Two new acylated dolabellane-type diterpenes, nigellamines B3 (9) and D (10), were isolated from Nigella sativa (Ranunculaceae) seeds using column chromatography and preparative HPLC. Their structures were determined based on chemical and physicochemical evidence, and confirmed using previously isolated related compounds as reference. Of the seed constituents, nigellamines A2 (2), A3 (3), A5 (5), B1 (6), and B2 (7) had in vitro triglyceride metabolism-promoting activities in the high glucose-pretreated human liver carcinoma cell line, HepG2.  相似文献

Isoniazid-naphthoquinone hybrids were synthesized and evaluated against a susceptible (H37Rv) strain and two isoniazid-resistant strains (INHR1 and INHR2) of Mycobacterium tuberculosis. The antimycobacterial activity of the derivatives was determined based on the resazurin microtiter assay and their cytotoxicity in adhered mouse monocyte macrophage J774.A1 cells (ATCC TIB-67). Of the twenty-two compounds evaluated against the three strains of M. tuberculosis, twenty-one presented some activity against the H37Rv and INHR1 (katG S315T) or INHR2 (inhA C(−5)T) strains. Compounds 1a, 2a, and 8a were effective against the INHR1 strain, and compounds 1a, 1b, 2a, 3a, 5a, 5b and 8a were effective against the INHR2 strain, with MICs in the range of 3.12–6.25 µg/mL. Compounds 1b and 5b were the most active against H37Rv, with MIC of 0.78 µg/mL. Based on the selectivity index, 1b and 5b can be considered safe as a drug candidate compounds. These results demonstrate that quinoidal compounds can be used as promising scaffolds for the development of new anti-TB drugs and hybrids with activity against M. tuberculosis-susceptible and INH-resistant strains.  相似文献

Diverse 2-pyridinyl quinolines 612 and 2-pyridinilvinyl quinolines 1317 were prepared using a straightforward synthesis based on the BiCl3-catalyzed multicomponent imino Diels–Alder (imino DA) reaction or a novel tandem imino DA/catalytic tetrahydroquinoline ring oxidation/Perkin condensation sequential process. All members of the series showed activities against dermatophytes and some of them possessed a broad spectrum of action. 2-(Pyridin-4-yl)quinoline 9 and 2-(2-pyridin-4-yl)vinyl)quinoline 16 showed the best MIC80 and MIC50 against the clinically important fungi Candida albicans and non-albicans Candida species. In turn, 6-ethyl-2-(pyridin-2-yl)quinoline 6 showed the best properties against standardized as well as clinical strains of Cryptococcus neoformans.  相似文献

A convergent strategy was followed to modify systematically carbazole based CB2 receptor ligands. The length of the N-(fluoroalkyl) group (n in 7), the length of the alkanamide (m in 7) and the substitution pattern of the phenyl moiety (X and Y in 7) were varied systematically. The highest CB2 affinity was found for the 2-fluoroethyl substituted carbazole derivative 20a (Ki = 5.8 nM) containing the propionamide and the 2-bromo-4-fluorophenyl moiety. According to docking studies 20a fits nicely into the binding pocket of the CB2 receptor, but elongation of the fluoroethyl side chain leads to a different binding mode of the ligands. The high CB2 affinity together with the high selectivity over the CB2 subtype qualifies the fluoroethyl derivative 20a to be developed as a PET tracer.  相似文献

Novel nonpeptide small molecule renin inhibitors bearing an N-isopropyl P1 motif were designed based on initial lead structures 1 and aliskiren (2). (P3–P1)-Benzamide derivatives such as 9a and 34, as well as the corresponding P1 basic tertiary amine derivatives 10 and 35 were found to display low nanomolar inhibition against human renin in vitro.  相似文献

A new dolabellane-type diterpene alkaloid, deoxynigellamine, together with three known dolabellane-type diterpene alkaloids, were isolated from the seeds of Nigella glandulifera. Three novel coordination polymers with different structural motifs, [Ag(pmtmb)]n (1), [Cd(pmtmb)2(H2O)]n (2) and [Cu(pmtmb)2]n (3), have been synthesized with a flexible asymmetrical bridging ligand, 4-(2-pyrimidylthiomethyl)benzoic acid (Hpmtmb). X-ray diffraction analyses show that 1 is a 2D layer containing unusual zigzag Ag chains based on mixed ligand-supported and ligand-unsupported Ag–Ag interactions, 2 is a necklace structure that further linked through hydrogen bonding to form a 2D sheet, and 3 is a 3D compact framework full assembled from 1D centipede-like chains via π–π stacking interactions. Interestingly, the ligand pmtmb adopts different configurations and coordination modes in the solid structures of these complexes. Furthermore, three complexes exhibit remarkable thermal stability and the complexes 1 and 2 exhibit intense green and purple luminescence, respectively.  相似文献

Phytochemical investigation of the whole plant of Hypericum curvisepalum led to the isolation of nine polycyclic polyprenylated acylphloroglucinols (19). Among them, curvisepalumiones A (1) and B (2) are two new compounds and their structures were elucidated based on extensive spectroscopic data analysis combined with Rh2(OCOCF3)4-induced electronic circular dichroism experiment. Cytotoxic assay showed that all the tested compounds except for 1 was moderately cytotoxic against human hepatocarcinoma cell line SMMC-7721 cell line, while compounds 3 and 4 also exhibited cytotoxicity toward MGC-803 cells. In addition, compound 2 exhibited weak inhibitory activity against Bacillus subtilis.  相似文献

Two new dihydronaphthalenones (1, 2) with four known compounds (36) were isolated as secondary metabolites of the endophytic fungus Nodulisporium sp. from Antidesma ghaesembilla. Their chemical structures were established based on 1D and 2D NMR spectroscopic data and HR-ESI–MS analyses. The relative configuration of 2 was confirmed by single crystal X-ray crystallographic analysis. A new isofuranonaphthalenone 2 displayed antimycobacterial and antimalarial activities with IC50 values of 3.125 and 11.3 μg/mL, respectively.  相似文献

Fourteen novel conjugates of 3,28-di-O-acylbetulins with AZT were prepared as anti-HIV agents, based on our previously reported potent anti-HIV triterpene leads, including 3-O-acyl and 3,28-di-O-acylbetulins. Nine of the conjugates (4953, 55, 56, 59, and 60) exhibited potent anti-HIV activity at the submicromolar level, with EC50 values ranging from 0.040 to 0.098 μM in HIV-1NL4-3 infected MT-4 cells. These compounds were equipotent or more potent than 3-O-(3′,3′-dimethylsuccinyl)betulinic acid (2), which is currently in Phase IIb anti-AIDS clinical trial.  相似文献

Suspension cultures of Abronia nana were established to produce C-methylisoflavones. A new C-methylrotenoid, named abronione A (2), was isolated along with three known rotenoids, boeravinone D (1), boeravinone A methyl ether (3), and mirabijalone D (4). The IC50 values of compounds 1, 2, and 4 on β-secretase (BACE1) were 4.77, 62.21, and 4.24 μM, respectively, whereas 3 was inactive. At concentrations up to 1.0 mM, the compounds did not inhibit other proteases such as trypsin, chymotrypsin, and elastase, indicating that they were specific inhibitors of β-secretase. Compounds 1 and 4 were non-competitive inhibitors based on the Dixon plot and with Ki values of 5.01 and 4.28 μM, respectively. At 50 μM, compound 4 inhibited Aβ1–42 production by 43.7% in APPSW-N2a cells.  相似文献

Using various chromatographic techniques, 23 triterpene saponins (1–23) were isolated from an ethanol extract of Stauntonia hexaphylla, including two new compounds (12 and 15). Their chemical structures were established by comprehensive spectroscopic methods such as 1D- and 2D-NMR, and HR-ESI-MS, and chemical reactions. The anti-inflammatory activities of the isolated saponins were determined using the nitric oxide (NO) assay. Compound 13 exhibited the greatest inhibitory effect (IC50?=?0.59?μM). In addition to NO, compound 13 suppressed the secretion of PGE2, IL-1β, and IL-6, but not TNF-α, and inhibited the protein expression of iNOS and COX-2 in LPS-activated RAW264.7 cells. The chemical derivatives of the isolated compounds were studied using structure–activity relationships. The results suggested that compound 13 isolated from S. hexaphylla might be useful for treating inflammation. This is the first comprehensive study of saponins from the leaves of S. hexaphylla based on anti-inflammatory extract screening guidelines.  相似文献

Complexes [Bi(2AcPh)Cl2]·0.5H2O (1), [Bi(2AcpClPh)Cl2] (2), [Bi(2AcpNO2Ph)Cl2] (3), [Bi(2AcpOHPh)Cl2]·2H2O (4), [Bi(H2BzPh)Cl3]·2H2O (5), [Bi(H2BzpClPh)Cl3] (6), [Bi(2BzpNO2Ph)Cl2]·2H2O (7) and [Bi(H2BzpOHPh)Cl3]·2H2O (8) were obtained with 2-acetylpyridine phenylhydrazone (H2AcPh), its -para-chloro-phenyl- (H2AcpClPh), -para-nitro-phenyl (H2AcpNO2Ph) and -para-hydroxy-phenyl (H2AcpOHPh) derivatives, as well as with the 2-benzoylpyridine phenylhydrazone analogues (H2BzPh, H2BzpClPh, H2BzpNO2Ph, H2BzpOHPh).Upon coordination to bismuth(III) antibacterial activity against Gram-positive and Gram-negative bacterial strains significantly improved except for complex (4).The cytotoxic effects of the compounds under study were evaluated on HL-60, Jurkat and THP-1 leukemia, and on MCF-7 and HCT-116 solid tumor cells, as well as on non-malignant Vero cells. In general, 2-acetylpyridine-derived hydrazones proved to be more potent and more selective as cytotoxic agents than the corresponding 2-benzoylpyridine-derived counterparts.Exposure of HCT-116 cells to H2AcpClPh, H2AcpNO2Ph and complex (3) led to 99% decrease of the clonogenic survival. The IC50 values of these compounds were three-fold smaller when cells were cultured in soft-agar (3D) than when cells were cultured in monolayer (2D), suggesting that they constitute interesting scaffolds, which should be considered in further studies aiming to develop new drug candidates for the treatment of colon cancer.  相似文献

The 70% aqueous methanolic extract of the Chinese plant Aristolochia manshuriensis was found to contain two novel substituted phenanthrene compounds, SCH 546909 (1), and another phenanthrene glycoside (2). The structures of 1 and 2 were established based on NMR studies. They were identified as inhibitors of the CDK2 enzyme. Compound 1 was found to be a potent inhibitor of the CDK2 enzyme with an IC50 of 140 nM, whereas compound 2 was found to be less active with an IC50 of >10 μM.  相似文献

A new class of corticotropin releasing factor 1 (CRF1) receptor antagonists characterized by a tricyclic core ring was designed and synthesized. Novel tricyclic derivatives 2ae were designed as CRF1 receptor antagonists based on conformation analysis of our original 2-anilinobenzimidazole CRF1 receptor antagonist. The synthesized tricyclic derivatives 2ae showed CRF1 receptor binding activity with IC50 values of less than 400?nM, and the 1,2,3,4-tetrahydropyrimido-[1,2-a]benzimidazole derivative 2e was selected as a lead compound with potent in vitro CRF1 receptor binding activity (IC50?=?7.1?nM). To optimize the pharmacokinetic profiles of lead compound 2e, we explored suitable substituents on the 1-position and 6-position, leading to the identification of compound 42c-R, which exhibited potent CRF1 receptor binding activity (IC50?=?58?nM) with good oral bioavailability (F?=?68% in rats). Compound 42c-R exhibited dose-dependent inhibition of [125I]-CRF binding in the frontal cortex (5 and 10?mg/kg, p.o.) as well as suppression of locomotor activation induced by intracerebroventricular administration of CRF in rats (10?mg/kg, p.o.). These results suggest that compound 42c-R successfully binds CRF1 receptors in the brain and exhibits the potential to be further examined for clinical studies.  相似文献

A single-crystal, X-ray diffraction study was performed on a nonalkenic, cyclic trimer (C18H18O9, 4) of levoglucosenone, in order to confirm its chemical structure. Crystals of 4 are orthorhombic, with unit-cell parameters of a = 792.20, b = 1874.35, c = 2383.02 pm, space group P212121, and z = 8. The structure was solved by direct methods, and refined by least-squares to R = 0.032, based on 2990 unique reflections. Each asymmetrical unit contains two symmetry-independent molecules of 4 and one of acetone. The previously assigned chemical structure and stereochemistry of 4 were found to be correct.  相似文献

The crystal structures of the title compounds Sb(C9H6NO)2(S2COC2H5) (1) and Sb(S2COC2H5)3 (2) have been determined by three dimensional X-ray diffraction techniques and refined by a least squares method; final R 0.049 for 2911 reflections [I ? 3σ(I)] for (1) and R 0.047, Rw 0.046 for 846 reflections [I ? 2σ(I)] for (2). Crystals of (1) are triclinic, space group P1, a = 10.825(2), b = 11.131(2), c = 8.911(1) Å, α = 109.45(1), β = 95.92(1) and γ = 93.02(1)° with Z = 2. Crystals of (2) are rhombohedral, space group R3, arhomb = 10.138(3) Å and α = 103.43(2)°. The environment of the Sb atom in (1) is based on a pentagonal bipyramidal geometry consisting of the six donor atoms of the three chelating ligands and a stereochemically active lone-pair of electrons which occupies the remaining axial position. The xanthate ligand chelates the Sb atom almost symmetrically with two long SbS bonds of 3.059(2) and 3.171(2) Å. In contrast the xanthate ligands in (2) chelate the Sb atom with asymmetric SbS bonds of 2.511(2) and 3.002(3) Å.  相似文献

The competitive reactions of mononucleobase cations SP-4-2-[PtCl(9-EtGua)(NH3)(quinoline)]+, 1, and trans-[PtCl(9-EtGua)(pyridine)2]+, 2, with 5′-guanosine monophosphate (5′-GMP) and N-Acetylmethionine (N-AcMet) were studied by 1H NMR Spectroscopy. The results confirmed the previously observed kinetic selectivity for sulfur over nitrogen binding. The symmetric bis(pyridine) complex reacted faster than the ammine/quinoline moiety – the estimated half-times for reaction with 5′-GMP and N-AcMet were, respectively, 7.4 and 2.3 h for 1 and 4.90 and <0.75 h for 2. Thus modification of the planar amine can enhance sulfur selectivity – based on the observed rates a S/N selectivity ratio of 3.2 is obtained for 1 but >6.5 for 2. Applications of these findings were extended to study the reaction of 1 and 2 with Ubiquitin. One principal adduct corresponding to chloride displacement is observed for both species and in this case little difference in rate is observed. The likely binding site is the unique methionine residue. The percentage of platinum-bound ubiquitin is higher for 1 and 2 than the parent dichlorides trans-[PtCl2(NH3)(quinoline)] and trans-[PtCl2(pyridine)2]. The results suggest that systematic ligand modification can modulate sulfur donor specificity and suggest possible structural features for design of platinum-based bifunctional DNA–protein cross-linking agents, rather than the DNA–DNA cross-linking principally adopted by the anticancer drug cisplatin and congeners.  相似文献

Two new ent-kaurane diterpenoids, namely 3α,16α-dihydroxy-ent-kauran-19-oic acid (1) and 3α,9β-dihydroxy-ent-kauran-19-oic acid (2), together with eight known compounds were isolated and identified from the leaves of Sphagneticola trilobata. Their structures were elucidated on the basis of extensive spectroscopic analysis including 1D, 2D NMR and MS techniques. Known compounds 4, 5 and 810 were obtained from S. trilobata for the first time. These compounds were tested for their in vitro α-glucosidase inhibitory activity and their tyrosinase inhibitory potential. Compounds 2, 3 and 6 were found to show in vitro α-glucosidase inhibitory activity with IC50 values ranging from 0.398 to 0.476 mM, which were close or more potent than reference compound acarbose (IC50 0.410 mM). Compounds 2 and 6 were further revealed to show in vitro tyrosinase inhibitory activity (IC50 29.25 and 40.74 μM) but inferior to that of the positive control kojic acid (IC50 12.55 μM).  相似文献

Thiophene bioisosteres of potent GluN2B receptor negative allosteric modulators were prepared and evaluated pharmacologically. The five-step synthesis of 4,5,7,8-tetrahydro[7]annuleno[b]thiophen-6-one (10) was considerably improved by carboxylation of thiophene-3-carboxylic acid (8) in the first reaction step. Reductive amination and alkylation led to three homologous series of secondary and tertiary phenylalkylamines 5, 11 and 12. Metalation, reaction with 1-formylpiperidine and subsequent reduction provided hydroxymethyl derivatives 15 and 16, which had been designed as bioisosteres of phenols. 2-Bromo derivatives 18 were obtained by bromination of ketone 10 with NBS and subsequent reductive amination. High GluN2B affinity was achieved with [7]annuleno[b]thiophenes bearing a 3-phenylpropylamino or 4-phenylbutylamino moiety (e.g. 5c: Ki = 5.9 nM; 11d: Ki = 9.0 nM). Tertiary ethylamines 12 showed lower GluN2B affinity than tertiary methylamines 11 or secondary amines 5 (e.g. 5c: Ki = 5.9 nM; 11c: Ki = 6.0; 12c: Ki = 51 nM). A Br-atom or a hydroxymethyl moiety in 2-position were less tolerated by the GluN2B receptor. Very similar relationships between the structure and GluN2B affinity and structure and σ affinity, in particular σ2 affinity, were detected. A slight preference for the ifenprodil binding site of GluN2B receptors over σ1 and σ2 receptors was found for methylamines 11c (≈2-fold) and 11d (≈1.5–2-fold) as well as for bromo derivative 18c (≈3-fold).  相似文献
