The germination of Amaranthus paniculatus seeds was inhibited by applying paclobutrazol, a specific inhibitor of gibberellin biosynthesis. This inhibition was markedly counteracted by gibberellin A3 (GA3), suggesting that endogenous gibberellins are required for germination in this species. The inhibitory effect of paclobutrazol was also overcome by ethephon (2-chloroethylphosphonic acid) or the precursor of ethylene biosynthesis, ACC (1-aminocyclopropane-l-carboxylic acid). Thus the physiological effect of gibberellin can be mimicked by ethylene released from ethephon or synthesised from exogenous ACC. Methyl jasmonate (JA-Me) inhibited or retarded germination of Amaranthus caudatus seeds in darkness at 24°C, Ethephon, ACC and gibberellins (GA3 or GA4+7) partially or completely reversed this inhibition depending on the concentration of JA-Me applied. Both ethephon and the gibberellins were more effective than ACC. Dormant Amaranthus retroflexus seeds do not germinate in the dark at temperatures below 35°C. Fully dormant seeds germinate only at 35–40°C whereas non-dormant ones germinate within a wider range of temperatures (15 to 40°C). Germination of non-dormant seeds requires at least 10% oxygen, but the sensitivity of seeds to oxygen deprivation increases with increasing depth of dormancy. 10–6 to 10–4 M ethephon, 10–3 M 1-aminocyclopropane 1-carboxylic acid (ACC) and 10–3 M gibberellic acid (GA3) break this dormancy. In the presence of 10–3 M GA3 dormant seeds are able to germinate in the same range of temperatures as non-dormant seeds. The stimulatory effect of GA3 is less dependent on temperature than that of ethephon, while ACC stimulates germination only at relatively high temperatures (25–30°C). With the development of pineapple [Ananas comosus (L.) Merr.] as a fresh fruit crop, it became common to force inflorescence development with ethephon [(2-chloroethyl)phosphonic acid] or ethylene throughout the year. Environmental induction (EI) of inflorescence development disrupts scheduling of fruit harvest and may cause significant losses if small plants are induced, resulting in fruits that are too small to be marketable. Our objective was to identify plant growth regulators (PGRs) that could inhibit EI. Because circumstantial evidence indicates that EI occurs in response to naturally produced ethylene or changes in plant sensitivity to it, most work was done with PGRs that inhibit ethylene biosynthesis or block ethylene action. The synthetic auxin 2-(3-chlorophenoxy)propionic acid (CPA) was included because in one study it reduced the percentage of EI. GA3, aminooxyacetic acid (AOA), aminoethoxyvinylglycine (AVG), daminozide [butanedioic acid mono-(2,2-dimethylhydrazide)], and silver thiosulfate (STS) had no effect on EL CPA, paclobutrazol [(2RS,3RS)-1-(4-chlorophenyl)methyl-4,4-dimethyl-2(1h-1,2,4-triazol-1-yl)penten-3-ol], and uniconazole [(E)-(p-chlorophenyl)-4,4-dimethyl-2-(1,2,4-triazol-1-yl)-1-penten-3-ol] delayed or inhibited EI of pot-grown pineapple plants. Uniconazole and paclobutrazol inhibited growth and ethylene production by leaf basal-white tissue, and either or both effects could account for the inhibition of EI. Production of 1-aminocyclopropane-1-carboxylic acid (ACC) was unaffected by these compounds, but the activity of ACC oxidase, which converts ACC to ethylene, was inhibited and probably accounts for the reduced ethylene production by leaf basal-white tissue. CPA stimulated ethylene production by stem apical tissue approximately fourfold relative to the control. ACC oxidase activity and the malonyl-ACC (MACC) content in stem apical tissue were also greater than in the control, indicating that CPA greatly stimulated the production of ACC and its sequestration into MACC. The mechanism by which CPA delayed or inhibited EI is not known. CPA, paclobutrazol, and uniconazole appear to have some potential for inhibiting EI of pineapple. Germination of lettuce seeds has obvious thermoinhibition, but the mechanism for thermoinhibition of seed germination is poorly understood. Here, we investigated the interactions of nitrate, abscisic acid (ABA) and gibberellin on seed germination at high temperatures to understand further the mechanism for thermoinhibition of seed germination. Our results showed that lettuce (Lactuca sativa L. ‘Jianye Xianfeng No. 1’) seeds exhibited notable thermoinhibiton of germination at ≥17°C in darkness, and at ≥23°C in light, but the thermoinhibited seeds did not exhibit secondary dormancy. Thermoinhibition of seed germination at 23 or 25°C in light was notably decreased by 5 and 10 mM nitrate, and the stimulatory effects were markedly prevented by nitric oxide (NO) scavenger 2-phenyl-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide. The sensitivity of seed germination to exogenous ABA increased with increasing temperature. Thermoinhibition of seed germination was markedly decreased by fluridone (an inhibitor of ABA biosynthesis) and GA3, and was increased by diniconazole (an inhibitor of the ABA-catabolizing enzyme ABA 8′-hydroxylase) and paclobutrazol (an inhibitor of GA biosynthetic pathway). The effect of fluridone in decreasing thermoinhibition of seed germination was obviously antagonized by paclobutrazol, and that of GA3 was notably added to by fluridone, and that of nitrate was antagonized by paclobutrazol, diniconazole and ABA and was added to by GA3 and fluridone. Our data show that thermoinhibition of lettuce seed germination is decreased by nitrate in a NO-dependent manner, which is antagonized by ABA, diniconazole and paclobutrazol and added by fluridone.  相似文献

Freshly harvested, dormant seeds of Amaranthus retroflexus were unable to germinate at 25 and 35 °C. To release their dormancy at the above temperatures, the seeds were stratified at a constant temperature (4 °C) under laboratory conditions or at fluctuating temperatures in soil or by outdoor burial in soil. Fully dormant, or seeds stratified or buried (2006/2007 and 2007/2008) for various periods were treated with exogenous gibberellic acid (GA3), ethephon and abscisic acid (ABA). Likewise, the effects of these regulators, applied during stratification, on seed germination were determined. The results indicate that A. retroflexus seed dormancy can be released either by stratification or by autumn–winter burial. The effect of GA3 and ethylene, liberated from ethephon, applied after various periods of stratification or during stratification, depends on dormancy level. GA3 did not affect or only slightly stimulated the germination of non-stratified, fully dormant seeds at 25 and 35 °C respectively. Ethylene increased germination at both temperatures. Seed response to GA3 and ethylene at 25 °C was increased when dormancy was partially removed by stratification at constant or fluctuating temperatures or autumn–winter burial. The response to GA3 and ethylene increased with increasing time of stratification. The presence of GA3 and ethephon during stratification may stimulate germination at 35 °C. Thus, both GA3 and ethylene can partially substitute the requirement for stratification or autumn–winter burial. Both hormones may also stimulate germination of secondary dormant seeds, exhumed in September. The response to ABA decreased in parallel with an increasing time of stratification and burial up to May 2007 or March 2008. Endogenous GAn, ethylene and ABA may be involved in the control of dormancy state and germination of A. retroflexus. It is possible that releasing dormancy by stratification or partial burial is associated with changes in ABA/GA and ethylene balance and/or sensitivity to these hormones.  相似文献

The influence of after-ripening, sodium nitroprusside, potassium ferricyanide, cyanide, paclobutrazol and nitrite on germination of seeds of Nicotiana benthamiana was investigated as well as the influence of plant hormones such as gibberellins and abscisic acid. Dormancy of N. benthamiana seeds was broken by all treatments except treatments with abscisic acid, paclobutrazol and gibberellic acid (GA3). Gibberellins had an interesting effect on dormancy breakage of studied seeds which was dependent on use of particular gibberellin: GA3 or GA4+7. Unlike GA3, GA4+7 had broken seed dormancy.  相似文献

Tissue-culture-propagated own-rooted cv. Spartan apple trees (Malus domestica Borkh.) planted in 1979 were treated in 1983 and 1985 via a soil-line trunk drench with the plant growth retardant paclobutrazol [(2RS, 3RS)-1-(4-chlorophenyl)-4.4-dimethyl-2-(1,2, 4-triazol-1-yl) pentan-3-ol]. Seeds of immature fruits from untreated and treated trees were sampled in 1989 ca 75 days after full bloom. After seeds were freeze-dried, gibberellins (GAs) were extracted, purified and fractionated via C18 reversed-phase high-performance liquid chromatography (HPLC). Gibberellins A1, A3, A4, A7, A8, A9, A15, A17, A19, A20, A24, A34, A35, A44, A51, A53, A54, A61, A62, A63 and A68 were identified by using C18 HPLC, gas chromatography-selected ion monitoring and Kovats retention indices. Eight of the GAs identified were also quantified by using deuterated internal standards. The paclobutrazol applications caused a 55% reduction of vegetative shoot elongation in 1989, but both treated and untreated trees had developed a biennial bearing pattern by that time (heavy bloom or “on year’in 1989). Levels of early 13-hydroxylation pathway GAs, viz. GA53, GA19, GA20, GA1 and also GA3, were not altered by treatment. However, GA4, GA7 and GA9 were increased 13.4, 6.5 and 3.8 times, respectively, in seeds of fruit from treated compared to untreated trees.  相似文献

Ethephon (Eth), gibberellin A3, A4 + 7 (GA3, GA4 + 7), and 6-benzyladenine (BA) removed secondary dormancy of Amaranthus caudatus seeds. The GAs and BA potentiated the effect of ethephon or 1-aminocyclopropane-1-carboxylic acid (ACC), an ethylene biosynthesis precursor, in terms of the rate or final percent of germination. Aminoethoxyvinylglycine (AVG), an ACC synthase activity inhibitor, was observed to simultaneously inhibit the release from dormancy effected by GA3 or BA as well as the ethylene production stimulated by these regulators. Breaking of secondary dormancy by GA3, GA4 + 7 or BA was prevented by 2,5-norbornadiene (NBD), an inhibitor of ethylene binding. Ethylene completely or markedly reversed the inhibitory effect of NBD. We thus conclude that the removal of secondary dormancy in Amaranthus caudatus seeds by gibberellin or benzyladenine involves ethylene biosynthesis and action.  相似文献

The N-substituted phthalimide AC 94377 (1-(3-chlorophthalimido)-cyclohexanecarboxamide) was equally effective as a mixture of the gibberellins A4 and A7 (GA4/7) in breaking dormancy and stimulating germination of celery seeds when either was used in combination with ethephon or daminozide as a seed soak. Whereas seedlings emerging from GA4/7-treated seeds became etiolated in comparison with those from untreated seeds, those from AC 94377-treated seeds showed normal development. Preharvest sprays of gibberellic acid (GA3) increased the height of mature plants in comparison with untreated controls by about 16 per cent whereas AC 94377 was ineffective. The yield from GA3-treated plots was about 10 per cent greater than that from AC 94377-treated plots.  相似文献

Unimbibed Amaranthus caudatus seeds were found to contain stachyose, raffinose, verbascose, sucrose, galactinol, myo-inositol, glucose and fructose, while no galactose, maltose and maltotriose was detected. During imbibition, seed concentrations of verbascose, stachyose, raffinose, galactinol, myo-inositol (temporary) and fructose (transient) were observed to decrease; concentrations of galactose and maltose remained fairly constant, while those of sucrose, glucose and maltotriose increased, the increase in sucrose concentration was only temporary. Effects of gibberellin A3 (GA3) at 3 × 10−4 M and ethephon at 3 × 10−4 M alone or in the presence of methyl jasmonate (Me-JA) at 10−3 M on concentrations of soluble sugars during germination of A. caudatus seeds were examined. Me-JA was found to inhibit seed germination and fresh weight of the seeds, but did not affect sucrose, myo-inositol, galactose and maltose concentrations during imbibition for up to 20 h. The exogenously applied GA3 was observed to enhance germination, stachyose breakdown and glucose concentration after 20 h of incubation. Ethephon stimulated seed germination as well as utilisation of stachyose, galactinol (both after 14 and 20 h) and raffinose (after 14 h of incubation). Although the stimulatory effect of either GA3 or ethephon on seed germination was blocked by Me-JA; these stimulators increased mobilisation of raffinose and stachyose, but only ethephon enhanced both glucose and fructose after 14 and/or 20 h of incubation in the presence of Me-JA. The maltose concentration was increased by both GA3 and ethephon alone and in the presence of Me-JA. Of the growth regulators studied, ethephon alone and/or in combination with Me-JA significantly increased the concentrations of glucose, fructose, galactose, maltose and maltotriose. The differences in sugar metabolism appear to be linked to ethylene or GA3 applied simultaneously with Me-JA.  相似文献

Leaf chlorophyll (Chl, A, B) and total soluble protein were assayed in greenhouse-grown 1.5-year-old trees of 2 citrus types, trifoliate orange (Poncirus trifoliata (L.) Raf.) and sour orange (Citrus aurantium L.) exposed to 12 h (day/night) photoperiods in growth chambers under high (30°/21°C, day/night; noncold-hardening) and low (16°/5°C; cold-hardening) temperature regimes. Trees were sprayed 2 × per week for 5 weeks with one of the following solutions at 100 M: napthaleneacetic acid (NAA), paclobutrazol (2RS, 3RS)-1-(4-chlorophenyl-4,4-dimethyl-2-(1,2,4-triazol-1-yl)pentan-3-ol) (PPP333), benzyl-adenine (BA), abscisic acid (ABA), gibberellic acid (GA3), minerals only (N, P, K, S, Ca, Mg) and BA (+) minerals. NAA, PP333, ABA and GA3 decreased Chl A, B and soluble protein in both citrus types under cold-hardening conditions in contrast to increases with the use of BA and BA (+) minerals especially in trifoliate orange. Both BA and GA3 increased Chl A, B and protein synthesis under high temperature in both citrus types. Under noncold-hardening temperatures, GA3 enhanced Chl A, B but sharply reduced foliar protein concentration. Dieback of both cultivars following exposure to temperatures down to –6.7°C was decreased 7% by NAA sprays during noncold-hardening temperatures. Cold tolerance of noncoldhardened trifoliate orange trees was also improved with ABA and PP333. Foliar sprays of NAA (sour orange) and PP333 and BA (+) minerals (trifoliate) increased cold tolerance of cold-hardened trees by 8%. Results indicate that spray applications of growth regulators influence physiological factors associated with foliar functioning and cold tolerance in citrus during different temperature regimes.Summary Growth promoters (BA) and inhibitors (NAA) have the potential to promote cold hardines through either a strong stimulatory effect on foliar physiology or a marked inhibition of growth in general. This suggests that each growth regulator may possess an independent role in the cold-hardiness phenomenon and may also interact with physiological processes other than soluble protein and chlorophyll metabolism. Growth Regulators and the Phytochrome-Mediated Dormancy of Celery Seeds   总被引:2,自引:0,他引:2  
Seeds of five celery (Apium graveolens L.) cultivars germinated at 15°C in the light or dark but at 22°C only in the light. This light requirement was overcome by treatment with a mixture of the gibberellins GA4 and GA7 (GA4/7) but interactions of cytokinins, daminozide, ethephon, EDTA and N-phenyl-N′-4-pyridylurea (NC5392) with GA4/7 were observed. Varietal differences in response to GA4/7 concentration and the requirement for cytokinins were related to the upper temperature limits for germination of the different cultivars. Seeds of cultivars responding to low concentrations of GA4/7 appeared to contain less natural inhibitor than those requiring either high concentrations of GA4/7 or cytokinin in addition to low GA4/7. The cytokinin requirement for germination was partially removed by leaching the seeds with water. Interaction studies with applied hormones indicated that in seeds incubated in the light inhibition by abscisic acid was partially alleviated by N6-benzyladenine but not by GA4/7 application. The implications of these results are discussed in relation to the involvement of natural plant hormones in the dormancy mechanism of celery seeds.  相似文献

Seeds from heavily fruiting (on-year), mature untreated, and paclobutrazol-treated apple trees (Malus domestica Borkh. cv. Spartan) were sampled in mid-June 1987, mid-July 1987, and mid-July 1990. After seeds were freeze-dried, gibberellins (GAs) were extracted, purified, and fractionated via C18 reversed-phase high-performance liquid chromatography (HPLC). Nine GAs (GA1, GA3, GA4, GA7, GA8, GA9, GA19, GA20, and GA53) were quantified by the use of deuterated GA internal standards. Paclobutrazol trunk drench treatments reduced vegetative shoot elongation in the seasons that seeds were sampled by 55% or more. Between June 17, 1987 and July 15, 1987, the dry weight of seeds from both untreated and treated trees increased about 2.5 times and there were reductions, on a per seed basis, of GA4 in seeds from both untreated and treated trees, of GA7 in seeds from treated trees, and of GA9 in seeds from untreated trees. However, GA9 increased in seeds from treated trees. Changes in levels of some of the early-13-hydroxylation pathway GAs (GA15 GA3, GA8, GA19, GA20, and GA53) also occurred during the month. For mid-July harvested seeds, the pattern, with some exceptions, was that 2 years after paclobutrazol treatment (1987), levels of early-13-hydroxylation pathway GAs in seeds from treated trees were lower compared to controls but after 5 years (1990) their levels tended to increase. For the non-13-hydroxylated GAs (GA4, GA7, and GA9), 2 years after paclobutrazol treatment, GA4 levels were equal in seeds from untreated and treated trees, GA7 decreased in seeds from treated trees compared with controls, but GA9 levels increased. Levels of these three GAs were higher in seeds from treated trees 5 years after treatment (1990) but levels of GA4, GA7, and GA9 dramatically increased in seeds from treated trees 4 years after treatment (1989), as we previously reported.  相似文献

A reduction in concentration of gibberellins has been implicated in the phase change from juvenile to mature forms of ivy (Hedera helix L.). Attempts were made to increase the effective internal concentration of gibberellins by exogenous application of GA3, and to decrease them by various applications of abscisic acid (ABA) and paclobutrazol (PP333), alone or in combination with GA3. ABA and GA3 were fed directly into the xylem of ivy plants by a wick system (a less drastic procedure than the defoliation or decapitation used by earlier workers) whereas PP333 was applied as a soil drench.Mature ivy responded to the application of GA3 by reversion to the juvenile form, although this reversion was incomplete with respect to leaf lobing and red (anthocyanin) pigmentation and could occur spontaneously without the application of GA3. Contrary to expectation, applications of ABA and PP333 caused the stimulation of growth in juvenile ivy. No adult characteristics were induced. As similar concentrations of ABA and PP333 produced severe retardation of growth (which could be alleviated by the application of GA3) in other species, it is suggested that ivy may be an unsuitable model system for the investigation of phase change in woody plants.  相似文献

The effects of 5-bromo-2-deoxyuridine (BUdR, thymidine analogue), AMO-1618 (2-isopropyl-4-dimethylamino-5-methylphenyl-1-piperidine carboxylate methyl chloride), a growth retardant, and p-chlorophenoxyisobutyric acid (PCIB, an antiauxin) on growth (dry weight increase) and fiber development in unfertilized cotton (Gossypium hirsutum L.) ovules grown in vitro have been studied. BUdR (5 M) causes about 70% inhibition of fiber production, with little effect on ovule growth, if applied during the first 6 d of culture in the presence of GA3 and IAA. AMO-1618, when used with GA3 alone, causes only a small reduction in both dry weight and fiber production, but when used with IAA alone reduces both fiber production and dry weight, the effect on the latter being predominant. In the presence of both IAA and GA3, AMO-1618 causes a small decrease in fiber production but a major decrease in dry weight. PCIB completely inhibits fiber growth but has little effect on dry weight, especially when GA3 is present. The germination of whole seeds of celery (Apium graveolens L.) was inhibited by paclobutrazol, ancymidol and lower concentrations of uniconazole. The growth retardants daminozide, AMO 1618 and chlormequat chloride inhibited the germination of cut seeds only, indicating that the seed coat prevents the penetration of these compounds at the examined concentrations. Application of a mixture of the gibberellins A4 and A7 (GA 4/7) reversed the inhibition of all the examined growth retardants. Cytokinins (artificial or natural) had no effect when applied alone and did not interact with GA4/7 in the light. However, in the dark the cytokinins at some concentrations and GA4/7 had a synergistic effect in reversing the inhibition caused by growth retardants to whole seeds or in promoting the germination of whole seeds. Endogenous gibberellins (GAs) were extracted from safflower (Carthamus tinctorius L.) stems and detected by capillary gas chromatography-mass spectrometry from which GA1, GA3, GA19,, GA20, GA29, and probably, GA44 were detected. The detection of these GAs suggests that the early 13-OH biosynthetic pathway is prevalent in safflower shoots. Deuterated GAs were used as internal standards and GA concentrations were determined in stems harvested at weekly intervals. GA1 and GA19 levels per stem increased but concentrations per gram dry weight decreased over time. GA20 was only detected in young stem tissue.Gibberellic acid (GA3) was also applied in field trials and both GA3 and the GA biosynthetic inhibitor, paclobutrazol, were applied in growth chamber tests. GA3 increased epidermal cell size, internode length, and increased internode cell number causing stem elongation. Conversely, paclobutrazol reduced stem height, internode and cell size, cell number and overall shoot weight. In field tests, GA3 increased total stem weight, but decreased leaf weight, flower bud number and seed yield. Thus, GA3 promoted vegetative growth at the expense of reproductive commitment. These studies collectively indicate a promotory role of GAs in the control of shoot growth in safflower, and are generally consistent with gibberellin studies of related crop plants. Summary Dormancy in intact seeds of Ruellia humilis is broken by chilling or by treatment with gibberellic acid (GA3). Embryos are nondormant and will grow when the seed coat is removed completely. Embryos from chilled or GA3-treated seeds have more growth potential than embryos from nontreated seeds. Dormancy is ascribed to the mechanical restriction of the embryo by the seed coat. Chilling and treatment with GA3 break dormancy by increasing the expansive force of the embryo; thus, chilled or GA3-treated embryos exert enough expansive force to break through the seed coat, whereas nontreated embryos do not.This work was supported by an NIH Biomedical Sciences Support Grant to the University of Kentucky.  相似文献
