Chlorella sp. strain 3.83, a symbiotic Chlorella isolated from the heliozoan Acanthocystis turfacea, excreted between 8% and 16% of assimilated 14CO2 as maltose in the light (15000 lx), with a pH optimum around 4.8. This percentage increased when the illuminance was lowered (36% at 1700 lx). Release of [14C]maltose continued in darkness and could be inhibited by the uncoupler carbonyl cyanide p-trifluoro-methoxyphenylhydrazone and by diethylstilbestrol. Net efflux of maltose was observed even at a concentration ratio of extracellular/intracellular maltose of 7.8. Exogenous [14C]maltose (5 mM) was taken up by the cells with a rate <2% of that of simultaneous maltose release, indicating a practically unidirectional transport. Summary Incubation of bean hook plasma membrane vesicles in the presence of L-[14C]ascorbate (ASC) resulted in a specific recovery of significant levels of the ligand with the vesicles. The strong decrease in radioactive ASC detected after hypotonic disruption of the vesicles or after an assay at 4 °C indicated that ASC was probably transported from the medium into the lumen of the membrane vesicles. The concentration kinetics of this presumptive transport process revealed a saturation curve which best fitted a biphasic model. Each phase in this model showed Michaelis-Menten type kinetics. The kinetic parameters for the different phases were calculated to be 14 and 79 M (K m1 andK m2) and 26 and 53 pmol/min · mg protein (V max1 andV max2). High concentrations of iso-ascorbate, dehydroascorbate (DHA) or non-labelled ASC significantly reduced the uptake of the radioactive vitamin. It was demonstrated that sugar or amino acid carriers are not involved in the ASC transport reaction. Generation of transmembrane cation gradients (H+, K+, Ca2+, Na+) or addition of sulfhydryl reagents (pCMBS or NEM) did not affect the ASC uptake in any way. The aim of this work was to discover how leucoplasts from suspension cultures of soybean (Glycine max L.) oxidize hexose monophosphates. Leucoplasts were isolated from protoplast lysates on a continuous gradient of Nycodenz with a yield of 28% and an intactness of 80%. Incubation of the leucoplasts with 14C-labelled substrates led to 14CO2 production, that was dependent upon leucoplast intactness, from [U-14C]glucose 6-phosphate, [U-14C]glucose 1-phosphate, [U-14C] fructose 6-phosphate and [U-14C]glucose+ATP, but not from [U-14C]fructose-1,6-bisphosphate or [U-14C]triose phosphate. The yield from [U-14C]glucose 6-phosphate was at least four times greater than that from any of the other substrates. When [1-14C]-, [2-14C]-, [3,4-14C]-, and [6-14C]glucose 6-phosphate were supplied to leucoplasts significant 14CO2 production that was dependent upon leucoplast intactness was found only for [1-14C]glucose 6-phosphate. It is argued that soybean cell leucoplasts oxidize glucose 6-phosphate via the oxidative pentose phosphate pathway with very little recycling, and that in these plastids glycolysis to acetyl CoA is negligible.S.A.C. thanks the Science and Engineering Research Council for a research studentship.  相似文献

Protoplasts isolated from beetroot tissue took up glucose preferentially whereas sucrose was transported more slowly. The 14C-label from [14C]glucose and [14C]sucrose taken up by the cells could be detected rapidly in phosphate esters and, after feeding of [14C]glucose was found also in sucrose. The temperature-dependent uptake process (activation energy EA about 50 kJ · mol–1) seems to be carrier mediated as indicated by its substrate saturation and, for glucose, by competition experiments which revealed positions C1, C5 and C6 of the D-glucose molecule as important for effective uptake. The apparent Km(20° C) for glucose (3-O-methylglucose) was about 1 mM whereas for sucrose a significantly lower apparent affinity was determined (Km about 10 mM). When higher concentrations of glucose (5 mM) or sucrose (20 mM) were administered, the uptake process followed first-order kinetics. Carrier-mediated transport was inhibited by N,N-dicyclohexylcarbodiimide, Na-orthovanadate, p–chloromercuribenzenesulfonic acid, and by uncouplers and ionophores. The uptake system exhibited a distinct pH optimum at pH 5.0. The results indicate that generation of a proton gradient is a prerequisite for sugar uptake across the plasma membrane. Protoplasts from the bundle regions in the hypocotyl take up glucose at higher rates than those derived from bundle-free regions. The characteristics of transmembrane transport of 14C-labelled indol-3yl-acetic acid ([1-14C]IAA) were compared in Chlorella vulgaris Beij., a simple unicellular green alga, and in Chara vulgaris L., a branched, multicellular green alga exhibiting axial polarity and a high degree of cell and organ specialization. In Chara thallus cells, three distinguishable trans-plasmamembrane fluxes contributed to the net uptake of [1-14C]-IAA from an external solution, viz.: a non-mediated, pH-sensitive influx of undissociated IAA (IAAH); a saturable influx of IAA; and a saturable efflux of IAA. Both saturable fluxes were competitively inhibited by unlabelled IAA. Association of [3H]IAA with microsomal preparations from Chara thallus tissue was competitively inhibited by unlabelled IAA. Results indicated that up-take carriers occurred in the membranes at a much higher density than efflux carriers. The efflux component of IAA net uptake by Chara was not affected by several phytotropins (N-1-naphthylphthalmic acid, NPA; 2-(1-pyrenoyl)benzoic acid; and 5-(2-carboxyphenyl)-3-phenylpyrazole), which are potent non-competitive inhibitors of specific auxin-efflux carriers in more advanced plant groups, and no evidence was found for a specific association of [3H]NPA with Chara microsomal preparations. It was concluded that Chara lacked phytotropin receptors. Net uptake of [1-14C]IAA also was unaffected by 2,3,5-triiodobenzoic acid except at concentrations ( 10–1 mol · m–3) high enough to depress cytoplasmic pH (determined by uptake of 5,5-dimethyloxazolidine-2,4-dione). Chlorella cells accumulated [1-14C]IAA from an external solution by pH-sensitive diffusion of IAA across the plasma membrane and anion (IAA) trapping, but no evidence was found in Chlorella for the occurrence of IAA carriers. These results indicate that carrier systems capable of mediating the transmembrane transport of auxins appeared at a very early stage in the evolution of green plants, possibly in association with the origin of a differentiated, multicellular plant body. The incubation of Chlorella cells with glucose causes the induction of an uptake system, which is specific for the basic amino acids arginine and lysine. Both amino acids are taken up in the positively charged form and with high affinity (K m values 2 M and 7 M, respectively). The transport of arginine depolarizes the membrane by 20–30 mV. The charge compensation is achieved within a few seconds after arginine addition by the proton pump, later on K+ efflux serves for charge compensation. No evidence for arginine-proton symport was found, neither by inhibitor studies nor by use of other Chlorella strains which have a slower-responding proton pump. The accumulation of arginine is appreciably higher than it should be according to the thermodynamic force of the membrane potential. The major fluxes of carbohydrate metabolism were estimated during starch breakdown by ripening bananas (Musa cavendishii Lamb ex Paxton). Hands of bananas, untreated with ethylene, were allowed to ripen in the dark at 21° C. Production of CO2 and the contents of starch, sucrose, glucose and fructose of intact fruit were determined for a period of 10 d that included the climacteric. The detailed distribution of label was determined after supplying the following to cores of pulp from climacteric fruit: [U-14C]-, [1-14C]-, [3,4-14C]-and [6-14C]glucose, [U-14C]glycerol, 14CO2. The data obtained were used to estimate the following fluxes, values given as mol hexose · (g FW)–1 · h–1 in parenthesis: starch to hexose monophosphates (5.9) and vice versa (0.4); hexose monophosphates to sucrose (7.7); sucrose to hexose (4.7); hexose to hexose monophosphate (3.8); glycolysis (0.5–1.6); triose phosphate to hexose monophosphates (0.14); oxidative pentose-phosphate pathway (0.48); CO2 fixation in the dark (0.005). Uptake of sucrose into vacuoles of suspension cells of Saccharum sp. (sugarcane) was investigated using a vacuole-isolation method based on osmotic- and pH-dependent lysis of protoplasts. Vacuoles took up sucrose at high rates without the influence of tonoplast energization on sucrose transport. Neither addition of ATP or pyrophosphate nor dissipation of the membrane potential or the pH gradient by ionophores changed uptake rates appreciably. Generation of an ATP-dependent pH gradient across the tonoplast was measured in vacuoles and tonoplast vesicles by fluorescence quenching of quinacrine. No H+ efflux could be measured by addition of sucrose to energized vacuoles or vesicles so that there was no evidence for a sucrose/H+ antiport system. Uptake rates of glucose and other sugars were similar to those of sucrose indicating a relatively non-specific sugar uptake into the vacuoles. Sucrose uptake was concentration-dependent, but no clear saturation kinetics were found. Incorporation and release of 14C-label in prenylquinones of Chlorella was investigated under steady state conditions. After one hour of 14CO2-photosynthesis all plastid quinones investigated were labeled. The highest label was found in phylloquinone (18%) while -tocopherol exhibits the lowest label (0.38%). Among the plastoquinones, plastohydroquinone-9 shows a higher labeling degree (5.1%) and a faster labeling kinetic than plastoquinone-9 (1.6%). After replacement of 14CO2 against 12CO2 the total radioactivity in plastohydroquinone-9, -tocopherol and phylloquinone decreases but in -tocoquinone and plastoquinone-9 proceeds further. From this labeling kinetic we conclude, that newly synthesized [14C]-tocopherol molecules are converted to [14C]-tocoquinone and [14C]plastohydroquinone-9 molecules to [14C]plastoquinone-9. From their 14C-incorporation kinetic half-lives could be calculated for all prenylquinones in the same ranges as previously found for the chlorophylls and carotenoids (Grumbach et al., 1978). Half-lives are shorter in plastohydroquinone-9 (30 min) and plastoquinone-9 (40 min) than in phylloquinone (55 min), -tocoquinone (50 min) and -tocopherol (220 min). This means that all prenyl-lipids such as chlorophyll a, -and -carotene, plastohydroquinone-9 and plastoquinone-9 which are more directly involved in the process of photosynthesis are subject to a continuous and higher turnover than the xanthophyll and -tocopherol. From the fast labeling kinetic and short half-lives of -tocoquinone and especially phylloquinone with a labeling degree of 12% after one hour of 14CO2 photosynthesis we suppose that perhaps these two prenylquinones are also involved in the photosynthetic activity of chloroplasts.  相似文献

Maize (Zea mays L.) and pearl millet (Pennisetum americanum (L.) Leeke) seedlings were exposed to [15N]nitrate for 1-h periods at eight times during a 24-h period (16–8 h light-dark for maize; 14–10 h for millet). Influx of [15N]nitrate as well as its reduction and translocation were determined during each period. The efflux of previously absorbed [14N]nitrate to the uptake solution was also estimated. No marked diurnal changes in [14N]nitrate efflux or [15N]nitrate influx were evident in maize. In contrast, [14N]nitrate efflux from millet increased and eventually exceeded [15N]nitrate influx during the late dark and early light periods, resulting in net nitrate efflux from the roots. The dissimilarity of their diurnal patterns indicates that influx and efflux are independently regulated. In both species, [15N]nitrate reduction and 15N translocation to shoots were curtailed more by darkness than was [15N]nitrate influx. In the light, maize reduced 15% and millet 24% of the incoming [15N]nitrate. In darkness, reduction dropped to 11 and 17%, respectively. Since the accumulation of reduced-15N in shoots declined abruptly in darkness, whereas that in roots was little affected, it is suggested that in darkness [15N]nitrate reduction occurred primarily in roots. The preincubation of Chlorella cells with glucose caused a tenfold increase of the maximal uptake rate of ammonium without change in the K m (2 M). A similar stimulation of ammonium uptake was found when the cells were transferred to nitrogen-free growth medium. The time-course of uptake stimulation by glucose revealed a lag period of 10–20 min. The turnover of the ammonium transport system is characterized by a half-life time of 5–10 h, but in the presence of light 30% of uptake activity stayed even after 50 h. 6-Deoxyglucose was not able to increase the ammonium uptake rate. These data together were interpreted as evidence for induction of an ammonium transport system by a metabolite of glucose. Mechanistic studies of the ammonium transport system provided evidence for the electrogenic uptake of the ammonium ion. The charge compensation for NH 4 + entry was achieved by immediate K+ efflux from the cells, and this was followed after 1 min by H+ extrusion. Ammonium accumulated in the cells; the rate of uptake was sensitive to p-trifluoromethoxy-carbonylcyanide-phenylhydrazon and insensitive to methionine-sulfoxime. Uptake studies with methylamine revealed that methylamine transport is obviously catalyzed by the ammonium transport system and, therefore, also increased in glucose-treated Chlorella cells.Abbreviation p.c. packed cells  相似文献

Cultures of Acetobacterium woodii and Clostridium thermoaceticum growing on fructose or glucose, respectively, were found to produce small, but significant amounts of carbon monoxide. In the gas phase of the cultures up to 53 ppm CO were determined. The carbon monoxide production was completely inhibited by 1 mM cyanide. Cultures and cell suspensions of both acetogens incorporated 14CO specifically into the carboxyl group of acetate. This CO fixation into C1 of acetate was unaffected by cyanide (1 mM). The findings are taken to indicate that CO (in a bound form) is the physiological precursor of the C1 of acetate in acetate synthesis from CO2. The cyanide inhibition experiments support the hypothesis that the cyanide-sensitive carbon monoxide dehydrogenase may serve to reduce CO2 to CO rather than to incorporate the carbonyl into C1 of acetate.  相似文献

Tissue slices from developing potato tubers (Solanum tuberosum L.) and developing cotyledons of faba bean (Vicia faba L.) were incubated with specifically labelled [13C]glucose and [13C]ribose. Enriched[13C]glucose released from starch granules was analysed by nuclear magnetic resonance (NMR). Spectral analyses were also performed on sucrose purified by high-performance liquid chromatography. In both tissues a low degree of randomisation (< 11 % in potato and < 14% in Vicia) was observed between carbon positions 1 and 6 in glucose released from starch when material was incubated with [13C]glucose labelled in positions 6 and 1, respectively. Similarly, with [2-13C]glucose a low degree of randomisation was observed in position 5. These findings indicate that extensive transport of three-carbon compounds across the amyloplast membrane does not occur in storage organs of either species. This is in agreement with previously published data which indicates that sixcarbon compounds are transported into the plastids during active starch synthesis. When [1-13C]ribose was used as a substrate, 13C-NMR spectra of starch indicated the operation of a classical pentose-phosphate pathway. However, with [2-13C]glucose there was no preferential enrichment in either carbon positions 1 or 3 relative to 4 or 6 of sucrose and starch (glucose). This provides evidence that entry of glucose in this pathway may be restricted in vivo. In both faba bean and potato the distribution of isotope between glucosyl and fructosyl moieties of sucrose approximated 50%. The effect of exogenous glucose on the major fluxes of carbohydrate metabolism in cores of climacteric fruit of banana (Musa cavendishii Lamb ex Paxton) was determined with the intention of using the effects in the application of top-down metabolic control analysis. Hands of bananas, untreated with ethylene, were allowed to ripen in the dark at 21 °C. Cores were removed from climacteric fruit and incubated in 100 or 200 mM glucose for 4 or 6 h. The rates of starch breakdown, sucrose and fructose accumulation and CO2 production were measured. The steady-state contents of hexose monophosphates, adenylates and pyruvate were determined. In addition, the detailed distribution of label was determined after supply of the following: [U-14C]-, [1-14C]-, [3,414C]and [6-14C]glucose, and [U-14C]glycerol. The data were used to estimate the major fluxes of carbohydrate metabolism. Supply of exogenous glucose led to increases in the size of the hexose-monophosphate pools. There was a small stimulation of the rate of sugar synthesis and a major increase in the rate of starch synthesis. Starch breakdown was inhibited. Respiration responded to the demand for ATP by sugar synthesis. The transport of [14C]phenylacetic acid (PAA) in intact plants and stem segments of light-grown pea (Pisum sativum L. cv. Alderman) plants was investigated and compared with the transport of [14C]indiol-3yl-acetic acid (IAA). Although PAA was readily taken up by apical tissues, unlike IAA it did not undergo long-distance transport in the stem. The absence of PAA export from the apex was shown not to be the consequence of its failure to be taken up or of its metabolism. Only a weak diffusive movement of PAA was observed in isolated stem segments which readily transported IAA. When [1-14C]PAA was applied to a mature foliage leaf in light, only 5.4% of the 14C recovered in ethanol extracts (89.6% of applied 14C) had been exported from the leaf after 6.0 h. When applied to the corresponding leaf, [14C]sucrose was readily exported (46.4% of the total recovered ethanol-soluble 14C after 6.0 h). [1-14C]phenylacetic acid applied to the root system was readily taken up but, after 5.0 h, 99.3% of the recovered 14C was still in the root system.When applied to the stem of intact plants (either in lanolin at 10 mg·g-1, or as a 10-4 M solution), unlabelled PAA blocked the transport through the stem of [1-14C]IAA applied to the apical bud, and caused IAA to accumulate in the PAA-treated region of the stem. Applications of PAA to the stem also inhibited the basipetal polar transport of [1-14C]IAA in isolated stem segments. These results are consistent with recent observations (C.F. Johnson and D.A. Morris, 1987, Planta 172, 400–407) that no carriers for PAA occur in the plasma membrane of the light-grown pea stem, but that PAA can inhibit the carrier-mediated efflux of IAA from cells. When [1-14C]indol-3yl-acetic acid ([1-14C]IAA) was applied to the upper surface of a mature foliage leaf of garden pea (Pisum sativum L. cv. Alderman), 14C effluxed basipetally but not acropetally from 30-mm-long internode segments excised 4 h after the application of [1-14C]IAA. This basipetal efflux was strongly inhibited by the inclusion of 3.10–6 mol· dm3 N-1-naphthylphthalamic acid (NPA) in the efflux buffer. In contrast, when [14C] sucrose was applied to the leaf, the efflux of label from stem segments excised subsequently was neither polar nor sensitive to NPA. The [1-14C]IAA was initially exported from mature leaves in the phloem — transport was rapid and apolar; label was recovered from aphids feeding on the stem; and label was recovered in exudates collected from severed petioles in 20 mM ethylenediaminetetraacetic acid. No 14C was detected in aphids feeding on the stems of plants to which [1-14C]IAA had been applied apically, even though the internode on which they were feeding transported considerable quantities of label. Localised applications of NPA to the stem strongly inhibited the basipetal transport of apically applied [1-14C]IAA, but did not affect transport of [1-14C]IAA in the phloem. These results demonstrate for the first time that IAA exported from leaves in the phloem can be transferred into the extravascular polar auxin transport pathway but that reciprocal transfer probably does not occur. In intact plants, transfer of foliar-applied [1-14C]IAA from the phloem to the polar auxin transport pathway was confined to immature tissues at the shoot apex. The membrane potential (Δψ) of rat liver mitochondria dropped upon addition of carbonyl cyanide p-trifluoromethoxyphenylhydrazone (FCCP) but was gradually and fully restored to the original value by the subsequent addition of dithioerythritol. Concomitantly, Ca2+ released from mitochondria was reaccumulated and the oxidative phosphorylation process completely recoupled. Neither of these effects has been observed with dinitro-o-cresol or 2,4-dinitrophenol, uncouplers which, unlike FCCP, do not react with thiols. Δψ abolished by FCCP was also restored, though incompletely, by albumin; a prompt and complete restoration was however achieved upon subsequent addition of dithioerythritol. Dithioerythritol also completely and rapidly restored the Δψ decreased by addition of diazene dicarboxylic acid bisdimethylamide (diamide).  相似文献

Protoplasts isolated from leaves of peas (Pisum sativum L.) and of Vicia faba L. produced 1-aminocyclopropane-1-carboxylic acid (ACC) from endogenous substrate. Synthesis of ACC and conversion of ACC to ethylene was promoted by light and inhibited by 3-(3,4-dichlorophenyl)-1,1-dimethylurea and carbonyl cyanide m-chlorophenylhydrazone. Aminoethoxyvinylglycine inhibited ethylene synthesis to a minor extent when given during incubation of the protoplasts but was very effective when added both to the medium in which the protoplasts were isolated and to the incubation medium as well. Radioactivity from [U-14C]methionine was incorporated into ACC and ethylene. However, the specific radioactivity of the C-2 and C-3 atoms of ACC, from which ethylene is formed, increased much faster than the specific radioactivity of ethylene. The respiratory uncouplers carbonyl cyanide p-trifluoromethoxyphenylhydrazone (FCCP) and carbonyl cyanide m-chlorophenylhydrazone (CCCP) affect the activities of two mitochondrial ion channels from mouse liver. At micromolar concentrations, the phenylhydrazones block the voltage-dependent 100-pS channel, mCS, and induce the multiple-conductance-level channel, MCC. The binding site(s) involved in perturbation of channel activities are probably distinct from the sites involved in uncoupling of oxidative phosphorylation which occurs at nanomolar concentrations of the phenylhydrazones. The effects of FCCP and CCCP on the mitochondrial ion channels could be partially reversed by washing with fresh media and were always reversed by perfusion with dithiothreitol. These results indicate that the effects of the phenylhydrazones on mitochondrial ion channels may be related to the ability of these compounds to act as sulfhydryl reagents and not to their protonophoric and uncoupling activity.  相似文献
