The influence of silicon on the growth of maize plants cultivated in hydroponics in the presence of cadmium (5 μM) was investigated. Four different treatments were used: Control (C), Cadmium (Cd), Silicon (Si) and Cadmium plus Silicon (Cd + Si). The Si concentration was 35 mM. Thirteen-day-old plants were harvested. Growth parameters (length of primary seminal root, leaf area of first and second fully developed leaves, fresh and dry weight of below- and above-ground parts of the plants), and Cd concentration and total amount of Cd in the below- and above-ground parts were determined. In roots, the development of the endodermal barrier was observed by fluorescent staining with Fluorol yellow 088.Inhibitory effects of Cd on plant growth were observed. Silicon treatment in the absence of Cd had positive effects on most of observed growth parameters compared with the control. Moreover, Si in the Cd + Si treatment improved all growth parameters compared with the cadmium treatment. Silicon increased the cell-wall extensibility both in Si and Cd + Si treatments when compared with the control. Alleviation of the Cd-inhibitory effect on maize plants by Si was not due to exclusion of Cd from the plant; in contrast, Cd concentration in below- and above-ground plant parts and the total amount of Cd per plant were significantly higher in the Cd + Si plants than in the Cd treatment. The increased Cd content in Cd + Si plants was correlated with the development of the endodermis; during the second stage of endodermal development, suberin lamellae were formed at a greater distance from the root apex in the Cd + Si than in the Cd treatment. Silicon itself did not influence the development of suberin lamellae in the maize roots compared with the control.  相似文献

A long-term experiment was conducted to investigate the alleviative effects of silicon (Si) on cadmium (Cd) toxicity in garlic plants grown in pots. Cd and Si were introduced into soil before sowing. Cd was added at a rate of 20 mg kg?1 soil, and Si was applied at two rates: 50 mg SiO2 kg?1 (Si1) and 500 mg SiO2 kg?1 (Si2). There were totally six treatments consisting of CT (control, no added Cd or Si), Si1, Si2, Cd, Cd + Si1, and Cd + Si2. The results showed that Si addition did not affect the growth of garlic plants under control conditions. Under Cd stress, the plant growth and PSII quantum efficiency were inhibited, and they were significantly improved in the presence of added Si. Added Si at Si1 level did not change the soil pH and Cd availability, while it increased Cd accumulation in both shoot and bulb, and improved Cd tolerance. Si added at Si2 level increased the soil pH and decreased Cd availability, and decreased Cd accumulation in different parts of the plant. Added Si had no effect on the activities of soil catalase, urease or invertase regardless of Cd presence. The results suggest that Si could increase Cd tolerance of garlic plants, and the tolerance increase was attributed to not only decreased Cd availability but also in planta detoxification mechanism. There is no evidence indicating that Si-mediated increase of Cd tolerance is related to improved soil microorganism environment as observed in biotic stress conditions.  相似文献

Background and Aims

Silicon (Si) has been shown to ameliorate the negative influence of cadmium (Cd) on plant growth and development. However, the mechanism of this phenomenon is not fully understood. Here we describe the effect of Si on growth, and uptake and subcellular distribution of Cd in maize plants in relation to the development of root tissues.

Methods

Young maize plants (Zea mays) were cultivated for 10 d hydroponically with 5 or 50 µm Cd and/or 5 mm Si. Growth parameters and the concentrations of Cd and Si were determined in root and shoot by atomic absorption spectrometry or inductively coupled plasma mass spectroscopy. The development of apoplasmic barriers (Casparian bands and suberin lamellae) and vascular tissues in roots were analysed, and the influence of Si on apoplasmic and symplasmic distribution of 109Cd applied at 34 nm was investigated between root and shoot.

Key Results

Si stimulated the growth of young maize plants exposed to Cd and influenced the development of Casparian bands and suberin lamellae as well as vascular tissues in root. Si did not affect the distribution of apoplasmic and symplasmic Cd in maize roots, but considerably decreased symplasmic and increased apoplasmic concentration of Cd in maize shoots.

Conclusions

Differences in Cd uptake of roots and shoots are probably related to the development of apoplasmic barriers and maturation of vascular tissues in roots. Alleviation of Cd toxicity by Si might be attributed to enhanced binding of Cd to the apoplasmic fraction in maize shoots.  相似文献

Silicon (Si) frequently accumulates in plants tissues, mainly in roots of dicotyledons, such as cowpea. By contrast, Cadmium (Cd) is a metal that is extremely toxic to plant metabolism. This research aims to investigate if the deposition of Si in root can reduce Cd contents and minimize its negative effects on leaves, measuring gas exchange, chlorophyll fluorescence, antioxidant metabolism, photosynthetic pigments and growth, which may explain the possible role of Si in the attenuation of Cd toxicity in cowpea. This study had a factorial design, with all factors completely randomized and two Cd concentrations (0 and 500 µM Cd, termed as – Cd and + Cd, respectively) and three Si concentrations (0, 1.25 and 2.50 mM Si). Si reduced Cd contents in the roots and in other plant organs, such as stems and leaves. The Si contents were highest in roots, followed by stems and leaves, which was explained by the passive absorption of Si. The application of Si promoted increase in both the macro- and micronutrient contents in all tissues, suggesting that Si mitigates the effect of Cd on nutrient uptake. Si attenuated Cd-mediated effects on light absorption of photosystem II (PSII), increasing the effective quantum yield of PSII photochemistry and the electron transport rate. Additionally, toxic effects induced by Cd on gas exchange were mitigated by the action of Si. Plants treated with Cd + Si showed increase in the activities of antioxidant enzymes and reductions in oxidant compounds; these modifications were promoted by Si via detoxification mechanisms. Increases in the photosynthetic pigments and growth of plants treated with Si and exposed to Cd stress were detected and were due to the reduced deterioration of cell membranes and maintenance of chloroplasts, which had positive repercussions on growth and development. This study validated the hypothesis that the accumulation of Si in roots induces benefits on metabolism and alleviates the toxic effects caused by Cd in leaves of cowpea.  相似文献

The effects of Cd, in combination with salicylic acid (SA) and sodium nitroprusside (SNP), on ryegrass seedlings were studied. Exposure of plants to 0.1 mM CdCl2 for 2 weeks resulted in toxicity symptoms such as chlorosis and necrotic spots on leaves. The addition of 0.2 mM SA or 0.1 mM SNP slightly alleviated the toxic effects of Cd. After application of both SA and SNP, these symptoms significantly decreased. Treatment with Cd resulted in a decrease of dry weight of roots and shoots, chlorophyll content, net photosynthetic rate (P n), transpiration rate (T r), and the uptake and translocation of mineral elements. In Cd-treated plants, levels of lipoxygenase activity and malondialdehyde, hydrogen peroxide (H2O2), and proline contents significantly increased, whereas the activities of antioxidant enzymes, such as superoxide dismutase, guaiacol peroxidase, catalase, and ascorbate peroxidase, decreased in both roots and shoots. The results indicated that Cd caused physiological stresses in ryegrass plants. The Cd-stressed plants exposed to SA or SNP, especially to SA + SNP, exhibited improved growth compared with Cd-stressed plants. Application of SA or SNP, especially the combination SA + SNP, considerably reduced root-to-shoot translocation of Cd and increased the activities of antioxidant enzymes in both roots and shoots of Cd-stressed plants. The interaction of SA and SNP increased chlorophyll content, P n and T r in leaves, and the uptake and translocation of mineral elements, and decreased lipid peroxidation and H2O2 and proline accumulation in roots and shoots. These results suggest that SA or SNP, and, in particular, their combination counteracted the negative effects of Cd on ryegrass plants.  相似文献

We have investigated the influence of silicon on higher zinc concentration reducing the growth of aboveground parts by ca 50 % in young maize plants (hybrid Novania) grown in hydroponics. Eight different treatments were used: control, Zn (800 μM ZnSO4·7H2O), Si1/Si2.5/Si5 (1/2.5/5 mM Na2SiO7) and Zn+Si (combination of zinc and all silicon concentrations). The concentration of Zn and Si and their distribution in plants was determined. The growth parameters (length of primary seminal root, leaf area of first and second leaves, fresh and dry weight of below- and above-ground plant parts) of plants grown in various Zn+Si treatments were significantly decreased in comparison to all other treatments. Increasing concentration of Si in combination with Zn treatment and selected hybrid (Novania) resulted in increased physiological stress in comparison to Zn treatment. However, roots and shoots of all Zn+Si treated plants contained significantly lower amount of Zn than Zn treatment. The Si concentration in roots was the same in Si and Zn+Si plants. In general, higher amount of Si was observed in shoots than in roots of Si1- and Si2.5-treated plants and opposite was observed in Si5-treated plants. In spite of significantly decreased root and shoot accumulation of Zn in the presence of Si, no positive effect of Si on Zn toxicity in young maize plants under experimental conditions used in this work and used maize hybrid was observed.  相似文献

Potassium (K+) plays important roles in the development of plants and the response to various environmental stresses. However, the involvement of potassium in alleviating heavy metal stress in tobacco remains elusive. Greenhouse hydroponic experiments were conducted to evaluate the alleviating effects of K+ on tobacco subjected to cadmium (Cd) toxicity using four different K+ levels. Dose-dependent increases of plant biomass were found in both 0-μM Cd and 5-μM Cd treatments under different K+ levels, with the exception of the 1-mM KHCO3 (K3) treatment. The best mitigation effect was recorded with the 0.5-mM K+ (K2) treatment, which greatly alleviated Cd-induced growth inhibition, photosynthesis reduction, and oxidative stress. Compared with K0 treatment (no KHCO3 addition), K2 treatment significantly reduced Cd uptake and translocation after 5 and 10 days of Cd treatment. Moreover, the net photosynthetic rate, intracellular CO2 concentration, stomatal conductance, and transpiration rate as well as K+, zinc, manganese, copper, and iron concentrations in both shoots and roots after 10 days of Cd treatment significantly improved under the K2 treatment, and malondialdehyde accumulation in both shoots and roots was repressed, compared with K0 + Cd. Superoxide dismutase was found to play key roles in alleviating Cd-induced oxidative pressure in shoots of plants in K2 treatment under Cd treatment. Our findings advocate a positive role for K+ in reducing pollutant residues for safe production, especially in soils slightly or moderately polluted with Cd.  相似文献

A hydroponic experiment was carried out to study the physiological mechanisms of N-acetyl cysteine (NAC) in mitigating cadmium (Cd) toxicity in two barley (Hordeum vulgare L.) genotypes, Dong 17 (Cd-sensitive) and Weisuobuzhi (Cd-tolerant). Addition of 200 μM NAC to a culture medium containing 5 μM Cd (Cd + NAC) markedly alleviated Cd-induced growth inhibition and toxicity, maintained root cell viability, and dramatically depressed O 2 ·? and ·OH, and malondialdehyde accumulation, significantly reduced Cd concentration in leaves and roots, especially in the sensitive genotype Dong 17. External NAC counteracted Cd-induced alterations of certain antioxidant enzymes, e.g., brought root superoxide dismutase and glutathione reductase, leaf/root peroxidase and glutathione peroxidase activities of the both genotypes down towards the control level, but elevated Cd-stress-depressed leaf catalase in Dong 17 and root ascorbate peroxidase activities in both genotypes. NAC counteracted Cd-induced alterations in amino acids and microelement contents. Furthermore, NAC significantly reduced Cd-induced damage to leaf/root ultrastructure, e.g. the shape of chloroplasts in plants treated with Cd + NAC was relatively normal with well-structured thylakoid membranes and parallel pattern of lamellae but less osmiophilic plastoglobuli compared with Cd alone treatment; nuclei of root cells were better formed and chromatin distributed more uniformly in both genotypes. These results suggested that under Cd stress, NAC may protects barley seedlings against Cd-induced damage by directly and indirectly scavenging reactive oxygen species and by maintaining stability and integrity of the subcellular structure.  相似文献

Impatiens walleriana plants accumulate sufficiently high concentrations of cadmium (Cd) for this species to be considered a potential Cd hyperaccumulator. Rooted cuttings were grown hydroponically for 25 and 50 days in solutions spiked with various Cd concentrations. The subcellular distribution and chemical forms of Cd in different organs were analyzed, and its upward translocation was also assessed. The plants accumulated large amounts of Cd; the Cd concentration in the roots and shoots reached 120–1900 and 60–1600 mg/kg, respectively. Regardless of the growth period, the Cd accumulated in the roots was primarily compartmentalized in the soluble fraction or ethanol and deionized water extractable chemical forms with high migration abilities. Translocation to the shoots was followed by an association of Cd mainly in the cell wall or with pectate and protein. The roots’ Cd showed a high migration capacity for predicting the shoots’ Cd concentrations. Different exposure periods significantly affected the subcellular distribution of Cd in the stems, and thus the upward translocation.  相似文献

Endodermal cell walls of the three dicotyledoneous species Pisum sativum L., Cicer arietinum L. and Ricinus communis L. were isolated enzymatically and analysed for the occurrence of the biopolymers lignin and suberin. From P. sativum, endodermal cell walls in their primary state of development (Casparian strips) were isolated. Related to the dry weight, these isolates contained equal amounts of suberin (2·5%) and lignin (2·7%). In contrast, the endodermal cell walls of C. arietinum and R. communis, which were nearly exclusively in their secondary state of development, contained significantly higher proportions of suberin (10–20%) and only traces of lignin (1–2%). The results of the chemical analyses were supported by a microscopic investigation of Sudan III-stained root cross-sections, showing a Casparian strip restricted to the radial walls of the endodermis of P. sativum and well-pronounced red suberin lamellae in C. arietinum and R. communis roots. Compared with recently investigated monocotyledoneous species, higher amounts of suberin by one order of magnitude were detected with the secondary state of development of dicotyledoneous species. Furthermore, the carbohydrate and protein contents of primary (Clivia miniata Reg. and Monstera deliciosa Liebm.), secondary (C. arietinum and R. communis) and tertiary endodermal cell walls (Allium cepa L. and Iris germanica L.) were determined. The relative carbohydrate content of secondary endodermal cell walls was low (14–20%) compared with the content of primary (42–50%) and tertiary endodermal cell walls (60%), whereas the protein content of isolated endodermal cell walls was high in primary (13%) and secondary (8%) and low in tertiary endodermal cell walls (0·9–2%). The results presented here indicate that the quantitative chemical composition of primary, secondary, and tertiary endodermal cell walls varies significantly. Finally, cell wall proteins are described as an additional important constituent of endodermal cell walls, with the highest concentrations occurring in primary (Casparian strips) and secondary endodermal cell walls.  相似文献

运用差速离心法和化学试剂逐步提取法,分析了Cd和Zn在皖景天根、茎和叶的亚细胞分布及其化学形态.结果表明:10 μmol·L-1 Cd处理下,Cd在皖景天细胞内的主要分布位点是其可溶部分;在100 μmol·L-1 Cd处理下,Cd在根中主要分布在细胞壁、茎中主要分布在细胞壁和可溶部分、叶中超过90%的Cd分布在可溶部分.高Cd浓度处理时,皖景天根、茎和叶的细胞壁中Cd分布比例增加,而可溶部分Cd分布比例相对减少.在1和800 μmol·L-1 Zn处理条件下,Zn在皖景天根、茎和叶的主要分布位点是可溶部分;高Zn浓度处理时,皖景天叶、茎和根的可溶部分和细胞壁中Zn的分布比例无明显变化.细胞器中Zn和Cd分布都很少.Cd在皖景天根、茎和叶内主要以氯化钠提取态和水提取态存在,Zn在皖景天根、茎和叶内以多种化学形态存在.  相似文献

The aim of this study was to investigate the effects of silicon in alleviating cadmium stress in maize plants grown in a nutrient solution and to evaluate the potential of the spectral emission parameters and the ratio of red fluorescence (Fr) to far-red fluorescence (Ffr) in assessing the beneficial effects of Si. An experiment was carried out using a nutrient solution with a toxic dose of Cd and six doses of Si; biomass, Cd, Si, and photosynthetic pigments of the plants were measured. Chlorophyll (Chl) a fluorescence analysis demonstrated that Si alleviated Cd toxicity in plants. Chl fluorescence measurements were sensitive in detecting such effects even when significant changes in biomass production and concentrations of photosynthetic pigments were not observed. The spectral emission and the Fr/Ffr ratio were sensitive to the effects of Si. Silicon caused a reduction in the translocation of Cd to the shoots of maize plants.  相似文献

Ultrastructure and development of apoplastic barriers within indeterminate root nodules formed by Vicia faba L. were examined by light and electron microscopy. The nodule outer cortex is separated from the inner cortex by a heavily suberized nodule endodermis, which matures in submeristematic regions and possesses suberin lamellae. Unsuberized passage cells are present near vascular strands, which are surrounded by a vascular endodermis attached on the inner side of the nodule endodermal cell walls. The vascular endodermis appears immediately below the meristematic apex in developmental state I (Casparian bands), gradually develops suberin lamellae, and attains developmental state II at the base of the nodule. For chemical analysis apoplastic barrier tissues were dissected after enzymatic digestion of non-impregnated tissues. Root epidermal and endodermal cell walls as well as nodule outer cortex could be isolated as pure fractions; nodule endodermal cell walls could not be separated from vascular endodermal cell walls and enclosed xylem vessels. Gas chromatography-flame ionization detection and gas chromatography-mass spectrometry were applied for quantitative and qualitative analysis of suberin and lignin in isolated cell walls of these tissues. The suberin content of isolated endodermal cell walls of nodules was approximately twice that of the root endodermal cell walls. The suberin content of the nodule outer cortex and root epidermal cell walls was less than one-tenth of that of the nodule endodermal cell wall. Substantial amounts of lignin could only be found in the nodule endodermal cell wall fraction. Organic solvent extracts of the isolated tissues revealed long-chain aliphatic acids, steroids, and triterpenoid structures of the lupeol type. Surprisingly, extract from the outer cortex consisted of 89% triterpenoids whereas extracts from all other cell wall isolates contained not more than 16% total triterpenoids. The results of ultrastructural and chemical composition are in good correspondence and underline the important role of the examined tissues as apoplastic barriers.  相似文献

Apoplastic transport barriers in the roots of rice (Oryza sativa L. cv. IR64) and corn (Zea mays L. cv. Helix) were isolated enzymatically. Following chemical degradation (monomerization, derivatization), the amounts of aliphatic and aromatic suberin monomers were analysed quantitatively by gas chromatography and mass spectrometry. In corn, suberin was determined for isolated endodermal (ECW) and rhizo-hypodermal (RHCW) cell walls. In rice, the strong lignification of the central cylinder (CC), did not allow the isolation of endodermal cell walls. Similarly, exodermal walls could not be separated from the rhizodermal and sclerenchyma cell layers. Suberin analyses of ECW and RHCW of rice, thus, refer to either the entire CC or to the entire outer part of the root (OPR), the latter lacking the inner cortical cell layer. In both species, aromatic suberin was mainly composed of coumaric and ferulic acids. Aliphatic suberin monomers released from rice and corn belonged to five substance classes: primary fatty acids, primary alcohols, diacids, omega-hydroxy fatty acids, and 2-hydroxy fatty acids, with omega-hydroxy fatty acids being the most prominent substance class. Qualitative composition of aliphatic suberin of rice was different from that of corn; (i) it was much less diverse, and (ii) besides monomers with chain lengths of C(16), a second maximum of C(28) was evident. In corn, C(24) monomers represented the most prominent class of chain lengths. When suberin quantities were related to surface areas of the respective tissues of interest (hypodermis and/or exodermis and endodermis), exodermal cell walls of rice contained, on average, six-times more aliphatic suberin than those of corn. In endodermal cell walls, amounts were 34 times greater in rice than in corn. Significantly higher amounts of suberin detected in the apoplastic barriers of rice corresponded with a substantially lower root hydraulic conductivity (Lp(r)) compared with corn, when water flow was driven by hydrostatic pressure gradients across the apoplast. As the OPR of rice is highly porous and permeable to water, it is argued that this holds true only for the endodermis. The results imply that some caution is required when discussing the role of suberin in terms of an efficient transport barrier for water. The simple view that only the quantity of suberin present is important, may not hold. A more detailed consideration of both the chemical nature of suberins and of the microstructure of deposits is required, i.e. how suberins impregnate wall pores.  相似文献

Summary Mineral distribution in the roots of wheat (Triticum aestivum L. cv. Wheaton) was investigated using X-ray microanalysis of bulk frozen hydrated roots in SEM and of freeze substituted sections in TEM. Results obtained using the two methods agreed reasonably well. A total often elements were detected: Na, Mg, Si, P, S, Cl, K, Ca, Mn, and Fe. Of these Si, P, Ca, and Mn were incorporated into biomineralized structures. Silica was deposited in the endodermal walls in the older parts of the root. Silicon was also detected in the large central metaxylem lumina in the basal zone of the root, and in the smaller peripheral metaxylem and the immediately contiguous pericycle and outer parenchyma cells bridging the small metaxylem vessels to the endodermal layer. In the basal zone of the root some of the inner cortical cells contained intracellular electron opaque deposits. A municipal solid-waste bottom slag was used to grow maize plants under various abiotic stresses (high pH, high salt and high heavy metal content) and to analyse the structural and chemical adaptations of the cell walls of various root tissues. When compared with roots of control plants, more intensive wall thickenings were detected in the inner tangential wall of the endodermis. In addition, phi thickenings in the rhizodermis in the oldest part of the seminal root were induced when plants were grown in the slag. The role of the phi thickenings may not be a barrier for solutes as an apoplastic dye could freely diffuse through them. The chemical composition of cell walls from endodermis and hypodermis was analysed. Slag-grown plants had higher amounts of lignin in endodermal cell walls when compared to control plants and a higher proportion of H-type lignin in the cell walls of the hypodermis. Finally, the amount of aliphatic suberin in both endo- and hypodermal cell walls was not affected by growing the plants on slag. The role of these changes in relation to the increase in mechanical strengthening of the root is discussed.  相似文献

Background and Aims

Apoplasmic barriers in plants fulfil important roles such as the control of apoplasmic movement of substances and the protection against invasion of pathogens. The aim of this study was to describe the development of apoplasmic barriers (Casparian bands and suberin lamellae) in endodermal cells of Arabidopsis thaliana primary root and during lateral root initiation.

Methods

Modifications of the endodermal cell walls in roots of wild-type Landsberg erecta (Ler) and mutants with defective endodermal development – scarecrow-3 (scr-3) and shortroot (shr) – of A. thaliana plants were characterized by light, fluorescent, confocal laser scanning, transmission and cryo-scanning electron microscopy.

Key Results

In wild-type plant roots Casparian bands initiate at approx. 1600 µm from the root cap junction and suberin lamellae first appear on the inner primary cell walls at approx. 7000–8000 µm from the root apex in the region of developing lateral root primordia. When a single cell replaces a pair of endodermal and cortical cells in the scr-3 mutant, Casparian band-like material is deposited ectopically at the junction between this ‘cortical’ cell and adjacent pericycle cells. Shr mutant roots with an undeveloped endodermis deposit Casparian band-like material in patches in the middle lamellae of cells of the vascular cylinder. Endodermal cells in the vicinity of developing lateral root primordia develop suberin lamellae earlier, and these are thicker, compared wih the neighbouring endodermal cells. Protruding primordia are protected by an endodermal pocket covered by suberin lamellae.

Conclusions

The data suggest that endodermal cell–cell contact is required for the spatial control of Casparian band development. The effects of Cd on the growth and Cd uptaking in corn (Zea mays L.) were explored under different Cd stress. The results showed that no reduction in shoot and root dry matter yields were noted when the plants were grown at Cd supply levels ≤100 μmol l?1 nutrient solution. The Cd concentration in the shoots and roots of corn increased sharply with increasing external Cd supply levels, peaked at 50 μmol l?1, and then decreased slowly with further increasing Cd levels due to high Cd toxic effects on root growth. The concentrations of chlorophyll a, chlorophyll b and chlorophyll a + b in the leaf of corn decreased slowly with increasing external Cd supply levels. Proline concentrations of corn increased when the plants were grown under the external Cd influence. The lower concentration of Cd treatments did not influenced the growth of corn significantly, and increased the uptake of Cd, the higher levels of Cd supply caused significantly physiological resposes and decreased the Cd uptaking.  相似文献
