以沙质草地优势物种猪毛蒿、胡枝子和糙隐子草为研究对象,利用开顶式生长室(OTC)模拟增温,研究降水减少20%、40%和60%与增温的交互作用对3种典型植物叶片光合气体交换特征及叶绿素荧光特征的影响,以揭示沙质草地3种优势植物对气候变化的响应规律。结果显示:(1)与自然温度相比,OTC模拟增温增加了猪毛蒿C_(i),显著降低了胡枝子G_(s)、P_(n)和T_(r)、糙隐子草G_(s)和P_(n)、猪毛蒿WUE和L_(s),也显著降低了猪毛蒿和胡枝子F_(v)/F_(m)和F_(v)/F_(o)。(2)无论增温与否,随着降水减少幅度的增加,猪毛蒿G_(s)和P_(n)呈下降趋势,且中度以上的干旱胁迫下(降水减少>40%)胡枝子和糙隐子草P_(n)显著低于对照。(3)在自然温度条件下,轻度干旱胁迫时(降水减少20%)猪毛蒿T_(r)显著低于对照,重度干旱胁迫时(降水减少60%)其WUE、F_(v)/F_(m)和F_(v)/F_(o)显著低于对照;重度干旱胁迫时,胡枝子C_(i)显著高于对照,差异幅度达10.7%,L_(s)显著低于对照,轻度干旱胁迫时(降水减少20%)其F_(v)/F_(m)和F_(v)/F_(o)显著高于中度以上的干旱胁迫;中度以上的干旱胁迫下糙隐子草T_(r)和G_(s)显著低于对照,重度干旱胁迫时,其C_(i)、F_(v)/F_(m)和F_(v)/F_(o)显著低于对照,WUE和L_(s)显著高于对照。(4)增温与降水减少交互作用下,所有处理猪毛蒿C_(i)均高于对照,差异幅度分别达4.5%,6.0%和8.4%;胡枝子T_(r)均显著低于对照,差异幅度达57.8%;重度干旱胁迫时猪毛蒿L_(s)和WUE显著低于对照,糙隐子草F_(v)/F_(m)和F_(v)/F_(o)随降水减少而降低,中度以上的干旱胁迫时其值显著低于对照。(5)相关性分析表明,3个优势物种的P_(n)与F_(v)/F_(m)和F_(v)/F_(o)均呈显著正相关关系,其中猪毛蒿和糙隐子草的P_(n)—F_(v)/F_(m)和P_(n)—F_(v)/F_(o)斜率明显高于胡枝子。研究表明,气候变暖会在一定程度上加剧降水减少对沙质草地3种群落优势物种光合作用的抑制。  相似文献

Two new species of oribatid mites of the genus Scapheremaeus (Oribatida, Cymbaeremaeidae), Scapheremaeus gibbus sp. n. and Scapheremaeus luxtoni sp. n., are described from New Zealand. Scapheremaeus gibbus sp. n. is morphologically most similar to Scapheremaeus humeratus Balogh & Mahunka, 1967, but differs from the latter by the number of notogastral, genital and adanal setae, morphology of bothridial setae, position of adanal lyrifissures and absence of humeral processes. Scapheremaeus luxtoni sp. n. is morphologically most similar to Scapheremaeus yamashitai Aoki, 1970, but differs from the latter by the morphology of notogastral and rostral setae, morphology of leg solenidia φ2 and development of humeral processes. The species Scapheremaeus zephyrus Colloff, 2010 is recorded for the first time in New Zealand. An identification key to the known New Zealand species of Scapheremaeus is provided.  相似文献

The mode of interaction of aqueous dispersions of phospholipid vesicles is investigated. The vesicles (average diameter 950 Å) are prepared from total lipid extracts of Escherichia coli composed of phosphatidylethanolamine, phosphatidylglycerol and cardiolipin. One type of vesicle contains trans-Δ9-octadecenoate, the other type trans-Δ9-hexadecenoate as predominant acyl chain component. The vesicles show order?disorder transitions at transition temperatures, Tt = 42° C and Tt = 29° C, respectively. A mixture of these vesicles is incubated at 45° C and lipid transfer is studied as a function of time using the phase transition as an indicator. The system reveals the following properties: Lipids are transferred between the two vesicle types giving rise to a vesicle population where both lipid components are homogeneously mixed. Lipid transfer is asymmetric, i.e. trans-Δ9-hexadecenoate-containing lipid molecules appear more rapidly in the trans-Δ9-octadecenoate-containing vesicles than vice versa. At a given molar ratio of the two types of vesicles the rate of lipid transfer is independent of the total vesicle concentration. It is concluded that lipid exchange through the water phase by way of single molecules or micelles is the mode of communication of these negatively charged lipid vesicles.  相似文献

The effect of NH4Cl addition on intracellular pH (pH i ) was determined by flow cytometric measurements of the fluorescence of a pH-sensitive dye. The effects of NH4Cl on growth were determined for batch growth of cells in flasks in an incubator. The addition of NH4Cl caused a cytoplasmic acidification. A new lower steady-state value of pH i was attained within 20–40 min of NH4Cl addition. A correlation was found between the effects of NH4Cl on growth and on pH i : whereas 3 mM NH4Cl had little effect on growth and on pH i , 10 mM NH4Cl caused a substantial growth inhibition and a pH i decrease of 0.2–0.3 units. The effects of NH4Cl on growth and on pH i were found to be independent of the external pH value (pH e over the range 6.8 to 7.6, except that 10 mM NH4Cl was more toxic at pH e 7.6. The addition of NH4Cl caused an increase in the average cell volume at pH e 7.6, but had no effect on the average cell volume at pH e 's 6.8 and 7.2. For comparison, the effects of pH e alone on growth and on pH i were determined. There was little difference in cell growth at pH e 's 6.8, 7.2 and 7.6. At pH e 6.6, there was a substantial growth inhibition. Some measurements of the effects of pH e on pH i were made, although the steady-state value of pH i as a function of pH e was not determined due to limitations in the pH i -measuring technique. These measurements showed that pH i remained constant from pH e 7.6 to 6.8, but fell by 0.2 units at pH e 6.6, in agreement with the growth results.  相似文献

The taxonomic position of two antarctic dorylaimid species Amblydorylaimus isokaryon (Loof, 1975) Andrássy, 1998 and Pararhyssocolpus paradoxus (Loof, 1975), gen. n., comb. n. are discussed on the basis of morphological, including SEM study, morphometric, postembryonic and sequence data of 18S rDNA and the D2-D3 expansion fragments of large subunit rDNA. The evolutionary trees inferred from 18S sequences show insufficient resolution to determine the assignment of the two species to particular families, moreover Pararhyssocolpus paradoxus gen. n., comb. n. (=Rhyssocolpus paradoxus) previously regarded as a member of Nordiidae or Qudsianematidae, showed distant relationship both to Rhyssocolpus vinciguerrae and Eudorylaimus spp. The phylogram inferred from 28S sequences revealed that Amblydorylaimus isokaryon is a member of a well-supported group comprised of several Aporcelaimellus spp., while, no close relationships could be revealed for the Pararhyssocolpus paradoxus gen. n., comb. n. to any nematode genus. On the basis of molecular data and morphological characteristics, some taxonomic changes are proposed. Amblydorylaimus isokaryon is transferred from family Qudsianematidae to family Aporcelaimidae, and a new monotypic genus Pararhyssocolpus gen. n. is proposed, attributed to Pararhyssocolpidae fam. n. The diagnosis of the new family is provided together with emended diagnosis of the genera Amblydorylaimus and Pararhyssocolpus gen. n. Data concerning distribution of these endemic genera in the Antarctic region are also given.  相似文献

Seaweeds have been focused as potential and promising resources to develop novel pharmaceuticals. The present study was aimed to investigate the bioactive principles of Sargassum crassifolium (S. crassifolium) through organic solvents methanol and petroleum ether extractions individually. The present study also extended to determine the antibacterial potentiality of the bioactive principles from methanolic extract (ME) and petroleum ether extract (PEE) of S. crassifolium against a set of human pathogenic bacteria. Gas chromatography-mass spectrometry (GC–MS) and Fourier transform infrared spectroscopy (FT-IR) analysis of the ME and PEE were exhibiting unique bioactive constituents. The antibacterial effect of ME and PEE were showed the moderate spectrum of activity when compared to the standard streptomycin disc against the screened human pathogenic bacteria. The bacterial sensitivity to the ME was sequenced as Bacillus subtilis > Pseudomonas aeruginosa > Escherichia coli > Klebsiella pneumoniae > Staphylococcus aureus > Streptococcus pyogenes. Furthermore, the spectrum of activity of PEE was showing more or less similar pattern of action with almost equal potency. The spectrum of activity of PEE extract was in the order Bacillus subtilis > Pseudomonas aeruginosa > Escherichia coli > Staphylococcus aureus > Streptococcus pyogenes > Klebsiella pneumoniae.  相似文献

Experiments with intact plants of Lolium perenne previously grown with 14NO3 revealed significant efflux of this isotopic species when the plants were transferred to solutions of highly enriched 15NO3. The exuded 14NO3 was subsequently reabsorbed when the ambient solutions were not replaced. When they were frequently replaced, continual efflux of the 14NO3 was observed. Influx of 15NO3 was significantly greater than influx of 14NO3 from solutions of identical NO3 concentration. Transferring plants to 14NO3 solutions after a six-hour period in 15NO3 resulted in efflux of the latter. Presence of Mg2+, rather than Ca2+, in the ambient 15NO3 solution resulted in a decidedly increased rate of 14NO3 efflux and a slight but significant increase in 15NO3 influx. Accordingly, net NO3 influx was slightly depressed. A model in accordance with these observations is presented; its essential features include a passive bidirectional pathway, an active uptake mechanism, and a pathway for recycling of endogenous NO3 within unstirred layers from the passive pathway to the active uptake site.  相似文献

Many species in the families of Rosaceae, Solanaceae, and Scrophulariaceae exhibit gametophytic self-incompatibility, a phenomenon controlled by two polymorphic genes at the S-locus, style-S (S-RNase) and pollen-S (SFB). Sequences of both genes show high levels of diversity, characteristic of genes involved in recognition of self-incompatibility systems in plants. In this study, S 24 -RNase and SFB 24 alleles were cloned from Prunus armeniaca cv. Chuanzhihong (Chinese apricot). Sequence comparisons of deduced amino acid sequences revealed that the P. armeniaca S 24 -haplotype has different SFB alleles, but shares a single S-RNase allele with P. armeniaca S 4 -haplotype. Moreover, P. armeniaca S 24 -RNase haplotype has a single and three different alleles with S 1 -RNase of P. tenella (dwarf almond) and S 1 -RNase of P. mira (smooth pit peach), respectively. The functionalities of SFB 24 and SFB 4 have been evaluated by pollen tube growth and controlled field tests of P. tenella and P. mira. Genetic analysis of the two intercrosses showed that progenies segregated 1:1 into two S-genotype classes, which is consistent with the expected ratio for semi-compatibility. These findings imply that the allelic function of the S 24 -haplotype is identical to that of the S 4 -haplotype in a self-incompatibility reaction. Thus, these two Prunus S-haplotypes are in fact two neutral variants of the same S-haplotype. The evolution of the S-allele is also discussed in terms of both functions and differences between S 24 - and S 4 -haplotypes in Prunus.  相似文献

Two new species of Mongolbittacus Petrulevičius, Huang & Ren, 2007, Mongolbittacus speciosus sp. n. and Mongolbittacus oligophlebius sp. n., and two new species of Exilibittacus Yang, Ren & Shih, 2012, Exilibittacus foliaceus sp. n. and Exilibittacus plagioneurus sp. n., in the family Bittacidae, are described and illustrated based on five well-preserved fossil specimens. These specimens were collected from the late Middle Jurassic Jiulongshan Formation of Daohugou, Inner Mongolia, China. These new findings enhance our understanding of the morphological characters of early hangingflies and highlight the diversity of bittacids in the Mid Mesozoic ecosystems.  相似文献

We conducted a molecular analysis of the Vrn-B1 gene in two near-isogenic lines (NILs) carrying the dominant Vrn-B1 S and Vrn-B1 Dm alleles from the Saratovskaya 29 and Diamant 2 cultivars, respectively. These lines are characterized by different times of ear emergence. PCR analysis and subsequent sequencing of the regulatory regions of Vrn-B1 revealed the full identity of the promoter region in both alleles. Simultaneously, we found significant differences in the structure of the first intron of the Vrn-B1 S allele when compared to Vrn-B1 Dm ; specifically, the deletion of 0.8 kb coupled with the duplication of 0.4 kb. We suggest that these changes in intron 1 of Vrn-B1 S caused earlier ear emergence in the corresponding NIL. The unusual structure of intron 1 within the Vrn-B1 S allele was described for the first time in this study. The allele Vrn-B1 Dm was almost identical with the previously studied sequence of the Vrn-B1a allele of T. aestivum, Triple Dirk B. We designated the new Vrn-B1 S allele as Vrn-B1c. PCR analysis of the Vrn-B1 gene in 26 spring wheat cultivars of both Russian and foreign breeding revealed that 16 of them contain the Vrn-B1a allele and 6 contain the Vrn-B1c allele. Other cultivars studied contained the recessive vrn-B1 gene, except for Novosibirskaya 67. This study demonstrates that the traditional system of Vrn-1 markers does not fully encompass the allelic diversity of these genes because none of the cultivars containing the Vrn-B1c allele gave a PCR product using the previously developed set of primers for identification of the Vrn-B1 locus. We showed that the newly characterized Vrn-B1c allele is widely distributed among different genotypes of spring wheat. The findings indicate the impact of structural changes in the first intron of Vrn-1 on the vernalization response and heading time.  相似文献

A new method determining the activity of tannin acyl hydrolase (tannase) was made. This method was based on the change in optical density of substrate tannic acid at 310 mμ. In this method, the error of measurement was about 1~3%, and many samples could be tested at one time because of its simplicity.

The procedure was as follows; To four parts of substrate (0.350 w/v% of tannic acid dissolved in 0.05m citrate buffer, pH 5.5), one part of the enzyme solution was added.

After t minutes reaction at 30°C, 0.1 part of the mixture was added to ten parts of 90% ethanol.

The optical density of the ethanol solution at 310 mμ was measured. Tannase activity (unit/ml) was given by following equation. u=114×Et1?Et2t2?t1

Where Et1 and Et2 mean the optical density of the ethanol solution at 310 mμ prepared after t1 and t2 minutes reaction, and one unit of the enzyme means the amount of the enzyme which is able to hydrolyze one μ mole of the ester bond in tannic acid in one minute.

The substrate tannic acid used in this determining method was purified. It was composed of one mole of glucose and nine moles of gallic acid, and eight moles of which formed four moles of m-digallic acid.  相似文献

Antagonistic capability of Trichoderma harzianum was improved through UV-irradiation. Four different type of mutants, T. harzianum - Ma (Th-Ma), T. harzianum - Mb(Th-Mb), T. harzianum - Mc (Th-Mc), T. harzianum - Md (Th-Md) of T. harzianum and the parent strain (Th-P) were selected for further studies. Th-Ma and Th-Mb showed more antagonistic capability against Macrophomina phaseolina than its parent strain Th-P in dual culture. Biochemical analysis of these four mutants and the parent strain showed that Th-Ma releases higher level of two lytic enzymes i.e. chitinases and cellulases and Th-Mb produces more β-1,3-glucanase activity than the parent strain. Culture filtrate of Th-Ma also showed antifungal properties. Study of the competitive saprophytic ability (CSA) of these four mutants and the parent strain were also made. Th-Ma exhibited higher CSA than the parental isolate while Th-Md had less CSA than all other mutants and the parent strain of T. harzianum.  相似文献

1. Evolutionary changes in the structure of an enzyme that provide an increase in its Km value are considered. Provided that Km increases as a result of increases in the forward rate constants of the catalysis relative to the reverse rate constants, the enzyme catalyses the conversion of a fixed concentration of its substrate more rapidly when its structure provides that Km>[S] than when Km<[S]. 2. Catalytic efficiency of enzymes is discussed in terms of the simplest plausible model, the Haldane [(1930) Enzymes, Longmans, London] reversible three-step model: [Formula: see text] The rate equation for the forward reaction of this model (formation of P) may be written in the simple form: [Formula: see text] Keq. is the equilibrium constant (=[P]eq./[S]eq.), and kcat.=V/[E]T, where [E]T is the total enzyme concentration. 3. To assess the effectiveness of an enzyme, it is necessary only to determine the extent to which the constraints of a particular kinetic mechanism permit v2 (v when Km»[S]) to approach vd (the diffusion-limited rate). 4. The value of the optimal rate of catalysis (vopt., the maximal value of v2) is dictated by the equilibrium constant for the reaction, Keq.; v2=vd/a, where [Formula: see text] when k+1 is assumed equal to k−3, and vopt.=vd/amin.. When Keq.≥1, it is necessary that k+2»k−1 for a to take its minimum value, amin.; when Keq.«1, it is necessary only that k+2»Keq.·k−1, i.e. a can equal amin. even if k+2<k−1. When Keq.»1, vopt.=vd; when Keq.=1, vopt.=vd/2, and when Keq.«1, vopt.=Keq.·vd. 5. The analysis, together with predicted effects of evolutionary pressure, suggests that in practice the rates of the fastest enzyme-catalysed freely reversible reactions might be expected to be lower than the value of k+1[E]T[S] by about an order of magnitude, particularly if Keq.<1. 6. The existing literature suggests that, in general, appropriate values of Km have evolved for the provision of high rates of catalysis but that many values of kcat. are not large enough to provide optimal rates of catalysis unless the value of k+1 in vivo is lower than its value in free solution.  相似文献

Chitosan (Ch) was chemically modified with ethylenesulfide (Es) under solvent-free conditions to give (ChEs), displaying a high content of thiol groups due to opening of the three member cyclic reagent. Elemental analysis showed a decrease in nitrogen content. This result indicated the incorporation of two ethylenesulfide molecules for each unit of the polymeric structure of the precursor biopolymer. Infrared spectroscopy, thermogravimetry, and 13C NMR in the solid state demonstrated the effectiveness of the reaction, with signals at 30 ppm for ChEs due to the change in the methylene group environment. Divalent metal uptake by chemically modified biopolymer gave the order Cu > Ni > Co > Zn, reflecting the corresponding acidity of these cations in bonding to the sulfur and the basic nitrogen atoms available on the pendant chains. The equilibrium data were fitted to Freundlich, Temkin, and Langmuir models. The maximum monolayer adsorption capacity for the cations was found to be 1.54 ± 0.02, 1.25 ± 0.03, 1.13 ± 0.01, and 0.83 ± 0.03 mmol g−1, respectively. The Langmuir model best explained the cation–sulfur bond interactions at the solid–liquid interface. The thermodynamics for these interactions gave exothermic enthalpic values of −43.02 ± 0.03, −28.72 ± 0.02, −26.27 ± 0.04, and −17.32 ± 0.02 kJ mol−1, respectively. The spontaneity of the systems is given by negative Gibbs free energies of −31.2 ± 0.1, −32.7 ± 0.1, −31.7 ± 0.1, and −32.2 ± 0.1 kJ mol−1, respectively, in spite of the unfavorable negative entropic values of −39 ± 1, −13 ± 1, −18 ± 1, and −49 ± 1 J K−1 mol−1 due to solvent ordering in the course of complexation. This newly synthesized biopolymer is presented as a chemically useful material for cation removal from aqueous solution.  相似文献

Methods of estimating within- and between-population gene diversity in parthenogenetic species using mini- and microsatellite DNA markers and modified Wright's F ST statistic are presented with special reference to model populations of lizards of the genus Darevskia(D. dahli, D. armeniaca, D. unisexualis). We used DNA fingerprinting data for several populations of these species examined earlier. The effects of variation in M13 minisatellite, (GACA) n - and (TCC) n -microsatellite loci on the formation of within-population gene diversity in parthenogenetic species D. dahli and D. armeniaca were shown to be different. The equality of the realized gene diversity Hand its maximum possible value H max in two populations of D. dahli (H max = 0.032, H = 0.031, P 0.0431; H max = 0.024, H = 0.027, P = 0.09) and D. armeniaca (H max = 0.05, H = 0.053, P = 0.03; H max= 0.054, H = 0.055, P= 0.02) suggests that variation in (GACA) n loci substantially contributes to the maintenance of within-population genetic diversity. Analysis of between-population genetic diversity using loci M13, (GACA) n , and (TCC) n showed differentiation of D. dahli populations from northeastern and northwestern Armenia (F ST = 0.0272, P = 3 × 10–13) and genetic homogeneity of the Armenian and introduced to the Ukraine populations of D. armeniaca characteristic of one clone (F ST = 0, P = 1).  相似文献

Consider the family Gn of all n neuron networks whose dynamical behaviors are described by Caianiello's neuronic equations, and also its subfamily Rn of all reverberating networks each of whose neuronic equations have only periodic solution (states), i.e., without having any transient states. This paper is specifically concerned with characterizations of the subfamily Rn. First, we show that Rn is contained in a subfamily Sn of Gn consisting of all self-dual networks. We introduce Chow's matrix corresponding to each network of Sn, using Chow parameters and some algebraic operations {2α} applied to the coefficient matrix of the network in Sn, such as interchanges of coefficients between two neurons or changes of their signs. Then we give some necessary conditions on Chow's matrix under which any network in Sn belong to Rn, and a necessary and sufficient condition on the coefficient matrix. We also discuss relations between Chow's matrix and the maximum period of reverberations. In particular, it is shown that Chow's matrix of a network in Rn is symmetric if and only if the maximum period of reverberations is less than three. By virtue of these results, we propose two methods of construction of networks in Rn. The first method is an inductive construction. The second is based on the algebraic operations.  相似文献

Current knowledge about the abundance, growth, and primary production of the seagrass Cymodocea nodosa (Ucria) Ascherson is biased towards shallow (depth <3 m) meadows although this species also forms extensive meadows at larger depths along the coastlines. The biomass and primary production of a C. nodosa meadow located at a depth of 8–11 m was estimated at the time of maximum annual vegetative development (summer) using reconstruction techniques, and compared with those available from shallow meadows of this species. A depth-referenced data base of values at the time of maximum annual development was compiled to that end. The vegetative development of C. nodosa at 8–11 m depth was not different from that achieved by shallow (depth <3 m) meadows of this species. Only shoot density, which decreased from 1637 to 605 shoots m−2, and the annual rate of elongation of the horizontal rhizome, which increased from 23 to 71 cm apex−1 year−1, were different as depth increased from <3 to 8–11 m. Depth was a poor predictor of the vegetative development and primary production of C. nodosa. The biomass of rhizomes and roots decreased with depth (g DW m−2 = 480 (±53, S.E.) − 32 (±15, S.E.) depth (in m); R2 = 0.12, F = 4.65, d.f. = 35, P = 0.0381) which made total biomass of the meadow to show a trend of decrease with depth but the variance of biomass data explained by depth was low. The annual rate of elongation of the horizontal rhizome showed a significant positive relationship with depth (cm apex−1 year−1 = 18 (±5.1, S.E.) + 5.0 (±1.33, S.E.) depth (in m); R2 = 0.50, F = 14.07, d.f. = 14, P = 0.0021). As shoot size and growth did not change significantly with depth, the reduction of shoot density should drive any changes of biomass and productivity of C. nodosa as depth increases. The processes by which this reduction of C. nodosa abundance with depth occur remain to be elucidated.  相似文献
