After removal of the embryo from developing seeds of Vicia fabaL. and Pisum sativum L., the ‘empty’ ovules werefilled with a substitute medium (pH 5.5) and the effect of theosmolality of the medium on K+ and Mg2+ release from the seedcoat was examined. After removal of the embryo from developing seeds of Pisum sativum,the ‘empty’ ovules (seed coats without enclosedembryo) were filled with a solution (pH 5.5) containing mannitol(usually 400 mM) to which various salts were added. A solutioncontaining two isotopes ((a) [2H]-sucrose/[–14C]aminoisobutyricacid (AIB) or (b) [3H]valine/[14C]asparagine mixture) was administeredto the plant via the petiole subtending the fruiting node, and[2H]solute and [14C]solute unloading from the seed coat wasmeasured, in pulse-labelling experiments of about 5 h. The presenceof 25 or 50 mM K+ in the ‘empty’ ovule enhancedthe release of sucrose from the seed coat particularly duringthe first hours of the experiment, but the stimulating effectof K+ on the release of labelled solutes derived from aminoacids was much smaller. The presence of 25 mM CaCl2 did notaffect the release of sucrose or amino acids from the seed coat.The effect of K+ on sucrose and amino acid release is explainedas an inhibition of sucrose and amino acid resorption from theseed coat apoplast into seed coat cells, after unloading fromthe seed coat unloading sites. It is suggested that amino acidrelease is much less affected by K+ than sucrose release, becausefar less resorption of amino acids by seed coat parenchyma cellstakes place during amino acid transport into the seed coat cavity. The extent of post-phloem solute transport through the coatsymplasts of developing seeds of Vicia faba L. and Phaseolusvulgaris L. was evaluated. For Vicia seed coats, the membrane-impermeantfluorochrome, CF, moved radially from the chalazal vein to reachthe chlorenchyma and thin-walled parenchyma transfer cell layers.Thereafter, the fluorochrome moved laterally in these two celllayers around the entire circumference of the seed coat. Transferof CF from the chalazal vein was inhibited by plasmolysis ofattached ‘empty’ seed coats. In contrast, the spreadof phloem imported CF was restricted to the ground parenchymaof Phaseolus seed coats. Fluorochrome loaded into the outermostground parenchyma cell layer was rendered immobile followingplasmolysis of excised seed-coat halves. Phloem-imported [14C]sucroseand the slowly membrane permeable sugar, L-[14C]glucose, werepartitioned identically between the vascular and non-vascularregions of intact Vicia seed coats. For 14C-photosynthates,these partitioning patterns in attached ‘empty’Vicia seed coats were unaffected by PCMBS, but inhibited byplasmolysis. Tissue autoradiographs of intact Phaseolus seedcoats demonstrated that a pulse of 14C-photosynthate moved fromthe veins to the grounds tissues. In excised Vicia seed coats,preloaded with 14C-photosynthates, the cellular distributionof residual 14C-label was unaffected by PCMBS. In contrast,PCMBS caused the 14C-photosynthate levels to be elevated inthe veins and ground parenchyma relative to the branch parenchymaof Phaseolusseed coat halves. Based on the above findings, itis concluded that the phloem of Vicia seed coats is interconnectedto two major symplastic domains; one comprises the chlorenchyma,the other the thin-walled parenchyma plus thin-walled parenchymatransfer cells. For Phaseolusseed coats, the phloem forms amajor symplastic domain with the ground parenchyma. After removal of the embryo from developing ovules of Viciafaba L. and Pisum sativum L., seed-coat exudates were collectedand the amino acid fraction of the exudate was analyzed. InV. faba, alanine was the most important compound of the aminoacid fraction. In P. sativum, alanine and glutamine were thetwo most important components, whereas only small amounts ofasparagine were present. Comparison with published data suggeststhat seed-coat exudates may differ from phloem sap in the relativeimportance of these amino acids. Abstract After removal of the embryo from developing seeds of Vicia faba L. and Pisum sativum L., the ‘empty’ ovules were filled with a substitute medium (pH 5.5) and the effect of the osmolality of this solution on assimilate transport was exandned. In pulse-labelling experiments with a mixture of [3H]sucrose and [14C]α-andnoisobutyric acid (AIB), a solute concentration of 400 mol m?3 (100 mol m3? sucrose + 300 mol m?3 mannitol) was too low to maintain sugar and andno acid transport into empty ovules of V. faba in a very early stage of development (embryo dry weight < 100 mg) on the same level as transport into intact ovules within the same fruit. A 550-mol m?3 solution could maintain the normal rate of transport. In experiments with seeds in a more advanced stage of development (embryo dry weight > 250 mg), transport of labelled sucrose and AIB into empty ovules filled with a 400-mol m?3 solution was practically equal to transport into intact ovules within the same fruit. Experiments without isotopes, on sugar and andno acid release from the seed coat, confirmed the important role of the osmotic environment. A very low osmolality of the solution (e.g. 50 mol m?3 mannitol) enhanced net efflux of assimilates from excised seed coats and cotyledons, by inhibiting resorption from the apoplast.  相似文献

Excised seed-coat halves and cotyledons of developing seedsof Pisum sativum L. were incubated in a bathing medium (pH 5·5),in order to measure the release or uptake of sucrose and aminoacids. Net efflux of sucrose and amino acids was reduced bya 250 mol m –3 mannitol solution and a 400 mol m –3solution, in comparison with a 100 mol m–3 control. Thiseffect could not be observed in the case of the amino acid analogue-aminoisobutyric acid (AIB). Net uptake of labelled sucroseor valine by cotyledons and seed coats was enhanced by a highosmolality of the bathing medium. The data on AIB and the datafrom uptake experiments support the view that net efflux ofassimilates is reduced by a high solute concentration in theapoplast (e.g. 400 mol m–3 mannitol), via a stimulationof carrier-mediated sucrose and amino acid uptake into cotyledonaryand seed coat tissues. In experiments with attached empty ovulesof pea in a very early stage of development, sugar release fromthe seed coat was enhanced by a low osmolality of the apoplastsolution (e.g. 100 mol m–3 mannitol, in comparison witha 400 mol m –3 control). This paradoxical effect may beobserved when the stimulatory effect on net assimilate effluxfrom seed coat tissues is exceeding the inhibitory effect onassimilate import into the seed coat. The amino acid composition of the EDTA-induced phloem exudatereaching the fruit and the seed, and of the solutes releasedby the seed coat during fruit development were determined inglasshouse-grown pea (Pisum sativum L. cv. Finale) suppliedeither with nitrate-free nutrients (nodulated plants) or withcomplete medium (non-nodulated plants). The EDTA-promoted exudationtechnique was used supposedly to collect phloem sap and theempty seed technique supposedly to collect the solutes secretedby the seed coat to the embryo sac cavity. In young seeds embryosac liquid was sampled directly from the embryo sac. The maincarbohydrate transported and secreted was sucrose. The mainamino acids reaching the fruit were asparagine, glutamine, andhomoserine. Their proportions were steady during a day-nightcycle and throughout fruit development. Amino acid compositionchanges occurred first in the pathway from fruit stalk to seedfunicle, due to the formation of threonine (probably from homoserine)and in the seed coat due to production of glutamine, alanineand valine which, together with threonine were the main secretedamino acids. The temporary nitrogen reserves of the pod walland seed coat were remobilized as asparagine during senescence.Phloem exudate of nodulated plants showed a higher (about twice)proportion of asparagine but lower proportions of homoserineand glutamine than in EDTA-induced phloem exudate of nitrate-fedplants. The two types of nitrogen nutrition also produced somechanges in relative proportions of threonine and homoserinesecreted by the seed coat. Mechanism of Photosynthate Efflux from Vicia faba L. Seed Coats   总被引:2,自引:0,他引:2  
In order to develop a tentative model of the mechanism of photosynthateefflux from the vascular region of Vicia faba L. seed coats,wash-out experiments were performed after removal of the embryo. The sulphydryl group modifiers, pCMBS and NEM, reduced 14C-photosynthateefflux by 40% and 50%, respectively. Their inhibitory effectcould be prevented or reduced (in the latter case) by includingDTT in the bathing solution. Maltose competed with sucrose forefflux; a concentration of 300 mol m–3 inhibited 14C-photosynthaterelease by 35%. The cations K+ , Na+ Mg2+ and TPP+ enhancedefflux significantly, whereas the countenon Cl had noeffect. The presence of the protonophore CCCP (0·1 molm–3) led to a reduction of efflux by 50% net proton extrusiondropped by 34%. To a lesser extent, an efflux inhibition wasalso achieved by decreasing the cytoplasmic pH with the weakacid DM0. In contrast, alterations in the external pH causedonly a feeble response. The ATPase inhibitor, EB, decreasedphotosynthate efflux and H+ extrusion. DES reduced efflux slightly,presumably by affecting ATPase activity as well as energy metabolism. Based on these findings, it is proposed that a sucrose/protonantiport mechanism could be responsible for photosynthate effluxfrom Vicia faba seed coats. The effect of p -chloromercuribenzenesulfonic acid (PCMBS), carbonylcyanide- m -chlorophenylhydrazone (CCCP) and a high apoplastic pH (pH 7.5 compared with pH 5.5) on the release of sugars (sucrose and glucose) and amino acids from attached and detached seed coats of Pisum sativum L. cv. Marzia into a bathing solution was measured by means of the 'empty seed coat technique'. PCMBS reduced the release of sugars and amino acids from attached as well as from detached seed coats, suggesting that carrier-mediated transport might be involved. CCCP reduced sugar release from attached seed coats while amino acid release was hardly affected. In experiments with detached seed coats CCCP had no effect on release of either sugar or amino acids, suggesting that it is not energy-dependent. Raising the pH of the bathing solution from pH 5.5 to pH 7.5 slightly increased sugar release from both attached and detached seed coats while amino acid release was not affected. This might indicate a role of the apoplastic pH in regulating sugar release from the seed coat via a retrieval mechanism. The presented data indicate that there are important differences between sugars and amino acids with respect to transport processes in the seed coat. This is supported by the observation that the rate of amino acid release from the seed coat was higher than the rate of sugar release. The release data of detached seed coats were subjected to compartmental analysis in order to calculate rate constants for release from cell compartments. In the case of sugars, the half-times for emptying the cytoplasmic and vacuolar compartment were 0.8 h and 12.5 h. respectively. For amino acids the half-times were 0.5 h for emptying the cytoplasmic and 3.8 h for emptying the vacuolar compartment.  相似文献

Seed coat development was studied on two nearisogenic linesof peas (Pisum sativum L.): RbRb (wild type, round seed) andrbrb (wrinkled seed). A mutation at the rb locus modifies thedry seed shape and reduces the starch content of the embryo.This mutation is now known to affect the activity of ADPGlucosepyrophosphorylase, a key enzyme in the starch biosynthetic pathway.We have investigated the effects of the rb mutation on seedcoat development and found that the mutation reduces the growthrate and starch content in this organ. However, experimentson the kinetics of 14C-sucrose loading showed that starch synthesisfrom unloaded sucrose occurred in the seed coat for both mutantand wild-type lines. In addition, the sucrose concentrationwas increased and amino acid concentration decreased such thatthe nutritional balance of the embryos was affected. However,osmolality of the seed coat cells was not affected, suggestinga regulatory process which allows the maintenance of the importof assimilates in the seeds of either line. A study has been made of the changes during development in thetotal nitrogen, uncombined amino acid, starch and ethanol-solublesugar contents of field bean cotyledons from two large-seededcultivars, Minica and Felix, and two lines of the small-seededcultivar Dacre, which differed in protein concentration of themature seed. In all cultivars the concentration of total nitrogen,uncombined amino acids and ethanol-soluble sugars decreasedwhile that of starch increased during development. However,the pattern of decrease in the proportion of total nitrogenvaried between the large and the small-seeded cultivars. Fromday 40 (after pollination) onwards a higher percentage of ethanol-solublesugars but a lower percentage of starch was found in the large-seededcultivars. Throughout development the Dacre line with the ‘high’protein concentration in the mature seed maintained a higherpercentage of total nitrogen and a correspondingly lower percentageof ethanol-soluble sugars and starch than the ‘low’protein line. A cytochemical study of naphthol AS-D esterases in vegetativeshoot apices of Pisum sativum and Vicia faba L. has shown thepresence of carboxyl esterases (E.C. 3.1.1.1 [EC] .) in those meristemcells already committed to form vascular elements. Experiments were undertaken with embryo-less ovules of Pisumsativum to study the influence of apoplastic osmolality on seedcoat import and seed coat unloading.11CO2 pulse labelling alongwith collimated monitoring of plant tissues were used with attachedovules to measure continuously and simultaneously total podimport, import into a modified ovule and photo-assimilate washoutfrom the seed coat of the ovule into a flow-through bathingsolution.Our results indicated that seed coat import was immediatelyaffected by a change in the applied bathing solution osmolality,with a decrease in osmolality lowering seed coat import andan increase in osmolality increasing import. 11C-photo-assimilatewashout from attached ovules was found to respond in a similarmanner to the apoplastic osmolality. However, the osmotic effecton 11C-washout was a delayed response and it appears that themajority of this observed response was due to the alterationin seed coat tracer import. Further experiments with 14C-labelled,excised seed coat halves (i.e. no further import) supportedthis hypothesis by demonstrating that seed coat unloading (measuredas 14C-photo-assimilate washout) was actually enhanced at alow solution osmolality. PCMBS had no effect on seed coat importor washout in attached, modified ovules, suggesting that photo-assimilateunloading from seed coats of Pisum does not involve a carrierprotein. Studies of the spatial distribution of imported 14Cin Pisum seed coats further suggest that this unloading, intothe apoplast, occurs from non-phloem cell types, and that themovement of photo-assimilates from the sieve elements to theterminal unloading site occurs via symplastic transport. ‘Physiological maturity’, i.e. the time when seedsreach their maximum dry weight during development, occurredwhen maturation drying on the parent plant in the field hadreduced seed moisture content to approximately 60 per cent infaba bean (Vicia faba L.), lentil (Lens culinaris Medic.), chickpea(Cicer arietinum L.), white lupin (Lupinus albus L.), soya bean(Glycine max [L.] Merr.) and pea (Pisum sativum L.) The onsetof desiccation-tolerance, i.e. the ability of seeds to germinatefollowing harvest and rapid artificial drying, coincided withphysiological maturity, except in pea where it occurred a littleearlier at about 70 per cent moisture content. Maximum seedquality as determined by maximum viability, minimum seedlingabnormalities and maximum seedling size occurred in pea, chickpeaand lupin when seeds were harvested for rapid drying at physiologicalmaturity; but for maximum seed quality in the other speciesmaturation drying had to proceed further - to about 45 per centmoisture content in soya bean and to about 30 per cent moisturecontent in lentil and faba bean seed crops. Much of this variationamongst the six species, however, was due to differences inthe variation in maturity within each seed crop. Results forindividual pods showed that peak maturity, i.e. maximum seedquality following harvest and rapid artificial drying, was achievedin all six species once maturation drying had reduced the moisturecontent of the seeds to 45–50 per cent. In pea, faba beanand soya bean there was a substantial decline in viability andan increase in seedling abnormalities when harvest was delayedbeyond the optimal moisture content for harvest.  相似文献

Interpretation of tracer washout from an attached empty seedcoat depends on whether photoassimilate within the apoplastof the seed coat is absorbed by the seed coat tissues. Usingsucrose trapping procedures, we were unable to see any evidencefor sucrose uptake from the seed coat apoplast which would beneeded to provide the seed coat with its carbohydrate requirementsif phloem unloading were into the apoplast. Once released intothe apoplast photoassimilate is unavailable to the seed coattissue. Changes between equimolar solutions of sorbitol andsorbitol/sucrose mixes induced small transient responses inseed coat unloading which suggest that sorbitol and sucrosehad different reflection coefficients and gave water relationresponses with rapid, and fatiguable, osmoregulation withinthe seed coat. Immediate inhibition of seed coat unloading with PCMBS is reported,followed by inhibition of import into the entire pod. PCMBSappears to be xylem mobile, thereby quickly being dispersedthroughout the entire experimental pod. A complex CCCP responseis reported, which is consistent with immediate inhibition ofsymplastic transport followed by membrane disruption. AlthoughCCCP inhibited seed coat unloading, there was no effect on ovuleimport. The process of sugar and amino acid release by the seed coat of Pisum sativum L. cv. Marzia was studied. Prior to measuring the release of solutes by the seed coat of developing ovules, the embryo was removed from each ovule studied. After this surgical treatment, each "empty" seed coat was filled with the appropriate solution (pH 5.5) with or without inhibitor. Both KCN and p-chloromercuribenzenesulfonic acid (PCMBS) strongly inhibited the release of sucrose and p -aminoisobutyric acid (AIB) by the seed-coat. These data support the view that phloem unloading is an energy-dependent process sensitive to the sulfhydryl group modifier PCMBS. In pulse-labelling experiments, addition of high concentrations of unlabelled sucrose (200 m M ) and AIB (25 m M ) to the solution filling the seed coat cavity did not diminish the release of labelled solutes by the unloading sites of the seed coat. This observation presents evidence against the view that phloem unloading into a strong sink is related to low sugar concentrations in the apoplast.  相似文献

In developing seed ofVicia faba L., solutes imported throughthe phloem of the coats move symplastically from the sieve elementsto a specialized set of cells (the thin-walled parenchyma transfercells) for release to the seed apoplast. Potassium (K+) is thepredominant cation released from the seed coats. To elucidatethe mechanisms of K+ efflux from seed coat to seed apoplast,whole-cell currents across the plasma membranes of protoplastsof thin-walled parenchyma transfer cells were measured usingthe whole-cell patch-clamp technique. Membrane depolarizationelicited a time-dependent and an instantaneous outward current.The reversal potential (ER of the time-dependent outward currentwas close to the potassium equilibrium potential (EK and itshifted in the same direction as EK upon changing the externalK+ concentration, indicating that this current was largely carriedby an efflux of K+. The activation of the time-dependent outwardK+ current could be well fitted by two exponential componentsplus a constant. The instantaneous outward current could alsobe carried by K+ efflux as suggested by ion substitution experiments.These K+ outward rectifier currents elicited by membrane depolarizationare probably too small to represent the mechanism for the normalK+ efflux from seed coat cells. Membrane hyperpolarization morenegative than –80 mV activated a time-dependent inwardcurrent. K+ influx was responsible for the inward current asthe current reversed at membrane voltage close to EK and shiftedin the same direction as EK when external [K+] was varied. Activationof this K+inward rectifier current was well fitted with twoexponential components plus a constant. A regulating functionfor this current is suggested. Lateral root primordium development has been examined in primaryroots of Vicia faba L., Pisum sativum L., Zea mays L. and Phaseolusvulgaris L. Following their initiation from an estimated minimumnumber of 77–162, 20–57, 17 and 12 cells respectivelyin Vicia, Phaseolus, Pisum and Zea, the primordia rapidly increasedin cell number to emerge as secondary roots about 2.8–3.6days later depending on the species being examined. Cell doublingtimes were estimated directly from cell numbers at differenttimes following primordium inception and were found to increasewith increase in primordium size in each of the species investigated. The number of primordia formed per cm of root growth per daywas greatest in Zea and least in Pisum. A comparison of thedata obtained for Vicia with that in the literature led to theconclusion that although the number of primordia produced percm of root growth was independent of the rate of primary elongation,the number produced per day increased in a linear fashion withincrease in the rate at which the primary lengthened. Species that showed marked morphological and physiological responsesby their roots to Fe-deficiency (Strategy I plants) were comparedwith others that do not exhibit these responses (Strategy IIplants). Roots from Fe-deficient cucumber (Cucumis sativusL.‘Ashley’), tomato (Lycopersicon esculentumMill.T3238FER) and pea (Pisum sativumL. ‘Sparkle’) plantsproduced more ethylene than those of Fe-sufficient plants. Thehigher production of ethylene in Fe-deficient cucumber and peaplants occurred before Fe-deficient plants showed chlorosissymptoms and was parallel to the occurrence of Fe-deficiencystress responses. The addition of either the ethylene precursorACC, 1-aminocyclopropane-1-carboxylic acid, or the ethylenereleasing substance, Ethephon, to several Fe-sufficient StrategyI plants [cucumber, tomato, pea, sugar beet (Beta vulgarisL.),Arabidopsis(Arabidopsis thaliana(L.) Heynh ‘Columbia’), plantago(Plantago lanceolataL.)] promoted some of their Fe-deficiencystress responses: enhanced root ferric-reducing capacity andswollen root tips. By contrast, Fe-deficient roots from severalStrategy II plants [maize (Zea maysL. ‘Funo’), wheat(Triticum aestivumL. ‘Yécora’), barley (HordeumvulgareL. ‘Barbarrosa’)] did not produce more ethylenethan the Fe-sufficient ones. Furthermore, ACC had no effecton the reducing capacity of these Strategy II plants and, exceptin barley, did not promote swelling of root tips.
