The nuclear pore complexes are complex protein structures located in the nuclear envelope, where they control the nuclear-cytoplasmic transport, and inside the stacks of endoplasmic reticulum cisternae, annulate lamellae. After overexpression of some nucleoporins, numerous granules are visible in the cytoplasm. According to the published data, these granules are the annulate lamellae. In the current paper, the structural organization of POM121-containing granules was analyzed using correlative light and electron microscopy. The ultrastructural study demonstrates that POM121-containing granules are not annulate lamellae but aggregates of endoplasmic reticulum membranes. Thus, overexpressed POM121 is not able to induce the annulate lamella formation. The mechanisms of self-organization of non-functional structures (such as the aggregates of endoplasmic reticulum membranes described here) and possible involvement of these mechanisms in the formation of cellular structures are discussed.  相似文献

Characteristic intracytoplasmic lamellar bodies were found in a malignant gonadal stromal tumor. These bodies consisted of the stacks of up to 200 tubular cisternae arranged in parallel. Each cisterna had a circular section in tangential view and a diameter of about 85 nm. The cisternae on the outermost side of these lamellar bodies tended to be dilated and adorned with ribosomes. The ends of cisternae were often contiguous with rough-surfaced endoplasmic reticulum. The latter feature is also seen in annulate lamellae, but periodically spaced annuli or discontinuities characteristic of annulate lamellae were never observed. Furthermore, fine ribosomal granules resembling a rosary were recognizable along the whole circumference of the outer surface of each cisterna. The unique structure we describe is a cytoplasmic organelle which, like annulate lamellae, is closely associated with the endoplasmic reticulum and is presumed to be related to the genesis of rough-surfaced endoplasmic reticulum in tumor cells.  相似文献

Semi-isolated annulate lamellae were prepared from single newt oocytes (Triturus alpestris) by a modified Callan-Tomlin technique. Such preparations were examined with the electron microscope, and the negative staining appearance of the annulate lamellae is described. The annulate lamellae can be detected either adhering to the nuclear envelope or being detached from it. Sometimes they are observed to be connected with slender tubular-like structures interpreted as parts of the endoplasmic reticulum. The results obtained from negative staining are combined with those from sections. Especially, the structural data on the annulate lamellae and the nuclear envelope of the very same cell were compared. Evidence is presented that in the oocytes studied the two kinds of porous cisternae, namely annulate lamellae and nuclear envelope, are markedly distinguished in that the annulate lamellae exhibit a much higher pore frequency (generally about twice that found for the corresponding nuclear envelope) and have also a relative pore area occupying as much as 32% to 55% of the cisternal surface (compared with 13% to 22% in the nuclear envelopes). The pore diameter and all other ultrastructural details of the pore complexes, however, are equivalent in both kinds of porous cisternae. Like the annuli of the nuclear pore complexes of various animal and plant cells, the annuli of the annulate lamellae pores reveal also an eightfold symmetry of their subunits in negatively stained as well as in sectioned material. Furthermore, the annulate lamellae are shown to be a site of activity of the Mg-Na-K-stimulated ATPase.  相似文献

Annulate lamellae have been found in a primitive neuroectodermal tumor, a metastatic cerebellar tumor, a testicular seminoma, a retinoblastoma and three melanomas. These annulate lamellae are arranged in stacked parallel arrays in the cytoplasm of tumor cells. The number of annulate lamellae observed to comprise a single stack varies from 2--4 in the seminoma tumor to 5--18 in the cerebellar tumor. Although the functional significance of annulate lamallae is still unknown, in many instances they have been found to be continuous with rough-surfaced cisternae of the endoplasmic reticulum and ribosomes have been demonstrated on the surface of annulate lamellae. This may suggest that annulate lamellae participate in protein synthesis.  相似文献

Summary Structures identified as annulate lamellae, lamellar bodies and subsurface cisternae were found in neurons of the hyperstriatum accessorium of the avian forebrain. Annulate lamellar arrays with up to six lamellae were present in the larger somata. The lamellae were made up of fused smooth-surfaced cisternae forming pores or annuli and were surrounded by a dense filamentous to granular material. Stacks of nonfenestrated, parallel, regularly spaced cisternae, designated as lamellar bodies, also appeared in the cytoplasm. When flattened they were reminiscent of the electron dense subsurface cisternae. Continuity could be demonstrated between peripherally located subsurface cisternae and lamellar bodies. The dense filamentous to finely granular substance was also located between these structures. Annulate lamellae, lamellar bodies and subsurface cisternae were always observed in conjunction with the rough endoplasmic reticulum. The functional significance of these structural associations is considered.  相似文献

Summary During meiotic maturation, the cortex of oocytes of Xenopus laevis undergoes structural reorganization, visualized in this study by freeze-fracture electron microscopy. In the full-grown but immature oocyte, annulate lamellae are dispersed throughout the subcortex of the egg, 5 to 20 m from the plasma membrane. The annulate lamellae consist of well-organized stacks of membrane with visible pores. Stimulation of meiotic maturation by progesterone leads to disruption of the annulate lamellae and formation of an elaborate cortical endoplasmic reticulum which surrounds the cortical granules and intertwines throughout the cortex of the mature egg. Pore-like structures similar to those previously observed in the subcortical annulate lamellae are observed in the mature cortical endoplasmic reticulum. The cortical endoplasmic reticulum is often in close apposition with the plasma membrane and with membranes of cortical granules, but no junctions are visualized. This study provides further evidence that the cortical endoplasmic reticulum develops during progesterone-stimulated meiotic maturation in vitro, and that the annulate lamellae are precursors to the cortical endoplasmic reticulum.  相似文献

Cytoplasmic annylate lamellae were found in the islet organ of a cyclostome, the hagfish (Myxine glutinosa), predominantly in cells interpreted as young proliferating beta-cells, and also in endocrine cells and enterocytes of the bile duct and gut and in the endothelial cells of small blood vessels. A close association was observed annulate lamellae and granular endoplasmic reticulum. Both in cells with and in those without annulate lamellae, crystalline inclusions of proteinaceous nature were seen in granular endoplasmic reticulum. These inclusions were occasionally closely associated to annulate lamellae, and a direct continuity could be seen between granular endoplasmic reticulum and the outer nuclear membrane surrounding an inclusion partially situated in the perinuclear cisterna. Rod-shaped structures and rounded electron dense bodies were seen in the nuclei of some islet parenchymal cells. The presence of annulate lamellae in the islet organ and associated tissues of Myxine glutinosa is believed to be related to the very high phylogenetic age of this species. The close association observed between annulate lamellae, granular endoplasmic reticulum, crystalline inclusions, and sometimes also nuclear membranes, may be of functional significance.  相似文献

Summary A multilamellate body (MLB), bearing close resemblance to an array of annulate lamellae, has been observed in several adenohypophysial cell types of the teleost, Hemihaplochromis philander. In longitudinal section, each MLB comprises a ladder-like row of 12–50 sausage-shaped profiles, termed lamellae. A few lamellae in each section show connections with the endoplasmic reticulum. Apposition of paired lamellar membranes at regularly spaced intervals results in a beaded appearance, whereby sites of membrane apposition are probable pore sites. The MLB differs from annulate lamellae in having poorly developed pores and closer packing of lamellae. It is suggested that the MLBs described here, may represent annulate lamellae at a stage of development or break-down when pores are incomplete.Acknowledgement is made to the South African Council for Scientific and Industrial Research for financial aid towards this work. Summary Dictyosomes in three higher plant cell secretory systems,Zea root cap cells,Tradescantia pollen tubes and digestive glands ofDionaea fly traps, are shown to possess a structural polarity in terms of either the production of secretory vesicles, or the reaction of individual cisternae to the osmium-zinc-iodide impregnation procedure. These observations contradict previous claims that higher plant dictyosomes lack structural polarity. Doubts about the applicability of the endomembrane flow concept to higher plant cell dictyosomes are discussed in relation to the relative balance between carbohydrate and protein in secreted products. The lack of dictyosome-associated endoplasmic reticulum in some of these plant cell systems is confirmed.  相似文献

Post-fertilization events leading to the cleavage of the zygote of the sea-urchin, Arbacia punctulata were examined with the light and electron microscopes. Prior to prophase of the first cleavage division, endoplasmic reticulum and annulate lamellae become organized around the zygotic nucleus to produce a crescent-shaped structure which is defined as the streak (Harvey, '56). With the advent of prophase the streak undergoes morphogenic events which lead to the formation of the mitotic asters. During this transition there is a loss of annulate lamellae and a concomitant increase in endoplasmic reticulum. Annulate lamellae are not found as a part of the mitotic apparatus and are not again observed within the embryo until the two cell stage. During telophase, karyomeres are formed which consist of chromosomes delimited by a porous bilaminar envelope. Blastomere nuclei are produced following the fusion of the outer laminae, and subsequently by the fusion of the inner laminae of the envelopes encompassing the karyomeres.  相似文献

The ultrastructure of neurons of the diffuse supraoptic nucleus of the hamster has been studied. These neurons show two specializations of the endoplasmic reticulum: annulate lamellae and whorl bodies. From one to three whorl bodies are found in the same neuron. The annulate lamellae and the whorl body cisterns are continuous with the cisterns of the rough endoplasmic reticulum. These neurons present an extraordinarily developed rough endoplasmic reticulum, small mitochondria, neurosecretory vesicles and a Golgi complex filled with electron-dense material. Astrocytic processes of different thickness surround the neurosecretory cells.  相似文献

Summary Hindgut epithelial cells of the aquatic isopod,Asellus communis, and the terrestrial isopod,Armadillidium vulgare, possess annulate lamellae. The organelle is present in non-dividing cells of intermolt adult organisms. The lamellae exhibit dense pore areas traversed by diaphragms, and the lamellae are associated with elements of endoplasmic reticulum.This research was supported in part by a grant to E. R. Pineal glands from young adult hamsters (Mesocricetus auratus Water-house) kept on a 14-10 light-dark photoperiod contain annulate lamellae. These annulate lamellae are apparently limited to the light pinealocytes. They are found in close proximity to nuclei, the Golgi apparatus, and agranular endoplasmic reticulum. There is also an interesting association or microtubules with the annulate lamellae. It seems reasonable that the presence of annulate lainellae in the pinealocytes may have some correlation with the physiological function or functions of these cells.  相似文献

The endoplasmic reticulum (ER) of rooster's spermatids was analyzed during spermiogenesis, which was subdivided into eight distinct steps on the basis of changes observed with the electron microscope in the nucleus, acrosome-perforatorium system, manchette, and flagellum. In steps 1 and 2, spermatids' ER cisternae presented the following specializations: A loose network of tubular cisternae was distributed throughout the cytoplasm. Six to eight tight networks of anastomosed tubular cisternae parallel to each other were closely stacked to form a discoid body (1.5-2.5 microns in diameter and 0.5-0.8-micron thick) in which spheroidal vesicles (0.4 micron in diameter) were inserted. Close to and connected with this body, called the alveolar body, there was a stack of annulate lamellae. Large, flattened ER cisternae were seen singly or in piles of two or three running parallel to the nuclear surface. A collection of tubular ER cisternae faced plaques of thickened plasma membranes. These elements of the ER system appear continuous with each other. During steps 3-5 of spermiogenesis, no modification of the alveolar body-annulate lamellae complex was noted; the large flattened ER cisternae disappeared, however, and the broad network of tubular cisternae developed markedly. During steps 6 and 7, the latter network of tubular cisternae fragmented into vesicles that swelled to give a vacuolated appearance to the cytoplasm. The alveolar body-annulate lamellae complex remained visible until late step 7, when it disintegrated just before spermiation. Thus the system of ER cisternae underwent marked structural modifications during spermiogenesis.  相似文献

本文研究了环孔片层在金鱼卵母细胞发育过程中的一些形态变化。描述卵母细胞发育的五个阶段中每个阶段环礼片层的分布以及排列的情况。结果表明随着卵母细胞的生长,环孔片层的出现率和每束中片层的数目都逐渐增加。除了在卵原细胞以外,环礼片层一般都分布在远离细胞核的卵质中。尚观察到孔结合物质和片层间物质与多核蛋白体有联系。我们从观察结果推测金鱼卵母细胞的环孔片层可能是由粗面内质网衍生而成。  相似文献

Résumé L'évolution en l'absence d'hormone cérébrale provoque, dans les ovocytes de N. diversicolor, la formation de lamelles annelées cytoplasmiques (L.A.). Le processus débute par l'apparition de lames de réticulum à proximité du noyau (15 jours d'évolution à 15° C). Les lames se rapprochent ensuite du noyau et des pores apparaissent, situés le plus souvent face à ceux de l'enveloppe nucléaire (20ème jour). La différenciation des lames du réticulum en L.A. progresse ensuite depuis les éléments les plus proches du noyau vers les plus externes.Dans le cytoplasme des ovocytes ayant évolué à la température de 20° C, on assiste en outre à la formation d'empilements de citernes ergastoplasmiques. Ces citernes, non situées à proximité du noyau, subissent également une transformation progressive en L.A.L'étude cytochimique effectuée par digestion enzymatique a montré que la pepsine dégrade fortement le matériel dense des lamelles annelées. Ce matériel semble donc renfermer une teneur élevée en protéines auxquelles pourrait se trouver associé du RNA.
Mode of formation of the annulate lamellae of oocytes of Nereis diversicolor developing in the absence of brain hormone
Summary Experimental elimination of brain activity leads to the formation of cytoplasmic annulate lamellae (L.A.) in oocytes of N. diversicolor. This process begins with the appearance of cisternae of endoplasmic reticulum in the vicinity of the nucleus (15 days of development at 15° C). Then, the cisternae move closer to the nucleus and pores appear, often facing the pores of the nuclear envelope (20th day). The differentiation of endoplasmic reticulum into L.A. progresses from the elements closest to the nucleus to the more peripheral ones.In the cytoplasm of oocytes which developed at 20° C, stacks of cisternae of rough endoplasmic reticulum have been observed. These cisternae, which are not localized near the nucleus, also undergo a progressive transformation into annulate lamellae.A cytochemical study, based on enzymatic digestion, shows that pepsin strongly digests the dense material of L.A. This material, therefore, seems to contain a high amount of proteins, perhaps associated with RNA.

Twelve nontumorous adenohypophyses and 36 various pituitary adenomas, removed by surgery, have been investigated by electron microscopy in order to shed some light on annulate lamellae, primarily on their ultrastructural features, incidence, origin, fate and functional significance. No annulate lamellae were found in the nontumorous adenohypophyses and in 33 pituitary adenomas. They were, however, detected in two adenomas consisting of undifferentiated cells and one adenoma composed of sparsely granulated prolactin cells indicating that these unique membrane configurations cannot be regarded as an exceedingly rare finding and, furthermore, that they may be disclosed not only in undifferentiated but occasionally in highly differentiated cells. Annulate lamellae may arise from endoplasmic reticulum and/or nuclear envelope and consist of arrays of smooth walled double membrane sheets exhibiting regularly spaced interruptions as well as continuities with the endoplasmic reticulum. No relationship was established between annulate lamellae and adenohypophysial secretory activity. Our findings seem to be consistent with the view that annulate lamellae are present in those cells which have the tendency to proliferate.  相似文献

This review is devoted to annulate lamellae, a specific compartment of endoplasmic reticulum that occurs, presumably, in actively growing and rapidly dividing cells (oocytes, embryonic and tumor cells). We summarized both earlier and recent data on the dustribution of annulate lamellae in various cell types, on their morphology, and the distribution of interaction with intracellular structures at various treatments. As the annulate lamellae contain cytoplasmic pore complexes, a special attention was paid to their relation with nuclear pores. Possible functions of the annulate lamellae in intracellular processes and, particularly, in nuclear envelope assembly, are discussed.  相似文献

HeLa cells from conventional culture media have been studied in thin sections with the electron microscope; in many cases cells were examined in sets of sections cut in series. The fine structure of the cells is described including three unusual features not hitherto reported. It has been found that numerous cells contained rows of parallel smooth surfaced cisternae spaced about 150 mµ apart and communicating with rough surfaced elements of the endoplasmic reticulum. These cisternae resembled "annulate lamellae" but did not contain regular arrays of pores. In many cells an area of juxtanuclear cytoplasm was occupied by a membranous structure composed of closely applied pairs of narrow cisternae either arranged in concentric rings or else extending in several directions in a haphazard manner. Sparse particles were present on the outer membranes of each pair of cisternae. Communications between the double cisternae and other membrane-bounded structures were not observed. A small number of cells contained areas of cytoplasm devoid of organelles and filled with amorphous fuzzy material. The observations recorded are discussed.  相似文献
