Forest soils and canopies are major components of ecosystem CO2 and CH4 fluxes. In contrast, less is known about coarse woody debris and living tree stems, both of which function as active surfaces for CO2 and CH4 fluxes. We measured CO2 and CH4 fluxes from soils, coarse woody debris, and tree stems over the growing season in an upland temperate forest. Soils were CO2 sources (4.58 ± 2.46 µmol m?2 s?1, mean ± 1 SD) and net sinks of CH4 (?2.17 ± 1.60 nmol m?2 s?1). Coarse woody debris was a CO2 source (4.23 ± 3.42 µmol m?2 s?1) and net CH4 sink, but with large uncertainty (?0.27 ± 1.04 nmol m?2 s?1) and with substantial differences depending on wood decay status. Stems were CO2 sources (1.93 ± 1.63 µmol m?2 s?1), but also net CH4 sources (up to 0.98 nmol m?2 s?1), with a mean of 0.11 ± 0.21 nmol m?2 s?1 and significant differences depending on tree species. Stems of N. sylvatica, F. grandifolia, and L. tulipifera consistently emitted CH4, whereas stems of A. rubrum, B. lenta, and Q. spp. were intermittent sources. Coarse woody debris and stems accounted for 35% of total measured CO2 fluxes, whereas CH4 emissions from living stems offset net soil and CWD CH4 uptake by 3.5%. Our results demonstrate the importance of CH4 emissions from living stems in upland forests and the need to consider multiple forest components to understand and interpret ecosystem CO2 and CH4 dynamics.  相似文献

To determine the effects of vermicompost leachate (VCL) on resistance to salt stress in plants, young tomato seedlings (Solanum lycopersicum, cv. Ailsa Craig) were exposed to salinity (150 mM NaCl addition to nutrient solution) for 7 days after or during 6 mL L??1 VCL application. Salt stress significantly decreased leaf fresh and dry weights, reduced leaf water content, significantly increased root and leaf Na+ concentrations, and decreased K+ concentrations. Salt stress decreased stomatal conductance (gs), net photosynthesis (A), instantaneous transpiration (E), maximal efficiency of PSII photochemistry in the dark-adapted state (Fv/Fm), photochemical quenching (qP), and actual PSII photochemical efficiency (ΦPSII). VCL applied during salt stress increased leaf fresh weight and gs, but did not reduce leaf osmotic potential, despite increased proline content in salt-treated plants. VCL reduced Na+ concentrations in leaves (by 21.4%), but increased them in roots (by 16.9%). VCL pre-treatment followed by salt stress was more efficient than VCL concomitant to salt stress, since VCL pre-treatment provided the greatest osmotic adjustment recorded, with maintenance of net photosynthesis and K+/Na+ ratios following salt stress. VCL pre-treatment also led to the highest proline content in leaves (50 µmol g??1 FW) and the highest sugar content in roots (9.2 µmol g??1 FW). Fluorescence-related parameters confirmed that VCL pre-treatment of salt-stressed plants showed higher PSII stability and efficiency compared to plants under concomitant VCL and salt stress. Therefore, VCL represents an efficient protective agent for improvement of salt-stress resistance in tomato.  相似文献

Impact of different levels of elevated CO 2 on the activity of Frankia (Nitrogen-fixing actinomycete) in Casuarina equisetifolia rooted stem cuttings has been studied to understand the relationship between C. equisetifolia, Frankia and CO2. The stem cuttings of C. equietifolia were collected and treated with 2000 ppm of Indole Butyric Acid (IBA) for rooting. Thus vegetative propagated rooted stem cuttings of C. equisetifolia were inoculated with Frankia and placed in the Open top chambers (OTC) with elevated CO2 facilities. These planting stocks were maintained in the OTC for 12 months under different levels of elevated CO2 (ambient control, 600 ppm, 900 ppm). After 12 months, the nodule numbers, bio mass, growth, and photosynthesis of C. equisetifolia rooted stem cuttings inoculated with Frankia were improved under 600 ppm of CO2. The rooted stem cuttings of C. equisetifolia inoculated with Frankia showed a higher number of nodules under 900 ppm of CO2 and cuttings without Frankia inoculation exhibited poor growth. Tissue Nitrogen (N) content was also higher under 900 ppm of CO2 than ambient control and 600 ppm levels. The photosynthetic rate was higher (17.8 μ mol CO2 m?2 s?1) in 900 ppm of CO2 than in 600 ppm (13.2 μ mol CO2 m?2 s?1) and ambient control (8.3 μ mol CO2 m?2 s?1). This study showed that Frankia can improve growth, N fixation and photosynthesis of C. equietifolia rooted stem cuttings under extreme elevated CO2 level conditions (900 ppm).  相似文献

The Caulobacter crescentus (NA1000) xynB5 gene (CCNA_03149) encodes a predicted β-glucosidase-β-xylosidase enzyme that was amplified by polymerase chain reaction; the product was cloned into the blunt ends of the pJet1.2 plasmid. Analysis of the protein sequence indicated the presence of conserved glycosyl hydrolase 3 (GH3), β-glucosidase-related glycosidase (BglX) and fibronectin type III-like domains. After verifying its identity by DNA sequencing, the xynB5 gene was linked to an amino-terminal His-tag using the pTrcHisA vector. A recombinant protein (95 kDa) was successfully overexpressed from the xynB5 gene in E. coli Top 10 and purified using pre-packed nickel-Sepharose columns. The purified protein (BglX-V-Ara) demonstrated multifunctional activities in the presence of different substrates for β-glucosidase (pNPG: p-nitrophenyl-β-D-glucoside) β-xylosidase (pNPX: p-nitrophenyl-β-D-xyloside) and α-arabinosidase (pNPA: p-nitrophenyl-α-L-arabinosidase). BglX-V-Ara presented an optimal pH of 6 for all substrates and optimal temperature of 50 °C for β-glucosidase and α-l-arabinosidase and 60 °C for β-xylosidase. BglX-V-Ara predominantly presented β-glucosidase activity, with the highest affinity for its substrate and catalytic efficiency (Km 0.24 ± 0.0005 mM, Vmax 0.041 ± 0.002 µmol min?1 mg?1 and Kcat/Km 0.27 mM?1 s?1), followed by β-xylosidase (Km 0.64 ± 0.032 mM, Vmax 0.055 ± 0.002 µmol min?1 mg?1 and Kcat/Km 0.14 mM?1s?1) and finally α-l-arabinosidase (Km 1.45 ± 0.05 mM, Vmax 0.091 ± 0.0004 µmol min?1 mg?1 and Kcat/Km 0.1 mM?1 s?1). To date, this is the first report to demonstrate the characterization of a GH3-BglX family member in C. crescentus that may have applications in biotechnological processes (i.e., the simultaneous saccharification process) because the multifunctional enzyme could play an important role in bacterial hemicellulose degradation.  相似文献

In this study, we have compared the photosynthetic characteristics of two contrasting species of Tradescantia plants, T. fluminensis (shade-tolerant species), and T. sillamontana (light-resistant species), grown under the low light (LL, 50–125 µmol photons m?2 s?1) or high light (HL, 875–1000 µmol photons m?2 s?1) conditions during their entire growth period. For monitoring the functional state of photosynthetic apparatus (PSA), we measured chlorophyll (Chl) a emission fluorescence spectra and kinetics of light-induced changes in the heights of fluorescence peaks at 685 and 740 nm (F 685 and F 740). We also compared the light-induced oxidation of P700 and assayed the composition of carotenoids in Tradescantia leaves grown under the LL and HL conditions. The analyses of slow induction of Chl a fluorescence (SIF) uncovered different traits in the LL- and HL-grown plants of ecologically contrasting Tradescantia species, which may have potential ecophysiological significance with respect to their tolerance to HL stress. The fluorometry and EPR studies of induction events in chloroplasts in situ demonstrated that acclimation of both Tradescantia species to HL conditions promoted faster responses of their PSA as compared to LL-grown plants. Acclimation of both species to HL also caused marked changes in the leaf anatomy and carotenoid composition (an increase in Violaxanthin?+?Antheraxantin?+?Zeaxanthin and Lutein pools), suggesting enhanced photoprotective capacity of the carotenoids in the plants grown in nature under high irradiance. Collectively, the results of the present work suggest that the mechanisms of long-term PSA photoprotection in Tradescantia are based predominantly on the light-induced remodeling of pigment-protein complexes in chloroplasts.  相似文献

Irradiance continuously fluctuates during the day in the field. The speed of the induction response of photosynthesis in high light affects the cumulative carbon gain of the plant and could impact growth and yield. The photosynthetic induction response and its relationship with the photosynthetic capacity under steady-state conditions (P max) were evaluated in 37 diverse soybean [Glycine max (L.) Merr.] genotypes. The induction response of leaf photosynthesis showed large variation among the soybean genotypes. After 5 min illumination with strong light, genotype NAM23 had the highest leaf photosynthetic rate of 33.8 µmol CO2 m?2 s?1, while genotype NAM12 showed the lowest rate at 4.7 µmol CO2 m?2 s?1. Cumulative CO2 fixation (CCF) during the first 5 min of high light exposure ranged from 5.5 mmol CO2 m?2 for NAM23 to 0.81 mmol CO2 m?2 for NAM12. The difference in the induction response among genotypes was consistent throughout the growth season. However, there was no significant correlation between CCF and P max among genotypes suggesting that different mechanisms regulate P max and the induction response. The observed variation in the induction response was mainly attributed to ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) activation, but soybean lines differing in the induction response did not differ in the leaf content of Rubisco activase α- and β-proteins. Future studies will be focused on identifying molecular determinants of the photosynthetic induction response and determining whether this trait could be an important breeding target to achieve improved growth of soybeans in the field.  相似文献

In order to achieve recognition as environmentally friendly production, flue gases should be used as a CO2 source for growing the microalgae Chlorella sorokiniana when used for hydrogen production. Flue gases from a waste incinerator and from a silicomanganese smelter were used. Before testing the flue gases, the algae were grown in a laboratory at 0.04, 1.3, 5.9, and 11.0 % (v/v) pure CO2 gas mixed with fresh air. After 5 days of growth, the dry biomass per liter algal culture reached its maximum at 6.1 % CO2. A second experiment was conducted in the laboratory at 6.2 % CO2 at photon flux densities (PFD) of 100, 230, and 320 μmol photons m?2 s?1. After 4 days of growth, increasing the PFD increased the biomass production by 67 and 108 % at the two highest PFD levels, as compared with the lowest PFD. A bioreactor system containing nine daylight-exposed tubes and nine artificial light-exposed tubes was installed on the roof of the waste incinerator. The effect of undiluted flue gas (10.7 % CO2, 35.8 ppm NO x , and 38.6 ppm SO2), flue gas diluted with fresh air to give 4.2 % CO2 concentration, and 5.0 % pure CO2 gas was studied in daylight (21.4?±?9.6 mol photons m?2 day?1 PAR, day length 12.0 h) and at 135 μmol photons m?2 s?1 artificial light given 24 h day?1 (11.7?±?0.0 mol photons m?2 day?1 PAR). After 4 days’ growth, the biomass production was the same in the two flue gas concentrations and the 5 % pure CO2 gas control. The biomass production was also the same in daylight and artificial light, which meant that, in artificial light, the light use efficiency was about twice that of daylight. The starch concentration of the algae was unaffected by the light level and CO2 concentration in the laboratory experiments (2.5–4.0 % of the dry weight). The flue gas concentration had no effect on starch concentration, while the starch concentration increased from about 1.5 % to about 6.0 % when the light source changed from artificial light to daylight. The flue gas from the silicomanganese smelter was characterized by a high CO2 concentration (about 17 % v/v), low oxygen concentration (about 4 %), about 100 ppm NO x , and 1 ppm SO2. The biomass production using flue gas significantly increased as compared with about 5 % pure CO2 gas, which was similar to the biomass produced at a CO2 concentration of 10–20 % mixed with N2. Thus, the enhanced biomass production seemed to be related to the low oxygen concentration rather than to the very high CO2 concentration.  相似文献

Xylanases (EC 3.2.1.8) are hydrolytic enzymes that have found widespread application in the food, feed, and paper-pulp industries. Streptomyces sp. FA1 xynA was expressed as a secreted protein in Pichia pastoris, and the xylanase was applied to the production of Chinese steamed bread for the first time. The optimal pH and the optimal temperature of XynA were 5.5 and 60 °C, respectively. Using beechwood as substrate, the K m and V max were 2.408 mg mL?1 and 299.3 µmol min?1 mg?1, respectively. Under optimal conditions, a 3.6-L bioreactor produced 1374 U mL?1 of XynA activity at a protein concentration of 6.3 g L?1 after 132 h of fermentation. Use of recombinant XynA led to a greater increase in the specific volume of the CSB than could be achieved using commercial xylanase under optimal conditions. This study provides the basis for the application of the enzyme in the baking industry.  相似文献

The altitude-related responses to the increased application of CO2, N, and their combination were investigated in two Abies faxoniana populations, which originated from a subalpine coniferous forest at elevations of 2,580 and 3,200 m using closed-top chambers. The two contrasting populations were subjected to two CO2 regimes (350 and 700 µmol mol?1) and two N levels (0 and 5 g N m?2 year?1). Their net photosynthetic rate, non-structural carbohydrate concentration, and photosynthetic N use efficiency (PNUE) increased under elevated CO2. However, the increases detected in the high-elevation (HE) population were significantly greater than those found in the low-elevation (LE) population. Under elevated CO2 and N application, the maximal carboxylation rate (V cmax) increased in HE population, whereas no effects were found on V cmax in LE population. The C to N ratio decreased under N application in both populations. N application also induced the HE population to show greater increases in free amino acids, soluble proteins, N concentration, and PNUE than LE population. These results suggested that the population from HE was more sensitive to elevated CO2 and (or) N application than LE population. Results of this study provided valuable knowledge for predicting forest development under increased atmospheric CO2 concentration and (or) N deposition.  相似文献

The aim of this study was to evaluate the effects of different intensities and quality of light and explant type on the growth of and volatile compounds in Lippia gracilis in vitro. The treatments were as follows: light intensities of 26, 51, 69, 94, or 130 µmol m?2 s?1 from fluorescent lamps and light-emitting diode (LED) lamps at different wavelengths, namely, white, red, blue, and combinations of red and blue light at ratios of 2.5:1 and 1:2.5, respectively, and two explant types, namely, nodal and apical segments. On the 30th day of culture on half-strength Murashige and Skoog (Physiol Plant 15(3):473–497, 1962) medium, growth, production of photosynthetic pigments, chlorophyll a and b, total chlorophyll, carotenoids, and volatile constituents (using headspace gas chromatography-mass spectrometry) were analyzed. The light quality and intensity significantly influenced the in vitro growth of L. gracilis. The apical segments were superior in all parameters evaluated compared to nodal segments. The number of segments plantlet?1, root length, and leaf, shoot, root, and total weight were higher with increasing light intensity, especially under the 94 µmol m?2 s?1 treatment, for both explant types. The red light showed the highest leaf (32.28 mg plantlet?1) and total (58.33 mg plantlet?1) dry weight of all the light qualities. Major constituents, namely, ρ-cymene, γ-terpinene, thymol, carvacrol, and E-caryophyllene, were identified, regardless of light conditions. The amount and composition of volatile compounds varied according to light intensity and quality. Low intensity (26 µmol m?2 s?1) increased γ-terpinene content (12.42%) and concomitantly decreased carvacrol (38.52%). Blue LED light showed higher production of carvacrol (48.11%).  相似文献

L-asparaginase gene from Bacillus subtilis strain R5 (Asn-R5), comprising 990 nucleotides corresponding to a polypeptide of 329 amino acids, was cloned and expressed in Escherichia coli. Recombinant Asn-R5 was produced in soluble and active form exhibiting a specific activity of 223 μmol min?1 mg?1. The optimal temperature and pH for L-asparaginase activity of Asn-R5 were 35 °C and 9.0, respectively. Asn-R5 displayed a 50% activity with D-asparagine and 2% with L-glutamine compared to 100% with L-asparagine. No activity could be detected when D-glutamine was used as substrate. Half-life of the enzyme was 180 min at 35 °C and 40 min at 50 °C. There was no effect of metal ions and EDTA on the activity indicating that Asn-R5 enzyme activity is not metal ion dependent. The Km and Vmax values were 2.4 mM and 265 μmol min?1 mg?1, respectively. Activation energy for reaction catalyzed by Asn-R5 was 28 kJ mol?1. High L-asparaginase activity and thermostability of recombinant Asn-R5 may be beneficial for industrial production and application.  相似文献

The freshwater macrophyte, Ottelia alismoides, is a bicarbonate user performing C4 photosynthesis in the light, and crassulacean acid metabolism (CAM) when acclimated to low CO2. The regulation of the three mechanisms by CO2 concentration was studied in juvenile and mature leaves. For mature leaves, the ratios of phosphoenolpyruvate carboxylase (PEPC) to ribulose-bisphosphate carboxylase/oxygenase (Rubisco) are in the range of that of C4 plants regardless of CO2 concentration (1.5–2.5 at low CO2, 1.8–3.4 at high CO2). In contrast, results for juvenile leaves suggest that C4 is facultative and only present under low CO2. pH-drift experiments showed that both juvenile and mature leaves can use bicarbonate irrespective of CO2 concentration, but mature leaves have a significantly greater carbon-extracting ability than juvenile leaves at low CO2. At high CO2, neither juvenile nor mature leaves perform CAM as indicated by lack of diurnal acid fluctuation. However, CAM was present at low CO2, though the fluctuation of titratable acidity in juvenile leaves (15–17 µequiv g?1 FW) was slightly but significantly lower than in mature leaves (19–25 µequiv g?1 FW), implying that the capacity to perform CAM increases as leaves mature. The increased CAM activity is associated with elevated PEPC activity and large diel changes in starch content. These results show that in O. alismoides, carbon-dioxide concentrating mechanisms are more effective in mature compared to juvenile leaves, and C4 is facultative in juvenile leaves but constitutive in mature leaves.  相似文献

Effects of three levels of photosynthetic photon flux (PPF: 60, 160 and 300 μmol m−2s−1) were investigated in one-month-old Phalaenopsis plantlets acclimatised ex vitro. Optimal growth, chlorophyll and carotenoid concentations, and a high carotenoid:chlorophyll a ratio were obtained at 160 μmol m−2s−1, while net CO2 assimilation (A), stomatal conductance (g), transpiration rate (E) and leaf temperature peaked at 300 μmol m−2s−1, indicating the ability of the plants to grow ex vitro. Adverse effects of the highest PPF were reflected in loss of chlorophyll, biomass, non-protein thiol and cysteine, but increased proline. After acclimatisation, glucose-6-phosphate dehydrogenase, shikimate dehydrogenase, phenylalanine ammonia-lyase (PAL) and cinnamyl alcohol dehydrogenase (CAD) increased, as did lignin. Peroxidases (POD), which play an important role in lignin synthesis, were induced in acclimatised plants. Polyphenol oxidase (PPO) and β-glucosidase (β-GS) activities increased to a maximum in acclimatised plants at 300 μmol m−2s−1. A positive correlation between PAL, CAD activity and lignin concentration was observed, especially at 160 and 300 μmol m−2s−1. The study concludes that enhancement of lignin biosynthesis probably not only adds rigidity to plant cell walls but also induces defence against radiation stress. A PPF of 160 μmol m−2s−1was suitable for acclimatisation when plants were transferred from in vitro conditions.  相似文献

We investigated the effect of carbon dioxide (CO2)-ambient (350 µmol CO2 mol?1) and CO2-enriched (1500 µmol CO2 mol?1) conditions of in vitro photoautotrophic system on two cultivars, ‘RRIM600’ and ‘RRIT413’ of rubber tree (Hevea brasiliensis) in an acclimatization process of 45 days. Survival percentage of in vitro rubber tree plantlets derived from somatic embryos under ambient CO2 was better than those under CO2-enriched conditions, especially in cv. ‘RRIT413’. Subsequently, the survival rate of ex vitro transplanted plantlets was similar to the in vitro plantlets and abnormal morphological characters such as light-green leaves (SPAD), small leaves in cv. ‘RRIT413’ acclimatized under CO2-enriched conditions were demonstrated 30 days after the plantlets were transferred into the soil. Maximum quantum yield of PSII, photon yield of PSII, stomatal conductance and transpiration rate in cv. ‘RRIT413’ acclimatized under CO2-enriched conditions were sharply declined by 39.0, 50.6, 47.1 and 45.8%, respectively as compared to those acclimatized under ambient CO2 conditions. In contrast, the in vitro acclimatized plantlets of cv. ‘RRIM600’ were un-responsive under both ambient- and enriched-CO2 conditions. In conclusion, genotypic dependent in response to CO2 enriched conditions in in-vitro acclimatization of rubber tree plantlets was evidently demonstrated as a key result to regulate plant growth and development in ex vitro environments. Interestingly, soluble sugar contents (sucrose, glucose and fructose) were increased after transplanting the plantlets of cv. ‘RRIM600’ acclimatized under CO2-enriched condition into the soil and thus, can be considered as an adaptive indicator of ex vitro adaptation.  相似文献

Fucoxanthin is a carotenoid that exerts multiple beneficial effects on human health. However, reports comparing microalgae culture conditions and their effect on growth and fucoxanthin production are still limited. Isochrysis galbana and Phaeodactylum tricornutum cultures in different light (62.0, 25.9, 13.5, or 9.1 μmol photons m-2 s-1), mixing conditions (1 vvm aeration or 130 rpm agitation), and media compositions (F/2 and Conway medium) were studied for comparison of cellular growth and fucoxanthin production on F/2 medium. I. galbana showed a better adaptation to tested culture conditions in comparison with P. tricornutum, reaching 2.15?×?107?±?4.07?×?106 cells mL-1 and a specific growth rate (μ) of 1.12?±?0.05 day-1 under aerated conditions and 62.0 μmol photons m-2 s-1 light intensity. Fucoxanthin concentration was about 25 % higher in P. tricornutum cultures under 13.5 μmol photons m-2 s-1 light intensity and aerated conditions, but the highest fucoxanthin total production was higher in I. galbana, where 3.32 mg can be obtained from 1 L batch cultures at the 16th day under these conditions. Moreover, higher cell densities (~32.41 %), fucoxanthin concentration (~42.46 %), and total production (~50.68 %) were observed in I. galbana cultures grown in Conway medium, if compared with cultures grown in F/2 medium. The results show that the best growth conditions did not result in the best fucoxanthin production for either microalgae, implying that there is not a direct relationship between cellular growth and fucoxanthin production. Moreover, the results suggest that I. galbana cultures on Conway medium are strong candidates for fucoxanthin production, where 1.2 to 15 times higher fucoxanthin concentration are observed in comparison to macroalgal sources.  相似文献

Invasive plants can influence ecosystem processes such as greenhouse gas (GHG) emissions from wetland systems directly through plant-mediated transfer of GHGs to the atmosphere or through indirect modification of the environment. However, patterns of plant invasion often co-vary with other environmental gradients, so attributing ecosystem effects to invasion can be difficult in observational studies. Here, we assessed the impact of Phragmites australis invasion into native shortgrass communities on methane (CH4) emissions by conducting field measurements of CH4 emissions along transects of invasion by Phragmites in two neighboring brackish marsh sites and compared these findings to those from a field-based mesocosm experiment. We found remarkable differences in CH4 emissions and the influence of Phragmites on CH4 emissions between the two neighboring marsh sites. While Phragmites consistently increased CH4 emissions dramatically by 10.4 ± 3.7 µmol m?2 min?1 (mean ± SE) in our high-porewater CH4 site, increases in CH4 emissions were much smaller (1.4 ± 0.5 µmol m?2 min?1) and rarely significant in our low-porewater CH4 site. While CH4 emissions in Phragmites-invaded zones of both marsh sites increased significantly, the presence of Phragmites did not alter emissions in a complementary mesocosm experiment. Seasonality and changes in temperature and light availability caused contrasting responses of CH4 emissions from Phragmites- versus native zones. Our data suggest that Phragmites-mediated CH4 emissions are particularly profound in soils with innately high rates of CH4 production. We demonstrate that the effects of invasive species on ecosystem processes such as GHG emissions may be predictable qualitatively but highly variable quantitatively. Therefore, generalizations cannot be made with respect to invader-ecosystem processes, as interactions between the invader and local abiotic conditions that vary both spatially and temporally on the order of meters and hours, respectively, can have a stronger impact on GHG emissions than the invader itself.  相似文献

On the Brazilian coast, the red alga Gracilaria caudata J. Agardh is exploited for agar production. In view of its economic importance for potential mariculture, this work aimed to elucidate and characterize ecotypes along an extended Brazilian coastline by evaluating the effects of irradiance (70 and 150 μmol photons m?2 s?1) on the number of differentiated branches, growth rates (GRs), photosynthesis parameters, and pigment content of female gametophytes and tetrasporophytes from three distinct geographical areas: Ceará State (CE), Bahia State (BA), and São Paulo State (SP). Compared to low irradiance, high irradiance promoted higher GRs and a greater number of differentiated branches, as well as maximum electron transport rate (ETRmax) and maximum photosynthesis (P max) in both phases. However, irrespective of population or irradiance, tetrasporophytes had higher ETRmax (3.30 μmol e? m?2 s?1) than gametophytes (2.54 μmol e? m?2 s?1), corroborating the hypothesis that tetrasporophytes have better physiological performance than gametophytes, implicating correspondingly better adaptive abilities. Under high irradiance, regardless of the period of cultivation (14 or 28 days), strains from CE presented the highest GR (15% day?1) when compared to strains from SP (14% day?1) and BA (13% day?1). CE strains also had the highest number of differentiated branches and P max. Based on these results, we suggest that CE strains are the best candidates for future experimental mariculture tests. Importantly, overall differences in physiological performance among the strains from different populations give evidence of intraspecific diversity, thus supporting the hypothesis of ecotypic differentiation and allowing this study to define different G. caudata ecotypes.  相似文献

In the current scenario of climate change and increasing water scarcity there is an increased need to combine research efforts for the development of abiotic stress resistant crops, specifically plants able to support water deficit (WD). Polyamines (PAs) have been described as being involved in the regulation of many physiological processes and a variety of stress responses in plants. Arginine decarboxylase (ADC) is considered a key enzyme of the polyamine (PA) biosynthetic pathway. In this study, a T2 transgenic homozygous line of Medicago truncatula expressing the oat Adc under the control of CaMV 35S was obtained and was shown to have higher leaf accumulation of putrescine, spermidine and norspermidine compared to wild type plants. The photosynthetic parameters, leaf internal CO2 concentration (Ci), net CO2 assimilation rate (A), transpiration (E) and stomatal conductance (gs) of transformed and untransformed lines during WD and water deficit recovery experiments were measured by IRGA (infrared gas analyzer) and compared over time. Two light intensities were used, growth light intensity (391 μmol m?2 s?1) and saturating light intensity (1044 μmol m?2 s?1). Independently of the light intensity, and under WD, the transgenic line stood out with increased Ci, A, E and gs; suggesting a possible benefit of the augmented PAs under such disturbing environmental conditions. We showed that the constitutive expression of the oat Adc gene improve the physiological responses to WD and that WD recovered transgenic plants had higher seed yield, suggesting a possible benefit of PA metabolism manipulation in legumes.  相似文献

Palmitoleic acid (C16:1Δ9), contributes greatly to human health, industrial chemicals and biodiesel. The filamentous oleaginous microalgae Tribonema sp. has been identified as a highly efficient producer of palmitoleic acid. Temperature and light regime were adapted to regulate the palmitoleic acid content in this study. Strain T. minus was able to grow well at all the tested temperatures, even at 5 °C. The optimum temperature for palmitoleic acid accumulation (54.25 % of total fatty acid) was 25 °C. Moreover, both light intensity and photoperiod affect the growth, lipid content and fatty acid files of T. minus. The culture exposed to 240 μmol photons m?2 s?1 with a photoperiod of 24:0 showed the highest biomass (6.87 g L?1) and biggest lipid content (61.27 % of dry weight), whereas the most amount of palmitoleic acid (50.47 % of total fatty acid) was detected at 120 μmol photons m?2 s?1. These findings make tangible contributions to culture T. minus for commercial production of lipid or palmitoleic acid.  相似文献
