Aims: To monitor variations in the bacterial community and fermentation products of maize silage within and between bunker silos. Methods and Results: Silage samples were collected in 2008 and 2009 from three dairy farms, wherein the farmers arranged for a contractor to produce maize silage using bunker silos. Silage was prepared using a lactic acid bacteria (LAB) inoculant consisting of Enterococcus faecium, Lactobacillus plantarum and Lactobacillus buchneri. Eight samples were collected from each bunker silo; 4 ‘outer’ and 4 ‘inner’ samples were collected from near the top and the bottom of the silo. The dry matter, lactic acid, acetic acid, ethanol, 1‐propanol and 1,2‐propanediol contents differed between bunker silos in both sampling years. Higher acetic acid, 1‐propanol and 1,2‐propanediol contents were found in the bottom than the top layers in the 2008 samples, and higher lactic acid content was found in the top than the bottom layers in the 2009 samples. The bacterial community varied more between bunker silos than within a bunker silo in the 2008 samples, whereas differences between the top and the bottom layers were seen across bunker silos in the 2009 samples. The inoculated LAB were uniformly distributed, while several nonconventional silage bacteria were also detected. Lactobacillus acetotolerans, Lactobacillus panis and Acetobacter pasteurianus were detected in both years. Stenotrophomonas maltophilia was detected in the 2008 samples, and Lactobacillus reuteri, Acinetobacter sp. and Rahnella sp. were detected in the 2009 samples. Conclusions: Although differences were seen within and between bunker silos, the bacterial community may indicate a different relationship between bunker silos and sampling locations within a bunker silo from that indicated by the fermentation products. Significance and Impact of the Study: Analysis of bacterial community can help understand how diverse non‐LAB and LAB species are involved in the ensiling process of bunker‐made maize silage.  相似文献

Despite the importance of meju as a raw material used to make Korean soy sauce (ganjang) and soybean paste (doenjang), little is known about the bacterial diversity of Korean meju. In this study, the bacterial communities in meju were examined using both culture-dependent and independent methods in order to evaluate the diversity of the bacterial population. Analyses of the 16S rRNA gene sequences of the bacterial strains isolated from meju samples showed that the dominant species were related to members of the genera Bacillus, Enterococcus, and Pediococcus. The community DNAs extracted from nine different meju samples were analyzed by barcoded pyrosequencing method targeting of the V1 to V3 hypervariable regions of the 16S rRNA gene. In total, 132,374 sequences, with an average read length of 468 bp, were assigned to several phyla, with Firmicutes (93.6%) representing the predominant phylum, followed by Proteobacteria (4.5%) and Bacteroidetes (0.8%). Other phyla accounted for less than 1% of the total bacterial sequences. Most of the Firmicutes were Bacillus and lactic acid bacteria, mainly represented by members of the genera Enterococcus, Lactococcus, and Leuconostoc, whose ratio varied among different samples. In conclusion, this study indicated that the bacterial communities in meju were very diverse and a complex microbial consortium containing various microorganisms got involved in meju fermentation than we expected before.  相似文献

Zeugodacus cucurbitae (Coquillet) is one of the most significant and widespread tephritid pest species of agricultural crops. This study reports the bacterial communities associated with Z. cucurbitae from three geographical regions in Southeast Asia (Thailand, Peninsular Malaysia, and Sarawak). The bacterial microbiota were investigated by targeted 16S rRNA gene (V3–V4 region) sequencing using the Illumina Mi-Seq platform. At 97% similarity and filtering at 0.001%, there were seven bacterial phyla and unassigned bacteria, comprising 11 classes, 23 orders, 39 families and 67 genera. The bacterial diversity and richness varied within and among the samples from the three geographical regions. Five phyla were detected for the Sarawak sample, and six each for the Thailand and Peninsular Malaysia samples. Four phyla—Actinobacteria, Bacteroidetes, Firmicutes, and Proteobacteria—were represented in all the fruit fly specimens, forming the core members of the bacterial community. Proteobacteria was the predominant phylum, followed by Bacteroidetes, Firmicutes, and Actinobacteria. Fifty-three genera were represented in the Thailand sample, 56 in the Peninsular Malaysia sample, and 55 in the Sarawak sample. Forty-two genera were present in all the three geographical regions. The predominant core members were order Enterobacteriales (Proeteobacteria), and family Enterobacteriaceae (Enterobacteriales). Klebsiella (Enterobacteriaceae) was the predominant genus and K. oxytoca the predominant species with all specimens having?>?10% relative abundance. The results indicate the presence of a great diversity as well as core members of the bacterial community associated with different populations of Z. cucurbitae.

地形气候因子与物种分布的关系影响着高寒植被群落的演替,同时对山地水源涵养林功能和结构的维持具有重要的意义。以祁连山东段水源涵养林为研究对象,基于野外植物群落物种组成及地形气候因子调查数据,运用数量分类与排序等方法,探究了高寒植物群落特征及其与地形气候因子的关系。结果表明:65个调查样方中出现181个植物种,隶属40科,124属。科的物种组成及占总物种数比例分别为菊科30个种,占16.57%;蔷薇科17个,占9.44%;禾本科13个种,占7.22%;豆科11个种,占6.11%。毛茛科10个种,占5.56%。单属种82个,占总属数的66.13%。群落层片由乔木层、灌木层和草本层组成,乔木8种,灌木25种,草本148种。乔木层优势种有青海云杉、祁连圆柏、红桦。灌木层优势种有金露梅、山生柳、匙叶小檗、高山绣线菊。草本层优势种有甘肃薹草、珠芽蓼、早熟禾、唐松草、甘青蒿。TWINSPAN将高寒植被群落划分为7个群丛类型:群丛I红桦-红花蔷薇-甘肃薹草B.albosinensis-Rosa moyesii-C.kansuensis,群丛II青海云杉-匙叶小檗-甘肃薹草P.crassifolia-B.vernae-C.kansuensis,群丛III祁连圆柏-高山绣线菊-珠芽蓼Sabina chinensis-Spiraea alpine-P.viviparum,群丛IV高山绣线菊+鬼箭锦鸡儿-珠芽蓼S.alpine+Caragana jubata-P.viviparum,群丛V沙棘+甘青蒿+鼠掌老鹳草Hippophae rhamnoides+A.tangutica+Geranium sibiricum,群丛VI沙棘+苦荬菜+苦荞麦H.rhamnoides+Ixeris polycephala+Fagopyrum tataricum,群丛VII沙棘-冰草+西北沼委陵菜H.rhamnoides-Agropyron cristatum+Comarum salesovianum。7个群丛在DCA排序图上聚集分布,反映了较好的环境梯度。CCA排序结果表明,海拔是祁连山高寒植被群落植物种分布的最重要环境因子,其次是降水、温度、坡向、坡度。  相似文献

Aims

To isolate and characterize bacteriocins produced by predominant species of lactic acid bacteria from faeces of elderly subjects.

Methods and Results

Screening over 70 000 colonies, from faecal samples collected from 266 subjects, using the indicator organisms Lactobacillus bulgaricus LMG 6901 and Listeria innocua DPC 3572, identified 55 antimicrobial‐producing bacteria. Genomic fingerprinting following ApaI digestion revealed 15 distinct strains. The antimicrobial activities associated with 13 of the 15 strains were sensitive to protease treatment. The predominant antimicrobial‐producing species were identified as Lactobacillus salivarius, Lactobacillus gasseri, Lactobacillus acidophilus, Lactobacillus crispatus and Enterococcus spp. A number of previously characterized bacteriocins, including ABP‐118 and salivaricin B (from Lact. salivarius), enterocin B (Enterococcus faecium), lactacin B (Lact. acidophilus), gassericin T and a variant of gassericin A (Lact. gasseri), were identified. Interestingly, two antimicrobial‐producing species, not generally associated with intestinally derived microorganisms were also isolated: Lactococcus lactis producing nisin Z and Streptococcus mutans producing mutacin II.

Conclusion

These data suggest that bacteriocin production by intestinal isolates against our chosen targets under the screening conditions used was not frequent (0·08%).

Significance and Impact of the Study

The results presented are important due to growing evidence indicating bacteriocin production as a potential probiotic trait by virtue of strain dominance and/or pathogen inhibition in the mammalian intestine.  相似文献

Dajiang is a traditional fermented food prepared from soybeans that is still popular in northeast China. Although recent studies have revealed that a variety of bacterial species contribute to the production of fermented soybean products, little is known about bacterial communities involved in the fermentation of dajiang made in northeast China. In this study, 14 samples of naturally fermented dajiang were analyzed by denaturing gradient gel electrophoresis (DGGE) to determine the diversity of the bacteria involved in fermentation. Our results indicate that lactic acid bacteria, including Lactobacillus plantarum, uncultured Leuconostoc mesenteroides, Leuconostoc gasicomitatum, Enterococcus faecium, and Tetragenococcus halophilus, were the predominant species. This is the first report of Enterococcus spp. and Leuconostoc spp. in the Chinese fermented soybean paste dajiang using DGGE. The presence of Bacillus spp. (including Bacillus firmus), Oceanobacillus spp., and Paenibacillus glycanilyticus in the dajiang samples may be due to their salt tolerance. Potentially pathogenic Alphaproteobacteria and Staphylococcus epidermidis strains were also detected in this study. Moreover, three uncultured bacterial clones were found in some samples and require further study. The results reveal a high level of bacterial diversity in dajiang.  相似文献

[目的]为详细了解水稻不同组织内生细菌群落多样性。[方法]对宁粳43号内生细菌的总DNA提取后,采用高通量测序技术对水稻内生细菌的16S rRNA基因进行了序列测定,分析了水稻不同组织部位内生细菌群落结构特征。[结果]叶部共获得内生细菌OTUs 610个,茎部411个,根部174个。物种分类显示,叶部内生细菌种类隶属于22门40纲103目198科399属,其中优势类群是红球菌属(Rhodococcus)和乳酸杆菌属(Lactobacillus),它们的相对丰度分别为21.00%和9.19%;茎部内生细菌种类隶属于19门31纲85目169科306属,其中优势类群是红球菌属和罗尔斯通菌属(Ralstonia),它们的相对丰度分别为19.25%和13.52%;根部内生细菌种类隶属于9门19纲44目82科140属,其中优势类群是肠杆菌属(Enterobacter)和埃希氏杆菌属(Escherichia),它们的相对丰度分别为81.13%和10.89%。根茎叶中相同的OTU有78个,放线菌门(Actinobacteria)与大多数细菌具有相关性。根系内生细菌中具有调控各种代谢网络功能的物种丰度高于茎部和叶部。[结论]不同水稻组织内生细菌具有丰富的群落多样性,其中叶部的内生细菌物种最丰富,根系参与各种代谢调控的细菌丰度最高,各个组织部位的优势菌属各不相同,变形菌门是最重要的水稻内生细菌。  相似文献

Mezcal is an alcoholic beverage obtained from the distillation of fermented juices of cooked Agave spp. plant stalks (agave must), and each region in Mexico with denomination of origin uses defined Agave species to prepare mezcal with unique organoleptic characteristics. During fermentation to produce mezcal in the state of Tamaulipas, not only alcohol-producing yeasts are involved, but also a lactic acid bacterial community that has not been characterized yet. In order to address this lack of knowledge on this traditional Mexican beverage, we performed a DGGE-16S rRNA analysis of the lactic acid bacterial diversity and metabolite accumulation during the fermentation of a typical agave must that is rustically produced in San Carlos County (Tamaulipas, Mexico). The analysis of metabolite production indicated a short but important malolactic fermentation stage not previously described for mezcal. The denaturing gradient gel electrophoresis (DGGE) analysis of the 16S rRNA genes showed a distinctive lactic acid bacterial community composed mainly of Pediococcus parvulus, Lactobacillus brevis, Lactobacillus composti, Lactobacillus parabuchneri, and Lactobacillus plantarum. Some atypical genera such as Weissella and Bacillus were also found in the residual must. Our results suggest that the lactic acid bacteria could strongly be implicated in the organoleptic attributes of this traditional Mexican distilled beverage.  相似文献

Fine roots ( ≤ 2 mm diameter) are of great value when investigating belowground interactions among different plant species and soil nutrient cycling in forest ecosystems. However, fine root separation and species identification are labor-intensive and time-consuming processes. This study aimed to evaluate the aptitude of near-infrared reflectance spectroscopy (NIRS) in predicting tree species composition in fine root mixed samples. The coniferous species Cunninghamia lanceolata and Pinus massoniana, the deciduous species Alniphyllum fortunei and Liquidambar formosana, and the evergreen broadleaved species Cyclobalanopsis glauca represent the five subtropical tree species selected for this investigation. To obtain near-infrared reflectance spectral data, 20 samples taken in the field and 70 artificially mixed samples of the five species were produced after root samples were oven-dried and ground. Calibration was performed with partial least squares regression and leave-one-out cross-validation. Root mass proportions of the mixed samples showed good predictive capacity for C. lanceolata, P. massoniana, and C. glauca with low root mean square error of prediction ( < 6.82%) and high determination coefficients (R2>0.944). Predictions for A. fortunei and L. formosana were acceptable with R2>0.819. NIRS shows potential in predicting tree species composition with suitable accuracy.  相似文献

【目的】研究传统药用植物刺山柑(Capparis spinosa L.)不同部位细菌群落结构、物种组成和多样性特征,为药用植物微生物资源的开发及微生物与宿主互作提供理论依据。【方法】本研究以刺山柑地上部植物组织(果实、茎)和地下部土壤(根际土壤、非根际土壤)为研究材料,采用高通量测序技术分析刺山柑不同部位细菌的16S rRNA基因多样性,比较其细菌群落结构和物种组成特征。【结果】刺山柑4种样本共获得的3 649个操作分类单元(operational taxonomic unit,OTU),属于34门、88纲、248目、464科和1 051属。土壤样本的细菌多样性大于植物组织,细菌群落多样性以根际土壤、非根际土壤、茎和果实的顺序逐渐降低,果实内生细菌群落多样性始终最低,显著低于根际土壤。不同部位相对丰度较高的细菌门如下:植物组织中以变形菌门为主,根际土壤中为变形菌门和放线菌门,非根际土中为厚壁菌门和放线菌门。无色杆菌属(Achromobacter)、欧文氏菌属(Erwinia)、肠球菌属(Enterococcus)、微小杆菌属(Exiguobacterium)、乳杆菌属(Lactobacillus)和克雷伯氏菌属(Klebsiella)主要存在于刺山柑植物组织中。游动球菌属(Planomicrobium)、库克菌属(Kocuria)、类芽孢杆菌属(Paenibacillus)、链霉菌属(Streptomyces)、微枝形杆菌属(Microvirga)和节杆菌属(Arthrobacter)主要分布于土壤中。β多样性分析结果表明,刺山柑植物组织和土壤的细菌群落结构具有显著差异,同类型样本的细菌群落结构相似。【结论】刺山柑土壤样本中细菌群落的多样性和丰富度均高于植物组织,刺山柑不同部位的细菌群落组成不同。本研究对刺山柑不同部位细菌群落结构进行了初步分析,鉴定了各部位细菌群落中的核心菌群,为以后挖掘刺山柑的功能研究和利用提供了准确的微生物信息。  相似文献

The genus Viola, particularly Melanium section, rich in metallophytes, is an excellent taxon for study of microevolutionary and adaptation processes. Pollen, ovule, and microstructural floral characters were investigated by LM, SEM, and CLSM in seven endemic Albanian violets, five serpentinophytes (Viola albanica, V. dukadjinica, V. albanica × V. dukadjinica, V. raunsiensis, and V. macedonica), two from chalk soil (V. aetolica and V. schariensis), and in their closest relatives (V. lutea ssp. sudetica, V. tricolor ssp. tricolor, and V. arvensis) for their taxonomic usefulness and adaptive value. Three among analyzed characters were common in all Albanian violets however not unique. Serpentinophytes, V. aetolica and V. schariensis possessed hairs deep inside the spur, developed pollen heteromorphism, both increase the chance of pollination in unpredictable conditions and had strongly developed tannin rich layer in the outer integument of the young ovules with a protective role. They also all exhibited high pollen viability (86.9 ± 10.2%), high frequency of normally developed, enlarged (fertilized) ovules in ovary (65.0 ± 24.0%), but also high frequency of degenerations in developing ovules (40.4 ± 9.8%). Several flower characters may be adaptive in the unfavorable, high altitude environment, including serpentine soils. High pollen viability and normally developed fertilized ovules are sufficient for Albanian species maintenance.  相似文献

This study identified the bacterial community composition associated with Barbour's seahorses (Hippocampus barbouri). Seahorses and samples from the surrounding environment (sediment and water) were collected from Cantiasay Island in Surigao del Norte, Philippines. Genomic DNA was isolated from these samples, and the V1–V3 region of the 16S rRNA gene was amplified and sequenced on the Illumina MiSeq platform. There were 929 803 sequence reads corresponding to 859 operational taxonomic units (OTUs), which were obtained from the six genomic libraries. The results showed that the most abundant OTUs were affiliated to the phylum Proteobacteria, particularly those belonging to the genera Shewanella, Pseudomonas, Acinetobacter and Aeromonas, which were found to be dominant in seahorse gut samples. The only exception was for the skin of male seahorses, which was dominated by members belonging to the genus Bacillus (phylum Firmicutes). These findings on bacterial community structure and composition may shed light on therapeutic claims of Barbour's seahorses and help in the conservation of this species.  相似文献

Three different genes were sequenced from isolates of five plant-pathogenic Verticillium species, Verticillium albo-atrum, Verticillium dahliae, Verticillium longisporum, Verticillium nigrescens, and Verticillium tricorpus. The sequences covered parts of the mitochondrial cytochrome b gene (cob), the mitochondrial small subunit rRNA gene (rns) and the nuclear ITS2 region. When the sequences were combined, the five species clustered in five monophyletic groups, with V. nigrescens distantly related to the other species while V. tricorpus displayed a somewhat closer relationship to the three remaining species. V. albo-atrum, V. dahliae and V. longisporum were found to be very similar to each other, with V. albo-atrum and V. longisporum displaying the closest relationship. The species affiliation of V. longisporum is discussed.  相似文献

Microbial communities within subsurface uranium (U) deposits were explored to understand the nature of community composition and their potential role in biogeochemical cycle and bioremediation. Geochemical analysis revealed that the U ores were mainly hosted on metamorphosed chlorite-biotite schists, containing varied organic carbon and elevated level of several heavy metals (U, Cu, Cr, Zn, etc.). Microbial diversity as explored by 16S rRNA gene clone library and DGGE analyses revealed predominance of Proteobacteria, Acidobacteria, Bacteroidetes along with Firmicutes and candidate division OP9 within the domain Bacteria and Euryarchaeota within the domain Archaea. Among the physiochemical parameters, level of organic carbon showed considerable impact on influencing community diversity and composition. Samples from Jaduguda with high organic carbon showed abundance of bacteria known for metabolizing different carbon compounds and affiliated to unclassified uncultured members of Chitnophagaceae (Bacteroidetes), Gp4 of Acidobacteria and unclassified β-Proteobacteria and halophilic, nitrate-reducing γ-Proteobacteria. A relatively diverse assemblage of species capable of autotrophic/heterotrophic N2 fixation, CH4 utilization, H2(0)/Fe(II)/Mn(II)/S(0) oxidation, NO3/Fe(III) reduction, U and other metal precipitation/mineralization and affiliated to families Rhizobiaceae, Bradyrhizobiaceae, Caulobacteraceae, Comamonadaceae and genera Acinetobacter, Marinobacter and Alcanivorax constituted the Bagjata samples. Distribution and interrelations among abundance of various bacterial groups detected in other U mines/radioactive waste sites were compared with our data. Overall, the study reported distinct compositions of indigenous microbial communities among the samples from two mines and provided a better insight in geomicrobiology of U deposits.  相似文献

Bemisia tabaci‐transmitted geminiviruses are one of the major threats on cassava and vegetable crops in Africa. However, to date, few studies are available on the diversity of B. tabaci and their associated endosymbionts in Africa. More than 28 species have been described in the complex of B. tabaci cryptic species; among them, 2 are invasive pests worldwide: MED and MEAM1. In order to assess the species diversity of B. tabaci in vegetable crops in Senegal, several samplings in different localities, hosts and seasons were collected and analyzed with nuclear (microsatellite) and mitochondrial (COI) markers. The bacterial endosymbiont community was also studied for each sample. Two species were detected: MED Q1 and MEAM1 B. Patterns of MED Q1 (dominance on most of the samples and sites, highest nuclear and mitochondrial diversity and broader secondary endosymbiont community: Hamiltonella, Cardinium, Wolbachia and Rickettsia), point toward a predominant resident begomovirus vector group for MED Q1 on market gardening crops. Furthermore, the lower prevalence of the second species MEAM1 B, its lower nuclear and mitochondrial diversity and a narrower secondary endosymbiont community (Hamiltonella/Rickettsia), indicate that this genetic group is exotic and results from a recent invasion in this area.  相似文献

Aims: To characterize the bacterial communities in commercial total mixed ration (TMR) silage, which is known to have a long bunk life after silo opening. Methods and Results: Samples were collected from four factories that produce TMR silage according to their own recipes. Three factories were sampled three times at 1‐month intervals during the summer to characterize the differences between factories; one factory was sampled 12 times, three samples each during the summer, autumn, winter and spring, to determine seasonal changes. Bacterial communities were determined by culture‐independent denaturing gradient gel electrophoresis. All silages contained lactic acid as the predominant acid, and the contents appeared stable regardless of factories and product seasons. Acetic acid and 1‐propanol contents were different between factories and indicated seasonal changes, with increases in warm seasons compared to cool seasons. Both differences and similarities existed among the bacterial communities from each factory and product season. Lactobacillus parabuchneri was found in the products from three of four factories. Various sourdough lactic acid bacteria (LAB) were identified in commercial TMR silage; Lactobacillus panis, Lactobacillus hammesii, Lactobacillus mindensis, Lactobacillus pontis, Lactobacillus frumenti and Lactobacillus farciminis were detected in many products. Moreover, changes owing to product season were distinctive, and Lact. pontis and Lact. frumenti became detectable in summer products. Conclusion: Sourdough LAB are involved in the ensiling of commercial TMR silage. Silage bacterial communities vary more by season than by factory. The LAB species Lact. parabuchneri was detected in the TMR silage but may not be essential to the product’s long bunk life after silo opening. Significance and Impact of the Study: Commercial TMR silage resembles sourdough with respect to bacterial communities and long shelf life. The roles of sourdough LAB in the ensiling process and aerobic stability are worth examining.  相似文献

The microbial diversity of Japanese- and Chinese-fermented soybean pastes was investigated using nested PCR–denaturing gradient gel electrophoresis (DGGE). Five Japanese-fermented soybean paste samples and three Chinese-fermented soybean paste samples were analyzed for bacteria and fungi. Extracted DNA was used as a template for PCR to amplify 16S rRNA and 18S rRNA genes. The nearly complete 16S rRNA and 18S rRNA genes were amplified using universal primers, and the resulting products were subsequently used as a template in a nested PCR to obtain suitable fragments for DGGE. Tetragenococcus halophilus and Staphylococcus gallinarum were found to dominate the bacterial microbiota in Japanese samples, whereas Bacillus sp. was detected as the predominant species in Chinese samples. DGGE analysis of fungi in soybean pastes determined the presence of Aspergillus oryzae and Zygosaccharomyces rouxii in most of the Chinese and Japanese samples. Some differences were observed in the bacterial diversity of Japanese- and Chinese-fermented soybean pastes.  相似文献

The cultivable bacterial communities associated with four South China Sea sponges—Stelletta tenuis, Halichondria rugosa, Dysidea avara, and Craniella australiensis in mixed cultures—were investigated by microbial community DNA-based DGGE fingerprinting and 16S rDNA phylogenetic analysis. Diverse bacteria such as α-, γ-, δ-Proteobacteria, Bacteroidetes, and Firmicutes were cultured, some of which were previously uncultivable bacteria, potential novel strains with less than 95% similarity to their closest relatives and sponge symbionts growing only in the medium with the addition of sponge extract. According to 16S rDNA BLAST analysis, most of the bacteria were cultured from sponge for the first time, although similar phyla of bacteria have been previously recognized. The selective pressure of sponge extract on the cultured bacterial species was suggested, although the effect of sponge extract on bacterial community in high nutrient medium is not significant. Although α- and γ-Proteobacteria appeared to form the majority of the dominant cultivable bacterial communities of the four sponges, the composition of the cultivable bacterial community in the mixed culture was different, depending on the medium and sponge species. Greater bacterial diversity was observed in media C and CS for Stelletta tenuis, in media F and FS for Halichondria rugosa and Craniella australiensis. S. tenuis was found to have the highest cultivable bacterial diversity including α-, γ-, δ-Proteobacteria, Bacteroidetes, and Firmicutes, followed by sponge Dysidea avara without δ-Proteobacteria, sponge Halichondria rugosa with only α-, γ-Proteobacteria and Bacteroidetes, and sponge C. australiensis with only α-, γ-Proteobacteria and Firmicutes. Based on this study, by the strategy of mixed cultivation integrated with microbial community DNA-based DGGE fingerprinting and phylogenetic analysis, the cultivable bacterial community of sponge could be revealed effectively.  相似文献

Culture-dependent and -independent approaches were employed to identify the bacterial community structure from olive-mill wastewater produced from three olive-fruit varieties. The 233 bacterial isolates recovered were phylogenetically related to 38 members of Firmicutes, Actinobacteria, α-Proteobacteria, β-Proteobacteria, γ-Proteobacteria, and Bacteroidetes. Employing a novel microarray-based approach (PhyloChip) a high bacterial diversity was revealed consisting of 18 different phyla with representatives from 99 different families. The bacterial diversity in olive-mill wastewater from the three olive tree varieties was dominated by α-, β-, γ-, δ-, ε-Proteobacteria, Firmicutes, Bacteroidetes, Chloroflexi, Cyanobacteria, and Actinobacteria. This in-depth analysis of the indigenous microbiota indicated a cultivar-specific bacterial profile. Interestingly, the common bacterial taxa present in all three varieties examined were restricted indicating that the bacterial communities present in the olive-mill wastewater are greatly influenced by the olive-fruit variety.  相似文献

Lactobacillus equi, Lactobacillus hayakitensis, Lactobacillus johnsonii, and Weissella confusa/cibaria were the dominant species in 12 South African horses. The Bifidobacterium-group was detected in the feces of only one of the 12 horses. Sequencing of the nested-PCR amplicon identified the Bifidobacterium-group as Parascardovia denticolens. Cell numbers of L. equi, L. hayakitensis, and W. confusa/cibaria were consistent in all samples. P. denticolens, Bifidodobacterium pseudolongum, and a phylogenetic relative of Alloscardovia omnicolens were rarely detected. L. equigenerosi, a dominant species in Japanese horses, was detected in the fecal samples of only one horse.  相似文献
