Ship groundings and ice-breakers can cause pollution of the polar environment with antifouling biocides such as diuron and Irgarol 1051. The present study used pulse amplitude modulated fluorometry to compare single and joint toxicities of diuron and Irgarol 1051 on two freshwater taxa of microalgae (Chlorella and Chlamydomonas) originating from Arctic and temperate regions. 30 min acute toxicity tests using chlorophyll a (Chl a) fluorescence revealed that Arctic strains of microalgae were more sensitive to herbicides than their temperate counterparts. Diuron and Irgarol 1051 had equal toxicities in the Arctic species, while Irgarol 1051 was more toxic (EC50 = 5.55–14.70 μg L−1) than diuron (EC50 = 12.90–>40 μg L−1) in the temperate species. Toxicity assessment of various mixtures of diuron and Irgarol 1051 revealed antagonistic, additive, and synergistic effects. Our data suggest that herbicides can adversely affect photosynthesis in Arctic microalgae at relatively low levels, and their impact can increase under complex mixture conditions.  相似文献

The effects of NH4+ or NO3 on growth, resource allocation and nitrogen (N) uptake kinetics of two common helophytes Phragmites australis (Cav.) Trin. ex Steudel and Glyceria maxima (Hartm.) Holmb. were studied in semi steady-state hydroponic cultures. At a steady-state nitrogen availability of 34 μM the growth rate of Phragmites was not affected by the N form (mean RGR = 35.4 mg g−1 d−1), whereas the growth rate of Glyceria was 16% higher in NH4+-N cultures than in NO3-N cultures (mean = 66.7 and 57.4 mg g−1 d−1 of NH4+ and NO3 treated plants, respectively). Phragmites and Glyceria had higher S/R ratio in NH4+ cultures than in NO3 cultures, 123.5 and 129.7%, respectively.Species differed in the nitrogen utilisation. In Glyceria, the relative tissue N content was higher than in Phragmites and was increased in NH4+ treated plants by 16%. The tissue NH4+ concentration (mean = 1.6 μmol g fresh wt−1) was not affected by N treatment, whereas NO3 contents were higher in NO3 (mean = 1.5 μmol g fresh wt−1) than in NH4+ (mean = 0.4 μmol g fresh wt−1) treated plants. In Phragmites, NH4+ (mean = 1.6 μmol g fresh wt−1) and NO3 (mean = 0.2 μmol g fresh wt−1) contents were not affected by the N regime. Species did not differ in NH4+ (mean = 56.5 μmol g−1 root dry wt h−1) and NO3 (mean = 34.5 μmol g−1 root dry wt h−1) maximum uptake rates (Vmax), and Vmax for NH4+ uptake was not affected by N treatment. The uptake rate of NO3 was low in NH4+ treated plants, and an induction phase for NO3 was observed in NH4+ treated Phragmites but not in Glyceria. Phragmites had low Km (mean = 4.5 μM) and high affinity (10.3 l g−1 root dry wt h−1) for both ions compared to Glyceria (Km = 6.3 μM, affinity = 8.0 l g−1 root dry wt h−1). The results showed different plasticity of Phragmites and Glyceria toward N source. The positive response to NH4+-N source may participates in the observed success of Glyceria at NH4+ rich sites, although other factors have to be considered. Higher plasticity of Phragmites toward low nutrient availability may favour this species at oligotrophic sites.  相似文献

Functional indicators are being increasingly used to assess waterway health but their responses to pressure in non-wadeable rivers have not been widely documented or applied in modern survey designs that provide unbiased estimates of extent. This study tests the response of river metabolism and loss in cotton strip tensile strength across a land use pressure gradient in non-wadeable rivers of northern New Zealand, and reports extent estimates for river metabolism and decomposition rates. Following adjustment for probability of selection, ecosystem respiration (ER) and gross primary production (GPP) for the target population of order 5–7 non-wadeable rivers averaged −7.3 and 4.8 g O2 m−2 d−1, respectively, with average P/R < 1 indicating dominance by heterotrophic processes. Ecosystem respiration was <−3.3 g O2 m−2 d−1 for 75% of non-wadeable river length with around 20% of length between −10 and −20 g O2 m−2 d−1. Cumulative distribution functions of cotton strength loss estimates indicated a more-or-less linear relationship with river km reflecting an even spread of decay rates (range in k 0.0007–0.2875 d−1) across non-wadeable rivers regionally. A non-linear relationship with land cover was detected for GPP which was typically <5 g O2 m−2 d−1 where natural vegetation cover was below 20% and greater than 80% of upstream catchment area. For cotton strength loss, the relationship with land cover was wedge-shaped such that sites with >60% natural cover had low decay rates (<0.02 d−1) with variability below this increasing as natural cover declined. Using published criteria for assessing waterway health based on ER and GPP, 232–298 km (20–29%) of non-wadeable river length was considered to have severely impaired ecosystem functioning, and 436–530 km (42–50%) had no evidence of impact on river metabolism.  相似文献

Soil salinity is recognized worldwide as a major threat to agriculture, particularly in arid and semi-arid regions. Producers and decision makers need updated and accurate maps of salinity in agronomically and environmentally relevant ranges (i.e., <20 dS m−1, when salinity is measured as electrical conductivity of the saturation extract, ECe). State-of-the-art approaches for creating accurate ECe maps beyond field scale (i.e., 1 km2) include: (i) Analysis Of Covariance (ANOCOVA) of near-ground measurements of apparent soil electrical conductivity (ECa) and (ii) regression modeling of multi-year remote sensing canopy reflectance and other co-variates (e.g., crop type, annual rainfall). This study presents a comparison of the two approaches to establish their viability and utility. The approaches were tested using 22 fields (total 542 ha) located in California’s western San Joaquin Valley. In 2013 ECa-directed soil sampling resulted in the collection of 267 soil samples across the 22 fields, which were analyzed for ECe, ranging from 0 to 38.6 dS m−1. The ANOCOVA ECa-ECe model returned a coefficient of determination (R2) of 0.87 and root mean square prediction error (RMSPE) of 3.05 dS m−1. For the remote sensing approach seven years (2007–2013) of Landsat 7 reflectance were considered. The remote sensing salinity model had R2 = 0.73 and RMSPE = 3.63 dS m−1. The robustness of the models was tested with a leave-one-field-out (lofo) cross-validation to assure maximum independence between training and validation datasets. For the ANOCOVA model, lofo cross-validation provided a range of scenarios in terms of RMSPE. The worst, median, and best fit scenarios provided global cross-validation R2 of 0.52, 0.80, and 0.81, respectively. The lofo cross-validation for the remote sensing approach returned a R2 of 0.65. The ANOCOVA approach performs particularly well at ECe values <10 dS m−1, but requires extensive field work. Field work is reduced considerably with the remote sensing approach, but due to the larger errors at low ECe values, the methodology is less suitable for crop selection, and other practices that require accurate knowledge of salinity variation within a field, making it more useful for assessing trends in salinity across a regional scale. The two models proved to be viable solutions at large spatial scales, with the ANOCOVA approach more appropriate for multiple-field to landscape scales (1–10 km2) and the remote sensing approach best for landscape to regional scales (>10 km2).  相似文献

The effect of two commonly used fertilizers, DAP (diammonium phosphate) and urea was studied on the freshwater flagellate Euglena gracilis using the automatic biotest ECOTOX. NOEC and EC50 values for various parameters like motility, velocity, cell shape and gravitaxis were calculated. The NOEC and EC50 values obtained for DAP were much lower than those for urea; i.e. DAP showed a stronger inhibitory effect as compared to urea. The inhibition caused by DAP increased with increasing exposure time over 24 h but urea showed no augmentation with increasing exposure time. Application of DAP resulted in an increased pH and high concentrations of ammonia but urea did neither affect the pH nor affect the ammonia concentration. Recovery experiments in deionized water after urea application showed a reconstitution of motility after 72 h. After an application of 1.35 g L−1 (24 h EC50 for motility) DAP motility recovered after 72 h but motility did not recover when the concentration was doubled (2.7 g L−1). The EC50 values obtained were compared with the EC50/LC50 values reported for other aquatic organisms and were found to be comparable with the reported values.  相似文献

In the current work nanoparticles (NPs) of α-amylase were generated in an aqueous solution using high-intensity ultrasound, and were subsequently immobilized on polyethylene (PE) films, or polycarbonate (PC) plates, or on microscope glass slides. The α-amylase NPs coated on the solid surfaces have been characterized by ESEM, TEM, FTIR, XPS and AFM. The substrates immobilized with α-amylase were used for hydrolyzing soluble potato starch to maltose. The amount of enzyme introduced in the substrates, leaching properties, and the catalytic activity of the immobilized enzyme were compared. The catalytic activity of the amylase deposited on the three solid surfaces was compared to that of the same amount of free enzyme at different pHs and temperatures. α-Amylase coated on PE showed the best catalytic activity in all the examined parameters when compared to native amylase, especially at high temperatures. When immobilized on glass, α-amylase showed better activity than the native enzyme over all pH and temperature values studied. However, the immobilization on PC did not improve the enzyme activity at any pH and any temperature compared to the free amylase. The kinetic parameters, Km and Vmax were also calculated. The amylase coated PE showed the most favorable kinetic parameters (Km = 5 g L−1 and Vmax = 5E−07 mol mL−1 min−1). In contrast, the anchored enzyme-PC exhibited unfavorable kinetic parameters (Km = 16 g L−1, Vmax = 4.2E−07 mol mL−1 min−1). The corresponding values for amylase-glass were Km = 7 g L−1, Vmax = 1.8E−07 mol mL−1 min−1, relative to those obtained for the free enzyme (Km = 6.6 g L−1, Vmax = 3.3E−07 mol mL−1 min−1).  相似文献

Aerobic granulation is a process in which suspended biomass aggregate and form discrete well-defined granules in aerobic systems. To investigate the properties and kinetics of aerobic granular sludge, aerobic granules were cultivated with glucose synthetic wastewater in a series of sequencing batch reactors (SBR). The spherical shaped granules were observed on 8th day with the mean diameter of 0.1 mm. With the organic loading rate (OLR) being increased to 4.0 g COD L−1 d−1, aerobic granules grew matured with spherical shape. The size of granules ranged from 1.2 to 1.8 mm, and the corresponding settling velocity of individual granule was 24.2–36.4 m h−1. The oxygen utilization rate (OUR) of mature granules was 41.90 g O2 kg MLSS−1 h−1, which was two times higher than that of activated sludge (18.32 g O2 kg MLSS−1 h−1). The experimental data indicated that the substrate utilization and biomass growth kinetics generally followed Monod's kinetics model. The corresponding kinetic coefficients of k (maximum specific substrate utilization rate), Ks (half velocity coefficient), Y (growth yield coefficient) and Kd (decay coefficient) were determined as follows, kc = 23.65 d−1, Kc = 3367.05 mg L−1, KN = 0.038 d−1, KN = 29.65 mg L−1, Y = 0.1927–0.2022 mg MMLS (mg COD)−1 and Kd = 0.00845–0.0135 d−1, respectively. Those properties of aerobic granules made aerobic granules system had a short setup period, high substrate utilization rate and low sludge production.  相似文献

The filamentous fungus Paecilomyces lilacinus was grown on n-hexadecane in submerged (SmC) and solid-state (SSC) cultures. The maximum CO2 production rate in SmC (Vmax = 11.7 mg CO2 Lg−1 day−1) was three times lower than in SSC (Vmax = 40.4 mg CO2 Lg−1 day−1). The P. lilacinus hydrophobin (PLHYD) yield from the SSC was 1.3 mg PLHYD g protein−1, but in SmC, this protein was not detected. The PLHYD showed a critical micelle concentration of 0.45 mg mL−1. In addition, the PLHYD modified the hydrophobicity of Teflon from 130.1 ± 2° to 47 ± 2°, forming porous structures with some filaments <1 μm and globular aggregates <0.25 μm diameter. The interfacial studies of this PLHYD could be the basis for the use of the protein to modify surfaces and to stabilize compounds in emulsions.  相似文献

The physiological reasons associated with differential sensitivity of C3 and C4 plant species to soil compaction stress are not well explained and understood. The responses of growth characteristics, changes in leaf water potential and gas exchange in maize and triticale to a different soil compaction were investigated. In the present study seedlings of triticale and maize, representative of C3 and C4 plants were subjected to low (L – 1.10 g cm−3), moderate (M – 1.34 g cm−3) and severe (S – 1.58 g cm−3) soil compaction level. Distinct differences in distribution of roots in the soil profile were observed. Plants of treatments M or S in comparison to treatment L, showed a decrease in leaf number, dry mass of stem, leaves and roots, and an increase in the shoot to root ratio. A drastic decrease in root biomass in M and S treatments in the soil profile on depth from 15 to 40 cm was observed. Any level of soil compaction did not influence the number of seminal and seminal-adventitious roots but decreased their length. The number and total length of nodal roots decreased with compaction. Changes of growth traits in M and S treatments in comparison to the L were greater for maize than for triticale and were accompanied by daily changes in water potential (ψ) and gas exchange parameters (PN, E, gs). Differences between M and S treatments in daily changes in ψ for maize were in most cases statistically insignificant, whereas for triticale, they were statistically significant. Differences in the responses of maize and triticale to soil compaction were found in PN, E and gs in particular for the measurements taken at 12:00 and 16:00. The highest correlation coefficients were obtained for the relationship between leaf water potential and stomatal conductance, both for maize and triticale, which indicates the close association between stomata behavior and changes in leaf water status.  相似文献

The reaction of [Cu(tren)(OH2)](ClO4)2 with KCN gave a mononuclear complex [Cu(tren)(CN)](ClO4) (1) (tren = tris(2-aminoethyl)amine). Using 1 as a building block, one pentanuclear compound, [{Cu(tren)(NC)}4Ni](ClO4)6 (2) and two trinuclear complexes, [{Cu(tren)NC}2Co(tren)](ClO4)5 · 2H2O (3), [{Cu(tren)CN}2NiL](ClO4)4 (4) (L = 3,10-bis(2-hydroxyethyl)-1,3,5,8,10,12-hexaazacyclotetradecane) were prepared and characterized by single crystal X-ray analysis. In 1, Cu(II) atom adopts a distorted trigonal bipyramidal (TBP) geometry. In 2, the Ni(II) atom occupies the center of the pentanuclear compound with a square-planar coordination geometry. In 3, the six-coordinated Co(III) atom presents a distorted octahedral geometry with four nitrogen atoms from tren and two carbon atoms of bridged cyano groups in cis-positions. In 4, the nickel atom is located in an inversion center and coordinated with two [(tren)CuCN]+ moieties through cyano-bridging ligands. Magnetic susceptibility measurements of 24 show that the magnetic interactions between the heterometallic ions are antiferromagnetical coupling through the cyano bridges with g = 2.25, J = −0.142 cm−1 and J = −0.167 cm−1 for 2, g = 2.06, J = −0.094 cm−1 for 3, and g = 2.20, J = −33.133 cm−1 for 4. Nutrient-use efficiency (NUE) within forests of the mangroves Rhizophora stylosa and Avicennia marina was estimated in arid Western Australia using litter fall rates and rates of leaf CO2 exchange. Litter fall rates ranged from 9.8 to 34.4 t DW ha−1 y−1 but equated to only 13–41% (mean = 30%) of net canopy primary production. Foliar N:P ratios were in most instances ≥16, suggesting P limitation. NUE for N based on litter fall rates were significantly less (NUEL = 167–322 g DW g−1 N) than those based on photosynthesis measurements (NUEP = 234–448 g DW g−1 N), suggesting that NUE estimates for nitrogen based on litter fall data are underestimates. NUEP estimates for N were significantly greater for R. stylosa than for A. marina. NUE for P were not significantly different, with NUEL ranging from 2905 to 5053 g DW g−1 P and NUEP ranging from 1632 to 4992 g DW g−1 P. Both sets of NUE are at the higher end of the range of estimates calculated for most other forests and equivalent to those for wet tropical mangroves. These arid-zone trees live in low-nutrient habitats, but it appears that selection on components of NUE (i.e. traits that reduce nutrient loss) rather than on NUE itself equates to a lack of clear patterns in NUE between different environments, emphasizing the flexible nature of nutrient allocation in woody plants. NUE in R. stylosa correlated inversely with mature leaf N and P content, implying that NUE in this species is maximized by the synthesis of low-nutrient leaves, i.e. a nutrient retention strategy, whereas such does not appear to be the case for A. marina. This strategy translates into a direct advantage in terms of net primary productivity for R. stylosa. This idea is supported by evidence of longer nutrient residence times for R. stylosa than for A. marina.  相似文献

Eight varieties of maize, namely Awassa 511 (A511), Birkata, composite of best families (CBF), Dendane, drought tolerant population (DTP1), Guto, Katumani and Keroshet planted in 1995 cropping season were compared for grain and crop residue yield and nutritive value of the stover. Significant differences (p < 0.05) were observed in grain and crop residue yields of the varieties. Grain yield was highest (6.99 t ha−1) in DTP1 and lowest (2.21 t ha−1) in Keroshet. Stover and total crop residue yields were significantly higher (p < 0.05) in A511 than in CBF and Dendane. The stem was the largest portion in the stover varying from 31% in Dendane to 50% in A511. The stover of Birkata showed a high crude protein (CP) and low ash and fibre contents compared to the other varieties. The CP content varied from 28 g kg DM−1 (Guto) to 61 g kg DM−1 (Birkata). Crop residue yield and quality parameters, except CP content of the stover, were not negatively correlated with grain yield. The potential utility index varied from 61% (Keroshet) to 74% (CBF). The varieties with higher potential utility indices, except Guto, also had relatively higher solubility, 48 h DM degradability and effective degradability compared to the other varieties. The overall results showed evidence of varietal differences in grain and stover yield and stover quality and indicated the possibility of selecting for maize varieties that combine high grain yield and desirable stover characteristics.  相似文献

Protocorm cultures of Dendrobium candidum were established in balloon type bubble bioreactors using Murashige and Skoog (MS) medium with 0.5 mg l−1 α-naphthaleneacetic acid (NAA), 2.5% (w/v) sucrose, 5:25 mM NH4:NO3 and 1% (v/v) banana homogenate for the production of biomass and bioactive compounds. In 3 l bioreactor containing 2 l medium, a maximum protocorm biomass (21.0 g l−1 dry biomass) and also optimum quantities of total polysaccharides (389.3 mg g−1 DW), coumarins (18.0 mg g−1 DW), polyphenolics (11.9 mg g−1 DW), and flavonoids (4.5 mg g−1 DW) were achieved after 7 weeks of culture. Based on these studies, 5 and 10 l bioreactor cultures were established to harvest 80 g and 160 g dry biomass. In 10 l bioreactors, the protocorms grown were accumulated with optimal levels of polysaccharides (424.1 mg g−1 DW), coumarins (15.8 mg g−1 DW), polyphenols (9.03 mg g−1 DW) and flavonoids (4.7 mg g−1 DW). The bioreactor technology developed here will be useful for the production of important bioactive compounds from D. candidum.  相似文献

Continuous anaerobic fermentations were performed in a novel external-recycle, biofilm reactor using d-glucose and CO2 as carbon substrates. Succinic acid (SA) yields were found to be an increasing function of glucose consumption with the succinic acid to acetic acid ratio increasing from 2.4 g g−1 at a glucose consumption of 10 g L−1, to 5.7 g g−1 at a glucose consumption of 50 g L−1. The formic acid to acetic acid ratio decreased from an equimolar value (0.77 g g−1) at a glucose consumption of 10 g L−1 to a value close to zero at 50 g L−1. The highest SA yield on glucose and highest SA titre obtained were 0.91 g g−1 and 48.5 g L−1 respectively. Metabolic flux analysis based on the established C3 and C4 metabolic pathways of Actinobacillus succinogenes revealed that the increase in the succinate to acetate ratio could not be attributed to the decrease in formic acid and that an additional source of NADH was present. The fraction of unaccounted NADH increased with glucose consumption, suggesting that additional reducing power is present in the medium or is provided by the activation of an alternative metabolic pathway.  相似文献

Pseudomonas putida 33 wild strain, subjected to gamma ray mutagenesis and designated as P. putida 300-B mutant was used as microbial rhamnolipid-producer by using distant carbon sources (viz. hydrocarbons, waste frying oils ‘WFOs’, vegetable oil refinery wastes and molasses) in the minimal media under shake flask conditions. The behavior of glucose as co-substrate and growth initiator was examined. The 300-B mutant strain showed its ability to grow on all the substrates tested and produced rhamnolipid surfactants to different extents however; soybean and corn WFOs were observed to be preferred carbon sources followed by kerosene and paraffin oils, respectively. The best cell biomass (3.5 g l−1) and rhamnolipids yield (4.1 g l−1) were obtained with soybean WFO as carbon source and glucose as growth initiator under fed-batch cultivation showing an optimum specific growth rate (μ) of 0.272 h−1, specific product yield (qp) of 0.318 g g−1 h and volumetric productivity (PV) of 0.024 g l−1 h. The critical micelle concentration of its culture supernatant was observed to be 91 mg rhamnolipids l−1 and surface tension as 31.2 mN m−1.  相似文献

Salinity is one of the serious abiotic stresses adversely affecting the majority of arable lands worldwide, limiting the crop productivity of most of the economically important crops. Sweet basil (Osmium basilicum) plants were grown in a non-saline soil (EC = 0.64 dS m−1), in low saline soil (EC = 5 dS m−1), and in a high saline soil (EC = 10 dS m−1). There were differences between arbuscular mycorrhizal (Glomus deserticola) colonized plants (+AMF) and non-colonized plants (−AMF). Mycorrhiza mitigated the reduction of K, P and Ca uptake due to salinity. The balance between K/Na and between Ca/Na was improved in +AMF plants. Growth enhancement by mycorrhiza was independent from plant phosphorus content under high salinity levels. Different growth parameters, salt stress tolerance and accumulation of proline content were investigated, these results showed that the use of mycorrhizal inoculum (AMF) was able to enhance the productivity of sweet basil plants under salinity conditions. Mycorrhizal inoculation significantly increased chlorophyll content and water use efficiency under salinity stress. The sweet basil plants appeared to have high dependency on AMF which improved plant growth, photosynthetic efficiency, gas exchange and water use efficiency under salinity stress. In this study, there was evidence that colonization with AMF can alleviate the detrimental salinity stress influence on the growth and productivity of sweet basil plants.  相似文献

The main photosynthesis and respiration parameters (dark respiration rate, light saturated production rate, saturation irradiance, photosynthetic efficiency) were measured on a total of 23 macrophytes of the Thau lagoon (2 Phanerogams, 5 Chlorophyceae, 10 Rhodophyceae and 6 Phaeophyceae). Those measurements were performed in vitro under controlled conditions, close to the natural ones, and at several seasons. Concomitantly, measurements of pigment concentrations, carbon, phosphorous and nitrogen contents in tissues were performed. Seasonal intra-specific variability of photosynthetic parameters was found very high, enlightening an important acclimatation capacity. The highest photosynthetic capacities were found for Chlorophyceae (e.g. Monostroma obscurum thalli at 17 °C, 982 μmol O2 g−1 dw h−1 and 9.1 μmol O2 g−1 dw h−1/μmol photons m−2 s−1, respectively for light saturated net production rate and photosynthetic efficiency) and Phanerogams (e.g. Nanozostera noltii leaves at 25 °C, 583 μmol O2 g−1 dw h−1 and 2.6 μmol O2 g−1 dw h−1/μmol photons m−2 s−1 respectively for light saturated net production rate and photosynthetic efficiency). As expected, species with a high surface/volume ratio were found to be more productive than coarsely branched thalli and thick blades shaped species. Contrary to Rd (ranging 6.7–794 μmol O2 g−1 dw h−1, respectively for Rytiphlaea tinctoria at 7 °C and for Dasya sessilis at 25 °C) for which a positive relationship with water temperature was found whatever the species studied, the evolution of P/I curves with temperature exhibited different responses amongst the species. The results allowed to show summer nitrogen limitation for some species (Gracilaria bursa-pastoris and Ulva spp.) and to propose temperature preferences based on the photosynthetic parameters for some others (N. noltii, Zostera marina, Chaetomorpha linum).  相似文献

Cheese whey powder (CWP) solution with different CWP or sugar concentrations was fermented to ethanol in a continuous fermenter using pure culture of Kluyveromyces marxianus (DSMZ 7239). Sugar concentration of the feed CWP solution varied between 55 and 200 g l−1 while the hydraulic residence time (HRT) was kept constant at 54 h. Ethanol formation, sugar utilization and biomass formation were investigated as functions of the feed sugar concentration. Percent sugar utilization and biomass concentrations decreased and the effluent sugar concentration increased with increasing feed sugar concentrations especially for the feed sugar contents above 100 g l−1. Ethanol concentration and productivity (DP) increased with increasing feed sugar up to 100 g l−1 and then decreased with further increases in the feed sugar content. The highest ethanol concentration (3.7%, v v−1) and productivity (0.54 gE l−1 h−1) were obtained with the feed sugar content of 100 g l−1 or 125 g l−1. The ethanol yield coefficient (YP/S) was also maximum (0.49 gE gS−1) when the feed sugar was between 100 and 125 g l−1. The growth yield coefficient (YX/S) decreased steadily from 0.123 to 0.063 gX gS−1 when the feed sugar increased from 55 to 200 g l−1 due to adverse effects of high sugar contents on yeast growth. The optimal feed sugar concentration maximizing the ethanol productivity and sugar utilization was between 100 and 125 g l−1 under the specified experimental conditions.  相似文献

α-Glucuronidase (EC 3.2.1.139) of family GH 115 from Scheffersomyces stipitis is a valuable enzyme for the modification of water-soluble xylan into insoluble biopolymers, due to its unique ability to act on polymeric xylans. The influence of growth rate on the production of α-glucuronidase by recombinant Saccharomyces cerevisiae MH1000pbk10D-glu in glucose-limited fed-batch culture was studied at 14 and 100 L scale. At and below the critical specific growth rate (μcrit) of 0.12 h−1 at 14 L scale, the biomass yield coefficient (Yx/s) remained constant at 0.4 g g−1 with no ethanol production, whereas ethanol yields relative to biomass (keth/x) of up to 0.54 g g−1 and a steady decrease in Yx/s were observed at μ > 0.12 h−1. Production of α-glucuronidase was growth associated at a product yield (kα-glu/x) of 0.45 mg g−1, with the highest biomass (37.35 g/L) and α-glucuronidase (14.03 mg/L) concentrations, were recorded during fed-batch culture at or near to μcrit. Scale-up with constant kLa from 14 to 100 L resulted in ethanol concentrations of up to 2.5 g/L at μ = 0.12 h−1. At this scale, α-glucuronidase yield could be maximised at growth rates below μcrit, to prevent localised high glucose concentration pockets at the feed entry zone that would induce oxido-reductive metabolism. This is the first report where recombinant production of α-glucuronidase (EC 3.2.1.139) by S. cerevisiae was optimised for application at pilot scale.  相似文献

The effect of nutrient addition on the growth of E. najas was evaluated in a dose response experiment using sand amended with phosphorus (P) and nitrogen (N), and in enrichment trials with N and P amendments to natural sediments. Plants, water and sediment came from lagoons of the Upper Paraná River Floodplain and from Itaipu Reservoir (Brazil). Relative growth rates (RGRs) of E. najas shoots, based on dry mass (DM), varied from 0.03 to 0.060 d−1 for both nutrients. Root:shoot biomass ratios were related to sediment exchangeable P (r = −0.419; P = 0.03) and N (r = −0.54; P = 0.006), however root RGR was not related to sediment nutrient concentrations. When natural sediments were amended with N and P, neither shoot nor root RGRs differed among treatments for substrata from either the reservoir or the floodplain lagoons (P > 0.05). Comparison of nutrient concentrations measured in natural sediments collected from several sites in both the Upper Paraná River Floodplain (range 49–213 μg P g−1 DM; 36–373 μg N g−1 DM) and Itaipu Reservoir (range 43–402 μg P g−1 DM; 7.9–238 μg N g−1 DM) showed that sediment N and P from these systems usually exceeded minimum requirements necessary for E. najas growth, as measured in the dose response experiment. Together, these results indicate that E. najas, at least in early stages of development, responds to sediment nutrient amendments and relies upon bottom sediments to meet its N and P requirements and that for at least two Brazilian ecosystems, growth of this species is not limited by insufficient sediment N or P. Thus, reducing N and P in water is not enough to control E. najas growth in short time periods in these ecosystems.  相似文献
