The pleurocarpus feather moss, Hylocomium splendens, is one of two co-dominant moss species in boreal forest ecosystems and one of the most common mosses on earth, yet little is known regarding its capacity to host cyanobacterial associates and thus contribute total ecosystem N. In these studies, we evaluated the N-fixation potential of the H. splendens–cyanobacteria association and contrasted the N-fixation activity with that of the putative N-fixing moss–cyanobacteria association of Pleurozium schreberi. Studies were conducted to: quantify N-fixation in H. splendens and P. schreberi in sites ranging from southern to northern Fennoscandia; assess N and P availability as drivers of N-fixation rates; contrast season-long N-fixation rates for both mosses; and characterize the cyanobacteria that colonize shoots of H. splendens. Nitrogen-fixation rates were generally low at southern latitudes and higher at northern latitudes (64–69°N) potentially related to anthropogenic N deposition across this gradient. Nitrogen fixation in H. splendens appeared to be less sensitive to N deposition than P. schreberi. The season-long assessment of N-fixation rates at a mixed feather moss site in northern Sweden showed that H. splendens fixed a substantial quantity of N, but about 50% less total N compared to the contribution from P. schreberi. In total, both species provided 1.6 kg fixed N ha−1 year−1. Interestingly, H. splendens demonstrated somewhat higher N-fixation rates at high fertility sites compared to P. schreberi. Nostoc spp. and Stigonema spp. were the primary cyanobacteria found to colonize H. splendens and P. schreberi. These results suggest that H. splendens with associated Nostoc or Stigonema communities contributes a significant quantity of N to boreal forest ecosystems, but the contribution is subordinate to that of P. schreberi at northern latitudes. Epiphytic cyanobacteria are likely a key factor determining the co-dominant presence of these two feather mosses across the boreal biome.  相似文献

Traditionally it has been thought that most boreal forest communities lack a significant input of biologically fixed nitrogen. Recent discoveries of nitrogen fixation by cyanobacteria associated with mosses have resulted in a re-evaluation of this view. While it is recognized that rates of nitrogen fixation in mosses can be highly variable, there is little understanding as to why this occurs. I monitored nitrogen fixation, using acetylene reduction, in wet lowland and dry upland boreal forest communities, in central Canada, over a growing season. At the peak of nitrogen fixation in mid summer, Sphagnum capillifolium had an 11 times higher rate of fixation than Pleurozium schreberi. Variation in canopy openness and precipitation had no effect on rates of fixation over the growing season. In P. schreberi fixation rates did not vary between sites. Temperature had a positive effect on fixation rates in both S. capillifolium and P. schreberi, but the effect was 4 times more pronounced in S. capillifolium. Seasonal rates of nitrogen fixation were estimated at 193 mg N m−2 for S. capillifolium and 23 mg N m−2 for P. schreberi. With moderate increases in climate warming, predicted increases in nitrogen fixation in S. capillifolium are sufficient to raise its decomposition rate. Increased temperatures may therefore act synergistically to change boreal systems from a sink to a source of carbon.  相似文献

In vitro cultures of Berberis buxifolia were established using thidiazuron (4.5, 23 and 45 mM) or picloram (4 and 40 mM) as plant growth regulators for sustaining growth. For producing berberine, a two-stage culture was performed. In the first step, thidiazuron or picloram were used for biomass production followed by the production stage where benzylaminopurine (4.4 mM) was added as a plant growth regulator. Berberine yields (102 mg g−1 DW) and in vitro shoot cultures (200 mg g−1 DW) were significantly lower than those of whole plants in the field (416 mg g−1 DW). The highest productivity (0.18 mg 1−1 day−1) was attained using picloram (either 4 on 40 mM) in the first stage for producing biomass.  相似文献

Rapid growth of ground vegetation following clear-cutting is important to site productivity because vegetation retains nutrients in the ecosystem and can decrease nutrient leaching prior to stand re-establishment. Aboveground biomass, nutrient contents (N, P, K and Ca) and species composition of ground vegetation were determined 1 year before and for 7 years after clear-cutting of a mixed forest dominated by Norway spruce [Picea abies (L.) H. Karst.] in eastern Finland. The biomass of the feather mosses [Pleurozium schreberi Brid. and Hylocomium splendens (Hedw.) B. S.& G.] and the dwarf shrubs (Vaccinium myrtillus L. and V. vitis-idaea L.), which had dominated the ground vegetation in the mature forest, significantly decreased after clear-cutting. However, with the exception of H. splendens, these species had recovered within 3–5 years. The biomass of Deschampsia flexuosa (L.) Trin. considerably increased soon after clear-cutting, and Epilobium angustifolium L. appeared 3–5 years after cutting. These species contributed to the retention of nutrients not simply because of their biomass but also because of higher nutrient concentrations in their tissues. Total biomass and nutrient contents of the ground vegetation exceeded those of the pre-cutting levels. The proportion of ground vegetation biomass and nutrient contents represented by mosses decreased after cutting, while V. myrtillus, although reduced after cutting, remained a marked nutrient sink. The results suggest that H. splendens is the most sensitive species to cutting, but the biomass of P. schreberi, V. myrtillus and V. vitis-idaea return to initial levels soon after clear-cutting as do the nutrient contents of ground vegetation.  相似文献

Expansion of Phragmites australis (Cav.) Trin. ex Steud. (common reed) into stands of Typha spp. (cattail; Typha australis L. and T. x glauca) is common in the wetlands of northwestern Indiana (USA). To understand this phenomenon better, we investigated the production of shoot sprouts and proportional allocation of biomass as well as a potential role for the water table in the relative dominance of each species. The reduction in sprouts from rhizomes upon vegetative expansion of Phragmites appeared to be the most likely process causing the decline of Typha. The latter had a shoot density of 39/m2 in plots without Phragmites, but this dropped to 13 shoots m−2 in plots that had been invaded by Phramites. Such a decline was likely caused by reduced reserves; e.g., the belowground biomass of Typha decreased from 11.3 g m−2 without Phragmites to 8.1 g m−2 with Phragmites. The latter also reduced its belowground biomass but not its shoot density in the presence of Typha. The mean weight of Phragmites shoots was 2.9 g, and nearly all produced inflorescences. Meanwhile, Typha failed to develop spadices despite its shoots having a greater biomass (7 g). This suggests that Phragmites is more efficient than Typha in shoot growth. Springtime flooding appeared to promote the sprout of Typha shoots from shallow rhizomes (≈18 cm below the soil surface), whereas the shoot density of Phragmites showed no correlation with water level in that season. Deep-rooted Phragmites (≈39 cm) occurred on both high and low water-table sites, whereas the shallow-rooted Typha was limited to only the former. Phragmites will likely continue its expansion, by vegetative sprouts from rhizomes, into Typha wetlands.  相似文献

Abstract

Growth rates of the moss species Hylocomium splendens, Pleurozium schreberi, Hypnum cupressiforme, Ctenidium molluscum and Rhytidium rugosum were determined in terms of dry matter production and stem elongation. Sample sites were chosen to obtain a wide variety of altitudinal and climatic conditions in Austria. The annual increases were detected by using modified Petri dishes, external markers and morphological criteria. Individual shoots of H. splendens and P. schreberi showed a significant decrease of biomass increment with rising altitudes. Measurements of biomass and annual production per area of populations at various altitudes showed a reverse pattern. Central European individuals of H. splendens on average produced 15% more biomass than populations from Northern Europe. There is a strong correlation between stem elongation and temperature for all investigated mosses except R. rugosum. C. molluscum and R. rugosum showed maximum growth rates in shaded sites, with fairly high humidity, although their main occurrence in Austria is in open, mainly dry stands.  相似文献

The response patterns of net photosynthesis to moisture level of mosses in xeric habitats were compared with those in mesic habitats, in order to determine whether the former species are better adapted to the xeric condition with regard to carbon gain. Moss species examined wereRhacomitrium lanuginosum andR. barbuloides in xeric open habitats andDicranum japonicum, Hypnum plicatulum, Ptilium crista-castrensis, Pleurozium schreberi andHylocomium splendens in mesic habitats on the coniferous forest floor in the upper subalpine zone of Mt. Fuji. Three additional xerophytic species collected at other localities,Ptychomitrium polyphylloides, Grimmia pilifera andHedwigia ciliata, were also examined. Five species in the xeric habitats showed an optimum range of moisture level for net photosynthesis, 2 to 3g·g−1. On the other hand, species in the forest showed a wider optimum range, 3 to 8g·g−1. Net photosynthetic rate at the moisture level of 0.5g·g−1 was positive in xerophytic mosses, but negative in most forest mosses. Moisture levels where external capillary water disappeared and drop of water potential began was determined by blotting water-saturated shoots with membrane filters. These moisture levels were low in the xerophytic mosses and high in the forest mosses, although there were some exceptions. It was concluded that mosses in xeric habitats are better adapted for the efficient use of water for photosynthesis than those in mesic habitats.  相似文献

Aquatic carnivorous plants usually grow in shallow dystrophic waters poor in inorganic N and P. Utricularia australis was chosen as a model plant for its prolific distribution and great ecological plasticity. The photosynthetic CO2 compensation point and factors associated with investment in carnivory and capture of prey were measured in 17 U. australis micropopulations in Třeboň basin, Czech Republic, together with water chemistry factors at these sites differing greatly in their trophic level, water hardness, and prey availability. Apical shoot growth rate was estimated at some oligotrophic sites. The micropopulations differed greatly in the proportion of traps with animal prey (2.7–70%, mean 26%), trap proportion to total biomass (1.4–42%, mean 26%), mean trap biomass (0.7–63 μg trap−1, mean 19 μg), and maximum trap size (1–3 mm, mean 2.0 mm). CO2 compensation points ranged from 0.7 to 6.1 μM (mean 2.6 μM). A weak HCO3 use (compensation point 0.51 mM) was found in plants growing in alkaline water. Trap biomass proportion did not correlate significantly with prey capture and CO2 compensation points with ambient [CO2]. A very rapid apical growth (2.5–4.2 new nodes day−1) occurred in sand pits. Thus, HCO3 use in U. australis can be induced by growing at very high pH. CO2 compensation points resembled those known in other aquatic non-carnivorous plants. They did not reflect carnivory. In spite of very rapid apical shoot growth, the relative growth rate of U. australis can be zero in oligotrophic habitats without prey.  相似文献

Efficient plant regeneration via shoot tip provided a basis for the optimization of the genetic transformation protocol. Therefore, experiments were conducted to establish an efficient in vitro regeneration protocol in summer squash for genetic co-transformation. 6-benzylaminopurine at 0.05 mg l−l was found to be optimum concentration of direct regeneration from shoot tip. Effective root system was induced in shootlets in indole-3-aceticacid 0.5 mg l−l. Two vectors namely pCAMBIA 2200 harboring marker gene nptII and pCAMBIA 0390 harboring gene, encoding C-repeat binding factor (cbf1) were used for co-transformation taking shoot tips as explants from in vitro germinated seeds. Explants were selected after co-cultivation on kanamycin supplemented medium and shoots and roots were induced. The transgenic plants were confirmed by polymerase chain reaction (PCR) and further southern blot analysis confirmed the integration of nptII and cbf1 genes in genome of summer squash with co-transformation efficiency of 0.7 percent.  相似文献

In order to determine the seasonal growth and biomass ofTrapa japonica Flerov, field observations were carried out at Ojaga-ike Pond, Chiba, Japan, during 1979 and 1980. In spring, the plant showed exponential growth (c. 0.080 g g−1 day−1) and shoot elongation was as rapid as 10 cm day−1. The plant attained its maximum biomass (380.5±35.1 g m−2) in late August, and about 50% of this was concentrated in the topmost 30-cm stratum (645.7±33.1 g m−3); maximum total stem length exceeded 6m. The plant produced large (500–800 mg per fruit), but small numbers of nut-like fruit (maximum, 5 fruits per rosette). Defoliation occurred almost linearly with time at a rate of 30.6 leaves m−2 day−1; annual net leaf production was estimated to be about twice as large as the seasonal maximum leaf biomass. While the number of leaves per rosette showed moderate seasonal change, rosette density, rosette area and leaf dry weight changed considerably during the year. From the negative log-log correlation between mean total leaf dry weight per rosette and rosette density, density-dependent rosette growth was assumed. The cause of the wide spread of this species in aquatic habitats is briefly discussed in terms of its seed size and morphology.  相似文献

Quasi steady state growth of Lactococcus lactis IL 1403 was studied in glucose-limited A-stat cultivation experiments with acceleration rates (a) from 0.003 to 0.06 h−2 after initial stabilization of the cultures in chemostat at D = 0.2–0.3 h−1. It was shown that the high limit of quasi steady state growth rate depended on the acceleration rate used—at an acceleration rate 0.003 h−2 the quasi steady state growth was observed until μ crit = 0.59 h−1, which is also the μ max value for the culture. Lower values of μ crit were observed at higher acceleration rates. The steady state growth of bacteria stabilized at dilution rate 0.2 h−1 was immediately disrupted after initiating acceleration at the highest acceleration rate studied—0.06 h−2. Observation was made that differences [Δ(μ − D)] of the specific growth rates from pre-programmed dilution rates were the lowest using an acceleration rate of 0.003 h−2 (< 4% of preset changing growth rate). The adaptability of cells to follow preprogrammed growth rate was found to decrease with increasing dilution rate—it was shown that lower acceleration rates should be applied at higher growth rates to maintain the culture in the quasi steady state. The critical specific growth rate and the biomass yields based on glucose consumption were higher if the medium contained S 0 = 5 g L−1 glucose instead of S 0 = 10 g L−1. It was assumed that this was due to the inhibitory effect of lactate accumulating at higher concentrations in the latter cultures. Parallel A-stat experiments at the same acceleration and dilution rates showed good reproducibility—Δ(μ − D) was less than 5%, standard deviations of biomass yields per ATP produced (Y ATP), and biomass yields per glucose consumed (Y XS) were less than 15%.  相似文献

Summary The proliferation and survival of avocado nodal cultures of juvenile origin were affected by the form and concentration of nitrogen. Optimum growth was achieved on modified Murashige and Skoog medium containing 67% KNO3 and 33% NH4NO3 with total N of 40 mM supplemented with 100 mg l−1 myo-inositol, 1 mg l−1 thiamine HCl, 30 g l−1 sucrose, and 4.44 μM BA with a 16-h photoperiod (120–150 μmol m−2 s−1). Proliferating shoots and plantlets were photosynthetically active. Better shoot growth and accumulation of higher biomass occurred in a CO2-enriched environment than under ambient CO2 conditions. CO2 assimilation efficiency, however, was higher under the latter conditions than in a CO2-enhanced environment, e.g., 31±7 and 17±2 μmol CO2 m−2 s−1, respectively. The net CO2 assimilation rates of in vitro grown plantlets were comparable to those of seedlings ex vitro.  相似文献

Nodulation and the subsequent nitrogen fixation are important factors that determine the productivity of legumes. The beneficial effects of nodulation can be enhanced when rhizobial inoculation is combined with plant-growth-promoting bacteria (PGPB). The PGPB strain Bacillus thuringiensis-KR1, originally isolated from the nodules of Kudzu vine (Pueraria thunbergiana), was found to promote plant growth of field pea (Pisum sativum L.) and lentil (Lens culinaris L.) under Jensen’s tube, growth pouch and non-sterile soil, respectively, when co-inoculated with Rhizobium leguminosarum-PR1. Coinoculation with B. thuringiensis-KR1 (at a cell density of 106 c.f.u. ml−1) provided the highest and most consistent increase in nodule number, shoot weight, root weight, and total biomass, over rhizobial inoculation alone. The enhancement in nodulation due to coinoculation was 84.6 and 73.3% in pea and lentil respectively compared to R. leguminosarum-PR1 treatment alone. The shoot dry-weight gains on coinoculation with variable cell populations of B. thuringiensis-KR1 varied from 1.04 to 1.15 times and 1.03 to 1.06 times in pea and lentil respectively, while root dry weight ratios of coinoculated treatments varied from 0.98 to 1.14 times and 1.08 to 1.33 times in pea and lentil respectively, those of R. leguminosarum-PR1 inoculated treatment at 42 days of plant growth. While cell densities higher than 106 c.f.u. ml−1 had an inhibitory effect on nodulation and plant growth, lower inoculum levels resulted in decreased cell recovery and plant growth performance. The results of this study indicate the potential of harnessing endophytic bacteria of wild legumes for improving the nodulation and growth of cultivated legumes.  相似文献

Mesic–dry tundras are widespread in the Arctic but detailed assessments of net primary production (NPP) and ecosystem carbon (C) stocks are lacking. We addressed this lack of knowledge by determining the seasonal dynamics of aboveground vascular NPP, annual NPP, and whole-ecosystem C stocks in five mesic–dry tundras in Northern Sweden with contrasting microtopography, altitude, and dominant species. Those measurements were paralleled by the stock assessments of nitrogen (N), the limiting nutrient. The vascular production was determined by harvest or in situ growing units, whereas the nonvascular production was obtained from average species growth rates, previously assessed at the sites. Results showed that aboveground vascular NPP (15–270 g m−2), annual NPP (214–282 g m−2 or 102–137 g C m−2) and vegetation biomass (330–2450 g m−2) varied greatly among communities. Vegetation dominated by Empetrum hermaphroditum is more productive than Cassiope tetragona vegetation. Although the large majority of the apical NPP occurred in early-mid season (85%), production of stems and evergreen leaves proceeded until about 2 weeks before senescence. Most of the vascular vegetation was belowground (80%), whereas most of the vegetation production occurred aboveground (85%). Ecosystem C and N stocks were 2100–8200 g C m−2 and 80–330 g N m−2, respectively, stored mainly in the soil turf and in the fine organic soil. Such stocks are comparable to the C and N stocks of moister tundra types, such as tussock tundra. The green microalga Chlorella zofingiensis can produce the ketocarotenoid astaxanthin under heterotrophic culture conditions. Here we report the growth-associated biosynthesis of astaxanthin in this biotechnologically important alga. With glucose as sole carbon and energy source, C. zofinginesis grew fast in the dark with rapid exhaustion of nitrogen and carbon sources from media, leading to a high specific growth rate (0.034 h−1). Cultures started at a cell concentration of about 3.4 × 109 cells l−1 reached, after 6 days, standing biomass values of 1.6 × 1011 cells or 8.5 g dry weight l−1. Surprisingly, the biosynthesis of astaxanthin was found to start at early exponential phase, independent of cessation of cell division. A general trend was observed that the culture conditions benefiting cell growth also benefited astaxanthin accumulation, indicating that astaxanthin was a growth-associated product in this alga. The maximum cell dry biomass and astaxanthin yield were 11.75 g l−1 and 11.14 mg l−1 (about 1 mg g−1), simultaneously obtained in the fed-batch culture with a combined glucose–nitrate mixture addition, which were the highest ever reported in dark-heterotrophic algal cultures. The possible reasons why dark-heterotrophic C. zofingiensis could produce astaxanthin during the course of cell growth were discussed.  相似文献

The spatial and temporal variation of microphytobenthic biomass in the nearshore zone of Martel Inlet (King George Island, Antarctica) was estimated at several sites and depths (10–60 m), during three summer periods (1996/1997, 1997/1998, 2004/2005). The mean values were inversely related to the bathymetric gradient: higher ones at 10–20 m depth (136.2 ± 112.5 mg Chl a m−2, 261.7 ± 455.9 mg Phaeo m−2), intermediate at 20–30 m (55.6 ± 39.5 mg Chl a m−2, 108.8 ± 73.0 mg Phaeo m−2) and lower ones at 40–60 m (22.7 ± 23.7 mg Chl a m−2, 58.3 ± 38.9 mg Phaeo m−2). There was also a reduction in the Chl a/Phaeo ratio with depth, from 3.2 ± 3.2 (10–20 m) to 0.7 ± 1.0 (40–60 m), showing a higher contribution of senescent phytoplankton and/or macroalgae debris at the deeper sites and the limited light flux reaching the bottom. Horizontal differences found in the biomass throughout the inlet could not be clearly related to hydrodynamics or proximity to glaciers, but with sediment characteristics. An inter-summer variation was observed: the first summer presented the highest microphytobenthic biomass apparently related to more hydrodynamic conditions, which causes the deposition of allochthonous material.  相似文献

Summary An efficient protocol for in vitro propagation of an aromatic and medicinal herb Ocimum basilicum L. (sweet basil) through axillary shoot proliferation from nodal explants, collected from field-grown plants, is described. High frequency bud break and maximum number of axillary shoot formation was induced in the nodal explants on Murashige and Skoog (1962) medium (MS) containing N6-benzyladenine (BA). The nodal explants required the presence of BA at a higher concentration (1.0 mg·l−1, 4.4 μM) at the initial stage of bud break; however, further growth and proliferation required transfer to a medium containing BA at a relatively low concentration (0.25 mg·gl−1, 1.1 μM). Gibberellic (GA3) at 0.4 mg·l−1 (1.2 μM) added to the medium along with BA (1.0 mg·l−1, 4.4 μM) markedly enhanced the frequency of bud break. The shoot clumps that were maintained on the proliferating medium for longer durations, developed inflorescences and flowered in vitro. The shoots formed in vitro were rooted on half-strength MS supplemented with 1.0 mg·l−1 (5.0 μM) indole-3-butyric acid (IBA). Rooted plantlets were successfully acclimated in vermi-compost inside a growth chamber and eventually established in soil. All regenerated plants were identical to the donor plants with respect to vegetative and floral morphology.  相似文献

The effect of the root-inoculum size and axuin concentration on growth of adventitious roots and accumulation of ginsenosides were studied during suspension cultures of ginseng (Panax ginseng C.A. Meyer). Of the various concentrations of indole-3-butyric acid (IBA) and γ-naphthaleneacetic acid (NAA) used as supplementary growth regulators along with Murashige and Skoog medium, 25 μM IBA was found suitable for lateral root induction and growth, as well as accumulation of ginsenosides. Inoculum size of 5 g L−1 was found suitable for optimal biomass (10.5 g L−1 dry biomass) and ginsenosides (5.4 mg g−1 DW) accumulation. Of the various length of root inocula tested (chopped to 1–3, 4–6, 7–10 mm and un-chopped), root inocula of 7–10 mm was found suitable for biomass and ginsenoside accumulation.  相似文献

In vitro cultured plants from Oldenlandia affinis were established from seeds and grown on a hormone-free medium. In vitro plants produced the cyclic peptide kalata B1 in concentrations of 0.67 mg g−1 dry weight after growth of 30 days. This was approximately 50% of the concentration analysed in green house plants (shoot tips), where different concentrations have been determined in leaves (1.82 mg g−1), shoot tips (1.36 mg g−1), stems (0.36 mg g−1), and in flowers (0.16 mg g−1). Callus and cell suspension cultures could be initiated from aseptic root, stem and leaf explants of O. affinis seedlings and plants. Different light intensities were shown to affect culture growth as well as chlorophyll synthesis. The friable callus was then used for the establishment of a cell suspension culture. Fresh and dry weight measurements showed that growth was optimal on MS medium supplemented with 0.4 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-d). Leaf suspensions cultured on this medium showed a 4-fold increase of biomass by the first week of incubation. No quantifiable amounts of kalata B1 were produced under these conditions. Morphological differentiation seems to be essential for cyclic peptide production. Therefore, several undifferentiated as well as organised cell lines of O. affinis have been developed. These cell lines will constitute a worthwhile starting point for the optimisation of kalata B1 synthesis in liquid media to the objective of producing cyclic peptides under controlled and defined conditions in bioreactors.  相似文献
