Chenopodium album pollen is one of the main sources of pollen allergy in desert and semi-desert areas and contains three identified allergens, so the aim of this study is comparison of the diagnostic potential of C. album recombinant allergens in an allergenic cocktail and C. album pollen extract. Diagnostic potential of the allergenic cocktail was investigated in 32 individuals using skin prick test and obtained results were compared with the acquired results from C. album pollen extract. Specific IgE reactivity against the pollen extract and allergenic cocktail was determined by ELISA and western blotting tests. Inhibition assays were performed for the allergenic cocktail characterization. The exact sensitization profile of all patients was identified which showed that 72, 81 and 46% of allergic patients had IgE reactivity to rChe a 1, rChe a 2 and rChe a 3, respectively. Almost all of C. album allergic patients (30/32) had specific IgE against the allergenic cocktail. In addition, there was a high correlation between IgE levels against the allergenic cocktail and IgE levels against the pollen extract. The allergenic cocktail was able to completely inhibit IgE binding to natural Che a 1, Che a 2 and Che a 3 in C. album extract. In addition, positive skin test reactions were seen in allergic patients that tested by the allergenic cocktail. The reliable results obtained from this study confirmed that the allergenic cocktail with high diagnostic potential could be replaced with natural C. album allergen extracts in skin prick test and serologic tests.  相似文献

The relevance of allergy skin prick testing in the diagnosis and treatment of seasonal allergic rhinitis and pollen asthma can usefully be interpreted in relation to the timing and duration of seasonal symptoms and the presence of pollen and mould spores in the air. This calendar has been constructed from three years continuous observations of pollen and Alternaria mould spore counts between January 1993 and December 1995, using a Burkard 7‐day volumetric spore trap. Of the total airborne pollen, tree pollen comprises 65%, weeds and herbs 11% and grasses 18%. Unidentified pollen, “other”; group, accounts for 6% of the total airborne pollen. The most numerous of the tree pollen is that of the introduced trees cypress (Cupressus spp.) and privet (Ligustrum spp.). Grass pollen is seen in small numbers throughout the winter but shows a rapid increase in spring to peak in mid to late November. Weeds pollinate from early spring through to summer. Alternaria mould, which is a risk factor for childhood asthma, occurs mainly in late spring and summer but is present in small numbers intermittently throughout winter.  相似文献

An aerobiological survey was performed in Berhampore town of West Bengal, India, to know the frequency of three common airborne pollen, namely Acacia auriculiformis, Eucalyptus citriodora and Madhuca indica using an ASTIR one day volumetric sampler. Acacia pollen showed its peak concentration in September, followed by Madhuca in April, while Eucalyptus showed its two peaks between September–October and January–April. Meteorological factors like temperature, RH, rainfall played an important role in release and dispersal of pollen. Skin prick tests with the antigenic extracts of the three pollen types, showed their allergenic potentialities. The highest markedly positive reactions were exhibited by Eucalyptus (34.04%), followed by Madhuca (22.93%) and Acacia (21.87%). 30–60% (NH4)2 SO4 cut fraction (Fraction II) of each pollen type showed maximum positivity in skin prick test. Biochemical analysis showed that Acacia pollen was richer in protein and carbohydrate, than the other two types. The total protein component of the above types were studied by SDS-PAGE showing different protein bands with a range of molecular weight 29–110 KD. In isolated fraction II (allergenically most potent) of Eucalyptus and Madhuca different protein band of 43–200 KD were obtained, while a single protein band of 57 KD was obtained for Acacia. The investigation of airborne pollen and fungalspore concentrations was carried out in Cracowbetween 1997–1999. For this study thevolumetric method has been employed (Burkard).At the same time the clinical diagnosis ofpollen allergy in 40 patients was obtained onthe basis of an interview, positive skin pricktests with pollen extracts and increasedspecific IgE level. An increase in seasonalallergy symptoms in all patients occurred fromthe middle of May to the middle of August.Eighty eight percent of the patients (35 out of40) were sensitive to Poaceae pollen and about50% of them were additionally sensitive totree and herb pollen excluding grasses. Forpatients with additional allergy to tree pollenthe seasonal symptoms started at the end ofMarch (Betula) while for patients withadditional allergy to herb pollen it wasextended to the middle of September (Artemisia).Five people out of 40 revealed positive skinreactions to Alternaria spores and anincrease in specific IgE level. Positive skinreaction to Cladosporium spores with noincrease in specific IgE level occurred in 2patients. The increase in seasonal allergysymptoms in people sensitive to Alternariaspores noted in July and August could becaused not only by these spores but also byPoaceae pollen.  相似文献

Summary Concentrations of airborne pollens recorded in Trieste in 1989 are evaluated in relation to allergic complaints in 113 patients with skin prick tests positive to one pollen species. Analysis of the result enable the two most important allergens to be confirmed as: Poaceae in 70.7% of patients and Parietaria in 18.6%. Few people had monosensitation to Compositae (Artemisia), Corylaceae and Fagaceae.Sympotoms are related to the flowering period when pollen levels climbed to daily averages of 15–20 grains/m3 but they started later than airborne peak concentrations and finished later than pollen decline. Pollen concentration recording can be a useful way to predict the clinical manifestation in sensitive patients but other factors are involved in determining symptoms like subjective mucosal reactions, polysensitization, patients living and working environment.  相似文献

The air that we inhale contains simultaneously a multiple array of allergenic pollen. It is well known that such allergens cause allergic reactions in some 15 of the population of the Western World. However little is known about the quantitative aspect of this phenomenon. What is the lowest concentration of pollen that might trigger allergic responses? As people are exposed to heterogeneous and variable environments, clarification of the partial contribution of each of the major airborne pollen allergens and determination of its role in invoking allergy are of prime importance. Objectives: (1) Assessment of a possible correlation between the concentration of airborne pollen and incidence of allergy. (2) Estimation of the lowest average concentrations for various species of airborne pollen that elicit allergic symptoms when exceeded. (3) Determination of the extent of the variations in manifestation of allergy symptoms that can be explained by fluctuations in the concentration of individual species of airborne pollen. Methods: The study was conducted during 14months with a rural population in Israel. The participants completed a detailed questionnaire and were skin prick tested with the common airborne allergens. The appearance of clinical symptoms, i.e. nasal, bronchial, ocular or dermal, were reported daily by the patients. Concentrations of the airborne pollen and spores were monitored in the center of activity of the residents during one day every week, using three Rotorod pollen traps. The pollen grains were identified by light microscopy. Results: The pollen spectrum was divided into time-blocks presenting the main pollination periods of the investigated species. The correlation between the concentration of airborne pollen of the relevant species and the clinical symptoms of the patients was determined for each time block. The correlation differed for different clinical symptoms and for different pollen allergens. Highest correlation with airborne pollen counts was found for patients with nasal and bronchial symptoms. The onset of the clinical symptoms by sensitive patients started, in each of the relevant groups, once the weekly average concentration of the airborne pollen crossed a threshold level. Under the limitations of the present study, this level was estimated to be 2–4 pollen m–3 air for olive, 3–5 pollen m–3 air for grasses, 4–5 pollen m–3 air for Artemisia, 10–20 pollen m–3 air for pecan and 50–60 pollen m–3 air for cypress. Conclusions: Fluctuations in specific airborne pollen grains explained up to 2/3 of the variation in clinical allergy responses. Those were: 69 of the variation for cypress (March–April), 66 for the grasses (March–April), 49 for the pecan (May–June) and 62 for Artemisia (Autumn).  相似文献

Zygophyllum fabago is a herbaceous plant withairborne pollen found widely in the MurciaRegion, in the Southeast of Spain. Although itsallergenicity has been recently reported,little is known of its involvement inpollinosis. Aerobiological study andsensitization in pollinotics weremeasured using a Hirst volumetric trap. Wehave measured the atmospheric concentrationsof this pollen and other allergenicpollen types in our region, between March 1993 andMarch 1997. Z. fabago pollen wascollected for a morphometric study of thepollen grain, and a lyophilized extract wasprepared for skin prick tests. We haveconducted skin tests with different pollen typesfrom our region and with Z. fabago in1736 patients with symptomssuggesting pollinosis. The size of the pollengrain averages 15.17 × 17.35 µm. Thepollination period extends from May to August,with a mean accumulated concentration of 448grains/m3. Out of 1736 pollinotics,263 (15.15%) showed a positive skin test forZ. fabago, 6 were monosensitized and 257were sensitized to other common pollen types fromour Region. Specific IgE to Z. fabago wasequal or higher than 0.35 ku/l in 86.56% ofsensitized patients. Chenopodiaceae pollinateduring spring and autumn and sensitize a largernumber of patients; Urticaceae reach thehighest pollen concentrations for a longerperiod but are not the primary cause ofpollinosis. This study shows that Z. fabagopollen becomes airborne, elicits an IgEresponse and, like other pollens, contributestowards triggering allergic symptoms.It should therefore be considered arelevant allergen and accordingly beincluded in skin test procedures.  相似文献

Abstract

Cupressaceae is one of the most widespread families in the Mediterranean region, widely used as ornamental trees both in cities and extra urban areas. The family is also known to produce a considerable amount of pollen grains. Nevertheless, Cupressaceae pollen is generally scarce in sediments and it is attested to be one of the most underrepresented pollen in ancient spectra. Cupressaceae pollen diffusion was detected by means of two sampling methods: moss cushions, which are able to accumulate pollen grains for several successive years, and volumetric sampler, which records the airborne pollen day by day. The study was conducted in the city of Florence (Italy), repeating moss samplings at the same sites for three times. The comparison between the two sampling methods revealed the low preservation of this pollen type and suggested that even low quantities of this pollen may indicate the local presence of the source plants. In order to test the contribution of the local pollen sources to the background of the airborne pollen, the pollen content of moss cushions was also related to the number of Cupressaceae plants surroundings each sampling site. The results indicate a noticeable influence of the plants growing in a short-range distance.  相似文献

Summary A comparison betweenParietaria pollen count and allergic symptoms of rhino-conjunctivitis in the early season was used in utilized to determine a «threshold-value» for this pollen. Clinical data were obtained from diary-cards of 34 allergic patients and pollen data from a volumetric sampling, carried out by means of a Hirst-Burkard pollen-trap. A significant correlation (r=0.98) was found between pollen count and symptom scores. Mild symptoms were registered with concentrations above 10–15 pollens/m3. Severe symptoms occurred when pollen count exceeded 80/m3/24 h., and over 90% of patients recorded symptoms. The importance of the late reactions and of the total allergenic airborne content are emphasized.  相似文献

The annual sums of daily airborne pollen concentrations fluctuate from year to year. It has been suggested that for some taxa there is a regular or alternating pattern in these fluctuations. On the other hand, environmental changes may lead to decreasing or increasing trends in airborne pollen concentrations. These two phenomena can only be studied reliably on the basis of long-term volumetric observations of abundantly occurring pollen types. For this study two arboreal (Betula and Quercus) and two herbaceous (Poaceae and Urtica) types were chosen.

For some of these pollen types a weak but significant trend is observed. For the arboreal types (Quercus and Betula) there appears to be a rather constant biennial fluctuating rhythm.  相似文献

This work aims to investigate the presence of airborne grass pollen and to identify antigenic and allergenic profiles from eight different grass species collected in the Porto region (Portugal). Poaceae airborne pollen, sampled using a Hirst-type volumetric trap during 2003–2007, was the second most abundant type, and high concentrations were found from April to August. Pollen proteins extracted from the eight grass species collected were separated by SDS-PAGE, being the allergenic profile investigated by immunoblotting using sera from atopic patients and maize profilin polyclonal antibody (ZmPRO3). Pollen extract profiles showed several bands ranging from 10 to 97 kDa. In immunoblotting studies, a low molecular weight protein (12–13 kDa) was recognized by profilin antibody. Also, in all pollen extracts except Zea mays, the IgE binding proteins of 12–13 kDa were detected in sera from the 25 patients with different sensitization profiles presenting high IgE values (>80 kU/l). This protein can be considered as a potential causal agent of the allergic respiratory diseases.  相似文献

Summary The present article reports on pollen embryogenesis resulting in haploid plants in 14 different F1 lines of Nicotiana tabacum. The response for pollen embryogenesis was high in cultures where the F1 lines were derived from mutants. Present observations are in agreement with previous reports on Secale cereale where one of the parents is from a mutant MII. These observations indicate that plant genotype is the main factor for differences in the induction of pollen embryogenesis.  相似文献

Airborne pollen is an important and potent source of aeroallergens. The aim of the study was to conduct a 2-year aerobiological survey in Calcutta, India, for knowing the concentration and seasonal periodicity of Lantana camara (LC) pollen. The sensitization due to this pollen among seasonal respiratory allergic patients and its chemical composition was studied. An aerobiological survey was conducted with a volumetric Burkard sampler from 2004 to 2006. Protein components of LC pollen were characterized by sodium dodecyl sulfate polyacrylamide gel electrophoresis and IgE immunoblotting. Allergenic activities were determined by in vivo (skin prick test) and in vitro (enzyme-linked immunosorbent assay. In vitro inhibition tests were performed to evaluate cross-reactivity. LC pollen was present from March to May and from September to December contributing up to 10.5% to the total aeropollen load during peak month. Horizontal profile showed highest concentration for nearest (0.5 m) rotorod and it was decreased by half in a distance within 4.5–6.5 m from plot edge. LC pollen contained 7.5% carbohydrate, 19.3% lipid with proline and valine as dominant amino acid. Among 1,500 adult respiratory allergic patients tested, 7.93% showed higher level of positive reaction. IgE binding proteins of 22, 42, 45 and 95 kD were revealed. LC pollen showed remarkable cross-reactivity with other local Verbenaceae pollen taxa (Clerodendron viscosum, Tectona grandis and Vitex negundo). This is the first study on LC pollen regarding its aerobiological, clinical and immuno-biochemical aspects; it should be helpful for the diagnosis and therapy of patients susceptible to LC pollen.  相似文献

Aerobiological sampling through volumetric pollen traps requires the use of an appropriate adhesive, whose selection is a critical factor in these studies. This study compares the differences between two adhesives that have been widely used in aerobiological studies with the aim of providing a persuasive argument for using petroleum jelly as a viable alternative to silicone diluted with the banned substance carbon tetrachloride. The study was performed using Hirst-type pollen traps in an indoor controlled environment to avoid interference attributed to outdoor meteorological parameters. Furthermore, a gravimetric study was conducted simultaneously as a control. Four different pollen types with different pollen sizes and similar flowering seasons were studied (Acer, Platanus, Quercus and Rumex). Differences between pollen traps were minimised by interchanging the drums in each pollen trap every 24 hours. Gravimetric results showed homogeneous airborne pollen distribution in the study. No statistically significant differences were found between the results for the sample adhesives for Quercus and Platanus, whereas significant differences were observed for Rumex and Acer. In the case of Acer, more pollen grains were recorded using the petroleum jelly, while for Rumex, more pollen grains were recorded with silicone. After this experiment we consider that petroleum jelly can be used as a viable alternative adhesive to the banned silicone diluted with carbon tetrachloride.  相似文献

Background

Current diagnostics for allergies, such as skin prick and radioallergosorbent tests, do not allow for inexpensive, high-throughput screening of patients. Additionally, extracts used in these methods are made from washed pollen that lacks pollen surface materials that may contain allergens.

Methodology/Principal Findings

We sought to develop a high-throughput assay to rapidly measure allergen-specific IgE in sera and to explore the relative allergenicity of different pollen fractions (i.e. surface, cytoplasmic, commercial extracts). To do this, we generated a protein microarray containing surface, cytoplasmic, and commercial extracts from 22 pollen species, commercial extracts from nine non-pollen allergens, and five recombinant allergenic proteins. Pollen surface and cytoplasmic fractions were prepared by extraction into organic solvents and aqueous buffers, respectively. Arrays were incubated with <25 uL of serum from 176 individuals and bound IgE was detected by indirect immunofluorescence, providing a high-throughput measurement of IgE. We demonstrated that the allergen microarray is a reproducible method to measure allergen-specific IgE in small amounts of sera. Using this tool, we demonstrated that specific IgE clusters according to the phylogeny of the allergen source. We also showed that the pollen surface, which has been largely overlooked in the past, contained potent allergens. Although, as a class, cytoplasmic fractions obtained by our pulverization/precipitation method were comparable to commercial extracts, many individual allergens showed significant differences.

Conclusions/Significance

These results support the hypothesis that protein microarray technology is a useful tool for both research and in the clinic. It could provide a more efficient and less painful alternative to traditionally used skin prick tests, making it economically feasible to compare allergen sensitivity of different populations, monitor individual responses over time, and facilitate genetic studies on pollen allergy.  相似文献

The aim of this study was to construct a picture of the influence of meteorological conditions on the start and duration of the airborne Betulaceae pollen season and the pollen concentrations in the atmosphere of Zagreb, Croatia. The study during three seasons (2002–2004) used a 7‐day Hirst‐type volumetric pollen and spore trap. Total annual airborne pollen of Alnus, Corylus and Betula greatly varied from year to year. The differences in the dates of onset of airborne pollen presence of Alnus, Corylus and Betula noted in Zagreb in 2002–2004 were controlled by weather conditions, particularly temperature and precipitation. In all years studied, airborne pollen peaks were recorded on days with temperature above 0°C and without or minimal precipitation. The mean number of days with airborne pollen concentrations exceeding levels which provoke symptoms of an allergic reaction was 15, 16 and 29 days for alder, hazel and birch, respectively. The results of the present study may provide useful data for allergologists to reach accurate diagnoses, and timely information on concentrations of airborne pollen types and concentrations for individuals with pollen hypersensitivity.  相似文献

Estimations based upon geostatistics and mapping have enabled the construction of a spatial model to predict the presence of biological particles in a particular region. This methodological proposal has been tested in a case study, at a regional scale, of airborne Olea pollen, using the data acquired from␣various sampling stations that are designed for the aerobiological monitoring of pollen levels. These sampling stations have been set up in cities throughout the region of Andalusia (southern Spain) at sites with very different characteristics in terms of biogeography, bioclimate, topography and vegetation. Pollen counts were made daily at all sites during 2003 using a volumetric spore-trap. Data were comparatively analysed in classical diagrams and by means of spatial-temporal maps. Space-time models were constructed using three coordinates, x, y (the UTM coordinates of each sampling station) and z, (the aerobiological data compiled for a specific period). The aerobiological data were interpolated by applying the traditional geostatistical method of Kriging. The introduction of the variable “space” into the model allowed us to predict pollen levels in different areas throughout the region. The interpolation method was used to make weekly estimations of Olea pollen values in areas where there was no aerobiological sampling station. In addition, the maps generated present a two-dimensional vision of the study area, showing that bioclimatic diversity of this region promotes a step-wise flowering of Olea.  相似文献

The aim of this study was to determine the onset, length and end of the ragweed pollen season, taking into account diurnal, day-to-day, monthly and annual pollen variations, the effect of some meteorological parameters on atmospheric pollen concentrations and possible differences in the airborne pollen season and concentration due to sampling site. Airborne pollen was collected at three sites in central Croatia (Zagreb, Samobor and Ivanić Grad) during three pollen seasons (2002–2004). Seven-day Hirst-type volumetric pollen traps were used for pollen sampling. Ragweed pollen was the third most abundant pollen type to occur in the atmosphere of central Croatia. Total Ambrosia pollen concentration was the highest in the 2003 pollen season and the lowest in 2004 at all sampling sites. Maximum emissions were restricted to August and September. Intradiurnal periodicity showed a peak from 1000 to 1200 hours. The concentration of ragweed pollen during the pollen season was greatly influenced by temperature and precipitation: on rainy days accompanied by temperature decline, the air pollen concentration decreased abruptly. The results of this study are aimed at helping to alleviate the symptoms of allergic reactions in individuals with ragweed pollen hypersensitivity, thus improving their quality of life.  相似文献

The present study seeks to compare daily and hourly airborne pollen concentrations at eight different stations in Castilla-La Mancha (central Spain) and Extremadura (south-western Spain) and assess pollen distribution sources. Sampling stations were located 69–440 km apart in a straight line in Albacete, Toledo, Talavera de la Reina and Ciudad Real in Castilla-La Mancha, and in Badajoz, Plasencia, Santa Amalia and Zafra in Extremadura. Airborne pollen was collected using Hirst-type volumetric spore traps. Quercus was the most abundant pollen type at all stations except for Ciudad Real, where Olea pollen predominated. Comparisons of daily data between pairs of stations revealed statistically significant positive correlations in all cases for Poaceae and Olea. Comparisons of hourly data between stations indicated greater differences than daily data. Analysis of correlation coefficients and straight-line distances between stations revealed a strong negative correlation. Analysis of total airborne pollen data for the eight sampling stations suggests that airborne pollen concentrations decrease from west to east and from south to north, partly reflecting dominant wind patterns. A clear correlation was observed between airborne pollen concentrations and the surface area covered by olive crops in a 50 km radius around the sampling stations.  相似文献
