing in soil parent material and soil texture, was analyzed using terminal restriction fragment length polymorphism (T-RFLP) of 16S rRNA genes. Nine sandy to silty soils from North-East Germany could clearly be distinguished from each other, with a relatively low heterogeneity in the community structure within the field replicates. There was a relationship between the soil parent material, i.e. different glacial and aeolian sediments, and the clustering of the profiles from different sites. A site-specific grouping of T-RFLP profiles was also found for the rhizosphere samples of the same field sites that were planted with potatoes. The branching of the rhizosphere profiles corresponded partly with the soil parent material, whereas the effect of the plant genotype was negligible. Selected terminal restriction fragments differing in their relative abundance within the nine soils were analyzed based on the cloning of the 16S rRNA genes of one soil sample. A high phylogenetic diversity observed to include Acidobacteria, Betaproteobacteria, Bacteroidetes, Verrucomicrobia, and Gemmatimonadetes. The assignment of three out of the seven selected terminal restriction fragments to members of Acidobacteria suggested that this group seems to participate frequently in the shifting of community structures that result from soil property changes.  相似文献

A greenhouse study was conducted to monitor microbial community dynamics related to contaminant concentration and plant growth during the phytoremediation of pyrene. Soil microbial communities in both bulk and rhizosphere soils were assessed using a polymerase chain reaction--denaturing gradient gel electrophoresis (PCR-DGGE). However, no correlation between pyrene concentration and bacterial community shifts was observed. Even though plants significantly enhanced pyrene degradation (undetectable after 91 d for planted treatments compared with 82 mg/kg for unplanted treatments after 147 d with initial concentration of 758 mg/kg), biodegradation may have been accomplished by the pre-existing microbial community. It is also possible that the method was unable to pick up subtle community shifts, considering that the carbon source from pyrene was only 3.7% of the existing soil organic matter. This research suggests that plants significantly enhance degradation and mineralization of pyrene in soil, although there was no conclusive evidence supporting any specific bacterial activity responsible for this enhancement.  相似文献

Root-microbe interactions are considered to be the primary process of polyaromatic hydrocarbon (PAH) phytoremediation, since bacterial degradation has been shown to be the dominant pathway for environmental PAH dissipation. However, the precise mechanisms driving PAH rhizostimulation symbiosis remain largely unresolved. In this study, we assessed PAH degrading bacterial abundance in contaminated soils planted with 18 different native Michigan plant species. Phenanthrene metabolism assays suggested that each plant species differentially influenced the relative abundance of PAH biodegraders, though they generally were observed to increase heterotrophic and biodegradative cell numbers relative to unplanted soils. Further study of >1800 phenanthrene degrading isolates indicated that most of the tested plant species stimulated biodegradation of a broader range of PAH compounds relative to the unplanted soil bacterial consortia. These observations suggest that a principal contribution of planted systems for PAH bioremediation may be via expanded metabolic range of the rhizosphere bacterial community.  相似文献

The ability of facultative anerobes to respire nitrate has been ascribed mainly to the activity of a membrane-bound nitrate reductase encoded by the narGHJI operon. Respiratory nitrate reduction is the first step of the denitrification pathway, which is considered as an important soil process since it contributes to the global cycling of nitrogen. In this study, we employed direct PCR, cloning, and sequencing of narG gene fragments to determine the diversity of nitrate-reducing bacteria occurring in soil and in the maize rhizosphere. Libraries containing 727 clones in total were screened by restriction fragment analysis. Phylogenetic analysis of 128 narG sequences separated the clone families into two main groups that represent the Gram-positive and Gram-negative nitrate-reducing bacteria. Novel narG lineages that branch distinctly from all currently known membrane bound nitrate-reductase encoding genes were detected within the Gram-negative branch. All together, our results revealed a more complex nitrate-reducing community than did previous culture-based studies. A significant and consistent shift in the relative abundance of the nitrate-reducing groups within this functional community was detected in the maize rhizosphere. Thus a substantially higher abundance of the dominant clone family and a lower diversity index were observed in the rhizosphere compared to the unplanted soil, suggesting that a bacterial group has been specifically selected within the nitrate-reducing community. Furthermore, restriction fragment length polymorphism analysis of cloned narG gene fragments proved to be a powerful tool in evaluating the structure and the diversity of the nitrate-reducing community and community shifts therein.  相似文献

对山东菏泽赵楼牡丹园栽培的不同品种牡丹(Paeonia suffruticosa)根围丛枝菌根(ar-buscular mycorrhiza,AM)真菌孢子密度、种属构成、种丰度、分布频度、物种多样性指数及其物种组成相似性等进行了研究.结果表明:不同品种牡丹根围AM真菌的种属构成、种丰度和分布频度等均不相同,其中,从‘凤丹'和‘赵粉'根围中各分离到10种AM真菌,从‘乌龙捧盛'和‘洛阳红'根围中分离到9种,从‘胡红'根围中分离到8种;‘凤丹'根围AM真菌孢子密度最高,为59个·50 g-1土,‘胡红'最低,为47个·(50 g)-1土;‘赵粉'的物种多样性指数最高(1.89),‘胡红'最低(1.71).‘凤丹'和‘胡红'的菌根侵染率最高,为63.6%,‘乌龙捧盛'最低,为52.7%.不同牡丹品种之间AM真菌种类组成的相似性系数在0.71~0.95,其中‘乌龙捧盛'和‘凤丹'的相似性系数最高(0.95),而‘胡红'与‘洛阳红'的相似性系数最低(0.71).牡丹基因型能改变AM真菌群落结构特征.  相似文献

To examine the relationship between plant species composition and microbial community diversity and structure, we carried out a molecular analysis of microbial community structure and diversity in two field experiments. In the first experiment, we examined bacterial community structure in bulk and rhizosphere soils in fields exposed to different plant diversity treatments, via a 16S rRNA gene clone library approach. Clear differences were observed between bacterial communities of the bulk soil and the rhizosphere, with the latter containing lower bacterial diversity. The second experiment focused on the influence of 12 different native grassland plant species on bacterial community size and structure in the rhizosphere, as well as the structure of Acidobacteria and Verrucomicrobia community structures. In general, bacterial and phylum-specific quantitative PCR and PCR-denaturing gradient gel electrophoresis revealed only weak influences of plant species on rhizosphere communities. Thus, although plants did exert an influence on microbial species composition and diversity, these interactions were not specific and selective enough to lead to major impacts of vegetation composition and plant species on below-ground microbial communities.  相似文献

The aim of this study was to investigate the removal of ibuprofen in laboratory scale constructed wetlands. Four (planted and unplanted) laboratory‐scale horizontal subsurface flow constructed wetlands were supplemented with ibuprofen in order to elucidate (i) the role of plants on ibuprofen removal and (ii) to evaluate the removal performance of a bioaugmented lab scale wetland. The planted systems showed higher ibuprofen removal efficiency than an unplanted one. The system planted with Juncus effusus was found to have a higher removal rate than the system planted with Phalaris arundinacea. The highest removal rate of ibuprofen was found after inoculation of gravel previously loaded with a newly isolated ibuprofen‐degrading bacterium identified as Sphingobium yanoikuyae. This experiment showed that more than 80 days of CW community adaptation for ibuprofen treatment could be superseded by bioaugmentation with this bacterial isolate.  相似文献

为探究不同积累型小麦品种对根际微生物群落结构及功能多样性的影响,以镉低积累型小麦济麦22和镉高积累型小麦冀5265为研究材料,采用分离培养法和Biolog-Eco微平板法分析根际细菌数量、可培养优势群落结构以及微生物群落功能多样性。结果表明:污染土壤济麦22根际总细菌数量和抗Cd细菌数量均显著高于冀5265,而非污染土壤中两品种间无差异。污染土济麦22根际发现较多产脲酶和高镉抗性菌株(200 mg/L)。污染土济麦22根际优势菌多为Arthrobacter sp.和Bacillus sp.,冀5265根际优势菌主要为Streptomyces sp.;非污染土济麦22与冀5265根际优势菌群相似,均以Bacillus sp.为主。Biolog试验结果表明,两个小麦品种根际微生物群落对碳源的利用能力存在差异,济麦22根际微生物AWCD值、Mc Intosh指数、Shannon-Wiener指数、Simpson指数在污染土和无污染土中均显著高于冀5265。因此,污染土壤中不同积累型小麦品种根际微生物群落结构及功能多样性均存在差异,该研究结果对于揭示高低积累型小麦根际微生物机制提供了重要参考依...  相似文献

We used a next-generation, Illumina-based sequencing approach to characterize the bacterial community development of apple rhizosphere soil in a replant site (RePlant) and a new planting site (NewPlant) in Beijing. Dwarfing apple nurseries of ‘Fuji’/SH6/Pingyitiancha trees were planted in the spring of 2013. Before planting, soil from the apple rhizosphere of the replant site (ReSoil) and from the new planting site (NewSoil) was sampled for analysis on the Illumina MiSeq platform. In late September, the rhizosphere soil from both sites was resampled (RePlant and NewPlant). More than 16,000 valid reads were obtained for each replicate, and the community was composed of five dominant groups (Proteobacteria, Acidobacteria, Bacteroidetes, Gemmatimonadetes and Actinobacteria). The bacterial diversity decreased after apple planting. Principal component analyses revealed that the rhizosphere samples were significantly different among treatments. Apple nursery planting showed a large impact on the soil bacterial community, and the community development was significantly different between the replanted and newly planted soils. Verrucomicrobia were less abundant in RePlant soil, while Pseudomonas and Lysobacter were increased in RePlant compared with ReSoil and NewPlant. Both RePlant and ReSoil showed relatively higher invertase and cellulase activities than NewPlant and NewSoil, but only NewPlant soil showed higher urease activity, and this soil also had the higher plant growth. Our experimental results suggest that planting apple nurseries has a significant impact on soil bacterial community development at both replant and new planting sites, and planting on new site resulted in significantly higher soil urease activity and a different bacterial community composition.  相似文献

Removal of pharmaceutical compounds in tropical constructed wetlands   总被引:2,自引:0,他引:2  
The ability of tropical horizontal subsurface constructed wetlands (HSSF CWs) planted with Typha angustifolia to remove four widely used pharmaceutical compounds (carbamazepine, declofenac, ibuprofen and naproxen) at the relatively short hydraulic residence time of 2-4 days was documented. For both ibuprofen and naproxen, pharmaceutical compounds with low Dow values, the planted beds showed significant (p < 0.05) enhancement of removal efficiencies (80% and 91%, respectively, at the 4 day HRT), compared to unplanted beds (60% and 52%, respectively). The presence of plants resulted in the removal of these pharmaceutical compounds from artificial wastewater. The more oxidizing environment in the rhizosphere might have played an important role, but other rhizosphere effects, beside rhizosphere aeration, appeared to be important also. Carbamazepine, considered one of the most recalcitrant pharmaceuticals, and declofenac showed low removal efficiencies in our CW, and this is attributable to their higher hydrophobicity. The fact that the removal of these compounds could be explained by the sorption onto the available organic surfaces, explains why there was no significant difference (p > 0.05) in their removal efficiencies between planted as compared to unplanted beds. No statistical significant differences (p > 0.05) were observed for the removal efficiencies of any of the pharmaceuticals tested for the 2-day HRT as compared to that corresponding to 4-day HRT. The rather efficient removal shown by the wetlands in this study (with HRTs of 2-4 days), indicates that such a CW system may be more practically used (with less land requirements) in tropical regions for removing conventional pollutants and certain pharmaceutical compounds from wastewater effluents.  相似文献

Soil microbes are known to be key drivers of several essential ecosystem processes such as nutrient cycling, plant productivity and the maintenance of plant species diversity. However, how plant species diversity and identity affect soil microbial diversity and community composition in the rhizosphere is largely unknown. We tested whether, over the course of 11 years, distinct soil bacterial communities developed under plant monocultures and mixtures, and if over this time frame plants with a monoculture or mixture history changed in the bacterial communities they associated with. For eight species, we grew offspring of plants that had been grown for 11 years in the same field monocultures or mixtures (plant history in monoculture vs. mixture) in pots inoculated with microbes extracted from the field monoculture and mixture soils attached to the roots of the host plants (soil legacy). After 5 months of growth in the glasshouse, we collected rhizosphere soil from each plant and used 16S rRNA gene sequencing to determine the community composition and diversity of the bacterial communities. Bacterial community structure in the plant rhizosphere was primarily determined by soil legacy and by plant species identity, but not by plant history. In seven of the eight plant species the number of individual operational taxonomic units with increased abundance was larger when inoculated with microbes from mixture soil. We conclude that plant species richness can affect below‐ground community composition and diversity, feeding back to the assemblage of rhizosphere bacterial communities in newly establishing plants via the legacy in soil.  相似文献
