Responses of the photosynthetic activity of Phaeodactylum tricornutum (Bacillariophyta) to organic carbon glycerol were investigated. The growth rate, photosynthetic pigments, 77 K fluorescence spectra, and chloroplast ultrastructure of P. tricornutum were examined under photoautotrophic, mixotrophic, and photoheterotrophic conditions. The results showed that the specific growth rate was the fastest under mixotrophic conditions. The cell photosynthetic pigment content and values of Chl a/Chl c were reduced under mixotrophic and photoheterotrophic conditions. The value of carotenoid/Chl a was enhanced under mixotrophic conditions, but was decreased under photoheterotrophic conditions. In comparison with photoautotrophic conditions, the fluorescence emission peaks and fluorescence excitation peaks were not shifted. The relative fluorescence of photosystem (PS) Ⅰ and PS Ⅱ and the values of F685/F710 and F685/F738 were decreased. Chloroplast thylakoid pairs were less packed under mixotrophic and photoheterotrophic conditions. There was a strong correlation between degree of chloroplast thylakoid packing and the excitation energy kept in PS Ⅱ. These results suggested that the PS Ⅱ activity was reduced by glycerol under mixotrophic conditions, thereby leading to repression of the photosynthetic activity.  相似文献

Long-term preservation of recalcitrant seeds is very difficult because the physiological basis on their desiccation sensitivity is poorly understood. Survival of Antiaris toxicaria axes rapidly decreased and that of immature maize embryos very slowly decreased with dehydration. To understand their different responses to dehydration, we examined the changes in mitochondria activity during dehydration. Although activities of cytochrome (Cyt) c oxidase and malate dehydrogenase of the A. toxicaria axis and maize embryo mitochondria decreased with dehydration, the parameters of maize embryo mitochondria were much higher than those of A. toxicaria, showing that the damage was more severe for the A. toxicaria axis mitochondria than for those of maize embryo. The state I and III respiration of the A. toxicaria axis mitochondria were higher than those of maize embryo, the former rapidly decreased, and the latter slowly decreased with dehydration. The proportion of Cyt c pathway to state III respiration for the A. toxicaria axis mitochondria was low and rapidly decreased with dehydration, and the proportion of alternative oxidase pathway was high and slightly increased with dehydration. In contrast, the proportion of Cyt c pathway for maize embryo mitochondria was high, and that of alternative oxidase pathway was low. Both pathways decreased slowly with dehydration.  相似文献

The relation between oxidative damage and viability loss of excised embryonic axes of Antiaris toxicaria subjected to rapid drying with silica gel at 15 °C was studied.Changes of survival rate,accumulation of thiobarbituric acid-reactive substances (TBARs),activities of superoxide dismutase (SOD),catalase (CAT),ascorbate peroxidase (APX),glutathione reductase (GR) and the permeability of cell membrane that was determined as relative electrolyte leakage (REL) were measured.The half-life moisture content (MCL50) was 0.41 g H2O/g DW (dry weight basis).During drying,the activities of SOD,CAT and APX increased until MCL50,and declined thereafter.The generation speed of ·O2-,and content of H2O2 and TBARs remained steadily or even decreased at MC levels higher than MCL50,demonstrating a low oxidative level in these axes.There was no significant correlation between viability loss and accumulation of reactive oxygen species or lipid peroxidation within the dehydration process until MCL50.Whereas the increase in REL from the beginning of the drying process indicated that the cell membrane was damaged.In conclusion,under rapid drying with silica gel the viability loss of excised recalcitrant A.toxicaria axes seemed to be triggered by mechanical or physical damage,rather than metabolic damage.  相似文献

The response of Prorocentrum donghaiense and Thalassiosira weissfiogii pigments under nitrate (N) and phosphate (P) limitation were studied using HPLC and in vivo fluorescence protocols in batch cultures. For P. donghaiense, the pigment ratio was kept stable under different nutrient conditions from the results of HPLC. For T. weissflogii, there was a lower ratio of chlorophyllide to Chl a during the exponential phase, but the reverse during the stationary phase. Different members of the phytoplankton had different pigments response mechanisms under nutrient limitation. From the results of in vivo fluorescence, the ratio of peridinin to Chl a for P. donghaiense increased in nutrient-free culture, while it was kept stable for nutrient-limited cultures during the exponential phase. For T. weissflogii, the ratio of fucoxanthin to Chl a for each culture increased during the exponential phase, but the ratio under N limitation was apparently lower than that for P limitation during the stationary phase. The results indicate that both pigment ratios from HPLC and in vivo fluorescence of To weissflogii were changed greatly under different nutrient conditions, which suggests that both ratios could be used as indicators of algal physiological status in different nutrient conditions.  相似文献

We present a computerized method for the semi-automatic detection of contours in ultrasound images.The novelty of our study is the introduction of a fast and efficient image function relating to parametric active contour models.This new function is a combination of the gray-level information and first-order statistical features,called standard deviation parameters.In a comprehensive study,the developed algorithm and the efficiency of segmentation were first tested for synthetic images.Tests were also performed on breast and liver ultrasound images.The proposed method was compared with the watershed approach to show its efficiency.The performance of the segmentation was estimated using the area error rate.Using the standard deviation textural feature and a 5×5 kernel,our curve evolution was able to produce results close to the minimal area error rate(namely 8.88% for breast images and 10.82% for liver images).The image resolution was evaluated using the contrast-to-gradient method.The experiments showed promising segmentation results.  相似文献

The critically endangered tree Schizolaena tampoketsana is confined to a few diminished and degraded forest fragments on the Malagasy highlands. This habitat is vulnerable to loss due to frequent fires in the surrounding grassland that threaten to spread into the forest. One of these fragments is the focus a conservation project and here the managers aim to conserve S. tampoketsana by restoring its forest habitat to its former extent as evidenced by remnant woody plants. To inform this activity the survival and early-stage growth of seedlings of four locally native tree species were compared under contrasting conditions of proximity to the remaining forest and shade. After 12 months, seedlings of three species(Baronia taratana, Eugenia pluricymosa, Uapaca densifolia) survived better and experienced improved growth in height in grassland close to the existing forest rather than distant from it, and two survived better with shade rather than unshaded. A number of mechanisms could explain these results including reduced exposure to desiccating sunlight and winds and better soil and greater water availability close to the forest. The seedlings of one species(Nuxia capitata) survived well under all conditions. This study suggests that reforestation in these dry highlands is most feasible adjacent to remnant forest fragments and in microhabitats that minimize water loss, though young plants of some tree species may be capable of surviving in harsher conditions.  相似文献

In this investigation,we propose and analyze a virus dynamics model with multi-stages of infected cells.The model incorporates the effect of both humoral and cell-mediated immune responses.We consider two modes of transmissions,virus-to-cell and cell-to-cell.Multiple intracellular discrete-time delays have been integrated into the model.The incidence rate of infection as well as the generation and removal rates of all compartments are described by general nonlinear functions.Wc derive five threshold parameters which determine the existence of the equilibria of the model under consideration.A set of conditions on the general functions has been established which is sufficient to investigate the global stability of the five equilibria of the model.The global asymptotic stability of all equilibria is proven by utilizing Lyapunov function and LaSalle’s invariance principle.The theoretical results are illustrated by numerical simulations of the model with specific forms of the general functions.  相似文献

Four different structural models of artificial joints were developed and the finite element method (FEM) was employed to investigate their mechanical characteristics under static and dynamic conditions. The materials used in the FEM calculation were ultra-high molecular weight polyethylene (UHMWPE), 316L stainless steel, CoCrMo alloy and Ti6A14V alloy. The stress distribution, strain, and elastic deformation under static and dynamic conditions were obtained. Analysis and comparison of the ~alculation results of different models were conducted. It is shown that with the same parameters the model of a metallic femur head covered with an artificial cartilage layer is more similar to the structure of the natural human joint and its mechanical characteristics are the best of the four models.  相似文献

Glutaminase is widely distributed among microorganisms and mammals with important functions. Lit-tle is known regarding the biochemical properties and functions of the deamidating enzyme glutami-nase in cyanobacteria. In this study a putative glutaminase encoded by gene slr2079 in Synechocystis sp. PCC 6803 was investigated. The slr2079 was expressed as histidine-tagged fusion protein in Es-cherichia coli. The purified protein possessed glutaminase activity, validating the functional assign-ment of the genomic annotation. The apparent Km value of the recombinant protein for glutamine was 26.6 ± 0.9 mmol/L, which was comparable to that for some of other microbial glutaminases. Analysis of the purified protein revealed a two-fold increase in catalytic activity in the presence of 1 mol/L Na . Moreover, the Km value was decreased to 12.2 ± 1.9 mmol/L in the presence of Na . These data demon-strate that the recombinant protein Slr2079 is a glutaminase which is regulated by Na through in-creasing its affinity for substrate glutamine. The slr2079 gene was successfully disrupted in Synecho-cystis by targeted mutagenesis and the △slr2079 mutant strain was analyzed. No differences in cell growth and oxygen evolution rate were observed between △slr2079 and the wild type under standard growth conditions, demonstrating slr2079 is not essential in Synechocystis. Under high salt stress condition, however, △slr2079 cells grew 1.25-fold faster than wild-type cells. Moreover, the photosyn-thetic oxygen evolution rate of △slr2079 cells was higher than that of the wild-type. To further charac-terize this phenotype, a number of salt stress-related genes were analyzed by semi-quantitative RT-PCR. Expression of gdhB and prc was enhanced and expression of desD and guaA was repressed in △slr2079 compared to the wild type. In addition, expression of two key enzymes of ammonium assimi-lation in cyanobacteria, glutamine synthetase (GS) and glutamate synthase (GOGAT) was examined by semi-quantitative RT-PCR. Expression of GOGAT was enhanced in △slr2079 compared to the wild type while GS expression was unchanged. The results indicate that slr2079 functions in the salt stress re-sponse by regulating the expression of salt stress related genes and might not play a major role in glutamine breakdown in Synechocystis.  相似文献

The inhibitory effect of parvovirus H-1 on the colonyforming ability.in vitro of QGY-7703,a cultured human hepatoma cell line,and on the formation and growth of its tumors in nude mice was studied.With higher multiplicity of infection(MOI) of H-1 given,survival of the QGY-7703 cells was found to be decreased.H-1 DNA amplification level at 30h postinfection(p.i.) was detected to be 7.4 times higher than that at 2h by dispersed cells assay,while the cells were delayed to enter into S phase.Plaques were formed in the indicator cells(new-born human kidney cell line,NBK) by progeny H-1 virus particles released from the infected QGY-7703 cells by infectious cell center assay.The formation of tumors in nude mice by QGY-7703 cells which were injected s c at 2h postinfection was observed to by prevented in 2 proups with given MOI 25 and 50.The tumor growth of MOI 10 group occurred at a lower exponential rate than that of control,after a 20d latent period.It was evident that parvovirus H-1 exhibited a direct inhibitory effect on the formation and growth of human hepatoma cells in vivo as well as in vitro.  相似文献

Cultivars of maize (Zea mays L.) with different sensitivity to drought were exposed to 0.3 mol/L sorbitol (-1.4 MPa water potential) for 24 h. Exposure to water deficiency significantly reduced the growth of both shoots (coleoptile and hypocotyl) and roots. Shoot growth was inhibited more than the growth of roots. Osmotic stress enhanced accumulation of soluble sugars. Electrolyte leakage, a cell injury index, was slightly increased after 0.3 mol/L sorbitoh Respiration was measured in the presence and absence of 2,6-dlchloro-phenol indophenoh 2,6-Dichloro-phenol indophenol did not influence respiration rates, because statistically equal results were observed under both conditions. Total respiration (VT) decreased after osmoticum treatment. There were no significant differences in the VT among the cultlvars analysed. The decrease In VT was caused by a decline In the activities and capacities of both cytochrome (Vcyt, Vcyt) and alternative pathway (Valt, Valt) of respiration. A high residual respiration (Vres) was observed, up to 27% of total uninhibited respiration. The result of uncoupler use clearly indicated that coupling was maintained after 24 h of osmotic stress. The recovery of the respiration rate was comparable with that of non-stressed control rates. According to these observations, no possible mltochondrial damage is expected. Water deficiency did not induce a stimulation of the alternative oxidase, so we assume that the stimulation of the alternative pathway is not related to drought stress resistance; rather, the function of the alternative pathway is to balance carbon metabolism and electron transport in a response to a changing environment.  相似文献

Frequency is one of the fundamental parameters of sound.The frequency of an acoustic stimulus can be represented by a neural response such as spike rate,and/or first spike latency(FSL)of a given neuron.The spike rates/frequency function of most neurons changes with different acoustic ampli-tudes,whereas FSL/frequency function is highly stable.This implies that FSL might represent the fre-quency of a sound stimulus more efficiently than spike rate.This study involved representations of acoustic frequency by spike rate and FSL of central inferior colliculus(IC)neurons responding to free-field pure-tone stimuli.We found that the FSLs of neurons responding to characteristic frequency(CF)of sound stimulus were usually the shortest,regardless of sound intensity,and that spike rates of most neurons showed a variety of function according to sound frequency,especially at high intensities.These results strongly suggest that FSL of auditory IC neurons can represent sound frequency more precisely than spike rate.  相似文献

A greenhouse study was conducted to investigate the persistence of methoxyfenozide in pepper (Capsicum annuum L.) foliage. An aqueous suspension of methoxyfenozide was sprayed on pepper plants at concentrations of 72 and 144 mg of active ingredient (a.i.)/L. Foliage was collected at different intervals of time (0, 3, 5, 10, 20, 30, 40, and 50 days) after the treatment, and the methoxyfenozide residue was measured by high-performance liquid chromatography. The foliage was also used in bioas- says to determine the residual toxicity on and the consumption rate of the third-instar larvae of the beet armyworm, Spodoptera exigua (Htibner) (Lepidoptera: Noctuidae). The methoxyfenozide concentrations observed after 50 days had decreased to 19 and 69μg/g per sample, corresponding to a loss of 61% and 28% from the application concentrations of 72 and 144 mg a.i./L, respectively. When fitting a first-order kinetics degradation model, the half-life (DT50) of this compound was 76 days. Both application concentrations of methoxyfenozide caused a high mortality rate (〉97%) when the larvae were fed the pepper foliage collected at all of the time intervals. Lastly, at all of the time points, the consumption rate by the larvae was reduced to between 57% and 92% for both concentrations that were bioassayed. Our results indicate that, under the present greenhouse conditions, the degradation of methoxyfenozide was slower than that reported by other authors and that its residues were highly toxic to S. exigua larvae. The implications of these results for the management programs of this pest are discussed.  相似文献

The protein kinase SOS2 (Salt Overly Sensitive 2) is essential for salt-stress signaling and tolerance in Arabidopsis. SOS2 is known to be activated by calcium-SOS3 and by phosphorylation at its activation loop. SOS2 is autophosphorylated in vitro, but the autophosphorylation site and its role in salt tolerance are not known. In this study, we identified an autophosphorylation site in SOS2 and analyzed its role in the responses of Arabidopsis to salt stress. Mass spectrometry analysis showed that Ser 228 of SOS2 is autophosphorylated. When this site was mutated to Ala, the autophosphorylation rate of SOS2 decreased. The substrate phosphorylation by the mutated SOS2 was also less than that by the wild-type SOS2. In contrast, changing Ser228 to Asp to mimic the autophosphorylation enhanced substrate phosphorylation by SOS2. Complementation tests in a sos2 mutant showed that the S228A but not the $228D mutation partially disrupted the function of SOS2 in salt tolerance. We also show that activation loop phosphorylation at Thr168 and autophosphorylation at Ser228 cannot substitute for each other, suggesting that both are required for salt tolerance. Our results indicate that Ser 228 of SOS2 is autophosphorylated and that this autophosphorylation is important for SOS2 function under salt stress.  相似文献

The genomic DNA of bacteria is highly compacted in a single or a few bodies known as nucleoids. Here, we have isolated Escherichia coli nucleoid by sucrose density gradient centrifugation. The sedimentation rates, structures as well as protein/DNA composition of isolated nucleoids were then compared under various growth phases. The nucleoid structures were found to undergo changes during the cell growth; i. e., the nucleoid structure in the stationary phase was more tightly compacted than that in the exponential phase. In addition to factor for inversion stimulation(Fis), histone-like nucleoid structuring protein(H-NS), heat-unstable nucleoid protein(HU) and integration host factor(IHF) here we have identified, three new candidates of E. coli nucleoid, namely DNA-binding protein from starved cells(Dps), host factor for phage Qβ(Hfq) and suppressor of td- phenotype A(Stp A). Our results reveal that the major components of exponential phase nucleoid are Fis, HU, H-NS, Stp A and Hfq, while Dps occupies more than half of the stationary phase nucleoid. It has been known for a while that Dps is the main nucleoid-associated protein at stationary phase. From these results and the prevailing information, we propose a model for growth phase dependent changes in the structure and protein composition of nucleoid in E. coli.  相似文献

Aims Forests represent the most important component of the terrestrial biological carbon pool and play an important role in the global carbon cycle. The regional scale estimation of carbon budgets of forest ecosystems, however, have high uncertainties because of the different data sources, estimation methods and so on. Our objective was to accurately estimate the carbon storage, density and sequestration rate in forest vegetation in Jilin Province of China, in order to understand the role of the carbon sink and to better manage forest ecosystems. Methods Vegetation survey data were used to determine forest distribution, size of area and vegetation types regionally. In our study, 561 plots were investigated to build volume-biomass models; 288 plots of shrubs and herbs were harvested to calculate the biomass of understory vegetation, and samples of trees, shrubs and herbs were collected to analyze carbon content. Carbon storage, density and sequestration rate were estimated by two forest inventory data (2009 and 2014), combined with volume-biomass models, the average biomass of understory vegetation and carbon content of vegetation. Finally, the distribution patterns of carbon pools were presented using ArcGIS soft ware. Important findings Understory vegetation biomass overall was less than 3% of the tree layer biomass, varying greatly among different forest types and even among the similar types. The carbon content of trees was between 45.80% 52.97%, and that of the coniferous forests was higher than that of the broadleaf forests. The carbon content of shrub and herb layers was about 39.79% 47.25% and 40%, respectively. Therefore, the vegetation carbon conversion coefficient was 0.47 or 0.48 in Jilin Province, and the conventional use of 0.50 or 0.45 would cause deviation of ±5.26%. The vegetation carbon pool of Jilin Province was at the upper range of regional carbon pool and had higher capacity of carbon sequestration. The value in 2009 and 2014 was 471.29 Tg C and 505.76 Tg C, respectively, and the total increase was 34.47 Tg C with average annual growth of 6.89 Tg C•a1. The corresponding carbon sequestration rate was 0.92 t•hm 2•a1. The carbon density rose from 64.58 t•hm 2 in 2009 to 66.68 t•hm2 in 2014, with an average increase of 2.10 t•hm2. In addition, the carbon storage of the Quercus mongolica forests and broadleaved mixed forests, accounted for 90.34% of that of all forests. The carbon increment followed the order of young > over-mature > near mature > middle-aged > mature forests. The carbon sequestration rate of followed the order of over-mature > young > near mature > middle-aged > mature forests. Both the carbon increment and the carbon sequestration rate of mature forests were negative. Furthermore, spatially the carbon storage and density were higher in the east than in the west of Jilin province, while the carbon increment was higher in northeast and middle east than in the west. The carbon sequestration rate was higher in Tonghua and Baishan in the south, followed by Jinlin in the middle and Yanbian in the east, while Baicheng and Songyuan, etc. in west showed negative values.  相似文献

The climatic preferences of Pelias barani, a rare Pontic endemic viper, are analysed and a new locality record is reported. According to species distribution modeling, with the average test AUC was was 0.904 ± 0.068, bioclimatic variables such as Precipitation of driest month(45.5%), Temperature seasonality(18.9%), Precipitation seasonality(17.9%) and Maximum temperature of warmest period(14.7%) appear to have the most useful information on geographic distribution of Pelias barani. Distribution models of Pelias barani under current climatic conditions showed better adaptation to the northwest and northeast part of Turkey.  相似文献

Radicle emergence and reserves mobilization are two distinct programmes that are thought to control germination. Both programs are influenced by abscissic acid (ABA) but how this hormone controls seed germination is still poorly known. Phenotypic and microscopic observations of the embryo axis of Medicago truncatula during germination in mitotic inhibition condition triggered by 10 μM oryzalin showed that cell division was not required to allow radicle emergence. A suppressive subtractive hybridization showed that more than 10% of up-regulated genes in the embryo axis encoded proteins related to cell-wall biosynthesis. The expression of α-expansins, pectin-esterase, xylogucan-endotransglycosidase, cellulose synthase, and extensins was monitored in the embryo axis of seeds germinated on water, constant and transitory ABA. These genes were overexpressed before completion of germination in the control and strongly inhibited by ABA. The expression was re-established in the ABA transitory-treatment after the seeds were transferred back on water and proceeded to germination. This proves these genes as contributors to the completion of germination and strengthen the idea that cell-wall loosening and remodeling in relation to cell expansion in the embryo axis is a determinant feature in germination. Our results also showed that ABA controls germination through the control of radicle emergence, namely by inhibiting cell-wall loosening and expansion.  相似文献
