Plasma hormone levels were examined in 4 mature Zebu bulls of normal libido (HL) and 4 which were sexually inactive (LL). When used in an artificial insemination programme the 8 bulls had similar fertility. Basal levels of LH and testosterone (T) estimated from 8 sequential blood samples at 30 minute intervals were not different in HL and LL bulls. Exposure of the animals to an estrous cow did not stimulate LH release. Following sexual stimulation plasma T levels actually decreased by an average (±S.E) of 2.9 (±1.9) ng/ml in the HL group and increased by 3.9 (±1.6) ng/ml in the LL group. An injection of 1 mg GnRH (Hoechst) caused LH release of similar magnitude in HL and LL bulls. The elevation of plasma T which followed GnRH injection was significantly larger in HL bulls.Low libido was not associated with a deficiency of basal LH or T, nor with the ability of the pituitary to respond to GnRH.  相似文献

The objective of this study was to test the efficacy of an inexpensive, reusable internal artificial vagina (IAV) developed for breeding soundness evaluation of range beef bulls. In addition, sexual behavior during semen collection by IAV was compared to behavior during pasture breeding. Breeding soundness exams (BSEs) were conducted on 165 bulls in two consecutive years (96 in Year 1 and 69 in Year 2). In Year 1, BSEs were done twice in all bulls, once by a conventional protocol using electroejaculation (EEJ), followed by the IAV method, one week later. In Year 2, all BSEs were done on one day; 69 bulls by the IAV method followed by EEJ in 21 bulls that failed to serve the IAV. For semen collection using an IAV, mount cows were restrained in breeding crates and an IAV was inserted into the vagina just beyond the depth of the vestibular sphincter. After each copulation, the IAV was replaced for the next bull to be tested. Semen collection by IAV was successful for all bulls that mounted and penetrated cows during the testing period (54.3 and 69.6% of the bulls served the cows with IAVs in Year 1 and 2, respectively). Semen was collected successfully by EEJ from all bulls in both years. Differences were observed between semen collection methods in semen volume and percentage of sperm staining alive; however, the differences were opposite in Year 1 and 2 and, therefore, were probably due to natural variations in time and within bull rather than the method of semen collection. Semen collection by IAV allowed the detection of problems that prevented copulation in 8 bulls (4.8%) that were determined to be satisfactory potential breeders when semen was collected by EEJ. In Year 1, breeding observations were made at pasture for 15 bulls that served, and 15 bulls that did not serve cows with an IAV. Bulls that did not serve the IAV during the test period had fewer mounts, attempts to mount, and completed services at pasture than bulls that had served the IAV, indicating that such bulls are likely to be less efficient breeders. In conclusion, IAVs might be used as an alternative to EEJ for breeding soundness evaluation in about 50-70% of range bulls. Semen collection by IAV also allows observation of sex drive and mating ability thus improves the ability to select sound breeding bulls.  相似文献

In the first experiment, the effect of the stress of blood collection (via tail vessel puncture) on serum luteinizing hormone (LH) was evaluated in six nonsuckled first calf Brangus heifers. The animals were bled on days 22 and 31 postpartum at 15 minute intervals for a period of two hours. Blood was processed to yield serum and analyzed for LH via radioimmunoassay (RIA). There were no significant differences or fluctuations in serum LH levels between bleeding periods or between cows. Serum LH concentrations in nonsuckled cows were not affected by the stress of blood collection. In the second experiment, 24 first calf Brangus heifers were randomly assigned to one of four treatment groups. Treatment 1 cows were suckled once daily for approximately 30 min starting day 21 postpartum. Treatment 2 cows were suckled twice daily for approximately 30 min each time, starting 21 days postpartum. Treatment 3 cows were suckled once daily for approximately 30 min starting 30 days postpartum. Treatment 4 cows were suckled twice daily for approximately 30 min each time starting 30 days postpartum. Each cow was bled via tail vessel puncture on days one and nine following the start of each treatment. The blood sampling regime was similar to that used in Experiment 1 and consisted of four presuckling samples taken at 15 min intervals, one midsuckling sample (the calf was allowed to suckle for 15 min) and four postsuckling samples taken at 15 min intervals. Blood was collected, processed to yield serum and assayed for LH via RIA. Suckling intensity (SI) was found to have a significant effect on serum LH levels. The once daily suckled cows had higher (P<.01) mean serum LH levels than did the twice daily suckled cows (1.70 +/- .03 and 1.53 +/- .03 ng/ml, respectively). The LH concentrations decreased (P<.01) from the first to last bleeding time (BT). The mean serum LH levels for the presuckling, midsuckling and the first postsuckling samples were higher (P<.05) than the last postsuckling sample. The mean serum LH level for the first time period prior to suckling was higher (P<.05) than the last postsuckling sample. The mean serum LH level for the first time period prior to suckling was higher (P<.05) than the last two periods after suckling (1.73 +/- .08 ng/ml vs 1.51 +/- .06 and 1.41 +/- .06 ng/ml). Bleeding day (BD) and weaning day (WD) did not alter serum LH levels. The interactions found to be significant (P<.01) were SIxBD, SIxWD, BDxWD and BTxSIxBDxWD.  相似文献

Changes in LH, FSH, and testosterone concentrations, testicular firmness and resilience, and scrotal circumference were monitored in 16 Corriedale rams (8 experienced adult and 8 inexperienced young) for 20 days during which they were used to stimulate ewes. The experiment was conducted during November (mid-non-breeding season). Increases (P<0.05) were observed in LH and testosterone concentrations and in testicular firmness and resilience during the first 4 days when rams were in permanent contact with estrual ewes. During the following 13 days, when rams were in contact with non-estrual ewes (i.e. initially estrual ewes were no longer in estrus), LH and testosterone concentrations decreased. When initially anestrous ewes exhibited estrus 17 to 20 days later, concentrations of testosterone increased. Testicular firmness and resilience remained high throughout the period. We conclude that: (1) rams used to stimulate anestrous ewes show an increase in LH and testosterone concentrations beginning at 12 h after joining, and greater concentrations are maintained while estrual ewes and mating are allowed; and (2) estrous and mating activity are probably the most important stimuli for the increase in hormone concentrations.  相似文献

This study was conducted to determine whether exposure of ram lambs to estrual ewes during their first autumn and again as adults just before serving capacity tests (SCT) affected the outcome of the sexual performance tests. Treatments were either early exposure of Polypay ram lambs (i.e., 7-8-mo-old rams with ewes for 17 d [n=30] or no early exposure [n=30]), and late exposure (i.e., 16-19-mo-old rams with estrual ewes for 3 d) or no exposure to estrual ewes in a 2x2 factorial arrangement. Three serving capacity tests were conducted immediately after the early exposure period for individual ram lambs that were exposed to ewes early. Three sham sexual performance tests (i.e., four ram lambs placed in test pens for 30-min without ewes) were conducted with ram lambs that were not exposed to ewes early. All rams were evaluated during nine 30-min serving capacity tests over a 2-mo period at 16-19 mo of age to determine sexual performance. Prior to serving capacity tests, one half of the rams from each early exposure treatment were exposed to estrual-induced ewes for 3 d. Specific sexual behaviors (e.g., sniffs, flehmens, foreleg kicks, vocalizations, mount attempts, mounts, and ejaculations) were recorded during serving capacity tests. Number of sniffs, flehmens, foreleg kicks, vocalizations, and mount attempts were summed without estimating the value of importance and analyzed as courtship behaviors. Sexual performance data were analyzed with Mixed model procedures for repeated measures. During serving capacity tests, the early exposed rams exhibited more courtships (40.3+/-8.0 versus 23.4+/-4.6; P<0.05; LSM+/-estimated SE), mounts (11.3+/-1.0 versus 7.7+/-0.9; P<0.01), and ejaculations (3.3+/-0.2 or 2.4+/-0.2; P<0.01) than rams not exposed to ewes as ram lambs, respectively. We conclude that early exposure of 7-8-mo-old ram lambs to estrual ewes improves sexual performance in serving capacity tests at 16-19 mo of age in most rams whereas, late exposure to estrual ewes for 3 d prior to serving capacity tests did not improve sexual performance scores.  相似文献

In Experiment I, 38 crossbred suckled beef cows grazing fescue pastures and 34 crossbred beef cows grazing bluestem pastures were randomly allocated at the time of calving into a group with 4 teaser bulls or no bulls. Two blood samples were collected 7 d apart from the cows to determine cyclic activity 67 and 76 d after calving in the fescue and bluestem pastures, respectively. Progesterone greater than 1.0 ng/ml in one or both samples indicated cyclic activity. There was no difference in the percentage of cows cyclic among the different groups. The number of cyclic cows in the fescue pasture with bulls was 16/19 (84%); in the fescue pasture with no bulls, 14/19 (74%); in the bluestem pasture with bulls, 17/17 (100%); and in the bluestem pasture with no bulls, 16/17 (94%). Overall cyclic activity among all cows for teaser bull-exposed and no bull was similar, 33/36 (91%) and 30/36 (83%). Overall cyclic activity was greater (P < 0.05) in cows grazing bluestem (33/34), 97% than fescue pastures (30/38), 80%. Measurements of cyclic activity were initiated too late in the postcalving period to quantify differences in estrous activity between the bull and no bull treatment groups. Another trial was planned for the following year with a modified protocol. In Experiment II, blood samples were collected for progesterone concentrations soon after calving and were repeated at intervals to characterize both the occurrence and duration of estrous cycles. In this experiment, 29 crossbred suckled beef cows grazing fescue pastures were randomly allocated 12 d after calving (Day 0) into 1 of 2 groups with teaser bulls or without bulls. Nineteen crossbred beef cows grazing bluestem pastures were allocated similarly 10 d after calving (Day 0). Bulls were added to the groups with bulls in fescue and bluestem pastures on day 6 after the initial allocations. Blood samples were collected from all cows on Day 0 and every 3 d until Day 46. Means (+/- SEM) of the cumulative progesterone concentrations (ng/ml) per cow for the 16 samples from cows grazing fescue were 12.5 +/- 3.5 for cows exposed to bulls, 2.5 +/- 0.16 for cows not exposed to bulls, 27.6 +/- 4.42 for cows grazing bluestem pastures and exposed to bulls, and 16.0 +/- 2.75 for cows without exposure to bulls. Progesterone concentrations were higher in cows exposed to bulls (P < 0.01). The percentages of both short and normal cycles increased (P < 0.01) in groups exposed to bulls (88%, 21/24 and 63%, 15/24) when compared with the no bull groups (29%, 7/24 and 21%, 5/24), respectively. Paddock-mating performance was studied in seven beef bulls which were each joined to 28 or 29 females of mixed dairy and beef breeds for 8 weeks in single-sire joining groups in paddocks ranging from 4.0 to 28 ha. Seven unmated bulls were used as control animals. The estimated weight of testicular tissue per cow joined ranged from 12 to 32 g. Liveweight and testicular size and tone were recorded prior to, during and after joining. There was no apparent effect of mating on testicular characteristics of bulls, and pre-joining testicular size of the bulls was not significantly related to the calving results of the cows.Mating activity of the bulls was detected with a chin-ball harness and by behavioural observations covering eight intervals during the joining period. During the first 21 days (cycle I) of the joining period, 81% of the cows were mated and this increased to 89% by the end of the joining period. The overall return rate was 21%, the overall calving rate was 85% and no significant differences between the seven bulls were recorded in these two parameters. The relatively high level of sexual activity during cycle I apparently had no adverse effect on the fertility of the bulls since there was an even distribution over time of the return to first service of cows mated during cycle I. Mating activity of the bulls during cycle I had no significant effect on the mean percentage of time spent grazing or lying, but time spent standing was lower (P < 0.001) during cycle I than the remainder of the joining period.General activity, expressed as km travelled per day, was measured at regular intervals during the joining period for one working bull and one heifer in each of four mating groups. Mean activity of the bulls was significantly higher (11.3 km/day) during cycle I than the remainder of joining (2.45 km/day) and was significantly correlated (r = 0.8) to the mean number of cows mated per bull per day.  相似文献

With the objective of comparing the behavioral activities of synchronized cows in the presence or absence of a bull, 30 Indobrazil cows with a mature corpus luteum, identified by rectal palpation, were injected with 25 mg of prostaglandin F2α (PGF2α) approximately at Day 8 of the cycle for three consecutive cycles. Immediately after each injection, the animals were observed continuously for 100 h during three observation periods. Having established a hierarchy order in the herd, cows were allowed to interact with two mature bulls, one during the first cycle and the other during the second; in the third cycle, they were allowed to interact without the presence of a male. Behavioral activities such as butting, smelling the genital area, mounting, attempting to mount and following were recorded. No differences were detected between observation periods with respect to the accuracy of estrous detection, duration of the estrus period or interval from injection to onset of estrus. Sixty percent of the mounts recorded were initiated by cows high in the hierarchy order. Genital investigation was significantly more frequent when the bulls were present. The intensity of estrous behaviour varied in the same cows in the presence or absence of the bull. Furthermore, 20% of the cows injected three times showed no behavioral estrus. Mounting occurred more frequently at night in the absence of the bulls.  相似文献

Ten different volatile compounds were detected in bovine faeces using three chromatograms. The chemical profiles of estrus faeces were distinguished significantly from other phases by the presence of three specific substances, viz. acetic acid and propionic acid and 1-iodo undecane. The estrus specific synthetic compounds were rubbed onto the genital region of nonestrus animals (dummy cows), and the bulls were allowed to sniff the genital region and observed sexual behaviours. The statistical significance was higher (P<0.001) in bulls exhibiting repeated flehmen and mounting behaviours towards the mixture of acetic acid, propionic acid and 1-iodo undecane. The bioassay revealed that the fatty acids viz., acetic acid; propionic acid and 1-iodo undecane produced in faeces during estrus appear to be estrus specific. The results suggest that these compounds may be used as estrus indicator in cow, probably involved in bovine biocommunication.  相似文献

Serum lutinizing hormone (LH) levels in cattle during various reproductive states were measured by radioimmunoassay. A sharp LH peak observed at estrus (22.72 ± 5.68 ng/ml) was about 26 times higher than at other stages of the cycle (0.87 ± 0.06 ng/ml). LH levels during the first 90 days of pregnancy (0.75 ± 0.15 ng/ml) were similar to those of the estrous cycle, except during estrus, while those during the second (0.22 ± 0.07 ng/ml) and third trimesters of pregnancy (0.22 ± 0.08 ng/ml) were significantly lower. Higher levels than those of the cycling cows, except during estrus, were seen in ovariectomized cows (2.21 ± 0.56 ng/ml). Levels of LH in cows with cystic follicles (2.00 ± 0.49 ng/ml) were higher than the levels in the cycle. LH levels in bulls (1.29 ± 0.39 ng/ml) were comparable to that of estrous cows. Serum LH of calves increased with age from 1.00 ± 0.32 ng/ml (less than 30 days of age), to 2.30 ± 0.83 ng/ml (181 to 210 days of age), and the level after 151 days was significantly higher than that of the cyclic cows, except during estrus.  相似文献

A 2×4 factorial design was used to evaluate the effects of monensin (0 vs 200 mg) and estradiol-17β (E2) dose (0, 1, 2 vs 4 mg) on postpartum interval (PPI) to first estrus and on serum LH release at 21 days postpartum. Forty-eight spring calving Brangus cows were randomly stratified by calving date and sex of calf into two feeding groups within 24 hr of calving. Each group received 2.7 kg/head/day of a milo:cottonseed meal (4:1) mixture containing either 0 or 200 mg monensin. Coastal bermuda grass hay and water were available adlibitum. During the period of E2 treatment and bi-hourly blood sampling, suckling was controlled at 6-hr intervals.Mean cow weight and body condition score within cell changed less than 23 kg and 0.5 points, respectively, from day 1 to day 21 postpartum and were unaffected by treatment (P>0.10). PPI was reduced (P<0.01) and proportion of cows exhibiting estrual behavior by 85 days postpartum was increased (P<0.05) by treatment with 200 mg monensin and unaffected by E2 dose. Monensin fed cows had a longer (P<0.05) interval to LH response (ILH) and to peak LH (ILHP) at the 4 mg E2 dose. Monensin had no effect (P>0.10) on LH variables at 0, 1 or 2 mg E2.  相似文献

For herds using AI heat detection rate and calving rate are the two major determinants of compactness of calving, of the proportion of cows that fail to conceive in a defined breeding season. Numerous factors affect the expression of heat including, housing arrangement, floor surface, feet and leg problems and status of herd mates. The number of mounts a cow receives increases with the number of cows that are in heat simultaneously up to about 3-4 cows in heat. Generally, cows that are themselves in heat, coming into heat or were recently in heat are most likely to mount a cow that is in heat. Cows that are at the mid-stages of their cycles (day 5 to about day 16) are least likely to mount a cow that is in heat and consequently could be termed "poor heat detectors". Similarly, cows that are pregnant show less interest in mounting other cows that are in heat. In smaller herds and as more cows become pregnant the likelihood of more than one cow being heat on any given day becomes less, consequently, making heat detection more difficult. The single most important factor affecting heat detection efficiency is that those responsible for checking for heat should fully understand the signs of heat and be fully committed to heat detection for as long as it is planned to use Al. Technological aids to improve heat detection include the use of tail paint, oestrous synchronisation, vasectomised bulls, pressure activated heat mount detectors, radio telemetric devices, pressure sensitive mount count devices and pedometers. As herd size increases and labour become more expensive there will be a greater adoption of some of these technological aids.  相似文献

Normally cyclic heifers received 2500 i.u. PMSG i.m. at Day 10 of the oestrous cycle and 15 mg prostaglandin (PG) i.m. 48 h later. From 30 h after PG the LH concentration in the peripheral blood was estimated every hour using a rapid RIA method which allowed the LH concentration to be known within 4 h. Monoclonal antibody against PMSG was injected in the jugular vein of 29 heifers at 4.8 h after the maximum of the preovulatory LH peak; 28 heifers were not treated with anti-PMSG (controls). Peripheral blood concentrations of PMSG, LH, progesterone and oestradiol were compared. Ovaries were collected by ovariectomy at fixed times, 22-30 h after the LH peak, and numbers were counted of small (2-10 mm), large (greater than 10 mm) and ovulated follicles, and of follicles with a stigma. In anti-PMSG-treated cows, the PMSG concentration fell sharply to non-detectable levels within 2 h of the treatment, indicating that PMSG was neutralized in these cows at the onset of final follicular maturation. In all cows, the concentration of oestradiol showed a significant decrease at about 8 h after the LH peak. After anti-PMSG treatment ovulations took place from 24 until 30 h after the LH peak, whereas in control cows follicles had already ovulated at or before 22 h and ovulations continued until 30 h. At 30 h 90% of the follicles had ovulated in anti-PMSG-treated cows vs 72% in the controls, resulting in 15 and 8 ovulations per cow respectively (P less than 0.05). Also, administration of monoclonal antibody against PMSG synchronized final follicular maturation and shortened the period of multiple ovulations. In conclusion, neutralization of PMSG shortly after the preovulatory LH peak suppresses adverse effects of PMSG on final follicular maturation, leading to an almost 2-fold increase of the ovulation rate.  相似文献

The effect of suckling on serum and follicular fluid hormones and on follicular gonadotropin receptors was studied. Sixteen anestrous postpartum cows were assigned to 1 of 2 groups: suckled (S) or weaned (W). All calves were allowed to suckle ad libitum from parturition to 21 days postpartum when calves from W cows were weaned. All cows were ovariectomized on Day 25 postpartum. W cows had more (P less than 0.01) pulses of LH during the 96-h period from weaning until ovariectomy than S cows (6.3 vs. 1.3 pulses). Serum concentrations of prolactin (Prl), estrone (E1), estradiol-17 beta (E2) and progesterone (P) were not different (P greater than 0.10) between groups. Furthermore, there were n differences (P greater than 0.10) in follicular in contents of luteinizing hormone (LH), E1, E2 and P between the treatment groups. However, follicular fluid content of Prl was greater (P less than 0.05) in the W cows than in the S cows (123 vs. 65.1 ng/cow). The number of follicular LH receptors was greater (P less than 0.05) in the W cows than in the S cows (71.1 vs. 48.3 fmoles/mg protein) although the number of follicular follicle-stimulating hormone (FSH) receptors was not different (P greater than 0.10) between W cows and S cows (1531 vs. 1862 fmoles/mg protein). There were no correlation between serum hormone concentrations and follicular fluid hormone content; however, the numbers of follicular LH receptors and follicular fluid Prl content were highly correlated in the W cows (r = 0.85; P less than 0.05). It is concluded that removal of the suckling stimulus increases pulsatile LH release and the accumulation of Prl in the follicular fluid. These factors, either together or separately, may at least in part be responsible for the increase in follicular LH receptor concentrations that were observed in the W cows.  相似文献

Twenty-seven fall calving Brangus cows were randomly allotted to one of four treatment groups: nonsuckled monensin (NSM), suckled monensin (SM), nonsuckled control (NSC), and suckled control (SC). Cows were group fed 1.82 kg/hd/day concentrate and Coastal bermuda grass hay adlibitum. Monensin cows received 200 mg monensin/hd/day in the concentrate. At 0800 hr on day 21 postcalving, the calves were separated from the cows. Suckled monensin and SC cows were allowed to suckle their calves for 30 min at 6-hr intervals. Nonsuckled monensin and NSC cows were not suckled. Calves were given free access to the cows after 1400 hr on day 22 postpartum. At 0800 hr on day 22 postpartum, a blood sample was collected. A 100 μg GnRH challenge was administered IM at 0801 hr. Blood samples were collected at 15-min intervals for 6 hr postinjection. Changes in body weight and body condition from day 21 postpartum to the day of first estrus were not different (P>0.10) by dietary treatment. Monensin cows consumed 10.7% less hay than did the control cows. Serum luteinizing hormone (LH) following GnRH was greater (P<0.005) in suckled than nonsuckled cows. Control cows released more (P<0.005) LH in response to GnRH than did the monensin cows. The postpartum interval (to first estrus) for the monensin cows (92.4±14.7 days) was shorter (P<0.025) than the controls (138.5±9.5 days). A greater proportion (P<0.005) of the monensin cows (8 of 14) exhibited estrus by 90 days postpartum compared to the control cows (0 of 13). Monensin and suckling appear to exert independent and agonistic influences on pituitary function in the postpartum beef cow.  相似文献

An experiment was conducted to test the hypothesis that sterile mounts around insemination improves pregnancy rate to artificial insemination (AI) and to define the effects of age, season, time to complete AI and time of day of insemination. A total of 178 Simbrah females were randomly assigned by calving date and body condition to one of three treatments during two consecutive years: (1) mating stimuli with a sterile bull at the time the cows were detected in estrus; (2) mating stimuli immediately after completing AI; (3) without mating stimuli. All cows and heifers were maintained under the same conditions of handling and feeding within the two breeding seasons (winter 1995 and summer 1996). Vasectomized bulls were used for the sterile mounts. Cows and heifers that were given a sterile mount at the time of detection of estrus, had an increased pregnancy rate (60.0%) compared with females given a sterile mount after completing AI (25.4%) or females without the sterile mount (35.6%) (P < 0.01). Age, season, time to complete AI and time of day of AI were all non-significant (P > 0.05). Therefore, there is a biostimulatory effect of mating at the time beef cattle females are detected in estrus, on pregnancy rates to AI.  相似文献

An attempt was made to assess discomfort to the cow associated with milking machine function. Seven cows were presented with conditional discrimination tasks between alternative stimuli, using operant conditioning procedures. No discrimination between stimuli was evident. When allowed to choose between wide and narrow pulsation ratios, the 7 cows failed to do more than respond randomly. Four of the cows who were given a choice between potentially more or less aversive stimuli, pulsation vs. zero pulsation, showed no clear preference. The data suggest either that cows cannot discriminate within the experimental range of machine functions, i.e. that no measurable preference existed, or, under the experimental methods used, that cows could not learn the conditional discrimination required.  相似文献

Eight bulls were divided into two groups and injected with either charcoal-extracted steer blood serum or charcoal-extracted bovine follicular fluid (bFF). Ten-milliliter injections were given subcutaneous every 12 h for 4 wk. Jugular blood collected before, during and after the injection period was analyzed for follicle-stimulating hormone (FSH) and luteinizing hormone (LH) by radioimmunoassay. All bulls were exposed to restrained, estrual heifers for 15 min every 2 wk for 16 wk starting 4 wk before the first injection. The number of mounts and services by each bull was recorded. Semen was collected with an artificial vagina and evaluated on alternate weeks during the same period. The concentration of FSH in serum decreased (P < 0.05) by 12 h after the first injection and remained 61% lower than that of serum-injected bulls during the injection period. The concentration of FSH increased (P < 0.05) by 3 d after the last injection. Injections of bFF did not affect the concentration of LH in serum. Bovine follicular fluid injections significantly depressed FSH; however, libido, serving capacity, and semen characteristics were unchanged.  相似文献

Primiparous cows with low body condition at calving have an extended anovulatory period. Induction of ovulation and oestrus is possible with progesterone treatment but the response to this treatment differs between Friesian and Jersey breeds. The objective of this study was to describe changes in pulsatile LH secretion and the synchrony of developing ovarian follicles that occur during a progesterone treatment period of 5 days in primiparous anovulatory cows. The experimental model compared the progesterone treatment with spontaneous post-partum changes as well as a breed comparison in a factorial design.Thirty-six cows (Jersey n=19 and Friesian n=17) were managed to calve with a low body condition score (BCS<4. 5). Daily changes in ovarian follicle size were observed with transrectal ultrasonography in each cow from 8 days post-partum. Thirty of these cows were diagnosed to be anovulatory at 12-18 days post-partum (day 0) and allocated to a treatment (n=16) or a control group (n=14), balanced for breed. Each treated cow had a progesterone-releasing controlled internal drug-releasing (CIDR) device inserted vaginally for 5 days while control cows were left untreated. Changes in plasma LH concentrations were measured with intensive blood sampling over 8 h on days -1, 1, and 4. Blood samples were also collected daily (06:00 h) for determination of plasma progesterone as well as oestradiol concentrations on days 6 and 8.Treatment with progesterone was associated with a transient initial decrease (day 1) in both LH pulse frequency and mean LH concentrations after device insertion, but both had returned to pre-treatment levels by day 4. Jersey cows had a greater pulse frequency, but there was no breed difference in mean LH concentrations. Patterns of ovarian follicle growth were affected by progesterone treatment with an increase in diameter of the dominant follicle (DF) identified after treatment initiation. This followed an earlier emergence of a new DF after device insertion. Follicular response to progesterone was dependent on the diameter of the DF present at treatment initiation. Those follicles >/=9 mm were replaced by a new DF during treatment such that the DF observed at the time of device removal was large (>/=9 mm) and growing in 13/16 cases.Progesterone was not effective for the induction of an LH surge, ovulation and oestrus in anovulatory cows with a low BCS. However, treatment was associated with synchronous development of a DF so that it was large and growing at the end of the treatment period in most cases. This synchronous development may be due to the transient suppression of LH and the presence of an LH-dependent DF.  相似文献

Six Brahman and six Hereford long-term ovariectomized cows were bled via tail vessel at 15 minute intervals for a period of 4 hours. Serum was collected and analyzed via radioimmunoassay (RIA) for luteinizing hormone (LH) to determine if ovariectomized Brahman and Hereford cows have pulsatile LH patterns and if breed of animal influenced LH patterns. Brahman and Hereford ovariectomized cattle did have pulsatile LH patterns. Although the trend was for higher LH levels in Hereford than Brahman cows there were no significant differences in mean serum LH levels, number or magnitude of LH peaks or serum LH pulse height.Six Brahman and five Hereford long-term ovariectomized cows were injected (IM) with a single dose of 500μg of gonadotropin releasing hormone (GnRH). Animals were bled via tail vessel at 15 minute intervals for a period of 6 hours. Serum was assayed for LH via RIA to determine if ovariectomized Brahman and Hereford cows differ in GnRH induced LH response. All animals showed increased serum LH in response to GnRH injection within the first 15 minute collection interval. There were no significant differences in duration of response between ovariectomized Brahman or Hereford cows. Ovariectomized Brahman cows had significantly lower (P<.005) Lh values per period than ovariectomized Hereford cows. It was therefore concluded that ovariectomized Brahman cows were significantly less responsive to GnRH induced LH release than were ovariectomized Hereford cows, although duration and shape of the response curves were not different.  相似文献
