Potato plants (Solanum tuberosum L.) were grown in water culturein a controlled environment. Cooling (+8°C) of individualtubers decreased their growth rates and increased the growthrates of non-cooled tubers of the same plant. The carbohydrateconcentration in non-cooled and cooled tubers did not differsignificantly, but 14C-import from labelled photosynthate waslower in cooled than in non-cooled tubers. The markedly lowerconversion rate of ethanol-soluble 14C to starch in cooled,in comparison to non-cooled tubers, was not associated withsignificant differences in the in vitro activities of starchsynthase, ADPG-pyrophosphorylase and starch phosphorylase understandard assay conditions (+30°C). However, the Q10-valuesof the enzymes differed in vitro in the temperature range between30°C and 8°C, leading to a marked decrease in the activityratio of ADPG-pyrophosphorylase/starch phosphorylase in cooledtubers. In tubers differing in growth rates without manipulation, 14d after tuber initiation significant positive correlations werefound between 14C-concentration of tuber tissue and the in vitroactivities of starch synthase and ADPG-pyrophosphorylase anda significant negative correlation between 14C-concentrationand starch phosphorylase. In contrast, in tubers which wereanalysed 5 d after initiation, there were only small differencesbetween tubers in growth rate, 14C import and the activity ratioADPG-pyrophosphorylase/starch phosphorylase. From various directand indirect evidence it is concluded that the growth rate ofindividual tubers, and thus the sink strength, is at least inpart controlled by the activity of starch synthesizing enzymes. Cells of Anacystis nidulans grown at 30°C were incubatedwith 14C-Chlorella protein hydrolysate at the elevated temperatures(30–55°C) and the effect of heat shock treatment onprotein synthesis was studied. Incubation temperatures higherthan 45°C caused a significant decrease in the incorporationof amino acids into proteins. Sunflower plants (Helianthus annuus L.) grown at 30°C werecooled to 13°C in the light in atmospheric CO2 or low CO2,or in darkness. Photosynthetic rate at 30°C after coolingwhole plants in atmospheric CO2 for 12 h during a photoperiodwas significantly lower than at the start of the photoperiodcompared to plants cooled at low CO2, those cooled in the darkand those maintained at 30°C. Amounts of sucrose, hexosesand starch in leaves at 13°C increased throughout a 14 hphotoperiod to levels higher than in leaves at 30°C, whereamounts of sucrose and hexoses were stable or falling after4 h. Carbohydrate accumulation at 13°C during this photoperiodwas more than twice that at 30°C. After three photoperiodsand two dark periods at 13°C carbohydrate levels in leaveswere still as high as at the end of the first photoperiod, butless carbohydrate accumulated during the photoperiods than duringthe first photoperiod, and more was partitioned as starch. Amountsof soluble carbohydrate in roots were greater after 14 h at13°C than in roots of plants at 30°C. Loss of 14C fromleaves at 30°C as a proportion of 14CO2 fixed by them at30°C, decreased after exposure of plants to 13°C inthe light for 30 min prior to 14CO2feeding. Results indicatean effect of cold on the transport process that was light-dependent.It is inferred that the reduction in the proportion of 14C lostfrom leaves after 10 h cooling was due to reduced sink demand,whereas the rise in the proportion of 14C lost from leaves after24 h reflects reduced photosynthetic rate. The coincidence ofreduced photosynthetic rate with raised carbohydrate levelsin leaves maintained at 30°C throughout, whilst the restof the plant was cooled to 13°C in the light implies feedbackinhibition of photosynthesis. Excised florets of some hardy Rhododendron species did not toleratefreezing at –5°C when ice-inoculated due to intracellularfreezing. Florets in intact December buds, however, could besupercooled to about –30°C. When flower buds of R.japonicum were slowly cooled with daily decrements of 5°Cto temperatures ranging from 0 to –20°C, the exothermtemperatures of the florets drastically decreased. This wasaccompanied by a decrease in water content of florets and peduncleand an increase in that of scales. The water in florets andthe peduncle is thought to migrate to scales and other tissuesduring the early stages of freezing; the dehydrated floret hasa lower freezing point which enhances its supercooling abilityand the dehydrated peduncle helps to maintain the supercooledstate of the florets. This hypothesis would explain the dependenceon the cooling rate of supercooling in Rhododendron flower buds.Water migration within flower buds was observed in other hardyRhododendron species with some variation in ice formation siteand the quantity of migrated water. The exotherm temperatureof excised florets was inversely proportional to their watercontent. Dehydration of flower buds by wind at 0°C alsoenhanced their supercooling ability. Nodulated white clover plants (Trifolium repens L. cv. Huia)were grown for 71 d in flowing nutrient solutions containingN as 10 mmol m–3 NH4NO3, under artificial illumination,with shoots at 20/15°C day/night temperatures and root temperaturereduced decrementally from 20 to 5°C. Root temperatureswere then changed to 3, 7, 9, 11, 13, 17 or 25°C, and theacquisition of N by N2 fixation, NH4+ and NO3 uptakewas measured over 14 d. Shoot specific growth rates (d. wt)doubled with increasing temperature between 7 and 17°C,whilst root specific growth rates showed little response; shoot:root ratios increased with root temperature, and over time at11°C. Net uptake of total N per plant (N2 fixation + NH4++ NO3) over 14 d increased three-fold between 3 and 17°C.The proportion contributed by N2 fixation decreased with increasingtemperature from 51% at 5°C to 18% at 25°C. Uptake ofNH4+ as a proportion of NH4+ + NO3 uptake over 14 d variedlittle (55–62%) with root temperature between 3 and 25°C,although it increased with time at most temperatures. Mean ratesof total N uptake per unit shoot f. wt over 14 d changed littlebetween 9 and 25°C, but decreased progressively with temperaturebelow 9°C, due to the decline in the rates of NH4+ and NO3uptake, even though N2 fixation increased. The results suggestthat N2 fixation in the presence of sustained low concentrationsof NH4+ and NO4 is less sensitive to low root temperaturethan are either NH4+ or NO3 uptake systems. Some Low-temperature Effects on Sieve Tube Translocation in Salix viminalis   总被引:1,自引:0,他引:1  
The data presented on translocation of 14C-labelled compoundsin Salix viminalis show that above a temperature close to –4°C translocation occurs, whilst below –4 °C inmost cases it does not. Stoppages were irreversible when the temperature of the cooledstem was raised again to normal (approximately 20 °C) andappeared to involve some disruption of the phloem. The temperaturesat which the bark was found to freeze were found to be closelysimilar, with respect to level and variability, to the stoppingtemperature. Respiration measurements on isolated strips of bark at –2°C showed that oxygen uptake fell to approximately 5 percent of its value at 25 °C. The decreased level of radioactivity in the cooled regions ofthe stems is considered to be evidence that the exchange oflabelled compounds between the sieve tubes and the surroundingcells was slowed up at low temperatures.  相似文献

The effects of growth and assay temperature on unidirectionalK+ fluxes in excised roots of rye (Secale cereale cv. Rheidol)were studied using 86Rb+ as a tracer. Both K+ influx to thevacuole, estimated as K+ uptake between 3 and 12 h after transferof unlabelled roots to radioactive solution, and movement ofK+ to the xylem were determined directly. Other fluxes weredetermined on excised roots of plants, which had been labelledwith 86Rb+ since germination, by conventional triple exponentialefflux analysis. When assayed at 20°C, roots of plants previously grown at20°C(WG roots) had lower rates of net K+ uptake than rootsof low temperature-acclimated plants, grown with a temperaturediferential between roots (87°C) and shoots (20°C) eithersince germination (DG roots) or for 3 d prior to experiments(DT roots). This resulted from a greater unidirectional K+ effluxacross the plasma membrane and a reduced K+ flux to the xylemin WG roots, compared to DG or DT roots, rather than a decreasein unidirectional K+ influx or a decrease in the net K+ fluxto the vacuole. Indeed, although WG roots had lower rates ofK+ influx and K+ efflux across the tonoplast at 20°C thanDG or DT roots, roots of plants from all growth temperaturetreatments showed an equivalent net K+ flux to the vacuole. Although all unidirectional K+ fluxes in roots from plants grownunder all temperature regimes were reduced by lowering the temperatureof the root, these fluxes were differentially affected in rootsof plants from contrasting growth temperature treatments. Rapidcooling to 8°C of WG roots resulted in a lower rate of K+influx and a transient increase in K+ efflux across both theplasma membrane and tonoplast, compared to DG and DT roots.Furthermore, since the K+ flux to the xylem was lower in WGroots, the net K+ uptake at 8°C into WG roots was considerablyreduced compared to DG and DT roots. These results suggest thatlow temperature-acclimation of K+ fluxes in rye roots may involvea reduction in the temperature sensitivity of K+ influx anda curtailment of K+ efflux across both the plasma membrane andtonoplast at low temperatures. A cryophilic strain of the unicellular green alga Chlamydomonas, C. nivalis was found to be more resistant to the stresses both of freezing and thawing and of shrinkage and rehydration than was a mesophilic strain C. reinhardii. C. nivalis was found to have a higher degree of unsaturation of phospholipid fatty acids. Following freezing and thawing of C. reinhardii there was a direct correlation between reduction in cell viability and loss of membrane selective permeability. Activation of intracellular phospholipases occurred at an early stage of freezing injury. Attempts to cold harden C. reinhardii were unsuccessful. For C. reinhardii methanol was the only effective cryoprotectant for freezing to and thawing from ?196 °C and the effects of cooling rate upon cellular survival are presented.  相似文献

Nutritional requirements, functional response, development andreproduction of Cyclops vicinus were studied with exclusivelyalgal food. Phytoflagellates were found to be adequate foodresources for both juvenile development and egg production.Ingestion measurements were performed with Chlamydomonas reinhardii.The functional response data give evidence for low feeding efficiency,especially for the naupliar stages. A difference between naupliiand older instars was also found in their quantitative foodneeds. A higher threshold food concentration was observed fornaupliar development (0.4 mg C l–1) than for copepoditedevelopment (0.2 mg C l–1). The calculation of assimilationefficiencies suggests that the high food requirements are dueto low specific ingestion rates rather than poor assimilationefficiency. Development time decreased as algal density increasedand males developed more quickly than females at all food concentrations.Body size and carbon content increased with increasing foodconcentration. Continuous egg production was observed abovea food concentration of 0.5 mg C l–1. The results haveimplications for the life cycle of C. vicinus. Summer diapauseis interpreted as a strategy to avoid starvation of the juvenilestages. Acclimation of NO3 transport fluxes (influx, efflux)in roots of oilseed rape (Brassica napus L. cv. Bien venu) andtheir sensitivity to growth at low root temperature was studiedin relation to external NO3 supply, defined by constantconcentrations ranging from sub- to supra-optimal with respectto plant growth rate. Plants were grown from seed in flowingnutrient solutions containing 250 mmol m–3 NO3at 17°C for 20d, and solution temperature in half the cultureunits was then lowered decrementally over 3 d to 7°C. Threedays later plants were supplied with NO3 at 1, 10, 100or 1000 mmol m–3 maintained for 18 d. Dry matter productionwas decreased more by low root zone temperature than low [NO3]e. Root specific growth rates were inversely related to [NO3]eand shoot:root ratios increased with time at [NO3]e between10–1000 mmol m–3. Net uptake of NO3 at 17°Cwas twice that at 7°C, and at both temperatures it doubledwith increasing [NO3]e between 1–10 mmol m–3with further small increases at higher [NO3]e. Mean unitabsorption rates of NO3 between 0–6 d and 6–14d were linearly related (r2 of 0.79–0.99) to log10[NO].Steady-state Q10 (7–17°C) for uptake between 0–6d were 0.91, 1.62, 1.27, and 1.10, respectively, at [NO3]eof 1, 10, 100, and 1000 mmol m–3, compared with correspondingvalues of 0.98, 1.38, 1.68, and 1.89 between 6–14 d. Thedata indicated that net uptake rates at 7 and 17°C divergedover time at high [NO3]e. Short-term uptake rates from1 mol m–3 NO3 measured at 17°C were higherin plants grown with roots at 7°C than at 17°C; for7°C plants there was a strong inverse linear relationship(r2=0.94) between uptake rate and treatment log10 [NO3]ewhilst rates in 17°C plants were independent of prior [NO3]e. Rates of NO3 influx and efflux under different steady-stateconditions of NO3 supply and root temperature were calculatedfrom dilution of 15N added to culture solutions. Efflux wassubstantial relative to net uptake in all treatments, and wasinversely related to [NO3]e at 17°C but not at 7°C.Ratios of influx: efflux ranged from 1.6–2.9 at 17°Cand 1.3–1.8 at 7°C, indicating the proportionatelygreater impact of efflux at low root temperature. Ratios ofefflux: net uptake were 0.53–1.56 at 17°C and 1.21–3.58at 7°C. The apparent sensitivities of influx and effluxto steady-state root temperature varied with [NO3]e.Both fluxes were higher at 17°C than 7°C in the presenceof 100–1000 mmol m–3 NO3 but the trend wasreversed at 1–10 mmol m–3 NO. Concentrations oftotal N measured in xylem exudate were at least 2-fold higherat 7°C compared with 17°C, attributable mainly to higherconcentrations of NO3 glutamine and proline. The resultsare discussed in terms of acclimatory and other responses shownby the NO3 transport system under conditions of limitingNO3 supply and low root temperature. The rate of carbon import by tomato fruits has been relatedto their carbon metabolism by examining the effects of fruittemperature on the metabolism of imported assimilates. 14C–sucrose,–glucose, –fructose, –malic acid and –citricacid were injected individually into young growing tomato fruitswhich were subsequently maintained at 25 or 5 °C for 48h. Fruit temperature greatly affected the proportions of 14Clost from the fruits by export and respiration. Only 40 percent of the injected 14C from 14C–sugars and 20 per centfrom 14C–acids was recovered from fruits at 25 °C.Less than 10 per cent of the injected 14C was exported, thebalance being respired. In contrast, more than 50 per cent ofthe injected 14C was recovered from cooled fruits, in whichthe import rate of carbon was presumably reduced, and 20–36per cent of injected 14C was exported. Cooling enhanced thesynthesis of 14C–sucrose from injected 14C–hexosesand inhibited the incorporation of 14C into starch and insolubleresidue. When 14C–sugars were injected, radioactivityexported from the cooled fruits was detected as sucrose in thephloem of the peduncles; radioactivity was also detected instems and roots when fruits were cooled. In almost fully–grownfruits injected 14C–compounds were metabolized less readilythan in smaller fruits. Conversion of 14C–hexoses to 14C–sucrosewas again enhanced by cooling (5 °C, but was less in fruitsmaintained at 35 °C than in controls. Freezing of healthy and rust (Puccinia lagenophorae) infectedleaves of Senecio vulgaris was compared calorimetrically bythermal analysis. In fully expanded leaves the threshold freezingtemperature was in the range –6.8 to –8.4 °Cin controls but –3.0 to –5.1 °C in leaves withsporulating rust sori. Comparable values in expanding leaveswere –5.0 to –8.9 °C and –3.9 to –6.7°C for healthy and rusted tissues, respectively. The bulktissue freezing point was between –1.0 and –4.0°C in both fully expanded and expanding healthy leaves,and was increased by infection by between +0.2 and 2.5 °C.Whereas healthy leaves supercooled by 3.1–5.8 °C,rusted leaves supercooled by only 1.8–4.9 °C Supercoolingof control leaves was reduced by dusting with aeciospores, particularlywhen leaves were wounded to simulate the rupture of the surfacecaused by sporulation, but wounding alone had no significanteffect. Supercooling of distilled water was also significantlyreduced by aeciospores, suspended at a concentration of 105spores ml–1. It is concluded that rust-induced changes in leaf freezing inS. vulgaris grown in controlled environments were due to anincrease in the number of sites for ice nucleation, caused bythe presence of the aeciospores, and increased penetration ofice into internal tissues, resulting from damage to the cuticleand epidermis. Although data for frost resistance obtained inthe growth-room are similar to previous field observations,the role of the above mechanisms under field conditions remainsunproven. Respiratory activity and seasonal changes in carbohydrate contentof the storage organs of Rubus chamaemorus L. have been investigated.Leaf dark respiration rate increases in a non-linear mannerfrom 0·7 mg CO2 evolved dm–2 h–1 at 0 °Cto 4·6 rng CO2 evolved dm–2 hh–1 at 30 °C.Root and rhizome respiration rates increase from 1 µ1O2 uptake g–1 fresh weight h–1 at 0.7 ° C to10 µ10, uptake g–1 f. wt h–1 at 20 °C.Rhizome carbohydrate reserves decline from a September peakof 33 per cent alcohol insoluble d. wt to 16 per cent in May. K+ movements during the shutting and subsequent opening of trap-lobesin Aldrovanda vesiculosa were measured using 86Rb as a tracerfor K+. Immediately after the shutting, a large amount of 86Rbpre-loaded in the trap-lobes was detected in the hollow spaceinside the shut trap. This may indicate that much of the K+in the active motor cells leaks out during the shutting, resultingin turgor loss in the cells. 86Rb(K+) uptake in the trap wasactive. During the opening process, enhanced 86Rb uptake wasobserved. The time course of this uptake was similar to thatof the opening of the trap-lobes, and both courses were acceleratedby IAA. Enhanced K+ uptake may restore the turgor in activemotor cells. The quantity of K+ that moved during the shuttingor opening was estimated as 20% of that in the active motorcells in the open state of the trap-lobes. The K+ efflux acrossthe membranes of the active motor cells may be caused by a largeincrease in bulk flow triggered by an action potential, andwas estimated as 6,200 pmol.cm–2. 1 This paper is dedicated to the memory of Professor Joji Ashidawho established the physiology of rapid movement in Aldrovandavesiculosa. Mode of photosynthesis in Mesembryanthemum crystallinum changesfrom C3 to Crassulacean acid metabolism (CAM) when the plantswere stressed with high salinity. [14C]Pyruvate uptake for 30s into intact chloroplasts isolated from leaves of the CAM modeof M. crystallinum was enhanced more than 5-fold in the lightcompared with that in the dark. The stromal concentration ofpyruvate in the light reached to more than 2.5 times of themedium. In contrast, little or no pyruvate uptake occurred inchloroplasts from C3 leaves in either light or dark condition.The initial uptake rate (10 s incubation at 4°C) into theCAM chloroplasts in the light was about 3-fold higher than therate in the dark. Km and Vmax of the initial uptake in the lightwere 0.54 mM and 8.5 µmol (mg Chl)–1 h–1 respectively.These suggest that pyruvate was actively incorporated into theCAM chloroplasts against its concentration gradient across theenvelope in the light. When hydroponically grown M. crystallinumwere stressed by 350 mM NaCl, the capacity of chloroplasts forpyruvate uptake was induced in 6 d corresponding to the inductionof the activities of PEP-carboxylase and NAD(P)+-malic enzymesin response to salt stress. Plants of six contrasting genotypes of barley were raised fromvernalized (imbibed at 1 °C for 30 d) or non-vernalizedseeds and grown in 12 different controlled environments comprisingfactorial combinations of three photoperiods (10, 13 and 16h d–1), two day temperatures (18 and 28 °C) and twonight temperatures (5 and 13 °C). Except at longer daysfor Athenais or Arabi Abiad, the 28 °C day temperature wasgenerally supra-optimal and delayed awn emergence. At lowertemperatures and in photoperiods shorter than the critical value,PC, which delay awn emergence, the time from sowing to awn emergencefor five of the genotypes conformed to the equation 1/f=a +bT{macron}+cPwhere f is the time to awn emergence (d), T{macron} is meandiurnal temperature (°C), P is photoperiod (h d–1)and a, b and c are genotype-specific constants. In Arabi Abiad,however, significant responses to temperature were not detected.The low temperature pre-treatment of the seeds reduced the subsequenttime to awn emergence in Athenais and the autumn-sown genotypesAger, Arabi Abiad and Gerbel B, especially in longer days, buteither had no effect or tended to delay awn emergence in thespring-sown types Emir and Mona. In the spring-sown types PCwas outside the range investigated (i.e. > 16 h d–1),but in Ager it was approx. 13 h d–1 and in Gerbel B justover 13 h d–1. The composition and positional distribution of fatty acids inmonogalactosyldiacylglycerol, digalactosyldiacylglycerol, phosphatidylglyceroland sulphoquinovosyldiacylglycerol from two cyanobacteria, Anacystisnidulans and Synechococcus sp. grown at 25°C have been determinedand compared with measurements of the phase separation temperaturesof the lipids. Only monogalactosyldiacylglycerol in Anacystisand sulphoquinovosyldiacylglycerol in Synechococcus showed phaseseparation temperatures above 0°C. The fatty acid composition of phospholipids in the microsomesand the vanadate-sensitive H+-ATPase activity of the roots ofone-year-old Scots pine (Pinus sylvestris L.) seedlings werestudied during flushing in spring. The seedlings in hydroponiccultures were subjected to different root temperatures (5, 12or 20°C). The shoot was maintained at 20/15° C (day/night)during the 35 d experiment. After 35 d at 5° C, root growthwas totally inhibited and shoot growth partly inhibited. In roots grown at 5° C the fatty acid composition of themicrosomal phospholipids and the degree of fatty acid unsaturation(bond index) were unchanged, while in roots grown at 12 and20° C the fatty acid composition changed and bond indexdecreased. At those root temperatures, the most obvious changewas a decline in the proportion of linolenic acid (C18:3). Inthe new white roots grown either at 12°C or 20°C theproportion of C18:2 was higher and the proportion of C18:3 lowerthan in 1-year-old roots. Independently of root temperature,H+-ATPase activity, determined on a fresh weight basis, declinedto half of the original activity during the experiment. Thedecline in H+ -ATPase activity was most rapid during the firstweek. In the old roots the decline in H+-ATPase activity followedclosely the decline in amount of membrane protein. In new rootsH+-ATPase activity was high and increased with increasing roottemperature. Nodulated white clover plants (Trifolium repens L. cv. Huia)were grown as simulated swards for 71 d in flowing nutrientsolutions with roots at 11 C and shoots at 20/15 C, day/night,under natural illumination. Root temperatures were then changedto 3, 5, 7, 11, 13, 17 or 25 C and the total N2, fixation over21 d was measured in the absence of a supply mineral N. Alltreatments were subsequently supplied with 10 mmol m–2NO2 in the flowing solutions for 14 d, and the relativeuptake of N by N2, fixation and NO3 uptake was compared.Net uptake of K+ was measured on a daily basis. Root temperature had little effect on root d. wt over the 35-dexperimental period, but shoot d. wt increased by a factor of3.5 between 3 and 25 C, with the sharpest increase occurringat 7–11 C. Shoot: root d. wt ratios increased from 25to 68 with increasing temperature at 7–25 C. N2-fixationper plant (in the absence of NO2 ) increased with roottemperature at 3–13C, but showed little change above13 C. The ratios of N2 fixation: NO2 uptake over 14d (mol N: mol N) were 0.47–0.77 at 3–7 C, 092–154at 11–17 C, and 046 at 25 C, reflecting the dominanceof NO3 uptake over N2 fixation at extremes of high andlow root temperature. The total uptake of N varied only slightlyat 11–25 –C (095–110 mmol N plant–1),the decline in N2 fixation as root temperature increased above11 C was compensated for by the increase in NO 3 uptake.The % N in shoot dry matter declined with decreasing root temperature,from 32% at 13 C to 15% at 3 C. In contrast, concentrationsof N expressed on a shoot water content basis showed a modestdecrease with increasing temperature, from 345 mol m–3at 3 C to 290 mol m–3 at 25 C. The effects of temperature, photoperiod and light integral onthe time to first flowering of pansy (ViolaxwittrockianaGams)were investigated. Plants were grown at six temperatures (meansbetween 14.8 and 26.1 °C), combined with four photoperiods(8, 11, 14 and 17 h). The rate of progress to flowering increasedlinearly with temperature (up to an optimum of 21.7 °C)and with increase in photoperiod (r2=0.91, 19 d.f.), the latterindicating that pansies are quantitative long day plants (LDPs).In a second experiment, plants were sown on five dates betweenJuly and December 1992 and grown in glasshouse compartmentsunder natural day lengths at six temperatures (means between9.4 and 26.3 °C). The optimum temperature for time to floweringdecreased linearly (from 21.3 °C) with declining light integralfrom 3.4 MJ m-2d-1(total solar radiation). Data from both experimentswere used to construct a photo-thermal model of flowering inpansy. This assumed that the rate of progress to flowering increasedas an additive linear function of light integral, temperatureand photoperiod. Independent data from plants sown on threedates, and grown at five temperatures (means between 9.8 and23.6 °C) were used to validate this model which gave a goodfit to the data (r2=0.88, 15 d.f.).
