A sample of 27 males of Zoniopoda tarsata from Argentina was studied cytologically. The three largest autosomal pairs and the X were characterized by the presence of interstitial C-bands. Chiasma position relative to the bands was analyzed at diplotene and diakinesis. The frequency of interstitial, terminal and total chiasmata per cell was studied for the whole autosomal bivalent set, analysing the variations between stages and among individuals. The comparison of interstitial chiasma frequencies between stages and among individuals and the study of chiasma position relative to the bands in pairs 1, 2 and 3 indicated that chiasma distribution varied from diplotene to diakinesis. Therefore, terminalization does exist in this species and the movement may occur towards the centromere. The frequency of terminal associations at diplotene showed a high negative correlation (r=-0.89; p<10-5) with the number of interstitial chiasmata. This correlation would not be expected if the two kinds of association were produced by different (independent) mechanisms. Consequently, terminal associations were considered genuine chiasmata. The correlation between interstitial and total chiasmata was very much lower then the former (r=0.39; p=0.04). This fact, besides the relatively low variation for chiasma number, observed among individuals suggests that in this species the number of interestitial chiasmata, which are the most important in controlling the genetical recombination, is mainly regulated by changes in chiasma distribution, while variations in total chiasma frequency are of much lower magnitude.Member of Carrera del Investigador del Consejo Nacional de Investigaciones Cientificas y Técnicas (CONICET)  相似文献

Summary Ten genotypes, including inbreds, hybrids, and advanced populations, were examined in order to elucidate the relationship between position and frequency distribution of chiasmata and quantitative traits, including yield heterosis in common beans. The hybrid and advanced population groups were determined to possess 83% and 54% increased chiasma frequency, respectively in contrast to inbred lines. The increase in chiasma frequency of these populations was further manifested in a high number of interstitial chiasmata. The regular and superior chromosome behaviour of the hybrids was found to be positively associated with quantitative measures on bean yield, harvest index and bean yield efficiency. The results were discussed from the point of view that: a) increased interstitial chiasmata may provide an effective mechanism for maintaining genetic diversity and heterosis in hybrid populations; and b) heterosis for chiasma frequency and quantitative traits may be due to dispersed genes on the chromosomes having combined intra-and interallelic interactions. The data provide evidence for the existence of positive associations between interstitially localized chiasmata with its recombination potential and regular chromosome behaviour to bean yield heterosis. The role of enhanced interstitial chiasmata to promote higher levels of genetic variation and heterozygous advantage is discussed.  相似文献

Seven populations of the selfer Triticum longissimum (= Aegilops longissima) and five populations of the closely related outbreeder T. speltoides (= Ae. speltoides) were scored for chiasma frequencies in pollen mother cells. The populations of the selfer have significantly higher frequencies of chiasmata than the outbreeding populations. This difference becomes even clearer when interstitial chiasmata alone are compared. It is argued that an optimal degree of effective recombination is achieved by the balance between outbreeding and interstitial chiasmata. — There are wider differences between the selfing populations than between the outbreeding populations, but the differences between families (within populations) are small in both species. Variation between plants within families seems to be lower in the selfer, but nevertheless high enough to be inexplicable on the basis of selfing alone. — Small populations subject to hardship conditions show a higher frequency of chiasmata than others.  相似文献

Two populations ofTrimerotropis pallidipennis from Mendoza (MA) and Uspallata (UA) (Argentina), polymorphic with respect to centric shifts and B-chromosomes, were cytologically analysed. The medium-sized chromosomes from both populations were polymorphic for 7 (MA) and 5 (UA) inversions respectively. A clear tendency towards chiasma localisation was evident in both populations although they differ significantly with respect to mean frequencies of both total and interstitial chiasmata. The analysis of these frequencies in relation to the polymorphisms showed that in UA, the inversions do not affect chiasma formation while in MA, total chiasma frequency is negatively correlated in a significant fashion with the number of heteromorphic bivalents. This decrease is related, in non-B-carriers, to a significant increase of interstitial chiasmata. Therefore a redistribution of chiasmata, which could increase genetic recombination, is evident. The successful maintenance of polymorphisms in each population might depend, in part, on their effects on recombination, which could be related with the local requirements of genetic variability.  相似文献

Summary Meiosis of four telocentric-tertiary compensating trisomics of rye (Secale cereale L.) was studied with the purpose of evaluating their suitability for use in maintaining genic male-sterile lines applied in hybrid varieties. They had been constructed from four different reciprocal translocations and three different telocentrics. In one trisomic a slight, but significant tendency was demonstrated for preferential pairing of the two normal chromosomes associated with the compensating complex. This promotes the desired segregation into one normal and one compensating karyotype. In all trisomies, however, too high a frequency of failure of chiasma formation in a critical segment of the complex was evident. This is correlated with the ease of recovery of the trisomics, but results in undesired segregational products. Interstitial chiasmata leading to the formation of branched configurations were also present, more in some trisomics than in others. These also result in undesired segregations. The behaviour at meiosis was so closely correlated with the length of the chromosome segments involved that a prediction of the most favourable combination of telocentric and translocation can be made. The telocentric should be large, the corresponding translocated segment large and the interstitial segment small. The non-translocated arm of the translocated chromosome should be large and the second translocated segment small. The combinations of translocations and telocentrics had not been selected for these criteria and did not meet the requirements for practical application.  相似文献

Amblytropidia australis and Dichroplus elongatus were found to be polymorphic for supernumerary heterochromatin. In both, basic karyotypes are 2n=22+XO in males.Mitotically unstable extra chromosomes were detected in a population of A. australis. The Bs are telocentric and their number varies from O to 2 within individuals. Mean frequencies of interstitial and total chiasmata at diplotene were compared between individuals with and without Bs. The mean frequency of interstitial chiasmata increases with the number of Bs per cell.A supernumerary terminal segment in S10 pair was observed in a heterozygous condition in several individuals of D. elongatus. The localization and frequency of chiasmata at diplotene were studied. The paradigm that meiotic recombination and chiasmata have the same basis has been challenged, primarily for plants. High resolution genetic mapping frequently results in maps with lengths far exceeding those based on chiasma counts. In addition, recombination between specific homoeologous chromosomes derived from interspecific hybrids is sometimes much higher than can be explained by meiotic chiasma frequencies. However, almost the entire discrepancy disappears when proper care is taken of map inflation resulting from the shortcomings of the mapping algorithm and classification errors, the use of dissimilar material, and the difficulty of accurately counting chiasmata. Still, some exchanges, especially of short interstitial segments, cannot readily be explained by normal meiotic behaviour. Aberrant meiotic processes involving segment replacement or insertion can probably be excluded. Some cases of unusual recombination are somatic, possibly premeiotic exchange. For other cases, local relaxation of chiasma interference caused by small interruptions of homology disturbing synaptonemal complex formation is proposed as the cause. It would be accompanied by a preference for compensating exchanges (negative chromatid interference) resulting from asymmetry of the pairing chromatid pairs, so that one side of each pair preferentially participates in pairing. Over longer distances, the pairing face may switch, causing the normal random chromatid participation in double exchanges and the relatively low frequency of short interstitial exchanges. Meiotic segregation, recombination, and aneuploidy was assessed for sperm from a t(1;10)(p22.1;q22.3) reciprocal translocation carrier, by use of two multicolor FISH methods. The first method utilized three DNA probes (a telomeric and a centromeric probe on chromosome 1 plus a centromeric probe on chromosome 10) to analyze segregation patterns, in sperm, of the chromosomes involved in the translocation. The aggregate frequency of sperm products from alternate and adjacent I segregation was 90.5%, and the total frequency of normal and chromosomally balanced sperm was 48.1%. The frequencies of sperm products from adjacent II segregation and from 3:1 segregation were 4.9% and 3.9%, respectively. Reciprocal sperm products from adjacent I segregation deviated significantly from the expected 1:1 ratio (P < .0001). Our assay allowed us to evaluate recombination events in the interstitial segments at adjacent II segregation. The frequencies of sperm products resulting from interstitial recombination in chromosome 10 were significantly higher than those resulting from interstitial recombination in chromosome 1 (P < .006). No evidence of an interchromosomal effect on aneuploidy was found by use of a second FISH method that simultaneously utilized four chromosome-specific DNA probes to quantify the frequencies of aneuploid sperm for chromosomes X, Y, 18, and 21. However, a significant higher frequency of diploid sperm was detected in the translocation carrier than was detected in chromosomally normal and healthy controls. This study illustrates the advantages of multicolor FISH for assessment of the reproductive risk associated with translocation carriers and for investigation of the mechanisms of meiotic segregation of chromosomes.  相似文献

Three populations of the grasshopper Trimerotropis pallidipennis from southern Argentina have been studied cytologically. A very characteristic B-chromosome was found in all three. They also showed geographical variability in respect of the presence of pericentric inversions, and the inversion system was found to influence chiasma frequency. The Laguna Blanca population, which is on the hypothetical pathway the species is believed to have followed during its migration from northern to southern latitudes, has the same karyotype composition as the N. American form, with fixed inversions in the 3 largest autosomes and the X-chromosome. Its members have a high total chiasma frequency and a great number of interstitial chiasmata. The Sierra de la Ventana population, situated at the absolute eastern border of the species distribution is highly polymorphic with respect to the presence of inversions in the medium chromosomes. Its members have the lowest total chiasma frequency and a greatly reduced number of interstitial chiasmata. Situated geographically between the other two, the Choele-Choel population has the highest frequency of inversions and many of them are homozygous. Its members have a higher total chiasma frequency than that observed in specimens from Sierra de la Ventana, and a greatly reduced number of interstitial chiasmata, similar to that observed in individuals from the latter population.  相似文献

Summary Unequal bivalents were found in two of four populations of the short horned grasshopper, Calliptamus palaestinensis bdhr. sampled in Israel.The inequality of the homologues was due to an extra segment which was heterochromatic and apparently terminal. Pairing configurations at pachytene and position of chiasmata at later stages revealed however that the extra segment was interstitial, and the long member terminated in a minute segment homologous to the terminal part of its normal partner. The percentage of reductional divisions at anaphase I corresponded well with the percentage of terminal chiasmata (i.e. distal to the extra segment) observed at diakinesis.The assumption of an interstitial position of the extra segment would explain the previously puzzling examples of unequal bivalents in Orthoptera whose reductional divisions have not been readily accounted for otherwise.  相似文献

Summary The frequency and distribution of chiasmata was investigated in two fertile carriers of reciprocal translocations, one with a 46,XY,t(9;10)(p22;q24) karyotype and one with a 46,X,-Y,+der(Y),t(Y;10)(q12;q24) karyotype. In both cases the chromosomes involved in the translocation showed an increase in chiasma frequency in comparison to karyotypically normal controls and in both cases this increase was localised, affecting only one interstitial segment of each translocation quadrivalent. In the t(9;10) case chiasmata appeared in substantial numbers in a novel location, the proximal two thirds of 9p, while in the t(Y;10) case chiasmata appeared in a conventional location, the medial region of 10q, but at an increased frequency. Furthermore there was evidence for inter-chromosomal effects in the t(9;10) case.  相似文献

An effect of C-band pattern and polymorphism on chiasma distribution in pollen meiosis was recently demonstrated inScilla siberica. A further meiotic banding study has been performed in the alliesS. amoena, S. ingridae, andS. mischtschenkoana in order to analyze the effect, if any, of their specific C-band patterns and cytochemically different heterochromatin types on recombination. No clear evidence for a preferential formation of chiasmata adjacent to homozygous intercalary heterochromatin and no consistent reduction of chiasma frequency near strongly heterozygous intercalary heterochromatin blocks, as observed inS. siberica, could be found. Terminal C-band heteromorphism is suspected to cause distal chiasma defaults. The results suggest once more that there is no uniform effect of heterochromatin on crossover distribution.  相似文献

Substituting one of the (sub)metacentric chromosomes of a simple interchange complex by the two corresponding telocentrics marks one of the unchanged arms, one of the interchange segments and one of the interstitial segments. When the two telocentrics can be distinguished and when only infrequently MI configurations are formed for which more than a single origin is possible, rather exact estimates of the frequency of occurrence of chiasmata in the six segments can be obtained. When the two telocentrics can not be distinguished and in addition the frequency of MI configurations with more than one possible origin is not negligible, the analysis is more complex. With some previous knowledge of the interchange and some simplification an acceptable simple solution remains possible. — Of both cases an example for rye, Secale cereale, is given, together with an analysis of interference between these segments, and a conversion of probabilities of being bound at MI to genetic length.  相似文献

Symmetric and asymmetric protoplast fusion between long term cell suspension-derived protoplasts ofTriticum aestivum (cv. Jinan 177) and protoplasts ofHaynaldia villosa prepared from one-year-old embryogeneric calli was performed by PEG method. In asymmetric fusion, donor calli were treated with gamma ray at a dose of 40, 60, 80 Gy (1.3 Gy/min) respectively and then used to isolate protoplasts. Results of morphological, cytological, biochemical (isozyme) and 5S rDNA spacer sequence analysis revealed that we obtained somatic hybrid lines at high frequency from both symmetric and asymmetric fusion. Hybrid plants were recovered from symmetric and low dose γ-fusion combinations. GISH (genomicin situ hybridization) analysis proved exactly the existence of both parental chromosomes and the common occurrence of several kinds of translocation between them in the hybrid clones regenerated from symmetric and asymmetric fusion. And the elimination of donor DNA in hybrid clones regenerated from asymmetric fusion combinations was found to increase with the increasing gamma doses. It is concluded that transference and recombination of nuclear DNA can be achieved effectively by symmetric and asymmetric fusion, hybrids with small fragment translocation which are valuable in plant breeding can be obtained directly by asymmetric fusion.  相似文献

Meiosis involves reciprocal exchange of genetic information between homologous chromosomes to generate new allelic combinations. In cereals, the distribution of genetic crossovers, cytologically visible as chiasmata, is skewed toward the distal regions of the chromosomes. However, many genes are known to lie within interstitial/proximal regions of low recombination, creating a limitation for breeders. We investigated the factors underlying the pattern of chiasma formation in barley (Hordeum vulgare) and show that chiasma distribution reflects polarization in the spatiotemporal initiation of recombination, chromosome pairing, and synapsis. Consequently, meiotic progression in distal chromosomal regions occurs in coordination with the chromatin cycles that are a conserved feature of the meiotic program. Recombination initiation in interstitial and proximal regions occurs later than distal events, is not coordinated with the cycles, and rarely progresses to form chiasmata. Early recombination initiation is spatially associated with early replicating, euchromatic DNA, which is predominately found in distal regions. We demonstrate that a modest temperature shift is sufficient to alter meiotic progression in relation to the chromosome cycles. The polarization of the meiotic processes is reduced and is accompanied by a shift in chiasma distribution with an increase in interstitial and proximal chiasmata, suggesting a potential route to modify recombination in cereals.  相似文献

The seasonal variations of chiasma frequency and distribution have been studied in the lizard: Podareis sicula. In this species, as in Phyllodactylus (King & Hayman, Chromosoma 69: 131–154, 1978), chiasma frequencies vary following a definite annual cycle, and clearly different trends are shown by interstitial and terminal chiasmata.A comparison between these seasonal chiasma frequency variations and those of environmental temperature shows the existence of a clear correlation between these two parameters. However, this correlation is different in the two types of chiasmata, and may be different within the same type of chiasma depending on the period of the year.A more significant correlation is observed between chiasma cycles and annual variations of the haematic levels of sexual steroid hormones. In particular we observe a highly significant correlation between interstitial chiasma frequencies and testosterone concentration. A less precise correlation between terminal chiasma frequencies and estradiol concentration is also observed.In Podarcis, as in Phyllodactylus, the sperm that will be used for fertilization derive from the spermatocytes showing the highest rate of interstitial chiasmata. This supports the hypothesis that the cyclic variations in interstitial chiasma frequencies represent a mechanism to ensure an adequate level of variability in a given population. The above mentioned correlation between chiasma frequencies and steroia hormone concentrations suggests that the seasonal chiasma cycles are controlled by the same environmental and hormonal factors regulating the spermatogenetic cycle.  相似文献

The F3 and F4 progeny of four balanced tertiary trisomics (BTTs) of rye carrying the short translocation chromosomes of their corresponding interchanges were karyotyped and scored for the monogenic recessively inherited tigrina marker (ti) which is located terminally on the short arm of chromosome 5R. The segment carrying the dominant allele (Ti) was included in the extra translocation chromosome of each BTT, while the normal 5R pair carried the recessive allele. Recombination fractions between ti and the interchange breakpoint varied between 0.005±0.003 and 0.032±0.005 for the different BTTs. Interstitial chiasma formation could be demonstrated in only one BTT, in 1% of the first metaphase configurations without, however, resulting in recombination. Chiasma localization just proximal to the terminal C-band of 5RS was concluded to cause the low recombination observed. Less than 0.5% of the progeny consisted of plants with a deviant karyotype able to reach maturity, indicating a high level of meiotic stability of the trisomics. The results further suggest that selection towards low male and high female transmission rates of the extra chromosome is possible. The suitability of the BTTs investigated for use in hybrid breeding programs is evaluated.  相似文献

Cytological studies were carried out on two wild species (L. hirsutum and L. pennellii) and the cultivated species (L. esculentum) of tomato and their F1 hybrids. Both parents and hybrids show a diploid chromosome number of 2n=24. The meiotic behaviour of the cultivated species showed a high degree of chromosome homology resulting in a high level of chiasmata frequency per bivalent. In contrast, the two wild species showed a slight increase in uniyalent frequency and a decrease in bivalent formation and chiasmata frequency. The meiotic behaviour of the hybrids showed a high level of univalents and low levels of bivalents as well as trivalents. Highly significant decreases in chiasmata frequency and increases in meiotic abnormalities, especially in the L. esculentum X L. pennellii hybrid, also were detected. The high meiotic irregularity and low chiasmata frequency recorded in the second hybrid indicated the disharmony and difference between its parental genomes and also served to predict its sterility. With regard to degree of pairing recorded in the hybrids, there is a possibility that sterility in such cases may refer to genetic factors in addition to the previously mentioned reasons. Pollen fertility showed no great difference between L. esculentum and L. hirsutum and their F1 hybrid, but a significant decrease was recorded in the L. esculentum X L. pennellii hybrid, which was clearly associated with high meiotic irregularity, low chiasmata frequency and chromosome association.  相似文献

The metaphase I and anaphase I stages of meiosis of wheat×rye hybrids carrying the ph1b mutation were analyzed by genomic in situ hybridization. This technique allows distinction between three different types of wheat-rye associations in metaphase I configurations as well as detection of wheat-rye recombinant chromosomes in anaphase I cells. The frequency of associations between wheat and rye chromosomes greatly exceeded the level of wheat-rye recombination found in the three hybrids examined. Extremely distal associations, which account for about 50% of the total wheat-rye metaphase I chromosomal pairing, can explain such a discrepancy between metaphase I and anaphase I data. It is further discussed whether these associations reflect very distally located chiasmata or nonchiasmatic pairing. The sizes of the segments exchanged in wheat-rye recombinant chromosomes provide cytological evidence that wheat-rye recombination is restricted to the distal chromosomal regions. Colchicine induced tetraploids (4x=28) from diploidSecale cereale heterozygous for a translocation showed a strong tendency of non-preferential pairing for the interchanged chromosomes. The normal chromosomes associated in configurations up to quadrivalents, and the translocation complex formed multivalents up to octavalents. Most of the interchanged chromosome associations were characterized by their heteromorphic nature. The percentage of the chromosomes in the interchange complex forming multivalent associations was far higher than that of the remaining twenty chromosomes. Abnormalities were observed at anaphase I and II in the pollen mother cells. The tetraploids appeared to be completely sterile. It is suggested that the high frequency of multivalent formation may be explained on the basis that the interchange might have involved a region of localized chiasmata. The absence of polyploidy in the genusSecale as against its widespread occurrence in the related grass genera may be accounted for, in part, on the basis of non-preferential pairing.  相似文献
