Growth and ion accumulation were measured in callus culturesof Cicer arietinum L. cv. BG-203, grown on media supplementedwith 0–200 mol m–3 NaCl. Fresh and dry weights decreasedat concentrations ranging from 100–200 mol m–3,the reduction being greater during the third and fourth weeksof culture. Slight stimulation of growth was observed at 25and 50 mol m–3 NaCl. There was also a decrease in tissuewater content (fresh weight: dry weight) at 100–200 molm–3 NaCl. The concentration of Na+ and Cl in thetissue increased with increasing salinity of the medium. Mostof the accumulation of these ions occurred by the first weekwhile significant growth inhibition became apparent by onlythe third week of culture. Tissue K+ and Mg2+ decreased withincreasing salinization, the decrease being greater in K+ levels.Levels of Ca2+, however, were maintained throughout the experimentalrange. Millhouse, J. and Strother, S. 1987. Further characteristicsof salt-dependent bicarbonate use by the seagrass Zostera muelleri.—J.exp. Bot. 38: 1055–1068. The contribution of HCO3to photosynthetic O2 evolutionin the seagrass Zostera muelleri Irmisch ex Aschers. increasedwith increasing salinity of the bathing seawater when the inorganiccarbon concentration was kept constant. K1/2 (seawater salts)for HCO3 -dependent photosynthesis was 66% of seawatersalinity. Both short- and long-term pretreatment at low salinitiesstimulated photosynthesis in full strength seawater. Twentyfour hours pre-incubation of seagrass plants in 3·0 molm–3 NaHCO3 resulted in increased photosynthesis at allsalinities, apparently due to stimulation of HCO3 use(K1/2 (seawater salts) = 26%). Vmax (HCO3) was not affectedby low salinity pretreatment. The kinetics of HCO3 stimulationby the major seawater cations was investigated. Ca2+ was themost effective cation with the highest Vmax (HCO3) andwith K1/2(Ca2+) = 14 mol m–3. Mg2+ was also very effectiveat less than 50 mol m–3 but higher concentrations wereinhibitory. This inhibition cannot be accounted for solely byprecipitation of MgCO3. Na+ and K+ were both capable of stimulatingHCO3 use. Stimulation was in two distinct parts. Up to500 mol m–3, both citrate and chloride salts gave similarresults (K1/2(Na+) 81 mol m–3, Vmax(HCO3) 0·26µmol O2 mg–1 chl min–1), but use of citratesalts above 500 mol m–2 caused a second stimulation ofHCO3 use (K1/2(Na+) 830 mol m–3, Vmax(HCO3)0·68 µmol O2 mg–1 chl min–1). Vmax(HCO3)for the second-phase Na+ or K+ stimulation was of the same orderas for Ca2+-stimulated HCO3 use. To further characterizesalt-dependent HCO3 use, the sensitivity of photosynthesisto Tris and TES buffers was investigated. The effects of Trisappear to be due to the action of Tris+ causing stimulationof HCO3 -dependent photosynthesis in the absence of salt,but inhibition of HCO3 use in saline media. TES has noeffect on photosynthesis. External carbonic anhydrase, althoughimplicated in salt-dependent HCO3 use in Z. muelleri,could not be detected in whole leaves. Collection of Pure Phloem Sap from Wheat and its Chemical Composition   总被引:5,自引:0,他引:5  
Wheat Phloem sap was collected without contamination from thestylets of small brown planthopper severed by a YAG laser beam.The sugar, amino acid and inorganic ion composition was determinedusing only one µl of the sap. The sap had a high sucrose level (251 mM), and also a high K+level (299 mM). Total amino acid compounds in the sap reached262 mM. The dominant amino acids were glutamic acid, asparticacid and serine, while r-amino butylic acid was absent. Themajor anion in the sap was Cl and its concentration was25.1 mM. Nitrate was also present at a concentration of 8.1mM. These results suggested that the sap obtained from the cut endof the stylets of the small brown planthopper was a phloem originof wheat. The amino acid analog p-fluorophenylalanine (FPA) inhibitedsugar and K+ secretion by nectary glands. FPA specifically reducedthe net excretion of Na+ and Cl by the salt glands ofthe halophyte Limonium vulgare and 36Clexcretion by theglands of the pitcher walls of the carnivorous plant Nepenthes.Net uptake and net accumulation of Na+ and Cl by Limoniumleaf tissue and 36Cl accumulation in Nepenthes pitchertissue were much less inhibited than excretion. The resultsare discussed in relation to literature reporting similar specificeffects of FPA on transport of ions from the symplast of barleyroots into the dead xylem elements. Osmotic adjustment and decreases inthe bulk tissue elasticity occurred to a similar extent in bothpopulations with increasing salinity. Cl concentrationsincreased with increasing salinity in both populations, whereasleaf Na+ concentrations increased only in the inland population,but were higher at all salinities in the marsh population. K+concentrations changed little with increasing salinity. Prolineconcentrations increased only at the highest salinity level,and did not difler significantly between populations. The physiological basis for salt tolerance has been studiedin the euryhaline alga Enteromorpha prolifera. Levels of inorganicions and organic (compatible) solutes have been measured. K+makes the major contribution towards the internal osmotic potentialof the cell, while Cl and, in particular, Na+ contentsare low. Levels of the organic solute ß-dimethylsulphonio-propionate(DMSP) are high but are fairly insensitive to changes in theexternal salinity. Levels of amino-acids, calcium, phosphateand sulphate contribute relatively little towards the internalosmotic potential of the alga. As salinity is altered there are marked changes in the tissuewater content and volume. These changes directly affect theconcentration of the osmotic solutes within the cell. In diluteseawaters there is an increase in turgor as there is littlechange in the internal solute content of the cell compared tovalues in normal sea water. Inorganic ions, in particular K+,and organic solutes are accumulated in concentrated seawaters,although concentrations greater than 2·00 x seawaterresult in a reduction in the internal osmotic potential of thecell, mainly through loss of K+. Following 20 d of exposure to 75 or 150 mol m–3 NaCl Sorghumbicolor (L.) Moench plants become capable of growing in mediumcontaining 300 mol m–3 NaCl. Control plants, which havenot been pretreated, or plants pretreated for less than 20 ddie within 2 weeks when exposed to 300 mol m–3 NaCl. Weconsider this induction of a capacity to survive in and toleratea high NaCl concentration as an adaptation to salinity. We suggestthat adaptation to salinity is more than osmotic adjustmentand that it takes longer to develop than osmotic adjustment.Concomitantly with the appearance of the ability to grow inhigh salinity, adaptation also comprises the development ofa capacity to regulate internal Na+ and Cl concentrations,even when external salinity is high. Shoot mean relative growthrates are similar for both control plants and for adapted plantsgrowing in 300 mol m–3 NaCl, although their shoot Na+and Cl concentrations are quite different. Based on thesedata, we propose that adaptation of Sorghum to high salinityresults from a modulation of genome expression occurring duringextended exposure to non-lethal NaCl concentrations. The effect of NaCl salinity on growth and development of somaticembryos of Sapindus trifoliatus L. was examined. Incorporationof 25 and 50 mol m–3 NaCl into the medium greatly increasedthe growth and development of somatic embryos and both theseconcentrations favoured the production of secondary embryoids.However, supplementation of 100 mol m–3 NaCl to the mediumdid not have any significant effect on the growth and developmentof somatic embryos. On the other hand, the culturing of proembryostructures in medium containing 200 mol m–3 NaCl resultedin complete death within 7 d of salt exposure. Analysis of somatic embryos revealed that, upon salinization,they accumulated Na+ and Cl in significant amounts butthe content of Na+ was much less compared to that of Cl.Addition of NaCl (up to 50 mol m–3) in the medium resultedin a considerable increase in the K+ content of somatic embryos.The content of proline in somatic embryos, however, increasedsubstantially in response to salinization. The amount of freesterols, steryl glycosides, steryl esters, and phospholipidsalso rose to higher values in salt-affected somatic embryos.The results suggest that somatic embryos of S. trifoliatus cantolerate concentrations of NaCl up to 100 mol m–3 withoutaffecting growth and that they have sufficient cellular mechanismsto tolerate salinity at relatively high levels. Ion absorption by Chara corallina from solutions containingpredominantly KC1 or RbCl at up to 100 mol m–3 resultedin accumulation of salts and turgor regulation. Turgor regulationdid not occur in solutions containing Na+ or Li+salts. Duringion absorption from various salts of K+ and Rb+ vacuolar cationconcentration exceeded Cl concentration. This differencewas shown to be balanced by the synthesis and accumulation ofmalate. Vacuolar malate concentration reached 48 mol m3,with accumulation occurring at rates of up to 0.45 mol m–3h–1. Malate accumulation was inhibited by low externalpH and was dependent upon external HCO3 concentration.The synthesis of malic acid and its subsequent dissociationimposed a severe acid load on the cell. Biophysical regulationof cellular pH was achieved by a H+efflux at a rate of about40 nmol m–2 s–1from the cell. The results presentedargue against cytoplasmic Cl, HCO3 or pH regulatingmalate accumulation in Chara and it is suggested that malatetransport across the tonoplast may regulate malate accumulation. The growth of Atriplex amnicola, its water and ion relations,and carbohydrate use were investigated in response to the interactiveeffects of salinity and root zone hypoxia in an experiment conductedin nutrient culture. One week of hypoxia in the root zone atboth 50 and 400 mol m–3 NaCl caused the cessation of rootgrowth, a reduction in shoot growth, and adversely affectedwater relations, but not ion relations or carbohydrate concentrations.Two weeks of hypoxia at 400 mol m–3 NaCl resulted in thedeath of root tips, a 20–fold increase in the resistanceto water flow from the exterior of the roots to the leaves,and a further deterioration in water relations. There was alsoa doubling of Cl concentrations in the xylem sap anda doubling of Na+ and Cl concentrations in the leaves.An increase in the concentration of starch in the leaves, andsugars in the leaves, stems and roots, indicated that therewere problems with carbohydrate use rather than supply. Underthe prevailing conditions of low vapour pressure deficit, iontoxicity was the most probable cause of injury to A. amnicolain hypoxic solutions at high salinity. The response of A. amnicolato the interactive effects of salinity and hypoxia were similarto those reported for non-halophytes, but occurred at highersalinities. The physiological basis for salt tolerance has been studiedin the euryhaline marine alga Enteromorpha intestinalis. Adaptationto dilute and concentrated seawaters has been investigated inthree separate populations of this alga: marine, rock pool andestuarine. Internal K+, Na+ and Cl levels have been determined usingtracer efflux analyses. K+ has been shown to be the major osmoticsolute within this alga. Cellular levels of Cl and, inparticular, Na+ are low although levels in the cell wall arehigh. Levels of these ions varied considerably between the separateplants; K+ levels within marine plants of E. intestinalis aretwo to four times those found in the other populations. Thetertiary sulphonium compound ß-dimethylsulphonio-propionateis maintained at relatively high levels, although it remainsfairly insensitive to change in the external salinity. Changes in the tissue water content and cell volume are large,particularly within the estuarine plants. The thin cell wallsof these plants allow large changes in volume in the diluteconditions experienced in an estuary, while low turgor preventscell rupture. Thicker cell walls and small cells of the marineand rock pool plants assist in tolerating high and low externalosmotic potential—the estuarine plants respond poorlyto concentrated seawater. Maize plants, subjected to 0, 80, 120 and 160 meq l–1salinity using NaCl, showed adverse effects on viability, germinationand tube growth of pollen, besides enhancing the bursting ofpollen. The endogenous levels of various metabolites in pollenwere also affected. Pollen grains from salinized plants hadmore soluble carbohydrates, free amino acids, especially proline,phenols and DNA and less starch, protein and RNA compared tothe non-saline controls. Salinity also resulted in the accumulationof ions such as Na+, K+ and Cl while it caused a reductionin the boron content of pollen. These metabolic disturbancespossibly lead to decreased viability, germination and tube growthof pollen thereby resulting into a reduction in reproductivecapacity of the plants under salt stress. Salt Tolerance in the Succulent, Coastal Halophyte, Sarcocornia natalensis   总被引:2,自引:0,他引:2  
The effects of 0, 50, 100, 200, 300, 400 and 500 mol m–3NaCl on growth and ion accumulation in the succulent, coastalhalophyte Sarcocornia natalensis (Bunge ex Ung.-Sternb.) A.J. Scott were investigated. Increase in salinity from 0 to 300 mol m–3 NaCl stimulatedproduction of fresh, dry, and organic dry mass, increased succulenceand shifted resource allocation from roots to shoots. Growthwas optimal at 300 mol m–3 and decreased with furtherincrease in salinity. Water contributed to a large proportion of the increase in freshmass. Inorganic ions, especially Na+ and Cl– contributedsubstantially to the dry mass. At 300 mol m–3 NaCl inorganicions contributed to 37% of total dry mass and NaCl concentrationin the shoots was 482 mol m–3. Expressed sap osmotic potentialsdecreased from –2.10 to –3.95 MPa as salinity increasedfrom 0 to 300 mol m–3 NaCl. Massive accumulation of inorganicions, especially Na+ and Cl, accounted for 86% of theosmotic adjustment at 300 mol m–3 NaCl. Salinity treatments decreased the concentrations of K+ in shoots.Plant Na+ :K+ ratios increased steadily with salinity and reacheda maximum of 16.6 at 400 mol m3 NaCl. It is suggested that the exceptional salt tolerance of S. natalensisis achieved by massive inorganic ion accumulation which providessufficient solutes for osmoregulation, increased water fluxand turgor-induced growth. The contribution of membrane transport to regulation of cytoplasmicpH in Chara corallina has been measured during proton-loadingby uptake of butyric acid. In the short-term (i.e. up to 20min) uptake of butyric acid is not affected by removal of externalK+, Na+ or Cl but over longer periods uptake is decreased(by 20–50% in different experiments) in the absence ofexternal Na+ or, sometimes, K+. Influxes of both Na+ and K+increase temporarily after addition of butyrate, Na+ immediatelyand K+ after a lag. Effects on Cl influx are small butCl efflux increases enormously after a short lag. Anapproximate comparison of internal butyrate with changes inthe concentration of K+, Na+, and Cl suggests that initially(i.e. for a few min) cytoplasmic pH is determined by bufferingand possibly by some decarboxylation of organic acids (biochemicalpH regulation), and that biophysical pH regulation involvingefflux of H+ balanced by influxes of K+, Na+ and especiallyefflux of Cl progressively becomes dominant. When butyric acid is washed out of the cells, cytoplasmic pHis restored completely or partially (depending on the butyrateconcentration used) and this is independent of the presenceor absence of external Cl. Where Cl is present,its influx is relatively small. It is suggested that cytoplasmicpH is then controlled biochemically, involving the synthesisof an (unidentified) organic acid and the accumulation of acidicanions in place of butyurate lost from the cell. During thesecond application of butyrate, net Cl efflux is small:it is suggested that control of cytoplasmic pH then involvesdecarboxylation of the organic acid anions. The questions of the source of Cl lost from the cell(cytoplasm or vacuole) and of possible cytoplasmic swellingassociated with the accumulation of butyrate are discussed. The early time-course of turgor adjustment following a hyper-or hypo-osomotic shock was examined in the brackish-water charophyteLamprothamnium papulosum. The response to a reduction in turgorwas a five to seven-fold stimulation of the influxes of Cl,K+ and Na+. The distribution of radioactive tracers in the cellsuggested that the ionic composition of the cytoplasm was strictlycontrolled during turgor adjustment. Metabolic activity wasrelatively unaffected by the loss of turgor. high fluxes throughthe cytoplasm, and a cytoplasmic K concentration possiblyas high as 280 mol m–3. Osmotic adjustment to a lower salinity was achieved by largeincreases in the passive effluxes of K+ and Cl ratherthan by decreases in their influxes. The membrane remained hyperpolarized during hyperosmotic adjustmentbut depolarized after a hypo-osmotic change. The response of jojoba [Simmondsia chinensis (Link) Schneid]plants to salinity was studied in solution culture. At concentrationsof 0, 100, 200 and 600 m-mol l–1 NaCI it was found thatjojoba plants have high tolerance to NaCl. The growth of theseplants was not affected by salinity. They accumulated largeamounts of Cl, Na+ and proline. These amounts decreasedrapidly in plants transferred back to control medium. Potassiumcontent decreased in NaCl-treated plants and tended to increaserapidly to the control level in plants transferred to controlmedium. The effect of salinity on water balance was not appreciable.As suggested for other xerophytic species, it could be assumedthat the high tolerance of jojoba to salinity plays an importantrole in its ability to endure periods of drought. The role ofproline during or after stress remains an open question. The Cl fluxes across the plasma membrane and the Clcontent of the acid–resistant alga Dunaliella acidophila(optimal growthat pH 1.0, positive membrane potential) werestudied in the presence of 0.01–300 mM Cl. Up to40 mM Cl in the medium, theinternal Cl concentrationis higher than that predicted by the electrochemical equilibrium,whereas at higher external Cl concentrations internalCl levels are lower than expected for the electrochemicalequilibrium. Growth in the absence of Cl is significantlylower than in the standard growth medium (2.2 mM Cl)and this reduction cannot be overcome by the addition ofothermonovalent anions such as Br or NO3 The latterimplies a specific Cl requirement in addition to therole of Cl as apermeant anion during ion translocations.Growth and photosynthesis tolerate an excess of Cl upto 300 mM (without stepwiseadaptation to increasing salinity).The uptake of Cl (measured by tracer techniques) exhibitsMichaelis–Menten kinetics (KM = 0.75 mM Cl) andis stimulated by light and high H+ concentrations. Internalacidification by acetic acid causes an inhibition of Cluptake. The uptake of Cl is inhibited by the monovalentanions Br, I, and NO3 with K1, values notvery much different from the KM. value for Cl. The aniontransport inhibitors SITS and DIDS do not affect photosynthesis,but strongly suppressthe uptake of Cl. The Clchannel blockers A–9–C and NPPB cause inhibitionsof Cl uptake as well as of photosynthesis andthe ATPpool. FCCP strongly depresses the internal ATP–pool withouta marked effect on Cl uptake. Cl efflux was inhibitedbyDIDS and SITS, but stimulated or inhibited by FCCP, dependingon the external Cl concentration. Results are in agreementwiththe hypothesis that Cl uptake into D. acidophila is dueto catalysed diffusion and is primarily independent of the hydrolysisofATP. Cl efflux is assumed to be coupled to an activepump. Data suggest tight co–operativity between the systemsresponsiblefor Cl uptake and Cl efflux, with thecytoplasmic pH and the membrane potential being important mediators. Uptake and partitioning through the xylem and phloem of K+,Na+, Mg2+ , Ca2+ and Cl were studied over a 9 d intervalduring late vegetative growth of castor bean (Ricinus communisL.) plants exposed to a mean salinity stress of 128 mol m–3NaCl. Empirically based models of flow and utilization of eachion within the whole plant were constructed using informationon ion increments of plant parts, molar ratios of ions to carbonin phloem sap sampled from petioles and stem internodes andpreviously derived information on carbon flow between plantsparts in xylem and phloem in identical plant material. Salientfeatures of the plant budget for K+ were prominent depositionin leaves, high mobility of K+ in phloem, high rates of cyclingthrough leaves and downward translocation of K+ providing theroot with a large excess of K+ . Corresponding data for Na+showed marked retention in the root, lateral uptake from xylemby hypocotyl, stem internodes and petioles leading to low intakeby young leaf laminae and substantial cycling from older leavesback to the root. The partitioning of the anionic componentof NaCl salinity, Cl, contrasted to that of Na+ in thatit was not substantially retained in the root, but depositedmore or less uniformly in stem, petiole and leaf lamina tissues.The flow pattern for Mg2+ showed relatively even depositionthrough the plant but some preferential uptake by young leaves,generally lesser export than import by leaf laminae, and a returnflow of Mg2+ from shoot to root considerably less than the recordedincrement of the root. Ca2+ partitioning contrasted with thatof the other ions in showing extremely poor phloem mobility,leading to progressive preferential accumulation in leaf laminaeand negligible cycling of the element through leaves or root.Features of the response of Ricinus to salinity shown in thepresent study were discussed with data from similar modellingstudies on white lupin (Lupinus albus L.) and barley (Hordeumvulgare L.) Fusicoccin (FC) inhibited net excretion of Cl by theglands of the pitchers of the carnivorous plant Nepenthes hookeriana;of Na+ and Cl by the salt glands of the halophytes Limoniumvulgare and L. pectinatum and of K+ in the nectar of Acer platanoidesflowers. It had no effect on K+ elimination with nectar of Impatienswalleriana (extrafloral nectaries) and Abutilon striatum. Abscisicacid (ABA) stimulated net excretion of K+ and Cl in Nepenthesand of Na+ and Cl in Limonium but had no effects on K+in nectar. Thus, FC and ABA had opposing effects on ion excretionby the salt eliminating glands of Limonium and Nepenthes. Bothcompounds, however, had similar effects on sugar secretion ofnectary glands which was either inhibited or unaffected by FCand ABA. It is suggested that the effects of FC and ABA on ion excretionby gland cells could be reconciled with literature showing FC-stimulationand possible ABA-inhibition of proton pumps at the plasmalemmaof plant cells. This paper describes experiments designed to investigate theeffects of increases in external osmotic pressure on the electrophysiologicalbehaviour of the plasmalemma in cells of the brackish-wateralga, Chara inflata. The electrical conductance of the plasmalemmaof these cells of, due to the diffusion of ions, consists mainlyof K+, Cl and leak components. The addition of sorbitolat concentrations in the range 40–280 mol m–3 tothe external solution bathing the cells, progressively and reversiblydepolarized the cell membrane and increased the total membraneconductance, chiefly due to increases in each of the separateionic conductances. At concentrations higher than about 280mol m–3 when the cells became plasmolysed, the effectsof sorbitol on the electrical properties of the cell ceasedto be fully reversible. When the membrane electrical potentialdifference is stepped in a negative direction with a voltage-clamp,the resulting inward current has voltage-dependent componentsconsisting of an inactivating K+ current, an activating Clcurrent and a constant leak current. The addition of sorbitolto the external solution modified these currents by increasingtheir magnitude, by increasing the half-time of the inactivationof the K+ current, and by decreasing the half-time of activationof the Cl current.
