Computer control of carbon-nitrogen ratio in Spirulina platensis   总被引:2,自引:0,他引:2  
An on-line computer was used to control the ratio of carbon to nitrogen in algal biomass. An indirect method of growth and biomass estimation was utilized. This was based on balancing the amount of CO(2) carbon in and out of the algal bioreactor. It was shown that growth conditions govern the morphology and composition of Spirulina platensis. Cells grown under light limitation were narrower, had high levels of phycocyanin pigments, and were packed full of small lipid granules. Whereas cells grown under nitrogen limitation lost their characteristic blue-green color, had reduced levels of phycocyanin, were fatter, and were packed full of larger lipid granules.  相似文献

Mangueira Lagoon, located in the extreme south of Brazil, has water with physicochemical characteristics such as alkaline pH and carbonate levels propitious for the growth of the cyanobacterium Spirulina platensis. Previously published studies have shown that Mangueira Lagoon water supplemented with small quantities of carbon and nitrogen is suitable for S. platensis cultivation and can significantly reduce production costs. We studied mixed cultures of Spirulina platensis and the toxic cyanobacterium Microcystis aeruginosa using a 2(3) factorial design in which the three factors were the initial biomass concentration of S. platensis and M. aeruginosa and the type of culture medium (100% Zarrouk's medium or 80% Mangueira Lagoon water plus 20% Zarrouk's medium). The highest S. platensis maximum specific growth rate (mu(max)) occurred in the culture with the highest M. aeruginosa biomass concentration and when undiluted culture medium was used (micro(max) = 0.283 d(-1)). The highest M. aeruginosa specific death rate (k) was obtained in the presence of S. platensis (k = 0.555 d(-1)) and was independent of the initial M. aeruginosa biomass concentration and culture medium, demonstrating that S. platensis cultures are not susceptible to contamination by M. aeruginosa. The culture medium had no significant influence (p > 0.05) on S. platensis micro(max) values, indicating that production costs could be reduced by using a medium consisting of 80% Mangueira Lagoon water plus 20% Zarrouk's medium.  相似文献

Accumulation of Se in mixotrophic culture of Spirulina platensis was investigated in this study. Results indicated that glucose was better than acetate as an organic carbon source for mixotrophic culture of S. platensis. Supplementation of glucose (2 gL(-1)) significantly enhanced the biomass concentration (2.57 gL(-1)) and the production of phycocyanin (0.279 gL(-1)) and allophycocyanin (0.126 gL(-1)) in S. platensis, which were much higher than those of photoautotrophic culture (1.08 gL(-1), 0.119 gL(-1) and 0.042 gL(-1), respectively). Stepwise addition of Se during the growth phase avoided the inhibitory effect of high Se concentration on the growth of S. platensis. The Se enrichment favored the production of phycocyanin and allophycocyanin in the algal cells. The highest Se yield (1033 microgL(-1)) was obtained at an accumulative Se concentration of 250 mgL(-1), with organic Se percentage, biomass concentration, phycocyanin and allophycocyanin yields of 92.3%, 2.55 gL(-1), 0.295 gL(-1) and 0.153 gL(-1), respectively. These results indicated that the application of mixotrophic culture S. platensis with stepwise addition of Se to the medium could offer an effective and economical way for the production of high Se-enriched algal products.  相似文献

An integrated process for the indoor production of 13C labeled polyunsaturated fatty acids (PUFAs) from Phaeodactylum tricornutum is presented. The core of the process is a bubble column photobioreactor operating with recirculation of the exhaust gas using a low-pressure compressor. Oxygen accumulation in the system is avoided by bubbling the exhaust gas from the reactor in a sodium sulfite solution before returning to it. To achieve a high 13C enrichment in the biomass obtained, the culture medium is initially stripped of carbon, and labeled 13CO(2) is automatically injected on-demand during operation for pH control and carbon supply. The reactor was operated in both batch and semicontinuous modes. In semicontinuous mode, the reactor was operated at a dilution rate of 0.01 h(-1), resulting in a biomass productivity of 0.1 g l(-1) per day. The elemental analysis of the inlet and outlet flows of the reactor showed that 64.9% of carbon was turned into microalgal biomass, 34.9% remained in the supernatant mainly as inorganic compounds. Only 3.8% of injected carbon was effectively fixed as the target labeled product (EPA). Regarding the isotopic composition of fatty acids, results showed that fatty acids were not labeled in the same proportion, the higher the number of carbons the lower the percentage of 13C. Isotopic composition of EPA ranged from 36.5 to 53.5%, as a function of the methodology used (GC-MS, EA-IRMS or gas chromatography-combustion-isotope ratio mass spectrometry (GC-IRMS)). The low carbon uptake efficiency combined with the high cost of 13CO(2) make necessary to redefine the designed culture system to increase the efficiency of the conversion of 13CO(2) into the target product. Therefore, the possibility of removing 12C from the fresh medium, and recovering and recirculating the inorganic carbon in the supernatant and the organic carbon from the EPA depleted biomass was studied. The inorganic carbon of the fresh medium was removed by acidification and stripping with N(2). The inorganic carbon of the supernatant was recovered also by acidification and subsequent stripping with N(2). The operating conditions of this step were optimized for gas flow rate and type of contactor. A carbon recovery step for the depleted biomass was designed based on the catalytic oxidation to CO(2) using CuO (10 wt.%) as catalyst with an oxygen enriched atmosphere (80% O(2) partial pressure). In this way, the carbon losses reduced an 80.2% and the efficiency of the conversion of carbon in EPA was increased to 19.5%, which is close to the theoretical maximum. Further increase in 13CO(2) use efficiency is only possible by additionally recovering other labeled by-products present in the biomass: proteins, carbohydrates, lipids, and pigments.  相似文献

Organic carbon (C) and nitrogen (N) are essential for heterotrophic soil microorganisms, and their bioavailability strongly influences ecosystem C and N cycling. We show here that the natural 15N abundance of the soil microbial biomass is affected by both the availability of C and N and ecosystem N processing. Microbial 15N enrichment correlated negatively with the C : N ratio of the soil soluble fraction and positively with net N mineralization for ecosystems spanning semiarid, temperate and tropical climates, grassland and forests, and over four million years of ecosystem development. In addition, during soil incubation, large increases in microbial 15N enrichment corresponded to high net N mineralization rates. These results support the idea that the N isotope composition of an organism is determined by the balance between N assimilation and dissimilation. Thus, 15N enrichment of the soil microbial biomass integrates the effects of C and N availability on microbial metabolism and ecosystem processes.  相似文献

Yeast Trichosporon cutaneoum K-1 were cultivated on milk serum supplemented with nitrogen and phosphorus sources. The culture showed a high biosynthetic capacity. The specific growth rate (mu) of yeast on milk serum was 0.40-0.45 hr-1. During lactose assimilation in the synthetic medium mu was 0.43 hr-1 and the economic coefficent of the biomass yield was 54%. It was shown that assimilation first involved lactose and then organic acids. The composition of the resulting biomass was identified.  相似文献

Spirulina platensis was batch cultivated in a novel 3.5l magnetic air-lift photobioreactor with external-loop on which 0-0.55 T electromagnetic field (EMF) was imposed to investigate the EMF effects on the algal growth and nutrition composition. At the same time, the correlative mechanism was discussed mainly based on C, N, P uptake. It was found that, EMF has double-side effects on S. platensis cultivation depend on EMF intensity. 0.25 T EMF stress was found to be suitable for the algal cultivation enhancement increasing the maximum cell dry weight by 22% in a time period two days less than that of the control at 35 degrees C and 252 micromol photonsm(-2)S(-1). In addition, the nutritional composition of S. platensis was improved in both essential amino acids such as histidine and trace elements Ni, Sr, Cu, Mg, Fe, Mn, Ca, Co and V. It was suggested it was possible to optimize the cultivation process of S. platensis by imposing suitable weak EMF which may enhance the nutrition assimilation e.g. C, N, P and minerals by S. platensis.  相似文献

The effect of elevated CO2 on the carbon and nitrogen distribution within perennial ryegrass (L. perenne L.) and its influence on belowground processes were investigated. Plants were homogeneously 14C-labelled in two ESPAS growth chambers in a continuous 14C-CO2 atmosphere of 350 and 700 L L-1 CO2 and at two soil nitrogen regimes, in order to follow the carbon flow through all plant and soil compartments.After 79 days, elevated CO2 increased the total carbon uptake by 41 and 21% at low (LN) and high nitrogen (HN) fertilisation, respectively. Shoot growth remained unaffected, whereas CO2 enrichment stimulated root growth by 46% and the root/soil respiration by 111%, irrespective of the nitrogen concentration. The total 14C-soil content increased by 101 and 28% at LN and HN, respectively. The decomposition of the native soil organic matter was not affected either by CO2 or by the nitrogen treatment.Elevated CO2 did not change the total nitrogen uptake of the plant either at LN or at HN. Both at LN and HN elevated CO2 significantly increased the total amount of nitrogen taken up by the roots and decreased the absolute and relative amounts translocated to the shoots.The amount of soil nitrogen immobilised by micro-organisms and the size of the soil microbial biomass were not affected by elevated CO2, whereas both were significantly increased at the higher soil N content.Most striking was the 88% increase in net carbon input into the soil expressed as: 14C-roots plus total 14C-soil content minus the 12C-carbon released by decomposition of native soil organic matter. The net carbon input into the soil at ambient CO2 corresponded with 841 and 1662 kg ha-1 at LN and HN, respectively. Elevated CO2 increased these amounts with an extra carbon input of 950 and 1056 kg ha-1. Combined with a reduced decomposition rate of plant material grown at elevated CO2 this will probably lead to carbon storage in grassland soils resulting in a negative feed back on the increasing CO2 concentration of the atmosphere.  相似文献

Nitrogen and phosphorus concentration in the effluent of a wastewater treatment plant can vary significantly, which could affect the growth kinetic and chemical composition of microalgae when cultivated in this medium. The aim of this work was to study the rate of growth, nutrient removal and carbon dioxide biofixation as well as biomass composition of Scenedesmus obliquus (S. obliquus) when it is cultivated in wastewater at different nitrogen and phosphorus ratio, from 1:1 to 35:1. A more homogeneous method for calculating productivities in batch reactors was proposed. The proper N:P ratio for achieving optimum batch biomass productivity ranged between 9 and 13 (263 and 322 mg L?1 d?1 respectively). This was also the ratio range for achieving a total N and P removal. Above and below this range (9–13) the maximum biomass concentration changed, instead of the specific growth rate.The maximum carbon dioxide biofixation rate was achieved at N:P ratio between 13 and 22 (553 and 557 mg CO2 L?1 d?1 respectively). Lipid and crude protein content, both depend on the aging culture, reaching the maximum lipid content (34%) at the lowest N:P (1:1) and the maximum crude protein content (34.2%) at the highest N:P (35:1).  相似文献

* A previously published model of crop nitrogen (N) status based on intercepted photosynthetically active radiation (R(i), mol per plant) suggested that plant organic N accumulation is related to R(i) by a constant ratio, defined hereafter as the radiation use efficiency for N (NRUE). The aim of this paper was to compare the effects of N nutrition and CO2 enrichment on NRUE and RUE (radiation use efficiency for biomass accumulation). * In three unrelated glasshouse experiments, tomato plants (Solanum lycopersicum) grown in hydroponics were fed for 28 d (exponential growth) with full solutions containing constant NO3(-) concentrations ([NO3(-)]) ranging from 0.05 to 15 mol m(-3), both under ambient or CO2-enriched (1000 microl l(-1)) air. * Each experiment comprised five harvests. Low [NO3(-)] (< 0.3 mol m(-3)) limited growth via leaf area (LA) restriction and decreased light interception. CO2 enrichment enhanced dry weight and LA. RUE was not affected by [NO3(-)], but increased under CO2-enriched air. By contrast, NRUE was not affected by [NO3(-)] or CO2 enrichment. * It is suggested that the radiation efficiency for organic N acquisition (NRUE) did not depend on C or N nutrition for young plants grown under unstressed conditions.  相似文献

We tested the prediction that the abundance and diversity of arbuscular mycorrhizal (AM) fungi are influenced by resource availability and plant community composition by examining the joint effects of carbon dioxide (CO(2) ) enrichment, nitrogen (N) fertilization and plant diversity on AM fungi. ? We quantified AM fungal spores and extramatrical hyphae in 176 plots after 7 yr of treatment with all combinations of ambient or elevated CO(2) (368 or 560 ppm), with or without N fertilization (0 or 4 g Nm(-2) ), and one (monoculture) or 16 host plant species (polyculture) in the BioCON field experiment at Cedar Creek Ecosystem Science Reserve, Minnesota, USA. ? Extramatrical hyphal lengths were increased by CO(2) enrichment, whereas AM spore abundance decreased with N fertilization. Spore abundance, morphotype richness and extramatrical hyphal lengths were all greater in monoculture plots. A structural equation model showed AM fungal biovolume was most influenced by CO(2) enrichment, plant community composition and plant richness, whereas spore richness was most influenced by fungal biovolume, plant community composition and plant richness. ? Arbuscular mycorrhizal fungi responded to differences in host community and resource availability, suggesting that mycorrhizal functions, such as carbon sequestration and soil stability, will be affected by global change.  相似文献

Nodulated lupins (Lupinus angustifolius cv. Wonga) were hydroponically grown under conditions of low phosphate (LP) or adequate phosphate (HP) to assess the effect of phosphoenolpyruvate carboxylase (PEPC)-derived organic acids on nitrogen assimilation in LP nodules. LP conditions are linked to altered organic acid metabolism, by the engagement of PEP metabolism via PEPC. In LP nodules, the enhanced organic acid synthesis may reduce the available organic carbon for nitrogen assimilation. The diversion of carbon between the organic acid- and amino acid pools was assessed through key nodular enzymes and (14)CO(2) metabolism. Under LP conditions, increased rates of organic acid synthesis via PEPC and malate dehydrogenase (MDH), coincided with reduced nitrogen assimilation via aspartate aminotransferase (AAT), aspartate synthetase (AS) and glutamine synthetase (GS)/glutamate synthase (GOGAT) activities. There was a preferential metabolism of nodular (14)CO(2) into organic acids and particularly into malate. High malate levels were associated with reduced N(2) fixation and synthesis of amino acids. These results indicate that phosphorus deficiency can enhance malate synthesis in nodules, but that excessive malate accumulation may inhibit N(2) fixation and nitrogen assimilation.  相似文献

The effect of an additional carbon source, lactate, on Penicillium camembertii growth on glutamate as both carbon and nitrogen sources was examined. Glutamate (and lactate) was present in excess in both media. Throughout the whole culture, similar growth time-courses were recorded on both media, indicating the absence of a lactate effect on growth. During the first part of growth, corresponding to an increasing amount of viable biomass, the rate of glutamate consumption remained high, as well as the related ammonium production, indicating its use as a carbon source in addition to being nitrogen source. The low growth rates recorded during the last part of growth resulted in low glutamate consumption, while lactate consumption continued mainly by a maintenance mechanism for the energy supply. A clear differentiation appeared therefore between the carbon source and the energy source: glutamate was mainly used as C source (and N source) for biosynthesis, while lactate was mainly assimilated for energy supply. Carbon and nitrogen yield examinations confirmed this result. Indeed, the C/N ratio found for P. camembertii cellular material (8.14) was about twice that of glutamate (4.29). From this, about half of the available nitrogen was used for biomass formation during growth on glutamate-lactate based medium, as experimentally confirmed (constant yield nitrogen from biomass on nitrogen from glutamate was found (0.49), while the excess nitrogen was released as ammonium). The constant and close to unit (0.99) yield carbon from CO2 on carbon from lactate, also recorded during growth on glutamate-lactate based medium, confirmed that lactate was mainly used as an energy source.  相似文献

* Greater fine-root production under elevated [CO2] may increase the input of carbon (C) and nitrogen (N) to the soil profile because fine root populations turn over quickly in forested ecosystems. * Here, the effect of elevated [CO)] was assessed on root biomass and N inputs at several soil depths by combining a long-term minirhizotron dataset with continuous, root-specific measurements of root mass and [N]. The experiment was conducted in a CO(2)-enriched sweetgum (Liquidambar styraciflua) plantation. * CO2) enrichment had no effect on root tissue density or [N] within a given diameter class. Root biomass production and standing crop were doubled under elevated [CO2]. Though fine-root turnover declined under elevated [CO2], fine-root mortality was also nearly doubled under CO2 enrichment. Over 9 yr, root mortality resulted in 681 g m(-2) of extra C and 9 g m(-2) of extra N input to the soil system under elevated [CO2]. At least half of these inputs were below 30 cm soil depth. * Increased C and N input to the soil under CO2 enrichment, especially below 30 cm depth, might alter soil C storage and N mineralization. Future research should focus on quantifying root decomposition dynamics and C and N mineralization deeper in the soil.  相似文献

The effect on decomposition of 4 different levels of nitrogen in aerial tissue ofSpartina alterniflora, collected at the end of its growing season litter, was studied in laboratory percolators for 56 days at 20C. The CO2 evolution and the release of organic nitrogen and organic carbon were monitored. From these data, the ash-free dry weight (AFDW), nitrogen (N) content, and carbonnitrogen (C/N) ratio were calculated at various times during decomposition. Fungal biomass, bacterial biomass, and the relative autoradiographic activity of bacteria were measured at the end of the study. Decomposition was significantly affected by the nitrogen content of the litter. A 55% increase in plant N increased overall weight loss and k by 50% and 40%, respectively. Furthermore, k (calculated from time course weight loss data) responded linearly to the 4 different levels of nitrogen inSpartina tissue. Fungi appear to dominate the microbial community. At the end of the experiment, fungal biomass was between 2.23 and 3.08% of the AFDW, and was calculated to contain 12 to 22% of the nitrogen in the litter. Bacterial biomass was 1/10 of the fungal biomass, and 12–17% of the bacteria were active. The total microbial biomass was not affected by increased plant nitrogen. In the course of decomposition, the organic nitrogen and carbon were highest in the effluent water in all treatments during the first 8 days. The respiration rate (CO2 evolution) first increased to a maximum at day 18 and then decreased to a constant rate (1–2 mg C/day/g detritus). Respiration was highest in the high N litter. The C/N ratio in all treatments increased from the start to day 8, then decreased to day 20. In low N litter, C/N then increased again as a result of increased total organic nitrogen (TON) loss relative to carbon mineralization. In the high N, this was reversed.  相似文献

Bench-grafted Fuji/M.26 apple (Malus domestica Borkh.) trees were fertigated with different concentrations of nitrogen by using a modified Hoagland's solution for 45 d. CO(2) assimilation and photosystem II (PSII) quantum efficiency in response to incident photon flux density (PFD) were measured simultaneously in recent fully expanded leaves under low O(2) (2%) and saturated CO(2) (1300 micromol mol(-1)) conditions. A single curvilinear relationship was found between true quantum yield for CO(2) assimilation and PSII quantum efficiency for leaves with a wide range of leaf N content. The relationship was linear up to a quantum yield of approximately 0.05 mol CO(2) mol(-1) quanta. It then became curvilinear with a further rise in quantum yield in response to decreasing PFD. This relationship was subsequently used as a calibration curve to assess the rate of non-cyclic electron transport associated with Rubisco and the partitioning of electron flow between CO(2) assimilation and photorespiration in different N leaves in response to intercellular CO(2) concentration (C(i)) under normal O(2) conditions. Both the rate of non-cyclic electron flow and the rate of electron flow to CO(2) or O(2) increased with increasing leaf N at any given C(i). The percentage of non-cyclic electron flow to CO(2) assimilation, however, remained the same regardless of leaf N content. As C(i) increased, the percentage of non-cyclic electron flow to CO(2) assimilation increased. In conclusion, the relationship between PSII quantum efficiency and quantum yield for CO(2) assimilation and the partitioning of electron flow between CO(2) assimilation and photorespiration are not affected by N content in apple leaves.  相似文献

A newly isolated strain of Cunninghamella echinulata grown on glucose produced significant quantities of biomass and cellular lipids in media with high C/N ratio. The oil yield from glucose consumed increased after nitrogen exhaustion in the growth medium, but gamma-linolenic acid (GLA) content in cellular oil systematically decreased during the lipid accumulation process. When lipid accumulation was completed, GLA concentration in the cellular lipids progressively increased. The highest GLA production (720 mg/l) was achieved in medium with a C/N ratio equal to 163. C. echinulata was also able to grow on orange peel. The C/N ratio in the orange peel decreased from 50 to 26 during solid-state fermentation. Maximum oxygen uptake was observed during assimilation of reducing sugars, whereas a polygalacturonase activity was detected after reducing sugars had been exhausted. The maximum GLA production was 1.2-1.5 mg/g of fermented peel, calculated on a dry weight basis. After enrichment of the pulp with inorganic nitrogen and glucose, an increase in the production of oil and GLA was observed.  相似文献

This work describes the long-term acclimation of the halotolerant microalga Dunaliella viridis to different photon irradiance, ranging from darkness to 1500 μmol m−2 s−1. In order to assess the effects of long-term photoinhibition, changes in oxygen production rate, pigment composition, xanthophyll cycle and in vivo chlorophyll fluorescence using the saturating pulse method were measured. Growth rate was maximal at intermediate irradiance (250 and 700 μmol m−2 s−1). The increase in growth irradiance from 700 to 1500 μmol m−2 s−1 did not lead to further significant changes in pigment composition or EPS, indicating saturation in the pigment response to high light. Changes in Photosystem II optimum quantum yield (Fv/Fm) evidenced photoinhibition at 700 and especially at 1500 μmol m−2 s−1. The relation between photosynthetic electron flow rate and photosyntetic O2 evolution was linear for cultures in darkness shifting to curvilinear as growth irradiance increased, suggesting the interference of the energy dissipation processes in oxygen evolution. Carbon assimilation efficiencies were studied in relation to changes in growth rate, internal carbon and nitrogen composition, and organic carbon released to the external medium. All illuminated cultures showed a high capability to maintain a C:N ratio between 6 and 7. The percentage of organic carbon released to the external medium increased to its maximum under high irradiance (1500 μmol m−2 s−1). These results suggest that the release of organic carbon could act as a secondary dissipation process when the xanthophyll cycle is saturated.
