Clearance of bacteria in the bronchoalveolar lavage, the level and functional activity of IgA and changes in the cellular composition of BAL were examined in mice after supralaryngeal immunization and subsequent challenge with Klebsiella pneumoniae. More than 60% of the bacterial inoculum was removed by nonspecific mechanisms within 90 min after inoculation; within the time interval 1.5-3.5 h, clearance was significantly accelerated in locally immunized mice. The enhancement of clearance effectiveness is specific and increases proportionally with the length of immunization (1 less than 2 less than 4 weeks); it is of short duration and towards the end of the 3rd week after immunization, in 73% of immunized animals, the clearance values did not differ from values found in controls. The local immunization did not influence the total level of IgA in BAL, the formation of specific IgA antibody was minimal, in vivo binding of IgA to klebsiella could not be demonstrated. In immunized mice, a significant increase in the numbers of PMN and lymphocytes, as well as an increased activity of phagocytic cell (PMN, MP) was found in BAL. The time interval of 1.5-3.5 h after challenge bounds the space for mechanisms, activated by local immunization in lower airways. The actual participation of individual factors in the accelerated elimination of bacteria from the lumen of airways, remains unclear so far.  相似文献

The evidence base for drug treatment of hypertension is strong. Early trials using thiazide diuretics suggested a shortfall in prevention of coronary heart disease. The superiority of newer drugs has been widely advocated but trial evidence does not support an advantage of beta-blockers, angiotensin converting enzyme inhibitors, calcium channel blockers or alpha-blockers for this outcome. Even meta-analyses have failed to clarify matters. If this issue is to be settled, bigger and better trials of longer duration in high-risk patients are needed. Meanwhile, the importance of rigorous blood pressure control using multiple drugs has been established. This should be the focus of our attention rather than agonising over differences in cause-specific outcomes that may not be generalisable to all patient populations.  相似文献

We constructed a mathematical model to explain from which instars soldiers should be produced to maximize the growth rate of a termite colony. The model is based on the demography of the lower termite's colony: many of them feed inside the nest. The model predicts the following: (1) When the colony is young (it still has enough food and needs a high ratio of soldiers to workers) it will produce soldiers from young instars. (2) When the colony is old (it does not have enough food any more and needs a lower ratio of soldiers to workers) it will produce soldiers from old instars. This prediction fits well with the fact investigated by past empirical researches. With the samples of colonies of Neotermes koshunensis, we measured the antennal joint number of soldiers of each colony. We plotted the antennal joint number (1) of the average and (2) of the maximum one for each colony, against the total number of colony members. For both in each colony, we confirmed the consistency between the model and the samples: as the colony matures, it produces soldiers from older instars. The model also explains why higher termites produce soldiers from fixed instars. Their life style, in which they go outside to collect food, does not require a change in their soldier production strategy.  相似文献

Data from 13 randomised trials on the effect of sodium restriction on blood pressure were analysed. The hypotensive effect of sodium restriction was found to be small and restricted largely to systolic blood pressure, which fell by an average of 3.6 mm Hg (range 0.5-10.0 mm Hg). The reduction increased with age and in those with higher blood pressure. Sodium restriction therefore seems to be of limited use in those who are most eligible for non-pharmacological treatment of high blood pressure--namely, young patients with mild hypertension.  相似文献

β-d-Xylosidase catalyzes hydrolysis of xylooligosaccharides to d-xylose residues. The enzyme, SXA from Selenomonas ruminantium, is the most active catalyst known for the reaction; however, its activity is inhibited by d-xylose and d-glucose (K i values of ~10?2?M). Higher K i’s could enhance enzyme performance in lignocellulose saccharification processes for bioethanol production. We report here the development of a two-tier high-throughput screen where the 1° screen selects for activity (active/inactive screen) and the 2° screen selects for a higher K i(d-xylose) and its subsequent use in screening ~5,900 members of an SXA enzyme library prepared using error-prone PCR. In one variant, termed SXA-C3, K i(d-xylose) is threefold and K i(d-glucose) is twofold that of wild-type SXA. C3 contains four amino acid mutations, and one of these, W145G, is responsible for most of the lost affinity for the monosaccharides. Experiments that probe the active site with ligands that bind only to subsite ?1 or subsite +1 indicate that the changed affinity stems from changed affinity for d-xylose in subsite +1 and not in subsite ?1 of the two-subsite active site. Trp145 is 6 Å from the active site, and its side chain contacts three active-site residues, two in subsite +1 and one in subsite ?1.  相似文献

In adult endotherms—birds and mammals—mortality rates are lower in flying than non-flying species. Rates vary with size, but allowing for this they are higher in penguins than flying birds, and in rodents than bats. These observations are most simply explained in terms of predation since flight, allowing movement in three dimensions, increases the chances of escape.
There are various problems in obtaining and comparing data on rates of mortality and this partly explains a wide scattering of points. Nevertheless the main results are statistically significant, mostly at P<0.001. Amongst birds, there are several other significant differences. Mortality rates are lower at lower latitudes, and in aquatic compared to terrestrial species, with cliff-nesters having lower rates than other aquatic birds and co-operative breeders than other terrestrial ones. No latitudinal effect was detected in mammals. The exceptionally low rates of mortality in bats and swifts are attributed to their being particularly hard to catch.  相似文献

Thermal biofeedback may be a useful adjunctive technique for enhancing cutaneous blood flow in patients with lower-extremity vascular complications of diabetes. However, autonomic, sensory, and/or motor neuropathies may impair vasomotion and limit the ability to alter blood flow and achieve significant foot warming with thermal biofeedback. We examined nerve function associated with four common types of diabetic neuropathy (sympathetic–autonomic, vagal–autonomic, sensory, and motor), hypothesizing that both sympathetic–autonomic and sensory neuropathies would limit the acquisition of biofeedback-mediated foot warming. Twenty-four participants with diabetes mellitus (19 with type II and 5 with type I) received a nerve conduction study and neurological evaluation of the upper and lower extremities. Hand temperature, foot temperature, and electrodermal gradient at the toes were monitored across six thermal biofeedback sessions. Participants were able to significantly raise p < .01) foot temperatures across sessions, an average of 2.2°F. Consistent with our hypotheses, 41% of the variance in foot warming was explained by lower-extremity sympathetic–autonomic and sensory nerve function tests. This study demonstrated that a general diabetic population, including patients with mild-to-moderate neuropathy, can increase skin perfusion with thermal biofeedback. As hypothesized, lower-extremity sympathetic–autonomic and sensory neuropathies interfered with foot warming.  相似文献

Mitochondrial decay plays a central role in the aging process. Although certainly multifactorial in nature, defective operation of the electron transport chain (ETC) constitutes a key mechanism involved in the age-associated loss of mitochondrial energy metabolism. Primarily, mitochondrial dysfunction affects the aging animal by limiting bioenergetic reserve capacity and/or increasing oxidative stress via enhanced electron leakage from the ETC. Even though the important aging characteristics of mitochondrial decay are known, the molecular events underlying inefficient electron flux that ultimately leads to higher superoxide appearance and impaired respiration are not completely understood. This review focuses on the potential role(s) that age-associated destabilization of the macromolecular organization of the ETC (i.e. supercomplexes) may be important for development of the mitochondrial aging phenotype, particularly in post-mitotic tissues.  相似文献

Immunochemical studies with light microscopy, confocal microscopy, and electron microscopy were used to examine proteins associated with caveolin (Cav) in canine lower esophageal sphincter. The main Cav was Cav-1. It appeared to be colocalized at the cell periphery, in punctate sites, with immunoreactivity to antibodies against different COOH- and NH2-terminal epitopes of neuronal nitric oxide (NO) synthase (nNOS). One COOH-terminal-directed antibody, made in guinea pig, was used to colocalize other immunoreactivities. Those that apparently colocalized with nNOS were L-Ca2+ channels, the PM Ca2+ pump, and, in part, calreticulin and calsequestrin. The large-conductance Ca2+-activated K+ (BK(Ca)) channels were located in discrete peripheral sites, some with Cav. Immunoreactivities not fully colocalized with nNOS were to the sarcoplasmic reticulum Ca2+ pump, connexins 43, 40, and 45, and vinculin. In patch-clamp studies, NO-driven outward currents, mainly through BK(Ca) channels, were inhibited by antibodies to Cav-1 and not by calmodulin and were restored by an NO donor. Several Ca2+-handling molecules are localized at the PM with and/or near Cav. This may allow intracellular calcium concentration levels to be controlled differently than those in the cytosol near caveolae.  相似文献

PURPOSE OF REVIEW: Fish oils rich in n-3 fatty acids reduce serum triglyceride levels. This well known effect has been shown to be caused by decreased very low-density lipoprotein triglyceride secretion rates in kinetic studies in humans. Animal studies have explored the biochemical mechanisms underlying this effect. Triglyceride synthesis could be reduced by n-3 fatty acids in three general ways: reduced substrate (i.e. fatty acids) availability, which could be secondary to increase in beta-oxidation, decreased free fatty acids delivery to the liver, decreased hepatic fatty acids synthesis; increased phospholipid synthesis; or decreased activity of triglyceride-synthesizing enzymes (diacylgylcerol acyltranferase or phosphatidic acid phosphohydrolase). RECENT FINDINGS: Rarely were experimental conditions used in rat studies physiologically relevant to the human situation in which 1.2% energy as n-3 fatty acids lowers serum triglyceride levels. Nevertheless, the most consistent effect of n-3 fatty acids feeding in rats is to decrease lipogenesis. Increased beta-oxidation was frequently, but not consistently, reported with similar numbers of studies reporting increased mitochondrial compared with peroxisomal oxidation. Inhibition of triglyceride-synthesizing enzymes was only occasionally noted. SUMMARY: As the vast majority of studies fed unphysiologically high doses of n-3 fatty acids, these findings in rats must be considered tentative, and the mechanism by which n-3 fatty acids reduce triglyceride levels in humans remains speculative.  相似文献

Following over 20 years of research on the climatic effects on biodiversity we now have strong evidence that climate change affects phenology, fitness, and distribution ranges of different taxa, including birds. Bird phenology likely responds to changes in local weather. It is also affected by climatic year‐to‐year variations on larger scales. Although such scale‐related effects are common in ecology, most studies analyzing the effects of climate change were accomplished using climatic information on a single spatial scale. In this study, we aimed at determining the scale‐dependent sensitivity of breeding phenology and success to climate change in a migratory passerine bird, the barn swallow (Hirundo rustica). For both annual broods, we investigated effects of local weather (local scale) and the North Atlantic Oscillation (NAO, large scale) on the timing of breeding and breeding success. Consistent with previous studies in migratory birds we found that barn swallows in Eastern Germany bred progressively earlier. At the same time, they showed reduced breeding success over time in response to recent climatic changes. Responses to climatic variation were observed on both local and large climatic scales, but they differed with respect to the ecological process considered. Specifically, we found that the timing of breeding was primarily influenced by large‐scale NAO variations and to a lesser extent by local weather on the breeding grounds. Conversely, climatic conditions on the local scale affected breeding success, exclusively. The observed decrease in breeding success over years is likely a consequence of scale‐related mismatches between climatic conditions during different breeding phases. This provides further evidence that a species' response of earlier breeding may not be enough to cope with climate change. Our results emphasize the importance of considering the response of ecological processes along different climatic scales in order to better understand the complexity of climate change effects on biodiversity.  相似文献

Preferred room temperatures were determined for 16 winter swimmers who had been exposed to cold shock daily by bathing in the sea (0–6°C) during at least two winter seasons. Each subject participated in one comfort experiment (2 1/2 h) in an environmental test chamber, where the preferred ambient temperature was determined by adjusting it according to his wishes. The subjects were sedentary and clothed in a standard uniform. Skin temperatures, rectal temperature and evaporative weight loss were measured. The results were compared with similar data for two other groups: (a) a control group of 64 normal subjects and (b) 16 persons who had been working 8 h a day at 8°C during at least 6 months in the meat packing industry. The results showed no significant differences between the ambient temperatures preferred by the three groups of subjects. This indicated that man cannot be adapted to prefer lower ambient temperatures by exposing him daily to cold.  相似文献

Service providers and their customers agree on certain quality of service guarantees through Service Level Agreements (SLA). An SLA contains one or more Service Level Objectives (SLO)s that describe the agreed-upon quality requirements at the service level. Translating these SLOs into lower-level policies that can then be used for design and monitoring purposes is a difficult problem. Usually domain experts are involved in this translation that often necessitates application of domain knowledge to this problem. In this article, we propose an approach that combines performance modeling with regression analysis to solve this problem. We demonstrate that our approach is practical and that it can be applied to different n-tier services. Our experiments show that for a typical 3-tier e-commerce application in a virtualized environment, the SLA can be met while improving CPU utilization by up to 3 times.

The ammonoid subfamily Peltoceratinae is a well-constrained group restricted to the Middle Upper Jurassic; it had a near global distribution. In Kutch (India), they were the most important marine macroinvertebrate group during the upper Callovian–lower Oxfordian. Previous reports suffered from excessive splitting due to failure to account for the large intraspecific variability and sexual dimorphism of these ammonoids. The present systematic revision, based on numerous specimens including many near-complete adult specimens, enables to recognize and describe three species of the genus Peltoceras (P. athleta, P. ponderosum, and P. kumagunense) and two species of the genus Peltoceratoides (P. semirugosus and P. propinquus), as well as identifying their macroconchs and microconchs. We rediscovered Peltoceras athleta, which was previously described by Waagen (1875) but often overlooked by subsequent workers. We were able to identify several morphs within this species that are stunningly similar with those previously reported from France. The presence of P. athleta sensu stricto prompted us to reinstate the Athleta Zone in Kutch, improving intercontinental biostratigraphic correlation. Also, it highlights that the genus Peltoceras has a different biostratigraphic longevity in Kutch: in Europe it is restricted to the upper Callovian, whereas in India it also occurs in the lower Oxfordian.  相似文献
