Summary In an ultrastructural and cytochemical study of tentoxin-treatedSorghum bicolor (L.) Moench, both bundle sheath and mesophyll plastids were severely affected, Plastids from chlorotic leaf areas lacked most internal membranes yet had plastid ribosomes and large fibrillar areas of plastid DNA. In recovered areas (mottled yellow and green), cells were found that had plastids of near-normal ultrastructure as well as the severely affected plastid-types found in chlorotic leaf areas. Polyphenol oxidase (PPO) cytochemistry of these mottled leaf areas indicated that all recovered mesophyll plastids had PPO whereas all the abnormal mesophyll plastids showed no activity. Because bundle sheath plastids ofSorghum have no PPO activity at any developmental stage, yet are affected by tentoxin, PPO cannot be uniquely affected by this toxin. We suggest that tentoxin may affect the transport of cytosolic proteins into the plastid.  相似文献

Polyphenol oxidases (PPOs; EC 1.14.18.1 or EC 1.10.3.2) catalyze the oxidation of phenolics to quinones, highly reactive intermediates whose secondary reactions are responsible for much of the oxidative browning that accompanies plant senescence, wounding, and responses to pathogens. To assess the impact of PPO expression on resistance to Pseudomonas syringae pv. tomato we introduced a chimeric antisense potato PPO cDNA into tomato (Lycopersicon esculentum L.). Oxidation of caffeic acid, the dominant o-diphenolic aglycone of tomato foliage, was decreased ca. 40-fold by antisense expression of PPO. All members of the PPO gene family were downregulated: neither immunoreactive PPO nor PPO-specific mRNA were detectable in the transgenic plants. In addition, the antisense PPO construct suppressed inducible increases in PPO activity. Downregulation of PPO in antisense plants did not affect growth, development, or reproduction of greenhouse-grown plants. However, antisense PPO expression dramatically increased susceptibility to P. syringae expressing the avirulence gene avrPto in both Pto and pto backgrounds. In a compatible (pto) interaction, plants constitutively expressing an antisense PPO construct exhibited a 55-fold increase in bacterial growth, three times larger lesion area, and ten times more lesions cm–2 than nontransformed plants. In an incompatible (Pto) interaction, antisense PPO plants exhibited 100-fold increases in bacterial growth and ten times more lesions cm–2 than nontransformed plants. Although it is not clear whether hypersusceptibility of antisense plants is due to low constitutive PPO levels or failure to induce PPO upon infection, these findings suggest a critical role for PPO-catalyzed phenolic oxidation in limiting disease development. As a preliminary effort to understand the role of induced PPO in limiting disease development, we also examined the response of PPO promoter::-glucuronidase constructs when plants are challenged with P. syringae in both Pto and pto backgrounds. While PPO B inducibility was the same in both compatible and incompatible interactions, PPO D, E and F were induced to higher levels and with different expression patterns in incompatible interactions.  相似文献

Polyphenol oxidase (PPO; EC 1.10.3.2 or EC 1.14.18.1) takes part in the response of tomato plants (Lycopersicon esculentum Mill.) to wounding and herbivore attack, mediated by the octadecanoid wound-signaling pathway. Wounding and methyl jasmonate (MeJA) induce expression of ppo genes and markedly increase the level of the enzyme. We report that pretreatment with MeJA also markedly increased the ability of isolated tomato chloroplasts to import and process PPO precursors (pPPO). Pea (Pisum sativum L.) chloroplasts showed no such response. Wounding or ethylene alone was ineffective but ethylene was synergistic with MeJA. Treatment with MeJA conferred a strong binding of pPPO, or its processing intermediate, to thylakoids and subsequent translocation into the lumen and processing to the mature protein. The effect on PPO import and translocation was evident after 8–16 h exposure to MeJA. Membrane-bound pPPO was cross-linked to a proteinaceous component of the thylakoid translocation apparatus, apparently induced by MeJA. The import and processing of other nuclear-encoded thylakoid proteins were not affected by MeJA in tomato. A 90-kDa protein that co-fractionated with thylakoids was induced along with the increase in competence for PPO import, and was identified as the proteinase-inhibitor multicystatin. Abstract

The ubiquitous type-3 copper enzyme polyphenol oxidase (PPO) has found itself the subject of profound inhibitor research due to its role in fruit and vegetable browning and mammalian pigmentation. The enzyme itself has also been applied in the fields of bioremediation, biocatalysis and biosensing. However, the nature of PPO substrate specificity has remained elusive despite years of study. Numerous theories have been proposed to account for the difference in tyrosinase and catechol oxidase activity. The “blocker residue” theory suggests that bulky residues near the active site cover CuA, preventing monophenol coordination. The “second shell” theory suggests that residues distant (~8?Å) from the active site, guide and position substrates within the active site based on their properties e.g., hydrophobic, electrostatic. It is also hypothesized that binding specificity is related to oxidation mechanisms of the catalytic cycle, conferred by coordination of a conserved water molecule by other conserved residues. In this review, we highlight recent developments in the structural and mechanistic studies of PPOs and consolidate key concepts in our understanding toward the substrate specificity of PPOs.  相似文献

Summary Leaves of young seedlings of a number of tall cultivars of wheat, lacking the dwarfing Rht genes, readily responded to a brief 2 min exposure to CO, as assessed by in vivo aerobic assay of nitrate reductase. This test depends on the inhibition of cytochrome c oxidase by CO, which in turn renders cytosolic NADH available for the reduction of nitrate to nitrite in vivo. Semi-dwarf cultivars of wheat (Rht present) did not respond to CO in this way. Since CO forms a complex only with reduced cytochrome a3, the results indicate differences in the redox state of cytochrome a3, during in situ respiration of leaves from tall and semi-dwarf plants which are likely to be under genetic control.  相似文献

Wheat spikelets detached from the spike at anthesis were cultured on solidified media and successfully produced mature grains. These grains resembled normal grains and contained well-developed, embryos. Lower concentrations of glutamine favored dry weight increase in developing grains. Such grains were indistinguishable from grains from greenhouse-grown plants in germination on moist blotting sheets. The technique of individual spikelet culture can be used to study physiology and development of wheat grains and kernels and to study host-pathogen interactions in wheat floret diseases such as Karnal bunt.  相似文献

Higher polyphenol oxidase (PPO) activity in wheat kernels and flour has been implicated in the time dependent darkening of various end-products. Previous study conducted on a bread wheat (Triticum aestivum L.) doubled haploid (DH) mapping population derived from Chara (medium-high PPO) and WW2449 (low PPO) identified a major QTL for PPO activity located on the long arm of chromosome 2A. Physical mapping of SSR markers accounting for up to 84% of phenotypic variation for PPO activities suggests that the candidate PPO locus is localised in the deletion bin delimited by 2AL 0.77–0.85. In order to develop functional gene markers, nine wheat ESTs mapped to this deletion bin and partial PPO reference genes were explored for their sequence identities and linkage with PPO locus in a mapping population. In the present study, two markers: one SNP and one CAPS based upon BQ161439 sequence variation between the parents were identified which exhibited a tight linkage (0–0.6 cM) with the PPO loci designated as XTc1 and XPPO- LDOPA. We also mapped the reference PPO gene (GenBank AY526268) characterised from developing kernels of wheat, on the long arm of chromosome 2A which exhibited a complete linkage with XPPO- L DOPA locus. Results suggest that PPO variation displayed in the DH population from Chara/WW2449 is due to the same reference PPO gene. Allelic homoplasy of tightly linked markers, indicated that these markers are ‘diagnostic’ for the selection of low PPO gene in a range of germplasm being used in different Australian breeding programs. Identification and validation of ‘functional gene markers’ would facilitate in enhancing the selection efficiency for low PPO activity in wheat breeding programs.  相似文献

Summary Diallel crosses between five divergent vulgare wheat cultivars were made in order to evaluate the mode of inheritance and combining ability of grain proteins. Significant differences in grain protein content were found between cultivars and their hybrids. It was established that the inheritance of seed protein in the F1 generation included both additive and non-additive gene action.  相似文献

A kinetic study of the activity of mushroom polyphenol oxidase in an organic system was carried out to obtain detailed enzyme kinetic data in relation to optimization of reaction conditions and substrate specificity. A simple method for consistent measurement of reaction rates in the heterogeneous enzyme/organic solvent system (consisting of immobilized polyphenol oxidase and a hydrated solution of the substrate in chloroform) was designed. The aqueous content of the system was optimized using p-cresol as the substrate. With this system, a crude extract of Agaricus bisporus was used to hydroxylate and oxidize a range of selected p-substituted phenolic substrates, yielding o-quinone products. Michaelis-Menten kinetics were used to obtain apparent K(M) and V(max) values with respect to each of these substrates. Results from this analysis indicated a correlation between the enzymic kinetic parameters obtained and the steric requirements of the substrates, which could be rationalized in terms of the restricted flexibility of the enzyme when it is in chloroform and also in terms of substrate and solvent hydrophobicity. In the course of the investigation UV molar absorption coefficients of several o-quinones were measured by a novel method: (1)H nuclear magnetic resonance (NMR) spectroscopy was employed to determine component concentrations in reaction mixtures resulting from the transformation of phenols by polyphenol oxidase in chloroform. Thus the UV molar absorption coefficients could be obtained directly, avoiding the necessity to isolate the water-sensitive, unstable o-quinones. (c) 1993 John Wiley & Sons, Inc.  相似文献

Retrotransposon-based molecular markers have been developed to study bread wheat ( Triticum aestivum) and its wild relatives. SSAP (Sequence-Specific Amplification Polymorphism) markers based on the BARE-1/ Wis-2-1A retrotransposons were assigned to T. aestivum chromosomes by scoring nullisomic-tetrasomic chromosome substitution lines. The markers are distributed among all wheat chromosomes, with the lowest proportion being assigned the D wheat genome. SSAP markers for BARE-1/ Wis-2-1A and three other wheat retrotransposons, Thv19 , Tagermina and Tar1, are broadly distributed on a wheat linkage map. Polymorphism levels associated with these four retrotransposons vary, with BARE-1/ Wis-2-1A and Thv19 both showing approximately 13% of bands polymorphic in a mapping population, Tagermina showing approximately 17% SSAP band polymorphism and Tar1 roughly 18%. This suggests that Tagermina and Tar1 have been more transpositionally active in the recent evolutionary past, and are potentially the more useful source of molecular markers in wheat. Lastly, BARE-1 / Wis-2-1A markers have also been used to characterise the genetic diversity among a set of 35 diploid and tetraploid wheat species including 26 Aegilops and 9 Triticum accessions. The SSAP-based diversity tree for Aegilops species agrees well with current classifications, though the Triticum tree shows several significant differences, which may be associated with polyploidy in this genus.Communicated by M.-A. Whether the two tetraploid wheat species, the well known Triticum turgidum L. (macaroni wheat, AABB genomes) and the obscure T. timopheevii Zhuk. (AtAtGG), have monophyletic or diphyletic origin from the same or different diploid species presents an interesting evolutionary problem. Moreover, T. timopheevii and its wild form T. araraticum are an important genetic resource for macaroni and bread-wheat improvement. To study these objectives, the substitution and genetic compensation abilities of individual T. timopheevii chromosomes for missing chromosomes of T. aestivum Chinese Spring (AABBDD) were analyzed. Chinese Spring aneuploids (nullisomic-tetrasomics) were crossed with a T. timopheevii x Aegilops tauschii amphiploid to isolate T. timopheevii chromosomes in a monosomic condition. The F1 hybrids were backcrossed one to four times to Chinese Spring aneuploids without selection for the T. timopheevii chromosome of interest. While spontaneous substitutions involving all At- and G-genome chromosomes were identified, the targeted T. timopheevii chromosome was not always recovered. Lines with spontaneous substitutions from T. timopheevii were chosen for further backcrossing. Six T. timopheevii chromosome substitutions were isolated: 6At (6A), 2G (2B), 3G (3B), 4G (4B), 5G (5B) and 6G (6B). The substitution lines had normal morphology and fertility. The 6At of T. timopheevii was involved in a translocation with chromosome 1G, resulting in the transfer of the group-1 gliadin locus to 6At. Chromosome 2G substituted for 2B at a frequency higher than expected and may carry putative homoeoalleles of gametocidal genes present on group-2 chromosomes of several alien species. Our data indicate a common origin for tetraploid wheat species, but from separate hybridization events because of the presence of a different spectrum of intergenomic translocations.  相似文献

Leaf rust, caused by Puccinia triticina Eriks., is an important foliar disease of common wheat (Triticum aestivum L.) worldwide. Pyramiding several major rust-resistance genes into one adapted cultivar is one strategy for obtaining more durable resistance. Molecular markers linked to these genes are essential tools for gene pyramiding. The rust-resistance gene Lr41 from T. tauschii has been introgressed into chromosome 2D of several wheat cultivars that are currently under commercial production. To discover molecular markers closely linked to Lr41, a set of near-isogenic lines (NILs) of the hard winter wheat cultivar Century were developed through backcrossing. A population of 95 BC3F2:6 NILs were evaluated for leaf rust resistance at both seedling and adult plant stages and analyzed with simple sequence repeat (SSR) markers using bulked segregant analysis. Four markers closely linked to Lr41 were identified on chromosome 2DS; the closest marker, Xbarc124, was about 1 cM from Lr41. Physical mapping using Chinese Spring nullitetrasomic and ditelosomic genetic stocks confirmed that markers linked to Lr41 were on chromosome arm 2DS. Marker analysis in a diverse set of wheat germplasm indicated that primers BARC124, GWM210, and GDM35 amplified polymorphic bands between most resistant and susceptible accessions and can be used for marker-assisted selection in breeding programs.  相似文献

Micellar catalysis of polyphenol oxidase in AOT/cyclohexane   总被引:4,自引:0,他引:4  
The catalytic behaviour of mushroom polyphenol oxidase has been studied in dioctylsulphosuccinate (AOT)/cyclohexane reverse micelles. The steady-state conditions were accomplished up to 20 min and 17 μg protein in the assay towards 4-methylcatechol and no loss of specific activity was observed relative to aqueous medium. The pH activity profile of the enzyme was kept in reverse micelles as in water, showing a plateau between 5 and 6.5. The stability of polyphenol oxidase to pH was also studied and about 20% inactivation was found in reverse micelles relative to aqueous medium at neutral pHs. Moreover there was a decrease of stability at acidic pHs. The optimum Wo obtained was 20 and the enzyme was nearly independent of the surfactant concentration at constant Wo.

Kinetic studies of polyphenol oxidase towards several substrates showed that the substrate inhibition by p-cresol and 4-methylcatechol observed in buffer was not kept in AOT/cyclohexane reverse micelles. Moreover, the Km increased and the catalytic efficiency (V/Km) of the enzyme decreased as the hydrophobicity of substrates was increased.  相似文献

Summary Tissue browning that frequently results in the early death of bamboo shoots in vitro correlated directly with polyphenol oxidase (PPO, EC 1.10.3.1) activity and inversely with titratable acidity. It was unrelated to the level of endogenous phenols. During the course of culture, timing of PPO activity paralleled that of explant browning. Browning was highest among shoots cultured in a medium of pH 8, which was consistent with the pH optinum of the bamboo enzyme. The pH optimum was first determined with the crude enzyme, then verified with two purified isozymes. Stability of the bamboo PPO was also highest at pH 10. PPO activities of the severely browning Dendrocalamus latiflorus, the moderately browning Phyllostachys nigra, and the relatively non-browning Bambusa oldhamii were inhibited strongly by ascorbic acid, cysteine, sodium diethyldithiocarbamate, and sodium sulfite. But characterization of bamboo PPO according to enzyme inhibitors was not possible because enzyme extracts of the three species gave varied responses to the traditional substances. Nutrient medium addenda of some PPO inhibitors, namely ascorbic acid, cysteine, kojic acid, and thiourea, mainly enhanced browning. However, ferulic acid at 3 mM and lower concentrations reduced the number of brown shoots per culture, although not the percentage of cultures that browned. Polyvinylpyrrolidone failed completely to suppress browning. The two purified isozymes showed different temperature optima for PPO activity: 60°C and 65°C. The purified isozymes displayed a substrate preference for dopamine, or a cathecol oxidase characteristics.  相似文献

Polyphenol oxidases (PPOs) are involved in the time-dependent darkening and discolouration of Asian noodles and other wheat end products. In this study, a doubled haploid (DH) population derived from Chara (moderately high PPO activity)/WW2449 (low PPO activity) was screened for PPO activity based on l-DOPA and l-tyrosine assays using whole seeds. Both these assays were significantly genetically correlated (r=0.91) in measuring the PPO activity in this DH population. Quantitative trait loci (QTLs) analysis utilising a skeleton map enabled us to identify a major QTL controlling PPO activity based on l-DOPA and l-tyrosine on the long arm of chromosome 2A. The simple sequence repeat (SSR) marker GWM294b explained over 82% of the line mean phenotypic variation from samples collected in both 2000 and 2003. Four SSR markers were validated for PPO linkage in genetically diverse backgrounds and proven to correctly predict the PPO activity in more than 92% of wheat lines. Physical mapping using deletion lines of Chinese Spring has confirmed the location of the GWM294b, GWM312 and WMC170 on chromosome 2AL, between deletion breakpoints 2AL-C to 0.85. In order to identify functional gene markers, data searches for alignments between rice BAC/PAC clones assembled on chromosome 1 and 4, chromosome 7, and (1) the wheat expressed sequence tags mapped in deletion bin (2AL-C to 0.85) and (2) the coding sequence of a previously cloned wheat PPO gene were made and found significant sequence similarities with the PPO gene or common central domain of tyrosinase. Available PPO gene sequences in the National Centre for Biotechnology Information (NCBI) database have revealed that there is a significant molecular diversity at the nucleotide and amino acid level in the wheat PPO genes.Electronic Supplementary Material Supplementary material is available for this article at .  相似文献

Summary Coriolus versicolor, a white-rot Basidiomycete, secretes cellulolytic and ligninolytic enzymes as well as polyphenol oxidase (PPO). Whereas the former degrade wood polymers, the latter can convert diphenols to diquinones and oligomerize syringic acid, a lignin derivative. Certain phenolic compounds can serve as disease-resistance factors controlling the proliferation of wood-decay fungi within host tissues. BecauseC. vesicolor can be batch-cultured, overproduction and enhanced secretion of enzymes of biological and commercial interests are feasible. Reported here are the results of attempts to define the timed appearances of intracellular and extracellular PPO, to assess substrate specificity as well as distinguish synthesis versus activation of intracellular PPO and to partially purify extracellular PPO. These efforts were to provide data enabling cell-free synthesis of PPO, cloning of the gene(s) for the oxidase and the establishment of its subcellular route of secretion. Whereas two protein peaks (6 and 12 days in a 16 day time-course) were observed for dialyzed mycelial homogenates, the homogenates' PPO specific activity rose between 4 and 12 days and then declined. Total extracellular protein content climbed from 6 to 15 days for dialyzed growth medium and the medium's PPO specific activity rose at 4 days post-inoculation and except at 9 days increased linearly to 15 days. When aliquots of dialyzed 12 and 15 day media were added to PPO assay mixtures containing catechol and either syringic or gallic acids, statistically significant differences in PPO specific activity between phenolic substrates were noted. Supplementation of cultures with 1.91 g cycloheximide ml growth medium–1 (control, growth medium only) together with 0.5 Ci [14C]-leucine revealed that cycloheximide inhibited PPO activity and suppressed [14C]-leucine incorporation into TCA-insoluble cytoplasmic protein. As for PPO partial purification, growth medium dialysis followed by 0–30% (NH4)2SO4 fractionation and subsequent 12 000×g dialyzate centrifugation yielded a 3.27-fold enhancement in PPO specific activity within the 12 000×g supernatant. Chromatography of the latter upon DEAE-Sephadex indicated that PPO exchanged with the DEAE counterion as it could be eluted with high ionic strength salt. These results suggest that: the occurrences of intracellular and extracellular PPO are time-dependent, intracellular PPO is de novo synthesized, the preferred substrate for extracellular PPO appears to be catechol and extracellular PPO can be partially purified by a combination of dialysis and ammonium sulfate fractionation as well as possibly DEAE chromatography and/or Sephadex G-150 gel filtration.  相似文献

A different expression pattern of polyphenol oxidases has been observed during storage in cultivars of potato (Solanum tuberosum L.) featuring different length of dormancy: a short-dormant cultivar showed, at the end of the dormancy, both the highest polyphenol oxidase activity and the largest number of enzyme isoforms. An isoform of polyphenol oxidase isolated at the end of the physiological dormancy from a short-dormant cultivar has been purified to homogeneity by means of column chromatography on phenyl Sepharose and on Superdex 200. The purification factor has been determined equal to 88, and the molecular mass of the purified isoform has been estimated to be 69 and 340 kDa by SDS polyacrylamide gel electrophoresis and gel filtration on Superdex 200, respectively, indicating this PPO isoform as a multimer. The corresponding zymogram features a diffused single band at the cathodic region of the gel and the pI of this polyphenol oxidase has been calculated equal to 6.5.  相似文献

Summary The genetic diversity of two diploid wheat species, Triticum monococcum and Triticum urartu (2n=2x=14), was assessed using random primers and the polymerase chain reaction (PCR). Electrophoretic analysis of the amplification products revealed a higher incidence of polymorphism in T. urartu than T. monococcum. Pair-wise comparisons of unique and shared polymorphic amplification products, were used to generate Jaccard's similarity coefficients. These were employed to construct phenograms using an unweighted pair-group method with arithmetical averages (UPGMA). The UPGMA analysis indicated a higher similarity among T. monococcum than T. urartu. Analysis of RAPD data appears to be helpful in determining the genetic relationships among genotypes.Contribution of the College of Agricultural Sciences, Texas Tech University Journal No. T-4-334.
