Yegorova and colleagues (1978) showed that a mutant strain of Drosophila melanogaster (ebony) was more sensitive to UV-induced killing of embryos and also less proficient in photoreactivating (PR) ability than a wild-type (Canton-S) strain and that the genes governing UV sensitivity and PR ability were different and presumably located on the autosomes. The experiments reported in the present paper were designed to compare the patterns of sensitivity of these 2 strains and their hybrids to X-irradiation. The sensitivity of the larvae to the killing effects of X-irradiation, and of male and female germ-cell stages to the X-ray induction of genetic damage was studied.It was found that the larvae of the ebony strain are more sensitive to X-ray-induced killing than those of the Canton-S strain. The frequencies of radiation-induced dominant lethals and sex-linked recessive lethals are higher in spermatozoa sampled from ebony males than in those of Canton-S males. In spermatozoa sampled from hybrid males, the yields of dominant lethals are no higher than in those sampled from Canton-S males and do not seem to depend on the origin of the X-chromosome. There are no statistically significant differences between the ebony and Canton-S strains in the sensitivity of their spermatozoa to the induction of autosomal translocations.Stage-7 oocytes sampled from ebony females are more sensitive to the X-ray induction of dominant lethality than are those from Canton-S females; oocytes sampled from hybrid females manifest a level of sensitivity that is significantly lower than that in either parental strain. The frequencies of X-chromosome losses induced in in this germ-cell stage are significantly lower in ebony than in Canton-S females at least at the exposure level of 3000 R at which 3 experiments were carried out. There are no measurable differences in the amount of dominant lethality induced in stage-14 oocytes of ebony, Canton-S and hybrid females.When X-irradiated Berlin-K males are mated to ebony or Canton-S females, the yields of dominant lethals are higher when ebony females are used, showing that there is a “maternal effect” for this kind of damage. Such a maternal effect is also found for sex-linked recessive lethals (irradiated Muller-5 males mated to ebony or Canton-S females). However, when irradiated ring-X-chromosome-carrying males are mated to ebony or Canton-S females, the frequencies of paternal sex-chromosome losses (scored as XO males) are lower when ebony females are used.These results have been interpreted on the assumption that the ebony strain is homozygous for recessive, autosomal genes that confer increased radiosensitivity and that the Canton-S strain carries the normal, wild-type alleles for these genes. The higher yields of dominant and recessive lethals in mature spermatozoa and of dominant lethals in stage-7 oocytes are a consequence of an enhanced sensitivity to the mutagenic (in particular, to the chromosome-breaking) effects of X-irradiation and/or of defective repair of radiation-induced genetic damage. The lower yield of XO males from irradiated stage-7 oocytes of ebony females is probably a consequence of a defect in the repair of chromosome-breakage effects, resulting in the conversion of potential X losses in females into dominant lethals. The “maternal effects” for dominant lethals, sex-linked recessive lethals and for the loss of ring-X chromosomes are assumed to have a common causal basis, namely, a defective repair of chromosome-breakage events in the females of the ebony strain.  相似文献

The repair-deficient mutant, mei-9a in Drosophila melanogaster was investigated regarding its effect on spontaneous and X-ray-induced chromosome loss in male postmeiotic cells. From matings of males carrying a mei-9a or an ordinary ring-X and a doubly marked Y chromosome (BSYy+) with mei-9a or ordinary females, the spontaneous frequencies of complete loss, partial loss, and inferred ring-X loss (based on shifts in sex ratio female:male) were significantly higher with mei-9a than with non-mei-9a. When males were given 3000 rad X-irradiation, frequencies of induced partial loss, inferred ring-X loss and the reduction in the number of progeny per female were significantly greater with mei-9a than with non-mei-9a. The results provide evidence that the mei-9a is a potentiator of both spontaneous and X-ray-induced chromosome lesions in sperm of the Drosophila male. Evidence is presented which implicates the presence of mei-9a in the P1 female and not the male as (at least) largely responsible for the characteristic mei-9a effects.  相似文献

This paper reports the results of a study on the mutagenic profile of HMPA in Drosophila melanogaster. HMPA produced all types of genetic damage tested for in post-meiotic cells of treated males; at the concentrations used, recessive lethals and ring-X losses were induced at significant rates while 2–3 translocations, entire and partial Y-chromosome losses only occurred at low rates. From a comparison with alkylation-induced mutational spectra, we note a number of peculiarities of HMPA mutagenesis:

1. (1) there is no storage effect on HMPA-induced translocations;

2. (2) the ratio of F2-lethals: F3-lethals varies from 6 : 1 to 9 : 1, indicating a low capacity of HMPA for delayed mutations;

3. (3) the use of the DNA-repair-deficient mei-9L1 females instead of an excision-proficient control strain has no influence on the recovery of mutations )recessive lethals) induced in males;

4. (4) the high frequencies of chromosome loss (CL) induced by HMPA, which are mostly due to ring-X loss, leads us to speculate that one (or more) of its metabolites acts as a DNA-crosslinking agent. In experiments on maternal effects with mei-9L1 females, there is a 20–40% reduction in the rates of induced CL. Conversely, with mei-41D5 females, there is a weak increase in CL frequencies.

5. (5) HPLC analysis of DNA reacted with [14C]HMPA exhibits no methylation at the O6 or the N-7 of guanine. This finding, together with the observed inactivity of hexaethylphosphoramide (HEPA) in the recessive lethal assay, suggests that the formation of DNA-bound forms from HMPA may not be the result of simple methylation reactions. This conclusion is supported by the genetic data, i.e., the lack of a storage effect on HMPA-induced chromosome rearrangements.

Consistent with a hypothesis by Brodberg et al. (1983) to explain the action of cisplatin in Drosophila, comparisons of the spectra of genetic alterations produced by HMPA, A 139 (bifunctional) and Thio-TEPA (trifunctional) in the assay for chromosome loss suggest the involvement of two distinct mechanisms in the formation of ring-X loss by crosslinking agents. One pathway concerns induction of chromosome loss as a consequence of sister-chromatid exchanges (SCEs). The second mechanism may be due to DNA adducts or a single adduct responsible for both a fraction of CL and for induced partial Y-loss (PL). Inactivation of the mei-9+ function has two consequences: SCE-mediated ring-X loss frequency is lowered in mei-9 females in comparison to the repair-proficient control strain, while the opposite effect is indicated for that fraction of ring-X loss generated by the second mutational pathway. Additional complicating factors include the observation of a dual role of storage in our study: the proportion of SCE-related chromosome losses decreases with increasing time of storage, but that produced by the alternative pathway increases. Thus, the CL frequencies actually recorded under the heading ‘chromosome loss’ appear as the net result of two mechanisms counteracting each other in their effects.  相似文献

The nature of the N2 effect for ICR-170, i.e., the greater mutagenic and lethal activities of this agent in the presence of N2 than O2, has been studied at the ad-3 region of Neurospora crassa. The characteristics of the N2 effect for ICR-170 were that (1) the N2 effect with ICR-170 was displayed in conidia when N2 was administered during, but not before or after, ICR-170 treatment, (2) the highly increased mutagenic and lethal activities of ICR-170 in the presence of N2 were due to an anoxic condition rather than to the presence of N2 per se, (3) the high killing activity of ICR-170 under N2 was due largely to increased cytoplasmic inactivation, (4) the N2 effect was a general phenomenon at the ad-3 region of N. crassa, and (5) the highly ICR-170-induced mutation in conidia under N2 was attributable to an actual enhancement in the mutagenic activity of ICR-170 rather than to selective killing. With regard to the mechanisms of the N2 effect with ICR-170, results indicate that this effect (1) was not due to more extracellular oxidative degradation of ICR-170 molecules in the presence of O2, or to a greater uptake of ICR-170 by conidia under N2, but (2) was due to the inhibition of conidial respiration under an anoxic environment.  相似文献

Photostimulation of N2 fixation in the blue-green alga Anabaena cylindrica was investigated using monochromatic light and by comparing action spectra of C2H2 reduction with that of photosynthetic O2 evolution. Maximum nitrogenase activity per unit energy was measured at a wavelength which corresponds to the maximum light absorption of chlorophyll a. The action spectrum of C2H2 reduction indicates a primary involvement of Photosystem I in N2 fixation by blue-green algae.  相似文献

Muller-5 males were irradiated with X-rays in nitrogen, in air or in oxygen (followed by nitrogen or oxygen post-treatments in the nitrogen and oxygen series) and were mated to females of a repair-proficient strain (mei+) or to those of a strain known to be deficient in excision repair of UV damage (in somatic cells). The latter strain, designated as mei-9a, is also known to be sensitive, in the larval stages, to the killing effects of UV, X-rays and to a number of chemical mutagens. The frequencies of sex-linked recessive lethals and autosomal translocations induced in the spermatozoa of males were determined and compared. The frequencies of sex-linked recessive lethals in the mei-9 control groups were consistently higher than in the mei+ groups. Irradiation in air or in nitrogen led to significantly higher yields of recessive lethals when the irradiated males were mated to mei-9 females, whereas, after irradiation in oxygen, the yields were similar with both kinds of female. No significant differences in the frequencies of reciprocal translocations were observed between the mei+ and mei-9 groups after irradiation of the males in nitrogen, in air or in oxygen. Likewise, no differential effects of the contrasting post-treatments (nitrogen versus oxygen), either for recessive lethals or for translocations, could be discerned. These results are considered to support the notion that the kinds of genetic damage induced in mature spermatozoa in air or in nitrogen are qualitatively similar (at least with respect to the component(s) that lead to the production of recessive lethal mutations), but clearly different when induced in an oxygen atmosphere. The enhanced yields of recessive lethals with mei-9 females (after irradiation of the males either in air or in nitrogen) has been interpreted on the assumption that the mei-9 mutant is also deficient for the repair of X-ray-induced, recessive lethal-generating premutational lesions. Possible reasons for the lack of differences between the mei+ and mei-9 groups with respect to translocation yields and for the absence of measurable differences in response between the contrasting post-treatments (after irradiation of the males in nitrogen) are discussed.  相似文献

Summary x-rayed adult males ofDrosophila melanogaster were left with untreated females from 2 to 3 days after which the males were discarded. Sex-linked recessive lethals and translocations were scored in progeny produced during the first 2 or 3 days following irradiation, and after storage of the spermatozoa in the females for 6 days.The results obtained show that the frequencies of sex-linked lethals and of translocation involving the two large autosomes and the x-chromosome were unchanged by storage. In experiments in which Y, 2, 3 translocations were scored both the 2–3, and the Y translocations showed a slight increase. The composition of the lacunal air in Phragmites australis (Cav.) Trin. ex Steudel during a diel cycle and during prolonged exposures to light and dark, respectively, was studied in the laboratory. The experimental system was constructed so that samples of the lacunal air could be withdrawn with syringes at fixed points in the stems and in the rhizomes of an intact shootsystem. The composition of the samples (CO2, O2, N2) was analyzed by gas chromatography.

A steep gradient in O2 concentration was observed within the lacunae, with highest concentrations in the aerial stems (21%) and lowest concentrations in the deepest-growing rhizomes (3.6%). Inverse gradients of CO2 and N2 occurred in the opposite direction. The composition of the lacunal air exhibited a clear diel variation, with highest O2 and lowest CO2 and N2 concentrations during the daytime.

It is concluded that the transport of gases within the lacunae of P. australis is mediated by diffusion following the concentration gradients of the gases, and convective flow of air resulting from solubilization of respiratory CO2 in the interstitial waters and/or in the transpiration stream. During the daytime, the ventilation is accelerated, probably as a consequence of thermo-osmotic pressurization resulting from temperature differences between the aerial plant tissues and the ambient air.  相似文献

The influence of defects in DNA repair processes on X-ray-induced genetic damage in post-meiotic male germ cell stages of Drosophila melanogaster was studied using the 'maternal effects approach'. Basc males were irradiated in N2, air or O2 either as 48-h-old pupae (to sample spermatids) or as 3-4-day-old adults (to sample mature spermatozoa) and mated to females of 3 repair-deficient strains (mei-9a: excision-repair-deficient; mei-41D5: post-replication-repair-deficient; mus(1)101D1: post-replication-repair-deficient and impaired in DNA synthesis). Simultaneous controls involving mating of males to repair-proficient females (mei+) were run. The frequencies of sex-linked recessive lethals and of autosomal translocations were determined following standard genetic procedures. The responses elicited in the different crosses with repair-deficient females were compared with those in mei+ crosses. The main findings are the following: with mei-9 females, the frequencies of recessive lethals are higher after irradiation of spermatids in N2, but not after irradiation in air of O2 (relative to those in the mei+ crosses); this result is different from that obtained in earlier work with spermatozoa, in which cell stage, higher yields of recessive lethals were obtained after irradiation of males in either N2 or air; in the mei-9 crosses, there are no significant differences in response (relative to mei+) after irradiation of either spermatozoa or spermatids in O2; the translocation frequencies in the mei-9 crosses are similar to those in the mei+ crosses, irrespective of the treated germ cell stage or the irradiation atmosphere; irradiation of either spermatozoa or spermatids in N2, air or O2 does not result in any differential recovery of recessive lethals in the mei-41 relative to mei+ crosses; irradiation of spermatids in N2 and of spermatozoa in air leads to a higher recovery of translocations in the mei-41 crosses; and after irradiation of spermatids or spermatozoa in any of the gaseous atmospheres, the frequencies of recessive lethals and of translocations are lower in the mus-101 crosses. The differences in responses (between cell stages, in different gaseous atmospheres and with different repair-deficient females) are explained on the basis of both qualitative and quantitative differences in the composition of the initial lesions and the extent to which their repair may be affected by the defects present in the different repair-deficient females. 1. The methyl viologen-catalyzed Mehler reaction was investigated in intact cells of five species of blue-green algae and Chlamydomonas reinhardi.

2. In the presence of methyl viologen, all the blue-green algae except Anabaena flos-aquae show a light-dependent O2 consumption as well as a post-illumination O2 evolution. The rate of O2 consumption is stimulated by 1 mM KCN, an inhibitor of catalase, but the dark O2 evolution becomes suppressed.

3. A. flos-aquae shows a light-dependent methyl viologen-catalyzed O2 uptake which is not affected by 1 mM KCN. Furthermore, there is no release of O2 in the dark following illumination.

4. With C. reinhardi, the cells do not show any net O2 exchange during or after illumination. Addition of 1 mM KCN, however, results in an immediate O2 uptake in the light.

5. Based on the mechanism postulated for the Mehler reaction in isolated chloroplasts, it was deduced that the differences in the kinetics of the O2 exchange catalyzed by methyl viologen reflect differences in the endogenous catalase activity in these algae. Cells of A. flos-aquae are deficient in catalase activity whereas those of the other blue-green algae possess catalase, although at low activity. C. reinhardi, on the other hand, has high catalase activity in vivo.

6. These findings are corroborated by results obtained from O2 electrode measurements of catalase activity in cell-free extracts of these algae.

采用开顶式气室熏蒸法,设置自然条件下臭氧(O3)浓度(对照,约40 nmol·mol-1)、80、160及200 nmol·mol-14个臭氧浓度,观测了不同浓度臭氧条件下银杏叶片可见伤害、活性氧生成量、抗氧化酶活性及相关基因表达变化情况,分析大气臭氧浓度升高对植物活性氧代谢的影响.结果表明: 160和200 nmol·mol-1 O3熏蒸明显伤害银杏叶片,80 nmol·mol-1与对照无差异,无可见伤害.O3处理20 d后,160和200 nmol·mol-1条件下银杏叶片的超氧自由基(O2)产生速率显著高于80 nmol·mol-1和对照,而80 nmol·mol-1与对照无差异;O3处理40 d后,160和200 nmol·mol-1熏蒸下叶片过氧化氢(H2O2)含量显著高于80 nmol·mol-1和对照,而过氧化氢酶(CAT)活性显著高于80 nmol·mol-1和对照,各臭氧处理抗坏血酸过氧化物酶(APX)活性均低于对照.熏蒸40 d后,CAT、APX基因的转录表达持续加强;防御素(GbD)的表达强度则随着臭氧浓度的增加及熏蒸时间的延长而呈显著加强.高浓度臭氧胁迫可使银杏叶片活性氧生成量增加、抗氧化酶活性下降、相关基因表达水平上调,有明显可见叶片伤害.  相似文献

The effects of the radioresistance factor rar-3 on the X-ray induction of various types of genetic damage in immature oocytes (about stage7) of Drosophila melanogaster were studied.

The dose-reduction factors previously postulated for rar-3 with respect to dominant lethals (1.58), sex-linked recessive lethals (1.87), non-disjunction of major chromosomes (1.58), and homologous interchanges (1.58)_were confirmed experimentally. It is concluded that all effects attributed arbitrarily to rar-3 are contributed by the single genetic factor rar-3.

No difference were found in quality of sex-linked recessive lethals (Y suppression, distribution over the X) induced in either rar-3 or rar-3+. Recombination frequencies were normal in unirradiated rar-3.  相似文献

The nitrogenase activity (measured by N2 or acetylene reduction) of cell-free extracts of the photosynthetic bacterium Chromatium was coupled to photochemically generated ATP and reductant. The ATP was formed through cyclic photophosphorylation by bacterial chromatophores. The reductant (reduced ferredoxin) was generated by a heated preparation (incapable of O2 and ATP production) of spinach chloroplasts. The nitrogenase activity of Chromatium extracts was supported by reduced Chromatium or Clostridium pasteurianum ferredoxin but not by that of spinach chloroplasts.  相似文献

Summary Storing of triethylene melamine-treated mature spermatozoa in untreated females was found to result in increased frequencies of both sex-linked recessive lethals and translocations involving the Y, II and III chromosomes. Frequencies of these mutations in effectively unstored spermatozoa were determined from progenies produced using sperm 2–4 days after treatment. The increase in translocation frequencies was on the order of 12-fold in progenies from sperm utilized 11–13 days after treatment when the sperm were stored at 25°C, and 3- to 6-fold when comparable sperm were stored at 12.5°C. Consistent but much smaller increases in frequencies of sex-linked lethals were found, with the increase in lethals tending to be correlated with relative increase in translocation frequency in a given experiment. On the assumption that sex-linked lethals related to chromosome breakage would be expected to increase in frequency in the same proportion as do translocations, approximate agreement was obtained when the proportions of breakage-related lethals among unstored lethals were estimated from the data in the four experimental series. The data are thus consistent with the hypothesis that chromosome breaks but not point mutations are realized during storage of treated spermatozoa. The reduction of ferricytochrome c by O2 and CO2 was studied in the pH range 6.6–9.2 and Arrhenius as well as Eyring parameters were derived from the rate constants and their temperature dependence. Ionic effects on the rate indicate that the redox process proceeds through a multiply-positively charged interaction site on cytochrome c. It is shown that the reaction with O2 and correspondingly with O2 of ferrocytochrome c) is by a factor of approx. 103 slower than warranted by factors such as redox potential. Evidence is adduced to support the view that this slowness is connected with the role of water in the interaction between O2/O2 and ferri-ferrocytochrome c in the positively charged interaction site on cytochrome c in which water molecules are specifically involved in maintaining the local structure of cytochrome c and participate in the process of electron equivalent transfer.  相似文献

1 Metabolic rates (Vo2), body temperature (Tb), and thermal conductance (C) were first determined in newly captured Maximowiczi's voles (Microtus maximowiczii) and Djungarian hamsters (Phodopus campbelli) from the Inner Mongolian grasslands at a temperature range from 5 to 35 °C.

2 The thermal neutral zone (TNZ) was between 25 and 32.5 °C for Maximowiczi's voles and between 25 and 30 °C for Djungarian hamsters. Mean Tb was 37.0±0.1 °C for voles and 36.2±0.1 °C for hamsters. Minimum thermal conductance was 0.172±0.004 ml O2/g h °C for voles and 0.148±0.003 ml O2/g h °C for hamsters.

3 The mean resting metabolic rate within TNZ was 2.21±0.05 ml O2/g h in voles and 2.01±0.07 ml O2/g h in hamsters. Nonshivering thermogenesis was 5.36±0.30 ml O2/g h for voles and 6.30±0.18 ml O2/g h for hamsters.

4 All these thermal physiological properties are adaptive for each species and are shaped by both macroenvironmental and microenvironmental conditions, food habits, phylogeny and other factors.

In Drosophila melanogater six chemicals were tested for radioprotectiveeffect against X-ray-induced genetic damage such as sex-linked recessive lethals and autosomal translocations using Oster's ring-X chromosome stock. A 2-day brood pattern was followed to score the damage induced at different spermatogenic stages separately. In all cases the chemicals were injected before X-irradiation. 10-mM solution of reduced glutathione (GSH) provided statistically significant protection against sex-linked recessive lethals in all broods. In translocation tests this chemical reduced the frequency in all broods but the result is not statistically significant. Cysteamine (MEA) did not show any protective effect but the frequency of lethals was slightly reduced in the first and fourth broods. 2-Aminoethyl isothiuronium Br·HBr (AET) showed a statistically significant protective effect when the data of the replicate experiments were pooled. Negative results were obtained for 5-hydroxytryptamine (5-HT) in sex-linked lethal tests. Aminoethyl phosphorothioate (AEPT) reduced the frequencies of both sex-linked lethals and autosomal translocations in all broods consistently but the results are not statistically significant. In tests for both lethals and translocations the reduction was largest in the stages with highest radiosensitivity. N(3-Aminopropyl)aminoethyl phosphorothioate (3AP-AEPT) gave no protection.  相似文献

A mass spectrometer has been used to measure O2 production and consumption and net CO2 exchange near the CO2 compensation point in Chlorella pyrenoidosa and Chlamydomonas reinhardtii grown in air and in 2% (v/v) and 5% (v/v) CO2 in air. Some experiments were run in presence of -hydroxy-2-pyridinemethanesulfonic acid and, for others, data are presented from parallel assays for enzymic glycolate oxidation. Although the glycolate-oxidising enzyme was present in cells grown with and without CO2 enrichment, the activities measured were not compatible with direct measurements of O2 consumption in the light. This and other evidence argues against glycolate being the primary substrate for light-induced O2 consumption by these organisms.  相似文献

This paper presents results of the genetic and cytological analysis of 144 sex-linked recessive lethals, plus 1 non-lethal. All of them were induced by IR hybrid dysgenesis. This collection of mutants was pooled from experiments involving inducer chromosomes that differ in the chrosomal position of their I elements. Our results show that 30% of the recessive lethals are associated with chromosomal rearrangements which depend on the strength of the IR interaction. These lethals are induced on both inducer- and reactive-origin chromosomes, and their frequency is dependent on the structure of the inducer chromosome used. The IR-induced lethals occur along the entire length of the X chromosome. These sites probably correspond to specific loci which are more or less homologous with I. The complementation relationshups showed that some specific loci were more frequently involved in all the lethal mutations tested. The most sensitive loci are, in order of observation: l(1)J1, ct, f, ma1 and m. Among induced recessive lethals considered to be point mutation, complementation tests showed that many of them are in fact multilocius deficiencies which can be detected only at the molecular level.

The nitrous oxide (N2O) reductase (nos) gene cluster from Achromobacter cycloclastes has been cloned and sequenced. Seven protein coding regions corresponding to nosR, nosZ (structural N2O reductase gene), nosD, nosF, nosY, nosL, and nosX are detected, indicating a genetic organization similar to that of Rhizobium meliloti. To aid homology studies, nosR from R. meliloti has also been sequenced. Comparison of the deduced amino acid sequences with corresponding sequences from other organisms has also allowed structural and functional inferences to be made. The heterologous expression of NosD, NosZ (N2O reductase), and NosL is also reported. A model of the CuA site in N2O reductase, based on the crystal structure of this site in bovine heart cytochrome c oxidase, is presented. The model suggests that a His residue of the CuA domain may be a ligand to the catalytic CuZ site. In addition, the origin of the spectroscopically-observed Cys coordination to CuZ is discussed in terms of the sequence alignment of seven N2O reductases.  相似文献
