The biochemical and molecular responses of five commercially well-known pomegranate cultivars to severe water stress were studied. The cultivars were subjected to 14-day water stress by withholding irrigation, followed by re-watering for 7 days. Results showed clear differences in metabolites contents and activities of antioxidant enzymes among various pomegranate cultivars during severe water stress and recovery. According to our results, increased accumulation of proline in pomegranate was not related to osmotic adjustment during severe water stress. Except for ‘Ghojagh’, leaves grown under severe water stress conditions showed symptoms of oxidative stress such as reduced chlorophyll concentration. The improved performance of ‘Ghojagh’ under drought stress may be associated with an efficient osmotic adjustment. The up- or down regulated expression of cytosolic glutathione reductase (cytosolic GR) and glutathione peroxidase were observed under drought conditions. Moreover, the suppressed expression of cytosolic GR was also noted. Comparatively, ‘Rabab’ exhibited higher antioxidant capacity and an efficient ROS-scavenging mechanism under drought stress. Lower levels of membrane lipid peroxidation in ‘Ghojagh’ and ‘Rabab’ under drought stress and the marked reduction of malondialdehyde concentration after re-watering represents that these cultivars have a good tolerance to drought stress. As a first step towards the study of the biochemical and molecular responses of pomegranate plants to water stress, this research provides new information into the mechanisms of drought tolerance in the plants.

We investigated various physiological characteristics of two wheat (Triticum aestivum L.) cultivars differing in drought tolerance, i.e., Shannong16 (a drought-tolerant cultivar) and Weimai8 (a high-yield wheat cultivar under well-watered conditions), under field drought conditions. The experiments were conducted over a two-year period. Drought stress (DS) was imposed by controlling irrigation and sheltering the plants from rain. Compared with Weimai8, Shannong16 exhibited the better water balance, the higher osmotic adjustment, the slower degradation of chlorophyll, and the higher net photosynthetic rate under drought-stress conditions. At the same time, we observed that Shannong16 maintained more integrated chloroplast and thylakoid ultrastructure in flag leaves than Weimai8 under field drought stress. The different levels of antioxidant competence, indicated by MDA content, antioxidant enzyme activities, and the level of superoxide radicals observed in the two wheat cultivars may be involved in the different levels of drought resistance of these cultivars.

Drought stress is one of the most important factors limiting the survival and growth of plants in the harsh karst habitats of southwestern China. Detailed knowledge about the ecophysiological responses of native plants with different growth forms to drought stress could contribute to the success of re-vegetation programs. Two shrubs, Pyracantha fortuneana and Rosa cymosa, and four trees, Broussonetia papyrifera, Cinnamomum bodinieri, Platycarya longipes and Pteroceltis tatarinowii, were randomly assigned to four drought treatments, i.e. well-watered, mild drought stress, moderate drought stress, and severe drought stress. Midday water potential, the maximum quantum efficiency of PSII photochemistry (Fv/Fm), pigments, osmotic solutes (soluble sugars and proline), cellular damages, and antioxidant enzymes (superoxide dismutase, catalase and peroxidase) were investigated. Drought stress significantly decreased pigments content, but increased the ratio of carotenoids to total chlorophylls in the studied species. After prolonged severe drought stress, the two shrubs exhibited higher Fv/Fm, less reductions of midday water potential, and lower increases of malondialdehyde content and ion leakage than the four trees. Prolonged severe drought stress largely decreased accumulations of osmotic solutes and activities of antioxidant enzymes in the four trees, but significantly increased proline content and superoxide dismutase activity in the two shrubs and peroxidase activity in P. fortuneana. The positive relationships were observed among activities of antioxidant enzymes, and between contents of osmotic solutes and activities of antioxidant enzymes. These findings suggested that the two shrubs had higher tolerance to severe drought stress than the four trees due to higher capacities of osmotic adjustment and antioxidant protection.

Effects of the antioxidant system and chlorophyll fluorescence on drought tolerance of four common bean (Phaseolus vulgaris L.) cultivars were studied. The cultivars were positioned in the order of a decrease in their drought tolerance: Yakutiye, Pinto Villa, Ozayse, and Zulbiye on the basis of changes in the water potential, stomatal conductance, photosynthetic pigment content, and lipid peroxidation. Under drought conditions, the level of H2O2 was not changed in cv. Pinto Villa but decreased in other cultivars. Antioxidant enzymes (superothide dismutase (SOD), guaiacol peroxidase (GPX), catalase (CAT), ascorbate peroxidase (APX), and glutathione reductase (GR)) were generally activated in all cultivars. Interestingly, CAT, APX, and GR activities were not changed in cv. Pinto Villa, APX activity decreased in cv. Yakutiye, and CAT activity was not changed in cv. Zulbiye. The increases in SOD and GPX activities in cv. Ozayse were higher than in other cultivars. Drought stress reduced the effective quantum yield of PS2 (ΦPS2) and the photochemical quenching (qp), while it increased nonphotochemical quenching (NPQ) in all cultivars. The reduction or increase was more pronounced in cv. Zulbiye. There were generally significant correlations between qp, NPQ, and ROS scavenging by SOD and APX. Also, there were significant correlations between SOD and qp in tolerant cultivars and APX and qp in sensitive ones. The results indicate that activation of SOD and APX was closely related to the efficiency of PS2 in common bean cultivars. This interaction was essential for protection of photosystems and plant survival under drought.

Two cultivars of peanut (Arachis hypogaea L.) which were designated as resistant (Florispan) and sensitive (Gazipasa) according to their growth retardation under drought stress conditions were compared for their oxidative damage and antioxidant responses. Sixteen days-old peanut seedlings were subjected to PEG-6000 solutions of two different osmotic potentials; −0.4 and −0.8 MPa, and various growth parameters, photosystem II activity, changes in malondialdehyde (MDA), hydrogen peroxide (H2O2) and proline levels, activities of ascorbate peroxidase (APX), catalase (CAT), peroxidase (POX) and gluthatione reductase (GR) enzymes were determined. Both cultivars exhibited water deficit at −0.8 MPa osmotic potential of PEG-6000 and H2O2 levels significantly increased during exposure to −0.4 MPa osmotic potential. However, H2O2 levels were under control in both cultivars at exposure to −0.8 MPa osmotic potential. Significant proline accumulation was observed in the tissues of cv. Florispan at −0.8 MPa osmotic potential, whereas proline accumulation did not appear to be an essential part of the protection mechanism against drought in cv. Gazipasa. No significant variation in chlorophyll fluorescence values were detected in neither of the cultivars. Enzyme activity measurements revealed that Gazipasa copes well with lesser magnitudes of drought stress by increasing the activity of mainly APX, and during harsh stress conditions, only APX maintains its activity in the tissues. In cultivar Florispan, GR activity appears to take role in lesser magnitudes of drought stress, whereas CAT and APX activities appear to be very crucial antioxidative defenses during intense stress conditions. The results indicate that, the level of proline and activities of the enzymes CAT and APX are important mechanisms for the maintenance of drought tolerance in peanut plants.

Bermudagrass (Cynodon dactylon) is a widely used warm-season turfgrass and one of the most drought tolerant species. Dissecting the natural variation in drought tolerance and physiological responses will bring us powerful basis and novel insight for plant breeding. In the present study, we evaluated the natural variation of drought tolerance among nine bermudagrass varieties by measuring physiological responses after drought stress treatment through withholding water. Three groups differing in drought tolerance were identified, including two tolerant, five moderately tolerant and two susceptible varieties. Under drought stress condition, drought sensitive variety (Yukon) showed relative higher water loss, more severe cell membrane damage (EL), and more accumulation of hydrogen peroxide (H2O2) and malondialdehyde (MDA), while drought tolerant variety (Tifgreen) exhibited significantly higher antioxidant enzymes activities. Further results indicated that drought induced cell injury in different varieties (Yukon, SR9554 and Tifgreen) exhibited liner correlation with leaf water content (LWC), H2O2 content, MDA content and antioxidant enzyme activities. Additionally, Tifgreen plants had significantly higher levels of osmolytes (proline level and soluble sugars) when compared with Yukon and SR9554 under drought stress condition. Taken together, our results indicated that natural variation of drought stress tolerance in bermudagrass varieties might be largely related to the induced changes of water status, osmolyte accumulation and antioxidant defense system.

In previous works, we have established a correlation between antioxidant system response and tolerance to drought, osmotic stress and photooxidative stress of different wheat cultivars with contrasting drought tolerance. In the present work, a protocol to obtain and transform wheat protoplasts was established. Transgenic protoplasts with Manganese Superoxide Dismutase (Mn-SOD) (E.C.: 1.15.1.1) and Glutathione Reductase (GR) (E.C.: 1.6.4.2) overexpression in chloroplasts were obtained, and their responses to photooxidative stress were characterized. Protoplasts with Mn-SOD or GR overexpression, showed different responses and tolerance to photooxidative stress. Protoplasts with Mn-SOD overexpression showed lower levels of oxidative damage, higher level of endogenous hydrogen peroxide and a great induction of total SOD and GR activities during photooxidative treatments. In protoplasts with GR overexpression the oxidative damage provoked by the photooxidative treatment was similar to control protoplasts, the GSH content and GSH/GSH + GSSG ratio were higher than control and Mn-SOD transformed protoplast, and total SOD and GR activities were not induced. Our results suggest that the differential responses and tolerance to photooxidative stress given by Mn-SOD or GR overexpression, also depend on the effects of these enzyme activities over the cellular redox state balance, which modulate the responses to photooxidative stress.

Aquaporin proteins are part of the complex response of common bean (Phaseolus vulgaris L.) to drought which affects the quality and quantity of yield of this important crop. To better understand the role of aquaporins in common bean, drought-induced gene expression of several aquaporins was determined in two cultivars, the more drought tolerant Tiber and the less tolerant Starozagorski ?ern. The two bean cultivars were selected among 16 European genotypes based on the tolerance to drought determined by time needed for plants to wilt after withholding irrigation and yield at harvest. The expression patterns of two plasma membrane intrinsic proteins, PvPIP1;2 and PvPIP2;7, and two tonoplast intrinsic proteins, PvTIP1;1 and PvTIP4;1 in leaves of 21 day old plants were determined by RT-qPCR in both cultivars under three degrees of drought stress, and under rehydration and control conditions. Gene expression of all four examined aquaporins was down-regulated in drought stressed plants. After rehydration it returned to the level of control plants or was even higher. The responses of PvPIP2;7 and PvTIP1;1 during drought and rehydration were particularly pronounced. The gene expression of PvPIP2;7 and PvTIP4;1 during drought was cultivar specific, with greater down-regulation of these two aquaporins in drought tolerant Tiber. Under drought stress the relative water content and water potential of leaves were higher in Tiber than in Starozagorski plants. The differences in these physiological parameters indicate greater prevention of water loss in Tiber during drought, which may be associated with rapid and adequate down-regulation of aquaporins. These results suggest that the ability of plants to conserve water during drought stress involves timely and sufficient down-regulation of gene expression of specific aquaporins.

Crested wheatgrass (Agropyron cristatum L.) is a cool-season perennial grass, which has demonstrated its potential for use as turfgrass. However, limited information is available on its drought and salinity tolerance. The main purpose of this study was to investigate the changes in the antioxidant defence system and physiological traits of six Iranian crested wheatgrass genotypes under drought and salinity stresses. The experimental design comprised a split plot with water treatments (control well-watered, salinity stress and water stress) as main plots and genotypes as subplots. This study demonstrated the variations in drought and salinity tolerance among crested wheatgrass genotypes. ‘ACSKI’, ‘ACAMI’ and ‘ACDTI’ generally performed better than other genotypes under drought and salinity conditions, mainly by maintaining higher activities of antioxidant enzymes like superoxide dismutase, peroxidase, catalase, ascorbate peroxidase and non-enzyme antioxidants like glutathione, higher proline and total non-structural carbohydrates content. The increased, decreased, and unchanged activities of antioxidant enzymes in the crested wheatgrass genotypes indicates a different forms of metabolism of antioxidant enzymes in response to drought and salinity stress. In general, drought and salinity stress increases the malondialdehyde (MDA) content and hydrogen peroxide (H2O2) content; however, ‘ACSKI’, ‘ACAMI’ and ‘ACDTI’ genotypes could tolerate an increase in MDA content and H2O2 content; therefore, lower levels of MDA content and H2O2 content were observed. The results showed that increasing levels of diamine oxidase and polyamine oxidase have been associated with increasing drought and salinity tolerance. Based on the biochemical and physiological parameters that were evaluated, we concluded that the genotype ‘ACSKI’ was superior in terms of drought and salinity tolerance. This superiority was mainly a result of better enzymatic and non-enzymatic antioxidant defence system and better osmotic adjustment under stress conditions.

In order to investigate the effects of drought stress on germination components of barley cultivars, a laboratory experiment was conducted in a factorial randomized complete design with four replications. The controlled experiment included ten of Egyptian barley cultivars namely; (Giza 123, 124, 125, 126, 127, 129, 130, 134, 135 and 2000) as first factor. The second factor included 4 levels of drought stress inducer by applying 0, 5, 10 and 20% of polyethylene glycol-6000 (PEG) which is equivalent to four osmotic potential levels including ?0.001, ?0.27, ?0.54 and ?1.09 MPa, respectively. The results showed that, the highest reduction was related to the drought level of 20% PEG among the barley cultivars. The best cultivars in terms of germination traits were Giza 134, Giza 127, and Giza 126 this indicate their tolerance to drought stress and Giza 130, 135, 2000 cultivars was moderately tolerance and remaining is less tolerance. The protein band 27 kDa and 78 kDa showed high intensity after stress in almost all cultivars. Those two protein bands their exciting was very clear in treated barley leaf tissue. It could be related to dehydrine and oxygen evolving enhancer protein 2 (OEE2) which involved in drought stress tolerance response. Cultivars Giza 127, 130 and 134 showed highest tolerance response under drought stress. The antioxidant enzymes PAGE pattern of Peroxidase (POX), Sodium dismutase (SOD) and Ascorbate peroxidase (APX) for Barley cultivars under drought stress revealed a high activities for Giza 126, 127, 134, 136 and 2000 under ?0.5 MPa osmotic stress by PEG in most of their isoforms. Based on similarity coefficient values the highest values were 1.0 with 100% similarly between tolerant cultivars Giza 130 and Giza 127. Similarly between the susceptible cultivars 125 and Giza 129 was 60%.These data confirmed by the growth parameters which we ranked as tolerant to drought stress.

Salicylic acid (SA) controls growth and stress responses in plants. It also induces drought tolerance in plants. In this paper, four wheat (Triticum aestivum L.) cultivars with different drought responses were treated with SA in three levels of drain (90, 60, 30% of maximum field capacity) to examine its interactive effects on drought responses and contents of osmotic solutes that may be involved in growth and osmotic adjustment. Under drought condition, the cultivars Geza 164 and Sakha 69 had the plant biomass and leaf relative water content (LRWC) greater than the cultivars Gemaza 1 and Gemaza 3. In all cultivars, drought stress decreased the biomass, LRWC, and the contents of inorganic solutes (Ca, K, Mg) and largely increased the contents of organic solutes (soluble sugars and proline). By contrast, SA increased the biomass, LRWC and the inorganic and organic solute contents, except proline. Correlation analysis revealed that the LRWC correlated positively with the inorganic solute contents but negatively with proline in all cultivars. SA caused maximum accumulations of soluble sugars in roots under drought. These results indicated that SA-enhanced tolerance might involve solute accumulations but independently of proline biosynthesis. Drought-sensitive cultivars had a trait lowering Ca and K levels especially in shoots. Possible functions of the ions and different traits of cultivars were discussed.

Drought is a polygenically controlled stress and a major agricultural risk that reduces crop productivity and limits the successful insight of land potential throughout the world. This review article has been divided into two parts, i.e., effect of drought stress on physiology of wheat and potential drought mitigation approaches. In the first part, physiological responses of wheat to stress were discussed. Cell membrane stability, relative water content, early maturity, decreased leaf area, small plant size, increased dry weight and root–shoot ratio, and the whole-plant transpiration rate response to enhanced atmospheric vapor pressure deficit are physiological traits associated with drought tolerance in wheat. Reduction of relative water content closes stomata and thereby reduces stomatal conductance. Osmotic adjustment improves drought tolerance by allowing cell enlargement, plant growth, and stomata to stay partially open and by maintaining CO2 fixation under severe water deficit. The wheat plant accumulates several organic and inorganic solutes in its cytosol to lessen its osmotic potential for maintenance of cell turgor. Drought affects photosynthesis negatively by changing the inner structure of chloroplasts, mitochondria, and chlorophyll content and minerals. Destruction of the photosystem II (PSII) oxygen releasing complex and reaction center can disturb production and use of electrons, causing lipid peroxidation of cell membrane through the production of reactive oxygen species. In the second part, drought mitigation approaches were discussed. Seed, drought, bacterial, and hormonal priming are common approaches used to lessen the effects of water deficit. Physiological trait-based breeding, molecular breeding, marker-assisted backcrossing, aerial phenotyping, water budgeting, and resource allocation are modern approaches used to develop drought tolerant wheat cultivars. Wheat genotypes produced as a result of a combination of all these methodologies will increase food security regarding the currently changing climate.

Complex study of the effect of soil drought (72 h) and subsequent rehydration for 24 and 48 h on the activities of antioxidant and osmoprotective systems in the leaves of young plants of winter wheat (Triticum aestivum L.) cvs. Ballada (high productivity) and Beltskaya (low productivity) was carried out. Under drought conditions, the content of water in the leaves of cv. Ballada reduced to a lesser degree than in the leaves of cv. Beltskaya. Drought did not affect the rate of leaf growth in cv. Ballada but retarded leaf growth in cv. Beltskaya. Under drought conditions, the content of ascorbate reduced in cv. Beltskaya but was not changed in cv. Ballada; the content of glutathione increased by 19% in cv. Ballada and by 30% in cv. Beltskaya. Under drought conditions, ascorbate peroxidase activity was not changed in cv. Ballada whereas in cv. Beltskaya there was a tendency to its decrease. Glutathione reductase activity in the leaves of cv. Beltskaya increased stronger than in cv. Ballada. Substantial differences between cultivars in the accumulation of reducing sugars and sucrose under water deficit were observed. In both cultivars, drought induced an active proline accumulation. Observed differences in the cultivar responses to water stress evidently indicate differences in the strategy of their adaptation to drought. Drought did not affect the contents of chlorophyll and MDA in both cultivars. The data obtained allow a suggestion that, under conditions of moderate soil drought, the coordinated system of antioxidant defense and osmotic control functioned sufficiently effective; as a result, oxidative stress was not developed in both cultivars. Young plants of both cultivars differing in their responses to water deficit retained the ability to recover after rehydration.

Grass pea (Lathyrus sativus L.) is a legume crop known from its tolerance to various abiotic stresses, especially drought. In this study, we investigated: (1) the response of grass pea seedlings to osmotic stress generated in vitro by polyethylene glycol (PEG); (2) potential drought acclimatization mechanisms of two polish grass pea cultivars. Grass pea seeds of two cultivars were sown on media containing different PEG concentrations (0, 5.5, 11.0 mM) and cultivated for 14 days in controlled conditions. Plants’ dry matter increased under osmotic stress (regardless of PEG concentration). In turn, the highest dose of PEG caused a reduction in seedling growth in both cultivars. Furthermore, PEG caused the peroxidase activity increase in whole seedlings and catalase (CAT) activity in roots. However, differences between cultivars were noted in: CAT activity in shoots; while phenols and anthocyanin content as well as electrolyte leakage in shoots and roots. In turn, in both tested genotypes, accumulation of proline increased in shoots under osmotic stress. Obtained results indicate that the examined plants, although belonging to the same species, differ in acclimatization processes leading to elevated tolerance to osmotic stress.

Salicylic acid (SA) acts as an endogenous signal molecule responsible for inducing abiotic stress tolerance in plants. In this study, the role of SA in improving drought tolerance in two maize cultivars (Zea mays L.) differing in their tolerance to drought was evaluated. The plants were regularly watered per pot and grown until the grain filling stage (R2) under a rainout shelter. At stage R2, parts of the plants were treated with SA, after which drought stress was applied. Leaf samples were harvested on the 10th and 17th days of the drought. Some antioxidant enzyme activity, such as the superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), glutathione reductase (GR), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), hydrogen peroxide (H2O2) and malondialdehyde (MDA) content, was measured during the drought period. Exogenous SA prevented water loss and delayed leaf rolling in comparison with control leaves in both cultivars. As a consequence of drought stress, lipid peroxidation, measured in terms of malondialdehyde content, was prevented by SA. SA pretreatment induced all antioxidant enzyme activities, and to a greater extent than the control leaves, during drought. SA also caused a reduction in the ascorbate (ASC) and glutathione (GSH) content in two maize cultivars. The H2O2 level was higher in SA pretreated plants than the controls in both cultivars. Pretreatment with SA further enhanced the activities of antioxidant enzymes and the concentrations of non-enzymatic antioxidants in the tolerant cultivar compared with the sensitive cultivar. Results suggested that exogenous SA could help reduce the adverse effects of drought stress and might have a key role in providing tolerance to stress by decreasing water loss and inducing the antioxidant system in plants with leaf rolling, an alternative drought protection mechanism.

The development of drought tolerant wheat cultivars has been slow due to lack of understanding the diagnostic physiological parameters associated with improved productivity under water stress. We evaluated responses to PEG induced osmotic stress under hydroponics in D-genome synthetic derived and bread wheat germplasm with the main aim to unravel and identify some promising attributes having role in stress tolerances. Genotypes used in this study differed in their morpho-physiological and biochemical attributes. Tolerant genotypes exhibited the ability to ameliorate harmful effects of PEG induced osmotic stress through better osmotic adjustment achieved through substantial relative water content (RWC), lowered osmotic potential, relatively stable root length having maximum water extraction capacity, significant increase in osmoprotectant concentration and relatively enhanced antioxidant activities. The results clearly revealed the importance of synthetic derivatives over check cultivars and conventional wheats in terms of osmotic stress responses. Interestingly, synthetic-derived advanced lines with Aegilops tauschii in its parentage including AWL-02, AWL-04 and AWL-07 proved superior over the best rainfed check cultivar (Wa-01). It was concluded that synthetic-derived wheats has great potential to improve a range of stress adaptive traits. It could, therefore, be recommended to be a useful strategy for allowing modern bread wheat to become adapted to a wider range of environments in future climate change scenarios.
