Paramecia detect and accumulate in or disperse from some chemicals. Cells do this by changing frequency of turning and speed of swimming. There are at least two mechanisms by which cells respond: one dependent on ability to turn, one dependent on speed modulation. There are also two classes of chemicals: those that require the cells' ability to turn in order to cause accumulation and dispersal (type I), and those that apparently require only speed modulation (type II). Attractants of type I cause qualitatively similar changes in behavior to repellents of type II and the converse; therefore, assays are needed to distinguish between these two classes of chemicals, despite qualitatively similar behavior of some attractants and repellents. We examined two assays of paramecium chemoresponse, T-maze assay and well test, to understand how the T-maze distinguishes between attractants of type I and repellents of type II and why the well test does not.  相似文献

Abstract.  Behavioural assays to study insect attraction to specific odours are tedious, time consuming and often require large numbers of replications. Olfactometer and flight tunnel tests can usually only be conducted with one or two odour sources at a time. Moreover, chemical information on the odour sources has to be obtained in separate analytical studies. An olfactometer was developed in which six odours can be tested simultaneously for their relative attractiveness while during the assays, part of each test odour can be trapped for further analyses. The effectiveness of this six-arm olfactometer was tested by observing the responses of the solitary endoparasitoid Cotesia marginiventris (Cresson) to host-induced odours from young maize plants. For statistical analyses, we used log-linear models were adapted to account for overdispersion and possible positional biases. Female wasps responded extremely well in tests where they were offered a single odour source, as well as in tests with multiple choices. The responses of wasps released in groups were the same as those released individually and it was found that females did not attract or repel each other, but males preferred arms in which females had been released. Dose–response tests with varying numbers of plants or host larvae on plants revealed that the wasps responded in a dose-related manner, thus showing that the system is well suited to measure relative preference. The clear choices of the insects amongst six possibilities provided substantial statistical power. Gas chromatographic analyses of sampled air revealed clean and effective odour trapping, which largely facilitates the comparison of results from behavioural assays with the actual blends of volatiles that were emitted by the various odour sources. Advantages and disadvantages compared to other methods are discussed.  相似文献

Responses of the mosquitoes Aedes aegypti, Culex quinquefasciatus, and Culex nigripalpus to volatiles and compounds associated with bovine and avian blood that were presented in collagen membranes were evaluated in olfactometer and landing assays. The presence of attractants produced by blood was supported by more attraction of all species to blood than water controls in the olfactometer. Females of Ae. aegypti and Cx. quinquefasciatus were more attracted to bovine blood than to avian blood, but there was no difference in Cx. nigripalpus responses. In landing assays, significantly more females of all species landed on casings with blood than on water controls. There was no difference in landing of Ae. aegypti on bovine or avian blood. However, significantly more females of Cx. quinquefasciatus and Cx. nigripalpus landed on avian blood compared to bovine blood. Blood presented in collagen casings was an effective method for evaluating in-flight attraction and landing in all three species. In the olfactometer, several individual compounds elicited attraction in all species, but none were as attractive as blood for all species. In landing assays, several organic acids and sulfides elicited landing, with Ae. aegypti responding to the greatest number of compounds. These assay methods are effective for evaluation of volatile compounds from blood, and although responses were obtained to several compounds, none were as effective as blood in the olfactometer and landing assays.  相似文献

It is well established that the response regulator of the chemotaxis system of Escherichia coli, CheY, can undergo acetylation at lysine residues 92 and 109 via a reaction mediated by acetyl-CoA synthetase (Acs). The outcome is activation of CheY, which results in increased clockwise rotation. Nevertheless, it has not been known whether CheY acetylation is involved in chemotaxis. To address this question, we examined the chemotactic behaviour of two mutants, one lacking the acetylating enzyme Acs, and the other having an arginine-for-lysine substitution at residue 92 of CheY - one of the acetylation sites. The Deltaacs mutant exhibited much reduced sensitivity to chemotactic stimuli (both attractants and repellents) in tethering assays and greatly reduced responses in ring-forming, plug and capillary assays. Likewise, the cheY(92KR) mutant had reduced sensitivity to repellents in tethering assays and a reduced response in capillary assays. However, its response to the addition or removal of attractants was normal. These observations suggest that Acs-mediated acetylation of CheY is involved in chemotaxis and that the acetylation site Lys-92 is only involved in the response to repellents. The observation that, in the cheY(92KR) mutant, the addition of a repellent was not chemotactically equivalent to the removal of an attractant also suggests that there are different signalling pathways for attractants and repellents in E. coli.  相似文献

Abstract:  Four non-host plant extracts from Bifora radians , Arctium lappa , Humulus lupulus and Xanthium strumarium , were tested against adult grape berry moths, Paralobesia viteana , for their potential as repellents or oviposition deterrents. Responses were compared with those elicited by the major component of the P. viteana sex pheromone. Moths of both sexes exhibited varying electroantennogram (EAG) responses from 1.7 to 2.4 mV to volatile odours from plant extracts, with the greatest response to extracts of H. lupulus (2.4 mV). A multi-arm olfactometer was used to assay the behavioural response of moths to the same treatments. Male moths responded significantly to H. lupulus extract, although their strongest response was to the sex pheromone (30.0%). Female moths showed no behavioural response to the sex pheromone, attracting only 6.7% of moths, but they were attracted to extracts of H. lupulus , X. strumarium and A. lappa , with 25.0%, 21.7% and 15.0% of the released insects responding to these treatments, respectively. Choice tests were conducted to compare oviposition by P. viteana on untreated grapes and on grapes treated with one of the extracts. Despite the attraction in the olfactometer assays, H. lupulus extract significantly reduced egg laying; only 12.5% of the total deposited eggs were laid on berries treated with this extract. The extract of B. radians provided complete protection against oviposition.  相似文献

Anopheles gambiae Giles sensu stricto (Diptera: Culicidae) is the most important vector of human malaria in sub‐Saharan Africa, affecting the lives of millions of people. Existing tools such as insecticide‐treated nets and indoor‐residual sprays are not only effective, but also have limitations as a result of the development of resistance to insecticides and behavioural adaptations in biting time. Therefore, novel mosquito‐control tools are needed. Odour‐releasing traps have a potential for both trapping and surveillance purposes. Based on the outcome of ex vivo gene expression assays and in vivo electrophysiological assays on hundreds of volatile organic compounds, 29 ‘candidate behaviourally‐disruptive organic compounds' (cBDOCs) are selected, belonging to 10 chemical categories, to be assayed in the laboratory using dual‐choice olfactometers for the ability to modify the ‘attractiveness’ (i.e. significantly more insects being caught in the associated trap) of a basic volatile blend consisting of ammonia, lactic acid and tetradecanoic acid but without additional carbon dioxide. Compounds that either reduce or increase trap catches by the basic blend in the olfactometer experiments are tested under African conditions in choice experiments in a semi‐field facility in Kenya. The release rates of cBDOCs are determined gravimetrically to allow the calculation of aerial concentrations at the trap outlet. Aerial concentrations in the sub‐p.p.m. range are reported for the first time to influence mosquito behaviour. The results of the olfactometer and semi‐field assays generally correspond. Under semi‐field conditions, three compounds, 3‐heptanol (0.025 p.p.m.), 2‐methylpropanal (0.05 p.p.m.) and 4,5‐dimethylthiazole (0.73 p.p.m.), significantly increase trap catches relative to the basic blend consisting of ammonia, lactic acid, tetradecanoic acid and carbon dioxide. 2‐Acetylthiophene, 2‐nonanone and 2‐phenylethanol decrease the number of mosquitoes caught at all concentrations tested under semi‐field conditions. These compounds hold promise as attractants and spatial repellents to be applied in mosquito control programmes.  相似文献

Vertically transmitted bacterial symbionts are common in arthropods. However, estimates of their incidence and diversity are based on studies that test for a single bacterial genus and often only include small samples of each host species. Focussing on ladybird beetles, we collected large samples from 21 species and tested them for four different bacterial symbionts. Over half the species were infected, and there were often multiple symbionts in the same population. In most cases, more females than males were infected, suggesting that the symbionts may be sex ratio distorters. Many of these infections would have been missed in previous studies as they only infect a small proportion of the population. Furthermore, 11 out of the 17 symbionts discovered by us were either in the genus Rickettsia or Spiroplasma, which are rarely sampled. Our results suggest that the true incidence and diversity of bacterial symbionts in insects may be far greater than previously thought.  相似文献

Background

Entomopathogenic nematodes (EPNs) are tiny parasitic worms that parasitize insects, in which they reproduce. Their foraging behavior has been subject to numerous studies, most of which have proposed that, at short distances, EPNs use chemicals that are emitted directly from the host as host location cues. Carbon dioxide (CO2) in particular has been implicated as an important cue. Recent evidence shows that at longer distances several EPNs take advantage of volatiles that are specifically emitted by roots in response to insect attack. Studies that have revealed these plant-mediated interactions among three trophic levels have been met with some disbelief.

Scope

This review aims to take away this skepticism by summarizing the evidence for a role of root volatiles as foraging cues for EPNs. To reinforce our argument, we conducted olfactometer assays in which we directly compared the attraction of an EPN species to CO2 and two typical inducible root volatiles.

Conclusions

The combination of the ubiquitous gas and a more specific root volatile was found to be considerably more attractive than one of the two alone. Hence, future studies on EPN foraging behavior should take into account that CO2 and plant volatiles may work in synergy as attractants for EPNs. Recent research efforts also reveal prospects of exploiting plant-produced signals to improve the biological control of insect pests in the rhizosphere.  相似文献

Responses of Halobacterium halobium cells to chemical stimuli have been shown by a capillary technique. Cells were attacted by D-glucose and several amino acids and repelled by phenol. Certain chemicals, such as acetate, benzoate, indole, and NiSO4, that are known to act as repellents of Escherichia coli cells served as attractants for Halobacterium. In the presence of ethionine, sensitivity to attractants was reduced. Arsenate prevented the attraction by glucose without lowering the cellular adenosine 5'-triphosphate level. The ability for chemo-accumulation toward glucose and histidine was interfered with by the formation of photosensory systems. Light-induced motor responses and chemosensory behavior toward glucose and histidine became detectable in the late stationary growth phase only. The behavior toward acetate and indole was not connected to photobehavior in that way: both substances acted as attractants already in the late log phase. Inhibition of bacteriorhodopsin synthesis by L-nicotine allowed chemo-accumulation toward glucose and histidine already in the late logarithmic phase.  相似文献

We studied the response of female Callosobruchus chinensis to chemical cues emitted by cowpea seeds at different stages of bruchid infestation (uninfested, egg carrying, L1-, and L4-infested). Olfactory attractiveness was determined in Y-tube olfactometer assays by testing individual seed categories against either clean air or uninfested seeds. Oviposition preferences between uninfested and infested seeds were determined in petri-dish choice-experiments. The olfactometer assays revealed that weevils discriminate between seeds containing different stages of developing bruchids on the basis of olfactory cues. While odors from uninfested and egg-carrying seeds acted as attractants, odors from L1- and L4-infested seeds failed to induce a positive response by the bruchids. When given a choice between uninfested and infested seeds in the olfactometer, weevils preferred uninfested seeds over L1- and L4-infested seeds, but failed to distinguish between uninfested and egg-carrying seeds. In the oviposition experiment as well, bruchids showed a distinct preference for uninfested seeds when offered in combination with L1- and L4-infested seeds. This experiment further showed a reduced acceptance of egg carrying seeds. Our results indicate that C. chinensis females use chemical information during both host searching and host acceptance. Volatiles from uninfested or egg carrying seeds act as attractants, while deterrence increases as development of bruchid immature stages progresses.  相似文献

Colombia is one of the world's largest producers of coffee [Coffea arabica L. (Rubiaceae)]. The coffee berry borer (CBB), Hypothenemus hampei (Ferrari) (Coleoptera: Curculionidae: Scolytinae), is the main pest of coffee. This insect is controlled through an integrated pest management program that includes cultural, biological, and chemical control strategies. Despite research seeking CBB attractants and repellents, these potential management tools have not been successfully incorporated into control programs. This work proposes the use of plant functional diversity for CBB management, for which a number of plants related to coffee and weeds were selected. CBB preference to these plants was determined by olfactometry and volatile compounds emitted by them were identified. Field trials were performed to test CBB preference under field conditions. These trials determined the olfactory preference of CBB to coffee berries accompanied by material of the plants Crotalaria micans Link (Fabaceae), Lantana camara L. (Verbenaceae), Nicotiana tabacum L. (Solanaceae), Artemisia vulgaris L., Calendula officinalis L., Stevia rebaudiana (Bertoni) Bertoni, and Emilia sonchifolia (L.) DC. (all four Asteraceae). Under laboratory conditions N. tabacum, L. camara, and C. officinalis were identified as repellents for CBB in olfactometer assays, whereas E. sonchifolia functioned as attractant. Controlled field trials corroborated CBB repellency of N. tabacum and L. camara; both release volatile sesquiterpenes. Selected candidate attractants included E. sonchifolia plants, for showing attraction in the laboratory. The potential use of these plants in agroecological management of coffee plantations is discussed.  相似文献

Positive chemotaxis by Bdellovibrio bacteriovorus strain UKi2 was measured for 139 compounds. Twenty-one compounds were attractants; sensitive attraction was elicited by acetate, propionate, thioacetate, malonate, cis-oxalacetate, D-glucose-6-phosphate, acetyl coenzyme A, ammonium ion, barium ion, manganous ion, and potassium ion. Several of the attractants for B. bacteriovorus strain UKi2 also were attractants to strains 6-5-S and 114; however, strains 109D and 109J were not attracted by the compounds tested. Of 33 compounds tested, 8 were repellents for B. bacteriovorus strain UKi2: n-caproate, alanine, isoleucine, leucine, phenylalanine, tyrosine, cobaltous chloride, and hydronium ion. None of the organic repellents for strain UKi2 elicited repulson from strains 114 or 109D. However, all three strains of Bdellovibrio show aerotaxis. Several compounds were tested for their effects on viability and predacious growth of B. bacteriovorus strain UKi2. No simple correlation was found between attraction or repulsion and benefit or harm to bdellovibrios. The data are consistent with the view that in nature, the greatest survival value of chemotaxis for bdellovibros may be in aerotaxis, attraction to certain inorganic ions and acetate, and repulsion by hydronium ion.  相似文献

Efforts have been made to develop vertebrate odor‐based attractants to lure hematophagous triatomines into traps. However, more work is needed to reach a practical, cheap, and efficient odor lure. We carried out attraction and capture tests in a dual‐choice olfactometer and a pitfall trap. Here we report that a three‐component, CO2‐free, synthetic blend of vertebrate odor (consisting of ammonia, L(+) lactic acid and hexanoic acid, and known as Sweetscent®) significantly induces 3rd‐instar Rhodnius prolixus and Triatoma infestans nymphs to fall into the test capture‐tube of the olfactometer. Blend constituents presented singly or in two‐component blends did not evoke a response and, therefore, we propose that the insects respond specifically to the three‐component blend in a synergistic way. When tested in a pitfall trap in an experimental arena, this blend induced capture in 37.5% of the lured traps, whereas 9% of the nymphs tested were captured in a single night. No insects were captured in control traps. Our work represents a proof‐of‐concept regarding capture of triatomines using host odor‐based, CO2‐free synthetic mixtures as lures for pitfall traps. CO2‐free lures are more practical for field work than natural or CO2‐containing synthetic blends.  相似文献

Social insects and insects that provision nests are well known to have complex foraging behaviour involving repeated visits to learned locations. Other insects do not forage from a central location and are generally assumed to respond to resources by simple attraction without spatial memory. This simple response to resource cues is generally taken as giving rise to patterns of resource use that correspond directly to resource distribution. By contrast, the solitary parasitoid wasp Hyposoter horticola monitors the locations of multiple potential hosts (butterfly eggs) for up to several weeks, until the hosts become susceptible to parasitism. Essentially all hosts in the landscape are found, and one-third of them are parasitized, independent of host density. Here, we show that the wasps do not relocate hosts using odour markers previously left by themselves or other foragers, nor do they find the eggs anew repeatedly. Instead, the wasps relocate host eggs by learning the position of the eggs relative to visual landmarks. The anticipatory foraging behaviour presented here is a key to the wasp's exceptionally stable population dynamics.  相似文献

Repellents are highly effective in preventing humans from being bitten by mosquitoes and other blood‐feeding insects. Here we provide an overview of the representative in vitro bioassays for the laboratory testing of these agents. Currently, potential repellents are mostly evaluated on human skin, using the arm‐in‐cage method. However, for the testing of large numbers of chemicals for mosquito repellent activity, several in vitro bioassays have been established. Based on their components, as described in the recent literature, these can be categorized: (i) the natural tendency system; (ii) the membrane plus blood system; (iii) the skin plus attractants system; and (iv) the host plus netting system. The major advantages and disadvantages of these bioassays are discussed with the aim of establishing reliable and practical methods for discovering novel mosquito repellents.  相似文献

Introduction

Our ability to document insect preference for semiochemicals is pivotal in pest control as these agents can improve monitoring and be deployed within integrated pest management programmes for more efficacious control of pest species. However, methods used to date have drawbacks that limit their utility. We present and test a new concept for determining insect motivation to move towards, or away from, semiochemicals by noting direction and speed of movement as animals work against a defined energy landscape (environmentally dependent variation in the cost of transport) requiring different powers to negotiate. We conducted trials with the pine weevils Hylobius abietis and peach-potato aphids Myzus persicae exposed to various attractants and repellents and placed so that they either moved up defined slopes against gravity or had to travel over variously rough surfaces.

Results

Linear Mixed Models demonstrated clear reductions in travel speed by insects moving along increasingly energetically taxing energy landscapes but also that responses varied according to different semiochemicals, thus highlighting the value of energy landscapes as a new concept to help measure insect motivation to access or avoid different attractants or repellents across individuals.

Conclusions

New sensitive, detailed indicators of insect motivation derived from this approach should prove important in pest control across the world.  相似文献

Abstract A Y-tube olfactometer and a still-air olfactometer were developed to determine the attractiveness of several host plants for the vine weevil ( Otiorhynchus sulcatus (F.); Coleoptera: Curculionidae). Odours of weevil-damaged yew ( Taxus baccata ) and spindle trees ( Euonymus fortunei ) are attractive to the vine weevil, but Rhododendron and strawberry ( Fragaria  ×  ananassa ) are not. Undamaged Euonymus is attractive to the weevils in springtime but not in late summer. When clean air or undamaged Euonymus is the alternative, weevils strongly prefer weevil-damaged Euonymus foliage, and this preference is retained throughout the year. Hence, plant damage plays a role in attraction of the vine weevil. In contrast to the permanent attractiveness of weevil-damaged Euonymus , mechanically damaged plants gradually lose the attractiveness that they have early in the growing season. This suggests that emission of volatiles, produced by the plants in response to weevil damage, is important for attraction of the weevils because the weevils may use these plant odours to find suitable food plants throughout the season. Apart from weevil-damage-related plant volatiles, green leaf volatiles must also play a significant role, as indicated by the fact that weevils prefer: early season, undamaged Euonymus over clean air; early season, mechanically damaged Euonymus over undamaged Euonymus ; and, throughout the season, had no preference when mechanically damaged Euonymus is tested against weevil-damaged Euonymus . Thus, monitoring traps may be developed by the use of green leaf volatiles and/or herbivore-induced volatiles, as attractants.  相似文献

Codon usage bias refers to the phenomenon where specific codons are used more often than other synonymous codons during translation of genes, the extent of which varies within and among species. Molecular evolutionary investigations suggest that codon bias is manifested as a result of balance between mutational and translational selection of such genes and that this phenomenon is widespread across species and may contribute to genome evolution in a significant manner. With the advent of whole‐genome sequencing of numerous species, both prokaryotes and eukaryotes, genome‐wide patterns of codon bias are emerging in different organisms. Various factors such as expression level, GC content, recombination rates, RNA stability, codon position, gene length and others (including environmental stress and population size) can influence codon usage bias within and among species. Moreover, there has been a continuous quest towards developing new concepts and tools to measure the extent of codon usage bias of genes. In this review, we outline the fundamental concepts of evolution of the genetic code, discuss various factors that may influence biased usage of synonymous codons and then outline different principles and methods of measurement of codon usage bias. Finally, we discuss selected studies performed using whole‐genome sequences of different insect species to show how codon bias patterns vary within and among genomes. We conclude with generalized remarks on specific emerging aspects of codon bias studies and highlight the recent explosion of genome‐sequencing efforts on arthropods (such as twelve Drosophila species, species of ants, honeybee, Nasonia and Anopheles mosquitoes as well as the recent launch of a genome‐sequencing project involving 5000 insects and other arthropods) that may help us to understand better the evolution of codon bias and its biological significance.  相似文献

【目的】结合四臂嗅觉仪与轨迹跟踪分析系统,通过分析不同试验条件下黑腹果蝇Drosophila melanogaster的嗅觉行为,建立适合于果蝇等小型昆虫的嗅觉行为实验技术体系。【方法】采用Ethovision XT软件追踪黑腹果蝇在四臂嗅觉仪内的活动轨迹,分析各区域内的累计停留时长、运动速度;同时,考察气流速度和味源设置对苹果醋引诱及蓝桉Eucalyptus globulus叶油驱避黑腹果蝇效果的影响。【结果】适合于黑腹果蝇嗅觉行为分析的气流速度为300~1 200 mL/min;300 mL/min时黑腹果蝇个体间运动速度最为均一,因此推荐最适气流速度大小为300 mL/min。味源设置不会改变黑腹果蝇对味源物的嗅觉行为反应,但对味源物的引诱指数有一定的影响,单一臂放置味源物试验效果更佳。单头测试的实验效果优于多头试验。【结论】本研究建立的基于轨迹跟踪的嗅觉行为分析体系可用于昆虫信息化合物有效成分的筛选鉴定。在合适的气流条件下,单一臂设置味源化合物逐头进行测试试验效果更佳;多头试验可用于化合物的初步筛选。  相似文献
