We examined evolutionary mechanisms in the tetraploid Elymus caninus by comparing the phylogenetic relationships of 21 accessions suggested by sequence data from two single copy nuclear genes, the largest subunit of RNA polymerase II (RPB2) and phosphoenolpyruvate carboxylase (pepC), and one non-coding chloroplast region, TrnD/T. Elymus caninus is known combining two different genomes, an St genome and an H genome. Data from two single copy nuclear genes showed that there are two versions of the St genome in the species, St1 and St2. Most accessions combined one of these versions with an H genome version but two accessions had both versions of the St sequence for RPB2. This suggests that the RPB2gene may have been duplicated without chromosome doubling, possibly induced by transposable element. Our data also indicate that the H genome sequences in E. caninus have multiple origins, and a close phylogenetic relationship between Hordeum bogdanii and H sequences in some accessions of E. caninus. Thus, it is more likely that H. bogdanii is one of the major donors of the H copy in E. caninus. The maternal origin of E. caninus is the St genome species. There was no correlation between the geographic origin of the accessions and their sequence divergence.  相似文献

Artificial sexual hybrids between Elymus caninus (L.) L. and E. fibrosus (Schrebk) Tzvel. were experimentally examined in generations F1-F5. The possibility of genetic introgression between these species was shown. Morphologically, the hybrid plants can be assigned to either of the parental species or to variety E. caninus var. muticus (Holmb.) Karlsson. Some traits (spike density, leaf blade width, leaf blade pubescence, awns of lemmas) exhibited considerable variation. Polypeptide spectra of endosperm proteins were characterized in the initial parental biotypes and the hybrid progeny, using a gel-buffer SDS electrophoresis system. It was suggested that successful interspecies hybridization requires backcrosses or normalizing crosses. The possibility of sexual genetic exchange enables to utilize the gene pools of the two species to transfer the required traits in selection forage forms.  相似文献

Genetic diversity of 33 Elymus caninus accessions was investigated using isozyme, RAPD, and microsatellite markers. The three assays differed in the amount of polymorphism detected. Microsatellites detected the highest polymorphism. Six microsatellite primer pairs generated a total of 74 polymorphic bands (alleles), with an average of 15.7 bands per primer pair. Three genetic similarity matrices were estimated based on band presence or absence. Genetic diversity trees (dendrograms) were derived from each marker technique, and compared using Mantel's test. The correlation coefficients were 0.204, 0.267, and 0.164 between isozyme and RAPD distance matrices, RAPD and microsatellite distance matrices, and between isozyme and microsatellite distance matrices, respectively. The three methodologies gave differing views of the amount of variation present but all showed a high level of genetic variation in E. caninus. The following points may be drawn from this study whether based on RAPD, microsatellite, or isozyme data: (i) The Icelandic populations are consistently revealed by the three dendrograms. The congruence of the discrimination of this accession group by RAPD, microsatellite, and isozyme markers suggests that geographic isolation strongly influenced the evolution of the populations; (ii) The degree of genetic variation within accessions was notably great; and (iii) The DNA-based markers will be the more useful ones in detecting genetic diversity in closely related accessions. In addition, a dendrogram, which took into account all fragments produced by isozymes, RAPDs, and microsatellites, reflected better the relationships than did dendrograms based on only one type of marker.  相似文献

 Microsatellites are highly variable DNA sequences that can be used as markers for the genetic analysis of plants. The potential of microsatellite markers for use in a genetic diversity study in Elymus species was evaluated. Genomic libraries of Elymus caninus were constructed. The libraries were screened with two dinucleotide, (GA)n and (GT)n, and two trinucleotide repeats, (TCT)n and (CAC)n. A total of 19 positive clones were found for the two dinucleotide repeats; no positive clone was found for the trinucleotide repeats. Positive clones were sequenced to confirm the presence of microsatellites and to generate polymerase chain reaction (PCR) primers based on the sequences flanking the microsatellite. All sequenced (GA)n clones have repeats of n>10; over half of the (GT)n microsatellites have n<10 repeats. Primer pairs were designed and evaluated for 8 selected microsatellites. PCR products were amplified from 15 Elymus caninus accessions. The number of alleles found for the eight loci varied from 1 for ECGA89 and ECGT35 to 13 for ECGA22, as determined by non-denaturing polyacrylamide electrophoresis. Six microsatellite loci were found to be polymorphic in E. caninus. The eight primer pairs were tested on three other species; seven were successful in amplifying DNA from Elymus alaskanus and E. mutabilis, and four amplified DNA from E. caucasicus. The morphological similarity of Elymus trachycaulus to the Eurasian E. caninus has often been noted. This has lead to controversial and contradicting taxonomic treatments. Nevertheless, there has been no systematic investigation on molecular genetic similarity between E. trachycaulus and E. caninus. In this study, random amplified polymorphic DNA (RAPD) analysis was used to study the similarity between the two species. RAPD analysis of 38 samples representing E. caninus and E. trachycaulus complex yielded 111 interpretable RAPD bands. The Jaccard’s similarity values for E. caninus ranged from 0.38 between accessions H10345 and H10353 to 0.97 between accessions H8745 and H10096, with an average of 0.67. The Jaccard’s similarity values for E. trachycaulus complex ranged from 0.09 between E. trachycaulus ssp. subsecundus (PI 537321) and E. trachycaulus ssp. violaceus (PI 272612) to 0.78 between accessions PI 315368 and PI 372644, with an average of 0.43. The results from different analyses (NJ and PCA) were similar but not identical. The molecular genetic separation between E. caninus and E. trachycaulus was consistent. The PCA analysis clearly separated all E. caninus accessions from E. trachycaulus and its subspecies. The NJ analysis also showed separation between most accessions of E. caninus and E. trachycaulus. Further analysis excluding E. trachycaulus ssp. subsecundus and ssp. violaceus revealed that E. caninus species and E. trachycaulus species were clearly separated into two distinct groups. The RAPD data thus support the treatment of E. caninus and E. trachycaulus as distinct species. The analyses further indicate that E. violaceus is nested within E. trachycaulus, and more related to E. trachycaulus complex rather than to E. caninus.  相似文献

The AFLP method was used to study the inter-population variability of eight populations of Elymus caninus (L.) L. as well as three populations of Hordelymus europaeus (L.) Jess. ex Harz. In these studies a clear distinction was found between two phenotypes of E. caninus collected in the same locality. It also appeared that two populations of E. caninus representing the “pauciflorum” morphotype were clustered together, similarly as two populations of E. caninus exhibiting morphotype “caninus”. Additionally, the populations of the “pauciflorum” type were clustered together with all samples of H. europaeus. Furthermore, the same approach was applied to analyze the intra-population variability of E. caninus. The populations ranged from nearly uniform to as diverse as the samples collected from different localities. In some populations of this species the presence of off-type plants was revealed. Our data indicate the predominantly self-pollinating character of E. caninus and the possible genetic relationship between of E. caninus and H. europaeus. One of the challenges in evolutionary biology is to understand the evolution of speciation with incomplete reproductive isolation as many taxa have continued gene flow both during and after speciation. Comparison of population structure between sympatric and allopatric populations can reveal specific introgression and determine if introgression occurs in a unidirectional or bidirectional manner. Simple sequence repeat markers were used to characterize sympatric and allopatric population structure of plant species, Elymus alaskanus (Scribn. and Merr.) Löve, E. caninus L., E. fibrosus (Schrenk) Tzvel., and E. mutabilis (Drobov) Tzvelev. Our results showed that genetic diversity (HE) at species level is E. caninus (0.5355) > E. alaskanus (0.4511) > E. fibrosus (0.3924) > E. mutabilis (0.3764), suggesting that E. caninus and E. alaskanus are more variable than E. fibrosus and E. mutabilis. Gene flow between species that occurs within the same geographic locations versus gene flow between populations within species was compared to provide evidence of introgression. Our results indicated that gene flow between species that occur within the same geographic location is higher than that between populations within species, suggesting that gene flow resulting from introgressive hybridization might have occurred among the sympatric populations of these species, and may play an important role in partitioning of genetic diversity among and within populations. The migration rate from E. fibrosus to E. mutabilis is highest (0.2631) among the four species studied. Asymmetrical rates of gene flow among four species were also observed. The findings highlight the complex evolution of these four Elymus species.  相似文献

Genetic relationships among 24 Italian and Adriatic populations of barbs (genus Barbus ) were assessed using electrophoretic analysis of allozymes and mitochondrial DNA sequences of the cytochrome b gene. Results obtained with both markers were concordant, but they were not congruent with the current morphology-based systematics and taxonomy. Populations assigned to the same nominal taxa ( B. caninus , B. petenyi and B. rebeli ), were very divergent in both allozymes and mtDNA, indicating that these populations deserve recognition as different units for conservation and management. On the other hand, the two fluvio-lacustrine taxa considered as distinct species (i.e. B. plebejus and B. tyberinus ) are genetically very close to each other, showing no clear differences at either allozymes or mtDNA. The Pacific jack Caranx caninus is a common species fished by artisanal fishermen off the coast of Colima, México. During 2002, monthly samples of morphometric data and otoliths were taken to determine age and growth. Seven age groups were identified. The highest growth, 14.4 cm, takes place during the first year of life. During the second year, C caninus grows 11.76 cm; the third year 9.61 cm; the fourth 7.85 cm; the fifth 6.41 cm and sixth year 5.24 cm. The constants of von Bertalanffy's growth equation were: L(infinity) = 83.26 cm, W(infinity) = 18.138 g, K = 0.202, t(0) = -0.283 and A(0.95) = 15 years. Growth curves of other species of the same genus were calculated in order to compare them with the one obtained in the present work. The gonadosomatic index presented higher values during November and May. The periods of more intensive feeding are from August to February.  相似文献

Three types of markers-isozymes, RAPDs (random amplified polymorphic DNAs), and wheat microsatellites- were tested on wheat, spelt, and four wild wheat relatives (Aegilops cylindrica, Elymus caninus, Hordeum marinum, and Agropyron junceum). The aim was to evaluate their capability to provide specific markers for differentiation of the cultivated and wild species. The markers were set up for subsequent detection of hybrids and introgression of wheat DNA into wild relatives. All markers allowed differentiation of the cultivated from the wild species. Wheat microsatellites were not amplified in all the wild relatives, whereas RAPDs and isozymes exhibited polymorphism for all species. The dendrograms obtained with RAPD and isozyme data separated Swiss wheat cultivars from those collected in Austria and England, while no difference was found between Swiss spelt and wheat. RAPD data provided a weak discrimination between English and Austrian E. caninus. The microsatellite-based dendrogram discriminated populations of Ae. cylindrica, but no clear separation of H. marinum from E. caninus was revealed. The similarity matrices based on the three different sets of data were strongly correlated. The highest value was recorded between the matrices based on RAPDs and isozymes (Mantel's test, r = 0.93). Correlations between the similarity matrix based on microsatellites and matrices based on RAPDs and isozymes were lower: 0.74 and 0.68, respectively. While microsatellites are very useful for comparisons of closely related accessions, they are less suitable for studies involving less-related taxa. Isozymes provide interesting markers for species differentiation, but their use seems less appropriate for studies of within-species genetic variation. RAPDs can produce a large set of markers, which can be used for the evaluation of both between- and within-species genetic variation, more rapidly and easily than isozymes and microsatellites.  相似文献

The genome constitution of Icelandic Elymus caninus, E. alaskanus, and Elytrigia repens was examined by fluorescence in situ hybridization using genomic DNA and selected cloned sequences as probes. Genomic in situ hybridization (GISH) of Hordeum brachyantherum ssp. californicum (diploid, H genome) probe confirmed the presence of an H genome in the two tetraploid Elymus species and identified its presence in the hexaploid Elytrigia repens. The H chromosomes were painted uniformly except for some chromosomes of Elytrigia repens which showed extended unlabelled pericentromeric and subterminal regions. A mixture of genomic DNA from H. marinum ssp. marinum (diploid, Xa genome) and H. murinum ssp. leporinum (tetraploid, Xu genome) did not hybridize to chromosomes of the Elymus species or Elytrigia repens, confirming that these genomes were different from the H genome. The St genomic probe from Pseudoroegneria spicata (diploid) did not discriminate between the genomes of the Elymus species, whereas it produced dispersed and spotty hybridization signals most likely on the two St genomes of Elytrigia repens. Chromosomes of the two genera Elymus and Elytrigia showed different patterns of hybridization with clones pTa71 and pAes41, while clones pTa1 and pSc119.2 hybridized only to Elytrigia chromosomes. Based on FISH with these genomic and cloned probes, the two Elymus species are genomically similar, but they are evidently different from Elytrigia repens. Therefore the genomes of Icelandic Elymus caninus and E. alaskanus remain as StH, whereas the genomes of Elytrigia repens are proposed as XXH.  相似文献

In this study, we review the classification of two species, Elymus hispanicus and E. marginatus, which are restricted to highly valuable and sensitive Mediterranean ecosystems. The genomic composition of the two species is analysed by in situ hybridization. In addition, lodicule morphology and foliar anatomy of both species are compared with those of E. caninus, E. repens, E. sibiricus (i.e., the type species of Elymus s.s.) and Pseudoroegneria strigosa (i.e., the type species of Pseudoroegneria). The genomic formula 2n = 8x = 56; HStStSt is proposed for E. hispanicus and 2n = 4x = 28; StSt for E. marginatus. In this latter species, the absence of the ribosomal genes in one of the two St genomes suggests that diploidization may have occurred during the evolution the species. Regarding foliar anatomy, E. hispanicus, E. caninus, E. repens, and E. sibiricus shared several characteristics, but the leaf blades of E. marginatus proved anatomically more similar to those of Ps. strigosa. The data compiled support the contention that: (1) E. hispanicus belongs to Elymus s.s.; (2) E. marginatus should be transferred to Pseudoroegneria; and (3) the morphology of the lodicules should be carefully reconsidered for appropriately describing the boundaries between Elymus s.s. and Pseudoroegneria. The new combination Ps. marginata is proposed and a detailed iconography of the plant is provided.  相似文献

A serological survey for antibodies to Leptospira interrograns serovar hardjo was conducted on 574 serum samples from 10 native and 4 introduced wildlife species in south-eastern Australia. The microscopic agglutination (MA) test was used, and titres to hardjo antigen were detected in 33.5% of 352 brushtailed possums (Trichosurus vulpecula) sampled in several areas of Victoria. Prevalence of reactors ranged from 14 to 66% in 4 populations examined intensively. Serovar balcanica was isolated from possums with hardjo antibodies from two different areas. Of 20 wombats Vombatus ursinus) examined in Victoria, antibodies to hardjo were found in sera from 4 and titres to Pyrogenes and Pomona serogroups were detected in another. Hardjo antibodies were demonstrated in sera from 13 of 19 rusa deer (Cervus timorensis). Negative MA test results to hardjo antigens were recorded in 55 mountain possums (T. caninus), 63 macropods (Macropus spp.), 17 water rats (Hydrmys chrysogaster), 39 fallow deer (Dama dama), 2 hog deer (Axis porcinus) and 2 water buffalo (Bubalus bubalus). No MA antibodies to any of 16 leptospiral serogroups were detected in 17 water rats tested. Kidneys were examined from 330 of these animals and focal interstitial nephritis suggestive of leptospirosis was found in kidneys of 63 of 169 T. vulpecula, 3 of 55 T. caninus, 12 of 18 V. ursinus, 6 of 22 Macropus spp., 9 of 16 H. chrysogaster, 5 of 11 C. timorensis and 3 of 39 D. dama. A statistical association between focal interstitial nephritis and MA antibodies to hardjo was found in T. vulpecula.  相似文献

Phytochemical constituents isolated from Indian species of the genus Plectranthus reported up to 2009 are compiled. In India, the genus Plectranthus is found in all the habitats and altitudes, particularly in the Himalaya, the Southern Ghats, and the Nilgiri region. P. amboinicus, P. barbatus, P. caninus, P. mollis, P. coetsa, and P. incanus are the most common species found in India. Phytochemical studies of the genus revealed that Indian Plectranthus species are rich in essential oil, and that the most abundant secondary metabolites are diterpenoids, i.e., labdanes, abietanes, and ent-kauranes, as well as triterpenoids.  相似文献

Seven populations of Hordelymus europaeus and four populations of Leymus arenarius from Poland were subjected to examination of 36 morphological characters. This study showed that both species are relatively uniform and that morphological variation of their populations represents a continuum. Of those, three populations of either species were selected for analysis with molecular markers – RAPDs and AFLPs. These populations differed with respect of geographical location as well as syntaxa and habitat. RAPD-PCR was performed for individual plants and clearly grouped them according to the population origin. For either H. europaeus or L. arenarius - the studied populations differed in degree of their intrinsic variation while none of them as a whole was significantly different from the remaining ones. In AFLP analysis the studied populations were represented by DNA pools of several individual plants. Also this approach allowed discrimination among the population samples of both H. europaeus and L. arenarius. Both RAPDs and AFLPs were accordant in indication that H. europaeus exceeds L. arenarius with respect to variation accumulated at the DNA level. The metacercariae of Centrocestus formosanus, a minute intestinal trematode of mammals and birds, were detected in the freshwater fish, Puntius brevis, from Vientiane Municipality, Lao PDR. The metacercariae were experimentally fed to mice, and adult flukes were recovered in their small intestines 7 days later. The adult flukes were morphologically characterized by having 32 (rarely 34) circumoral spines arranged in 2 alternative rows, a large bipartite seminal vesicle, an oval-shaped ovary, and an X-shaped excretory bladder. Based on these characters, the adults were identified as Centrocestus formosanus (Nishigori, 1924). The taxonomic significance of C. formosanus, in relation to a closely related species, C. caninus (Leiper, 1913), is briefly discussed. It has been first verified by adult worm recovery that C. formosanus is prevalent in Vientiane areas of Lao PDR, taking the freshwater fish, P. brevis, as a second intermediate host.  相似文献

The ichthyofauna fished in Bahía de Navidad, Jalisco, Mexico in the Central Pacific was surveyed during 1998. Six thousand nine hundred and fourty-four organisms of 130 species were caught which weighted 3,231 kg. Nearly 30% of the species belonged to Carangidae. Haemulidae and Sciaenidae. The most important species in number and biomass were Microlepidotus brevipinnins, Caranx caninus and C. caballus. Species number, abundance and biomass fluctuated during the year. The largest number of species was caught in June (61), the minimum in March (33). January had the maximum abundance (1,397 organisms), while the minimum was obtained during August (251). The maximum biomass values were from January (556.5 kg), and the lowest from August (114.7 kg). Eighteen species accumulated 87% of the total abundance, while 20 species represented 86.3% of the biomass. There were large in differences species number, abundance and biomass nets of different mesh size.  相似文献

Seven tetraploid species ofElymus, viz.E. sibiricus, E. caninus, E. gmelinii, E. semicostatus, E. caucasicus, E. parviglume, andE. longearistatus subsp.canaliculatus, representing five sections were studied morphologically and used in interspecific hybridizations. The aim was to investigate whether the present sectional delimitation of the genus was in agreement with genomic data and if there was a correlation between genome constitution and morphology. The study revealed: (i) further information on the genomic affinities between the different species, (ii) that there is no congruence between genome constitution of the species and current sectional delimitation, and (iii) that there is a correlation between genome constitution and morphology in the palea apex shape and in the size of cilia of the palea.  相似文献
